BackgroundAnimal African Trypanosomosis (AAT) is a parasitic disease of livestock that has a major socio-economic impact in the affected areas. It is caused by several species of uniflagellate extracellular protists of the genus Trypanosoma mainly transmitted by tsetse flies: T. congolense, T. vivax and T. brucei brucei. In Burkina Faso, AAT hampers the proper economic development of the southwestern part of the country, which is yet the best watered area particularly conducive to agriculture and animal production. It was therefore important to investigate the extent of the infection in order to better control the disease. The objective of the present study was to assess the prevalence of trypanosome infections and collect data on the presence of tsetse flies.MethodsBuffy coat, Trypanosoma species-specific PCR, Indirect ELISA Trypanosoma sp and trypanolysis techniques were used on 1898 samples collected. An entomological survey was also carried out.ResultsThe parasitological prevalence of AAT was 1.1%, and all observed parasites were T. vivax. In contrast, the molecular prevalence was 23%, of which T. vivax was predominant (89%) followed by T. congolense (12.3%) and T. brucei s.l. (7.3%) with a sizable proportion as mixed infections (9.1%). T. brucei gambiense, responsible of sleeping sickness in humans, was not detected. The serological prevalence reached 49.7%. Once again T. vivax predominated (77.2%), but followed by T. brucei (14.7%) and T. congolense (8.1%). Seven samples, from six cattle and one pig, were found positive by trypanolysis. The density per trap of Glossina tachinoides and G. palpalis gambiensis was 1.2 flies.Conclusions/significanceOverall, our study showed a high prevalence of trypanosome infection in the area, pointing out an ongoing inadequacy of control measures.

Abstract Background Human African Trypanosomiasis (HAT) is an important neglected tropical disease caused by Trypanosoma spp. parasites transmitted by species of tsetse fly (Glossina spp). The most important vectors of HAT are riverine tsetse and these can be controlled by attracting them to stationary baits such as insecticide-impregnated traps or targets deployed along the banks of rivers. However, the geographical nature of some riverine habitats, particularly mangroves but also extensive lake and river networks, makes deployment of baits difficult and limits their efficacy. It is known that tsetse are attracted by the movement of their hosts. Our hypothesis was that mounting a target on canoes typically used in Africa (‘pirogues’) would produce an effective means of attracting-and-killing riverine tsetse in extensive wetland habitats. Methods In Folonzo, southern Burkina Faso, studies were made of the numbers of tsetse attracted to a target (75 × 50 cm) of blue cloth and netting mounted on a pirogue moving along a river, versus the same target placed on the riverbank. The targets were covered with a sticky film which caught tsetse as they contacted the target. Afin d’étudier la diversité spatio-temporelle des glossines vectrices de trypanosomoses en Côte d’Ivoire, des enquêtes ont été réalisées sur le fleuve Comoé dans trois sites localisés dans des zones écoclimatiques différentes : au nord, au centre et au sud. Elles ont eu lieu en saisons sèche et pluvieuse. Pour chaque site et saison, les glossines ont été capturées pendant cinq jours consécutifs à l’aide de pièges biconiques disposés suivant des transects perpendiculaires au fleuve Comoé. A Kafolo, en savane, trois espèces et sous-espèces ont été capturées indépendamment de la saison : Glossina palpalis gambiensis, G. tachinoides et G. medicorum. A Aboisso-Comoé et à Groumania, respectivement en zone de forêt et de transition forêt-savane, seule G. palpalis palpalis a été capturée. La densité apparente par piège, toutes espèces confondues, a été plus importante à Kafolo qu’à Groumania et à Aboisso-Comoé. Elle a été de 9,48 glossines (gl)/piège/jour (écart-type [ET] = 26,30) à Kafolo, de 0,79 gl/piège/ jour (ET = 2,65) à Groumania et de 0,18 gl/piège/jour (ET = 0,58) à Aboisso-Comoé en saison sèche. En saison pluvieuse, elle a été de 3,64 gl/piège/jour (ET = 9,76) à Kafolo et de 1,42 gl/piège/jour (ET = 2,35) à Groumania. Ces résultats montrent que la diversité des espèces de glossines le long du fleuve Comoé est fonction de la conservation du biotope, ainsi que l’illustre la disparition progressive de certaines espèces comme G. morsitans submorsitans qui n’a pas été capturée au cours de cette étude. Nous confirmons également que seul le vecteur majeur de la trypanosomiase humaine africaine, G. palpalis s.l., est capable de s’adapter à l’homme.

