Parallel evolution is common in nature and provides one of the most compelling examples of rapid environmental adaptation. In contrast to the recent burst of studies addressing genomic basis of parallel evolution, integrative studies linking genomic and phenotypic parallelism are scarce. Edaphic islands of toxic serpentine soils provide ideal systems for studying rapid parallel adaptation in plants, imposing strong, spatially replicated selection on recently diverged populations. We leveraged threefold independent serpentine adaptation of Arabidopsis arenosa and combined reciprocal transplants, ion uptake phenotyping, and available genome-wide polymorphisms to test if parallelism is manifested to a similar extent at both genomic and phenotypic levels. We found pervasive phenotypic parallelism in functional traits yet with varying magnitude of fitness differences that was congruent with neutral genetic differentiation between populations. Limited costs of serpentine adaptation suggest absence of soil-driven trade-offs. On the other hand, the genomic parallelism at the gene level was significant, although relatively minor. Therefore, the similarly modified phenotypes e.g., of ion uptake arose possibly by selection on different loci in similar functional pathways. In summary, we bring evidence for the important role of genetic redundancy in rapid adaptation involving traits with polygenic architecture.

Drought represents one of the most severe constraints to agricultural production; therefore, resistance to drought has the highest priority in the development of new cultivars. How insect pests respond to drought-resistant cultivars is poorly understood. In this study, by using the rose-grain aphid, Metopolophium dirhodum (Walker), in controlled laboratory conditions, we investigated the level of antibiosis of six commercially available cultivars of spring wheat (Triticum aestivum L.), which differed in their resistance to drought (drought-resistant cultivars: Septima, Jarissa and Seance; drought-sensitive cultivars: Quintus, Kabot and SW Kadrilj). Leaf morphological and structural traits were also measured for each cultivar to determine which leaf traits may be responsible for the observed differences. We used an age-stage, two-sex life table approach for comprehensive quantification of the aphid response to a particular cultivar and for projecting population growth. Although we found significant differences in all the life table and population growth parameters between the cultivars (SW Kadrilj was the most resistant to the aphids and Septima and Quintus were the most susceptible to the aphids), resistance to drought alone did not explain the observed variation. The cultivars also varied in most of the morphological and structural leaf traits measured. An increase in cuticle thickness at the cell junction above the major vein hampered the development of the aphid nymphs, which resulted in slower rates of population increase. Trichome density seemed to reduce the survival of the aphid nymphs. These two traits support not only drought resistance in general but also resistance to aphids and are potentially useful for selection in the future breeding of cereals.

Eutrophication has been suggested as a possible factor and nitrate, which is able to suppress non-symbiotic orchid seed germination even at very low concentrations, and could pose a serious threat for natural orchid populations. Early ontogenesis of all orchids entirely depends on orchid mycorrhizal symbiosis, and at this initial mycoheterotrophic stage, many terrestrial green orchids associate with polyphyletic fungal symbionts (i.e., mycobionts), collectively called “rhizoctonias.” We asked whether these fungi might also have some non-nutritional roles, i.e., whether they might confer resistance to eutrophication. To test this hypothesis, we co-cultivated seeds of the terrestrial orchid Dactylorhiza majalis with five rhizoctonias (two Tulasnella, two Ceratobasidium and one Serendipita isolate) at various ecologically meaningful nitrate concentrations (0 to 100 mg/L). With the exception of one Tulasnella isolate, all mycobionts supported the growth of protocorms and formed orchid mycorrhiza, i.e., intracellular hyphal pelotons, in the protocorms. Nitrate suppressed asymbiotic, as well as symbiotic, seed germination in all but one fungal treatment; the seeds co-cultivated with one of the Ceratobasidium isolates were indeed insensitive to nitrate. We conclude that nitrates also negatively affect symbiotic orchid germination, depending on the available compatible mycobionts. Thus, eutrophication with nitrate may decrease the number of orchid mycobionts capable of supporting seed germination.

We investigated selected species of the six subgenera of the genus Gunnera (Gunneraceae, Gunnerales, basal core eudicot) in order to confirm and to particularize for this taxon trait specifics on the occurrence of endodermis with Casparian bands in the organs of these representative species and to relate this endodermis to the structure of the vascular tissues. The general presence of endodermis with Casparian bands in stems and the vascular tissue arrangement varying from monostele to complex siphonostele is particularly specific for the genus. Petiolar endodermis was also characteristic and some species even had an endodermis surrounding veins in leaf lamina. Similarity of vascular tissue in stems and petioles in Gunnera species to seedless vascular plants is an interesting structural trait regarding phylogenic position of the genus. Only two species had simple, tetrarch to pentarch root vascular patterns common for eudicots, whereas all others were hexarch to multiple polyarch.

