Using the Cap Analysis of Gene Expression (CAGE) technology, the FANTOM5 consortium provided one of the most comprehensive maps of transcription start sites (TSSs) in several species. Strikingly, ~72% of them could not be assigned to a specific gene and initiate at unconventional regions, outside promoters or enhancers. Here, we probe these unassigned TSSs and show that, in all species studied, a significant fraction of CAGE peaks initiate at microsatellites, also called short tandem repeats (STRs). To confirm this transcription, we develop Cap Trap RNA-seq, a technology which combines cap trapping and long read MinION sequencing. We train sequence-based deep learning models able to predict CAGE signal at STRs with high accuracy. These models unveil the importance of STR surrounding sequences not only to distinguish STR classes, but also to predict the level of transcription initiation. Importantly, genetic variants linked to human diseases are preferentially found at STRs with high transcription initiation level, supporting the biological and clinical relevance of transcription initiation at STRs. De som har en ställföreträdare, vilka kallas för huvudmän, är en sårbar grupp eftersom de i många fall är beroende av andra för att kunna klara sig i sin vardag. Människor som av fysiska eller psykiska orsaker inte kan föra sin egen talan i rättsliga frågor kan få hjälp i form av en ställföreträdare enligt föräldrabalken. En ställföreträdare träder in i huvudmannens ställe i en rad situationer där huvudmannen behöver hjälp. Huvudmännens relationer till ställföreträdare har stor betydelse för huvudmännens levnadsvillkor i vardagen. Syftet med denna förstudie har varit att undersöka huvudmännens förväntningar på och erfarenheter av att leva med hjälp från god man eller förvaltare för att kunna klara sig i sin vardag. Förstudiens frågeställningar är följande: Vilka förväntningar har huvudmännen på sin ställföreträdare? Vilka erfarenheter har huvudmännen av hjälp från ställföreträdare i sin vardag? Hur upplever huvudmännen tilltron till sin egen förmåga att kunna klara sig på egen hand i sin vardag? Hur upplever huvudmännen tilltron till sin egen förmåga att kunna påverka vilken hjälp de kan få av sin ställföreträdare? Vilka utvecklingsområden finns avseende hjälp genom ställföreträdarskap och huvudmännens tilltro till sin egen förmåga? Ett praktiskt och erfarenhetsbaserat lärande och utveckling har stått i centrum i förstudiens design redan från början genom ett praktiknära och deltagarbaserat forskningsperspektiv. Både tematiskt öppna frågor och slutna frågor användes vid datainsamling. Beroende på deltagarnas behov och kognitiva funktionsnedsättning gavs kompletterande förklaringar av frågorna. Huvudmännens förväntningar på och erfarenheter av att leva med hjälp från god man eller förvaltare för att kunna klara sig i sin vardag visar intressanta resultat. Likheter och skillnader om hjälp från ställföreträdare ger en bra grund för vidare utvecklingsarbeten. En generell slutsats är att det görs en del goda arbeten för huvudmännen, men det behövs en del förbättringar också. Följande rekommendationer vill vi särskilt uppmärksamma: - Goda exempel och framgångsfaktorer i ställföreträdarskap bör tas tillvara och synliggöras - Vad som ingår i ställföreträdarskap enligt bestämmelser bör tydliggöras mer både för huvudmännen och ställföreträdare - Utbildningsinsatser för ställföreträdare bör ses över och kunskaper om olika typer av funktionsnedsättning som huvudmännen har ges tillräckligt - En heterogen grupp framträder där tilltron till förmågan och förväntningar och erfarenheter varierar till en viss del, vilket är ett tecken på att dialog, samråd och lyhördhet bör prioriteras i fortsatt utvecklingsarbeten Lättläst sammanfattning och rekommendationer Huvudmän är en sårbar grupp. Det är personer som har en fysisk eller psykisk funktionsnedsättning, som gör att det kan vara svårt att klara sin vardag. Här är frågorna vi ville ha svar på: - Vilka förväntningar har huvudmännen på sin ställföreträdare? - Vad tycker huvudmännen om den hjälp de får från sin ställföreträdare? - Hur mycket tro på att man klarar sin vardag själv har huvudmännen? - Hur mycket litar huvudmännen på att de själva klarar av att påverka vilken hjälp de kan få från sin ställföreträdare. - Vad kan ställföreträdare bli bättre på i framtiden? - Vad kan göras för att få huvudmännen att tro på sina egna förmågor att klara sin vardag? När vi ställde frågorna i förstudien ville vi göra det på ett så tydligt sätt som möjligt. Vi använde oss av bilder för att göra frågor och svar tydliga. Den som intervjuade läste upp frågorna högt om det var svårt att förstå frågan, eller om man inte kunde läsa själv. Om det ändå var svårt att förstå kunde intervjuaren förklara mera. Vi ställde öppna frågor, alltså frågor där man själv berättade vad man ville säga som svar. Vi ställde också frågor som handlade om ett visst område, där fick man välja ett av de svar som fanns att välja på. Sammanlagt var det 19 huvudmän som var med i förstudien. 17 var från Eskilstuna och 2 från Strängnäs. 13 huvudmän var män. 5 huvudmän var kvinnor, och en svarade inte på frågan om kön. Svaren vi fick ifrån huvudmännen i förstudien var intressanta. Vi såg tydligt att många av huvudmännen förväntade sig samma form av hjälp från sina ställföreträdare. Däremot såg vi att huvudmännen tyckte olika om hur de kunde påverka sina ställföreträdare. De som inte fick den hjälp de förväntade sig tyckte till exempel att ställföreträdare kan visa, prata och förklara mer vad de gör. Det skulle kunna göra att huvudmännen känner sig mer delaktig. Förstudien visade att ställföreträdare gör en del bra arbeten för sina huvudmän. Men det behövs en del förbättringar också. Vi föreslår därför: - att det som huvudmännen tycker fungerar bra är det viktigt att visa för andra. - att man tydligt berättar för ställföreträdare vad de får och ska göra mot och med sina huvudmän. - att man tydligt berättar för huvudmännen vad ställföreträdaren kan och ska hjälpa till med. - att ställföreträdare får utbildning och kunskap om olika funktionsnedsättningar som huvudmännen kan ha. - att ställföreträdare ska lyssna noga på sina huvudmän, därför att huvudmännen är olika och har olika möjligheter att säga vad de tycker och tänker. Context: Cardiometabolic complications in obesity may be linked to white adipose tissue (WAT) dysfunction. Transcriptomic studies of Sc WAT have reported that CCL18, encoding the CC chemokine ligand 18 (CCL18), is increased in obesity/insulin resistance but its functional role is unknown. Objective: Our objectives were to determine if CCL18 is secreted from Sc WAT and if secreted and/or serum levels associate with metabolic phenotypes. We also planned to define the primary cellular source and if CCL18 exerts effects on adipocytes. Design: This is a cohort study. Setting: The study took place in an outpatient academic clinic. Participants: A total of 130 obese women scheduled for bariatric surgery and 35 nonobese controls were included. Methods: Insulin sensitivity was assessed by hyperinsulinemic euglycemic clamp or homeostasis model assessment. CCL18 was analyzed in serum/WAT incubates by ELISA. Effects of recombinant CCL18 was determined in cultures of primary human adipocytes and the monocyte cell line THP-1 differentiated into M0/M1/M2 macrophages. Main Outcome Measure: Association with metabolic risk factors was measured. Results: CCL18 was secreted from WAT and the levels correlated positively with insulin resistance, Adult Treatment Panel III risk score and plasma triglycerides, independent of body mass index and better than other established adipocytokines. In 80 obese women, S-CCL18 levels were significantly higher in insulin resistant compared with insulin sensitive subjects. In WAT CCL18 mRNA was expressed in macrophages and correlated positively with immune-related genes, particularly those enriched in M2 macrophages. While CCL18 increased cyto-/chemokine expression in M0/M2-THP-1 cells, human adipocytes showed no responses in vitro. Conclusions: Circulating and WAT-secreted CCL18 correlates with insulin resistance and metabolic risk score. Because CCL18 is macrophage-specific and associates with adipose immune gene expression, it may constitute a marker of WAT inflammation.