BACKGROUND:African animal trypanosomosis (AAT) is a major constraint to sustainable development of cattle farming in sub-Saharan Africa. The habitat of the tsetse fly vector is increasingly fragmented owing to demographic pressure and shifts in climate, which leads to heterogeneous risk of cyclical transmission both in space and time. In Burkina Faso and Ghana, the most important vectors are riverine species, namely Glossina palpalis gambiensis and G. tachinoides, which are more resilient to human-induced changes than the savannah and forest species. Although many authors studied the distribution of AAT risk both in space and time, spatio-temporal models allowing predictions of it are lacking. METHODOLOGY/PRINCIPAL FINDINGS:We used datasets generated by various projects, including two baseline surveys conducted in Burkina Faso and Ghana within PATTEC (Pan African Tsetse and Trypanosomosis Eradication Campaign) national initiatives. We computed the entomological inoculation rate (EIR) or tsetse challenge using a range of environmental data. The tsetse apparent density and their infection rate were separately estimated and subsequently combined to derive the EIR using a "one layer-one model" approach. The estimated EIR was then projected into suitable habitat. This risk index was finally validated against data on bovine trypanosomosis. It allowed a good prediction of the parasitological status (r2 = 67%), showed a positive correlation but less predictive power with serological status (r2 = 22%) aggregated at the village level but was not related to the illness status (r2 = 2%). CONCLUSIONS/SIGNIFICANCE:The presented spatio-temporal model provides a fine-scale picture of the dynamics of AAT risk in sub-humid areas of West Africa. The estimated EIR was high in the proximity of rivers during the dry season and more widespread during the rainy season. The present analysis is a first step in a broader framework for an efficient risk management of climate-sensitive vector-borne diseases.

BACKGROUND: Tsetse flies of the Palpalis group are the main vectors of sleeping sickness in Africa. Insecticide impregnated targets are one of the most effective tools for control. However, the cost of these devices still represents a constraint to their wider use. The objective was therefore to improve the cost effectiveness of currently used devices. METHODOLOGY/PRINCIPAL FINDINGS: Experiments were performed on three tsetse species, namely Glossina palpalis gambiensis and G. tachinoides in Burkina Faso and G. p. palpalis in Côte d'Ivoire. The 1 × 1 m(2) black blue black target commonly used in W. Africa was used as the standard, and effects of changes in target size, shape, and the use of netting instead of black cloth were measured. Regarding overall target shape, we observed that horizontal targets (i.e. wider than they were high) killed 1.6-5x more G. p. gambiensis and G. tachinoides than vertical ones (i.e. higher than they were wide) (P < 0.001). For the three tsetse species including G. p. palpalis, catches were highly correlated with the size of the target. However, beyond the size of 0.75 m, there was no increase in catches. Replacing the black cloth of the target by netting was the most cost efficient for all three species. CONCLUSION/SIGNIFICANCE: Reducing the size of the current 1*1 m black-blue-black target to horizontal designs of around 50 cm and replacing black cloth by netting will improve cost effectiveness six-fold for both G. p. gambiensis and G. tachinoides. Studying the visual responses of tsetse to different designs of target has allowed us to design more cost-effective devices for the effective control of sleeping sickness and animal trypanosomiasis in Africa.

1. Tsetse flies (Diptera: Glossinidae) are K‐strategist species and deposit a single larva at 10‐day intervals in specific sites. As larviposition site selection strongly impacts reproductive success, the selection of larviposition sites is unlikely to be random and will be subject to strong selective processes, probably mediated by specific cues. 2. This study was designed to assess the existence of an aggregation effect in the palpalis group and to test its potential chemical nature (contact or volatile compounds). The larviposition site selection of Glossina palpalis gambiensis was studied according to the presence of conspecific and heterospecific larvae (morsitans group) buried in substrates. Three sets of experiments were performed with either individual or grouped gravid females and with or without physical access to the substrate. 3. In individual larviposition experiments, females were more likely to select trays with buried larvae than unconditioned sand (63.2% and 36.8%, respectively; P < 0.05). In the grouped experiment with substrate contact, females were more likely to larviposit in trays with conspecific (40%) or heterospecific (33%) buried larvae than in unconditioned sand (22%) or empty trays (5%; P < 0.05). The results were similar without substrate contact, but more pupae were deposited in empty trays (19%). 4. These results provide the first evidence for larval aggregation in G. p. gambiensis and suggest that larviposition site selection is mediated by volatile semiochemicals of larval or pupal origin. However, this larval aggregation does not seem to be species specific and therefore offers new options for the behavioural manipulation of these vectors. [ABSTRACT FROM AUTHOR]
