Sargahydroquinoic acid (SHQA), a bioactive compound found in certain Sargassum species, exhibits significant health benefits. This study optimized the extraction of SHQA from Sargassum yezoense using response surface methodology (RSM) and evaluated its antioxidant effects through in vitro and in vivo assays. A Box–Behnken design (BBD) was effectively employed to investigate the effects of incubation temperature, time, and ethanol concentration on SHQA yield, achieving a high coefficient of determination (R2 = 0.961). Analysis of variance (ANOVA) validated the model's reliability (F = 13.86, p = 0.005) and highlighted ethanol concentration as a highly significant factor (p < 0.001). Optimal extraction conditions were identified as 52.8 °C, 8.3 h, and 74.1% ethanol. The SHQA-maximized extract (SME) contained 67.8 ± 0.6 mg SHQA/g and 25.00 ± 1.01 mg phloroglucinol equivalent/g. SME exhibited antioxidant capacity of 26.45 ± 0.66 mg and 28.74 ± 2.30 mg vitamin C equivalent/g in ABTS and DPPH assays, respectively, and 0.29 ± 0.02 mM FeSO4 equivalent/g in the FRAP assay. Additionally, SME at 50 µg/mL and SHQA at 1 µg/mL inhibited reactive oxygen species (ROS) generation in an H2O2-induced zebrafish model. Background: Precision-cut lung slices (PCLS) are ex vivo models with preserved lung cell populations and maintained tissue architecture. PCLS are, therefore, a powerful tool in respiratory research to study molecular mechanisms that closely reflect whole tissue biology. High-quality RNA and protein extraction from PCLS is, however, challenging as agarose significantly interferes with the yield and purity of extracted material. The present study aimed to optimize QIAzol-based isolation protocol for high-yield and quality RNA, miRNA, and protein extraction from PCLS. Materials and methods: PCLS were prepared from 10 to 15-week-old Wistar rats and cultured for 7 days in Dulbecco's Modified Eagle Medium/Nutrient Mixture F-12 (DMEM/F-12) supplemented with 0.1% FBS, penicillin, and streptomycin. LDH release to PCLS culture media was measured to determine cellular cytotoxicity. To select the optimal miRNA/RNA isolation protocol, we tested two different times (10 min, 2 h) and temperatures (room temperature, 4 °C, and −20 °C) of precipitation with isopropanol. Finally, we also assessed isolation with GHCL (guanidinium hydrochloride) extraction buffer. To select the optimal protein isolation protocol, we tested protein precipitation for 10 min at room temperatures (21 ± 1 °C) with 1.5 volumes of isopropanol and 3 volumes of acetone per 1 volume of phenol-ethanol supernatant. Additionally, we also tested protein precipitation for 3 h at −20 °C with 3, 5, and 7 acetone volumes per 1 volume of phenol-ethanol supernatant. We also validated protein precipitation with back extraction buffer instead of 100% ethanol. To measure the general efficiency of the optimized QIAZ-4 protocol, we used native rat lungs. PCLS for the ex vivo model of allergic inflammation were treated with IL-13 at a concentration of 80 ng/ml. Results: Standard QIAzol isolation protocol provided RNA, miRNA, and protein with low yield and poor quality. We found that 2-h isopropanol precipitation at 4 °C with a high concentration of salts significantly increased the yield and quality of extracted RNA and miRNA and provided acceptable qPCR efficiency (between 90 and 110%). Surprisingly, 2-h isopropanol precipitation at −20 °C significantly increased qPCR efficiency above the acceptable range (average efficiency: 120.4%). As for protein extraction, we found that 3-h acetone precipitation at −20 °C provided the highest yield with linear protein detection on Westen Blot. Optimized QIAZ-4 provided significantly higher miRNA and RNA yield compared to standard QIAzol protocols. We also found a significantly increased expression of Eotaxin-1 in PCLS treated with IL-13 as compared to the untreated controls. The aim of the present research is to investigate the effect of ohmic‐assisted extraction (OAE) parameters on the Falcaria vulgaris extract (FVE) properties. Various parameters, including ethanol‐to‐water ratio (0%–100%), voltage gradient (10–30 V/cm), temperature (30°C–60°C), and extraction time (10–30 min), were applied as independent factors. Physicochemical process, antimicrobial properties, and energy analysis were accomplished to achieve OAE optimization. Results indicated that extraction yield (EY) and energy consumption (EC) significantly increased with increasing the voltage gradient and extraction time (p < 0.05). Also, EY significantly increased with increasing of process temperature and the ethanol concentration in the solvent (p < 0.05). The findings demonstrated that as the voltage gradient, process duration, temperature, and ethanol concentration in the solvent were increased, there was a significant initial increase followed by a decrease in both the total phenolic content (TPC) and the DPPH radical scavenging activity of FVE (p < 0.05). The optimization results indicated that the extraction process with an ethanol‐to‐water ratio of 65.5%, voltage gradient of 20.3 V/cm, temperature of 56.6°C, and extraction time of 22.1 min results in maximum extract TPC, DPPH, and EY, and minimum extraction EC. The OAE method showed greater EY and less EC and CO2 production compared to the hydrodistillation (HD) method. In general, OAE caused improved antioxidant and antimicrobial properties of the FVE. This study investigated the effect of high-pressure enzymatic treatment (HPET) on the quality characteristics such as soluble solid content, yield, flavonoid content, and phenolic acid content of mulberry fruits. Of particular note, first, the quality characteristics of the mulberry fruits were most significantly improved under the high-pressure treatment with mixture of Pectinex Be XXL and Ultra color. Second, in the flavonoids, a decrease in rutin content and increases in hyperoside and quercetin contents were confirmed by HPET, which may be the result of the two-step deglycosylation of rutin by pectinase activity. Finally, the extraction yield of mulberry fruits was maintained during HPET 0–36 h, and its bioactive compound profile was variously changed. Therefore, this study provided new clues as to how the HPET may promote changes in the extraction yield and bioactive substance profile of mulberry fruits and suggest the potential for use of natural materials based on mulberry fruit extracts. The selection of appropriate solvents in the extraction process plays a crucial role in determining the nature and quantity of secondary metabolites extracted from medicinal plants. This study investigates the impact of solvent polarity on the extraction yield, phenolic content, and antioxidant properties of seeds from four plants belonging to the Malvaceae family: Abutilon fruticosum, Abutilon pannosum, Sida ovata and Malvastrum coromendelianum. The