L'esposizione di vongole ad un germicida commerciale (1000 ppm) per 3 ore per ridurre la carica batterica non modifica l'attività catalasica nella ghiandola digestiva. Invece dosi molto inferiori del germicida saggiate in vitro su colture di emociti alterano profondamente la morfologia e la funzionalità cellulare. Pertanto questo trattamento sembra applicabile in vivo, anche se in vitro è chiaramente immunotossico. The exposure of clams to 1000 ppm strong oxidizing commercial germicide for 3 hours to reduce bacterial load did not modify the catalase activitiy in the midgut gland. Conversely, far lower germicide doses strongly affected the morphology and functionality of in vitro tested cultured haemocytes. The germicide treatment may be promising in vivo, in spite of its in vitro immunotoxicity.

In this study we compared the mitochondrial membranes from gills of Philippine clam Ruditapes philippinarum and of the Mediterranean mussel Mytilus galloprovincialis analyzing the lipid composition and the physico-chemical state of both membranes and liposomes prepared from the total lipids extracted from mitochondria. Fatty acid and sterol analyses were carried out as in a previous work. Physico-chemical studies of isolated membranes and liposomes were performed at three temperatures (10,20,30 °C) by using 2-dimethylamino-(lauroyl)-naphtalene (Laurdan) as fluorescent probe which is located at the polar headgroups level. Although both species exhibit the typical fatty acid pattern of marine invertebrates with high levels of n-3 PUFA, the fatty acid composition resulted deeply “species-oriented”. The different fatty acid profile could mirror, other than the different habitats and feeding habits, also a species-specific biosynthetic pattern. Sterol composition was more diversified in R. philippinarum than in M. galloprovincialis and in both species more than 50% sterols were different from cholesterol (57,9 and 55% respectively). Although mitochondrial membranes of both species were in a liquid-crystalline phase, in Ruditapes philippinarum the physical state was much more sensitive to temperature and less rigid than that in Mytilus galloprovincialis. Moreover liposomes prepared from mitochondrial total lipids of both species showed peculiar differences of the physical phase.

Although the bactericidal properties of the germicide employed on clams are tightly linked to its oxidative power, confirmed by the oxidative stress detected in in vitro treated haemocytes, the similarity of VOC profiles and TBARs levels in treated and control clams suggest that the organoleptic features are unaffected, thus shouldering this treatment before clam introduction into the market.

SUMMARY – The expression of HIF-1a and HSF1 was investigated in plasma samples (T0: at birth; T12: 12 hours after birth) of 9 neonates (4 healthy and 5 sick foals); plasma MDA concentration was also evaluated in 16 neonates (10 healthy and 6 sick foals). MDA concentration, HIF-1a and HSF1 expression decreased significantly at T12 if compared with T0 (p< 0.05). HIF-1a was significantly higher in sick foals at all sampling times (p< 0.05) and resulted correlated to pCO2 at T0 and to MDA at T12 (p< 0,05; R=0,83 e R=-0,72, respectively). HSF was significantly correlated to HCT and RBC at T0 (p< 0,05; R=0,68 e R=0,77, respectively). The parameters measured could be considered potential indexes of perinatal hypoxia.

Gli organismi viventi che tollerano ampie variazioni di ossigeno durante episodi di ipossia/anossia (euriossici), presentano adattamenti biochimici e fisiologici che ne consentono la sopravvivenza fino al ripristino delle normali condizioni di ossigenazione. Il ritorno alla normossia è caratterizzato da un aumento repentino del consumo di ossigeno che può indurre la formazione di specie radicaliche dell’ossigeno (ROS) con un conseguente danno ossidativo. Dal momento che i ROS sono continuamente prodotti, gli organismi hanno evoluto un sistema di difesa antiossidante costituito da componenti enzimatiche e non enzimatiche. I sistemi enzimatici maggiormente coinvolti sono rappresentati da superossido dismutasi (SOD), da catalasi (CAT) e dalla glutatione perossidasi Se-dipendente (GPx). Nel presente studio è stato utilizzato come animale sperimentale Scapharca inaequivalvis, una specie di mollusco bivalve presente in Adriatico, dal momento che la risposta alla riossigenazione degli animali euriossici è proponibile come un modello utile per lo studio del danno da ischemia/riperfusione. Gli animali sono stati esposti ad anossia per 3,6,24 ore e riossigenazione per 1,3,5,24 ore. Sulla ghiandola digestiva dei molluschi è stata investigata l’attività di SOD, CAT, GPx e G6PDH (tecniche spettrofotometriche), nonchè l’espressione di due isoforme della SOD, MnSOD e CuZnSOD (immunoblotting). I risultati hanno mostrato alti livelli di SOD sia in anossia che in riosssigenazione; CAT e GPx aumentano significativamente durante il recupero mostrando una correlazione inversa dal momento che competono per lo stesso substrato a dimostrazione di una attività di difesa contro lo stress ossidativo integrata ed efficiente. L’aumento del GPx, corrispondente ad un picco di attività della G6PDH alla dodicesima ora di riossigenazione, potrebbe essere riconducibile a fenomeni di perossidazione lipidica. Dall’immunoblotting si nota che solo la CuZnSOD viene maggiormente espressa dopo 6 e 24h di anossia e 3h di riossigenazione. Nei mammiferi in ischemia si assiste ad una caduta della concentrazione degli enzimi antiossidanti, il ritorno alla normossia (riperfusione o riossigenazione) è caratterizzato da un rapido aumento del consumo di ossigeno che induce la formazione di ROS. Gli episodi degenerativi a livello cellulare iniziano a comparire solo in seguito alla reintroduzione dell’ossigeno molecolare nel tessuto ischemico. La presenza di alti livelli di enzimi anche in anossia indica una risposta di S. inaequivalvis tipica degli animali eurossici e utile per lo studio dei processi ischemici in mammiferi.

