11. Assignment of the antibacterial potential of Ag2O/ZnO nanocomposite against MDR bacteria Proteus mirabilisand Salmonella typhiisolated from bone marrow transplant patients

Abstract

The rate of infectious diseases started to be one of the major mortality agents in the healthcare sector. Exposed to increased bacterial infection by antibiotic-resistant bacteria became one of the complications that occurred for bone marrow transplant patients. Nanotechnology may provide clinicians and patients with the key to overcoming multidrug-resistant bacteria. Therefore, this study was conducted to clarify the prevalence of MDR bacteria in bone marrow transplant recipients and the use of Ag2O/ZnO nanocomposites to treat participants of diarrhea brought on by MDR bacteria following bone marrow transplantation (BMT). Present results show that pathogenic bacteria were present in 100 of 195 stool samples from individuals who had diarrhea. Phenotypic, biochemical, and molecular analysis clarify that Proteus mirabilisand Salmonella typhiwere detected in 21 and 25 samples, respectively. Successful synthesis of Ag2O/ZnO nanocomposites with a particle enables to inhibition of both pathogens. The maximum inhibitory impact was seen on Salmonella typhi. At low doses (10−5g/l), it prevented the growth by 53.4%, while at higher concentrations (10−1g/l), Salmonella typhiwas inhibited by 95.5%. Regarding Proteus mirabilis, at (10−5g/l) Ag2O/ZnO, it was inhabited by 78.7%, but at higher concentrations (10−1g/l), it was inhibited the growth by 94.6%. Ag2O/ZnO nanocomposite was therefore found to be the most effective therapy for MDR-isolated bacteria and offered promise for the treatment of MDR bacterial infections that cause diarrhea.

Abstract

Background: The most hazardous wastewater sources in the northern part of the Middle Nile Delta, Egypt; receiving a massive amount of agricultural, industrial, and sewage drainage are Kitchener drain which is one of the tallest drainage systems, and Burullus Lake which represents the 2nd largest Egyptian coastal lake. Results: The current work is to determine the abundance and frequency of cyanophytes, chlorophytes, and bacillariophytes and the correlation between them and environmental abiotic components. Among sixty nine microalgal species, 19 species are belong Cyanophyta, 26 belong Chlorophyta and 24 belong Bacillariophyta. Genus Scenedesmus (Chlorophyta) was the most abundant in the study area (13 species), followed by Genus Oscillatoria (9 species) and Genus Navicula (7 species). Nostoc muscorum and Chlorella vulgaris were the most common and recorded in all sites (100% of the locations) under study. The application of the two-way indicator species analysis (TWINSPAN) and detrended correspondence analysis revealed agglomerating of 4 groups (communities) at 4th level of classification and reasonable segregation between these groups. Zinc, cadmium and lead were showed the highest levels (0.26 ± 0.03, 0.26 ± 0.06, and 0.17 ± 0.01 ppm, respectively). Conclusions: The correlation analysis between water and community variables indicated a high negative correlation of total algae richness with nickel (r = -0.936, p < 0.01). Cyanophyta and Bacillariophyta were correlated negatively (r = -0.842, p < 0.01). However, Chlorophyta showed a negative richness with each of Ni and Pb (r = -0.965, -0.873, respectively) on one hand and a high positive correlation was revealed (r = 0.964) with all environmental variables on the other hand. Abstract

The possibility of isolating from the cyanobacterium Thermosynechococcus elongates the intact monomeric photosystem 1 (PS1) complex that has preserved all subunits and the photochemical activity of the isolated trimeric complex is discussed. The dynamic equilibrium between monomers and trimers in the thylakoid membrane have shown a transient monomer population in the course of biogenesis and is also the basis for short-term adaptation of the cell to changing environmental conditions.

Abstract

Purification of highly active trimeric PS1 was optimized and speeded up from 32 to 15 hours, and new and mild methods were developed for the qualitative and quantitative production of monomeric PS1. This enabled for a detailed analysis in comparison with the trimeric form in terms of pigment/cofactor contents, subunit composition, photosynthetic electron transport and photocurrent of immobilized PS1. Both complexes showed almost identical spectroscopic characteristics of LD-spectra, redox potential, Chl \(\it a\)P700 ratio, carotenoid contents, subunit composition and activity. Modeling of kinetics measurements (time resolved fluorescence in the lower ps range) indicated the same origin of the long wavelength chlorophylls (LWCs) in both complexes. Optimization of photocurrent measurements of PS1, which had been immobilized on a gold surfaces by a \(\textit {His-tag}\), showed the higher efficiency of the artificial electron donor PMS in comparison with the native Cyt\( _{C6}\).
