1. Mobilizing sorghum genetic diversity: Biochemical and histological‐assisted design of a stem ideotype for biomethane production

Abstract: Abstract Sorghum currently contributes to the species portfolio that is supporting bioenergy production including anaerobic digestion. Although agro‐morphological ideotypes maximizing biogas production have been recently proposed, there is a crucial need to refine our understanding of the impacts of the stem composition and structure on this processing trait in order to ensure genetic gains in the mid to long terms. This study aims to assess the potential of Sorghum bicolor ssp bicolor stem genetic diversity to maximize genetic gains for biogas production and define a biogas stem ideotype. In this context, a panel of 57 genotypes, encompassing most of the stem composition variability available in cultivated sorghum, was characterized over five sites. Simultaneous histological and biochemical characterizations were performed. A high broad sense heritability associated with a moderate genetic variability was detected for stem biogas potential ensuring significant genetic gains in the future. In addition, the development of a stem histological phenotyping pipeline made it possible to describe the genetic diversity available for the internode anatomy and the repartition of key cell wall components. Consistently with previous studies, moderate to high heritability was observed for stem biochemical components. Genetic correlation, hierarchical clustering, and multiple stepwise regression analyses identified soluble sugar content as the first main driver of biogas potential genetic variability. Nevertheless, breeding programs should anticipate that biogas yield improvement will also rely on the monitoring of the cell wall components and their distribution in the stem jointly with the soluble sugar content. According to the assets of sorghum in terms of adaptation to environmental stresses and the present results regarding the identification of stem ideotypes suitable for different value chains, this species will surely play a key role to optimize the economic and environmental sustainability of the agrosystems that are currently facing the effects of climate change.

2. An 'overlooked' habitat for thermophilic bacteria: the phyllosphere

Abstract: Thermophilic microbes are present everywhere around us and their only known natural biotope is far away and most usually associated with geothermal energy. To answer this paradox, we explore the hypothesis that the phyllosphere (surface of leaves), due to its exposition to the sun, could well be a thermophilic habitat for microbes and thus a source of thermophilic microbes growing around 50°C – 60°C. To support this hypothesis, we reviewed the heat sources on earth and associated microbial habitats, as well as the difficult identification of thermophilic microbes. We further present an experiment to show the presence and activity of thermophilic bacteria in the phyllosphere. Leaves were collected from eleven tree species from five locations on three continents belonging to three different biomes. On fresh leaves, 16S rDNA sequencing reveals the presence of 0.2 to 7% of clearly identified thermophilic bacteria. Moreover, after incubation at 55°C under aerobic and anaerobic conditions, 16S rDNA sequencing reveals the presence of 4 to 99% of clearly identified thermophilic bacteria. The accumulation of observations provides coherence to our hypothesis and allows the emergence of a new vision of leaves as a thermophilic biotope. We then propose a life cycle of microbes belonging to the thermophilic biotope associated with leaf surfaces.

3. Multiplexed chemostat system for quantification of biodiversity and ecosystem functioning in anaerobic digestion.

Abstract: Continuous cultures in chemostats have proven their value in microbiology, microbial ecology, systems biology and bioprocess engineering, among others. In these systems, microbial growth and ecosystem performance can be quantified under stable and defined environmental conditions. This is essential when linking microbial diversity to ecosystem function. Here, a new system to test this link in anaerobic, methanogenic microbial communities is introduced. Rigorously replicated experiments or a suitable experimental design typically require operating several chemostats in parallel. However, this is labor intensive, especially when measuring biogas production. Commercial solutions for multiplying reactors performing continuous anaerobic digestion exist but are expensive and use comparably large reactor volumes, requiring the preparation of substantial amounts of media. Here, a flexible system of Lab-scale Automated and Multiplexed Anaerobic Chemostat system (LAMACs) with a working volume of 200 mL is introduced. Sterile feeding, biomass wasting and pressure monitoring are automated. One module containing six reactors fits the typical dimensions of a lab bench. Thanks to automation, time required for reactor operation and maintenance are reduced compared to traditional lab-scale systems. Several modules can be used together, and so far the parallel operation of 30 reactors was demonstrated. The chemostats are autoclavable. Parameters like reactor volume, flow rates and operating temperature can be freely set. The robustness of the system was tested in a two-month long experiment in which three inocula in four replicates, i.e., twelve continuous digesters were monitored. Statistically significant differences in the biogas production between inocula were observed. In anaerobic digestion, biogas production and consequently pressure development in a closed environment is a proxy for ecosystem performance. The precision of the pressure measurement is thus crucial. The measured maximum and minimum rates of gas production could be determined at the same precision. The LAMACs is a tool that enables us to put in practice the often-demanded need for replication and rigorous testing in microbial ecology as well as bioprocess engineering.

4. Needles of Pinus halepensis as biomonitors of bioaerosol emissions.

Abstract: We propose using the surface of pine trees needles to biomonitor the bioaerosol emissions at a composting plant. Measurements were based on 16S rRNA gene copy numbers of Saccharopolyspora rectivirgula, a bioindicator of composting plant emissions. A sampling plan was established based on 29 samples around the emission source. The abundance of 16S rRNA gene copies of S. rectivirgula per gram of Pinus halepensis needles varied from 104 to 102 as a function of the distance. The signal reached the background level at distances around the composting plant ranging from 2 km to more than 5.4 km, depending on the local topography and average wind directions. From these values, the impacted area around the source of bioaerosols was mapped.

