Abstract

Grapevine trunk pathogens (GTPs) cause serious damage to grapevines and have significant economic impacts. There is no effective protection against grapevine trunk diseases. Newly designed AgSe nanoparticles (NPs) and CuSe NPs, single‐element Ag NPs, Cu NPs, Se NPs and selected chemicals or chemical agents such as sodium arsenite, 8‐hydroxyquinoline, silver nitrate, colloidal silver, Altron Silver fertilizer and silver thiosulfate complex (NH4)3/Ag(S2O3)2 were tested in vitro against two serious GTPs Diaporthe eres, Eutypa lata and Diplodia seriataisolated from walnut. The most significant inhibition of fungal growth was observed for silver nitrate and AgSe NPs, which showed the highest level of half the maximum effective EC50 concentration with the lowest concentrations. In the case of silver nitrate at a concentration of 1000 mg L−1, 79% inhibition of mycelial growth was observed for the pathogen E. lata, 48% for D. seriata and 54% for D. eres. AgSe NPs, in which the concentration of silver is 2588 mg L−1 and that of selenium is 902 mg L−1, showed 68% inhibition of mycelial growth in the pathogen E. lata, 54% in D. seriata and 58% in D. eres. Abstract

Epigenetics is the study of heritable alterations in phenotypes that are not caused by changes in DNA sequence. In the present study, we characterized the genetic and phenotypic alterations of the bacterial plant pathogen Xanthomonas&nbsp, campestris pv. campestris (Xcc) under different treatments with several epigenetic modulating chemicals. The use of DNA demethylating chemicals unambiguously caused a durable decrease in Xcc bacterial virulence, even after its reisolation from infected plants. The first-time use of chemicals to modify the activity of sirtuins also showed some noticeable results in terms of increasing bacterial virulence, but this effect was not typically stable. Changes in treated strains were also confirmed by using methylation sensitive amplification (MSAP), but with respect to registered SNPs induction, it was necessary to consider their contribution to the observed polymorphism. The molecular basis of the altered virulence was deciphered by using dualRNA-seq analysis of treated Xcc strains infecting Brassica&nbsp, rapa plants. The results of the present study should promote more intensive research in the generally understudied field of bacterial epigenetics, where artificially induced modification by epigenetic modulating chemicals can significantly increase the diversity of bacterial properties and potentially contribute to the further development of the fields, such as bacterial ecology and adaptation.

Abstract

Changes in the bacterial spectrum of cabbage heads after storage under commonly used storage conditions were examined in this study. Cabbage seeds (Brassica oleracea var. capitata L.) were artificially inoculated with X. campestris pv. campestris (Xcc), a serious pathogen of cruciferous plants causing black rot. Isolation of bacterial cultures from Xcc-inoculated and non-inoculated cabbage heads were carried out in two time points-at the day of harvest and after four months of storage. According to our previous research and literature reports, the most frequent genera of bacteria were chosen for PCR testing, i.e., Bacillus cereus group, Bacillus subtilis group, Pseudomonas sp., and X. campestris pv. campestris. A few of the obtained bacterial cultures were negative for the four above-mentioned species. In those, other bacteria were identified by 16S rRNA sequencing. In both Xcc-inoculated and non-inoculated cabbage heads, changes of the bacterial spectrum over time were observed. The severity of Xcc infection of heads increased after four months of storage. Bacillus species represented the most frequently occurring bacterial genus. The presence of the Bacillus subtilis group increased significantly after storage in non-inoculated cabbage heads. The minor part of the other genera identified by sequencing in the first sampling were not detected in the stored cabbage heads. Abstract

Grapevine Pinot gris virus (GPGV) is a putative causal agent of grapevine leaf mottling and deformation disease that has been reported worldwide throughout the grapevine-growing regions. Fifty-four grapevines collected from five Algerian grapevine-growing regions were tested for the presence of GPGV in phloem tissues. Eight of the tested grapevines were infected by GPGV. Viromes of two selected Vitis vinifera cv. Sabel grapevines infected by GPGV and showing virus-like symptoms were analyzed by small RNA sequencing. Phylogenetic analyses of the partial coding sequence (cds) of the RNA-dependent RNA polymerase (RdRp) domain showed that all Algerian GPGV isolates were grouped with some already-described asymptomatic isolates. This study provides the first survey of the occurrence of GPGV in Algeria. Abstract

The character of parent material, organic matter and climatic conditions are the crucial factors of pedogenesis. They directly influence the intensity of weathering, transformation processes, elements release and geochemical cycles. The mineral transformation in a profile of the recent soil developed on loess (Haplic Luvisols, Siltic, Epidystric, Ochric) was studied. The land area Litovelské Pomoraví was chosen because of the stability of soil‐forming factors such as topography, relief, vegetation and climate. X‐ray diffraction (XRD) quantitative analysis of the fine earth (<2 mm) and XRD analysis of clay (<2 μm) and the fine clay fraction (<0.2 μm) were applied. The obtained results showed the most significant differences between the argic horizons (Bt1 and Bt2) and other soil horizons. The lower content of quartz and feldspars and higher content of dioctahedral micas and phyllosilicates in both Bt horizons was observed. It was assumed the clay illuviation/lessivage process was preceded by long‐term leaching of carbonate in humid continental conditions during almost the whole Holocene. The distribution of the clay fraction correlates with the distribution of phyllosilicates through the profile. Major clay minerals in the clay fraction (<2 μm) were illite and kaolinite, smectite, chlorite, vermiculite and several types of mixed‐layered phases. The analysis of the finer clay fraction (<0.2 μm) identified the new phase hydroxy‐interlayered minerals as the product of illite (vermiculite) weathering in humid continental conditions. Abstract

The susceptibility of twenty-four cabbage breeding lines to Xanthomonas campestris pv. campestris was evaluated. The selection of appropriate inoculation method was done on 4 cabbage cultivars ('Cerox', 'Sintex', 'Sonja' and 'Avak'). One month old plants were infected by 5 inoculation methods (spraying, injection by syringe, multiple pricking, carborundum abrasion and scissor clipping method). Four different bacterial isolates of Xcc (WHRI 3811, 3971A, 1279A; SU) and their mixture were evaluated for the aggressiveness on 'Cerox' and 'Sonja' cultivars. On the basis of obtained results, breeding lines of head cabbage were inoculated by mixture of all tested isolates using multiple pricking method. The disease severity of inoculated seedlings proved high susceptibility of young plants to the Xcc infection. The disease incidence determined 75 and 105 days after sowing showed changes for 16 of tested lines and indicated that resistance testing should be observed until mature stage. The study revealed five breeding lines (DP25, T1, IT10, Kalibos and Avak1) with disease incidence lower than 20 % as perspective sources of resistance for further breeding., info:eu-repo/semantics/openAccess