seed powder of each species was subjected to extraction using solvents with varying polarities, including acetone, chloroform, methanol, and water. Physicochemical analysis, extraction yield, quantitative phenolic content, and antioxidant activity were assessed. Total phenolic content (TPC) was determined using the Folin–Ciocalteu reagent, and antioxidant activity was measured using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay. Analysis of variance (ANOVA) revealed that the choice of extracting solvents significantly influenced (p < 0.05) extraction yield, phenolic content, and antioxidant properties. In conclusion, water and methanol emerged as the most effective solvents for achieving high extraction yields, phenolic compound content, and free radical scavenging activities. The results demonstrated a robust correlation between total phenolic content (mg of Gallic acid equivalent/g of sample) and free radical scavenging activity (IC50). The study highlights the substantial impact of solvent type, suggesting the potential benefits of using a combination of polar and nonpolar solvents to enhance the extraction efficiency of phytochemicals, phenolic content and antioxidant quality from medicinal plant seeds. The current study aimed to enhance the coagulation properties of camel milk using enzymatic extracts derived from Moringa oleifera seeds. Experiments were conducted to evaluate the clotting activities of Moringa extract on camel milk and compare them with cow milk. Our results regarding the characterisation of the enzymatic extract showed an extraction yield of 54.3±1.8%. The optimum coagulation conditions were determined to be pH 5 and a temperature of 55°C. In addition, the enzymes exhibited substantial clotting activity (2.54 RU) on both camel and cow milk. The cheese samples showed significant oxidative and antibacterial activity. This study aimed to develop an efficient method for the extraction of bioactive compounds from green tea waste (GTW) toward its potential application in the food industry. GTW, which is generated during the harvesting and processing of green tea products, accounts for a global annual loss of nearly 1 million tonnes. Notably, this waste is rich in polyphenolic compounds, particularly catechins, which are renowned for their significant health benefits. We assessed the optimization of catechin extraction from GTW employing hot water extraction (HWE), ultrasound‐assisted extraction (UAE), and ethanol extraction (EthE) techniques at different sample‐to‐solvent ratios (1:100, 1:50, and 1:20 w/v). The extraction temperature was set at 80°C for both HWE and UAE; however, for EthE, the temperature was slightly lower at 70°C, adhering to the boiling point of ethanol. High‐performance liquid chromatography was used to determine the extraction efficiency by quantifying various catechins (i.e., catechin, epicatechin [EC], epicatechin gallate [ECG], epigallocatechin [EGC], and epigallocatechin gallate [EGCG]). In terms of the concentration for individual catechins, EC was found to be the highest concentration detected, ranging from 30.58 ± 1.17 to 37.95 ± 0.84 mg/L in all extraction techniques and ratios of solvents, followed by EGCG (9.71 ± 1.40–20.99 ± 1.11 mg/L), EGC + C (7.95 ± 0.66–12.58 ± 0.56 mg/L), and ECG (1.85 ± 0.71–6.05 ± 0.06 mg/L). The findings of DPPH (2,2‐diphenyl‐1‐picryl‐hydrazyl) free radical assay illustrated that HWE demonstrated the highest extraction efficiency at all ratios, ranging from 61.41 ± 1.00 to 70.36 ± 1.47 mg/L. The 1:50 ratio exhibited the highest extraction yield (25.98% ± 0.75%) compared to UAE (24.16% ± 0.95%) and EthE (22.59% ± 0.26%). Moreover, this method of extraction (i.e., HWE) produced the highest total catechins and %DPPH reduction. Consequently, HWE was the most efficient method for extracting catechins from GTW, underscoring its potential for valorizing waste within the food manufacturing industry. [ABSTRACT FROM AUTHOR]

Ultrasound-assisted extraction (UAE) is a promising technique for isolating bioactive compounds from plant materials, as ultrasound disrupts cell walls through mechanical effects, and facilitates the release of cellular constituents. Despite its growing popularity, the efficiency of UAE for extracting limonin from citrus peels, particularly Wenling Gaocheng, remains understudied. In the present work, we systematically explored the influence of various operational parameters on the yield of limonin using UAE. The limonin content was quantified using a Waters 2695 HPLC system coupled with a Waters 2487 UV detector. The effects of six parameters on the UAE of limonin from Wenling Gaocheng peels was examined through single-factor experiments. We observed that the limonin yield first increased and then decreased with decreasing particle size. The yield also increased as the solvent/material ratio increased, but the rate of increase decreased. Among the tested solvent concentrations, 70% ethanol was found to be the most effective. The yield significantly improved with increasing ultrasonic intensity up to 0.2556 W/cm² before decreasing sharply. Between 20 and 70°C, the yield fluctuated slightly, and a gradual improvement was observed with longer extraction times. Medicinal and aromatic plants are valued for their natural compounds, being used in alternative treatments in medicine, as raw materials for various industries or products. Essential oils are obtained through the processing of some of these species. Their popularity has significantly increased in the last decade, especially due to studies on their beneficial effects on human health. The extraction method is crucial for the quality and function of essential oils, alongside other influencing factors. This paper presents a brief overview of the use of the DIC (Instant Controlled Pressure Drop) process as a "green" method for the extraction of essential oils from medicinal and aromatic plants. Abstract Psyllium mucilage was extracted from Plantago ovata Forsk and functional properties were investigated. Concerning this objective, Response Surface Methodology based on a 4-factor 3-level Box-Behnken design was used. The independent variables were as follows: extraction temperature (50-80 °C), extraction time (60-120 min), water-to- Psyllium seed ratio (50/1-100/1), and water pH (4-10). On the other hand, the dependent variables were decided as EY (extraction yield), SC (swelling capacity), EC (emulsion capacity), and ES (emulsion stability). Optimum extraction conditions based on numerical optimization were 79.99 °C extraction temperature, 60.02 min extraction time, 99.99:1 water-to-seed ratio, and 7.38 water pH. Within these optimum conditions, 29.54% EY, 25.47 mL g-1 SC, 68.39% EC, and 76.61% ES were predicted. The results indicated that Psyllium mucilage can be used as a promising additive as a stabilizing agent in food formulations.

