ABSTRACT: The aim of this study was to investigate the influence of the addition of prebiotics rice bran, inulin and hi-maize, on the survival of Lactobacillus acidophilus in alginate microparticles obtained by external ionic gelation followed by freeze-drying. The microparticles size ranged from 127.5μm to 234.6μm. Microparticles added from the different prebiotics demonstrated an increase in the protection of the microorganism, which presented greater viability against the gastrointestinal simulation. As for storage under different conditions, rice bran treatment at 25ºC kept probiotics viable for 30 days. Under storage conditions -18°C and 7°C, treatments containing prebiotics hi-maize and rice bran maintained viable probiotic microorganisms for a period of 60 days.

ABSTRACT: Technique of complex coacervation was used to produce microcapsules of Lactobacillus acidophilus La-5 encapsulated in gelatin and gum arabic which were then freeze-drying. Microcapsules were characterized using scanning electron and optical microscopy, and resistance of probiotics was evaluated during release into a simulated gastrointestinal tract and storage at different temperatures. The complex coacervation process produced microcapsules with a high encapsulation efficiency (77.60% and 87.53%), ranging from 127.14-227.05 μm with uniform distribution. Microencapsulation was an efficient approach to achieve significant protection of probiotics against simulated gastrointestinal conditions compared with free cells. Encapsulation also improved the viability of probiotics during storage at either −18 ºC for 120 days, 7 ºC for 105 days or 25 ºC for 45 days. Therefore, complex coacervation was demonstrated to be adequate and promising for encapsulation of probiotics.

Microwave-assisted solid sampling analysis coupled to flame furnace atomic absorption spectrometry (MW-SS-FF-AAS) was used for Cd and Pb determination in food-contact polymer samples, with the aim of minimizing reagents and laboratory waste. Operational parameters, such as the FF tube design, the oxygen flow rate, the flame stoichiometry, the sample mass, among others, were evaluated and optimized. Calibration was performed using only reference solutions, and the limits of quantification were 1.7 and 4.6 µg g−1 for Cd and Pb, respectively. Accuracy was assessed by the analysis of certified reference materials (CRMs), and by comparison with the results obtained by inductively coupled plasma mass spectrometry after microwave-assisted wet digestion. The MW-SS-FF-AAS results for the CRMs showed no statistical difference with the certified values, and good agreement was observed with the results of the digestion method. The consumption of rice milk has increased, mainly by individuals intolerant to lactose or allergic to cow milk. However, rice milk contains As. In this sense, the concentration of As in rice milk should be controlled. In the present study it is proposed a methodology for determination of As(III), dimethylarsenic (DMA), monomethylarsenic (MMA) and As(V) species in rice milk using LC-ICP-MS. The main features of the methodology are fast analysis, easy and simple sample preparation, where the sample is 3-fold diluted in the mobile phase and then filtered. The four arsenic species investigated were detected in the analysed samples, being As(V) the main species. The limit of quantification of the method ranges from 0.25 to 0.43 μg L−1 As. The analyte recovery ranged from 81 to 116% for samples spiked to 1.00 μg L−1 or 5.00 μg L−1 As and the relative standard deviation was better than 5%. Este estudo teve como objetivo avaliar a composição mineral de diferentes tipos de cascas de ovo, bem como a segurança microbiológica de amostras submetidas a diferentes métodos de higienização. Para a obtenção do pó de casca de ovo, as cascas foram lavadas, higienizadas, secas em estufa e trituradas em moinho. Cascas de ovo de granja (criação confinada), de coloração branca e vermelha, e cascas de ovo coloniais (caipira), provenientes da região central do Rio Grande do Sul, foram comparadas quanto a sua composição mineral. O Ca, mineral predominante na casca de ovo, se manteve em concentrações semelhantes nas diferentes amostras (cerca de 365mg g-1). As cascas de ovo de granja apresentaram maior concentração de Mg e menor concentração de Sr que as cascas de ovo coloniais. Não foram encontradas quantidades significativas de Fe, Cr, Mn, Mo, Ni, Se, Al, Cd e Pb nas amostras analisadas. Adicionalmente, tanto amostras higienizadas com imersão em hipoclorito e posterior fervura em água, quanto amostras nas quais a imersão em hipoclorito foi suprimida, não apresentaram contaminação por coliformes, estafilococos ou salmonela. Os resultados indicam que a casca de ovo pode ser utilizada na nutrição humana, já que é rica em Ca, não apresenta contaminação por metais tóxicos e, se processada de forma adequada, apresenta boa qualidade higiênico-sanitária

