Objetivo: Avaliar os efeitos moduladores e antioxidantes da ergotioneína (ERT) sobre vias de sinalização redox em camundongos falcêmicos. Materiais e métodos: Camundongos transgênicos para Anemia Falciforme (AF) foram gerados e mantidos na CEMIB/UNICAMP. Avaliou-se 3 grupos de camundongos humanizados: o controle, constituído de animais transgênicos para S+Fetal (SF); animais transgênicos para AF (SS) e transgênicos tratados com ERT (SS+ERT; VO 35 mg/kg), durante 60 dias. Os animais foram eutanasiados por aprofundamento da anestesia (CEUA 5955-1/2022) e a medula óssea foi utilizada para análise de RT-qPCR, em pseudo quintuplicatas, de membros de vias de sinalização centrais da biologia redox (ATF4, NFE2L2, FoxO3, CAT, SOD1, GPx1, PRDX1, TRX, TXNIP), do transportador específico de ERT (SLC22A4); e de um fator de iniciação da tradução eucariótica (EIF2α). O fold change do nível de mRNA foi calculado usando a fórmula 2-ΔΔCT. Para as análises estatísticas, adotou-se o General Linear Model, no formato one-way ANOVA, seguido pelo teste post hoc de Bonferroni. Resultados: No grupo SS foi observado uma indução dos fatores de transcrição NFE2L2 (̃9 vezes), FoxO3 (̃3 vezes) e das enzimas antioxidantes CAT (̃4 vezes) e GPX1 (̃6 vezes), enquanto para ATF4, TRX, SOD1 e PRDX1 houve uma inibição em relação ao grupo SF. Já no grupo SS+ERT observou-se uma expressão aumentada de ATF4 (̃2 vezes), TXNIP (̃6 vezes), CAT (̃8,5 vezes), SOD1 (̃1,2 vezes) e GPX1 (̃3,5 vezes), enquanto os níveis de NFE2L2, TRX1, PRDX1 se mantiveram similares ao controle. Interessantemente, houve uma indução da expressão do SLC22A4, tanto no grupo SS (̃7 vezes) quanto SS+ERT (̃13 vezes). Em relação ao EIF2α, sua expressão foi inibida nos SS e manteve-se similar ao controle em SS+ERT. Discussão: A AF é caracterizada pela geração incessante de espécies reativas de oxigênio, o que compromete a capacidade antioxidante das células eritroides. Assim, a fim de atenuá-lo, o mecanismo de resposta redox pode ocorrer de duas formas, pelo controle da atividade enzimática ou em nível transcricional, foco deste estudo, ao induzir as vias de sinalização redox e, consequente, aumentando síntese dos antioxidantes em situações de distresse oxidativo. Esta última, observada no grupo SS, como demonstrado pelos transcritos avaliados. No grupo SS+ERT, como esperado, a suplementação com ERT apresentou uma ação citoprotetora in vivo ao modular vias de adaptação redox. Resultados este que corroboram, nosso estudo prévio in vitro, em células K562 induzidas à diferenciação eritroide e submetidas a estresse oxidativo. Ainda, os altos níveis de SLC22A4 tanto nos SS quanto nos SS+ERT, demonstra que esse antioxidante é altamente acumulado em células hematopoiéticas. Ressaltando que a maior expressão desse transportador nos SS tratados sugere uma maior internalização da ERT após sua prolongada administração, reforçando seu papel crítico durante a hematopoiese. Atuação que é corroborada pela elevação de ATF4 e TXNIP que vem sendo propostos como novos reguladores da proliferação e diferenciação eritroide. Por fim, a análise da expressão de EIF2α, possibilita a proposição que os padrões observados em nível gênico se manteriam em nível proteico. Conclusão: ERT é um modulador com espectro ainda mais amplo do que o previamente disposto na literatura, atuando em vias essenciais para homeostasia redox e eritropoiese.