Objective: This study aimed to optimize the pectin extraction yield from immature fruits of Guanxi pomelo by L. plantarum fermentation and analysis of the properties of pectin extracted by L.s plantarum fermentation. Methods: Taking the extraction yield of pectin as the index, orthogonal test was used to optimize the process conditions of fermentation of immature fruits of Guanxi pomelo by L. plantarum, and the content of galacturonic acid, degree of esterification, protein, water-holding capacity, oil-holding capacity, emulsifying activity and emulsion stability of pectin were determined. Results: The optimal conditions for fermentation of immature fruits of Guanxi pomelo by L. plantarum were as follows: fermentation temperature of 37 ℃, L. plantarum fermentation inoculum of 14%, liquid-solid ratio of 25∶1 (mL/g), and fermentation time of 12 h. Under the control of these conditions, the extraction yield of pectin was 11.60%, and the content of galacturonic acid; the degree of esterification, protein, water-holding capacity, oil-holding capacity, emulsifying activity and emulsion stabilities were about 26.13%, 68.58%, 0.53 g/g, 7.01 g/g, 14.33% and 33%, respectively. Conclusion: The extraction yield of pectin extracted by fermentation of L. plantarum was similar to that of the acid process, and the obtained pectin was highly esterified pectin with good application properties.

Background: M. oleifera is an enriched plant with a variety of rich ingredients that play a very important part in the human diet. Thus, scientists have great interest in assessing the medicinal value of the plant to promote the preparation of new and advanced drugs. Hence, the preparation of plant extracts for experimental purposes is an initial step and key to achieving a quality research outcome. Materials and Methods: The primary objective of this study was to evaluate the screening of M. oleifera bioactive compounds using various solvents (methanol, ethanol, acetone, petroleum ether, chloroform, and water) in the extraction procedure and determine the quality and quantity of bioactive constituents. Results: The quantitative analyses showed that crude leaf, pod, and bark extracts revealed the presence of alkaloids, phenols, terpenoids, proteins, and carbohydrates in all extracts except for petroleum ether extract. Qualitative analysis of the detected phytochemicals reveals the highest concentrations were found in leaf extracts, where a high extraction yield was recorded in the aqueous extract. Conclusion: This study reveals that the presence or absence of particular phytochemicals is determined by the polarity of the solvents used for extraction. Also, the justification for M. oleifera to contain rich phytochemicals, including alkaloids, flavonoids, phenolics, terpenoids, tannins, and saponins, that are known to have pharmacological properties, was validated, and they can be explored for biological potential. Phaleria macrocarpa (Scheff.) Boerl., locally known as ‘Mahkota Dewa’, is a fruit commonly found in Malaysia. It demonstrates high potential for various biological activities, including antioxidant, anti-inflammatory, antibacterial, and anticancer properties. Nevertheless, there is a lack of research on the effect of essential oil extraction using different solvents on the antibacterial and antioxidant activities of this seed’s fruit. This study aims to investigate the extraction yield, volatile components, antibacterial, and antioxidant activities of essential oil extracted from P. macrocarpa seeds using ethyl acetate, hexane, and petroleum ether as solvents. Gas chromatography-mass spectrometry was used to analyse the volatile components, while the antibacterial activity was assessed against Listeria innocua and Serratia marcescens. The antioxidant activity was evaluated using total phenolic content (TPC), total flavonoid content (TFC), and the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay. Hexane showed the highest extraction yield (51.87 ± 0.21%), followed by petroleum ether (44.40 ± 0.14%) and ethyl acetate (27.08 ± 0.10%). The essential oils extracted with ethyl acetate, hexane, and petroleum ether contained 24, 26, and 21 volatile components, respectively. The major component, 1-(ethenylthio)-octane was found in concentrations ranging from 42.371 to 54.863 ppm, followed by 2,2-dimethyl-5-(3-methyl-2-oxiranyl) cyclohexanone, with concentrations between 30.771 and 40.783 ppm. The essential oil exhibited moderate antibacterial activity, as indicated by the zone of inhibition against L. innocua (8.00 ± 1.30 mm to 11.00 ± 1.40 mm) and S. marcescens (9.00 ± 1.00 mm to 14.00 ± 1.20 mm) compared to the positive control. Additionally, the essential oil demonstrated good antioxidant activity, with TPC, TFC, and DPPH values for hexane (65.56 ± 0.02 mg GAE/g, 4.46 ± 0.17 mg RE/g, 223.78 ± 29.69 μg/mL), ethyl acetate (72.99 ± 0.05 mg GAE/g, 19.47 ± 0.41 mg RE/g, 294.02 ± 12.70 μg/mL), and petroleum ether (70.29 ± 0.07 mg GAE/g, 4.83 ± 0.05 mg RE/g, 803.64 ± 27.76 μg/mL), respectively. Nonetheless, correlation analysis revealed a moderate relationship between DPPH and TFC. The essential oils from P. macrocarpa seeds demonstrates strong natural antibacterial and antioxidant potential, with hexane as the most efficient solvent. Exploring compounds like 1-(ethenylthio)-octane and 2,2- dimethyl-5-(3-methyl-2-oxiranyl)cyclohexanone may reveal medicinal applications and lead to innovative remedies. [ABSTRACT FROM AUTHOR]

The potential of Sphagnum sericeum moss extracts as an alternative treatment for childhood acute Lymphoblastic leukaemia (ALL) was investigated in this study. Aqueous, methanol, and chloroform Sphagnum sericeum extracts were analyzed for phytochemical composition and cytotoxic effects on T-cell ALL lines (Jurkat and CCL-119). Results show that the S. sericeum aqueous extract (SSAE) yielded the highest percentage, followed by the methanol extract (SSME) and the chloroform extract (SSCE). Phytochemical analysis identified alkaloids, glycosides, and terpenoids in all extracts. SSME demonstrated significant cytotoxicity towards ALL cells, with the lowest IC50 values observed at 72 hours. Notably, SSME induced morphological changes, including blebbing and cell fragmentation after 6 h, indicative of apoptosis and cell destruction. These findings suggest SSME's potential as a natural agent for treating paediatric ALL, offering a novel approach in cancer research and contributing insights into the phytochemical compounds and cytotoxic effects of S. sericeum extracts on T-ALL cell lines. Indigo quality is determined by its indigotin content. Another quality indicator is colour. For an evaluation of species, indigo samples from Indigofera tinctoria, Indigofera suffruticosa, Indigofera arrecta, Persicaria tinctoria, Strobilanthes cusia and Wrightia laevis cultivated in Austria and China were visually classified and analysed spectrophotometrically and using a L*a*b* measuring device. In addition to a standardised hot-extraction method without lime, some samples were extracted simulating traditional methods at ambient temperatures using lime. The highest indigotin contents were achieved with Indigofera arrecta (55%, Austria) and Strobilanthes cusia (56%, China). There were no statistically significant differences between the indigo extraction yields of the species cultivated in Austria, but