Objective: Smoking suppresses PD-1 expression in patients with rheumatoid arthritis (RA). In this study, we assess if smoking changed the epigenetic control over CD8+ T cell memory formation through a microRNA (miR) dependent mechanism. Methods: Phenotypes of CD8+ T cells from smokers and non-smokers, RA and healthy, were analyzed by flow cytometry. A microarray analysis was used to screen for differences in miR expression. Sorted CD8+ cells were in vitro stimulated with nicotine and analyzed for transcription of miRs and genes related to memory programming by qPCR. Results: CD27+CD107a−CD8+ T cells, defining a naïve-memory population, had low expression of PD-1. Additionally, the CD27+ population was more frequent in smokers (p = 0.0089). Smokers were recognized by differential expression of eight miRs. Let-7c-5p, let-7d-5p and let-7e-5p, miR-92a-3p, miR-150-5p, and miR-181-5p were up regulated, while miR-3196 and miR-4723-5p were down regulated. These miRs were predicted to target proteins within the FOXO-signaling pathway involved in CD8+ memory programming. Furthermore, miR-92a-3p was differentially expressed in CD8+ cells with naïve-memory predominance. Nicotine exposure of CD8+ cells induced the expression of miR-150-5p and miR-181a-5p in the naïve-memory cells in vitro. Additionally, nicotine exposure inverted the ratio between mRNAs of proteins in the FOXO pathway and their targeting miRs. Conclusions: Smokers have a high prevalence of CD8+ T cells with a naïve-memory phenotype. These cells express a miR profile that interacts with the memory programming conducted through the FOXO pathway.

Syftet med föreliggande studie är att undersöka vilka möjligheter och utmaningar några lärare i årskurs 1 anser att digitala verktyg medför i svenskundervisingen. Studien utgörs av en kvalitativ ansats i form av intervjuer. Studien visar på vikten av att arbeta med digitala verktyg i undervisningen då det gynnar elevers utveckling och lärande. Resultatet visar att digitala verktyg tillämpas i svenskundervisningen utifrån syftet att eleverna ges möjlighet till ett lustfyllt och varierande lärande som i sin tur leder till motivation hos dem till att vilja lära. I vilken omfattning lärare använder digitala verktyg varierar dock. En bidragande orsak till detta beskrivs vara beroende på vilken digital kompetens läraren innehar. Slutsatsen är att de tillfrågade lärarna är medvetna om digitala verktygs positiva påverkan för elevernas kunskapsutveckling. Trots detta anser de inte att det tillämpas i tillräcklig omfattning. Vi kan genom denna studie konstatera att följden av att många lärare inte innehar en tillräcklig digital kompetens leder till att deras användande av digitala verktyg i undervisningen begränsas. Detta leder vidare till att deras elever inte ges möjlighet att få utveckla sina digitala kunskaper. Detta tyder på att det finns ett behov av utvecklingsområden inom skolans verksamhet gällande lärares digitala kompetensutveckling.

Syftet med föreliggande studie är att undersöka hur och varför sex verksamma lärare i årskurs 2 tillämpar högläsning i undervisningen. Studien utgörs av en kvalitativ ansats och tolkas ur ett sociokulturellt perspektiv. Resultatet visar på vikten av att arbeta med högläsning i undervisningen då det gynnar språkutvecklingen. Resultatet visar även att högläsning tillämpas i undervisningen för att eleverna ska ges möjlighet till avkoppling. Hur de tillfrågade lärarna väljer att arbeta med en färdigläst bok varierar men det vanligaste förekommande är att eleverna får i uppgift att sammanfatta boken i form av samtal eller en skrivuppgift. Det framkommer dock att efterarbete med en färdigläst bok inte alltid sker, främst på grund av tidsbrist. Slutsatsen är att de tillfrågade lärarna är medvetna om högläsningens positiva påverkan för elevernas kunskapsutveckling. Trots detta upplevs högläsning inte tillämpas i tillräcklig omfattning. Vi kan vidare genom denna studie konstatera att följden av att högläsning vanligen inte är schemalagt är att det ofta bortprioriteras i undervisningen.