Objetivo: Demonstrar os efeitos protetores da melatonina (MEL) no controle do estresse oxidativo em modelo de camundongos transgênicos para anemia falciforme (AF). Materiais e métodos: Os camundongos foram gerados e mantidos no CEMIB/UNICAMP e divididos em três grupos: grupo controle constituído pelos animais S+Fetal (SF); animais transgênicos não tratados (SS); e animais transgênicos tratados com MEL (SS+MEL; VO 10 mg/kg) por 60 dias. Os animais foram eutanasiados por aprofundamento da anestesia (CEUA 5955-1/2022) e a medula óssea foi utilizada para análise de RT-qPCR, em pseudo quintuplicatas, de importantes genes de vias redox, tais como: NRF2, FOXO3, MST1, YWHAQ (14-3-3z), CAT, SOD1, GPX1, PRX1, TRX1 e TXNIP. A quantificação dos resultados se baseou na derivação da fórmula 2-ΔΔCT. As análises estatísticas foram realizadas pelo General Linear Model, no formato one-way ANOVA, seguido pelo teste post hoc de Bonferroni. Resultados: Os resultados demonstraram que a expressão do NRF2 (̃3 vezes), FOXO3 (̃3 vezes) e MST1 (̃6 vezes) foram induzidas nos SS, resultado este não observado nos SS+MEL; além disso, o YWHAQ foi induzido no SS+MEL (̃2 vezes) e se manteve similar ao controle no SS. Já para via TRX1/TXNIP, para os SS+MEL, a TRX1 se manteve similar ao SF e a TXNIP foi inibida (̃3 vezes), enquanto para os SS ocorreu uma inibição da TRX1 (̃3 vezes) e os níveis de TXNIP se mantiveram similares ao controle. Em relação aos genes que codificam antioxidantes, CAT e GPX1 foram induzidas 4 e 8 vezes, respectivamente, nos SS; porém, não quando sob tratamento com MEL. Já a SOD1 e PRX1 foram mantidos similares aos SF nos SS+MEL e inibidos nos SS. Discussão: Vários mecanismos moleculares têm sido propostos como contribuintes para uma elevada taxa oxidativa em pessoas com AF. Intermediários da homeostase redox não são apenas marcadores potenciais da gravidade da doença, mas também alvos importantes para terapias antioxidantes. Nossos resultados evidenciaram a atuação essencial dos fatores de transcrição NRF2 e FOXO3, em resposta ao distresse oxidativo característico da doença, como observado nos camundongos SS através da indução de seus transcritos associada a de genes sob seu controle, como das enzimas CAT e GPX1. Por outro lado, observamos uma restauração dos níveis de transcritos semelhante ao controle no grupo tratado com MEL. Já foi demonstrado que a MEL pode eliminar até dez espécies radicalares antes que o metabólito final seja eliminado do corpo, o que resulta em um efeito antioxidante em cascata. Esses resultados sugerem a noção de que a MEL atuou diretamente na detoxificação das espécies reativas no modelo usado, não sendo necessária a ativação das vias de sinalização redox. Além disso, alguns antioxidantes, como TRX, PRX1 e SOD1, que desempenham diversas outras funções celulares e são atualmente sugeridos como possíveis fatores de transcrição, foram mantidos com níveis de transcritos similares ao controle, demonstrando assim a modulação da MEL sobre genes envolvidos na resposta ao estresse oxidativo e outros processos celulares. Conclusão: A suplementação de camundongos transgênicos com MEL evidenciou o uso promissor dessa indolamina como estratégia terapêutica antioxidante na AF por meio da restauração dos níveis de transcritos de importantes genes envolvidos na manutenção redox para valores semelhantes ao grupo controle.