International audience; Background and AimsPyroloids, forest sub-shrubs of the Ericaceae family, are an important model for their mixotrophic nutrition, which mixes carbon from photosynthesis and from their mycorrhizal fungi. They have medical uses but are difficult to cultivate ex situ; in particular, their dust seeds contain undifferentiated, few-celled embryos, whose germination is normally fully supported by fungal partners. Their germination and early ontogenesis thus remain elusive.MethodsAn optimized in vitro cultivation system of five representatives from the subfamily Pyroloideae was developed to study the strength of seed dormancy and the effect of different media and conditions (including light, gibberellins and soluble saccharides) on germination. The obtained plants were analysed for morphological, anatomical and histochemical development.Key ResultsThanks to this novel cultivation method, which breaks dormancy and achieved up to 100 % germination, leafy shoots were obtained in vitro for representatives of all pyroloid genera (Moneses, Orthilia, Pyrola and Chimaphila). In all cases, the first post-germination stage is an undifferentiated structure, from which a root meristem later emerges, well before formation of an adventive shoot.ConclusionsThis cultivation method can be used for further research or for ex situ conservation of pyroloid species. After strong seed dormancy is broken, the tiny globular embryo of pyroloids germinates into an intermediary zone, which is functionally convergent with the protocorm of other plants with dust seeds such as orchids. Like the orchid protocorm, this intermediary zone produces a single meristem: however, unlike orchids, which produce a shoot meristem, pyroloids first generate a root meristem.

Root epidermis and apoplastic barriers (endodermis and exodermis) are the critical root structures involved in setting up plant-soil interface by regulating free apoplastic movement of solutes within root tissues. Probing root apoplast permeability with “apoplastic tracers” presents one of scarce tools available for detection of “apoplastic leakage” sites and evaluation of their role in overall root uptake of water, nutrients, or pollutants. Although the tracers are used for many decades, there is still not an ideal apoplastic tracer and flawless procedure with straightforward interpretation. In this article, we present our experience with the most frequently used tracers representing various types of chemicals with different characteristics. We examine their behaviour, characteristics, and limitations. Here, we show that results gained with an apoplastic tracer assay technique are reliable but depend on many parameters–chemical properties of a selected tracer, plant species, cell wall properties, exposure time, or sample processing.

There are various preparatory techniques for light microscopy permitting access to the inner structure of plant body and its development. Minute objects might be processed as whole-mount preparations, while voluminous ones should be separated into smaller pieces. Here we summarize some of the "classical" techniques to cut more voluminous objects into slices and access their inner structure either for simple anatomical analysis or for further processing (e.g., histochemistry, immunohistochemistry, in situ hybridization, enzyme histochemistry).

The multicellular plant body is a complex structure that is internally organized into organs and tissues specialized for particular functions. The outer boundary of a plant body is delimited by the epidermis, and its specific modifications allow controlled material exchange with the surrounding environment. The inner space of a plant body is subdivided into functional domains. Such division is known to take place in both the symplastic and apoplastic spaces of organs and tissues. The apoplast is composed mostly of intercellular spaces and porous cell walls. It surrounds the symplast, which is bordered by the plasma membrane. The internal subdivision of apoplastic space is carried out by so-called apoplastic barriers, which are cell layers with modified cell walls where lowered porosity decreases the passive flow of solutes, water, gasses, and regulatory molecules. There is a well-established role of the endodermis in the function of the root in vascular plants. However, the root endodermis is not the only “barrier” essential for plant function and development. Similar barriers are known to be present in stems, leaves, and the root periphery. This review focuses on the historical course of our understanding of the development, structure, and function of these protective layers.

