Nowadays there is a concern to improve the quality of education by including an interdisciplinary approach of concepts and their integration in the curriculum of scientific disciplines. The development of microbial fuel cells as a potential alternative for production of renewable energies gives undergraduate students the challenge of integrating interdisciplinary concepts in a hot topic of global interest as alternative energies. We present a laboratory experiment that has been part of a third-year undergraduate course in biology where students gained experience in assembling microbial fuel cells and the understanding of how they work. In this process, the students could integrate biological, biochemical, and electric concepts. In addition, the acquisition of manual skills and experimental design decisions are important for the development of future professionals.

Co-exposure soil studies of pollutants are necessary for an appropriate ecological risk assessment. Here, we examined the effects of two-component mixtures of metal oxide nanoparticles (NPs, ZnO or goethite) with chlorpyrifos (CPF) under laboratory conditions in short-term artificial soil assays using Eisenia andrei earthworms. We evaluated effects on metal accumulation, oxidative stress enzymes, and neurotoxicity related biomarkers in single and combined toxicity assays. Exposure to ZnO NPs increased Zn levels compared to control in single and combined exposure (ZnO NPs + CPF) at 72 h and 7 days, respectively. In contrast, there was no indication of Fe increase in organisms exposed to goethite NPs. One of the most notable effects on oxidative stress biomarkers was produced by single exposure to goethite NPs. This shows that the worms were more sensitive to goethite NPs than to ZnO NPs, regardless of nano-structure. Statistically significant differences between single and combined exposure were found for catalase and superoxide dismutase (goethite NPs) and for glutathione S-transferase (ZnO NPs) activities, mostly at 72 h. Acetylcholinesterase and carboxylesterase activities indicated that ZnO NPs were not neurotoxic to earthworms and similar degrees of inhibition were observed after single CPF and ZnO NPs + CPF exposure. These findings suggest a necessity to evaluate mixtures of NPs with co-existing contaminants in soil, and that the nature of metal oxide NPs and exposure time are relevant factors to be considered when assessing combined toxicity, as it may have an impact on ecotoxicological risk assessment.

Rapid diagnosis by using small, simple, and portable devices could represent one of the best strategies to limit the damage and contain the spread of viral, bacterial or protozoa diseases, principally when they can be transmitted by air and are highly contagious, as some respiratory viruses are. The presence of antibodies in blood or serum samples is not the best option for deciding when a person must be quarantined to stop transmission of disease, given that cured patients have antibodies, so the best diagnosis methods rely on the use of nucleic acid amplification procedures. Here we present a very simple device and detection principle, based on paper discs coupled to contactless conductivity (C4D) sensors, can provide fast and easy diagnostics that are needed when an epidemic outbreak develops. The paper device presented here solves one of the main drawbacks that nucleic acid amplification tests have when they are performed outside of central laboratories. As the device is sealed before amplification and integrally disposed in this way, amplimers release cannot occur, allowing repetitive testing in the physician’s practice, ambulances, or other places that are not prepared to avoid cross-contamination of new samples. The use of very low volume samples allows efficient reagent use and the development of low cost, simple, and disposable point-of-care diagnostic systems., Author summary In 2005, the World Health Organization (WHO) recognized Chagas Disease as a neglected tropical disease. Meanwhile the serological tests, recommended by WHO, can be performed for chronic disease diagnosis, the nucleic acid amplification tests must be performed for the detection of the acute phase of the disease. Although the existing laboratory diagnosis tests for Chagas Disease are sensitive and highly reproducible, they cannot be performed in rural, low infrastructure environments, where this disease prevails. In this sense, the use of simple and portable analytical devices may be able to offer an affordable solution to this problem, allowing fast sampling, diagnosis and treatment prescription in one simple and fast intervention, as the performed by short term medical missions. In this study we show for the first time a diagnosis test comprising low cost materials and employing a contactless and label-free conductivity detection system that is used to read the result of a nucleic acid amplification reaction. The test showed high sensitivity for Chagas Disease diagnosis showing the potential to be used in rural and low income places.

This study describes for the first time the development of 3D printed microfluidic devices with integrated electrodes for label-free counting of E. coli cells incorporated inside droplets based on capacitively coupled contactless conductivity detection (C4D). Microfluidic devices were fully fabricated by 3D printing in the T-junction shape containing two channels for disperse and continuous phases and two sensing electrodes for C4D measurements. The disperse phase containing E. coli K12 cells and the continuous phase containing oil and 1% Span® 80 were pumped through flow rates fixed at 5 and 60 μL min−1, respectively. The droplets with incorporated cells were monitored in the C4D system applying a 500-kHz sinusoidal wave with 1 Vpp amplitude. The generated droplets exhibited a spherical shape with average diameter of 321 ± 9 μm and presented volume of 17.3 ± 0.5 nL. The proposed approach demonstrated ability to detect E. coli cells in the concentration range between 86.5 and 8650 CFU droplet−1. The number of cells per droplet was quantified through the plate counting method and revealed a good agreement with the Poisson distribution. The limit of detection achieved for counting E. coli cells was 63.66 CFU droplet−1. The label-free counting method has offered instrumental simplicity, low cost, high sensitivity and compatibility to be integrated on single microfluidic platforms entirely fabricated by 3D printing, thus opening new possibilities of applications in microbiology.

