9. Larvicidal efficacy and mode of action of compound composition from Adiantum latifolium on Oryctes rhinoceros

- Abstract
Oryctes rhinoceros Linn., a well known pest of coconut palm, causes irrecoverable damage, reduction in yield and death of the plant. Insecticides with novel mode of action, more target selective and biodegradable are needed to replace the impacts of synthetic pesticides. Unexplored phytochemicals may have unknown site of action, which are selective to insects and, are safe for humans and the environment. Adiantum latifolium is a shade loving fern resistant to pathogens due to the presence of phytocompounds. The study was undertaken to find out the larvicidal efficacy of a compound composition of triterpenoids isolated from the leaves of A. latifolium against O. rhinoceros and its mode of action. The compounds showed antibacterial activity against gut bacteria of the larvae and larvicidal activity (LD50, 19.55 mg/kg), weight loss, 20-hydroxyecdysone titre and precocious metamorphosis of the larvae.. It also induced immunotoxicological effects via cellular and numerical changes of haemocytes in the haemolymph. The synergistic interaction of the triterpenoids is useful in the preparation of improved biopesticides.

White matter hyperintensities (WMH) are the most common brain-imaging feature of cerebral small vessel disease (SVD), hypertension being the main known risk factor. Here, we identify 27 genome-wide loci for WMH-volume in a cohort of 50,970 older individuals, accounting for modification/confounding by hypertension. Aggregated WMH risk variants were associated with altered white matter integrity (p=2.5x10-7) in brain images from 1,738 young healthy adults, providing insight into the lifetime impact of SVD genetic risk. Mendelian randomization suggested causal association of increasing WMH-volume with stroke, Alzheimer-type dementia, and of increasing blood pressure (BP) with larger WMH-volume, notably also in persons without clinical hypertension. Transcriptome-wide colocalization analyses showed association of WMH-volume with expression of 39 genes, of which four encode known drug targets. Finally, we provide insight into BP-independent biological pathways underlying SVD and suggest potential for genetic stratification of high-risk individuals and for genetically-informed prioritization of drug targets for prevention trials. White matter hyperintensities (WMH) are a common brain-imaging feature of cerebral small vessel disease. Here, the authors carry out a GWAS and followup analyses for WMH-volume, implicating several variants with potential for risk stratification and drug targeting.

- Abstract
White matter hyperintensities (WMH) are the most common brain-imaging feature of cerebral small vessel disease (SVD), hypertension being the main known risk factor. Here, we identify 27 genome-wide loci for WMH-volume in a cohort of 50,970 older individuals, accounting for modification/confounding by hypertension. Aggregated WMH risk variants were associated with altered white matter integrity (p = 2.5×10-7) in brain images from 1,738 young healthy adults, providing insight into the lifetime impact of SVD genetic risk. Mendelian randomization suggested causal association of increasing WMH-volume with stroke, Alzheimer-type dementia, and of increasing blood pressure (BP) with larger WMH-volume, notably also in persons without clinical hypertension. Transcriptome-wide colocalization analyses showed association of WMH-volume with expression of 39 genes, of which four encode known drug targets. Finally, we provide insight into BP-independent biological pathways underlying SVD and suggest potential for genetic stratification of high-risk individuals and for genetically-informed prioritization of drug targets for prevention trials.