Background Smoking is a risk factor for developing rheumatoid arthritis (RA), but the mechanism remains uncertain. We previously demonstrated that smoking lowers the T cell activation threshold by limiting programmed death protein 1 (PD-1) expression. Aim To investigate how smoking influence the levels of soluble PD-1 ligand (sPD-L1). Method Serum levels of sPD-L1 were measured in 246 RA patients and in 168 healthy subjects. The analysis was done with respect to inflammation, smoking, treatments, and autoantibody status. The effect of therapeutic TNF-inhibiting antibodies (TNFi) on sPD-L1 was studied in 16 RA patients at their first infliximab infusion. The expression of Fcγ-receptor (FcγR) subclass IIB and IIIA was analyzed with quantitative polymerase chain reaction in peripheral blood mononuclear cells (PBMCs) from 12 RA patients and 15 healthy controls, and in healthy PBMC exposed to IgG containing antibodies to cyclic citrullinated peptides (aCCP). Results The negative association between smoking and sPD-L1 in RA patients was established by multiple logistic regression (OR = 0.52, p = 0.038). Other covariates in the regression model were serum levels of IL-1β representing inflammation (OR = 1.6, p = 0.0076) and aCCP positivity (OR = 1.9, p = 0.047). First infliximab infusion repressed sPD-L1 (p = 0.023) in patients, and low levels of sPD-L1 were found in patients with early RA treated with TNFi (p = 0.018). Treatment with TNFi was associated with higher sPD-L1 in patients with long disease duration (p = 0.041) and restored levels in smokers. In vitro exposure to aCCP+ IgG suppressed sPD-L1 (p = 0.036), but aCCP+ patients with long disease duration had higher sPD-L1 (p = 0.016). High ratio of the inhibitory FcγR subclass IIB over the stimulatory IIIA resulted in low sPD-L1 release (p = 0.029). Smoking was associated with a higher FcγR IIB/IIIA ratio (p = 0.00062) and lower levels of sPD-L1 (p = 0.013). Conclusion In RA, serum sPD-L1 was related to systemic inflammation and aCCP positivity. Smoking altered the expression of FcγRs and limited sPD-L1 in RA patients, permitting inappropriate T cell responses. Differential regulation of sPD-L1 during the early and late RA may indicate transposition from acute to chronic inflammation.

Both value and flavor of wines can improve with time if stored properly. The factors with the most profound effect on the wine are light, humidity, temperature, vibrations, and ventilation. Light consisting of shorter wavelengths, under 450 nm, are dangerous for wine and can destroy the structure in wine. The humidity is supposed to be kept between 50-80 %, mainly to prevent the wine cork from drying out, since this would result in a shrinkage that will let in air to the bottle. When considering temperature, the ideal maturing temperature is 12-14 °C for both reds and whites. It is also essential to keep the wine at a stable temperature; it should not fluctuate more than 1.5 °C per day or 2-3 °C over a year. Larger amplitudes of vibrations cause an increased structural change in wine molecules. Therefore, they should be kept as small as possible. Ventilation is required since a reasonably small closed space with high humidity leads to a mold-prone environment which could be devastating to the wine. The wine market is shifting from bulk wine to becoming more wine enthusiast focus where it is believed the importance of a wine refrigerator will increase with this shift. The wine business is not scientifically originated. Several requirements are mainly “hearsay” and “common knowledge”. The aim of this paper was to identify scientifically founded facts to product develop wine refrigerators for a global market. Four different wine refrigerators called A, B, C, and D, from different brands, were tested in a laboratory from a wine requirement perspective with a focus on temperature, humidity, and electricity consumption. All tests were conducted for 24 hours. Both temperature and humidity were logged every 10 seconds and results were processed in MATLAB and presented in graphs. It was found that cabinets with separators held a more accurate temperature than those without one. It was also found that most of them held a temperature within the allowed fluctuation interval. Moreover, the air temperature fluctuation did not significantly affect the temperature inside the wine bottle indicating it is not important to further improve. The humidity was tested both with and without load but did not give a significant difference, which indicates the cabinets does not have a unique system to add humidity to the cabinet. Overall, all cabinets except one reached an average of at least 50 % relative humidity, which is the minimum requirement. The functionality of the humidity system of each unit is