In biohydrometallurgical processes, a mineral exhibits different oxidation phases due to the system's heterogeneity, especially in heap leach pads. Oxidation chemically modifies the mineral surface altering its interface, and thereby affecting the bacteria-mineral interaction, mineral reactivity and leaching velocity. Given that the mineral can be found in different oxidation states in heap bioleaching processes, three oxidation conditions of FeS2, in which iron and/or sulfur related compounds are formed on the mineral surface were assayed. This paper studies the interaction between the modified surface of a sulfide mineral, FeS2, and 0 K culture medium, typically used in biomining processes. Chemical and electrochemical changes on surfaces were characterized by subjecting them to weathering in an acidic culture medium (pH 1.8), without applying potential or current. The chemical species formed were identified by Raman spectroscopy. The modified pyrite surfaces showed significant interfacial transformations upon immersion in the culture medium, and the formation of passive chemical species, such as elemental sulfur, jarosite, phosphates and oxides, were identified. These interfacial modifications are correlated with changes in the open circuit potential (OCP) values during immersion of pyrite and surface modified pyrites in 0 K culture medium. Electrochemical characterization showed a decrease in mineral oxidation capacity, which directly affects the extent of leaching and possibly, of the interaction with other elements participating in the process, such as microorganisms. To study the interactions among bacteria and the pyrite mineral suffering different surface modifications, the attachment of the bioleaching bacterium Leptospirillum sp. was evaluated. A novel assay technique that involves quantification of lysozyme (Lys) through machine learning is put forward here. This article reports the tendency of the well- documented Ellington group anti-Lys aptamer, to produce aggregates when exposed to Lys. This property of apta-aggregation has been exploited here to develop an assay that quantifies the Lys using texture and area parameters from a photograph of the elliptical aggregate mass through machine learning. Two assay sets were made for the experimental procedure: one with high Lys concentration between 25–100 mM and another with low concentration between 1–20 mM. The high concentration set had a sample volume of 10 μl while the low concentration set had a higher sample volume of 100 μl, in order to obtain the statistical texture values reliably from the aggregate mass. The platform exhibited an experimental limit of detection of 1 mM and a response time of less than 10 seconds. Further, two potential operating modes for the aptamer were hypothesized for this aggregation property and the more accurate mode among the two was ascertained through bioinformatics studies.

Microbial fuel cells (MFCs) receive substantial research interest for their eco-friendly and energy conversion ability. Along with the production of electrical energy, a synergistic benefit of MFCs is also gained from their capability to reduce organic contaminant load in polluted water. The sub-categories of MFCs such as Sediment MFC and Plant MFC also have great prospects for bioremediation and energy conversion processes. However, the efficiency of these renewable energy-producing systems needs significant improvement for their real world application through commercial paradigm. This review mainly focuses on the power management systems (PMSs) that can be used in conjunction with the MFCs to improve its energy output for powering various ranges of electronic appliances. A PMS is a device that is constituted of some electronic components to work together in order to convert a low electrical input into a utilizable power output. The basic configuration of PMS mainly entails a charge pump, a supercapacitor, and a boost converter. Integration of these basic power management systems enables the enhancement of the output characteristic of MFCs to intermittently power small-scale appliances.

Hydrogen production was evaluated in two-chamber microbial electrolysis cells (MEC), where the chambers of the cell were separated using a new economical and environmentally friendly membrane made of poly (vinyl) alcohol/chitosan (PVA/CS). The MEC performance was compared to that of Nafion. The obtained results indicated that the MEC performance for hydrogen production did not show significant differences between the PVA/CS and Nafion membranes. MEC with PVA/CS showed the hydrogen production rate and hydrogen yield of 1277 ± 46 mL H2Lcat-1d-1 and 974 ± 116 mL H2 gacetate-1, respectively. The PVA/CS membrane allowed acetate removal that was 7% higher than that of Nafion due to the lower pH gradient and a lower voltage drop that increased the ion transfer across the membrane.