Abstract: Hydrolysis is considered the limiting step during solid waste anaerobic digestion (including co-digestion of sludge and biosolids). Mechanisms of hydrolysis are mechanistically not well understood with detrimental impact on model predictive capability. The common approach to multiple substrates is to consider simultaneous degradation of the substrates. This may not have the capacity to separate the different kinetics. Sequential degradation of substrates is theoretically supported by microbial capacity and the composite nature of substrates (bioaccessibility concept). However, this has not been experimentally assessed. Sequential chemical fractionation has been successfully used to define inputs for an anaerobic digestion model. In this paper, sequential extractions of organic substrates were evaluated in order to compare both models. By removing each fraction (from the most accessible to the least accessible fraction) from three different substrates, anaerobic incubation tests showed that for physically structured substrates, such as activated sludge and wheat straw, sequential approach could better describe experimental results, while this was less important for homogeneous materials such as pulped fruit. Following this, anaerobic incubation tests were performed on five substrates. Cumulative methane production was modelled by the simultaneous and sequential approaches. Results showed that the sequential model could fit the experimental data for all the substrates whereas simultaneous model did not work for some substrates. © 2019

Abstract: Anaerobic digestion is an increasingly widespread process for organic waste treatment and renewable energy production due to the methane content of the biogas. This biological process also produces a digestate (i.e., the remaining content of the waste after treatment) with a high fertilizing potential. The digestate composition is highly variable due to the various organic wastes used as feedstock, the different plant configurations, and the post-treatment processes used. In order to optimize digestate spreading on agricultural soils by optimizing the fertilizer dose and, thus, reducing environmental impacts associated to digestate application, the agronomic characterization of digestate is essential. This study investigates the use of near infrared spectroscopy for predicting the most important agronomic parameters from freeze-dried digestates. A data set of 193 digestates was created to calibrate partial least squares regression models predicting organic matter, total organic carbon, organic nitrogen, phosphorus, and potassium contents. Abstract: It is a real challenge for life cycle assessment practitioners to identify all relevant substances contributing to the ecotoxicity. Once this identification has been made, the lack of corresponding ecotoxicity factors can make the results partial and difficult to interpret. So, it is a real and important challenge to provide ecotoxicity factors for a wide range of compounds. Nevertheless, obtaining such factors using experiments is tedious, time-consuming, and made at a high cost. A modeling method that could predict these factors from easy-to-obtain information on each chemical would be of great value. Here, we present such a method, based on machine learning algorithms, that used molecular descriptors to predict two specific endpoints in continental freshwater for ecotoxicological and human impacts. The different tested machine learning algorithms show good performances on a learning database and the non-linear methods tend to outperform the linear ones. The cluster-then-predict approaches usually show the best performances which suggests that these predicted models must be derived for somewhat similar compounds. Finally, predictions were derived from the validated model for compounds with missing toxicity/ecotoxicity factors.HighlightsCharacterization factors (for human health and ecotoxicological impacts) were predicted using molecular descriptors.Several linear or non-linear machine learning methods were compared.The non-linear methods tend to outperform the linear ones using a train and test procedure. Cluster-then-predict approaches often show the best performances, highlighting their usefulness.This methodology was then used to derive characterization factors that were missing for more than a hundred chemicals in USEtox®.Graphical abstract

Abstract: International audience; In near-infrared spectroscopy (NIRS), the linear relationship between absorbance and an absorbing compound concentration has been strictly defined by the Bouguer-Beer-Lambert law only for the case of transmission measurements of nonscattering media. However, various quantitative calibrations have been successfully built both on reflectance measurements and for scattering media. Although the lack of linearity for scattering media has been observed experimentally, the sound multivariate statistics and signal processing involved in chemometrics have allowed us to overcome this problem in most cases. However, in the case of samples with varying water content, important modifications of scattering levels still make calibrations difficult to build due to nonlinearities. Moreover, even when calibration procedures are successfully developed, many preprocessing methods used do not guarantee correct spectroscopic assignments (in the sense of a pure chemical absorbance). In particular, this may prevent correct modeling and interpretation of the structure of water. In this study, dynamic near-infrared spectra acquired during a drying process allow the study of the physical effects of water content variations, with a focus on the first overtone OH absorbance region. A model sample consisting of aluminum pellets mixed with water allowed us to study this specifically, without any other absorbing interaction terms related to the dry mass-absorbing constituents. A new formulation of the Bouguer-Beer-Lambert law is proposed, by expressing path length as a power function of water content. Through this new formulation, it is shown that a better and simpler prediction model of water content may be developed, with more precise and accurate identification of water absorbance bands.

