- Abstract
Author summary Since the first outbreaks, Flavescence doree epidemics had been associated to the introduction of the North American leafhopper vector Scaphoideus titanus. We hereby show that the associated phytoplasma originated from European alders and that epidemics in grapevine are restricted to some phytoplasma genetic variants pre-existing in this wild asymptomatic plant host. The compatibility of this phytoplasma to the introduced S. titanus insect vector resulted from the adaptation of phytoplasma variable membrane proteins Vmps to leafhoppers of the same subfamily living on alders. Vmps organization is similar to adhesion related proteins (ARP) and seems to allow the duplication of pre-adapted repeated domains. This suggests a key role of the Vmp adhesins in the life-style of phytoplasmas infecting woody hosts that rely on the adaptation to new insect vectors to expand their plant-host range. Flavescence doree (FD) is a European quarantine grapevine disease transmitted by the Deltocephalinae leafhopper Scaphoideus titanus. Whereas this vector had been introduced from North America, the possible European origin of FD phytoplasma needed to be challenged and correlated with ecological and genetic drivers of FD emergence. For that purpose, a survey of genetic diversity of these phytoplasmas in grapevines, S. titanus, black alders, alder leafhoppers and clematis were conducted in five European countries. Out of 132 map genotypes, only 11 were associated to FD outbreaks, three were detected in clematis, whereas 127 were detected in alder trees, alder leafhoppers or in grapevines out of FD outbreaks. Most of the alder trees were found infected, including 8% with FD genotypes M6, M38 and M50, also present in alders neighboring FD-free vineyards and vineyard-free areas. Variability of vmpA and vmpB adhesin-like genes clearly discriminated 3 genetic clusters. Cluster Vmp-I grouped genotypes only transmitted by O. alni, while clusters Vmp-II and -III grouped genotypes transmitted by Deltocephalinae leafhoppers. Interestingly, adhesin repeated domains evolved independently in cluster Vmp-I, whereas in clusters Vmp-II and–III showed recent duplications. Latex beads coated with various ratio of VmpA of clusters II and I, showed that cluster II VmpA promoted enhanced adhesion to the Deltocephalinae Euscelidius variegatus epithelial cells and were better retained in both E. variegatus and S. titanus midguts. Our data demonstrate that most FD phytoplasmas are endemic to European alders. - Abstract
A survey of genetic diversity of “flavescence dorée” (FD)-related phytoplasmas in grapevines, alders and clematis as well as alder-feeding leafhoppers was conducted in France, Hungary, Germany, Italy and Serbia. Genotyping was based on the housekeeping gene map and on the vmp genes encoding surface variable membrane proteins. Transmission assays of the phytoplasmas were performed with alder and/or grapevine-feeding leafhoppers. The study demonstrated that European alders constitute an original reservoir of FD phytoplasma by hosting a high diversity of FD-related phytoplasma genotypes, also present in non viticultural areas. The alder phytoplasmas grouping in Vmp-I cluster were transmitted by the Macropsinae O. alni, but were not compatible with the FD phytoplasma vector on grapevine the Deltocephalinae Scaphoideus titanus. The alder phytoplasmas in Vmp-II and -III clusters were transmitted by the Deltocephalinae Allygus spp. and Orientus ishidae. Such pre-existing phytoplasmas were compatible with S. titanus transmissibility and can be responsible for the emergence of FD phytoplasma epidemics in grapevine. International audience; Impact of "bois noir" disease (BN) on performance of 'Chardonnay' grown in Eger (Hungary) is reported from plant (vegetative and reproductive parameters, fruit composition) to wine (wine quality, sensory analysis). BN-associated chlorophyll degradation and leaf rolling resulted in a loss in functioning surface of the canopy. Lack of lignification and non-viable buds evoked unpredictable crop production. Severity and extent of reduction in production varied between years, but the crop loss was always above 53%. In the case of BN-affected grapevines the reduced quantity resulted in a low quality of grape bunch composition, which enhanced unfavourable outcomes in wines. Differences in fruit composition and wine quality between healthy and BN affected grapevines were most pronounced in years with optimal weather conditions and the negative effect was covered up in poor years. In wine regions where mechanical harvesting is applied, harvesting of phytoplasma diseased vineyards can therefore not be selective and qualitative. International audience; Little is known about the mechanisms of insects - phytoplasma interactions which are driving the