Three of the primary bottlenecks, which should be consider for practical, point-of-need use of microbial fuel cell (MFC) analytical devices were surpassed in this work: i) the use of a diffusive barrier, hence, an electrogenic biofilm; ii) longer enrichment/stabilization times to produce a biofilm, made in a laboratory environment, over the electrode; and iii) difficulty comparing results obtained from MFCs based on electrogenic biofilms with standardized bioassays, a setback to be adopted as a new method. Here we show an easy way to determine water toxicity employing planktonic bacteria as biorecognition agents. The paper-based MFC contain an electron carrier (or mediator) to facilitate charge transfer from bacteria to the anode. In this way, there is no need to use biofilms. As far as we know this is the first paper-based MFC containing P. putida KT2440, a well characterized non-pathogenic bacteria previously used in standardized water toxicity bioassays. Results were obtained in 80 min and an effective concentration 50 of 9.02 mg L−1, calculated for Zn2+ (a reference toxic agent), was successfully compared with previously published and ISO standardized bioassays, showing a promising future for this technology. The practical design and cost (less than one U.S. dollar) of the paper-based MFC toxicity test presented will open new market possibilities for rapid and easy-to-use MFC analytical devices.

Chalcopyrite is the main ore mineral used in industrial copper extraction. However, when passivation processes occur during hydrometallurgical treatment, a large percentage of the mineral being treated is not solubilized, so the copper recovery is limited. Galvanic interactions between semiconductor minerals are the basis of many strategies to achieve a more efficient process for increasing copper dissolution. These interactions concern generally two metallic sulfides. The present study describes a new galvanic interaction between chalcopyrite-magnetite (CuFeS2-Fe3O4) in an acid microbial culture medium, used routinely in biomining processes. The electrochemical characterization of CuFeS2, Fe3O4 and a mineral containing CuFeS2-Fe3O4 is performed. Galvanic interactions are demonstrated by comparing Evans diagrams constructed from current transients obtained by imposing the potential pulses to each species studied. It is determined that CuFeS2 and Fe3O4 fulfil the role of anode and cathode, respectively, in the behavior of the corresponding mineral. Stripping voltammetry is used to quantify electro-dissolved ions; the electrooxidation of CuFeS2-Fe3O4 mineral in acid culture medium releases twice as many copper ions as pure chalcopyrite. This corroborates that the galvanic interactions prevent the formation of typical passivating components observed in chalcopyrite. Fil: Saavedra Olaya, Albert Ulises. The aim of this work was to use electrochemical methods, capable to follow the corrosion of minerals, in order to monitor the progressive attack of the bacteria on the mineral. The assay was performed in a three electrode cell, with pyrite as the working electrode. The tests were performed in the absence and presence of iron; when present it was in low concentration. In order to compare the bacterial attack with other conditions, the study was conducted in three systems: live bacteria in culture media, dead bacteria in culture media, and sterile culture media, used as a control. The initial bacterial concentration was 106 bact.mL-1. To follow the process, current and corrosion potential were calculated. Live bacteria system showed a continuous increase of current with respect to the incubation time, being up to 4-times higher in the condition with iron (the corrosion current is related to the increase of the mineral area, produced by the bacterial attack, which was corroborated identifying by SEM the bacterial fingerprint on the mineral). Dead bacteria and sterile culture medium showed no current increase with respect to time. In addition, voltammetric studies shown that in live bacteria system the surface area increased when the biofilm was present. Whereas, in the dead bacteria system only the presence of some organic compounds interacting with the mineral was detected. The controls (sterile culture medium) showed the presence of iron hydroxides complexes and elemental sulfur, product of chemical leaching and the initial phase of a passivation process. With this study we demonstrated that the leaching process can be monitored by electrochemical methods, where the process of bacterial-mineral interaction could be followed, and also simultaneously identifying the initial processes of passivation. Our work can be useful for the development of a device for the in situ monitoring of biomining processes.