Objetivo: Avaliar a dinâmica do metabolismo redox, ao longo do tempo, comparando eritrócitos falcêmicos e saudáveis, em um modelo de autoincubação celular prolongada; considerando o impacto sobre marcadores de hemólise e lesões oxidativas. Materiais e métodos: O estudo envolveu 12 brasileiros consentidos (19-42 anos), não aparentados e independente do sexo, dentre os quais, seis não apresentavam hemoglobinopatia (grupo controle - CG) e os demais possuíam anemia falciforme (grupo de estudo – AF). As amostras de sangue foram devidamente coletas e processadas, obtendo ao final, os eritrócitos lavados, os quais foram suspensos no seu próprio plasma na concentração celular de 40% (v/v) e submetidos a seis horas de incubação à 37°C, com dois períodos de análise (t0h e t6h). Ao final de cada período, a fragilidade osmótica, os níveis de hemoglobina livre [Hb livre (μM)] e atividade de enzimas antioxidantes, incluindo catalase [CAT (U/L)], glutationa peroxidase [GPx (U/mL)], glutationa redutase [GR (U/mL)], metahemoglobina redutase [metaHb-redutase (U/mL)] e tiorredoxina redutase [TrxR (U/mL)] foram avaliados por espectrofotometria. Ademais, a proporção de colesterol [Ch (nM)] para colesterol-aldeído [ChAld (uM)] foi analisada, como marcador de lesão oxidativa, utilizando HPLC-UV e HPLC-FD, respectivamente. Comparou-se os grupos celulares e os períodos de incubação isoladamente e em seguida a interação deles. Resultados: Na comparação entre os grupos, os eritrócitos falcêmicos apresentaram resistência osmótica na concentração de 5 g/L de NaCl, aumento de 33,8% nos níveis de Hb livre [AF:1,72 ± 0,30; p < 0,01], redução de 22,8% e 27,4% na atividade das enzimas CAT [AF:15,39 ± 4,02; p < 0,05] e GPx [AF: 2,91 ± 0,74; p < 0,01], respectivamente. Ademais, houve o aumento de 31,5% na atividade da metaHb-redutase [AF:3,75 ± 0,90; p < 0,01] e acúmulo de 22,5% de produtos oxidativos da membrana celular [AF: 2,45 ± 0,17; p < 0,01] nos eritrócitos falcêmicos. Entretanto, não houve interação significativa entre os grupos e o período de incubação, quanto as enzimas CAT (p = 0,93), GPx (p = 0,59), GR (p = 0,69), TrxR (p = 0,34), metaHb-redutase (p = 0,53), assim como nos níveis de Hb livre (p = 0,58) e na proporção ChAld/Ch (p = 0,12). Discussão: A estrutura e a bioquímica da hemoglobina S induzem um estado hiperoxidativo nos eritrócitos falcêmicos. Isso leva ao consumo de enzimas antioxidantes, danos oxidativos na membrana celular e aumento da hemólise, evidenciado na comparação entre os grupos CG e AF, desconsiderando o tempo. Entretanto, ao longo da incubação, houve um prejuízo semelhante das enzimas antioxidantes e um grau equivalente de lesão oxidativa nos grupos estudados, demonstrando que apesar das condições oxidativas distintas a que são submetidos, ambos os grupos eritrocíticos mantiveram uma dinâmica de metabolismo redox similar ao longo do tempo testado, fornecendo subsídio argumentativo para pesquisas futuras focadas na análise das características metabólicas dessas células durante a eritropoiese, a fim de averiguar a adaptabilidade celular nos precursores eritrocitários e possíveis alvos terapêuticos. Conclusão: A curiosa semelhança na dinâmica do metabolismo redox de eritrócitos falcêmicos e saudáveis sugere que os primeiros apresentam alta adaptabilidade celular em seus precursores, possibilitando respostas redox semelhantes aos saudáveis no período testado.