Uncaging promoter and enhancer dynamics In order to understand cellular differentiation, it is important to understand the timing of the regulation of gene expression. Arner et al. used cap analysis of gene expression (CAGE) to analyze gene enhancer and promoter activities in a number of human and mouse cell types. Regulation of adipose tissue stem cells (ASCs) and adipogenesis impact the development of excess body fat-related metabolic complications. Animal studies have suggested the presence of distinct subtypes of ASCs with different differentiation properties. In addition, ASCs are becoming the biggest source of mesenchymal stem cells used in therapies, which requires deep characterization. Using unbiased single cell transcriptomics we aimed to characterize ASC populations in human subcutaneous white adipose tissue (scWAT). The transcriptomes of 574 single cells from the WAT total stroma vascular fraction (SVF) of four healthy women were analyzed by clustering and t-distributed stochastic neighbor embedding visualization. The identified cell populations were then mapped to cell types present in WAT using data from gene expression microarray profiling of flow cytometry-sorted SVF. Cells clustered into four distinct populations: three adipose tissue-resident macrophage subtypes and one large, homogeneous population of ASCs. While pseudotemporal ordering analysis indicated that the ASCs were in slightly different differentiation stages, the differences in gene expression were small and could not distinguish distinct ASC subtypes. Altogether, in healthy individuals, ASCs seem to constitute a single homogeneous cell population that cannot be subdivided by single cell transcriptomics, suggesting a common origin for human adipocytes in scWAT. In the FANTOM5 project, transcription initiation events across the human and mouse genomes were mapped at a single base-pair resolution and their frequencies were monitored by CAGE (Cap Analysis of Gene Expression) coupled with single-molecule sequencing. Approximately three thousands of samples, consisting of a variety of primary cells, tissues, cell lines, and time series samples during cell activation and development, were subjected to a uniform pipeline of CAGE data production. The analysis pipeline started by measuring RNA extracts to assess their quality, and continued to CAGE library production by using a robotic or a manual workflow, single molecule sequencing, and computational processing to generate frequencies of transcription initiation. Resulting data represents the consequence of transcriptional regulation in each analyzed state of mammalian cells. Non-overlapping peaks over the CAGE profiles, approximately 200,000 and 150,000 peaks for the human and mouse genomes, were identified and annotated to provide precise location of known promoters as well as novel ones, and to quantify their activities.