in question. Most surprisingly, the high electricity consumption for all of the wine fridges entails potential improvement area of the cooling system that uses more energy than what is thought to be needed. A comparison with a regular domestic refrigerator shows great potential for improvement, especially since a wine fridge only needs to perform a temperature of 12 °C, while a domestic fridge keeps a temperature of approximately 4 °C. The vast amount of energy use could also be because of inadequate insulation, with leakage through or around the glass door. A closer look at the gaskets showed small unsealed spots where the gasket was not tightly against the cabinet, enabling ambient air to leak in. Overall, the energy consumption results highlighted an inefficient cooling cycle. The found development opportunity is to include a variable-speed compressor into the refrigeration unit to maintain proper internal climate regarding temperature, humidity, and energy demand. Furthermore, the results of this thesis highlight the light blockage solutions which seem non-considered in the tested refrigerators and what was found in the market. Både värde och smak hos vin kan öka över tid om det lagras på ett korrekt sätt. De faktorer som har mest påverkan på vinet är ljus, luftfuktighet, temperatur, vibrationer och ventilation. Ljus som består av korta våglängder under 450 nm är farliga för vinet och kan förstöra dess struktur. Luftfuktigheten bör hållas mellan 50 - 80 %, detta framförallt då korken annars kan torka ut vilket då kan leda till att korken krymper och luft kommer in i vinflaskan. Gällande temperaturen, är det ideala intervallet vid åldring av vin är 12 - 14 °C för både vitt och rött vin. Det är även viktigt att hålla vinet på en stabil temperatur, det bör inte fluktuera mer än 1,5 °C per dygn eller 2 - 3 °C över ett år. Ett forskningsarbete om vibrationer påvisar att ökade vibrationsamplituder orsakar ökade strukturella förändringar i vinets molekyler, därför bör de vara så små som möjligt. Ventilation är nödvändigt eftersom ett litet slutet utrymme med hög luftfuktighet leder till en miljö med mögelbenägenhet, vilket kan vara förödande för vinet. Vinmarknaden skiftar från bulkviner till att bli mer vinentusiast-fokuserade i köp där vikten av vinkyl tros öka i detta skifte. Hela vinindustrin är inte tungt forskningsgrundad där mycket tillgänglig kunskap ”hörsägen” och ”allmän kunskap”. Målet med denna studie var därmed att identifiera vetenskapligt grundade fakta för att produktutveckla vinkylar på en global marknad. Fyra olika vinkylar från olika märken, kallade A, B, C och D, testades utifrån ett vinperspektiv i ett laboratorium med fokus på temperatur, luftfuktighet och energikonsumtion. Samtliga tester utfördes under en period på 24 timmar där temperatur och luftfuktighet loggade var 10e sekund där resultaten sedan behandlades i MATLAB och presenterades i grafer. Det kom fram att skåp med separatorer höll en mer exakt temperatur än dom utan. Majoriteten av skåpen höll temperaturen inom det tillåtna fluktuationsintervallet under ett dygn. Slutligen visade resultaten att temperaturfluktuationerna i luften inte påverkade temperaturen i vinet inuti en vinflaska marginellt varpå slutsatsen blev att detta inte skall vara ett fokusområde vid fortsatt utveckling. Alla vinkylar testades med och utan last och påvisade inte en signifikant skillnad vilket indikerar att respektive kabinett inte inkluderas av ett unikt luftfuktighetssystem som tillför fukt till skåpet. Alla kylar utom en uppnådde ett genomsnitt av minst 50 % relativ luftfuktighet som definierades till minimikravet. Funktionen av respektive fuktighetssystem är ifrågasättbar där lite eller noll information gås att få. Mest överraskande, resultaten indikerade en hög energikonsumtion för alla vinkylar vilket indikerar en förbättringspotential av kylsystemet då den förbrukar mer energi än vad som är nödvändigt. Vid en jämförelse med ett vanligt konsumentkylskåp vidare indikerar samma slutsats då den ofta använder mindre energi trots att den skall åskakomma temperaturer runt 4 °C i jämförelse med en vinkyl som skall hålla temperaturer runt 12 °C. Stora energikonsumtionen kan också härstamma från bristfällig isolering med läckage genom och runt glasdörren. Empiriskt syntes små hålrum genom isolerande packningen där luft kunde läcka igenom. Däremot tros den största energikonsumtionen vara ett resultat av en ineffektiv kylcykel. Funna utvecklingsmöjligheten är att inkludera en variabel-hastighetskompressor i kylcykeln för att upprätthålla lämpligt internt klimat med avseende på temperatur, luftfuktighet och energikonsumtion. Avhandlingen påvisar även att ljusblockeringslösningen verkar ogenomtänkt i respektive testad kabinett och vad som är funnet på marknaden.

