Necrotic enteritis caused by Clostridium perfringens (C. perfringens) is characterized by poor performance and higher mortality rates in poultry farms. Novel dietary intervention involving bioactive molecules loaded into smart magnetized nano-system with a potent antioxidant function (quercetin-loaded Fe3O4-NPs), was evaluated for their impact on growth performance, intestinal immune and antioxidant defenses, and resistance against Clostridium perfringens in a necrotic enteritis challenge model. Four experimental groups comprising a total of 200 one-day-old Ross 308 broiler chickens were fed different diets: a control basal diet, a diet supplemented with quercetin (300 mg/kg), a diet with Fe3O4-NPs (60 mg/kg), and a diet with quercetin-loaded Fe3O4-NPs (300 mg/kg). These groups were then challenged with C. perfringens during the grower period. Dietary inclusion of quercetin-loaded Fe3O4-NPs prominently reduced C. perfringens colonization and its associated virulence genes expression, which subsequently restored the impaired growth performance and intestinal histopathological changes in challenged broilers. Quercetin-loaded Fe3O4-NPs supplemented group displayed higher Lactobacillus and Bifidobacterium counts, upregulation of intestinal host defense antimicrobial peptides related genes (avian β -defensin 6 and 12) and downregulation of intestinal inflammatory regulated genes (Interleukin-1 beta, C-X-C motif chemokine ligand 8, tumor necrosis factor- α , chemokine C–C motif ligand 20, inducible nitric oxide synthase and cycloox-ygenase-2). Intestinal redox balance was boosted via upregulation of catalase, superoxide dismutase, glutathione peroxidase and heme Oxygenase 1 genes along with simultaneous decrease in hydrogen peroxide, reactive oxygen species and malondialdehyde contents in groups fed quercetin-loaded Fe3O4-NPs. Overall, new nutritional intervention with quercetin-loaded Fe3O4-NPs impacted better immune and antioxidant defenses, attenuated C. perfringens induced necrotic enteritis and contributed to better performance in the challenged birds. Coccidiosis, caused by the protozoan Eimeria, is a significant disease in poultry farms worldwide, leading to substantial economic losses. Triazines, benzene-aceto-nitrile derivatives, are widely employed in the field of veterinary healthcare to combat the hazardous impacts of protozoan parasite infestation comprising coccidiosis. The current research was designed to investigate the pharmacokinetic profile of diclazuril, a member of triazines, in Japanese quails (Coturnix japonica) and domestic pigeons (Columba livia) following single oral administration at 0.3 mg/kg body weight. 78 Quails (male: female, 1:1, 7 weeks old) and 78 pigeons (male: female, 1:1, 4 weeks old) were randomly divided into 13 groups for each species (n=6 birds/group). Plasma samples were obtained at various time intervals (at time 0 [preceding diclazuril administration], and 0.5, 1, 4, 8, 12, 24, 48, 72, 96, 120, 144, and 168 hours after diclazuril administration) to determine its concentration utilizing high-performance liquid chromatography (HPLC). The noncompartmental approach was applied to assess the pharmacokinetic parameters via the aid of WinNonlin 8.3 software. In quails and pigeons, the peak plasma concentrations were 5.35 and 9.14 µg/mL attained at 8 hours, respectively. Additionally, the elimination half-lives (T1/2-z) were 30.74 and 26.48 hours, and the area under the plasma concentration-time curve from time zero to the last sample (AUC0-last) values were 155.67 and 343.57 µg h/mL, respectively. The mean residence time was 30.71 hours in quails and 39.68 hours in pigeons. Diclazuril exhibited favorable pharmacokinetic characteristics after oral administration at a dose of 0.3 mg/kg in quails and pigeons. However, to adjust the dosage regimen for curing coccidiosis, a future study is warranted to determine the clinical efficacy against coccidia infection. The pharmacokinetics of tylosin were investigated in 3 groups of ducks (n = 6). They received a single dose of tylosin (50 mg/kg) by intravenous (IV), intramuscular (IM), and oral administrations, respectively. Plasma samples were collected at various time points to 24 hr post‐administration to evaluate tylosin concentration over time. Additionally, tylosin residues in tissues and its withdrawal time were assessed using 30 ducks which received tylosin orally (50 mg/kg) once daily for 5 consecutive days. After IV administration, the volume of distribution, elimination half‐life, area under the plasma concentration–time curve, and the total body clearance were 7.07 ± 1.98 L/kg, 2.04 hr, 19.47 µg hr/ml, and 2.82 L hr−1 kg−1, respectively. After IM and oral administrations, the maximum plasma concentrations were 3.70 and 2.75 µg/ml achieved at 1 and 2 hr, and the bioavailability was 93.95% and 75.77%, respectively. The calculated withdrawal periods of tylosin were 13, 8, and 5 days for kidney, liver, and muscle, respectively. For the pharmacodynamic profile, the minimum inhibitory concentration for tylosin against M. anatis strain 1,340 was 1 µg/ml. The calculated optimal oral dose of tylosin against M. anatis in ducks based on the ex vivo pharmacokinetic/pharmacodynamic modeling was 61 mg kg−1 day−1. [ABSTRACT FROM AUTHOR]

Residues of veterinary drugs in poultry meat have serious health effects on humans (e.g., increase antimicrobial resistance, carcinogenicity, mutagenicity and hypersensitivity) which make the control of veterinary drug residues an important parameter in ensuring consumer protection. This work was performed to quantitatively determine minocycline residues in different tissues of chickens (kidneys, liver, muscles and fat) and sera following multiple oral doses of the drug using High Performance Liquid Chromatography (HPLC). Moreover, the study aimed to estimate the withdrawal time of this drug in chicken tissues. Twenty five healthy chicks (Cobb 500) were used. Twenty one chickens were given minocycline directly into the stomach at a dosage of 7 mg/kg body weight once daily for five successive days. Samples were analyzed at 1st, 3rd, 7th, 14th, 21st and 28th day after last oral dose. The results indicated a widespread distribution of minocycline in the tissue samples, which remained within the detectable limit till the 3rd day (49.20-135.20 μg/kg) in all tested tissues. While in kidneys, minocycline remained till the 7th day (11.80μg/kg) following the last oral administration of the drug. Therefore, it is recommended to pay attention to the proper withdrawal periods before marketing to ensure the hygienic suitability of broilers edibles for safe human consumption.