Abstract: International audience; Fast characterization of solid organic waste using near infrared spectroscopy has been successfully developed in the last decade. However, its adoption in biogas plants for monitoring the feeding substrates remains limited due to the lack of applicability and high costs. Recent evolutions in the technology have given rise to both more compact and more modular low-cost near infrared systems which could allow a larger scale deployment. The current study investigates the relevance of these new systems by evaluating four different Fourier transform near-infrared spectroscopic systems with different compactness (laboratory, portable, micro spectrometer) but also different measurement configurations (polarized light, at distance, in contact). Though the conventional laboratory spectrometer showed the best performance on the various biochemical parameters tested (carbohydrates, lipids, nitrogen, chemical oxygen demand, biochemical methane potential), the compact systems provided very close results. Prediction of the biochemical methane potential was possible using a low-cost micro spectrometer with an independent validation set error of only 91 NmL(CH4).gTS-1 compared to 60 NmL(CH4).gTS-1 for a laboratory spectrometer. The differences in performance were shown to result mainly from poorer spectral sampling; and not from instrument characteristics such as spectral resolution. Regarding the measurement configurations, none of the evaluated systems allowed a significant gain in robustness. In particular, the polarized light system provided better results when using its multi-scattered signal which brings further evidence of the importance of physical light-scattering properties in the success of models built on solid organic waste.

Abstract: International audience; This article aims at providing insight into the recirculation of (post-treated) solid digestate (SD) within digesters. Such a practice has been further identified as promising for anaerobically digesting recalcitrant feedstocks and for improving the energy efficiency of continuously stirred tank reactor (CSTR) agricultural biogas plants. Firstly, implementation strategies and the potential impact of SD direct recirculation on five CSTR biogas plants were investigated. According to the selected strategy, results indicated that plant methane production could rise by 0.6 to 6.3% or that a potential feedstock shortage of 64 to 1431 tons/year could be compensated. Secondly, the relevance of additional post-treatments for improving these initial results was evaluated. Thermo-chemical post treatments successfully increased SD biodegradability by 30 to 46% although their costs were not compensated by additional methane production. Short-term aerobic post-treatments failed in increasing SD biodegradability (up to 21% loss in biomethane potential). Hence, at full scale, a quick and direct recirculation of SD excluding any post-treatment appears to be the optimal condition to apply. Finally, conditions for the full-scale implementation of direct SD recirculation were theoretically studied. This practice has proved to increase the solid retention time by 11 to 38% and the plant total solid content by 6 to 20%. Thus, the critical point for its implementation should be the capacity of the plant mixing system to handle such an increase in solids. The relevance of SD recirculation needs to be determined on a case-by-case basis. Consequently, for the first time, this article provides a framework where the conditions can be identified for direct SD recirculation to be a relevant digestate management practice. Overall, this article demonstrates how direct SD recirculation can be a simple and low-cost mean for improving agricultural CSTR biogas plant efficiency. It also highlights the importance of achieving efficient digestate management in the biogas sector in order to reduce the costs of biogas production.

Abstract: International audience; In the context of organic waste management, near infrared spectroscopy (NIRS) is being used to offer a fast, non-destructive, and cost-effective characterization system. However, cumbersome freeze-drying steps of the samples are required to avoid water’s interference on near infrared spectra. In order to better understand these effects, spectral variations induced by dry matter content variations were obtained for a wide variety of organic substrates. This was made possible by the development of a customized near infrared acquisition system with dynamic highly-resolved simultaneous scanning of near infrared spectra and estimation of dry matter content during a drying process at ambient temperature. Using principal components analysis, the complex water effects on near infrared spectra are detailed. Water effects are shown to be a combination of both physical and chemical effects, and depend on both the characteristics of the samples (biochemical type and physical structure) and the moisture content level. This results in a non-linear relationship between the measured signal and the analytical characteristic of interest. A typology of substrates with respect to these water effects is provided and could further be efficiently used as a basis for the development of local quantitative calibration models and correction methods accounting for these water effects.

Abstract: International audience; Following treatment, amounts of pesticides can reach the atmosphere because of spray drift, volatilization from soil or plants, and/or wind erosion. Monitoring and risk assessment of air contamination by pesticides is a recent issue and more insights on pesticide transfer to atmosphere are needed. Thus, the objective of this work was to better understand and assess pesticide emission potentials to air through volatilization. The TyPol tool was used to explore the relationships between the global, soil and plant volatilization potentials of 178 pesticides, and their molecular properties. The outputs of TyPol were then compared to atmospheric pesticide concentrations monitored in various French regions. TyPol was able to discriminate pesticides that were observed in air from those that were not. Clustering considering parameters driving the emission potential from soil (sorption characteristics) or plant (lipophilic properties), in addition to vapor pressure, allowed better discrimination of the pesticides than clustering considering all parameters for the global emission potential. Pesticides with high volatilization potential have high total energy, and low molecular weight, molecular connectivity indices and polarizability. TyPol helped better understand the volatilization potentials of pesticides. It can be used as a first step to assess the risk of air contamination by pesticides.

Abstract: Total pressure (TP) affects the level of dissolved hydrogen gas in the fermentation medium leading to metabolic shifts in mixed microbial-culture-based systems. In this study, the effect on hydrogen production rate and yield was investigated at different TP of a hydrogen-producing system using a microbial non-sterile culture previously heat-treated. Four continuous stirred-tank reactors (CSTR) were operated in parallel on a mineral salts-molasses medium (21 g-COD. L−1) at 35 °C, pH 5.5 and hydraulic retention time (HRT) of 6 h. The TP was set at 80 kPa (R1), 100 kPa (R2), 120 kPa (R3) and 140 kPa (R4) for which reactor performances were estimated at steady-state conditions. As the increase of TP consequently increased the partial pressure of hydrogen ( p H 2 ), the hydrogen production rate (HPR) and yield (HY) were consistently negatively influenced. The highest HPR and HY (406.1 ± 36.8 mL-H2 h−1; 4.51 molH2 mol−1suc eq.) were achieved at low pressure conditions (80 kPa). The composition of the microbial community mainly represented by species from Sporolactobacillus and Clostridium genera, did not change with the increase and/or decrease of the TP, indicating a regulation at cellular but not population level.

