The aim of the MOCAST+ (MOnitoring mass variations by Cold Atom Sensors and Time measures) project, which was carried out during the years 2020–2022, was the investigation of the performance of a gravity field mission based on the integration of atomic clocks and cold atom interferometers. The idea was that the combined observations of the two sensors would be beneficial for the detection and monitoring of geophysical phenomena which have an impact on the time-variable part of the Earth gravity field models. Several different mission scenarios were simulated, considering different satellite configurations such as a Gravity Recovery and Climate Experiment (GRACE)-class formation and a Bender-class formation with either two or three in-line satellites along each orbit. Moreover, different atomic species (rubidium and strontium), different inter-satellite distances, different noise power spectral densities, and different observation rates were taken into account. For the gravity field estimation from the simulated data, the space-wise approach was exploited. The results showed that, as it could be expected, the Bender configuration provides significantly better monthly gravity field solutions, as compared to a ‘nominal’ configuration with two or three satellites in a GRACE-class formation. In this way, and pushing the quantum sensors technology to its limits, it is in fact possible to obtain results which are comparable with those from GRACE at low harmonic degrees, and are better at higher degrees with positive effects in the detectability of localized time variable phenomena, as well as in the determination of the static gravity field at a higher maximum spherical harmonic degree than the one achieved by Gravity Field and Steady-State Ocean Circulation Explorer (of course considering an equivalent mission life-time).

A multicenter phase II study for assessing the efficacy and the toxicity of hypofractionated radiotherapy with SIB plus temozolomide in patients with glioblastoma was carried out by the Brain Study Group of the Italian Association of Radiation Oncology. Twenty-four patients with newly diagnosed glioblastoma belonging to Recursive Partitioning Analysis classes III and IV were enrolled. The prescribed dose was 52.5 Gy in 15 fractions of 3.5 Gy and 67.5 in 15 fractions of 4.5 Gy to the SIB volume. Dose constraints for the hypofractionated schedule were provided. Radiotherapy was associated with concomitant and sequential temozolomide. Median overall survival (OS) was 15.1 months, while median progression-free survival (PFS) was 8.6 months. Actuarial OS at 12 months was 65.6% ± 0.09, whereas actuarial PFS at 12 months was 41.2% ± 0.10. Status of methylation of MGMT promoter resulted to be a significant prognostic factor for OS. Radiotherapy-related acute toxicity was not relevant. Three patients (12.5%) had G3 myelotoxicity that required temozolomide temporary interruption or dose reduction during the chemotherapy. However, chemotherapy was not definitely discontinued for toxicity in any case. One patient out of 24 (4.2%) developed radionecrosis that required surgical resection with no evidence of disease in the surgical specimen. This trial confirms that hypofractionated radiotherapy with SIB and association with temozolomide may be a reasonable and feasible option for good prognosis patients with GBM.

Social cognitive function, involved in the perception, processing, and interpretation of social information, has been shown to be crucial for successful communication and interpersonal relationships, thereby significantly impacting mental health, well-being, and quality of life. In this regard, assessment of social cognition, mainly focusing on four key domains, such as theory of mind (ToM), emotional empathy, and social perception and behavior, has been increasingly evaluated in clinical settings, given the potential implications of impairments of these skills for therapeutic decision-making. With regard to neurodegenerative diseases (NDs), most disorders, characterized by variable disease phenotypes and progression, although similar for the unfavorable prognosis, are associated to impairments of social cognitive function, with consequent negative effects on patients' management. Specifically, in some NDs these deficits may represent core diagnostic criteria, such as for behavioral variant frontotemporal dementia (bvFTD), or may emerge during the disease course as critical aspects, such as for Parkinson's and Alzheimer's diseases. On this background, we aimed to revise the most updated evidence on the neurobiological hypotheses derived from network-based approaches, clinical manifestations, and assessment tools of social cognitive dysfunctions in NDs, also prospecting potential benefits on patients' well-being, quality of life, and outcome derived from potential therapeutic perspectives of these deficits. To evaluate the feasibility and response to palliative radiotherapy delivered with static ports of tomotherapy--TomoDirect (TD) in patients affected with painful bone metastases from solid tumors.A prospective cohort of 130 patients (185 osseous lesions) was treated between 2010 and 2013 with TD. Three fractionation schedules were employed according to clinical decision-making (3 Gy × 10; 4 Gy × 5; 8 Gy × 1). Pain response was investigated at 2 weeks and 2 months (for evaluable patients). The Numeric Rating Scale (NRS-11) was used to assess pain. Response rates to radiotherapy were calculated following the criteria of the International Bone Metastases Consensus Group (IBMCG), accounting for the use of concomitant analgesics (response: complete or partial; non-response: stable pain, pain progression or "other"). Analgesic consumption was recalculated into the daily oral morphine-equivalent dose (OMED).Most of the patients had 1-2 bone metastases (91); those with multiple lesions mostly had a metachronous presentation (60%). Synchronous lesions were mainly approached with multiple plans (63%). Most treatments employed 3-4 fields (77%). Treatment times ranged from 255 to 939 s depending on fractionation, fields, and target lesions number. At 2 weeks, the median self-reported worst pain decreased significantly as median oral morphine-equivalent dose regardless of fractionation used. The response rate according to the IBMCG-based response categories ranged from 45 to 55%. Pain relief duration seems (response at 2 months) slightly inferior with the single fraction approach, with a higher re-treatment rate. At 2 weeks, the median self-reported worst pain and OMED significantly decreased regardless of fractionation (response rate: 49-55%). Pain relief decreased at 2 months, especially for single fraction (higher re-treatment rate).TD is a valid option to deliver palliative radiotherapy for painful bone metastases from solid tumors.

