- Abstract
Cancer is a complex disease that can also affect the younger population; however, it is responsible for a relatively high mortality rate of children and youth, especially in low- and middle-income countries (LMICs). Besides that, lipidomic studies in this age range are scarce. Therefore, we analyzed blood serum samples from young patients (12 to 35 years) with bone sarcoma (osteosarcoma) and compared their lipidomics to the ones from the control group of samples, named healthy control (HC group), using NMR and LC-MS techniques. Furthermore, differences in the lipidomic profiles between OS patients with and without metastasis indicate higher glycerophosphocholine (GPC) and glycerophospholipid (GPL) levels in osteosarcoma and increased cholesterol, choline, polyunsaturated fatty acids (PUFAs), and glycerols during the metastasis. These differences, detected in the peripheral blood, could be used as biomarkers for liquid biopsy.

This study aimed to identify barriers and facilitators to engaging with online interventions among 24 ethnoracially diverse college students self-identifying as being at risk for suicide. Thematic analysis was used to identify codes from focus groups. Participants shared mixed perceptions about online intervention use and effectiveness. Regarding barriers, they mentioned lack of motivation/self-discipline, artificiality, and lack of perceived need. Participants reported promotional information, incentives, and normalizing peer usage to facilitate uptake. Lastly, they mentioned program design, human coaching, and reminders to increase retention. These insights inform future research aimed at developing online intervention engagement models to increase uptake and retention. [ABSTRACT FROM AUTHOR]

International audience; The progress of science and technology has considerably increased the quality of life - especially the material standard of living - (e.g. disappearance of famines, increase in life expectancy) On the other hand, climate change, reduction of biodiversity, decrease of natural resources, and increase of environmental risks are some of the consequences of this human activities. Addressing these new challenges will require holistic comprehension and actionable approaches. The mitigation of these consequences requires effective responses in terms of transition. In this context, engineering and design students need time but also adapted curricula focused on sustainability and eco design concepts to support evolution of their skills, their knowledge and their culture. National audience; Jusqu'à présent, la recherche sur la soutenabilité s’est centrée principalement au niveau macro sur les acteurs institutionnels et les efforts de changement systémique. Au niveau micro, les chercheurs ont étudié le choix des produits par les consommateurs et leur élimination après usage [1].Le projet D-TechnoSS, financé par l'Agence nationale de la recherche (ANR) (ANR-20-CE10-0006), vise à développer une meilleure compréhension des mécanismes de prise de conscience et de changements des comportements associés à l’adoption de démarches de conception participatives dans une posture de soutenabilité forte.Le projet D-TechnoSS vise à explorer la proposition d’une intégration forte entre société et système de production. Dans le cadre du projet, il sera question de favoriser la prise de conscience des défis sociétaux, de faciliter le changement de comportements peu soutenables et d’accompagner le système productif dans on retour au seuil des limites planétaires. La méthode de recherche est basée sur une approche expérimentale, pour proposer un modèle d’évaluation de la soutenabilité dans une posture de soutenabilité forte.Le projet D-TechnoSS a donc pour ambition de mieux comprendre et accompagner les transitions sociales ainsi que l’adaptation nécessaire du système de production et des méthodes de conception.

