Abstract

Fitting statistical models to spatiotemporal data requires finding the right balance between imposing smoothness and following the data. In the context of p-splines, we propose a Bayesian framework for choosing the smoothing parameter which allows the construction of fully-automatic data-driven methods for fitting flexible models to spatiotemporal data. A computationally efficient implementation, exploiting the sparsity of the arising design and penalty matrices, is proposed. The findings are illustrated using a simulation and two examples, all concerned with the modelling of contaminants in groundwater, which suggest that the proposed strategy is more stable that competing strategies based on the use of criteria such as GCV and AIC.

Abstract

Purpose: To evaluate the effectiveness of mobile health applications (mHealth apps) containing a physical training component on physical function and physical activity in stroke rehabilitation. Materials and methods: A systematic literature search was conducted in three databases for studies published from inception to 12 July 2022. Clinical trials including mHealth apps with a physical training component were included using outcomes of physical function and physical activity. Quality of life was extracted as a secondary outcome. Results: Five RCTs, two non-RCTs, and four uncontrolled clinical trials were included with a total of 264 stroke survivors. Eleven apps were identified with a physical training component using features of gamification (six apps), exercise prescription (three apps), and physical activity (two apps). Six out of seven studies reported statistically significant improvements in physical function in favor of the experimental group, with the most robust findings for upper extremity function. For physical activity, statistically significant improvements were seen in the experimental groups. Only one study showed significant improvement in quality of life. Overall study quality was fair. Conclusions: mHealth apps containing a physical training component are promising for physical function and physical activity in stroke rehabilitation. Abstract

A period of 18 years of infrasonic recordings was analyzed from a microbarometer array (I18DK) in northwestern Greenland, near Qaanaaq. A huge number of infrasonic detections, over 700,000, have been made in I18DKs soundscape during the Arctic summers. Simultaneously identified were both calving events from marine-terminating glaciers and discharge related acoustics from a land-terminating glacier. This infrasonic activity is correlated to sea-surface and atmospheric temperature, respectively. Inter-yearly to daily variations were retrieved showing a strong variability in infrasonic detection rates and hence glacier activity. The highest number of infrasonic detections were found in recent years from the land-terminating glacier. The latter is supported by actual discharge measurements and partly by a discharge model. It is concluded that monitoring infrasound from glaciers can complement other techniques to remotely and passively get insights into glacier dynamics with high temporal and spatial resolution.

Abstract

This paper presents a new method called the functional distributional clustering algorithm (FDCA) that seeks to identify spatially contiguous clusters and incorporate changes in temporal patterns across overcrowded networks. This method is motivated by a graph-based network composed of sensors arranged over space where recorded observations for each sensor represent a multi-modal distribution. The proposed method is fully non-parametric and generates clusters within an agglomerative hierarchical clustering approach based on a measure of distance that defines a cumulative distribution function over temporal changes for different locations in space. Traditional hierarchical clustering algorithms that are spatially adapted do not typically accommodate the temporal characteristics of the underlying data. The effectiveness of the FDCA is illustrated using an application to both empirical and simulated data from about 400 sensors in a 2.5 square miles network area in downtown San Francisco, California. The results demonstrate the superior ability of the the FDCA in identifying true clusters compared to functional only and distributional only algorithms and similar performance to a model-based clustering algorithm. Abstract