ecological diversification of phytoplasmas. Phytoplasma surface proteins play an important role in phytoplasma life cycle and polymorphism of this protein might determine the transmission ability of different insect species. Strains of Candidatus Phytoplasma solani' from several European countries, based on variable membrane protein gene (stamp), were classified into four distinct phylogenetic clusters which clusters are present in different geographical areas. This may reflect the distribution of the different known insect vectors and the adaptation of the various 'Ca. P. solani' strain to a specific vector. To detect STAMP protein, monoclonal antibody 2A10 is available. This antibody was produced against StolburC, which is a representative strain of stamp clusters I. International audience; In order to evaluate the risk represented by the wild reservoir as a possible source of 'flavescence doree' outbreaks in Hungary, diverse wild perennial plants growing in vineyard areas were tested for the presence of 16SrV-C and D subgroup phytoplasmas. 16SrV phytoplasmas were detected by nested PCR-RFLP on the 16SrDNA in alders (86% infected) and in clematis (71% infected). In the frame of the bilateral Croatian-Hungarian project, viticulture regions with widespread Bois Noir (BN) and potential vector species were identified. Several areas adjoining state borders potentially having similar BN pathosystems were targeted for molecular characterization of pathogens in grapevines, weeds and insects. The total of 91 insect samples collected in 2010 was tested for phytoplasma presence. Most samples (71) consisted of individual Hyalestes obsoletus adults, out of which 25 were caught in the vineyard of Vukanovec (Međimurje) with Urtica dioica as a dominant weed of known importance for BN epidemiology. The rest of the insects were collected from other vineyards in Međimurje, Zagreb and Baranja with Convolvulus arvensis as a dominant weed. Insect populations screened in Međimurje included also Reptalus cuspidatus, whilst Baranja vineyards harboured more diverse insect populations including R. quinquecostatus, Anaceratagallia ribauti, Lygus rugulipennis, Cixius sp., Philaneus spumarius, and Laodelphax striatella, besides the above mentioned species. Total DNA was isolated by using MagNA Pure LC Instrument and MagNA Pure LC DNA Tissue Isolation Kit II (Roche) mostly from individual insects. Templates obtained from this magnetic-bead technology procedure were used for conventional direct and nested PCR employing P1/P7 (Deng and Hiruki, 1991 ; Schneider et al., 1995) and R16F2n/R2 (Gundersen and Lee, 1996) generic phytoplasma primers, respectively. Identification of stolbur phytoplasma DNA was performed by RFLP after digestion with the enzyme Tru1I (Fermentas). BN-positive insect samples were subjected to Tuf-typing (Langer and Maixner, 2004). In parallel, the DNA was tested by real-time PCR using modified Pelletier et al. (2009). BN phytoplasma DNA was amplified from 20 individual insects in conventional PCR-RFLP and real-time PCR experiments. The individual insects harbouring BN DNA were H. obsoletus from Vukanovec (2), Jastrebarsko (9) and Baranja (8), as well as a pooled sample of Laodelphax striatella from Baranja. Tuf-types were obtained for 15 H. obsoletus. Only one H. obsoletus from Vukanovec, a pathosystem dominated by U. dioica, was found to harbour VK-Type I. The rest was infected by VK-Type II, including the other positive H. obsoletus from Vukanovec, indicating that VK-Type II is more common and geographically more widespread along the Croatian-Hungarian border. Finding both VK-Types in insects caught in close proximity (Vukanovec) indicate that this ecological niche may involve both Urtica and Convolvulus type BN pathosystems. During 2010, several maize production areas in Hungary were surveyed for the occurrence of maize redness (MR) disease symptoms associated with stolbur phytoplasma, as well as for the presence of the known vector of the disease, a planthopper Reptalus panzeri (Low). Incidence of maize plants with symptoms of reddening was low in all surveyed areas. Altogether, 25 symptomatic maize plants were collected at 9 localities and tested for phytoplasma presence. In addition, from one locality specimens of cixiids R. panzeri and Hyalesthes obsoletus Signoret were collected and PCR analyzed. Presence of stolbur phytoplasma in MR symptomatic maize plants and stolbur-infected R. panzeri was identified at the single locality Monorierdo in central Hungary. In August 2010 potato plants expressing symptoms of phytoplasma infection were collected from three localities in Vojvodina province of Serbia and analyzed for phytoplasma presence. Phytoplasma detection was performed by PCR/RFLP analyzes of 16S rRNA gene and by stolbur-specific PCR amplification with Stol11 primers.