Orientadores: Sandra Martha Gomes Dias, Ana Carolina de Mattos Zeri Tese (doutorado) - Universidade Estadual de Campinas, Instituto de Biologia Resumo: Grandes avanços foram feitos ao longo dos anos em nossa compreensão sobre o câncer, mas muitos detalhes relativos aos mecanismos responsáveis pelo desenvolvimento tumoral ainda não são completamente entendidos. Recentemente, a capacidade de reprogramar o metabolismo energético foi reconhecida como um dos hallmarks da transformaçao tumoral. Seu estudo e compreensão tem aberto novas e promissoras portas para a busca de alvos terapêuticos. Cerca de 10-20% dos casos de câncer de mama diagnosticados correspondem ao subtipo triplo-negativo (TN), o qual não expressa os receptores dos hormônios estrogênio e progesterona e não apresenta amplificação de HER2. Os tumores TN apresentam um perfil molecular único, comportamento agressivo e pior prognóstico em relação a outros subtipos de câncer de mama. Além, os tumores TN não respondem aos tratamentos convencionais desenvolvidos para a doença, o que ressalta a necessidade de se procurar novas estratégias terapêuticas. Tumores TN tem captação aumentada de 2-deoxi-2- (18F) fluoro-D-glicose (FDG) e dependência de glutamina, o que se correlaciona com seu alto índice proliferativo e evidencia a intensificação da glicólise aeróbica ou efeito Warburg. Neste trabalho, objetivamos analisar a expressão gênica diferencial e de metabólitos de linhagens celulares e tecidos tumorais de mama TN e não-TN de maneira a se caracterizar a assinatura metabólica do subtipo TN. Por fim, objetivamos identificar potenciais novo alvos terapêuticos para o tratamento deste subtipo tumoral. Como resultados, verificamos que tumores TN apresentam características que potencialmente lhes garantem uma alta eficiência no uso de nutrientes, com alta captação e processamento de glicose, alto processamento de glutamina, re-aproveitamento de lactato e utilização de acetaldeídos como fonte energética e biossintética. No último caso, lhe grantindo, uma potencial via de quimioresistência. Há ainda a indicação de ativação de vias de síntese/modificação de lipídeos que podem ter uma função sinalizadora nas células. Por fim, foi gerada uma lista de 73 genes com expressão aumentada em tumores de mama TN (em relação a não-TN). Desta lista, GBP1, uma guanylate-binding protein, mostrou diminuir seletivamente a proliferação celular de linhagens TN quando silenciado. Sua função para o estabelecimento/progressão/sobrevivência do subtipo TN precisa ser avaliada de maneira a confirmá-la como alvo para o tratamento desta doença Abstract: Great advances have been made over the years in our understanding of cancer, but many details of the mechanisms involved for tumor development are not yet fully understood. Recently, the ability to reprogram the energy metabolism was recognized as one of the hallmarks of tumoral transformation. His study and understanding has opened promising new doors in the search for therapeutic targets. About 10-20% of cases of breast cancer diagnosed corresponds to triple negative (TN) subtype, which does not express the hormones receptors of estrogen and progesterone and does not have HER2 amplification. TN tumors have a unique molecular profile, an aggressive behavior and a poor prognosis compared to others breast cancer subtypes. In addition, TN tumors do not respond to conventional treatments developed for the disease, which highlights the need to look for new therapeutic strategies. TN tumors increase the uptake of 2-deoxy-2- (18F) fluoro-D-glucose (FDG) and has a glutamine dependence, which correlates with their high proliferative index and demonstrates the enhancement of aerobic glycolysis or Warburg effect. In this study, we aimed to analyze differential gene expression and metabolomics from cell lines and tumor breast tissues TN and not TN (N-TN) subtypes in order to characterize the metabolic signature. Finally, we aimed to identify potential therapeutic targets for treatment of this tumor subtype (TN). As a result, we found that TN tumors potentially have characteristics that guarantee them a high efficiency in the use of nutrients, with high uptake and processing of glucose, high glutamine processing, re-use lactate and use of acetaldehyde as energy and biosynthetic source. This last resource could be an important way of chemoresistance. There is also activation of synthesis and modification pathways of lipids that can have important signaling function in cells. Finally, we generated a list of 73 genes with increased expression in TN breast tumors (relative to non-TN) and from this list, GBP1 a guanylate-binding protein, shown to reduce selectively the cell proliferation in TN cell lines when muted. El presente trabajo tiene como finalidad la elaboración de una campaña de comunicación integral para la Asociación Peruana de Viajes y Turismo (APAVIT) en un contexto de crisis causado por la pandemia mundial del COVID-19. A través de esta campaña, se buscará captar a un público objetivo Millennial con el objetivo de dar a conocer a la asociación y motivarlos a viajar por el Perú a través de agencias de viaje cuando se reinicien las actividades en el sector turismo. The purpose of this work is to develop a comprehensive communication campaign for the Peruvian Association of Travel and Tourism Agencies (APAVIT) in a context of crisis caused by the global pandemic of COVID-19. Both have complicated diagnostics due to their phenotypic, biological, and genetic heterogeneity, unknown etiology, and the underlying biological pathways, and molecular mechanisms are still not completely understood. Given the multifactorial complexity of these disorders, identification and implementation of metabolic biomarkers would assist in their early detection and diagnosis and facilitate disease monitoring and treatment responses. To date, numerous studies have utilized metabolomics to better understand psychiatric disorders, and findings from these studies have begun to converge. La globalización y la transformación digital han llevado a que los clientes sean más exigentes, que busquen calidad y precios justos por los servicios o productos que necesitan. Buscan empresas enfocadas realmente al cliente, que entiendan sus necesidades y cuyos proveedores sean sus aliados. La presente investigación se centra en el mercado de los servicios legales en Colombia, principalmente en la ciudad de Bogotá. Se analiza la necesidad que tiene este tipo de empresa en retener al mejor talento humano, y así desarrollar una propuesta aplicable a una empresa de servicios legales, que contribuya a disminuir los niveles de rotación de la organización. Por medio de entrevistas, se busca identificar la percepción sobre los factores clave que pueden afectar o influir en las decisiones de los empleados para permanecer o renunciar a sus empleos. Inicialmente, se indaga acerca del nivel de satisfacción de los empleados, del nivel de compromiso y sobre si el ambiente laboral es positivo, entre otros factores. La investigación parte de la revisión de la literatura de diferentes conceptos que conllevan a la identificación y definición de las variables objeto de estudio, tales como: estructura organizacional, compensación y beneficios, salario emocional, ambiente de trabajo positivo, retos y desarrollo, reconocimiento y sentido de orgullo. La investigación es de tipo cualitativa y descriptiva, y busca construir espirales hermenéuticas de posibles interpretaciones y apreciaciones de los documentos leídos y de las entrevistas realizadas. Los análisis de los resultados de estas indagaciones permitirán distinguir los factores que se deben tener en cuenta para el desarrollo de la investigación, la cual se realiza en una muestra de asociados de todos los niveles de una firma de abogados de la ciudad de Bogotá., Globalization and digital transformation have led customers to be more demanding, seeking quality and fair prices for the services or products they need. They look for companies really focused on the customer, which understand their needs and whose suppliers are their allies. This investigation is focuses on the market of legal services in Colombia, mainly in Bogotá city. Is analyzed the need this kind of company has in retaining the best human talent, and then develop an applicable proposal to a legal services company, helping to reduce the rotation levels of the organization. Through interviews, we seek to identify the perceptions about the key factors that can affect or influence the decisions of employees to stay or give up their jobs. Initially, is inquired about the employee’s satisfaction level, the commitment level, and about if there is a positive work environment, among other factors. The research starts is based on the literature review of different concepts that lead to the identification and definition of variables under study, such as: organizational structure, compensation and benefits, emotional salary, positive work environment, challenges and development, recognition and sense of pride. The nature of this research is qualitative and descriptive, and it seeks to construct hermeneutical spirals of possible interpretations and appreciations of the read documents and of the carried-out interviews. The analysis of the results of these investigations will allow us to identify the factors that must be considered for the development of the research, which is carried out in a sample of different levels associates in a law firm in Bogotá city.