Although the mechanisms linking obesity to insulin resistance (IR) and type 2 diabetes (T2D) are not entirely understood, it is likely that alterations of adipose tissue function are involved. The aim of this study was to identify new genes controlling insulin sensitivity in adipocytes from obese women with either insulin resistant (OIR) or sensitive (OIS) adipocytes. Insulin sensitivity was first determined by measuring lipogenesis in isolated adipocytes from abdominal subcutaneous white adipose tissue (WAT) in a large observational study. Lipogenesis was measured under conditions where glucose transport was the rate limiting step and reflects in vivo insulin sensitivity. We then performed microarray-based transcriptome profiling on subcutaneous WAT specimen from a subgroup of 9 lean, 21 OIS and 18 obese OIR women. We could identify 432 genes that were differentially expressed between the OIR and OIS group (FDR ≤5%). These genes are enriched in pathways related to glucose and amino acid metabolism, cellular respiration, and insulin signaling, and include genes such as SLC2A4, AKT2, as well as genes coding for enzymes in the mitochondria respiratory chain. Two IR-associated genes, KLF15 encoding a transcription factor and SLC25A10 encoding a dicarboxylate carrier, were selected for functional evaluation in adipocytes differentiated in vitro. Knockdown of KLF15 and SLC25A10 using siRNA inhibited insulin-stimulated lipogenesis in adipocytes. Transcriptome profiling of siRNA-treated cells suggested that KLF15 might control insulin sensitivity by influencing expression of PPARG, PXMP2, AQP7, LPL and genes in the mitochondrial respiratory chain. Knockdown of SLC25A10 had only modest impact on the transcriptome, suggesting that it might directly influence insulin sensitivity in adipocytes independently of transcription due to its important role in fatty acid synthesis. In summary, this study identifies novel genes associated with insulin sensitivity in adipocytes in women independently of obesity. KFL15 and SLC25A10 are inhibitors of insulin-stimulated lipogenesis under conditions when glucose transport is the rate limiting step.