Microbial fuel cells (MFCs) can evolve in a viable technology if environmentally sound materials are developed and became available at low cost for these devices. This is especially important not only for the designing of large wastewater treatment systems, but also for the fabrication of low-cost, single-use devices. In this work we synthesized membranes by a simple procedure involving easily-biodegradable and economic materials such as poly (vinyl alcohol) (PVA), chitosan (CS) and the composite PVA:CS. Membranes were chemical and physically characterized and compared to Nafion®. Performance was studied using the membrane as separator in a typical H-Type MFCs showing that PVA:CS membrane outperform Nafion® 4 times (power production) while being 75 times more economic. We found that performance in MFC depends over interactions among several membrane characteristics such as oxygen permeability and ion conductivity. Moreover, we design a paper-based micro-scale MFC, which was used as a toxicity assay using 16 µL samples containing formaldehyde as a model toxicant. The PVA:CS membrane presented here can offer low environmental impact and become a very interesting option for point of need single-use analytical devices, especially in low-income countries where burning is used as disposal method, and toxic fluoride fumes (from Nafion®) can be released to the environment. Bacterial cell attachment that results in biofilm formation is the first step of bacteria-mineral interaction, and it is known that they strongly depend on the chemical characteristics of the mineral surface. In some industrial processes, like those used in biohydrometallurgy, the minerals are present in different chemical oxidation states, especially when heterogeneous low-grade ore deposits and mining tailings are used as starting materials for microbial inoculation. This study describes a strategy for monitoring bacterial attachment to pyrite (FeS2) and surface modified pyrite weathered in the culture growth medium used (pH 1.8), by means of a non-invasive electrochemical technique such as electrochemical impedance spectroscopy (EIS). The EIS evaluation of Leptospirillum sp. interaction with pyrite and surface modified pyrite weathered electrodes revealed significant changes at low frequencies, depending of the compounds presented over the four samples used, including unmodified pyrite and three surface modified samples. Once the frequency at which the adhesion process can become uncover was selected (0.1 and 0.05 Hz), the phase angle variation at such frequency was determined using different microbial concentrations. Different microbial attachment values were obtained for the different electrodes and related to the initial inoculum (2 × 108 cells/mL), as follow: FeS2/Fe(OH)n,S0 (38 ± 3.2%); FeS2/Fe(OH)n (31 ± 5.5%); FeS2 (27 ± 4.7%) and FeS2/S0 (18 ± 3.8%). Microbial attachment to unmodified and modified surfaces, evaluated by EIS, was corroborated with the traditional method of bacterial attachment evaluation by cell count in a Neubauer chamber (r2 = ~0.9). This strategy could be used in developing sensors for the fast and efficient bacterial attachment evaluation in minerals.

Spheres, tubes, and planar-shaped nanomaterials as Fe3O4 nanoparticles (MNPs), multiwalled carbon nanotubes (MWCNT), and graphene oxide (GO) were used for the first time to treat microfluidic paper-based analytical devices (μPADs) and create a biocompatible layer with high catalytic surface. Once glucose measurements are critical for diabetes or glycosuria detection and monitoring, the analytical performance of the proposed devices was studied by using bienzymatic colorimetric detection of this carbohydrate. The limit of detection values achieved for glucose with μPADs treated with MNPs, MWCNT, and GO were 43, 62, and 18 μM, respectively. The paper surface modification solves problems associated with the lack of homogeneity on color measurements that compromise the sensitivity and detectability levels in clinical diagnosis. Iron ions are monitored in processes such as bio-leaching, bio-oxidation, ferric leaching, passivation control, and others. The role of iron in different hydrometallurgy processes is very important because it affects strongly several industrial ore production, as chalcopyrite cooper extraction. In this work we present an amperometric FIA system that allows rapid quantification and speciation of iron in bio-mining processes. Linearity range, passivation of the working electrode and interferences were studied. We found a useful lineal range from 10 to 1500 mg L−1 for each of the ions, with a detection limit (Fe2+ and Fe3+) determined at 15 ± 2 mg L−1. High copper concentrations could interfere with the amperometric readings, though not in the copper/iron relationship encountered in this industry. Real samples, including an acid mine drainage sample, and the monitoring of the bio-oxidation kinetics of iron by Acidithiobacillus ferrooxidans were also quantified. In all cases, our results were compared with a standard colorimetric method that allows iron speciation (1,10-phenanthroline), showing good agreement between both methods. The electrochemical method presented here allows high sample throughput (ca. 45 samples h−1), fast analysis (ca. 1 min), and reagent free quantification of total iron, ferric and ferrous ions. Glassy carbon electrodes (GC) were modified with multiwalled carbon nanotubes (MWCNT/GC) and electrochemically treated first by applying an oxidation potential and then a reduction potential. The resulting electrodes were characterized via scanning electron microscopy, Raman spectroscopy, energy dispersive spectroscopy, and electrochemical techniques, particularly cyclic voltammetry using the redox probes Fe(CN)6 3− /4- and Ru(NH3)2+/3+ and electrochemical impedance spectroscopy using Fe(CN)6 3− /4-. These modified electrodes showed an electrochemical determination selective for dopamine (DA) and serotonin (5HT) in the presence of ascorbic acid (AA) and uric acid (UA), simultaneously measured, with a high reproducibility (an RSD of 1.7% for DA and 1.6% for 5HT) and a limit of detection (LOD) of 235 nmol L−1 for DA and 460 nmol L−1 for 5HT. The GC electrodes modified with oxidized MWCNT, subsequently reduced, showed higher selectivity towards the oxidation of DA and 5HT compared with GC bare electrodes or modified with MWCNT or oxidized MWCNT. Size, shape and surface characteristics strongly affect interfacial interactions, as the presented among iron oxide nanoparticles (NPs) aqueous colloids and bacteria. In other to find the forces among this interaction, we compare three types of surface modified NPs (exposing oxalate, arginine or cysteine residues), based on a simple synthesis and derivation procedure, that allows us to obtain very similar NPs (size and shape of the magnetic core). In this way, we assure that the main difference in the synthesized NPs are the oxalate or amino acid residue exposed, an ideal situation to compare their bacterial capture performance, and so too the interactions among them. Field emission scanning electron microscopy showed homogeneous distribution of particle sizes for all systems synthesized, close to 10 nm. Magnetization, zeta potential, Fourier transformed infrared spectrometry and other studies allow us further characterization. Capture experiments of Pseudomonas putida bacterial strain showed a high level of efficiency, independently of the amino acid used to wrap the NP, when compared with oxalate. We show that bacterial capture efficiency cannot be related mostly to the bacterial and NP superficial charge relationship (as determined by z potential), but instead capture can be correlated with hydrophobic and hydrophilic forces among them.