Hepatoblastomas exhibit the lowest mutational burden among pediatric tumors. We previously showed that epigenetic disruption is crucial for hepatoblastoma carcinogenesis. Our data revealed hypermethylation of nicotinamide N-methyltransferase, a highly expressed gene in adipocytes and hepatocytes. The expression pattern and the role of nicotinamide N-methyltransferase in pediatric liver tumors have not yet been explored, and this study aimed to evaluate the effect of nicotinamide N-methyltransferase hypermethylation in hepatoblastomas. We evaluated 45 hepatoblastomas and 26 non-tumoral liver samples. We examined in hepatoblastomas if the observed nicotinamide N-methyltransferase promoter hypermethylation could lead to dysregulation of expression by measuring mRNA and protein levels by real-time quantitative polymerase chain reaction, immunohistochemistry, and Western blot assays. The potential impact of nicotinamide N-methyltransferase changes was evaluated on the metabolic profile by high-resolution magic angle spinning nuclear magnetic resonance spectroscopy. Significant nicotinamide N-methyltransferase downregulation was revealed in hepatoblastomas, with two orders of magnitude lower nicotinamide N-methyltransferase expression in tumor samples and hepatoblastoma cell lines than in hepatocellular carcinoma cell lines. A specific TSS1500 CpG site (cg02094283) of nicotinamide N-methyltransferase was hypermethylated in tumors, with an inverse correlation between its methylation level and nicotinamide N-methyltransferase expression. A marked global reduction of the nicotinamide N-methyltransferase protein was validated in tumors, with strong correlation between gene and protein expression. Of note, higher nicotinamide N-methyltransferase expression was statistically associated with late hepatoblastoma diagnosis, a known clinical variable of worse prognosis. In addition, untargeted metabolomics analysis detected aberrant lipid metabolism in hepatoblastomas. Data presented here showed the first evidence that nicotinamide N-methyltransferase reduction occurs in hepatoblastomas, providing further support that the nicotinamide N-methyltransferase downregulation is a wide phenomenon in liver cancer. Furthermore, this study unraveled the role of DNA methylation in the regulation of nicotinamide N-methyltransferase expression in hepatoblastomas, in addition to evaluate the potential effect of nicotinamide N-methyltransferase reduction in the metabolism of these tumors. These preliminary findings also suggested that nicotinamide N-methyltransferase level may be a potential prognostic biomarker for hepatoblastoma. [ABSTRACT FROM AUTHOR]