Obesity affects gene expression and metabolism of white adipose tissue (WAT), which results in insulin resistance (IR) and type 2 diabetes. However, WAT is a heterogeneous organ containing many cell types that might respond differently to obesity-induced changes. We performed flow cytometry sorting and RNA expression profiling by microarray of major WAT cell types (adipocytes, CD45−/CD31−/CD34+ progenitors, CD45+/CD14+ monocytes/ macrophages, CD45+/CD14− leukocytes), which allowed us to identify genes enriched in specific cell fractions. Additionally, we included adipocytes and adipocyte progenitor cells obtained from lean and obese individuals. Taken together, we provide a detailed gene expression atlas of major human adipose tissue resident cell types for clinical/basic research and using this dataset provide lists of cell-type specific genes that are of interest for metabolic research.

Current worldwide figures suggest that obesity is pandemic. Understanding the underlying molecular mechanisms would help develop viable anti-obesity therapies. Here, we assess the influence of obesity-induced inflammation on white adipocyte cyclic guanosine monophosphate (cGMP) signaling, which is beneficial for adipocyte differentiation and thermogenesis. We find that murine gonadal and not inguinal fat is prone to obesity-induced inflammation. Correspondingly, the cGMP cascade is dysregulated in gonadal but not in inguinal fat of obese mice. Analysis of two independent human cohorts reveals a defective cGMP pathway only in visceral fat of obese subjects. Congruently, cGMP signaling is dysregulated in tumor necrosis factor α (TNF-α)-treated primary white adipocytes. TNF-α-mediated suppression of sGCβ1 is mediated via NF-κB, whereas PKG is repressed by JNK signaling. Additionally, TNF-α-activated JNK signaling suppresses PPARγ and aP2. Taken together, the intensity of obesity-induced inflammation dictates the amplitude of cGMP signaling dysregulation in white adipocytes through distinct pathways.

White adipose tissue (WAT) can develop into several phenotypes with different pathophysiological impact on type 2 diabetes. To better understand the adipogenic process, the transcriptional events that occur during in vitro differentiation of human adipocytes were investigated and the findings linked to WAT phenotypes. Single-molecule transcriptional profiling provided a detailed map of the expressional changes of genes, enhancers, and long noncoding RNAs, where different types of transcripts share common dynamics during differentiation. Common signatures include early downregulated, transient, and late induced transcripts, all of which are linked to distinct developmental processes during adipogenesis. Enhancers expressed during adipogenesis overlap significantly with genetic variants associated with WAT distribution. Transiently expressed and late induced genes are associated with hypertrophic WAT (few but large fat cells), a phenotype closely linked to insulin resistance and type 2 diabetes. Transcription factors that are expressed early or transiently affect differentiation and adipocyte function and are controlled by several well-known upstream regulators such as glucocorticosteroids, insulin, cAMP, and thyroid hormones. Taken together, our results suggest a complex but highly coordinated regulation of adipogenesis.