Indigofera arrecta and Persicaria tinctoria had statistically significantly higher indigotin extraction yields than Indigofera tinctoria and Indigofera suffruticosa. From the species extracted in China, Strobilanthes cusia showed higher values in all parameters than Indigofera tinctoria, Indigofera suffruticosa and Wrightia laevis. Compared with the standardised method, the method simulating local practice yielded more indigo but had a lower indigotin content; the indigotin extraction yields did not differ greatly. L*a*b* values enabled precise estimations of the indigotin content, making it an interesting option for quality control, as inexpensive, easy-to-handle L*a*b* measuring instruments have become available. The effects of ultrasonic power (0, 150, 300, 450, and 600 W) on the extraction yield and the structure and rheological properties of pepsin-soluble collagen (PSC) from albacore skin were investigated. Compared with the conventional pepsin extraction method, ultrasonic treatment (UPSC) significantly increased the extraction yield of collagen from albacore skin, with a maximum increase of 8.56%. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis revealed that peptides of low molecular weight were produced when the ultrasonic power exceeded 300 W. Meanwhile, secondary structure, tertiary structure, and X-ray diffraction analyses showed that the original triple helix structure of collagen was intact after the ultrasonic treatment. The collagen solutions extracted under different ultrasonic powers had significant effects on the dynamic frequency sweep, but a steady shear test suggested that the collagen extracted at 150 W had the best viscosity. These results indicate that an ultrasonic power between 150 and 300 W can improve not only the extraction yield of natural collagen, but also the rheological properties of the collagen solution without compromising the triple helix structure. Ultrasound-assisted extraction (UAE) is an innovative process for recovering valuable substances and compounds from plants and various biomaterials. This technology holds promise for resource recovery while maintaining the quality of the extracted products. The review comprehensively discusses UAE’s mechanism, applications, advantages, and limitations, focusing on extracting essential oils (EOs) from diverse terrestrial plant materials. These oils exhibit preservation, flavor enhancement, antimicrobial action, antioxidant effects, and anti-inflammatory benefits due to the diverse range of specific compounds in their composition. Conventional extraction techniques have been traditionally employed, and their limitations have prompted the introduction of novel extraction methods. Therefore, the review emphasizes that the use of UAE, alone or in combination with other cutting-edge technologies, can enhance the extraction of EOs. By promoting resource recovery, reduced energy consumption, and minimal solvent use, UAE paves the way for a more sustainable approach to harnessing the valuable properties of EOs. With its diverse applications in food, pharmaceuticals, and other industries, further research into UAE and its synergies with other cutting-edge technologies is required to unlock its full potential in sustainable resource recovery and product quality preservation.

The effect of microwave pretreatment and moisture levels of Camelina sativa seeds on the quality of extracted oil by cold press was investigated. The seed moistures were adjusted to 2.5%, 5.0%, 7.5%, and 10.0% and pretreated with microwaves for 0, 1, 2, and 3 min. Microwave pretreatment (3 min) of the seeds with 2.5% moisture increased the oil extraction yield by ∼11% compared to the control sample. The highest amount of acidity (0.564 g FFA·g−1 oil), peroxide value (2.4 meq O2·kg−1 oil), carotenoid (5.26 mg·kg−1 oil), and browning index (0.710) were found in the oil extracted from seeds with 10% moisture and 3 min microwave pretreatment. The total phenolic compound was increased by microwave pretreatment but was mitigated by the seed moisture content, and the highest amount (208.24 mg caffeic acid·100 g−1 oil) was observed at 3 min microwave pretreatment of the seeds with 2.5% moisture. Chlorophyll content decreased by both microwave pretreatment and seed moisture content in camelina oil. Generally, the fatty acid composition of the extracted oils was not affected by the seed pretreatments. In conclusion, pretreatment of the camelina seeds before oil extraction is suggested to obtain a high oil extraction yield with a good quality oil.

Nitrohumic acids (NHAs) are considered as promising slow-release N fertilizers. In this study, NHA was produced from six organic feedstocks, including coal, leonardite, municipal waste compost, sewage sludge, and apple and beech biochars via nitration using nitric acid. The nitration process was conducted in two ways: (i) after humic acid (HA) extraction (NHAD), and (ii) before HA extraction (NHAID). The prepared NHAs were then characterized using FT-IR and CHNS analysis. Additionally, N mineralization of NHAs was investigated in a sandy loam soil sample. The FT-IR spectra showed that both methods of nitration loaded nitro (NO2) groups into the HA structure. However, the CHNS analysis indicated the highest rate of N increase for NHAD (106%) and NHAID (113%) extracted from leonardite. Moreover, on average, the total acidity and carboxylic groups of the HAs increased by 7.5% and 14.5% after the nitration processes, respectively. The highest extraction yields of NHAD (26.1%) and NHAID (42.1%) were also obtained from leonardite. Although the extraction yield of NHAID was on average two times higher than that of NHAD, NHAD indicated a higher soil N availability (1.4 times). Passion fruit seed (PS) and rambutan seed (RS) contain sizable amounts of phenolic compounds (PCs) with bioactive compounds including antioxidant activity (AOA). The aim of this study was to optimize the extraction of PCs from PS and RS using response surface methodology (RSM). Preliminary results indicated that the solid to liquid (S-L) ratio of 1:40 (w/v) showed the highest yield (EY) for PS and RS, with values of 11.59 ± 0.08 and 14.55 ± 1.67 g/100 g samples, respectively. The S-L ratio of 1:30 (w/v) was selected as an economic logistic. The predicted response compared to experimental data for EY, EPC, DPPH, ABTS and FRAP of the extracts were 12.41 % (12.68 %), 715 (707), 85 (81), 539 (522) and 107 (160) mg GAE/100 g dry weight for RS and 13.68 % (14.41 %), 3141 (3235), 1327 (1434), 5793 (5564) and 1567 (1509) mg GAE/100 g dry weight for PS, respectively. Phaleria macrocarpa (Scheff.) Boerl. or 'Mahkota Dewa' is a popular plant found in Malaysia as it is a valuable source of phytochemicals and therapeutic properties. Drying is an essential step in the storage of P. macrocarpa fruits at an industrial level to ensure their availability for a prolonged shelf life as well as preserving their bioactive compounds. Hence, this study evaluates the effect of different temperatures on the drying kinetics, extraction yield, phenolics, flavonoids, and antioxidant activity of P. macrocarpa fruits. The oven-drying process was carried out in this study at temperatures of 40 °C, 50 °C, 60 °C, 70 °C, and 80 °C. Six thin-layer drying models (i.e., Lewis, Page, Henderson and Pabis, two-term exponential, Logarithmic, and Midilli and Kucuk