Background and aims Root absorptive characteristics rely on the presence of apoplastic barriers. However, little is known about the establishment of these barriers within a complex root system, particularly in a major portion of them - the lateral roots. In Zea mays L., the exodermis differentiates under the influence of growth conditions. Therefore, the species presents a suitable model to elucidate the cross-talk among environmental conditions, branching pattern and the maturation of barriers within a complex root system involved in the definition of the plant-soil interface. The study describes the extent to which lateral roots differentiate apoplastic barriers in response to changeable environmental conditions. Methods The branching, permeability of the outer cell layers and differentiation of the endo- and exodermis were studied in primary roots and various laterals under different types of stress of agronomic importance (salinity, heavy metal toxicity, hypoxia, etc.). Histochemical methods, image analysis and apoplastic tracer assays were utilized. Key results The results show that the impact of growth conditions on the differentiation of both the endodermis and exodermis is modulated according to the type/diameter of the root. Fine laterals clearly represent that portion of a complex root system with a less advanced state of barrier differentiation, but with substantial ability to modify exodermis differentiation in response to environmental conditions. In addition, some degree of autonomy in exodermal establishment of Casparian bands (CBs) vs. suberin lamellae (SLs) was observed, as the absence of lignified exodermal CBs did not always fit with the lack of SLs. Conclusions This study highlights the importance of lateral roots, and provides a first look into the developmental variations of apoplastic barriers within a complex root system. It emphasizes that branching and differentiation of barriers in fine laterals may substantially modulate the root system-rhizosphere interaction.

Phragmites australis and Glyceria maxima are fast-growing littoral grasses often competing for similar wetland habitats. Eutrophication affects their competitiveness, but the outcome is not generally predictable due to the complexity of interrelated factors. We hypotheses that pore water N:P and NH4 +:NO3 − modify their growth and metabolic responses to the trophic status of the habitat. The hypothesis was tested under standardized conditions of long-term sand cultures. Application of N + P up to extreme levels in combination with N:P

Two co-occurring dominant wetland helophytes and potential competitors, Phragmites australis and Glyceria maxima , were cultivated under N, P availabilities simulating the trophic status of wetlands with different fertility (oligo- and eutrophic). The long-term outdoor cultivation was performed with the goal to characterise the extent to which the nutrient enrichment affects plant growth, phenology, and particularly, the accumulation of N storage compounds in belowground organs of wetland rhizomatous plants prior to the onset of winter dormancy. In the present study, both species responded similarly to nutrient surplus. The enhanced growth, delayed shoot senescence, and delayed retranslocation of N into belowground organs were found in both species in eutrophic treatment. Furthermore, N levels remaining in dry leaves were proportionally related to those in living ones, being significantly higher in eutrophic treatment. The efficiency of N retranslocation from senescing leaves varied around 60% in both species and treatments. The formation of N reserves was, however, not disrupted in either species. Although plants in eutrophic treatments accumulated N in their belowground organs significantly later in the season (in the September–December period), the amount of accumulated N was sufficient to reach high belowground N standing stock. Considering formation of N reserves, the differences in species response to treatments were negligible. Phragmites and Glyceria accumulated similar belowground N standing stock prior to the winter. Glyceria may, however, additionally profit from N standing stock of over-wintering green leaves and from the potential of growth and N assimilation during a mild winter period, which is not possible in fully dormant Phragmites .

The ability to cope with NH4+-N was studied in the littoral helophytes Phragmites australis and Glyceria maxima, species commonly occupying fertile habitats rich in NH4+ and often used in artificial wetlands. In the present study, Glyceria growth rate was reduced by 16% at 179 μM NH4+-N, and the biomass production was reduced by 47% at 3700 μM NH4+-N compared to NO3−-N. Similar responses were not found in Phragmites. The amounts (mg g−1 dry wt) of starch and total non-structural carbohydrates (TNC) in rhizomes were significantly lower in NH4+ (8.9; 12.2 starch; 20.1; 41.9 TNC) compared to NO3− treated plants (28.0; 15.6 starch; 58.5; 56.3 TNC) in Phragmites and Glyceria, respectively. In addition, Glyceria showed lower amounts (mg g−1 dry wt) of soluble sugars, TNC, K+, and Mg2+ in roots under NH4+ (5.6; 14.3; 20.6; 1.9) compared to NO3− nutrition (11.6; 19.9; 37.9; 2.9, for soluble sugars, TNC, K+, and Mg2+, respectively), while root internal levels of NH4+ and Ca2+ (0.29; 4.6 mg g−1 dry wt, mean of both treatments) were only slightly affected. In Phragmites, no changes in soluble sugars, TNC, Ca2+, K+, and Mg2+ contents of roots (7.3; 14.9; 5.1; 17.3; 2.6 mg g−1 dry wt, means of both treatments) were found in response to treatments. The results, therefore, indicate a more pronounced tolerance towards high NH4+ supply in Phragmites compared to Glyceria, although the former may be susceptible to starch exhaustion in NH4+-N nutrition. In contrast, Glyceria's ability to colonize fertile habitats rich in NH4+ is probably related to the avoidance strategy due to shallow rooting or to the previously described ability to cope with high NH4+ levels when P availability is high and NO3− is also provided.