Abstract: The aim of this chapter is to provide a description of the main methods used to assess biological transformation of biomass. It will address saccharification tests which account for the ability of biomass to release sugars during enzymatic hydrolysis. Biochemical methane potential (BMP) tests, which are widely used to assess anaerobic digestibility of biomass for the production of methane and BMP prediction by near infra-red spectroscopy will be presented. Biohydrogen potential (BHP), used to assess the ability to produce biohydrogen from biomass by dark fermentation will be described. Respirometry tests accounting for assessing aerobic degradability of biomass will be described. Incubation tests used to assess carbon and nitrogen degradation in soils will also be presented.

Abstract: International audience; Life Cycle Assessment (LCA) characterises all the exchanges between human driven activities and the environment, thus representing a powerful approach for tackling the environmental impact of a production system. However, LCA practitioners must still choose the appropriate Life Cycle Impact Assessment (LCIA) method to use and are expected to justify this choice: impacts should be relevant facing the concerns of the study and misrepresentations should be avoided. This work aids practitioners in evaluating the adequacy between the assessed environmental issues and studied production system. Based on a geometrical standpoint of LCA framework, Life Cycle Inventories (LCIs) and LCIA methods were localized in the vector space spanned by elementary flows. A proximity measurement, the Representativeness Index (RI), is proposed to explore the relationship between those datasets (LCIs and LCIA methods) through an angular distance. RIs highlight LCIA methods that measure issues for which the LCI can be particularly harmful. A high RI indicates a close proximity between a LCI and a LCIA method, and highlights a better representation of the elementary flows by the LCIA method. To illustrate the benefits of the proposed approach, representativeness of LCIA methods regarding four electricity mix production LCIs from the ecoinvent database are presented. RIs for 18 LCIA methods (accounting for a total of 232 impact categories) were calculated on these LCIs and the relevance of the methods are discussed. RIs prove to be a criterion for distinguishing the different LCIA methods and could thus be employed by practitioners for deeper interpretations of LCIA results.

Abstract: International audience; Near-infrared spectrum analysis coupled with partial least squares regression can predict anaerobic digestion performances. Nonetheless, due to the complexity and diversity of organic matter, a detailed assessment of the effects of the composition of organic matter on biogas production remains a great challenge. Based on 275 samples representing a wide diversity of substrates, the application of the partial least square b coefficients to assess the effects of the involved molecules on the performances of anaerobic digestion processes is discussed. In particular, to accurately predict variables, there is a need to account for the whole near-infrared spectrum. The characterization of organic matter involving proteins, carbohydrate and lipid contents, chemical demand in oxygen, biodegradability, methane yield, and methane production kinetics data is demonstrated.

Abstract: The aim of this work is to evaluate biohydrogen production from agro-industrial wastewaters and by-products, by combining dark fermentation and microbial electrolysis in a two-step cascade process. Such coupling of both technologies constitutes a technological building block within a concept of environmental biorefinery where sustainable production of renewable energy is expected. Six different wastewaters and industrial by-products coming from cheese, fruit juice, paper, sugar, fruit processing and spirits factories were evaluated for the feasibility of hydrogen production in a two-step process. The overall hydrogen production when coupling dark fermentation and microbial electrolysis was increased up to 13 times when compared to fermentation alone, achieving a maximum overall hydrogen yield of 1608.6 ± 266.2 mLH2/gCODconsumed and a maximum of 78.5 ± 5.7% of COD removal. These results show that dark fermentation coupled with microbial electrolysis is a highly promising option to maximize the conversion of agro-industrial wastewaters and by-products into bio-hydrogen.

Abstract: International audience; Solid State Anaerobic Digestion (SSAD) of fungal pretreated wheat straw was evaluated in a leach bed reactor. During a first experiment, the effect of Substrate/Inoculum (S/I) ratios on the start-up phase was investigated. High S/I increased methane productivity but also raised the risk of reactor failure due to Volatile Fatty Acid (VFA) accumulation. With S/I ratios between 1.2 and 3.6 (Volatile Solid (VS) basis), the SSAD start-up using wheat straw was successful. Moreover, reactors were able to recover from acidification when the Total VFA/alkalinity ratio was lower than 2 gHAc_eq/gCaCO(3), with VFA concentrations lower than 10 and a pH close to 5.5. The conventional threshold of 0.6 gHAc_eq/gCaCO(3) for stable wet AD is therefore not adapted to SSAD. During a second experiment, after the wheat straw was submitted to a fungal pretreatment in a non-sterile pilot-scale reactor, it was digested with an S/I ratio of 2.8-2.9. Under batch SSAD conditions, the biodegradability of pretreated wheat straw was slightly improved in comparison to the control (254 versus 215 NmL/g VS, respectively). Considering mass losses occurring during the pretreatment step, suboptimal pretreatment conditions caused a slightly lower methane production (161 versus 171 NmL/gTS(initial) after 60-days anaerobic digestion). Nevertheless, pretreatment improved the start-up phase with lower acidification relative to controls. It would be particularly beneficial to improve the methane production in reactors with short reaction times.