To report the 1-year outcomes of a prospective phase II study on hypofractionated whole-breast intensity-modulated radiotherapy (IM-WBRT) with a simultaneous integrated boost (SIB) to the tumor bed delivered with static ports of tomotherapy (TomoDirect) (TD).A prospective cohort of 82 patients was enrolled between 2011 and 2012. Treatment schedule consisted of 45 Gy/20 fractions to the whole breast and 50 Gy/20 fractions to the surgical bed delivered concomitantly with TD over 4 weeks. A one-armed optimal two-stage Simon's design was selected to test the hypothesis that treatment modality under investigation would decrease acute skin toxicity over historical data using conventional fractionation and sequential boost. Primary endpoint was acute skin toxicity. Secondary endpoints included late toxicity, cosmesis, quality of life and local control.Median follow-up was 12 months (range 6-18). Maximum detected acute skin toxicity was G0 41 %; G1 53 %; G2 6 %; G31 %. With two G2-G3 acute skin toxicity events in the first stage and four in the second, the study fulfilled the requirements for the definition of the treatment approach under investigation as promising. Late skin toxicity was mild with noG2 events. Cosmesis was good/excellent in 91 % of patients and fair/poor in 9 %. Quality of life was preserved over time, with the exception of fatigue, which was transiently increased.Hypofractionated IM-WBRT with a SIB to the tumor bed delivered with TD provides consistent clinical results and it is able to reduce acute skin toxicity rate over conventionally fractionated and sequential boost tomotherapy-based IM-WBRT.

Although publications on circumnutation of the aerial parts of flowering plants are numerous and primarily from the time between Darwin (1880) and the 1950s, reports on circumnutation of roots are scarce. With the introduction of modern molecular biology techniques, many topics in the plant sciences have been revitalized; among these is root circumnutation. The most important research in this area has been done on Arabidopsis thaliana, which has roots that behave differently from those of many other plants; roots grown on inclined agar dishes produce a pattern of half waves slanted to one side. When grown instead on horizontally set dishes, the roots grow in loops or in tight right-handed coils that are characterized by a tight torsion to the left-hand. The roots of the few plants that differ from Arabidopsis and have been similarly tested do not present such patterns, because even if they circumnutate generally in a helical pattern, they subsequently straighten. Research on plants in space or on a clinostat has allowed the testing of these roots in a habitat lacking gravity or simulating the lack. Recently, molecular geneticists have started to connect various root behaviors to specific groups of genes. For example, anomalies in auxin responses caused by some genes can be overcome by complementation with wild-type genes. Such important studies contribute to understanding the mechanisms of growth and elongation, processes that are only superficially understood.