Methods: We performed meta-analyses of genome-wide association studies (GWAS) and examined associations of vascular risk factors and their genetic risk scores (GRS) with MRI-defined BI and a subset of BI, namely, small subcortical BI (SSBI), in 18 population-based cohorts (n = 20,949) from 5 ethnicities (3,726 with BI, 2,021 with SSBI). Top loci were followed up in 7 population-based cohorts (n = 6,862; 1,483 with BI, 630 with SBBI), and we tested associations with related phenotypes including ischemic stroke and pathologically defined BI. Results: The mean prevalence was 17.7% for BI and 10.5% for SSBI, steeply rising after age 65. Two loci showed genome-wide significant association with BI: FBN2, p = 1.77 × 10-8; and LINC00539/ZDHHC20, p = 5.82 × 10-9. Both have been associated with blood pressure (BP)-related phenotypes, but did not replicate in the smaller follow-up sample or show associations with related phenotypes. Age- and sex-adjusted associations with BI and SSBI were observed for BP traits (p value for BI, p [BI] = 9.38 × 10-25; p [SSBI] = 5.23 × 10-14 for hypertension), smoking (p [BI] = 4.4 × 10-10; p [SSBI] = 1.2 × 10-4), diabetes (p [BI] = 1.7 × 10-8; p [SSBI] = 2.8 × 10-3), previous cardiovascular disease (p [BI] = 1.0 × 10-18; p [SSBI] = 2.3 × 10-7), stroke (p [BI] = 3.9 × 10-69; p [SSBI] = 3.2 × 10-24), and MRI-defined white matter hyperintensity burden (p [BI] = 1.43 × 10-157; p [SSBI] = 3.16 × 10-106), but not with body mass index or cholesterol. GRS of BP traits were associated with BI and SSBI (p ≤ 0.0022), without indication of directional pleiotropy. Conclusion: In this multiethnic GWAS meta-analysis, including over 20,000 population-based participants, we identified genetic risk loci for BI requiring validation once additional large datasets become available. A paleomagnetic estimate of a depositional latitude for the lavas, as well as for the underlying contemporaneous glacial rocks, is reported. The paleolatitudinal estimate was derived from observations made of the Archaean-Paleoproterozoic 'Transvaal Supergroup' on the Kapvaal craton in South Africa. Within this supergroup are the Ongeluk lavas which conformably overlies glaciogenic deposits. The Ongeluk lavas are about 2.2 billion years old. It is hoped that such a study of the Earth's paleoclimate would provide a clue to the temporal and spatial distributions of Precambrian glaciations.

Oriented samples of the Lower Cambrian Bayan Gol Formation from Salaany Gol, Mongolia, were collected at roughly 5 m stratigraphic intervals for palaeomagnetic analysis. Progressive alternating-field and thermal demagnetization isolated two magnetic components: a present-field overprint, typically removed by 10 mT fields and [approximately]200 [degrees] C heating; and a high-coercivity, high-unblocking-temperature (550-600 [degrees] C), predominantly single-polarity component that was imparted to the rocks prior to early or middle Palaeozoic deformation. Single-polarity magnetization at Salaany Gol contrasts with results from Lower Cambrian rocks on the Siberian platform, previously considered correlative with the Bayan Gol Formation, which show a prominent change in polarity bias near the top of the Tommotian Stage. Two hypotheses can explain this discrepancy. First, the entire Bayan Gol Formation may correlate with the predominantly reversely polarized, lower half of the Tommotian Stage in Siberia. This model is consistent with plausible interpretations of [Delta]13C profiles for the Zavkhan basin and the Siberian platform. Alternatively, the characteristic magnetic direction from our samples may be a pre-fold overprint. If post-accretionary, then comparison with Siberian palaeomagnetic results suggest a Silurian-Devonian remagnetization age, and existing bio- and chemostratigraphic correlations provide the most reliable spatial and temporal links between the Zavkhan basin and the Siberian platform. If the observed magnetic directions are primary or an immediate overprint then they may be used to constrain the early Cambrian palaeogeography of the Zavkhan basin and the Palaeo-Pacific Ocean. Mean inclination of 62 [+ or -] 4 [degrees] corresponds to a palaeolatitude of 44 [+ or -] 5 [degrees], several thousand kilometres from the equatorial Siberian craton.

