Abstract

In cystic fibrosis (CF), airway mucus becomes thick and viscous, and its clearance from the airways is impaired. The gel-forming mucins undergo an ordered "unpacking/maturation" process after granular release that requires an optimum postsecretory environment, including hydration and pH. We hypothesized that this unpacking process is compromised in the CF lung due to abnormal transepithelial fluid transport that reduces airway surface hydration and alters ionic composition. Using human tracheobronchial epithelial cells derived from non-CF and CF donors and mucus samples from human subjects and domestic pigs, we investigated the process of postsecretory mucin unfolding/maturation, how these processes are defective in CF airways, and the probable mechanism underlying defective unfolding. First, we found that mucins released into a normal lung environment transform from a compact granular form to a linear form. Second, we demonstrated that this maturation process is defective in the CF airway environment. Finally, we demonstrated that independent of HCO3- and pH levels, airway surface dehydration was the major determinant of this abnormal unfolding process. This defective unfolding/maturation process after granular release suggests that the CF extracellular environment is ion/water depleted and likely contributes to abnormal mucus properties in CF airways prior to infection and inflammation.

Abstract

Summary: Leaf dark respiration (R dark ) is an important yet poorly quantified component of the global carbon cycle. Given this, we analyzed a new global database of R dark and associated leaf traits. Data for 899 species were compiled from 100 sites (from the Arctic to the tropics). Several woody and nonwoody plant functional types (PFTs) were represented. Mixed-effects models were used to disentangle sources of variation in R dark . Area-based R dark at the prevailing average daily growth temperature (T) of each site increased only twofold from the Arctic to the tropics, despite a 20°C increase in growing T (8-28°C). By contrast, R dark at a standard T (25°C, R dark 25 ) was threefold higher in the Arctic than in the tropics, and twofold higher at arid than at mesic sites. Species and PFTs at cold sites exhibited higher R dark 25 at a given photosynthetic capacity (V cmax 25 ) or leaf nitrogen concentration ([N]) than species at warmer sites. R dark 25 values at any given V cmax 25 or [N] were higher in herbs than in woody plants. The results highlight variation in R dark among species and across global gradients in T and aridity. In addition to their ecological significance, the results provide a framework for improving representation of R dark in terrestrial biosphere models (TBMs) and associated land-surface components of Earth system models (ESMs). © 2015 New Phytologist Trust.

Abstract

Most of the estimated 20 million people who are blind with cataracts live in rural areas of developing countries, where expert surgical resources are scarce. We have studied the use of multiflex open-loop anterior-chamber intraocular lenses (ACIOL)in high-volume low-cost surgery. Between 1992 and 1995, a total of 2000 people attending Lahan Eye Hospital, Nepal, with bilateral cataracts reducing vision to [is less than or equal to] 6/36 were randomly allocated to receive intracapsular extraction (ICCE) with aphakic spectacles, or ICCE with an ACIOL. We re-examined the cohort (1305/2000, 65%) between November 1996 and April 1997 and report the findings in this article. There were 13 new cases of poor visual outcome (best corrected vision [is less than] 6/60) arising after one year: 9 in the ACIOL group and 4 in the control group; odds ratio 2.1 (95% confidence interval, 0.59-9.55). The causes of poor outcome were as follows: ACIOL group -- retinal detachment (4 cases), cystoid macular oedema (2), epiretinal membrane (1), age-related macular degeneration (1), and late endophthalmitis (1); control group -- retinal detachment (2 cases), late endophthalmitis (1), and primary open-angle glaucoma with age-related macular degeneration (1). In rural areas of developing countries, well--manufactured multiflex open-loop ACIOLs can be implanted safely by experienced ophthalmologists after routine ICCE, avoiding the disadvantages of aphakic spectacle correction. Abstract

We conducted a meta-analysis of Parkinson's disease genome-wide association studies using a common set of 7,893,274 variants across 13,708 cases and 95,282 controls. Twenty-six loci were identified as having genome-wide significant association; these and 6 additional previously reported loci were then tested in an independent set of 5,353 cases and 5,551 controls. Of the 32 tested SNPs, 24 replicated, including 6 newly identified loci. Conditional analyses within loci showed that four loci, including GBA, GAK-DGKQ, SNCA and the HLA region, contain a secondary independent risk variant. In total, we identified and replicated 28 independent risk variants for Parkinson's disease across 24 loci. Although the effect of each individual locus was small, risk profile analysis showed substantial cumulative risk in a comparison of the highest and lowest quintiles of genetic risk (odds ratio (OR) = 3.31, 95% confidence interval (CI) = 2.55-4.30; P = 2 × 10-16). This first analysis of the two-stage IPDGC study focused on the set of loci that passed genome-wide significance in the first stage GWA scan. However, the second stage genotyping array, the ImmunoChip, included a larger set of 1,920 SNPs selected on the basis of the GWA analysis. Abstract

In our recent study of a seven-story reinforced concrete hotel in Van Nuys, California, which was damaged during Northridge 1994, earthquake, we showed that the prediction of where the localized damage may occur should be carried out by non-linear wave propagation methods (Gicev and Trifunac 2006). In this paper we describe how the rotations (local strains and drifts) in the building, depend upon the distribution of the stiffness along the building height, nonlinear properties of the reinforced concrete, and the nature of strong motion. To search for the optimum performance parameters, we vary the distribution of the velocity of propagation of SH waves from story to story. Five hypothetical buildings with: sinusoidal, linear, concave quadratic parabola, convex quadratic parabola and cubic parabola stiffness distributions, together with the actual distribution observed in the structure are considered. It is shown that the maximum rotations appear in the actual building, while the minimum rotations occur for the buildings with velocity distributions described by cubic parabola and quadratic convex parabola (maximum stiffness at the bottom) distributions. It is concluded that the design of the seven-story reinforced concrete hotel was not optimal. For design of new structures and for retrofit of old ones near faults, the stiffness distributions should resemble the cubic parabola or quadratic convex parabola distributions.

Abstract

This ecophysiological primer serves as an introductory chapter on the concepts applied in subsequent chapters reviewing the state of knowledge concerning the effects of climate change on ecological processes that affect impacts on terrestrial ecosystems and some socio-economic systems. Because many of the responses of terrestrial plants in the diverse ecosystems are similar, this primer discusses: the effects of important climatic driving forces that are likely to change; soil fertility as a modifying effect with a brief discussion on soil biological factors; and ecological interactions between the different organisms in each ecosystem.