In this study, the genes of the Aux/IAA family were used as functional markers to characterise bud break stages in a white poplar clone. In the first experiment, under greenhouse conditions, the sprouting of repressed sylleptic buds was obtained by pruning the shoot tip during growing season. Buds were collected at 0, 6, 24 and 48 h after pruning for molecular analyses. A decrease in transcript level of IAA4 and IAA8 genes was observed in the first and second bud below the cut after 6 and 24 h, respectively. In the second experiment, bud break of post-dormant proleptic buds was induced by forcing in climatic chamber. The first 6 and the following 6 buds below the apical one were sampled every 48 h during forcing. Anatomical studies were also carried out on buds and plants were equipped with stem and bud radial growth sensors to check their swelling. In both experiments, gene expression patterns showed a decrease and a successive increase in expression of IAA4 and IAA8 genes during bud break. The transient down-regulation of these genes was observed only in buds that formed new branches. Thus, similar molecular mechanisms are involved in bud break of both sylleptic and proleptic buds.

Over the last fifteen years, an increasing number of plant scientists have become interested in the Arabidopsis root growth pattern, that is produced on the surface of an agar plate, inclined from the vertical. In this situation, the roots wave intensely and slant preferentially towards one side, showing torsions in the epidermal cell files alternately right-and left handed. In addition, the pattern switches to the formation of large or strict coils when the plate is set horizontally. After this finding, different hypotheses were advanced attempting to explain the forces that shape these patterns. These basically appear to be gravitropism, circumnutation and negative thigmotropism. With regard to the symmetry, the coils and the slanting in the wild-type are essentially right-handed, but mutants were also reported which show a left-handed symmetry, while some do not show a regular growth pattern at all. This review article discusses the earlier as well as the most recent findings on the topic, and investigates the possibility of describing the different mechanisms shaping the root growth patterns via unifying hypothesis.

Arabidopsis thaliana roots grown on a vertically set plate do not elongate straight down the gravitational vector, but by making waves and coils, and by conspicuously slanting towards the right-hand. This behaviour, in a previous paper, was ascribed to the simultaneous effect of three processes: circumnutation, positive gravitropism and negative thigmotropism. However, when the plants are grown on the Random Positioning Machine (RPM), in conditions that are believed to simulate space microgravitational conditions closely, the roots do not show the usual pattern. In the wild type, the roots make large loops to the right-hand side, whereas in the gravitropic and auxinic mutants aux1, eir1, rha1, they just move randomly around the initial direction. Therefore, if the movements made on the RPM are those produced by the exclusion of gravitropism and negative thigmotropism, as is apparent, the conclusion is that Arabidopsis roots are animated by a form of chiral circumnutation, that is lacking in the auxinic and gravitropic mutants aux1, eir1 and rha1. In addition, the 1 g condition appears to reduce the scatter among the circumnutating tracks produced by the roots of the wild types, but not among those of the mutants. Because there is a scarcity of literature regarding circumnutation in roots, it is not known how widely root chiral circumnutation is spread, but it is known that, in some previously studied species, just random nutations are observed. Two kinds of nutating movements seem to exist in plant roots and, whereas the random process does not seem to be connected with auxin physiology and transport, the chiral process appears to be connected in the same way as gravitropism is.

To report the 4-year outcomes of a consecutive series of anal cancer patients treated with concurrent chemo-radiation delivered with intensity-modulated radiotherapy (IMRT), employing a simultaneous integrated boost (SIB) approach.A consecutive series of 54 patients was enrolled between 2007 and 2013. Treatment schedule consisted of 50.4 Gy/28 fractions (1.8 Gy daily) to the gross tumor volume, while the elective nodal volumes were prescribed 42 Gy/28 fractions (1.5 Gy/daily) for patients having a cT2N0 disease. Patients with cT3-T4/N0-N3 tumors were prescribed 54 (T3) or 60 (T4) Gy/30 fractions (1.8-2 Gy daily) to the gross tumor volume; gross nodal volumes were prescribed 50.4 Gy/30 fr (1.68 Gy daily) if sized ≤ 3 cm or 54 Gy/30 fr (1.8 Gy daily) if3 cm; elective nodal regions were given 45 Gy/30 fractions (1.5 Gy daily). Chemotherapy was administered concurrently according to the Nigro's regimen. Primary endpoint was colostomy-free survival (CFS). Secondary endpoints were local control (LC), disease-free survival (DFS), cancer-specific survival (CSS), overall survival (OS), and toxicity profile.Median follow up was 32.6 months (range 12-84). The actuarial probability of being alive at 4 years without a colostomy (CFS) was 68.9% (95% CI: 50.3%-84.7%). Actuarial 4-year OS, CSS, DFS, and LC were 77.7% (95% CI: 60.7-88.1%), 81.5% (95% CI: 64%-91%), 65.5% (95% CI: 47.7%-78.5%), and 84.6% (95% CI: 71.6%-92%). Actuarial 4-year metastasis-free survival was 74.4% (95% CI: 55.5%-86.2%). Maximum detected acute toxicities were as follows: dermatologic -G3: 13%; GI-G3: 8%; GU-G3: 2%; anemia-G3: 2%; neutropenia-G3:11%; G4: 2%; thrombocytopenia- G3:2%. Four-year G2 chronic toxicity rates were 2.5% (95% CI: 3.6-16.4) for GU, 14.4% (95% CI: 7.1-28) for GI, 3.9% (95% CI: 1%-14.5%) for skin, and 4.2% (95% CI: 1.1-15.9) for genitalia.Our study shows the feasibility of IMRT in the combined modality treatment of anal cancer, with comparable results to the literature with respect to LC, sphincter preservation and survival. Acute toxicity is lower if compared to series employing standard techniques. Our results support the use of IMRT on a routine basis for the treatment of anal cancer.