32. ESPECTRO OCULO-AURICULO-VERTEBRAL: UN REPORTE DE CASO.

- Abstract
El espectro oculo-auriculo-vertebral (OAVS, OMIM 164210) es una asociación no aleatoria de anomalías congénitas producidas por alteración en la morfogénesis del primer y segundo arcos faríngeos. La prevalencia es 1 en 3000-5000 nacidos vivos, con relación hombre:mujer 3:2. Dentro de las anomalías craneofaciales, usualmente unilaterales, se incluyen anomalías en orejas con o sin hipoacusia, microsomía hemifacial, hendiduras orofaciales y defectos oculares. Anomalías pulmonares, generalmente hipoplasia pulmonar, son raras y homolaterales a la microsomía hemifacial. El fenotipo clínico clásico incluye la tríada dermoides epibulbares/lipodermoides- microtia-asimetría facial. No existen criterios diagnósticos definitivos, considerándose como requisito único la microtia. El objetivo es presentar el caso de neonato femenino de 40.6 semanas, hijo de madre adolescente, sin antecedentes de importancia. Al nacimiento peso, talla y perímetro cefálico normales, APGAR 6, se evidenciaron malformación en la vía aérea alta, orejas de baja implantación, apéndices preauriculares bilaterales, globo ocular derecho ausente, blefarofimosis derecha, hipoplasia de hemicara derecha, retromicrognatia, cuello corto y radiografía de tórax con hipoplasia pulmonar derecha, costillas fusionadas, hemivértebras cervicales. Ecografías abdominal, renal, dorsolumbar y de cadera y cariotipo normales. Ecocardiograma normal con ductus arterioso permeable sin repercusión hemodinámica. Se realizó el diagnóstico clínico de OAVS con hipoplasia pulmonar, asociación raramente reportada en la literatura. [ABSTRACT FROM AUTHOR]

- Abstract
Cancer is a complex disease that can also affect the younger population; however, it is responsible for a relatively high mortality rate of children and youth, especially in low- and middle-income countries (LMICs). Besides that, lipidomic studies in this age range are scarce. Therefore, we analyzed blood serum samples from young patients (12 to 35 years) with bone sarcoma (osteosarcoma) and compared their lipidomics to the ones from the control group of samples, named healthy control (HC group), using NMR and LC-MS techniques. Furthermore, differences in the lipidomic profiles between OS patients with and without metastasis indicate higher glycerophosphocholine (GPC) and glycerophospholipid (GPL) levels in osteosarcoma and increased cholesterol, choline, polyunsaturated fatty acids (PUFAs), and glycerols during the metastasis. These differences, detected in the peripheral blood, could be used as biomarkers for liquid biopsy.