models) were evaluated to study the behaviour of oven-dried P. macrocarpa fruits based on the coefficient of determination ( R 2 ), root mean square error ( R M S E ), and chi-square ( χ 2 ). The quality of the oven-dried P. macrocarpa fruits was determined based on their extraction yield, total phenolic content (TPC), total flavonoid content (TFC), and antioxidant activity (2,2-diphenyl-1-picrylhydrazyl) using ultrasound-assisted extraction. The results showed that the time for moisture removal correspondingly increased in the oven-dried P. macrocarpa fruits. Apparently, the Midilli and Kucuk model is the most appropriate model to describe the drying process. The range of effective moisture diffusivity was 1.22 × 10 − 8 to 4.86 × 10 − 8   m 2 / s , and the activation energy was 32.33 kJ/mol. The oven-dried P. macrocarpa fruits resulted in the highest extraction yield (33.99 ± 0.05%), TPC (55.39 ± 0.03 mg GAE/g), TFC (15.47 ± 0.00 mg RE/g), and DPPH inhibition activity (84.49 ± 0.02%) at 60 °C based on the significant difference (p < 0.05). A strong correlation was seen between the antioxidant activity, TPC, and TFC in the oven-dried P. macrocarpa fruits. The current study suggests that the oven-drying method improved the TPC, TFC, and antioxidant activity of the P. macrocarpa fruits, which can be used to produce functional ingredients in foods and nutraceuticals. Lychee seeds (LS) and longan seeds (LoS) are excellent sources of phenolic compounds (PCs) with strong antioxidant activity (AOA). The aim of this study was to optimize the extraction conditions regarding extraction yield (EY), extractable phenolic compound (EPC), and AOA from LS and LoS using surface response methodology (RSM). Solvent concentration, extraction temperature, time, and solid to liquid ratio were optimized using RSM. Increasing the solid to solvent ratio from 1:05 to 1:40 (w/v), increased EY for LoS, however, EY did not change from 1:20 to 1:40 for LS. Solid–liquid ratio 1:20 was chosen for this study. Increasing the quantity of solvent leads to higher EPC and FRAP. The results showed that LoS exhibited higher AOA than LS measured as DPPH, ABTS, and FRAP, respectively. Ethanol concentrations and temperatures significantly (p < 0.05) affect EY, EPC, and AOA. The results (R2 > 0.85) demonstrated a good fit to the suggested models and a strong correlation between the extraction conditions and the phenolic antioxidant responses. The ethanol concentrations of 41 and 53%, temperatures of 51 and 58 °C, and the corresponding times of 139 and 220 min were the optimal conditions that maximized the EY, EPC, and AOA from LS and LoS. The optimal process conditions for solvent-free microwave extraction (SFME) of essential oils from Cinnamomum longepaniculatum deciduous leaves after moisture conditioning were established by response surface methodology (RSM). A Box-Behnken design (BBD) was applied to evaluate the effects of three independent variables: moisture content (A: 54%–74%), microwave power (B: 300–500 ​W) and microwave time (C: 20–40 ​min), on the extraction yield of essential oil. The compounds of the essential oils obtained by SFME, hydro-distillation and microwave-assisted hydro-distillation (MADE) were identified by gas chromatography-mass spectrometry (GC-MS), and the total lipids of C. longepaniculatum fresh leaves and deciduous leaves were analyzed. The correlation analysis of the response regression model indicated that quadratic polynomial model could be employed to optimize the extraction of essential oil. The optimal extraction condition was A: 58%, B: 400 ​W and C: 28 ​min. In the optimal condition, the maximum extraction yield was 4.475 mL/100 ​g dw and higher than that by MADE. The main compound of the essential oil was eucalyptol (63.54%), and total oxygenated compounds was 78.95%, mainly caused by SFME and the metabolism of endophytic bacteria with decreasing content of phospholipids and fatty acids. Analysis of variance under the extraction condition illustrated high fitness of the model and the success of RSM for optimizing and reflecting the expected process condition. SFME combined with moisture regulation was an effective method for extracting essential oil from C. longepaniculatum deciduous leaves.

Mycosporine-like amino acids (MAAs) are natural ultraviolet-absorbing compounds found in microalgae and macroalgae. MAA content changes seasonally and in response to environmental factors. We previously investigated MAAs from the red alga dulse (Devaleraea inkyuleei, formerly Palmaria palmata in Japan) in Usujiri, Hokkaido, Japan, from 2019 to 2020. At that time, some factors affecting MAA content were still unclear. In this study, we investigated MAA variation during the period from January to June 2021, and evaluated new methods of MAA extraction from dulse. We recorded a maximum MAA extraction yield (7.03 µmol/g dry weight) on 25 March 2021. Over the course of our three years of investigations from 2019 to 2021, we found that dulse was most suitable for MAA preparation from the middle of February to late April. In the later work reported in this paper, we improved our extraction method by using a lower-risk organic solvent (ethanol) rather than methanol. In addition, we evaluated MAA extraction using different levels of ethanol concentration (25, 50, and 99%) and different extraction times (2, 6, and 24 h). We found that extraction with 25% ethanol for 24 h increased MAA content by a factor of 3.2, compared with our previous extraction method. In summary, we determined the most suitable sampling period for Usujiri dulse, to extract the highest content of MAAs. We also improved the effectiveness of the extraction process.

Andrographis paniculata is widely used as a medicinal plant in many countries and andrographolide is the major bioactive compound extracted from A. paniculata leaf. This study purposely to optimize statistical andrographolide extraction using the accelerated solvent extraction (ASE) technique. The Box Behnken design (BBD) was chosen to determine the optimum ASE conditions for the extraction temperature (ºC), cycle number, and extraction time (min) to achieve the highest yield of andrographolide. The optimum ASE conditions were identified as: extraction temperature of 60 ºC, using 3 cycles and 5 min extraction time, with maximum conversion yield as high as 335.2 ± 0.2 mg/g determined by High Performance Liquid Chromatography (HPLC) with the squared correlation coefficients (R2) of 0.97. The findings revealed the ASE method significantly enhanced andrographolide extraction and agreed closely with the predicted value at 337.5 mg/g. Andrographolide was isolated by preparative HPLC technique. A. paniculata leaf extract and extracted andrographolide displayed moderate radical scavenging activity in 2, 2−Diphenyl−1−picrylhydrazyl hydrate (DPPH) assay with IC50 of 0.883 ± 1.597 mg/ml and 0.514 ± 0.285 mg/ml respectively as IC50 for ascorbic acid was 0.048 ± 0.004 mg/ml. A. paniculata extract and andrographolide inhibited the tyrosinase enzyme with IC50 of 0.749 ± 0.293 μg/ml and IC50 of 2.441 ± 2.026 μg/ml indicated stronger tyrosinase inhibition abilities than kojic acid, IC50 of 19.985 ± 0.557 μg/ml. These results suggest that A. paniculata leaf extract and andrographolide have greater potential as sources of biochemical compounds that can be used as skin depigmentation solutions.