We herein report on a case of synchronous bilateral breast cancer patient undergoing adjuvant intensity-modulated whole breast with static angle tomotherapy (TomoDirect). The patient was treated with a hypofractionated schedule employing a simultaneous integrated boost approach. Radiotherapy schedule was 45 Gy/20 fractions (2.25 Gy daily) to the bilateral whole breast and 50 Gy/20 fractions (2.5 Gy daily) to the 2 lumpectomy cavities. Treatment was delivered over 4 weeks. Dosimetric results were robust with consistent target coverage and adequate normal tissue avoidance. Treatment was generally well-tolerated and acute toxicity profile was mild. The present report highlights the promising clinical feasibility of TomoDirect for bilateral breast irradiation.

The apical hook that forms at the apex of most dicotyledonous seedlings during germination has in the past been variously described either as the consequence of positive gravitropism or as an autonomous process. Recently, however, emphasis has shifted more on the role of positive gravitropism as the inducer of this structure. To address the question on the nature and origin of the hook, experiments were performed on sunflower seedlings, not only under gravitational conditions but also by means of a clinostat, i.e. in the presence of multidirectionally compensated gravitational stimulation. The results of this work show that at least two processes are involved in the formation of a hook: positive gravitropism and an autonomous movement. Consequently, both Darwin, with his conviction that an innate process such as circumnutation was involved, and the supporters of positive gravitropism, were partly correct. To report the 2-year outcomes of whole breast intensity-modulated radiotherapy (IMRT) after conserving surgery for early breast cancer (EBC) delivered with static angle tomotherapy (TomoDirect) (TD).A prospective cohort of 120 EBC patients underwent whole breast IMRT with TD between 2010 and 2012. Radiation was delivered to a conventionally fractionated whole breast total dose of 50 Gy with TD, followed by a sequential conventionally fractionated tumor bed boost dose of 10-16 Gy with helical tomotherapy (HT). Clinical endpoints include acute and late toxicity, cosmesis, quality of life and local control.Median follow-up was 24 months (range 12-36 months); maximum detected acute skin toxicity was G0 22 %; G1 63 %; G2 12 % and G3 3 %. Predictors of acute dermatitis were as follows: volume of the whole breast minus boost volume receiving 105, 110 and 115 % of prescription dose, whole breast and boost volume, breast thickness and soft tissue thickness. Late skin toxicity was mild with noG2 events. Cosmesis was good/excellent in 91.7 % of patients and fair/poor in 8.3 %. Quality of life was preserved over time, but for fatigue, transiently increased.Adjuvant whole breast IMRT delivered sequentially with both TD and HT provides consistent clinical results. An observed unintended excessive dose outside the tumor bed might increase acute toxicity and eventually affect long-term clinical endpoints. The incorporation of the boost dose within the whole breast phase employing a simultaneous integrated boost (SIB) approach might mitigate this issue.

