Abstract Malaria has not yet been eradicated in Iran, and Plasmodium vivax (P. vivax) is the main cause of malaria in the country. This study aimed to investigate and analyze the amount of genetic diversity of Plasmodium vivax merozoite surface protein-5 (PvMSP-5) exon 1 gene in the southeast of Iran. Thirty-five patients with clinical symptoms of P. vivax malaria participated. The exon 1 of PvMSP-5 was amplified by PCR, and the PCR product of all isolates was sequenced, and genetic polymorphisms were determined using various genetic software. The analysis showed that studied isolates are different from one another in the DnaSP software version. Out of the 612 sites, 477 were monomorphic and 135 were segregated. The total number of mutations was 143. The singleton variable and the parsimony informative sites were 23 and 112, respectively. There were 17 specific haplotypes with haplotype diversity equal to 0.943. Nucleotide diversity was equal to 0.06766 in the isolates. The ratio of nonsynonymous (0.06446) to synonymous (0.07909) mutations was 0.815020. Tajima’s D, which expressed coding, and non-coding regions, was 0.72403, which was not deemed significant (P > 0.10). The analysis of intrapopulation diversity revealed nucleotide and haplotype diversity in the msp-5 gene of Iranian P. vivax isolates. In addition to balancing or purifying selection, intragenic recombination also contributed to the variation observed in exon 1 of PvMSP-5, according to the findings.

Introduction: Fungal aerosols from landfill sites can play a fundamental role in environmental pollution and health. The present study aimed to assess the dispersion of fungal aerosols from municipal solid waste disposal site. Materials and Methods: In this cross-sectional study, the concentration of fungal aerosols was determined in four geographical directions at distances of 250, 500, 750, and 1000 m around landfill site. Relative humidity and temperature were also measured. Moreover the concentration and type of the fungal taxa isolated from landfill site under different environmental and metrological conditions were evaluated. Results: The results showed that the maximum and minimum concentrations of fungal aerosol in the landfill site were 256.18 ± 59.7 CFU/m3 and 76.56 ± 23.2 CFU/m3, respectively. The most frequent fungi detected from municipal landfill site included Penicillium (43.67%), Cladosporium (33.54%), Yeast (7.60%), Aspergillus (5.91%), Curvularia (3.62%), Chrysosporium (1.57%), Alternaria (1.54%), Scopulariopsis (0.84%), and Ulocladium (0.60%) taxa. The maximum identified fungal aerosol concentration in the area around the solid waste landfill was 350 CFU/m3. Furthermore, the concentration of fungal aerosols in the environment was significantly related to relative humidity, wind direction, and temperature in spring and winter (P < 0.05). Conclusion: Municipal solid waste disposal site can be a potential source for fungal aerosol dispersion. Moreover, fungal aerosols concentration is correlated with wind direction and speed, relative humidity, and temperature.

Azorubine was selected as a contaminant in this study. The pH value of a factor it is very effective in destroying the color of Azorubine. Structural, morphological and optical properties of BFO using X-ray diffraction (IR-FT), scanning electron microscopy (SEM-FE) and Visible-UV spectroscopy were analyzed. Effects of different parameters on degradation photocatalytic dye of Azorubine dye at a concentration of 10 ppm including H2O2 content, type of light emitted, pH value and dose. The nanoparticles used were investigated and optimized. Degree of Azorubine dye degradation using BFO nanoparticles during. The time of 120 minutes reached 93.93% to show that this research is an effective photocatalytic method for destroys the color of azorubine.

Abstract Background Recently, the role of endogenous microbiota and the genotype-microbiota correlation in inflammatory bowel disease (IBD) pathogenesis have been highlighted. However, fungi, as the second most prevalent residents of the intestine, and their primary receptor, Dectin-1, are underrated. Thus, we conducted the first human study investigating the association of Leucine-rich repeat kinase 2 (LRRK2) polymorphism (rs11564258) with type and the extent of intestinal fungi in IBD patients. Material and methods A case–control study was performed on 79 ulcerative colitis (UC)-patients (case group) and 58 healthy subjects (HS group). DNA was extracted from blood samples of both groups and amplified with the primers designed for the specific locus containing the LRRK2 polymorphism (rs11564258) and then sequenced. Dectin-1 and LRRK2 mRNA expression levels were also determined. Furthermore, the type and prevalence of fecal yeast species were surveyed in case and control groups. Results A positive correlation was observed between rs11564258 polymorphism and UC susceptibility (p = 0.008 vs. HS). Patients with active UC had the highest rate of isolated fungal colonies (50.41%), followed by patients with non-active UC (24.6%) and HS (25%). These results showed a relationship between UC severity with the increased fungal load. Candida albicans had the highest prevalence in both UC (78.7%) and HS groups (55.8%). Whereas Saccharomyces cerevisiae was the second most common species detected in HS (15.23%), it was significantly reduced in the UC patient group (1.68%) (P = 0.0001). On the other hand, single nucleotide polymorphism (SNP, rs11564258) was not correlated with the increased fungal flora in the UC patients. Abstract Inflammatory bowel disease (IBD) is a chronic inflammatory disease affecting various parts of the gastrointestinal tract. A majority of the current evidence points out the involvement of intestinal dysbiosis in the IBD pathogenesis. Recently, the association of intestinal fungal composition With IBD susceptibility and severity has been reported. These studies suggested gene polymorphisms in the front line of host defense against intestinal microorganisms are considered to play a role in IBD pathogenesis. The studies have also detected increased susceptibility to fungal infections in patients carrying IBD-related mutations. Therefore, a literature search was conducted in related databases to review articles addressing the mycobiota-genotype association in IBD.