This study describes an automated data quality verification procedure supported by a database of power spectral densities (PSD) estimates for geophysical waveform data. The Royal Netherlands Meteorological Institute (KNMI) manages a 100-TB archive of continuous geophysical data collected from accelerometers, geophones, broadband seismometers, and infrasonic arrays deployed across the continental and Caribbean Netherlands. This rapidly expanding network at a scale of over 700 instruments makes the manual evaluation of data quality impractical and must be supported by data policies and automated methods. A technique is presented to compress and store PSD estimates in a database with a storage footprint of less than 0.05% of the raw data archive. Every week, the instrument performance is validated by comparing statistical properties of its latest monthly probabilistic PSD distribution to strict quality metrics. The criteria include thresholds based on global noise models, datalogger quantization noise models, constraints imposed by ambient noise conditions, and confidence intervals based on PSD estimates calculated from validated archived data. When a threshold is crossed, the station operator is alerted of the suspected degraded instrument performance, severely limiting the required amount of manual labor and associated human errors. The automated PSD assessment technique is applicable to accelerometers, geophones, broadband seismometers, infrasonic stations, and is demonstrated to be extendable to hydrophones, gravimeters, tiltmeters, and Global Navigation Satellite System receivers. The approach is therefore suitable for other geophysical monitoring infrastructures, for example, observational networks dedicated to continuous volcano monitoring. It is shown that it possible to detect degraded instrument performance that may otherwise remain undetected.

Abstract

Background & Aims: Continuing recalcitrance to therapy cements pancreatic cancer (PC) as the most lethal malignancy, which is set to become the second leading cause of cancer death in our society. The study aim was to investigate the association between DNA damage response (DDR), replication stress, and novel therapeutic response in PC to develop a biomarker-driven therapeutic strategy targeting DDR and replication stress in PC. Methods: We interrogated the transcriptome, genome, proteome, and functional characteristics of 61 novel PC patient–derived cell lines to define novel therapeutic strategies targeting DDR and replication stress. Validation was done in patient-derived xenografts and human PC organoids. Results: Patient-derived cell lines faithfully recapitulate the epithelial component of pancreatic tumors, including previously described molecular subtypes. Biomarkers of DDR deficiency, including a novel signature of homologous recombination deficiency, cosegregates with response to platinum (P <.001) and PARP inhibitor therapy (P <.001) in vitro and in vivo. We generated a novel signature of replication stress that predicts response to ATR (P <.018) and WEE1 inhibitor (P <.029) treatment in both cell lines and human PC organoids. Replication stress was enriched in the squamous subtype of PC (P <.001) but was not associated with DDR deficiency. Conclusions: Replication stress and DDR deficiency are independent of each other, creating opportunities for therapy in DDR-proficient PC and after platinum therapy.

Abstract

Gravity waves are an important part of the momentum budget of the atmosphere. Despite this, parameterizations of gravity wave spectra in atmospheric models are poorly constrained. Gravity waves are formed by jet streams, flow over topography and convection, all of which produce pressure perturbations as they propagate over the Earth’s surface, detectable by microbarometer arrays used for sensing infrasound. In this study, observations of gravity waves between 2007 and 2011 at an infrasound station in the Ivory Coast, West Africa are combined with meteorological data to calculate parameters such as intrinsic phase speed and wavenumber. Through spectral analysis, the seasonal and daily variations in all gravity wave parameters are examined. The gravity wave back azimuth varies with the migration of the Inter-Tropical Convergence Zone, a region of intense convection, supporting previous studies. Daily variations in gravity wave arrivals at the station can be linked to two distinct convective cycles over the land and ocean. This was achieved by combining the gravity wave parameters with lightning strikes detected by the Met Office’s Arrival Time Difference lightning detection system. Noise generated by turbulence in the middle of the day was found to attenuate smaller pressure amplitude gravity waves, artificially amplifying the daily variations in some gravity wave parameters. Detection of daily and seasonal variations in gravity wave parameters has the potential be used to improve the representation of gravity wave spectra in atmospheric models.

Abstract

This paper introduces the treethresh package offering accurate estimation, via thresholding, of potentially sparse heterogeneous signals and the denoising of images using wavelets. It gives considerably improved performance over other estimation methods if the underlying signal or image is not homogeneous throughout but instead has distinct regions with differing sparsity or strength characteristics. It aims to identify these different regions and perform separate estimation in each accordingly. The base algorithm offers code which can be applied directly to any one-dimensional potentially sparse sequence observed subject to noise. Also included are functions which allow two-dimensional images to be denoised following transformation to the wavelet domain. In addition to reconstructing the underlying signal or image, the package provides information on the believed partitioning of the signal or image into its differing regions.
