The new strategy for vaccine development such as the fused protein multi-epitope capable of preventing the reactivation of latent tuberculosis infection (LTBi) can be an effective strategy for controlling tuberculosis (TB) worldwide. This study was conducted to evaluate the immunity of experimentally infected BALB/c mice with Mycobacterium tuberculosis after injection of DNA construct. Nineteen female BALB/c mice were divided into three groups and injected with 0.50 mL of M. tuberculosis. After 3 weeks, lung and spleen samples from the infected mice were examined. The protective effects of light chain 3-fused protein multi-epitope against TB were evaluated for post-exposure and therapeutic exposure. The lungs and spleens of the mice were aseptically removed after death for histopathology analysis. The bacterial colonies were counted, and the cells were stained after 3 weeks of incubation. No significant differences were observed between the post-exposure and therapeutic exposure groups. The pathological changes in the lung tissue of mice in these groups included an increase in the thickness of interalveolar septa, hyperemia, and intraparenchymal pulmonary hemorrhage centers (positive control), scattered hyperemic areas (negative control), and hyperemia in the interstitial tissue, scattered hyperemic areas in the lung parenchyma and lymphocytic infiltration centers (experimental group). Flow cytometry of the post-exposure and therapeutic exposure models showed insignificant changes in all three groups. It seems necessary to develop a post-exposure and therapeutic exposure vaccine strategy that focuses on LTBi to prevent the progression of the active disease. Newcastle disease (ND) is a highly contagious viral infection affecting poultry production in many countries. Strict biosecurity and the administration of live attenuated vaccines against the ND virus (NDV) are the main implements of controlling programs. This study evaluated the efficacy and potency of the Razi Clone12IR Newcastle vaccine in specific pathogen-free (SPF) chickens. Chickens were vaccinated with either the Razi Clone12IR vaccine (group A1, n=20) or an imported Clone vaccine (B1, n=20) in the first week of life and boosted in the second week via eye drop, while negative control chickens received PBS (C1, n=20). Half of the birds in each group were challenged with the virulent NDV strain in the third post-vaccination week (A2, B2, and C2 groups). Specific antibody responses were determined in the collected sera by the hemagglutination inhibition (HI) assay for up to eight weeks. Cell-mediated immunity (CMI) was determined by the lymphocyte proliferation assay three and six weeks after the second vaccination. Sections of the tissues and organs, including the trachea, lungs, cecal tonsils, spleen, the bursa of Fabricius, liver, and small intestine, were subjected to histopathology. The immunized groups A1 and B1 showed significantly higher HI antibody titers before the challenge than the control group. In addition, lymphocyte proliferation responses significantly increased in the peripheral blood of the vaccinated groups. After the challenge, the A2 and B2 groups conferred good protection and drastically reduced virus shedding. No main lesions were noted in the tissues or organs of the vaccinated group in histopathology. In a few cases, mild microscopic lesions were observed, including the infiltration of inflammatory cells, which was related to the effect of the vaccine virus. These results indicate that the Razi Clone12IR vaccine is safe and can be an efficient tool for NDV infections by inducing protective humoral and CMI responses. [ABSTRACT FROM AUTHOR]

BACKGROUND: The grivet monkey (Chlorocebus aethiops), a non-human primate (NHP), has contributed significantly as an animal model in a variety of biological systems due to its similarities with human. OBJECTIVES: This study was designed to establish a detailed procedure for in vitro maturation (IVM) of germinal-vesicle stage oocytes, in vitro fertilization (IVF) and in vitro embryo culture (IVC) of grivet monkey. METHODS: The reproductive organs were obtained from10 adult male and 4 adult female grivet monkeys after the proper anesthesia. The ovarian follicles were aspirated by aspiration technique or by the means of oocyte recovery with centrifugation (ORC). For the sperm recovery, the epididymides were dissected from the testicles and the tails of the epididymides were minced in the sperm washing medium and incubated for 15 min. At the time of insemination, a portion of the pre-incubated spermatozoa (10 μL) was introduced into 90 μL of fertilization medium containing about 20 matured oocytes. RESULTS: The data on the oocyte recovery rate and IVM showed significant increase (P<0.05) in the COCs recovered via ORC technique (9.8±0.41 and 90.90%) comparedto the recovered COCs using aspiration (4.45±0.32, 80.00%). The results showed similarity in the rate of cleavage in both groups (ORC and aspiration) (71%). The rate of embryo development to the blastocyst stage was significantly higher in the ORC group (43%) compared to the aspiration group (33.33%). CONCLUSIONS: It is concluded that the oocyte recovery technique is of great importance in terms of non-human primates' COCs competence in the in vitro embryo production (IVEP). On the other hand, it is well documented in the current research that the commercial ART medium used in the human infertility clinics is well applicable for the grivet monkey IVEP. In this article we described necropsy findings of Cysticercus tenuicollis infection among a group of 65 wildcaught imported vervet monkeys from Tanzania into "Razi vaccine and serum research institute Karaj-Iran" for assessment of oral polio vaccine neurovirulence test (MNVT). The methods of diagnosis were based on observation of the C. tenuicollis, morphological characters of scolex as well as the shape, size and the number of the rostellar hooks and histopathological examination. Totally 7 female monkeys were detected positive (10.8%) for C. tenuicollis. Our results highlighted that, most cases of C. tenuicollis infection in vervet monkeys detected incidentally and cause mild or no disease, but well developed cysts could be organized in abdominal cavity causing peritoneal cysticercosis. It was suggested to set up more measures such as ultrasound imaging and serological tests, on next import shipment of the vervet monkeys, to prevent entrance of infected animals to the post quarantine holding areas.