One limiting aspect to make microbial fuel cells (MFCs) a viable technology is to obtain low cost and environmentally sound materials for their components. In this work we synthesized membranes by a simple procedure involving low price and biodegradable materials such as poly (vinyl alcohol) (PVA), chitosan (CS) and PVA:CS, all cross-linked with sulfuric acid; they were compared to Nafion®, as our reference/control membrane. PVA:CS show lower oxygen permeability in comparison to Nafion® membranes, a strong advantage in order to maintain anaerobic conditions in the anodic compartment of MFCs. Membranes were characterized in typical H-Type MFCs, and results show that PVA:CS membranes outperform Nafion® 4 times (power production) while being 75 times more economic. Moreover, we design a paper-based micro-scale MFC, which was assayed as a toxicity biosensor; we obtained results in less than 20 min using 16 μL volume samples containing formaldehyde as a model toxicant. The PVA:CS membrane presented here can offer low environmental impact (materials, fabrication and disposal) and become a very interesting option for point of need single use disposable analytical devices.

Modification of cellulosic paper with carbon nanotubes (CNT) was studied for the development of electronic and analytical devices. Interesting results were published by using a CNT aqueous solution and the capillary forces of filter paper to make conductive tracks, supercapacitors, potentiometric electrodes and chemometric sensors. In this report, we show for the first time an electrochemical characterization of CNT-CS-SDS paper electrodes constructed with an ink containing optimized proportions of multi-wall CNT, chitosan (CS) and sodium dodecyl sulfate (SDS), and we compared our data with CNT-SDS paper electrodes constructed with a previously reported ink. We achieved better reversibility (ΔE=131±14 mV, CVs) and reproducibility (RSD=3.63 %) with CNT-CS-SDS paper electrodes, when compared to CNT-SDS paper electrodes (ΔE=249±7 mV; RSD=6.8 %) used as controls. When electrodes were fold at 90° angle, CNT-CS-SDS paper electrodes showed lower RSD than CNT-SDS paper electrodes, 8.43 % and 21.5 % respectively. These results are in concordance with SEM analysis indicating a dense CS film in CNT-CS-SDS paper electrodes. As a proof of concept, we determine dopamine concentration by DPV in the presence of ascorbic and uric acids, the limit of detection calculated was 6.32 μM. Moreover, a bismuth-film was prepared by in situ plating of Bi into CNT-CS-SDS paper electrodes. ASV allowed us to detect Pb in the presence of Bi (10–200 ppb) with a limit of detection of 6.74 ppb. The present study was aimed to test an electrochemical sensing approach for the detection of an active chemolithotrophic metabolism (and therefore the presence of chemolithotrophic microorganisms) by using the corrosion of pyrite by Acidithiobacillus ferrooxidans as a model. Different electrochemical techniques were combined with adhesion studies and scanning electron microscopy (SEM). The experiments were performed in presence or absence of A. ferrooxidans and without or with ferrous iron in the culture medium (0 and 0.5 g L−1, respectively). Electrochemical parameters were in agreement with voltammetric studies and SEM showing that it is possible to distinguish between an abiotically-induced corrosion process (AIC) and a microbiologically-induced corrosion process (MIC). The results show that our approach not only allows the detection of chemolithotrophic activity of A. ferrooxidans but also can characterize the corrosion process. Microbial fuel cells were rediscovered twenty years ago and now are a very active research area. The reasons behind this new activity are the relatively recent discovery of electrogenic or electroactive bacteria and the vision of two important practical applications, as wastewater treatment coupled with clean energy production and power supply systems for isolated low-power sensor devices. Although some analytical applications of MFCs were proposed earlier (as biochemical oxygen demand sensing) only lately a myriad of new uses of this technology are being presented by research groups around the world, which combine both biological?microbiological and electroanalytical expertises. This is the second part of a review of MFC applications in the area of analytical sciences. In Part I a general introduction to biological-based analytical methods including bioassays, biosensors, MFCs design, operating principles, as well as, perhaps the main and earlier presented application, the use as a BOD sensor was reviewed. In Part II, other proposed uses are presented and discussed. As other microbially based analytical systems, MFCs are satisfactory systems to measure and integrate complex parameters that are difficult or impossible to measure otherwise, such as water toxicity (where the toxic effect to aquatic organisms needed to be integrated). We explore here the methods proposed to measure toxicity, microbial metabolism, and, being of special interest to space exploration, life sensors. Also, some methods with higher specificity, proposed to detect a single analyte, are presented. Electrogenic bacteria are organisms that can transfer electrons to extracellular electron acceptors and have the potential to be used in devices such as bioelectrochemical systems (BES). In this study, Dietzia sp. RNV-4 bacterium has been isolated and identified based on its biochemical, physiological and morphological characteristics, as well as by its 16S rRNA sequence analysis. Furthermore, the current density production and electron transfer mechanisms were investigated using bioelectrochemical methods. The chronoamperometric data showed that the biofilm of Dietzia sp. RNV-4 grew as the current increased with time, reaching a maximum of 176.6 ± 66.1 mA/m2 at the end of the experiment (7 d); this highly suggests that the current was generated by the biofilm. The main electron transfer mechanism, indicated by the cyclic voltammograms, was due to secreted redox mediators. By high performance liquid chromatography, canthaxanthin was identified as the main compound involved in charge transfer between the bacteria and the solid electrodes. Dietzia sp. RNV-4 was used as biological material in a microbial fuel cell (MFC) and the current density production was 299.4 ± 40.2 mA/m2. This is the first time that Dietzia sp. Here the historical relevance of bioassays as a unique and relevant analytical method used to evaluate the dosage of clinical used substances, as well of subtle amounts of high bioactive substances (as hormones) is revised. Most of the relevant applications of bioassays are summarized; differences with other biological based analytical systems, as biosensors, is detailed. The future goals and possibilities of biological based systems are discussed.