Age- and sex-adjusted associations with BI and SSBI were observed for BP traits (p value for BI, p[BI] = 9.38 x 10-25; p [SSBI] = 5.23 x 10-14 for hypertension), smoking (p[BI]= 4.4 x 10-10; p [SSBI] = 1.2 x 10 -4), diabetes (p[BI] = 1.7 x 10 -8; p [SSBI] = 2.8 x 10 -3), previous cardiovascular disease (p [BI] = 1.0 x 10-18; p [SSBI] = 2.3 x 10-7), stroke (p [BI] = 3.9 x 10-69; p [SSBI] = 3.2 x 10 -24), and MRI-defined white matter hyperintensity burden (p [BI]=1.43 x 10-157; p [SSBI] = 3.16 x 10-106), but not with body mass index or cholesterol. GRS of BP traits were associated with BI and SSBI (p <= 0.0022), without indication of directional pleiotropy. Conclusion In this multiethnic GWAS meta-analysis, including over 20,000 population-based participants, we identified genetic risk loci for BI requiring validation once additional large datasets become available. - Abstract
Introduction: There is conflicting evidence whether high-density lipoprotein cholesterol (HDL-C) is a risk factor for Alzheimer's disease (AD) and dementia. Genetic variation in the cholesteryl ester transfer protein (CETP) locus is associated with altered HDL-C. We aimed to assess AD risk by genetically predicted HDL-C. Methods: Ten single nucleotide polymorphisms within the CETP locus predicting HDL-C were applied to the International Genomics of Alzheimer's Project (IGAP) exome chip stage 1 results in up 16,097 late onset AD cases and 18,077 cognitively normal elderly controls. We performed instrumental variables analysis using inverse variance weighting, weighted median, and MR-Egger. Results: Based on 10 single nucleotide polymorphisms distinctly predicting HDL-C in the CETP locus, we found that HDL-C was not associated with risk of AD (P >.7). - Abstract
Applying HaploReg v4.1 analysis to the 536 variants resulted in 9 categories (Supplementary Table 8): miscRNA (1 variant); snoRNA (2 variants); microRNA (4 variants); snRNA (9 variants); pseudogenes (14 variants); sequencing in progress (43 variants); LINC RNA (86 variants); and 372 variants within protein coding genes. In addition, some variants annotate to the same gene resulting in a total of 139 genes (protein-coding or non-coding). Of those genes, 6 are exceptionally long, containing over a million base-pairs, the longest of which is PTPRT coded by 1117219bp. The shortest genes are the ones coding for micro (MIR3143) or small nuclear (U7) RNAs at 63bp each. There is only partial information regarding the chromatin state of each variant. However, from the information gathered in the analysis we observed 14 transcription start sites and 245 enhancers (Supplementary Table 8).

The suppressive effect of Sb interlayers in CdS/InP heterojunctions on the interface reaction and band offset modification was investigated using core and valence level spectra analysis. The results showed that the Sb interlayer effectively suppressed the interface reaction. However, the effect of the Sb interlayer on the charge distribution at the interface was negligible.

The deglacial history of the central sector of the last British–Irish Ice Sheet is poorly constrained, particularly along major ice-stream flow paths. The Tyne Gap Palaeo-Ice Stream (TGIS) was a major fast-flow conduit of the British–Irish Ice Sheet during the last glaciation. We reconstruct the pattern and constrain the timing of retreat of this ice stream using cosmogenic radionuclide (10Be) dating of exposed bedrock surfaces, radiocarbon dating of lake cores and geomorphological mapping of deglacial features. Four of the five 10Be samples produced minimum ages between 17.8 and 16.5 ka. These were supplemented by a basal radiocarbon date of 15.7 ± 0.1 cal ka BP, in a core recovered from Talkin Tarn in the Brampton Kame Belt. Our new geochronology indicates progressive retreat of the TGIS from 18.7 to 17.1 ka, and becoming ice free before 16.4–15.7 ka. Initial retreat and decoupling of the TGIS from the North Sea Lobe is recorded by a prominent moraine 10–15 km inland of the present-day coast. This constrains the damming of Glacial Lake Wear to a period before ∼18.7–17.1 ka in the area deglaciated by the contraction of the TGIS. We suggest that retreat of the TGIS was part of a regional collapse of ice-dispersal centres between 18 and 16 ka.