The administration of Cetuximab in combination with radiotherapy and chemotherapy has shown clear survival improvements within the locally advanced and the relapsed/metastatic settings respectively. These results have provided the clinical rational for the inclusion of Cetuximab into chemo-radiation regimens. Trials assessing the combination of Cetuximab with induction chemotherapy, concomitant chemo-radiotherapy or both are reviewed. Taken together, their results suggest that the addition of Cetuximab is promising in trials of induction chemotherapy, showing almost uniformly response rates higher than historical controls. In combination with concomitant hyperfractionated radiotherapy and Cisplatin the results of the RTOG 0522 trial do not suggest any benefit. However a positive effect cannot be excluded with other schedules. Although feasibility has been universally suggested, adding Cetuximab implies some toxicity enhancement. Single local and systemic toxicities are more frequent and supposedly the overall treatment intensity is increased. Moreover the drug-specific toxicities are potentially severe and deserve timely recognition and management.

Background This study investigates the use of TomoDirectTM 3DCRT for whole breast adjuvant radiotherapy (AWBRT) that represents a very attractive treatment opportunity, mainly for radiotherapy departments without conventional Linacs and only equipped with helical tomotherapy units. Methods Plans were created for 17 breast cancer patients using TomoDirect in 3DCRT and IMRT modality and field-in-field 3DCRT planning (FIF) and compared in terms of PTV coverage, overdosage, homogeneity, conformality and dose to OARs. The possibility to define patient-class solutions for TD-3DCRT employment was investigated, correlating OARs dose constraints to patient specific anatomic parameters. Results TD-3DCRT showed PTV coverage and homogeneity significantly higher than TD-IMRT and FIF. PTV conformality was significantly better for FIF, while no differences were found between TD-3DCRT and TD-IMRT. TD-3DCRT showed mean values of the OARs dosimetric endpoints significantly higher than TD-IMRT; with respect to FIF, TD-3DCRT showed values significantly higher for lung V20Gy, mean heart dose and V25Gy, while contralateral lung maximum dose and contralateral breast mean dose resulted significantly lower. The Central Lung Distance (CLD) and the maximal Heart Distance (HD) resulted as useful clinical tools to predict the opportunity to employ TD-3DCRT: positive correlations were found between CLD and both V20Gy and mean lung dose and between HD and both V25Gy and the mean heart dose. TD-3DCRT showed a significantly shorter mean beam-on time than TD-IMRT. Conclusions The present study showed that TD-3DCRT and TD-IMRT are two feasible and dosimetrically acceptable treatment approach for AWBRT, with an optimal PTV coverage and adequate OARs sparing. Some concerns might be raised in terms of dose to organs at risks if TD-3DCRT is applied to a general population. A correct patients clusterization according to simple quantitative anatomic measures, would help to correctly allocate patients to the appropriate treatment planning strategy in terms of target coverage, but also of normal tissue sparing.

Background Non-sinonasal neuroendocrine carcinomas (NSNECs) of the head and neck are considered an unfrequent clinico-pathological entity. Combined modality treatment represents an established therapeutic option for undifferentiated forms where distant metastasis is a common pattern of failure. Methods We report on a case of NSNEC treated with sequential chemo-radiation consisting of 6 cycles of cisplatin and etoposide followed by loco-regional radiation to the head and neck and simultaneous prophylactic cranial irradiation to prevent from intracranial spread, delivered with helical tomotherapy with the 'hippocampal avoidance' technique in order to reduce neuro-cognitive late effects. Results One year after the end of the whole combined modality approach, the patient achieved complete remission, with no treatment-related sub-acute and late effects. Conclusions The present report highlights the importance of multidisciplinary management for NSNECs of the head and neck, as the possibility to achieve substantial cure rates with mild side effects with modern radiotherapy techniques.

We report, in Arabidopsis thaliana plants, the effect of neutron irradiation on the transcription of a set of genes belonging to different physiological groups: auxin action, senescence, oxidative stress, and some aspects of photosynthesis. The results indicated that, in the wild-types, the effect on the ARF1, ARF2, and 19 genes was of down-regulation, whereas of the tested AUX/IAA only AUX/IAA7 showed up-regulation. Different results were obtained as regards the irradiation of the auxin transport mutants aux1 and eir1, because in these cases the ARF genes were up-regulated, whereas AUX/IAA7 was down-regulated in eir1. On the other hand, the senescence activated genes SAG12 and SAG13, and those connected to oxidative stress were up-regulated in the wild-type, but down-regulated in aux1. The gene CAB1, connected to photosynthesis, was also down-regulated in the wild-type, but up-regulated in aux1. Gene expression recovered in many cases almost to the initial condition in a time lapse of 24 hours, even though some effect persisted for a longer time. In particular, the state of juvenility of arf2 was extended by irradiation, whereas, in all the other cases, senescence was accelerated. The research indicates that through mutant selection or genetic engineering a true possibility exists of producing organism more suitable for life in space.