Abstract: The influence of hydraulic retention time (HRT) on the microbial communities was evaluated in an anaerobic sequencing batch reactor (AnSBR) using organic waste from a restaurant as the substrate. The relationship among Lactobacillus, Clostridium and Bacillus as key micro-organisms on hydrogen production from organic solid waste was studied. The effect of the HRT (8–48 h) on the hydrogen production and the microbial community was evaluated. Quantitative PCR was applied to determine the abundance of bacteria (in particular, Enterobacter, Clostridium and Lactobacillus genera). An AnSBR fermentative reactor was operated for 111 cycles, with carbohydrate and organic matter removal efficiencies of 80 ± 15·42% and 22·1 ± 4·49% respectively. The highest percentage of hydrogen in the biogas (23·2 ± 11·1 %), and the specific production rate (0·42 ± 0·16 mmol H2 gVSadded−1 d−1) were obtained at an HRT of 48 h. The decrease in the HRT generated an increase in the hydrogen production rate but decreasing the content of the hydrogen in the gas. HRT significantly influence the abundance of Enterobacter, Clostridium and Lactobacillus during the hydrogen production from food waste leading the hydrogen production as well as the metabolic pathways. The microbial analysis revealed a direct relationship between the HRT and the presence of fermentative bacteria (Enterobacter, Clostridium and Lactobacillus genera). Clostridium sp. predominated at an HRT of 48 h, while Enterobacter and Lactobacillus predominated at HRTs between 8 and 24 h. Significance and Impact of the Study Significance and Impact of the Study: It was demonstrated that hydrogen production using food waste was influenced by the hydraulic retention time (HRT), and closely related to changes in microbial communities together with differences in metabolic patterns (e.g. volatile fatty acids, lactate, etc.). The decrease in the HRT led to the dominance of lactic acid bacteria within the microbial community whereas the increase in HRT favoured the emergence of Clostridium bacteria and the increase in acetic and butyric acids. Statistical data analysis revealed a direct relationship existing between the HRT and the microbial community composition in fermentative bacteria. This study provides new insight into the relationship between the bioprocess operation and the microbial community to understand better and control the biohydrogen production.

Abstract: Mite infestation is a big threat for rose greenhouses. It is much easier to halt and destroy them if their future development can be predicted. Taking into account temperature, humidity, cropping practices, plant vigour and some other influent parameters, an expert is able to predict the future development of the mites. Unfortunately, not all greenhouses can call on an expert permanently to help them in their fight against mites. To help them we have developed a novel model to assess and anticipate mite invasions in greenhouses in the short term. The model, based on a Bayesian network, takes into account the environment and the parameters defining invasion status with their interactions Data have been collected using knowledge from horticultural experts. The model has been validated in real situations emanating from the field. We obtained a good correlation between forecasts and expert predictions for the 18 cases used in this study. Thus, using this model should help the growers to protect against mite outbreaks. It constitutes a framework for studies of other harmful pest invasions.

Abstract: With its high biomass yield potential and its adaptability to a large spectrum of crop management schemes (dedicated, double cropping), sorghum is a relevant candidate crop for anaerobic digestion. Moreover, the large genotypic variability of its biochemical composition offers opportunities to cultivate specific varieties that fit the expectations of different end-users. Within this context, the need to evaluate the variability of biomethane potential (BMP) among different genotypes cultivated at various geographical sites has become crucial. In this study, four sorghum genotypes were grown at three different sites and harvested at the same maturity stage (dough grain stage). Consistent BMP obtained from different assays enabled genotype comparisons. The methane potentials observed between genotypes and production sites ranged between 200 ± 5 NmLCH4/gTS and 259 ± 12 NmLCH4/gTS. Evaluation of the genotypic and cultivation site effects produced highly significant results, thus accounting for 36 and 34%, respectively, of the phenotypic variability.

Abstract: International audience; Volatile fatty acids (VFA), inorganic carbon (IC) and total ammonia nitrogen (TAN) are key variables in the current context of anaerobic digestion (AD). Accurate measurements like gas chromatography and infrared spectrometry have been developed to follow the concentration of these compounds but none of these methods are affordable for small AD units. Only titration methods answer the need for small plant monitoring. The existing methods accuracy was assessed in this study and reveals a lack of accuracy and robustness to control AD plants. To solve these issues, a new titrimetric device to estimate the VFA, IC and TAN concentrations with an improved accuracy was developed. This device named SNAC (System of titration for total ammonia Nitrogen, volatile fatty Acids and inorganic Carbon) has been developed combining the measurement of electrical conductivity and pH. SNAC were tested on 24 different plant samples in a range of 0–0.16 mol.L−1 TAN, 0.01–0.21 mol.L−1 IC and 0–0.04 mol.L−1 VFA. The standard error was about 0.012 mol.L−1 TAN, 0.015 mol.L−1 IC and 0.003 mol.L−1 VFA. The coefficient of determination R2 between the estimated and reference data was 0.95, 0.94 and 0.95 for TAN, IC and VFA respectively. Using the same data, current methods based on key pH points lead to standard error more than 14.5 times higher on VFA and more than 1.2 times higher on IC. These results show that SNAC is an accurate tool to improve the management of AD plant.