Background and objectives: TLR2, TLR4, and Dectin-1 (Clec7) are pattern recognition receptors (PRRs) expressed by intestinal epithelia cells and MYD88 is a signaling molecule of TLR2 and TLR4. They warn immune system about the presence of invading pathogens promoting initiation of inflammatory response. Because of colonic cancer risk, therapy of intestinal inflammation is of high importance. Natural products are suitable candidates among which Kombucha tea has shown healing effect on mice model of colitis. Methods: Filtered Kombucha tea was prepared from black tea and sucrose plus tea fungus and previously fermented Kombucha tea. The collection was fermented by incubation at 28 °C for 14 days and filtered. Colitis was induced in young and old mice by administration of 3.5% (w/v) dextran sodium sulfate in drinking water during 7 days; filtrated Kombucha tea was given orally to animals with colitis, at a dose volume of 10 mL/kg daily for 21 days. Dectin-1, f toll-like receptor (TLR)-2, 4, as PRRs, and MyD88, as PRR signaling molecule were measured and compared with the age-matched normal and colitis model. Results: Treatment with filtrated Kombucha tea significantly affected TLR2/TLR4 pathway and its downstream signaling molecules, MYD88 & dectin-1expression and subsequently inflammatory condition in dextran sodium sulfate-induced colitis. Conclusion: The results of the present study may indicate possible implication of Kombucha tea with TLRS which consequently produces the anti-colitis effects.

Background and Purpose: Seborrheic dermatitis (SD) is characterized by erythematous inflammatory patches that mostly appear in the sebaceous gland-rich skin areas. In addition to the key role of Malassezia species in SD, its contribution to other fungal microbiota has been recently addressed in the literature. Regarding this, the present study was conducted to identify and determine the fungal species associated with the incidence of SD. Materials and Methods: For the purpose of the study, fungal microbiome in scaling samples were collected from SD lesions and then analyzed based on the DNA sequencing of ITS regions. Results: In addition to Malassezia, several fungal species were detected in the samples collected from the SD lesions. According to the results, 15.5%, 13.3%, and 6.7% of the isolates were identified as Candida parapsilosis, Cryptococcus albidus var. albidus/ Rhodotorula mucilaginosa, and Penicillium polonicum, respectively. Conclusion: Based on the obtained results, C. parapsilosis was the most prevalent non-Malassezia species isolated from SD lesions. Our results provided basic information about a specific fungal population accounting for the incidence of SD.

Objective(s): Zonula occludens proteins (ZO-1 and ZO-2) are important intracellular tight junction (TJ)-associated proteins that link the cell cytoskeleton to the trans-membrane TJ proteins. Destruction of TJ proteins is called the “leaky gut syndrome” and has been observed in some of the gastrointestinal diseases such as the inflammatory bowel disease (IBD). So, therapeutic approaches aim to restore the expression of TJ proteins and reduce intestinal permeability. Healing effect of Kombucha tea (KT), so-called long-life mushroom, on the gastrointestinal system, particularly its extraordinary healing effects on intestinal ulcers has been purported traditionally and rarely reported scientifically. This study aimed to investigate the therapeutic effect of filtered KT (fKT) in young and old mice model of colitis. Materials and Methods: Leaky gut was induced in two groups of young and old age using dextran sodium sulfate in drinking water for seven days. Then, fKT was administered to the mice affected by colitis and compared with the age-matched normal and untreated animals with colitis. Results: Survival rate of the fKT-treated young and old animals with colitis increased and weight loss decreased. Accordingly, digestive disorders characterized by bleeding and diarrhea were improved in fKT-treated mice. Molecular and histological examination indicated that expression of ZO-1 and ZO-2 was significantly improved in fKT-treated mice. Conclusion: Our results suggest KT as a promising therapeutic candidate to reduce intestinal permeability. Young animals with colitis showed more severe clinical signs and less survival rate than old mice with colitis, but this group responded better to fKT treatment than the old mice.