The orphan receptor LRH-1 and the oxysterol receptors LXRα and LXRβ are established transcriptional regulators of lipid metabolism that appear to control inflammatory processes. Here, we investigate the anti-inflammatory actions of these nuclear receptors in the hepatic acute phase response (APR). We report that selective synthetic agonists induce SUMOylation-dependent recruitment of either LRH-1 or LXR to hepatic APR promoters and prevent the clearance of the N-CoR corepressor complex upon cytokine stimulation. Investigations of the APR in vivo, using LXR knockout mice, indicate that the anti-inflammatory actions of LXR agonists are triggered selectively by the LXRβ subtype. We further find that hepatic APR responses in small ubiquitin-like modifier-1 (SUMO-1) knockout mice are increased, which is due in part to diminished LRH-1 action at APR promoters. Finally, we provide evidence that the metabolically important coregulator GPS2 functions as a hitherto unrecognized transrepression mediator of interactions between SUMOylated nuclear receptors and the N-CoR corepressor complex. Our study extends the knowledge of anti-inflammatory mechanisms and pathways directed by metabolic nuclear receptor–corepressor networks to the control of the hepatic APR, and implies alternative pharmacological strategies for the treatment of human metabolic diseases associated with inflammation.

Transcriptional coregulators, rather than ligand signals, are suspected to confer context and pathway specificity to nuclear receptor signaling, but the identity of such specifying coregulators and the underlying molecular mechanisms remain largely enigmatic. Here we address this issue in metabolic oxysterol receptor LXR pathways and describe the selective requirement of GPS2 for ABCG1 cholesterol transporter gene transcription and cholesterol efflux from macrophages. We implicate GPS2 in facilitating LXR recruitment to an ABCG1-specific promoter/enhancer unit upon ligand activation and identify functional links to histone H3K9 demethylation. We further describe fundamental differences between ABCG1 and ABCA1 with regard to GPS2 in relation to other coregulators, which are likely to apply to additional LXR-regulated genes. Our work identifies a coregulator-dependent epigenetic mechanism governing the access of a nuclear receptor to communicating regulatory regions in the genome. The pathway and coregulator selectivity of this mechanism implies pharmacological possibilities for the development of selective LXR agonists.

SummaryWhite adipose tissue (WAT) morphology characterized by hypertrophy (i.e., fewer but larger adipocytes) associates with increased adipose inflammation, lipolysis, insulin resistance, and risk of diabetes. However, the causal relationships and the mechanisms controlling WAT morphology are unclear. Herein, we identified EBF1 as an adipocyte-expressed transcription factor with decreased expression/activity in WAT hypertrophy. In human adipocytes, the regulatory targets of EBF1 were enriched for genes controlling lipolysis and adipocyte morphology/differentiation, and in both humans and murine models, reduced EBF1 levels associated with increased lipolysis and adipose hypertrophy. Although EBF1 did not affect adipose inflammation, TNFα reduced EBF1 gene expression. High-fat diet intervention in Ebf1+/− mice resulted in more pronounced WAT hypertrophy and attenuated insulin sensitivity compared with wild-type littermate controls. We conclude that EBF1 is an important regulator of adipose morphology and fat cell lipolysis and may constitute a link between WAT inflammation, altered lipid metabolism, adipose hypertrophy, and insulin resistance.