Abstract: International audience; Understanding the environmental profile of a product computed from the Life Cycle Assessment (LCA) framework is sometimes challenging due to the high number of environmental indicators involved. The objective here, in guiding interpretation of LCA results, is to highlight the importance of each impact category for each product alternative studied. For a given product, the proposed methodology identifies the impact categories that are worth focusing on, relatively to a whole set of products from the same cumulated database. The approach extends the analysis of Representativeness Indices (RI) developed by Esnouf et al. (2018). It proposes a new operational tool for calculating RIs at the level of impact categories for a Life Cycle Inventory (LCI) result. Impact categories and LCI results are defined as vectors within a standardized vector space and a procedure is proposed to treat issues coming from the correlation of impact category vectors belonging to the same Life Cycle Impact Assessment (LCIA) method. From the cumulated ecoinvent database, LCI results of the Chinese and the German electricity mixes illustrate the method. Relevant impact categories of the EU-standardized ILCD method are then identified. RI results from all products of a cumulated LCI database were therefore analysed to assess the main tendencies of the impact categories of the ILCD method. This operational approach can then significantly contribute to the interpretation of the LCA results by pointing to the specificities of the inventories analysed and for identifying the main representative impact categories.

Abstract: International audience; The design and management of anaerobic digestion of sewage sludge (SS) require a relevant characterisation of the sludge organic matter (OM). Methods currently used are time-consuming and often insufficiently informative. A new method combining chemical sequential extractions (CSE) with 3D fluorescence spectroscopy was developed to provide a relevant SS characterisation to assess both OM bioaccessibility and complexity which govern SS biodegradability. CSE fractionates the sludge OM into 5 compartments of decreasing accessibility. First applied on three SS samples with different OM stability, fractionation profiles obtained were in accordance with the latter. 3D fluorescence spectroscopy revealed that the bioaccessible compartments were mainly constituted of simple and easily biodegradable OM while the unaccessible ones were largely made of complex and refractory OM. Then, primary, secondary and anaerobically digested sludge with different biodegradabilities were tested. Complexity revealed by 3D fluorescence spectroscopy was linked with biodegradability and chemical accessibility was correlated with sludge bioaccessibility.

Abstract: Optimization of feeding strategy is an essential issue of anaerobic co-digestion that can be greatly assisted with simulation tools such as the Anaerobic Digestion Model 1. Using this model, a set of parameters, such as the biochemical composition of the waste to be digested, its methane production yield and kinetics, has to be defined for each new substrate. In the recent years, near infrared analyses have been reported as a fast and accurate solution for the estimation of methane production yield and biochemical composition. However, the estimation of methane production kinetics requires time-consuming analysis. Here, a partial least square regression model was developed for a fast and efficient estimation of methane production kinetics using near infrared spectroscopy on 275 bio-waste samples. The development of this characterization reduces the time of analysis from 30 days to a matter of minutes. Then, biochemical composition and methane production yield and kinetics predicted by near infrared spectroscopy were implemented in a modified Anaerobic Digestion Model n°1 in order to simulate the performance of anaerobic digestion processes. This approach was validated using different data sets and was demonstrated to provide a powerful predictive tool for advanced control of anaerobic digestion plants and feeding strategy optimization.

Abstract: International Symposium on Effect-Related Evaluation of Anthropogenic Trace Substances - Concepts for Genotoxicity Neurotoxicity and Endocrine Effects, Aachen, october 2015; Understanding the fate and ecotoxicological effects of pesticides largely depends on their molecular properties. We recentlydeveloped BTyPol^ (Typology of Pollutants), a classification method of organic compounds based on statistical analyses. Itcombines several environmental (sorption coefficient, degradation half-life) and one ecotoxicological (bioconcentration factor)parameters, to structural molecular descriptors (number of atoms in the molecule, molecular surface, dipole moment, energy oforbitals, etc.). The present study attempts to extend TyPol to the ecotoxicological effects of pesticides on non-target organisms,based on data analysis from available literature and databases. It revealed that relevant ecotoxicological endpoints for terrestrialorganisms (e.g., soil microorganisms, invertebrates) that support a range of ecosystemic services are lacking as compared toaquatic organisms. The availability of ecotoxicological parameters was also lower for chronic than for acute ecotoxicity endpoints.Consequently, seven parameters were included for acute (EC50, LC50) and chronic (NOEC) ecotoxicological effects forone terrestrial (Eisenia sp.) and three aquatic (Daphnia sp., algae, Lemna sp.) organisms. In this new configuration, we usedTyPol to classify 50 pesticides into different clusters that gather molecules with similar environmental behaviors and ecotoxicologicaleffects. The classification results evidenced relationships between molecular descriptors, environmental parameters, andthe added ecotoxicological endpoints. This proof-of-concept study also showed that TyPol in silico classification can successfullyaddress new scientific questions and be expanded with other parameters of interest.