A new Arabidopsis mutant is characterized (rha1) that shows, in the roots, reduced right-handed slanting, reduced gravitropism and resistance to 2,4-D, TIBA, NPA and ethylene. It also shows reduced length in the shoot and root, reduced number of lateral roots and shorter siliques. The gene was cloned through TAIL-PCR and resulted in a HSF. Because none of the known gravitropic and auxinic mutants result from damage in a HSF, rha1 seems to belong to a new class of this group of mutants. Quantitative PCR analysis showed that the expression of the gene is increased by heat and cold shock, and by presence of 2,4-D in the media. Study of the expression through the GUS reporter gene revealed increased expression in clinostated and gravistimulated plants, but only in adult tissues, and not in the apical meristems of shoots and roots.

A new mutant of Arabidopsis named rha1 is characterized and the gene involved cloned. In roots, the mutant shows minimal right-handed slanting, reduced gravitropic response, notable resistance to 2,4-D, but scarce resistance to IAA and NAA. The roots also show a clear resistance to the auxin transport inhibitors TIBA and NPA, and to ethylene. Other characteristics are a reduced number of lateral roots and reduced size of shoot and root in the seedlings. The gene, cloned through TAIL-PCR, was found to be a heat-shock factor that maps on chromosome 5, close to and above the RFLP marker m61. The rha1 structure, mRNA, and translation product are reported. Since, so far, no other gravitropic mutant has been described as mutated in a heat-shock factor, rha1 belongs to a new group of mutants disturbed in slanting, gravitropism, and auxin physiology. As shown through the RT-PCR analyses of its expression, the gene retains the function connected with heat shock. If the characteristics connected with auxin physiology are considered, however, it is also likely that the gene, as a transcription factor, could be involved in root circumnutation, gravitropic response, and hormonal control of differentiation. Since GUS staining under the gene promoter was localized mainly in the mature tissues, rha1 does not seem to be involved in the first steps of gravitropism, but is rather related to the general response to auxin. The alterations in slanting (mainly due to reduced chiral circumnutation) and gravitropism lead to the supposition that the two processes may have, at least in part, common origins.

When primary Arabidopsis roots grow down a tilted agar plate, they do not elongate following the gravitational vector along a straight line, but instead they slant noticeably to the right-hand. This process is seen mostly in the ecotypes Wassilewskjia and Landsberg, whereas it is attenuated in the ecotype Columbia, and in some mutants is even inverted. The origin of the slanting of Arabidopsis roots, that evidently constitutes a form of chirality, has so far not been sufficiently clarified. In the present paper we describe it as the general result of the cumulative effects of positive gravitropism, circumnutation and a thigmotropic obstacle-avoiding movement, and in particular, as the consequence of the alternating movement of circumnutation of the root to the right and to the left of the gravitational vector. This movement, which does not appear symmetrical in its nature, since the waves made to the right-hand are complete whereas those made to the left-hand are reduced or aborted, appears to be the reason for the observed slanting. In addition, evidence is furnished supporting the hypothesis that the strong left-handed cell-files torsion, seen in right-handed coiling roots, is not the consequence of a primary process, but of an artifact, and is due to the adjustment of the three dimensional root circumnutating helix to the flat two dimensional agar surface.

When Arabidopsis seedlings are grown on a hard-agar plate, their primary roots show characteristic spiralling movements, apparent as waves, coils and torsions, together with a slanting toward the right-hand side. All these movements are believed to be the result of three different processes acting on the roots: circumnutation, positive gravitropism and negative thigmotropism. The basic movement of the roots is described as that of a growing right-handed helix, which, because of the root tip hitting the agar plate, is continuously switched from the right-hand to the left-hand of the growth direction, and vice versa. This movement also produces a slanting root-growth direction toward the right-hand because of the incomplete waves made by the right-handed root to the left-hand. By contrast, the torsions seen in the coils and waves are interpreted as artefacts that form as an adaptation of the three-dimensional root helix to the flat two-dimensional agar surface.