Muito tempo antes de ser conhecida a composição da Ilex paraguariensis (erva-mate), os indígenas utilizavam a mesma não somente pelo seu paladar, mas principalmente por conhecerem seus benefícios. Neste trabalho foi feita a determinação e avaliação dos valores de concentração de metais na primeira e segunda infusão em erva-mate comercializadas e industrializadas nos estados da região sul do Brasil. Para as infusões das amostras foi utilizada água destilada deionizada aquecida a 80o C. Para a determinação dos metais foi utilizada a espectrometria de massas com plasma indutivamente acoplado (ICP-MS). A concentração de cádmio, chumbo, cobalto, cromo, ferro e vanádio foram abaixo do limite de detecção (< LD = 0,3 μg.g-1). Na análise comparativa (ANOVA) da primeira infusão da erva-mate entre as diferentes regiões, verificou-se diferença significativa (p >0,05) para cobre e níquel. Quando aplicado o teste de Tukey, observou-se diferença signifi- cativa para cobre na primeira infusão da erva-mate oriunda do PR em relação à produzida no RS e SC. Para níquel, a diferença foi significativa entre as infusões obtidas de ervas-mates produzidas no RS e de SC. Para Zn foi observado a menor variação de concentração entre as regiões e as duas infusões. Considerando a concentração de elementos, nas regiões estudadas, estes estão presentes principalmente na primeira infusão diminuindo de forma significativa na segunda infusão e decrescendo mais nas demais. Desta forma o chimarrão, infusão da erva-mate, nos estados do sul do Brasil, encontra-se com baixa concentração de metais e, conseqüentemente, baixo risco a saúde para a população consumidora da tradição.

This study investigated the chemical composition of five different extracts of Aloysia triphylla and their activity against Aeromonas sp. The extracts were obtained from the dried leaves by pressurized CO2 extraction at 30, 50 and 70ºC, and 100, 150, and 200 bar, and analyzed by GC/FID and GC-MS. The antibacterial activity was assayed by the microdilution method. The tested microorganisms comprised seven Aeromonas isolates obtained from the kidney of infected silver catfish, Rhamdia quelen. The yield, chemical composition and antibacterial activity of the extracts were dependent on the extraction conditions. Mono and sesquiterpenoids were the major constituents of all the extracts and the highest extraction yield was obtained at 70ºC and 200 bar. A. triphylla presented moderate antibacterial activity against Aeromonas sp.

A utilização de lavoura e pastagens tem sido uma importante senão a principal alternativa de descarte do dejeto líquido de suínos. Porém, seu uso, muitas vezes, tem sido feito em quantidades superiores às exigências das culturas e com aplicações sequenciais que podem potencializar o efeito poluidor desse resíduo, já que o solo tem uma capacidade limitada de suporte. O presente estudo teve por objetivo determinar os teores totais de zinco (Zn), cobre (Cu), cromo (Cr), níquel (Ni), manganês (Mn) e cádmio (Cd) em áreas que são usadas como local de descarte de dejeto líquido de suínos. Para isso, foram coletadas amostras de solo em 13 propriedades rurais da região Oeste de Santa Catarina, usadas anualmente como local de descarte, e que possuíam um histórico de aplicação desse resíduo de 4 a 22 anos. Além disso, foi também realizada amostragem de solo em um experimento no município de Paraíso do Sul, RS, sob pastagem natural, que, durante um período de cinco anos, recebeu 28 aplicações de dejeto líquido de suínos. Zinco e cobre são os elementos que mais devem merecer a atenção com relação a um possível acúmulo no solo, em áreas com aplicação de dejeto líquido de suínos.The use of crop and pasture has been an important if not the main alternative of pig slurry disposal. However, its use has often been made in quantities exceeding the requirements of crops and with sequential applications that can enhance the polluting effect of this waste once the soil has a limited capacity of support. The present study aimed to determine the total content of zinc (Zn), copper (Cu), chromium (Cr), nickel (Ni), manganese (Mn) and cadmium (Cd) in areas that are used as a disposal site of pig slurry. For this, there were collected soil samples in 13 farms in the West Region of Santa Catarina used annually as a disposal site, and which had a history of application of the residue from 4 to 22 years. In addition, it was also conducted soil sampling in an experiment in the municipality of Paraíso do Sul, RS under natural pasture, that during five years received 28 applications of pig slurry. Zinc and copper are the elements that deserve more attention in relation to a possible accumulation in the soil, in areas with application of pig slurry.