Background and Purpose: Candida species are reportedly the most common human fungal pathogens. The incidence of urinary tract infections (UTIs) caused by Candida pathogens has increased in recent decades. However, such infections rarely occur in the absence of any predisposing factors. Regarding this, the aim of the present study was to identify the Candida species causing UTIs and determine the predisposing factors for candiduria. Materials and Methods: The current study was conducted on 1,450 urine samples obtained from patients suspected of UTI. Out of this number, 19 cases were candidiasis, and 2 cases were mixed infections caused by bacteria and fungi. Candida species were diagnosed differentially using the germ tube test, colony staining on CHROMagar medium, intracellular beta-glucosidase enzyme activity, and glucose absorption pattern. Then, the colonies with the same morphology were confirmed by the DNA sequencing of internal transcribed spacer regions. Results: According to the results, 38%, 28.6%, 14.3%, and 9.5% of the isolates were identified as C. albicans, C. glabrata, C. tropicalis, and C. kefir/C. krusei, respectively. The presence of one or more predisposing factors was proved in all patients in whom diabetes was the most prevalent predisposing factor (21.1%). Conclusion: Based on the obtained results, C. albicans species was the most prevalent fungal species. In addition, urinary fungal infections were less prevalent than bacterial urinary infections.

Background and objectives: Considering the increasing prevalence of depression, many studies are launched to investigate new antidepressant treatments. The present research has shown how psilocybin as an active compound of Psilocybe cubensis (Earle) Singer extract (PCE) can change the parameters related to depression and anxiety in animal models. Both serotonin (5-hydroxytryptamine: 5-HT) and glutamate modulate depressive-like behaviors and, therefore, we examined the possible interaction of psilocybin as 5-HT1 agonist with glutamate receptor N-methyl-D-aspartate (NMDA). Methods: Psilocybe cubensis extract of this mushroom was prepared by ethyl acetate. NMRI mice involved in all experiments and were treated with the vehicle, extract, or standard drug intraperitoneally. Open field (OFT), forced swimming (FST) and tail suspension tests (TST) were applied to measure the intended parameters. OFT was performed to verify the applied doses for measuring the following antidepressant activity. Results: PCE at the doses of 100 mg/kg significantly changed the locomotion, time spent in center and velocity of the animals in OFT. While treatment of the animals with PCE 10 and 40 mg/kg or ketamine 1 mg/kg did not alter the locomotor activity, co-administration of these subeffective amounts significantly reduced the immobility time in both FST and TST. Conclusion: These effects may indicate possible implication of psilocybin with NMDA receptor which consequently produces the antidepressant effects.

Background and Purpose: Seborrheic dermatitis is a chronic and recurrent superficial dermatitis in which Malassezia species play an important role. There are different Malassezia species, which have been recently reported to be resistant to common antifungals. Natural sources can be useful alternatives to reduce the emergence of this resistance. Kombucha tea is believed to have potential antimicrobial properties. Regarding this, the present study aimed to investigate theantifungal activity of Kombucha tea ethyl acetate fraction (KEAF) against Malassezia species obtained from the patients with seborrheic dermatitis. Materials and Methods: A total of 23 clinical isolates were identified by direct microscopic examination and Tween assimilation, and then confirmed by DNA sequencing of ITS regions for Malassezia species. Kombucha tea was fractionated using ethyl acetate (1:2 v/v). The minimum inhibitory concentration (MIC) microdilution assay was used to evaluate the anti-Malssezia activity of KEAF at three concentrations of 10, 40, and 80 mg/mL. Results: The results of the DNA sequence analysis indicated that M. furfur (39.13%) was the predominant species,followed by M. globosa (30.43%), M. sloofie (13.04%), M. sympodialis (13.04%), and M. restricta (4.34%), respectively. Furthermore, KEAF showed inhibitory activity against Malassezia species. Accordingly, KEAF had the lowest and highest MIC value against M. sloofie and M. restricta, respectively. Moreover, the inhibitory effect of the extract was equivalent to that of ketoconazole at 4.8 µg/mL. Conclusion: The findings of the current study highlighted the antifungal properties of KEAF. Therefore, this extract can be promoted as complementary medicine for the treatment of the infections caused by Malassezia.

As task-based language teaching focuses on real word tasks and the learners need to complete these tasks in the process of learning a foreign or second language, it helps target language fluency and student confidence. That is why second and foreign language teachers and researchers have shown interest in TBLT. This study attempts to investigate the effects of employing task-based writing instruction on Iranian EFL learners’ writing competence. The participants included 69 Iranian EFL learners at the intermediate level and they were placed randomly into a control group and an experimental group. The students in the experimental group performed writing tasks using task-based language teaching techniques, while those in the control group practiced writing skills using traditional writing exercises. To collect the pre-test and post-test data, the researchers administered the writing sections of two paper-based TOEFL tests and analyzed the data through Statistical Package for Social Sciences using descriptive statistics, t-test, and analysis of variance. The results showed significant improvements in the writing ability of the Iranian EFL learners who practiced writing skills using task-based language teaching techniques. Besides, using task-based writing techniques improved the Iranian EFL learners’ ability significantly in terms of different aspects of the writing competence, including sentence mechanics, language use, vocabulary, content, and organization.