The speciation of arsenic in fish has been widely investigated, but bioaccumulation and biotransformation of inorganic As in different tissues of Nile tilapia (Oreochromis niloticus) are not fully understood. The present study aimed to investigate the bioaccumulation of As in Nile tilapia, as well as to evaluate the distribution of the main arsenic species (As(III), As(V), MMA, DMA, and AsB) in liver, stomach, gill, and muscle, after controlled exposures to As(III) and As(V) at concentrations of 5.0 and 10.0 mg L -1 during periods of 1 and 7 days. Total As was determined by inductively coupled plasma mass spectroscopy (ICP-MS). For both exposures (As(III) and As(V)), the total As levels after 7-day exposure were highest in the liver and lowest in the muscle. Overall, the Nile tilapia exposed to As(III) showed higher tissue levels of As after the treatments, compared to As(V) exposure. Speciation of arsenic present in the tissues employed liquid chromatography coupled to ICP-MS (LC-ICP-MS), revealing that the biotransformation of As included As(V) reduction to As(III), methylation to monomethylarsonic acid (MMA) and dimethylarsinic acid (DMA), and subsequent conversion to nontoxic arsenobetaine (AsB), which was the predominant arsenic form. Finally, the interactions and antagonistic effects of selenium in the bioaccumulation processes were tested by the combined exposure to As(III), the most toxic species of As, together with tetravalent selenium (Se(IV)). The red blood cells (RBCs) are essential to transport oxygen (O 2 ) and nutrients throughout the human body. Changes in the structure or functioning of the erythrocytes can lead to several deficiencies, such as hemolytic anemias, in which an increase in reactive oxidative species generation is involved in the pathophysiological process, playing a significant role in the severity of several clinical manifestations. There are important lines of defense against the damage caused by oxidizing molecules. Among the antioxidant molecules, the enzyme peroxiredoxin (Prx) has the higher decomposition power of hydrogen peroxide, especially in RBCs, standing out because of its abundance. This review aimed to present the recent findings that broke some paradigms regarding the three isoforms of Prxs found in RBC (Prx1, Prx2, and Prx6), showing that in addition to their antioxidant activity, these enzymes may have supplementary roles in transducing peroxide signals, as molecular chaperones, protecting from membrane damage, and maintenance of iron homeostasis, thus contributing to the overall survival of human RBCs, roles that seen to be disrupted in hemolytic anemia conditions., (© 2023. Background: Interactions between the somatosensory and motor cortices are of fundamental importance for motor control. Although physically distant, face and hand representations are side by side in the sensorimotor cortex and interact functionally. Traumatic brachial plexus injury (TBPI) interferes with upper limb sensorimotor function, causes bilateral cortical reorganization, and is associated with chronic pain. Thus, TBPI may affect sensorimotor interactions between face and hand representations., Objective: The aim of this study was to investigate changes in hand-hand and face-hand sensorimotor integration in TBPI patients using an afferent inhibition (AI) paradigm., Method: The experimental design consisted of electrical stimulation (ES) applied to the hand or face followed by transcranial magnetic stimulation (TMS) to the primary motor cortex to activate a hand muscle representation. In the AI paradigm, the motor evoked potential (MEP) in a target muscle is significantly reduced when preceded by an ES at short-latency (SAI) or long-latency (LAI) interstimulus intervals. We tested 18 healthy adults (control group, CG), evaluated on the dominant upper limb, and nine TBPI patients, evaluated on the injured or the uninjured limb. A detailed clinical evaluation complemented the physiological investigation., Results: Although