In this work we present a device specifically designed to study microbial growth with several applications related to environmental microbiology and other areas of research as astrobiology. The Automated Measuring and Cultivation device (AMC-d) enables semi-continuous absorbance measurements directly during cultivation. It can measure simultaneously up to 16 samples. Growth curves using low and fast growing microorganism were plotted, including: Escherichia coli, and Haloferax volcanii, an halophilic archaeon., Comment: Draft version. This paper presents data obtained using an indigenous microbial community contained in anaerobic sediments (mud) collected from the shore of the Río de La Plata River (South America). After the sedimentary microbial fuel cells were assembled the evolution of current and power vs. time was studied. Two types of commercially available graphite materials were used as electrodes, which differ mainly in shape and size. In some experiments, an external carbon source (acetate) increased the power generation rate. The maximum power density observed in the aforementioned condition was 19.57 ± 0.35 and 8.72 ± 1.39 mW/m2 using rod and graphite disk electrodes, respectively. The better performance of the rod electrodes can be explained, at least in part, by an enhanced rate of mass transport by radial diffusion. This chapter presents a survey of developments and bioassay applications of individually addressable electrochemical arrays covering the past two decades and including over 90 references. Many different array designs have been fabricated, from macroscopic devices that have two to four sensing elements to ultramicroelectrode arrays containing as many as 12,000 individually addressable micrometer-sized working electrodes. Some inexpensively fabricated arrays are intended for single sample analysis and are disposable, while others are encapsulated for incorporation into reusable microfluidic systems. Devices and assays are grouped according to the types of modifications done to the sensing elements, and sections are included to consider unmodified elements as well as modifications with enzymes, antibodies or antigens, nucleic acids, in addition to viable cells and tissues. Some trends have been identified, and possible future directions are suggested.