There are few studies done on the work of social services on women exposed to violence. The work of social services with women exposed to violence has been a hot topic in the social policy debate. There have also been legislative changes to strengthen the work and help around women who are exposed to violent. The overall issues of the study focus on investigating the importance of social workers experiences in the work around the womens exposed to violence with the different types of policies, guidelines, action plan and regulations? We also wanted to explore how we can understand the importance of policies, guidelines, action plan and regulations and how it works when social workers use these in their work around women exposed to violence? The empirical study has mainly been gathered through qualitative semistructured interviews with professional social workers in the Swedish municipalities social services. We have interviewed six different social workers in four different municipalities all in the southern part of Sweden. Previous research has also been studied, national and international. The collected empiricism has been analyzed using the theory developed by Michael Lipsky, and we have also used Roine Johansson's interpretation of grassroots bureaucracy to gain a deeper understanding of our data material. We received data showing that the social worker decides on actions motivated by the policies, guidelines, action plans and regulations they relate to. We saw that the policies, guidelines, action plan and regulations that the social workers worked with were very different in their design, which meant that there were different structured structures within the organizations in the different municipalities, which resulted in differences in how to work. We also saw that the social worker had different experiences of how far and wide the use of these documents was, and that we could see the significance of social workers who experienced the documents in their work. The study depicts how social workers perceive that policies, guidelines, action plans and regulations affect their work around women exposed to violence, and how we can understand the importance of policies, guidelines, action plan and regulations and how it works when social workers use them in their work around women exposed to violence. The main conclusion in this study is that the social worker provides assistance, support and assistance to the woman as she fits into what the social worker's policies, guidelines, action plans and regulations describe. If the woman exposed to violence does not comply with the criteria such as policies, guidelines, action plans and regulations, then there are reasons for granting the victim of violence to receive support and assistance. The study also shows how clients are categorized to fit into the organization, they are interpreted renegotiated and adapted to fit the social services organization. The study also shows that some of the social workers use and follow the policies, guidelines, action plan and regulations more than others and it is also clear that they can be used in order to give more specialized efforts to the victim of violence. Det finns få studier kring socialtjänstens arbete med våldsutsatta kvinnor. Socialtjänstens arbete med våldsutsatta kvinnor har utgjort ett hett ämne i den socialpolitiska debatten. Det har även skett lagförändringar för att stärka arbetet och hjälpen kring de våldsutsatta kvinnorna. Studiens övergripande frågeställningar fokuserar på att undersöka vilken betydelse socialarbetaren upplever att olika typer av policy, riktlinjer, handlingsplan och föreskrifter har i sitt arbete kring de våldsutsatta kvinnorna. Vi ville även undersöka hur vi kan förstå betydelsen av policy, riktlinjer, handlingsplan och föreskrifter och hur det går till när socialarbetaren använder sig av dessa i sitt arbete kring våldsutsatta kvinnor. Studiens empiri har huvudsakligen samlats in genom kvalitativa, semistrukturerade intervjuer med yrkesverksamma och professionella socialarbetare inom Sveriges kommuners socialtjänster. Vi har intervjuat sex olika socialarbetare inom fyra olika kommuner alla inom området södra Sverige. Tidigare forskning har även studerats, nationell och internationell. Den insamlade empirin har analyserats med hjälp av teori som har utvecklats av Michael Lipsky och vi har även använt oss av Roine Johanssons tolkning av gräsrotsbyråkrati för att få en djupare förståelse för vårt datamaterial. Vi fick fram data som visar att socialarbetaren beslutar om insatser begrundat i den policy, riktlinjer, handlingsplaner och föreskrifter som finns att förhålla sig till. Vi såg att de policys, riktlinjer, handlingsplan och föreskrifter som socialarbetarna arbetade utefter var väldigt olika i sin utformning och det gjorde att det blev olika uppbyggda strukturer inom organisationerna i de olika kommunerna vilket resulterat i att det är skillnader i hur man arbetar. Vi kunde även se att socialarbetaren hade olika upplevelser av hur stor och bred användning dessa dokument har samt att vi kunde se vilken betydelse socialarbetarna upplevde att dokumenten hade i sitt arbete. Studien skildrar hur socialarbetare upplever att policy, riktlinjer, handlingsplaner och föreskrifter påverkar deras arbete kring våldsutsatta kvinnor. Studien skildrar även hur vi kan förstå betydelsen av policy, riktlinjer, handlingsplan och föreskrifter och hur det går till när socialarbetaren använder sig av dessa i arbetet kring våldsutsatta kvinnor. Den huvudsakliga slutsatsen i den här studien är att socialarbetaren ger insatser, stöd och hjälp till kvinnan när hon passar in i det som socialarbetarens policy, riktlinjer, handlingsplaner och föreskrifter beskriver. Om den våldsutsatta kvinnan inte stämmer överens med de kriterier som policy, riktlinjer, handlingsplaner och föreskrifter har så krävs det motiveringar för att bevilja att den våldsutsatta kvinnan ska få stöd och hjälp. Studien visar även hur klienterna kategoriseras för att passa in i organisationen, de tolkas omförhandlas och anpassas för att passa in i socialtjänstens organisation. Studien visar även att en del av socialarbetarna använder sig och följer de policy, riktlinjer, handlingsplan och föreskrifter mer än andra och det är även tydligt att de kan användas i syfte att ge den våldsutsatta kvinnan mer specialiserade insatser.