The extraction of essential oil from pine needles was optimized by response surface methodology, and the following optimal conditions were obtained: a fresh pine needle of 100 g, an extraction time of 2 h, a water dosage of 850 mL, and a NaCl concentration of 2.50%. The extraction yield of essential oil was 0.611% under optimal conditions, which was extremely close to the predicted value. The extraction yields of essential oil from needles of 12 common pines in Guangxi were compared. The contents of essential oil in needles of Pinus massoniana, Pinus crassicorticea, and Pinus taeda were relatively higher than other pines. A total of 44 chemical components were identified by GC-MS, including 12 monoterpenes, 14 sesquiterpenes, and 12 alcohols. The chemical components of essential oil from different pines have their own features, and it is speculated that they have good and diversified application potential in the fields of medicine, food, spices, and so on. The chemical compositions of essential oil with high extraction yield have similar characteristics. This phenomenon can be used as the basis and means for the selection of pines with high content of essential oil in needles.

Striped mullet (Mugil cephalus) roe is used for the production of high nutritional and added‐value delicacies. Its lipid fraction is rich in polyunsaturated fatty acids (PUFA) and bioactive compounds. This study examines scalable oil extraction methods for mullet roe oil extraction. Namely, solvent extraction (SE) using ethanol in two different temperatures, supercritical fluid extraction (SFE) using CO2‐ethanol mixture (SFE‐E) in two different temperatures, expeller oil press (EP) extraction, expeller oil press combined with ethanol extraction (EP‐SE) and wet reduction (WR) are examined. The methods are evaluated with regard to the oil yield and recovery, the oil oxidation and the composition in fatty acids, and polar compounds and unsaponifiable matter. EP‐SE and SE provide the highest oil recovery for tested extraction temperatures (76% and 65% respectively), followed by SFE‐E (46%) and EP (36%). Extracted oils present high PUFA content (28.5–33.9%). The type of extraction process and the process variables affect oil oxidation as well as the concentration of polar compounds and unsaponifiable matter. In terms of oxidation levels, 85% of the extracted oil samples were within the limits set by the Codex Alimentarius Commission. The potential of the examined methods for industrial mullet roe oil production is discussed. Practical applications: Oil rich in polyunsaturated fatty acids was extracted from stripped mullet roe. The work proposes several scalable extraction methods using mild conditions which could be applied to obtain edible and high nutritional value mullet roe oil with high recovery reaching up to 76%. The same methods could be employed also for mullet roe by‐products. The obtained results improve the knowledge regarding the potential of roe valorization for oil extraction as well as the effect of the extraction method on the oil yield, main composition features and the quality characteristics of oil extracted by mullet roe. In extra virgin olive oil production, it is essential to obtain a well-prepared olive paste which allows not only the extraction of the oil drops from the olives, but also the achievement of a high-quality oil while maintaining high yields. This work addresses the problem of determining the effect of three crushing machines on the viscosity of the olive paste: a hammer crusher, a disk crusher and a de-stoner were tested. The tests were repeated on both the paste leaving each machine and the paste to which water was added; this was done with the main aim of considering the different dilutions of the paste while entering the decanter. A power law and the Zhang and Evans model were used to analyse the rheological behaviour of the paste. The experimental results allow validation of the two models with a high (more than 0.9) coefficient of determination between experimental and numerical data. The results also show that the pastes obtained with the two classic crushing methods (hammers and disks) are almost identical, with a packing factor of about 17.9% and 18.6%, respectively. Conversely, the paste obtained with the de-stoner entails higher viscosity values and a smaller solid packing factor, of about 2.8%. At 30% dilution with water, the volume of the solid concentration dropped to about 11.6% for the hammer and disc crushers, while for the de-stoner it only reached 1.8%. This behaviour is also reflected in the evaluation of yields, which were 6% lower with the de-stoner. No significant differences regarding the legal parameters of oil quality were found using the three different crushing systems. Finally, this paper establishes some fundamental pillars in the research for an optimal model for identifying the rheological behaviour of the paste as a function of the crusher used. This research aimed to model the kinetic of ohmic heating extraction method of radish (Raphanus sativus L.) leaf extract in order to understand and optimize the extraction process .The effect of extraction parameters on extraction yield, total phenol content and DPPH scavenging activity was discussed, at a gradient voltage of 10 -30 V/cm, temperature of 30 -60°C, solvent ration (Ethanol to water) of 30 -100% and extraction time of 10 -30 min. Different empirical models such as first order, adsorption and sigmoid models presented to predict the kinetics of mass transfer without taking into account the underlying phenomena. Results indicated that the effect of gradient voltage, extraction time and solvent ratio (ethanol to water) on the extraction yield, total phenolic content and DPPH scavenging activity were significant (p˂0.05). Although increasing extraction temperature significantly resulted in a lower total phenolic content and DPPH scavenging activity of extract (p<0.05), however, this parameter no significant effect the extraction yield (p>0.05). Under optimum conditions, the experimental extraction yield and total phenol content were close to the predicted values calculated from the quadratic response surface model.Based on kinetics modeling that has been done, it can be said that the sigmoid kinetic model more can represent well the experimental results of radish leaf extract by the ohmic heating method when compared with the first -order and adsorption kinetic models. Indigo quality is determined by its indigotin content. Another quality indicator is colour. For an evaluation of species, indigo samples from Indigofera tinctoria, Indigofera suffruticosa, Indigofera arrecta, Persicaria tinctoria, Strobilanthes cusia and Wrightia laevis cultivated in Austria and China were visually classified and analysed spectrophotometrically and using a L*a*b* measuring device. In addition to a standardised hot-extraction method without lime, some samples were extracted simulating traditional methods at ambient temperatures using lime. The highest indigotin contents were achieved with Indigofera arrecta (55%, Austria) and Strobilanthes cusia (56%, China). There were no statistically significant differences between the indigo extraction yields of the species cultivated in Austria, but Indigofera arrecta and Persicaria tinctoria had statistically significantly higher indigotin extraction yields than Indigofera tinctoria and Indigofera suffruticosa. From the species extracted in China, Strobilanthes cusia showed higher values in all parameters than Indigofera tinctoria, Indigofera suffruticosa and Wrightia laevis. Compared with the standardised method, the method simulating local practice yielded more indigo but had a lower indigotin content; the indigotin extraction yields did not differ greatly. L*a*b* values enabled precise estimations of the indigotin content, making it an interesting option for quality control, as inexpensive, easy-to-handle L*a*b* measuring instruments have become available.