The haloarchaea Natrialba magadii and Haloferax volcanii, as well as the radiation-resistant bacterium Deinococcus radiodurans, were exposed to vacuum-UV (V-UV) radiation at the Brazilian Synchrotron Light Laboratory (LNLS). Cell monolayers (containing 105 - 106 cells per sample) were prepared over polycarbonate filters and irradiated under high vacuum (10-5 Pa) with polychromatic synchrotron radiation. N. magadii was remarkably resistant to high vacuum with a survival fraction of ((3.77 \pm 0.76) x 10-2), larger than the one of D. radiodurans ((1.13 \pm 0.23) x 10-2). The survival fraction of the haloarchaea H. volcanii, of ((3.60 \pm 1.80) x 10-4), was much smaller. Radiation resistance profiles were similar between the haloarchaea and D. radiodurans for fluencies up to 150 J m-2. For fluencies larger than 150 J m-2 there was a significant decrease in the survival of haloarchaea, and in particular H. volcanii did not survive. Survival for D. radiodurans was 1% after exposure to the higher V-UV fluency (1350 J m-2) while N. magadii had a survival lower than 0.1%. Such survival fractions are discussed regarding the possibility of interplanetary transfer of viable micro-organisms and the possible existence of microbial life in extraterrestrial salty environments such as the planet Mars and the Jupiter's moon Europa. At present, dwarf M stars are being considered as potential hosts for habitable planets. However, an important fraction of these stars are flare stars, which among other kind of radiation, emit large amounts of UV radiation during flares, and it is unknown how this events can affect life, since biological systems are particularly vulnerable to UV. In this work we evaluate a well known dMe star, EV Lacertae (GJ 873) as a potential host for the emergence and evolution of life, focusing on the effects of the UV emission associated with flare activity. Since UV-C is particularly harmful for living organisms, we studied the effect of UV-C radiation on halophile archaea cultures. The halophile archaea or haloarchaea are extremophile microorganisms, which inhabit in hypersaline environments and which show several mechanisms to cope with UV radiation since they are naturally exposed to intense solar UV radiation on Earth. To select the irradiance to be tested, we considered a moderate flare on this star. We obtained the mean value for the UV-C irradiance integrating the IUE spectrum in the impulsive phase, and considering a hypothetical planet in the center of the liquid water habitability zone. To select the irradiation times we took the most frequent duration of flares on this star which is from 9 to 27 minutes. Our results show that even after considerable UV damage, the haloarchaeal cells survive at the tested doses, showing that this kind of life could survive in a relatively hostile UV environment.

In this work, two archaea microorganisms (Haloferax volcanii and Natrialba magadii) used as biocatalyst at a microbial fuel cell (MFC) anode were evaluated. Both archaea are able to grow at high salt concentrations. By increasing the media conductivity, the internal resistance was diminished, improving the MFC's performance. Without any added redox mediator, maximum power (P (max)) and current at P (max) were 11.87/4.57/0.12 μW cm(-2) and 49.67/22.03/0.59 μA cm(-2) for H. volcanii, N. magadii and E. coli, respectively. When neutral red was used as the redox mediator, P (max) was 50.98 and 5.39 μW cm(-2) for H. volcanii and N. magadii, respectively. In this paper, an archaea MFC is described and compared with other MFC systems; the high salt concentration assayed here, comparable with that used in Pt-catalyzed alkaline hydrogen fuel cells, will open new options when MFC scaling up is the objective necessary for practical applications.

Since the 1970's, when the Viking spacecrafts carried out experiments aimed to the detection of microbial metabolism on the surface of Mars, the search for nonspecific methods to detect life in situ has been one of the goals of astrobiology. It is usually required that the methodology can detect life independently from its composition or form, and that the chosen biological signature points to a feature common to all living systems, as the presence of metabolism. In this paper we evaluate the use of Microbial Fuel Cells (MFCs) for the detection of microbial life in situ. MFCs are electrochemical devices originally developed as power electrical sources, and can be described as fuel cells in which the anode is submerged in a medium that contains microorganisms. These microorganisms, as part of their metabolic process, oxidize organic material releasing electrons that contribute to the electric current, which is therefore proportional to metabolic and other redox processes. We show that power and current density values measured in MFCs using microorganism cultures or soil samples in the anode are much larger than those obtained using a medium free of microorganisms or sterilized soil samples, respectively. In particular, we found that this is true for extremophiles, usually proposed to live in extraterrestrial environments. As aerobic respiration proceeds it consumes oxygen and produces carbon dioxide; and the relation between these two parameters, the respiratory quotient (RQ), is related to the type of substances being respired. Therefore, for a given water or wastewater origin, a more or less stable RQ is expected, making possible the estimation of BOD5 by means of CO2 production measurement. The microbial breathing intensity was continuously measured using a potentiometric CO2 electrode as transducer. We named this new biosensor BODstCO2. Proof of concept in this study was carried out with Saccharomyces cerevisiae or a BODSEED mixed microbial community as a biological recognition component. The effect of microbial load (0.5, 5, and 25 mg cm−2 membrane, dry weight) over the apparent linear range (up to 670 mg L−1 BOD5, 5 mg cm−2 membrane), detection limit (ca. 1 mg L−1 BOD5), stabilization time, reproducibility (typically better than 10%) and bio-membrane type (membrane or PVA hydrogel entrapment) were studied. When the Nernstian biosensor response was used for calibration, up to 20,000 mg L−1 glucose standard was measured without sample dilution. BOD calibrations were accomplished using the two more commonly used standard artificial wastewaters, GGA and OECD solutions. The results showed that the potentiometric CO2 electrode was an useful transducer, allowing us to build, calibrate and characterize a BOD-like biosensor. Moreover, limitations present at oxygen amperometric electrode (customarily used as BOD biosensor-based transducer) such as oxygen low solubility and its reduction at the cathode were avoided.