Wnt signaling regulates adipogenesis and adipocyte function. Secreted frizzled-related proteins (SFRPs) are a family of secreted proteins (SFRP1-5) that bind and inhibit Wnts. Several members, including SFRP5, have recently been implicated in adipocyte dysfunction in obesity.Our objective was to characterize the expression, secretion, and function of the SFRP family in human white adipose tissue (WAT) and fat cells.SFRP1-5 mRNA expression was measured in human sc and visceral WAT from lean and obese individuals and correlated to insulin sensitivity. SFRP secretion from WAT explants was assessed by ELISA. Gene expression of SFRPs in cultured adipocytes during and after differentiation was determined. Functional analyses were done by gene silencing or incubations with recombinant SFRPs.SFRP1-4, but not SFRP5, mRNA levels were altered in obesity. However, although SFRP1 was down-regulated and correlated positively with insulin sensitivity, SFRP2-4 were up-regulated, particularly in visceral WAT, and associated with insulin resistance. Only SFRP1, SFRP2, and SFRP4 were secreted from WAT, thereby constituting adipokines. Individual knockdowns of SFRP1, SFRP2, or SFRP4 during adipogenesis did not affect terminal differentiation. Incubations with SFRP1 reduced the secretion of the proinflammatory cytokines IL-6 and monocyte chemotactic protein-1 (MCP1) and increased the release of adiponectin.SFRP1, SFRP2, and SFRP4 are adipokines, the expression of which correlates with insulin sensitivity. For SFRP1, this may be related to effects on the secretion of IL-6, MCP1, and adiponectin. In contrast to recent murine findings implicating SFRP5 in metabolic dysfunction, this SFRP is neither regulated by obesity nor actively secreted from human WAT.

The orphan nuclear receptor Steroidogenic Factor-1 (SF-1, NR5A1) is a critical regulator of development and homeostasis of the adrenal cortex and gonads. We recently showed that a complex containing E3 ubiquitin ligase RNF31 and the known SF-1 corepressor DAX-1 (NR0B1) interacts with SF-1 on target promoters and represses transcription of steroidogenic acute regulatory protein (StAR) and aromatase (CYP19) genes. To further evaluate the role of SF-1 in the adrenal cortex and the involvement of RNF31 in SF-1-dependent pathways, we performed genome-wide gene-expression analysis of adrenocortical NCI-H295R cells where SF-1 or RNF31 had been knocked down using RNA interference. We find RNF31 to be deeply connected to cholesterol metabolism and steroid hormone synthesis, strengthening its role as an SF-1 coregulator. We also find intriguing evidence of negative crosstalk between SF-1 and both transforming growth factor (TGF) β and Wnt/β-catenin signaling. This crosstalk could be of importance for adrenogonadal development, maintenance of adrenocortical progenitor cells and the development of adrenocortical carcinoma. Finally, the SF-1 gene profile can be used to distinguish malignant from benign adrenocortical tumors, a finding that implicates SF-1 in the development of malignant adrenocortical carcinoma.

The aim of this study was to understand how the professional in social work handles the media scrutiny. When the media writes about social services there is seldom anything positive written about it, many social services organizations are instead heavily criticized and scrutinized by the media. When social services organizations are scrutinized by the media there is a crisis at work, and therefore should be handled as a crisis. Therefore, we describe how a crisis management might look like in an organization. The study is qualitative therefore we have interviewed four municipalities in Sweden that have been exposed by the media at a national level. We interviewed eight social workers and managers to get different opinions about the handling after media critic and scrutiny. We wanted to study how a social service handles the media when it been scrutinized, when the previous research only shows how social workers are affected by it. When the professionals in social services organization are scrutinized by the media there are frequently negative criticisms and that can lead to social services organizations legitimacy decreases. Therefor we will analyze our result from professional theory to see how the professional handles media scrutiny. We see similarities between a crisis and a media scrutiny and therefore we also use the concept of crisis. The results shows that it is important that colleagues support each other and that the professionals are open against media. The differences between the positions are that managers are the ones who have the ultimate responsibility for the medial scrutiny. The responsibility includes media contact; to ensure that there is enough support, information regarding the situation given out to everyone in the workplace and taking into account the security. To meet media in a better way is by have a media education there social workers and journalist can meet and exchanges knowledge and experience of each other works.