Neste trabalho 20 amostras comerciais sob a forma de comprimidos e 15sintéticas de cloridrato de metformina foram utilizadas e seus espectros adquiridos na faixa de 4000–650 cm-1 por espectroscopia difusa no infravermelho com transformada de Fourier (DRIFTS). Foram construídos e comparados modelos de calibração multivariada utilizando toda a região espectral analisada (modelo global PLS) e modelos otimizados empregando sub-regiões dos espectros FT-IR (modelos iPLS e siPLS). Os melhores resultados foram aqueles obtidos utilizando o algoritmo siPLS, dividindo o espectro em 40 intervalos e combinando 3 intervalos (37, 39 e 40), 4 variáveis latentes, e apresentou um valor de RMSEP inferior ao obtido utilizando o modelo global e o algoritmo iPLS. Assim, a aplicação das técnicas de seleção das regiões dos espectros de infravermelho (iPLS e siPLS) mostraram-se eficientes no desenvolvimento de metodologias mais simples, rápidas e não destrutivas para a análise das formulações farmacêuticas contendo metformida na forma de comprimidos, destacando-se a potencialidade destas técnicas no controle e fiscalização de medicamentos industrializados.In this work 20 metformin hydrochloride commercial pill samples and 14 synthetic samples were used and their spectra acquired in the 4000 – 650 cm-1 range by Diffuse Reflectance Infrared Fourier Transform Spectroscopy (DRIFTS). We analyzed data from historical controls treated with meglumine antimoniate to compare the frequency of adverse events observed in patients with cutaneous leishmaniasis treated with the same dose of meglumine antimoniate contaminated with heavy metals in an endemic area of the State of Bahia, Brazil. Group A patients were treated in 2000 with the drug produced by Eurofarma Laboratórios Ltda., São Paulo, Brazil (lot A) and group B patients were treated in 1996 with the reference drug produced by Rhodia Farma Ltda., São Paulo, Brazil (lot B). We observed an unusual higher frequency of skin reactions in group A patients. However, all type of adverse events observed in group A were also observed in group B. The physico-chemical analysis of these lots revealed that lot A had lower pH and higher concentration of total and trivalent antimony, lead, cadmium, and arsenic. Our findings suggest that the skin reactions could be attributed to heavy metal contamination of lot A.

A erva-mate (Ilex paraguariensis, St. Hill.) ingerida como infusão a quente, chamada de “chimarrão”, é uma cultura com grande valor comercial para os estados do sul do Brasil. Atualmente muitos estudos tem sido desenvolvidos buscando estabelecer os benefícios terapêuticos e efeitos colaterais à saúde humana da erva-mate, que podem estar relacionados à presença de metais. Neste trabalho foram determinadas as concentrações de metais essenciais e tóxicos na composição da erva-mate comercializada e consumida nos diferentes estados do sul do Brasil. Amostras provenientes do Rio Grande do Sul, Paraná e Santa Catarina foram analisadas para a determinação da concentração de Al, Ca, Cd, Co, Cr, Cu, Fe, K, Mg, Na, Ni, Pb, Sr, V, Zn por espectrometria de massa com plasma indutivamente acoplado (ICP-MS) e espectrometria de emissão óptica com plasma indutivamente acoplado (ICP-OES). Observou-se que a concentração média de todos os metais analisados nas amostras estava dentro dos limites toleráveis. Entretanto, foram observadas concentrações minerais diferenciadas entre as regiões estudadas. Desta forma, a erva-mate pode ser recomendada na utilização diária, podendo suprir necessidades básicas de ingestão desses metais essenciais sem o risco de problemas causados por elementos tóxicos.

15. Optimization of multivariate regression methods for quantification of sulphamethoxazole and trimethoprim in pharmaceutical formulations

A partial least-squares calibration (PLS) procedure in combination with infrared spectroscopy has been developed for simultaneous determination of sulphamethoxazole (SMZ) and trimethoprim (TMP) in powder raw material mixtures used for manufacturing commercial pharmaceutical products. Multivariate calibration modeling procedures using interval partial least squares (iPLS) were applied to select a spectral range that provided the lowest prediction error in comparison to the full-spectrum model. The experimental matrix was constructed using 52 synthetic samples and 13 commercial samples. The considered concentration ranges were 400 to 900 mg g-1 SMZ and 80 to 240 mg g-1 TMP. Spectral data were recorded between 650 and 4000 cm-1 with a 4 cm-1 resolution by Fourier transform infrared spectroscopy (FTIR) coupled with attenuated total reflection (ATR) accessory. The proposed procedure was compared with conventional procedure by high performance liquid chromatography (HPLC) using commercial samples containing SMZ and TMP. Results shown that PLS regression model combined to ATR-FTIR is a relatively simple, rapid and accurate procedure that could be applied to the simultaneous determination of SMZ and TMP in routine quality control of powder mixtures. A root mean square error of prediction (RMSEP) of 26.97 mg g-1 for SMZ and 12.23 mg g-1 for TMP was obtained after selection of better interval by iPLS.