Abstract: The main objective of this work was to develop a controller for biogas production in continuous anaerobic fixed-bed reactors, which used effluent total volatile fatty acids (VFA) concentration as control input in order to prevent process acidification at closed loop. To this aim, a fuzzy-logic-based control system was developed, tuned and validated in an anaerobic fixed-bed reactor at pilot scale that treated industrial winery wastewater. The proposed controller varied the flow rate of wastewater entering the system as a function of the gaseous outflow rate of methane and VFA concentration. Simulation results show that the proposed controller is capable to achieve great process stability even when operating at high VFA concentrations. Pilot results showed the potential of this control approach to maintain the process working properly under similar conditions to the ones expected at full-scale plants.

Abstract: EA BIOmE SUPDAT INRA; International audience; Chlordecone (C10Cl10O; CAS number 143-50-0) has been used extensively as an organochlorine insecticide but is nowadays banned and listed on annex A in The Stockholm Convention on Persistent Organic Pollutants (POPs). Although experimental evidences of biodegradation of this compound are scarce, several dechlorination products have been proposed by Dolfing et al. (2012) using Gibbs free energy calculations to explore different potential transformation routes. We here present the results of an in silico classification (TyPol - Typology of Pollutants) of chlordecone transformation products (TPs) based on statistical analyses combining several environmental endpoints and structural molecular descriptors. Starting from the list of putative chlordecone TPs and considering available data on degradation routes of other organochlorine compounds, we used different clustering strategies to explore the potential environmental behaviour of putative chlordecone TPs from the knowledge on their molecular descriptors. The method offers the possibility to focus on TPs present in different classes and to infer their environmental fate. Thus, we have deduced some hypothetical trends for the environmental behaviour of TPs of chlordecone assuming that TPs, which were clustered away from chlordecone, would have different environmental fate and ecotoxicological impact compared to chlordecone. Our findings suggest that mono- and di-hydrochlordecone, which are TPs of chlordecone often found in contaminated soils, may have similar environmental behaviour in terms of persistence.

Abstract: The development of anaerobic digestion involves both co-digestion of solid wastes and optimization of the feeding recipe. Within this context, substrate characterisation is an essential issue. Although it is widely used, the biochemical methane potential is not sufficient to optimize the operation of anaerobic digestion plants. Indeed the biochemical composition in carbohydrates, lipids, proteins and the chemical oxygen demand of the inputs are key parameters for the optimisation of process performances. Here we used near infrared spectroscopy as a robust and less-time consuming tool to predict the solid waste content in carbohydrates, lipids and nitrogen, and the chemical oxygen demand. We built a Partial Least Square regression model with 295 samples and validated it with an independent set of 46 samples across a wide range of solid wastes found in anaerobic digestion units. The standard errors of cross-validation were 90 mgO2⋅gTS−1 carbohydrates, 2.5 ∗ 10−2 g⋅gTS−1 lipids, 7.2 ∗ 10−3 g⋅gTS−1 nitrogen and 99 mgO2⋅gTS−1 chemical oxygen demand. The standard errors of prediction were 53 mgO2⋅gTS−1 carbohydrates, 3.2 ∗ 10−2 g⋅gTS−1 lipids, 8.6 ∗ 10−3 g⋅gTS−1 nitrogen and 83 mgO2⋅gTS−1 chemical oxygen demand. These results show that near infrared spectroscopy is a new fast and cost-efficient way to characterize solid wastes content and improve their anaerobic digestion monitoring.

Abstract: International audience; Following legislation, the assessment of the environmental risks of 30 000 to 100 000 chemical substances is required for their registration dossiers. However, their behavior in the environment and their transfer to environmental components such as water or atmosphere are studied for only a very small proportion of the chemical in laboratory tests or monitoring studies because it is time-consuming and/or cost prohibitive. Therefore, the objective of this work was to develop a new methodology, TyPol, to classify organic compounds, and their degradation products, according to both their behavior in the environment and their molecular properties. The strategy relies on partial least squares analysis and hierarchical clustering. The calculation of molecular descriptors is based on an in silico approach, and the environmental endpoints (i.e. environmental parameters) are extracted from several available databases and literature. The classification of 215 organic compounds inputted in TyPol for this proof-of-concept study showed that the combination of some specific molecular descriptors could be related to a particular behavior in the environment. TyPol also provided an analysis of similarities (or dissimilarities) between organic compounds and their degradation products. Among the 24 degradation products that were inputted, 58% were found in the same cluster as their parents. The robustness of the method was tested and shown to be good. TyPol could help to predict the environmental behavior of a "new" compound (parent compound or degradation product) from its affiliation to one cluster, but also to select representative substances from a large data set in order to answer some specific questions regarding their behavior in the environment.

Abstract: International audience; Solid organic waste represents an abundant, cheap, and available source of biodegradable substrates not yet exploited to produce biohydrogen by dark fermentation. The impact of the composition of solid organic waste on microbial metabolic pathways and subsequently on biohydrogen production, has not been clearly elucidated. The aim of this study is to determine the compositional features of different substrates that influence bioH2 production. For this, we measured Biological hydrogen potentials (BHP) on 26 different substrates and performed a multivariate statistical analysis of the experimental data using a partial least square regression. The results showed that the BHP values correlated well with the initial carbohydrate content measured after mild hydrolysis. A predictive model explaining more than 89% of the experimental variability was then built to predict the maximal biohydrogen yield with a high accuracy and for a large spectrum of organic waste. An explicative model showed that only carbohydrates, butyrate and lactate concentrations were significant variables explaining more than 98% of biohydrogen yield variability. Interestingly, an interaction term between carbohydrates and lactate concentrations was required to explain microbial pathways producing hydrogen.