Whereas biosensors have been usually proposed as analytical tools, used to investigate the surrounding media pursuing an analytical answer, we have used a biosensor-like device to characterize the microbial cells immobilized on it. We have studied the kinetics of transport and degradation of glucose at different concentrations and temperatures. When glucose concentrations of 15 and 1.5 mM were assayed, calculated activation energies were 25.2 and 18.4 kcal mol-1, respectively, in good agreement with previously published data. The opportunity and convenience of using Arrhenius plots to estimate the activation energy in metabolic-related processes is also discussed. Fil: Kormes, Diego J.. Universidad de Buenos Aires. Molecularly imprinted polymers (MIPs) are easily obtained by a non-covalent approach through the copolymerisation of suitable functional monomers and cross-linkers in the presence of the print molecule. Removal of the template leaves a polymer that selectively recognises it. However, experimental data have demonstrated that non-covalent molecularly imprinted polymers exhibit a heterogeneous binding sites distribution. In this work, two different imprinted polymers for chloramphenicol (CAP), obtained using different template concentrations, were evaluated for their kinetics and affinity binding properties against CAP in a batch approach. Experimental binding isotherms were fitted to Langmuir, Freundlich and Langmuir-Freundlich isotherms and the affinity distribution corresponding to the Freundlich isotherm was used to extract binding parameters. We describe a new, faster and convenient method to study some metabolic characteristics - by the successful application of immobilized yeast cells (S. cerevisiae) in a microbial biosensor-like device. Microbial biosensors consist of microorganisms immobilized on the surface of a membrane or in a gel, in close contact with a transducer. Almost all works published to date have used biosensors for analyses in which a concentration-related property of the external medium is measured. A different approach is presented here; we have successfully used S. cerevisiae and a carbon dioxide electrode as the main components of a biosensor-like device, used as a proof of concept, for a system useful to characterize metabolic parameters of the microbial cells immobilized on a carbon dioxide electrode. The biosensor-like device we are presenting allows us to calculate Michaelis-Menten parameters related to the kinetics of transport and degradation of several carbohydrates (i.e., glucose, fructose, galactose, sucrose and xylose, with K(m(app)) of 6.0, 5.8, 0.9, 2.0, and 147 mM, respectively), and the study of the kinetics of expression of non-constitutive proteins related to the transport and degradation of galactose.

Microbial fuel cells (MFCs) are bioelectrochemical systems (BES) capable of harvesting electrons from redox reactions involved in metabolism. In a previous work, we used chemoorganoheterotrophic microorganisms from the three domains of life - Bacteria, Archaea, and Eukarya - to demonstrate that these BES could be applied to the in situ detection of extraterrestrial life. Since metabolism can be considered a common signature of life "as we know it," we extended in this study the ability to use MFCs as sensors for photolithoautotrophic metabolisms. To achieve this goal, two different photosynthetic microorganisms were used: the microalgae Parachlorella kessleri and the cyanobacterium Nostoc sp. MFCs were loaded with nonsterilized samples, sterilized samples, or sterilized culture medium of both microorganisms. Electric potential measurements were recorded for each group in single experiments or in continuum during light-dark cycles, and power and current densities were calculated. Our results indicate that the highest power and current density values were achieved when metabolically active microorganisms were present in the anode of the MFC. Moreover, when continuous measurements were performed during light-dark cycles, it was possible to see a positive response to light. Therefore, these BES could be used not only to detect chemoorganoheterotrophic metabolisms but also photolithoautotrophic metabolisms, in particular those involving oxygenic photosynthesis. Additionally, the positive response to light when using these BES could be employed to distinguish photosynthetic from nonphotosynthetic microorganisms in a sample. Fil: Figueredo, Federico. The detection of pathogenic bacteria is key for the identification and prevention and of problems related to health and safety. Accurate and quick detection and quantification of pathogen microorganisms in food, drinking water and in clinical diagnostics using cost-effective methodologies are greatest challenges currently facing food industry, health and environmental fields. A rapid and sensitive biosensor, based on immunological methods, was developed for bacteria detection using LightScribe disc as platform. We are able to detect between 0 and 106 CFU/mL in only 1.5 hours without the need for sample pre-concentration. The cross-reactivity study and individual ready-to-eat meat samples were inoculated with live bacteria to demonstrate proof-of-concept for real samples.