hand-hand SAI was present in both the CG and the TBPI groups, hand-hand LAI was present in the CG only. Oxidative stress is a major cause of morbidity and mortality in human health and disease. In this review, we focus on the Forkhead Box (Fox) subclass O3 (FoxO3), an extensively studied transcription factor that plays a pleiotropic role in a wide range of physiological and pathological processes by regulating multiple gene regulatory networks involved in the modulation of numerous aspects of cellular metabolism, including fuel metabolism, cell death, and stress resistance. This review will also focus on regulatory mechanisms of FoxO3 expression and activity, such as crucial post-translational modifications and non-coding RNAs. Moreover, this work discusses and evidences some pathways to how this transcription factor and reactive oxygen species regulate each other, which may lead to the pathogenesis of various types of diseases. Therefore, in addition to being a promising therapeutic target, the FoxO3-regulated signaling pathways can also be used as reliable diagnostic and prognostic biomarkers and indicators for drug responsiveness., (© 2023. This study aimed to establish the importance of ergothioneine (ERT) in the erythroid adaptation mechanisms by appraising the expression levels of redox-related genes associated with the PI3K/AKT/FoxO3 and Nrf2-ARE pathways using K562 cells induced to erythroid differentiation and H 2 O 2 -oxidative stress. Cell viability and gene expression were evaluated. Two concentrations of ERT were assessed, 1 nM (C1) and 100 µM (C2), with and without stress induction (100 µM H 2 O 2 ). Assessments were made in three periods of the cellular differentiation process (D0, D2, and D4). The C1 treatment promoted the induction of FOXO3 (D0 and 2), PSMB5 , and 6 expressions (D4); C1 + H 2 O 2 treatment showed the highest levels of NRF2 transcripts, KEAP1 (D0), YWHAQ (D2 and 4), PSMB5 (D2) and PSMB6 (D4); and C2 + H 2 O 2 (D2) an increase in FOXO3 and MST1 expression, with a decrease of YWHAQ and NRF2 was observed. in C2 + H 2 O 2 (D2) an increase in FOXO3 and MST1 , with a decrease in YWHAQ and NRF2 was observed All ERT treatments increased gamma-globin expression. Statistical multivariate analyzes highlighted that the Nrf2-ARE pathway presented a greater contribution in the production of PRDX1 , SOD1 , CAT , and PSBM5 mRNAs, whereas the PI3K/AKT/FoxO3 pathway was associated with the PRDX2 and TRX transcripts. In conclusion, ERT presented a cytoprotective action through Nrf2 and FoxO3, with the latter seeming to contribute to erythroid proliferation/differentiation.

Melatonin (MEL) presents well-documented pleiotropic actions against oxidative stress (OS), acting indirectly through activation of transcription factors, e.g., FoxO3 and Nrf2. Thus, this study aimed to investigate the possible modulating effects of MEL on the redox signaling pathways PI3K/AKT/FoxO3 and Keap1/Nrf2/ARE in K562 erythroleukemic cells subjected to OS induction. For this, the viability, and transcript levels of genes involved in redox adaptation were evaluated in K562 cells in different periods of erythroid differentiation: under OS induction by hydrogen peroxide (100 µM H 2 O 2 ); treated with 1 nM (C1) and 1 mM (C2) MEL; and associated or not with stress induction. We observed a restoration of physiological levels of Nrf2 in both MEL concentrations under OS. The C1 was related to enhanced expression of antioxidant and proteasome genes through the Nrf2-ARE pathway, while C2 to the induction of FOXO3 expression, suggesting an involvement with apoptotic pathway, according to BIM transcript levels. The effects of MEL administration in these cells showed a period and dose-dependent pattern against induced-OS, with direct and indirect actions through different pathways of cellular adaptation, reinforcing the importance of this indolamine in the regulation of cellular homeostasis, being a promising therapeutic alternative for diseases that present an exacerbated OS.