The gravitropic response of plants can be divided into three steps: detection, transduction, and asymmetric growth. Of these the second is that on which we have the smallest knowledge. Even though we have in fact some support for a redistribution of auxin as the agent of the graviresponse (Cholodny-Went hypothesis), we still lack of a precise knowledge about the many steps which precede and follow auxin redistribution. We thereore today dispose of the powerful new techniques of molecular genetics, that can help us in dissecting the pathway. By these techniques, as is known, already a number of gravitropic mutants have been isolated in the model plant Arabidopsis, and, in addition, some of the genes involved have been cloned. These findings already permit some speculation about the gravitropic pathway, even though we still are far from an understanding of the process. With the aim thus of obtaining the information we need to elucidate the gravitropic pathway, in the last two years, we performed a screening of some significant collections of Arabidopsis mutants, isolating individuals disturbed in root gravitropism, and starting to clone the involved genes.

We propose a new method to model a non-stationary and slowly variable phenomenon. The case of a one-dimensional stochastic process is presented. Particular care is taken in order to prove that the condition of positive definiteness of the covariance function is satisfied. The estimate of the covariance function is afterwards obtained by means of local estimates. The method has been tested on a set of simulated data.

A new Arabidopsis thaliana (L.) Heynh. root mutant, named clg1, was isolated from the Feldmann-Du Pont T-DNA insertional mutagenesis collection. It is characterized by primary roots that make early right-handed coils, show increased right-handed slanting, and reduced positive gravitropism. The mutant roots however did not exhibit increased resistance to IAA, but only a modest increment of resistance to the auxins 2,4 D, and NAA, and to the auxin transport inhibitors TIBA and NPA. By contrast, the mutant roots showed a notable resistance to plant hormone ethylene (given as ACC). clg1 appears to be new, since it complements the most known auxin and gravitropism mutants, maps to chromosome 5, and shows a phenotype largely different from that of the known ethylene mutants. The increased right-handed slanting (chirality) can possibly be a consequence of the reduced gravitropic response, since gravitropism and slanting are competitive growth-direction leading forces. The increased resistance to ethylene, seems to indicate that this phythormone plays a role in the gravitropic response of roots (as already proposed for shoots), and possibly in the regulation of the connected signal transduction pathway. The gene involved in the clg1 mutation, which is recessive, was mapped, as above reported, to chromosome 5, close to the visible marker tt3.

La matiere azotee soluble dans le tampon citrate 0,2 mol/L a pH 8,0 de deux echantillons de 14 et 38 mois de Grana Padano, un fromage italien a pâte dure, a ete diafiltree sur membranes a porosite de 3 et 10 kDa. Deux fractions ont ete recueillies, contenant des peptides de masse moleculaire inferieure a 3 kDa et superieure a 10 kDa. La fraction inferieure a 3 kDa a ete fractionnee par chromatographie liquide haute performance (CLHP) en phase inverse sur colonne C18 et les peptides identifies en combinant la spectrometrie de masse a bombardement rapide et la degradation d'Edman. La plupart des 91 peptides identifies dans cette fraction A du fromage de 14 mois provenaient des caseines α s1 (44 peptides) et β (32 peptides). La majorite de ces derniers presentaient une origine commune dans la region 1-105/107 de la caseine β. Trente peptides resultaient de la degradation du peptide α s1 -CN (fl-79) forme par action de la plasmine sur la caseine α s1 . Treize peptides provenaient de la caseine α s2 et un seul de la para-K-caseine. Trente-huit peptides ont ete identifies dans la fraction inferieure a 3 kDa du fromage de 38 mois (dont 21 peptides identifies dans le fromage jeune): 8 issus de la caseine β, 27 de la caseine α s1 , 1 de la caseine α s2 et 2 de la para-κ-CN. La fraction superieure a 10 kDa a ete soumise a un fractionnement sur colonne Mono Q, puis a une separation par CLHP en phase inverse sur colonne C4. Les peptides ont ete identifies en combinant la spectrometrie de masse a l'electrospray et la degradation d'Edman. Les resultats concernant une fraction chromatographique sont presentes: les variants B et C de la caseine α s1 et plusieurs fragments de grosse taille tels que le peptide α s1 -CN-I ont ete identifies. Les resultats montrent que les differentes techniques de spectrometrie de masse permettent de caracteriser, mieux que d'autres techniques,la nature des peptides issus des caseines, quelle que soit leur taille.