The effects of ultrasonic power (0, 150, 300, 450, and 600 W) on the extraction yield and the structure and rheological properties of pepsin-soluble collagen (PSC) from albacore skin were investigated. Compared with the conventional pepsin extraction method, ultrasonic treatment (UPSC) significantly increased the extraction yield of collagen from albacore skin, with a maximum increase of 8.56%. The sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis revealed that peptides of low molecular weight were produced when the ultrasonic power exceeded 300 W. Meanwhile, secondary structure, tertiary structure, and X-ray diffraction analyses showed that the original triple helix structure of collagen was intact after the ultrasonic treatment. Xylan is a polysaccharide of industrial interest and its solubilization demands a high quantity of chemicals. A combination of chemical and biological treatment was applied to solubilize xylan, reducing hydrogen peroxide charge. Coniophora puteana, Gloeophyllum trabeum, and Pleurotus ostreatus were grown on non-milled sugarcane bagasse up to four months. After microorganisms growth, the chemical solubilization of xylan was performed applying 3% and 6% (m/v) of hydrogen peroxide. Using 3% of hydrogen peroxide after the biological pretreatment resulted in 99.74% of the xylan solubilization after the fourth month to C. puteana growth, and 48.63% of the xylan solubilized after the first month to G. trabeum growth. The results demonstrated that the biological pretreatment reduced the recalcitrance of the sugarcane bagasse by changing its structure, allowing to solubilize a higher amount of xylan, and applying half of the chemical reagent contributing to process feasibility. The purpose of this study was to optimize the effect of particle size, time and ultrasonic intensity on the extraction of phenolic compounds and energy efficiency. Sonication was performed with a VC750 Sonics processor at the following amplitudes: 30, 50 and 70%, which corresponds to the ultrasonic intensity, respectively: 1.6; 5.1 and 8.6 W∙cm−2. The frequency of ultrasound was 20 kHz. Extraction was carried out in a 5 s on–10 s off pulse system. The content of polyphenols and their antioxidant activity were assayed by the spectrophotometric method. Response surface methodology (RMS) was used to optimize the investigated variables. On the basis of the developed model, the highest polyphenols yield was obtained under the following extraction conditions (particle size 0.65 mm, extraction time 13.14 min, ultrasound intensity 6.92 W∙cm−2, which resulted in a maximum value of 15.24 mg GAE∙g−1 dry matter of hemp. Taking into account the lowest unit energy consumption, the best conditions were obtained for particle size of approx. 0.5 mm, extraction time 7.7 min, and ultrasound intensity 1.8 W∙cm−2, which resulted in a phenolic yield of 10.14 mg GAE∙g−1 dry matter of hemp. The best agreement between values of optimization variables within investigated criteria was obtained for the variable particle size. The establishment of operation protocols for olive oil (OO) extraction at non-industrial scale is crucial for research purposes. Thus, the present study proposes a simple and cost-effective method for OO extraction at the laboratory scale (LS) level. To validate the proposed methodology, industrial OO extraction (IS) was performed in parallel, using the same cultivars 'Galega vulgar' (GV), 'Cobrançosa' (COB) and 'Arbequina' (ARB) collected from the same orchards, within the same period. Obtained results showed highest extractability for COB and ARB, of about 53%, while GAL showed 50%. All produced OO showed values lower than the regulated limits for the physicochemical parameters (acidity, K232, K268 and ΔK), classifying them as extra virgin OO (EVOO). Highest total phenolic content was observed for COB, with no significant differences (p-value > 0.05) between extraction methods. Regarding fatty acid composition, oleic acid (C18:1) showed the lowest percentage for ARB, with about 66% and 68%, for LS and IS, respectively, and the highest for GV with about 72% for both LS and IS. Microwave thermal processing is a promising technology to greatly improve product quality by achieving high‐temperature short‐time (HTST) processing for solid foods. And the non‐thermal effect of microwave fields on nutritional quality is a major public concern. To distinguish the non‐thermal effect of microwave fields, the thermal effect of HTST processing should be revealed first. The objective of this study was to investigate the effects of different HTST processing on quality of Pacific saury fillets using extracted fatty acids as the indicator. A self‐developed thermal processing system was used to conduct the HTST processing with different heating rate (5.48–18.30°C/min), maximum heating temperature (123, 133 °C), and thermal processing level (F0 = 3.0 min, 6.0 min). Results showed that the extraction coefficient of lipids and fatty acids decreased with increasing heating rates, which implied less thermal damage of fish tissue, while higher thermal processing level increased these extraction coefficients. However, higher maximum processing temperature caused serious thermal damage of fatty acids, especially for PUFAs. Furthermore, changing pattern of each fatty acid during different HTST processing was revealed, which provided fundamental data for designing microwave thermal processing and exploring microwave non‐thermal effects. Andrographis paniculata is widely used as a medicinal plant in many countries and andrographolide is the major bioactive compound extracted from A. paniculata leaf. This study purposely to optimize statistical andrographolide extraction using the accelerated solvent extraction (ASE) technique. The Box Behnken design (BBD) was chosen to determine the optimum ASE conditions for the extraction temperature (°C), cycle number, and extraction time (min) to achieve the highest yield of andrographolide. The optimum ASE conditions were identified as: extraction temperature of 60 °C, using 3 cycles and 5 min extraction time, with maximum conversion yield as high as 335.2 ± 0.2 mg/g determined by High Performance Liquid Chromatography (HPLC) with the squared correlation coefficients (R²) of 0.97. The findings revealed the ASE method significantly enhanced andrographolide extraction and agreed closely with the predicted value at 337.5 mg/g. Andrographolide was isolated by preparative HPLC technique. A. paniculata leaf extract and extracted andrographolide displayed moderate radical scavenging activity in 2, 2-Diphenyl-1-picrylhydrazyl hydrate (DPPH) assay with IC50 of 0.883 ± 1.597 mg/ml and 0.514 ± 0.285 mg/ml respectively as IC50 for ascorbic acid was 0.048 ± 0.004 mg/ml. A. paniculata extract and andrographolide inhibited the tyrosinase enzyme with IC50 of 0.749 ± 0.293 µg/ml and IC50 of 2.441 ± 2.026 µg/ml indicated stronger tyrosinase inhibition abilities than kojic acid, IC50 of 19.985 ± 0.557 µg/ml. These results suggest that A. paniculata leaf extract and andrographolide have greater