Abstract: The aim of this work was to analyse the applicability of electrical conductivity sensors for on-line monitoring the start-up period of an anaerobic fixed-bed reactor. The evolution of bicarbonate concentration and methane production rate was analysed. Strong linear relationships between electrical conductivity and both bicarbonate concentration and methane production rate were observed. On-line estimations of the studied parameters were carried out in a new start-up period by applying simple linear regression models, which resulted in a good concordance between both observed and predicted values. Electrical conductivity sensors were therefore identified as an interesting method for monitoring the start-up period of anaerobic fixed-bed reactors due to its reliability, robustness, easy operation, low cost, and minimum maintenance compared with the currently used sensors.

Abstract: A hybrid (continuous–discrete) cascade control is proposed to regulate both, volatile fatty acids (VFA) and chemical oxygen demand (COD) concentrations in two-stage (acidogenic-methanogenic) anaerobic digestion (TSAD) processes. The outer loop is a discrete controller that regulates the COD concentration of the methanogenic bioreactor by using a daily off-line measurement and that modifies the set-point tracked by inner loop, which manipulates the dilution rate to regulate the VFA concentration of the acidogenic bioreactor, estimated by continuous on-line conductivity measurements, avoiding acidification. The experimental validation was conducted in a TSAD process for the treatment of tequila vinasses during 110 days. Results showed that the proposed cascade control scheme was able to achieve the VFA and COD regulation by using conventional measurements under different set-point values in spite of adverse common scenarios in full-scale anaerobic digestion processes. Microbial composition analysis showed that the controller also favors the abundance and diversity toward methane production.

Abstract: Received 1 September 2015, Revised 20 November 2015, Accepted 23 November 2015, Available online 7 January 2016; Fixed-biomass reactors present the main advantage of increasing the hydrogen production rates by supporting high organic loading rates. However, the use of complex natural inoculum often precludes a long-term operation through the rapid emergence of methanogens. In this study, an Anaerobic Sequencing Batch Biofilm Reactor (ASBBR) was inoculated with a simplified H2-producing culture to evaluate its influence on robustness and more particularly on methanogenic activity. An ASBBR was operated over 6 months with unsterile lactose-rich effluent and under Hydraulic Retention Time ranging from 1 to 34 h. Hydrogen performances in terms of productivity and yields ranged from 0.23 to 519 mmoleH2 L−1 d−1 and 0.01 to 7.11 moleH2 molelactose−1, respectively. No significant methane production was observed all along the experimental procedure, showing that inoculating with a simplified and highly enriched preculture could increase substantially the robustness of the process. Specific preparation of the inoculum may represent a solution to sustain long-term operation of biofilm-based reactors.

Abstract: International audience; In an integrated biorefinery concept, biological hydrogen and methane production from lignocellulosic substrates appears to be one of the most promising alternatives to produce energy from renewable sources. However, lignocellulosic substrates present compositional and structural features that can limit their conversion into biohydrogen and methane. In this study, biohydrogen and methane potentials of 20 lignocellulosic residues were evaluated. Compositional (lignin, cellulose, hemicelluloses, total uronic acids, proteins, and soluble sugars) as well as structural features (crystallinity) were determined for each substrate. Two predictive partial least square (PLS) models were built to determine which compositional and structural parameters affected biohydrogen or methane production from lignocellulosic substrates, among proteins, total uronic acids, soluble sugars, crystalline cellulose, amorphous holocelluloses, and lignin. Only soluble sugars had a significant positive effect on biohydrogen production. Besides, methane potentials correlated negatively to the lignin contents and; to a lower extent, crystalline cellulose showed also a negative impact, whereas soluble sugars, proteins, and amorphous hemicelluloses showed a positive impact. These findings will help to develop further pretreatment strategies for enhancing both biohydrogen and methane production.

Abstract: A Capillary Electrophoresis Single Strand Conformation Polymorphism (CE-SSCP) method based on functional [Fe–Fe]-hydrogenase genes was developed for monitoring the hydrogen (H 2 )-producing clostridial population in mixed-culture bioprocesses. New non-degenerated primers were designed and then validated on their specific PCR detection of a broad range of clostridial hydA genes. The hydA -based CE-SSCP method gave a specific and discriminating profile for each of the Clostridium strains tested. This method was validated using H 2 -producing mixed cultures incubated at temperatures ranging from 25 °C to 45 °C. The hydA CE-SSCP profiles clearly differed between temperatures tested. Hence, they varied according to variations of the H 2 production performances. The HydA sequences amplified with the new primer set indicated that diverse Clostridium strains impacted the H 2 production yields. The highest performances were related to the dominance of Clostridium sporogenes -like hydA sequences. This CE-SSCP tool offers highly reliable and throughput analysis of the functional diversity and structure of the hydA genes for better understanding of the H 2 -producing clostridial population dynamics in H 2 dark fermentation bioreactors.
