The occurrence of cryptic and pseudocryptic species, often living in sympatry, is widespread among microalgae. This phenomenon raises important questions about niche partitioning between these closely related species. To date, however, few studies have addressed the ecological mechanisms underlying sympatry in cryptic and pseudocryptic species. As a result, we have only a limited understanding of the factors that govern their distribution along environmental gradients. Here, we used the ribosomal internal transcribed spacer (ITS), 18S rRNA gene, and the RUBISCO LSU (rbcL) chloroplast gene sequence data together with cell wall morphology to show that estuarine populations of the widespread and common benthic diatom Navicula phyllepta Kütz. consist of pseudocryptic species. Growth rate measurements in function of salinity showed that N. phyllepta strains assigned to the different species differed in their tolerance to low salinities (5 practical salinity units, psu), which was reflected by their different (but widely overlapping) distribution in the Westerschelde estuary (the Netherlands). Multiple regression analyses of the factors determining the abundance of the different species in field samples revealed that, in addition to salinity, sediment type and ammonium concentrations were probably equally important. Our results show that N. phyllepta sensu lato comprises different species with specialized ecophysiological characteristics rather than generalists with a broad adaptability to different environmental conditions.

The diversity and spatio-temporal distribution of ammonia-oxidizing Archaea (AOA) and Bacteria (AOB) were investigated along a salinity gradient in sediments of the Westerschelde estuary. Sediment samples were collected from three sites with different salinities, and at six time points over the year. Denaturing gradient gel electrophoresis of PCR-amplified 16S rRNA and amoA gene fragments was used to identify the AOA and AOB present. Members of the AOA were mainly belonging to the Crenarchaeota Group 1, which were found at all sites, while members of the genus Nitrosomonas, which were abundant at the brackish sites, and of the genus Nitrosospira, which were present in early spring at the marine sites, were found to be the dominant AOB. Statistical analysis indicated that salinity and temperature were the main factors controlling the diversity and distribution of both AOA and AOB. Variability in net primary production rates was also correlated with species composition of both groups, but changes in the nitrite concentration only to the distribution of the AOA.

The physiological adaptation of the phosphate uptake behaviour of the cyanobacterium Anacystis nidulans (Synechococcus leopoliensis) has been studied under phosphate-deficient conditions. We show that, during additions of pulses of phosphate, the individual cells of a population potentially develop a coherent behaviour with respect to the kinetic and energetic properties of the high-affinity phosphate uptake system. These adaptive responses are independent of the amount of stored phosphate and the absolute external phosphate concentration, but depend on the exposure time to elevated phosphate concentrations. We demonstrate that the response to additions of pulses of phosphate triggers further metabolic alterations to CO2 fixation, and the flow of fixed carbon into the pool of low-molecular weight-compounds or glycogen. International audience; The composition and seasonal dynamics of biofilm-associated eukaryotic communities were analysed at the metre and kilometre scale along a salinity gradient in the Westerschelde estuary (The Netherlands), using microscopy and a genetic fingerprinting technique (PCR-DGGE). Microphytobenthic biomass, measured as chlorophyll a (chl a), varied seasonally over 2 orders of magnitude, being highest in spring. Communities were dominated by epipelic diatoms, in particular by members of the genus Navicula. In spring, a few smaller epipelic diatom species dominated during biomass peaks, while during the rest of the year, communities were more diverse and were characterised by larger species. The microphytobenthic community collapsed when grazers appeared, which happened concomitantly with a rise in temperature. Spring biomass development was associated with marked changes in porewater nutrient concentrations, especially towards the estuary mouth. In the DGGE data, diatoms, ciliates, amoebae, copepods, nematodes, annelids and platyhelminthes were detected. Ordination analysis of the species counts and DGGE data were largely congruent and indicated that on the scale of the whole estuary (i.e. km scale), taxonomic turnover in microphytobenthos composition was mainly associated with the salinity gradient. At smaller spatial scales, the position of sampling localities along the tidal exposure gradient appeared to be the main determinant of species turnover, in particular in the brackish reaches of the estuary.

When algal and cyanobacterial growth in lakes is limited by the supply of available phosphate, the external concentration of this nutrient decreases to nanomolar levels [1], and rapidly approaches a stationary value at which net uptake ceases for energetic reasons [2]. Phosphate uptake and subsequent growth is then only possible if the external phosphate concentration exceeds at least occasionally this stationary or threshold value, for example after excretion of this nutrient by zooplankton or fish.

This paper presents the results of studies into the use of the emerging techniques of nutrient induced fluorescence transients (NIFTs) and Fourier Transform InfraRed (FTIR) spectroscopy to determine the nutrient status of microalgae. Four species of microalgae were grown under conditions where growth rate was P-limited or P-replete, and NIFT responses and FTIR spectra in response to the re-supply of P (as $ {\rm PO}^{3-}_4 $ ) measured. These responses were compared to more conventional measures of algal nutrient status such as P-uptake rates, P quotas and transient effects of $ {\rm PO}^{3-}_4 $ on oxygen exchange. The NIFT technique and FTIR spectroscopy gave results that were consistent with those obtained by the other techniques. Furthermore, we were able to demonstrate NIFT responses in phytoplankton samples taken from Lake Lucerne (total ambient P $ \le $ 0.13 μM) but not from Lake Zurich (total P 0.55 μM). The potential and limitations of the various techniques are discussed.