potential as sources of biochemical compounds that can be used as skin depigmentation solutions. In general, hydrocolloids are a group of additives that play many roles in the food industry such as thickening, gelation and stabilization. Optimization of mucilage extraction from Sepestan fruit was performed using response surface methodology and Box–Benken statistical design. Extraction temperature in the range of 45–85 °C, pH in the range of 4–7, and liquid to solid ratio in the range of 5–15 ml/g as experimental variables were optimized base on different parameters including extraction yield, solubility, water absorption capacity, foam stability, oil absorption capacity, heat emulsion stability, turbidity and free radical scavenging. Results showed that the quadratic polynomial model was the most appropriate model for all the factors studied. Optimized conditions for extraction of Sepestan fruit mucilage (SFM) were determined as extraction temperature 57.49 °C, pH 4 and liquid to solid ratio 6.27 ml/g. In optimum conditions, the values of yield, solubility, water absorption capacity, foam stability, oil absorption capacity, heat emulsion stability, turbidity and antioxidant activity of SFM were determined as 8.83%, 27.30%, 28%, 0.04%, 28%, 443.4%, 0.13% and 36.83%, respectively. In a general conclusion, due to the reasonable yield of mucilage extraction and its pragmatic properties such as proper oil absorption, heat emulsion stability and antioxidant potential, its utilization may be recommended in food and pharmaceutical industry. Introduction: Excessive demand, environmental problems, and shortages in market-leader countries have led the citrus (essential) oil market price to drift to unprecedented high levels with negative implications for citrus oil-dependent secondary industries. However, the high price conditions have promoted market incentives for the incorporation of new small-scale suppliers as a short-term supply solution for the market. Essential oil chemical extraction via steam distillation is a valuable option for these new suppliers at a lab and small-scale production level. Nevertheless, mass-scaling production requires prediction tools for better large-scale control of outputs.Methods: This study provides an intelligent model based on a multi-layer perceptron (MLP) artificial neural network (ANN) for developing a highly reliable numerical dependency between the chemical extraction output from essential oil steam distillation processes (output vector) and orange peel mass loading (input vector). In a data pool of 25 extraction experiments, 14 output–input pairs were the in training set, 6 in the testing set, and 5 cross-compared the model’s accuracy with traditional numerical approaches.Results and Discussion: After varying the number of nodes in the hidden layer, a 1–9–1 MLP topology best optimizes the statistical parameters (coefficient of determination (R2) and mean square error) of the testing set, achieving a precision of nearly 97.6%. Our model can capture non-linearity behavior when scaling-up production output for mass production processes, thus providing a viable answer for the scalability issue with a state-of-the-art computational tool for planning, management, and mass production of citrus essential oils.

IntroductionMorinda citrifolia L. (Noni) as an evergreen plant is a rich source of natural polysaccharides.ObjectiveThe present work aims to investigate the maturation-related changes in polysaccharides of Morinda citrifolia L. (Noni) at five stages of maturity (stages from the lowest to highest degree – 1, 2, 3, 4, and 5).MethodsThe chemical composition (carbohydrate, protein, uronic acid, and sulfate radical) of Noni polysaccharides was determined by different chemical assays. Ion chromatography system was used to analyze the monosaccharide composition, and the molecular weight was measured by HPGPC. The polysaccharides were also analyzed by FT-IR and their radical scavenging effect against DPPH, hydroxyl radicals and ABTS was evaluated. The UV-vis assay and gel electrophoresis assay were performed to investigate the DNA damage protective effect.ResultsResults indicated the significant effect of fruit maturities on the extraction yields, molecular weights, uronic acid contents, sugar levels, monosaccharide compositions and proportions, antioxidant capacities, and DNA protective effects of Noni polysaccharides. However, no fruit maturity stage had prominent impact on the sulfuric radical contents and preliminary structure characteristics. Noni polysaccharides extracted at stage 5 (N5) had the largest extraction yield (8.26 ± 0.14%), the highest sugar content (61.94 ± 1.86%) and the most potent scavenging effect on DPPH (IC50: 1.06 mg/mL) and ABTS (IC50: 1.22 mg/mL) radicals. The stronger DPPH and ABTS radical scavenging activities of N5 might be contributed by its higher content of fucose and rhamnose and smaller molecular weight. Noni polysaccharides extracted at stage 4 (N4) showed the highest uronic acid content (4.10 ± 0.12%), and the superior performance in scavenging hydroxyl radicals and protecting DNA. The greater hydroxyl radical scavenging effect of N4 might be attributed to its higher percentage of the low molecular weight counterpart. Moreover, the DNA protective effects of N4 displayed a positive correlation with its hydroxyl radical scavenging ability.ConclusionOverall, stage 4 and stage 5 could be ideal stages of fruit maturity aiming at high-quality Noni polysaccharides extraction. This study provided valuable information for the selection of suitable Noni polysaccharides to cater for various industrial applications.

Extraction of proteins from industrial residues is a potential source for animal feed. Alkaline extraction combined with isoelectric precipitation may be a useful method for isolating protein from brewer's spent grain (BSG), an abundant by‐product from the brewing industry. The objective of this study was to examine the effect of temperature (30, 45 and 60°C), time (30, 60, 120 and 180 minutes), agitation (shaking and stirring), precipitation salinity (0.5 M sodium chloride), isolation mode (freeze drying and precipitation), pre‐treatment (defatting and delignification) and repeated extraction on the protein yield and protein content from BSG. Generally, the protein content decreased while protein yield increased with increasing extraction temperatures. Yield and content were maximised after 30 minutes reaching the highest protein content at 30oC (54.8%) and the highest protein yield at 60oC (10.5%). Precipitation with 0.5 M sodium chloride reduced the protein content to 36.4% and the yield to 4.2%. Freeze drying combined with triple extraction yielded a protein content of 20‐25%, together with a relatively high protein yield (ca. 45‐50%). Delignification increased the protein content from 27% to 32% whereas defatting reduced the overall protein yield from 45% to 38%. Combining isoelectric precipitation and single extraction was associated with a higher protein content (40‐55%) but poor protein yields (5‐10%).