Introduction: Studies have shown that the loss of the FOXO3 transcriptional function is involved in the pathophysiology of some chronic erythroid disorders, including beta-thalassemia (β-thal). Therefore, the single nucleotide polymorphism (SNP) rs3800231 (35-2764A > G) could contribute to alterations in its transcriptional activity, acting as a modifier of β-thal phenotypic manifestations., Objective and Method: In order to better understand the genotypic and/or allelic distributions among β-thal patients, we evaluated 83 β-thal heterozygous and 20 homozygous, compared to 117 individuals without hemoglobinopathies (control group). Additionally, we verified any influence of the FOXO3 polymorphism on clinical manifestations among β-thal homozygotes., Results: We obtained higher frequencies of the wild-type homozygous (AA) and the wild-type allele (A) in the β-thal group (p < 0.0001 and p = 0.00014, respectively). The most common clinical manifestations found among β-thal homozygotes were iron overload (90%), splenomegaly (65%) and bone complications (35%), e.g., osteopenia/osteoporosis. We observed that close to 80% of the patients presenting such manifestations had the genotype AA. However, we did not find any significant involvement of the FOXO3 polymorphism in clinical manifestation occurrences., Conclusion: Thus, we concluded that the SNP rs3800231 did not play a significant role as a modifier of the clinical manifestations observed in the β-thal homozygotes studied., (Copyright © 2020 Associação Brasileira de Hematologia, Hemoterapia e Terapia Celular. Objectives: The aim of this study was to evaluate the efficacy of two passive ultrasonic irrigation (PUI) methods and conventional manual irrigation (CMI) in root canal system (RCS) cleaning., Materials and Methods: Artificial single-rooted teeth were used. Four lateral canals were made 2 and 7 mm short from the apex. Root canals were filled with a contrast solution. The specimens were divided into four groups (n = 8): PUI1-intermittent flushing; PUI2-continuous flushing; CMI1-at 1 mm; and CMI2-at 3 mm. Micro-computed tomographic images were obtained. New images were taken after the irrigation protocols enabling the evaluation of the contrast solution volume (in mm(3)) within RCS (main and lateral root canals) in comparison to the initial analysis. The comparison among groups was performed using ANOVA and Tukey's tests with 5 % significance level., Results: There was no difference among PUI1, PUI2, and CMI1 regarding the contrast solution removal from RCS (p > 0.05). These methods demonstrated greater cleaning than CMI2. At the apical third, CMI2 showed lower cleaning capacity when compared to the other methods (p < 0.05)., Conclusions: PUI with intermittent or continuous flushing and CMI with the needle placed 1 mm from the working length were efficient in cleaning the main and lateral root canals., Clinical Relevance: Root canal irrigation plays an important role in cleaning and disinfection of the RCS. Effective apical irrigation is an important procedure during root canal treatment. PUI and CMI (at 1 mm short) were effective in cleaning of the main and simulated lateral root canals.

Objective: The aim of this study was to compare the cleaning of main and lateral canals using the irrigation methods: negative pressure irrigation (EndoVac system), passive ultrasonic irrigation (PUI) and manual irrigation (MI)., Materials and Methods: Resin teeth were used. After root canal preparation, four lateral canals were made at 2 and 7 mm from the apex. Root canals were filled with contrast solution and radiographed pre- and post-irrigation using digital radiographic system [radiovisiography (RVG)]. The irrigation protocols were: MI1-manual irrigation [22 G needle at 5 mm short of working length-WL]; MI2-manual irrigation (30G needle at 2 mm short of WL); PUI; EV1-EndoVac (microcannula at 1 mm short of WL); EV2-Endovac (microcannula at 3 mm short of WL). The obtained images, initial (filled with contrast solution) and final (after irrigation) were analyzed by using image tool 3.0 software. Statistical analysis was performed by analysis of variance (ANOVA) and Tukey tests (5% significance level)., Results: EV1 and EV2, followed by PUI showed better cleaning capacity than manual irrigation (MI1 and MI2) (p < 0.05)., Conclusion: Negative pressure irrigation and PUI promoted better cleaning of main and simulated lateral canals., Clinical Significance: Conventional manual irrigation technique may promote less root canal cleaning in the apical third. For this reason, the search for other irrigation protocols is important, and EndoVac and PUI are alternatives to contribute to irrigation effectiveness.

A birefringence technique is used to determine the average magnetic moments of magnetotactic bacteria in culture. Differences in are noted between live and dead bacteria, as well as between normal density and high density samples of live bacteria.