DAX-1 and SHP are two closely related atypical orphan members of the nuclear receptor (NR) family that make up the NR0B subfamily. They combine properties of typical NRs and of NR-associated coregulators: both carry the characteristic NR ligand-binding domain but instead of a NR DNA-binding domain they have unique N-terminal regions that contain LxxLL-related NR-binding motifs often found in coregulators. Recent structural data indicate that DAX-1 lacks a ligand-binding pocket and thus should rely on ligand-independent mechanisms of regulation. This might be true, but remains to be proven, for SHP as well. DAX-1 and SHP have in common that they act as transcriptional corepressors of cholesterol metabolism pathways that are related on a molecular level. However, the expression patterns of the two NRs are largely different, with some notable exceptions, and so are the physiological processes they regulate. DAX-1 is mainly involved in steroidogenesis and reproductive development, while SHP plays major roles in maintaining cholesterol and glucose homeostasis. This review highlights the key similarities and differences between DAX-1 and SHP with regard to structure, function and biology and considers what can be learnt from recent research advances in the field. Inledning: Efter flera redovisningsskandaler bestämde Svensk näringsliv, med flera, att instifta Kollegiet för Svensk Bolagsstyrning som övervakar Koden för Svensk Bolagsstyrning. Bland det viktigaste är bestämmelser angående intern kontroll. Intern kontroll är viktigt för alla bolag och dålig sådan anser många är en orsak till många företagskonkurser. 2005 instiftades denna kod vars krav tvingade noterade bolag att lämna en rapport angående deras IK och att utvärdera sin IK för första gången. Syfte: Syftet är att undersöka om bolagskodens införande har påverkat bolagens sätt att arbeta med intern kontroll av den finansiella rapporteringen, baserat på intern kontroll rapporterna samt att jämföra bolagen som lyder under enbart Koden med de som även lyder under SOX för att se om det finns en skillnad i hur mycket förändring som har skett under åren 2005-2008. Metod: Undersökningen är en kvalitativ studie med en induktiv ansats. Studien innefattar flera år så att vi kan se en förändring över tiden. Urvalet är 8 bolag varav 4 är noterade på både den Amerikanska och den svenska börsen och de andra 4 är enbart noterade på den Svenska börsen. Populationen är de 70 bolag som var från 2005 så kallade Kodbolag. Slutsatser: Kodens införande påverkar sättet företagen arbetar med intern kontroll. SOX-bolagen verkar ha blivit mer påverkade än kod-bolagen och detta beror troligtvis på den strängare lagstiftning som finns i USA. Även de bolag som avnoterades från SEC (amerikanska finansinspektionen) fortsatte med sitt arbete och utvecklade, i vissa fall, den bortom det som krävdes av SOX lagstiftningen. Kod-bolagens arbete med sin IK verkar blivit mer omfattande än vad det annars skulle ha varit, om det inte fanns några krav alls, men det verkar som den enklare standarden på rapporten inte har orsakat lika mycket förändring som skulle vara möjligt om högre krav ställdes på de Svenska bolagen.

Detta är ett 20 poäng stort examensarbete i Medieteknik utfört hösten 2005 samt våren 2006. Examensarbetets syfte är att skapa ett användargränssnitt till menysystemet för en produkt som skall komma med motionscyklar för hemmabruk. Examensarbetet har inneburit att genom hela designprocessen ha en hög grad av kontakt med potentiella slutanvändare till produkten. Detta har skett genom längre intervjuer, enkäter och tester. Gränssnittet skulle innehålla vissa funktioner som specificerades av uppdragsgivaren. Detta var till exempel vilka inställningar som bör kunna göras och vilka olika alternativ på cykling man har. Efter inledande dialog med potentiella slutanvändare och kvantitativ informationsinsamling togs ett antal designförslag fram som utvärderades av slutanvändare. Förslag på navigering och menyträd testades i experimentform, detta innebar att användarna ombads utföra ett antal uppgifter och fick förklara hur de tänkte lösa dessa. Slutligen sammanfogades resultaten till en prototyp till ett användargränssnitt. Det ingick även i examensarbetet att ta fram en ikon som skall visas då den personliga tränaren säger något till användaren under ett träningspass.