Nei teleostei il rame svolge funzioni vitali come cofattore in processi biologici-chiave, ma può anche manifestare effetti tossici, in dipendenza della biodisponibilità e del bioaccumulo. La forma Cu2+ del rame è quella a maggiore biodisponibilità. La tossicità del rame varia a seconda della specie ed è noto che gli stadi larvali dei pesci sono molto più sensibili al metallo rispetto agli adulti della stessa specie. Nella stessa specie tessuti e processi diversi possono inoltre manifestare effetti estremamente diversificati. Le principali vie di assunzione del metallo nei pesci sono rappresentate dalle branchie e dall’apparato digerente. I dati presentati fanno parte di una ricerca multidisciplinare che abbraccia un vasto spettro di indagini su diversi organi e tessuti e sono stati ottenuti in orate (Sparus auratus) esposte a CuSO4 (0,5 ppm nominali di Cu, salinità 33‰, t 10°C) e mantenute per 50 gg in un sistema statico, senza alimentazione. Parellelamente nelle stesse condizioni sono state mantenute due vasche con animali non esposti (controllo). Durante tutta la sperimentazione non è stata riscontrata mortalità, né visibili sofferenze o alterazioni comportamentali degli animali. Lipidi e acidi grassi. Le indagini condotte sul grasso periviscerale, più esposto rispetto ad altri distretti ai cambiamenti che si possono produrre in un organismo, evidenzia uno pettro degli acidi grassi tipico di un teleosteo allevato in acqua salata, alimentato quindi con diete contenenti grassi di origine vegetale come suggerito dalla cospicua presenza di 18:2n-6. Il digiuno protratto per tutti i 50 giorni di sperimentazione non ha modificato i rapporti reciproci tra i vari acidi grassi le cui percentuali al tempo 0 sono sovrapponibili con quelle di fine esperimento. La temperatura complessivamente bassa per tutta la durata dell’esperimento può aver ridotto al minimo le esigenze metaboliche e consentito il mantenimento dei valori iniziali anche dei saturi a corta catena. La stessa ridotta attività metabolica può aver interessato anche i processi attraverso cui si esplica l’azione del rame il cui effetto non è risultato quindi rilevabile su questo parametro. Espressione citocromo P450 (CYP1A): fegato - branchie. Indagini preliminari sul citocromo P450 (CYP 1A), eseguite mediante Western Blotting con anticorpi policlonali anti-fish in campioni di controllo e in campioni prelevati dopo 14 giorni di esposizione, hanno evidenziato in fegato e branchie due bande distinte di 49 e 61 kDa. Nel fegato la banda di 49 kDa non mostra differenze significative fra le condizioni esaminate mentre la banda di 61 kDa risulta più espressa nei trattati. Nelle branchie non si evidenziano differenze significative per quanto concerne la banda di più alto peso molecolare, mentre la banda inferiore sembra significativamente meno espressa nei trattati rispetto ai controlli.

To prove CYP1A-immunopositive protein presence and to evaluate MnSOD behaviour in haemocytes and haemolymph, specimens of C. gallina were exposed to 0.5 mg/L of Benzo[a]pyrene for 12 days. CYP1A-immunopositive protein showed two bands of 53.5 kDa and 63.8 kDa not differently expressed, whereas MnSOD activity decreased after 7 and 12 days of exposure.

The major animal responses to acute shock are known to involve changes in gene transcription and translation of heat shock protein (HSPs) . HSPs show different roles as chaperones in normal and damaged protein folding and in maintenance of receptor conformation. Marine littoral invertebrates tolerate a remarkable array of natural stresses, including anoxia, thermal and osmotic fluctuations. Several of these environmental stresses are responsible of HSP induction, that contribute to the tolerance of otherwise lethal conditions. This study was aimed at detecting HSP70 expression in Chamelea gallina tissues in response to oxygen, temperature and salinity changes.

The Antarctic zooplankton is adapted to low temperatures and the shortness of the primary production period. The lipid characterisation in these animals can increase the knowledge of biological and biochemical adaptation and of the transfer through trophic levels of molecules considered as environmental biomarkers. Therefore in the present study were analysed hydrocarbons, sterols, fatty acids (FAMEs) and waxes from the total lipid extracts of three zooplankton samples (1mm mesh Plankton Hamburg Net) collected on December 1997 in the stations PHN20, 22 and 30 in the "Continental Slope" sector of the Ross Sea.

Il cancro allo stomaco è la seconda forma più comune di tumore al mondo. Le variazioni a carico della frazione lipidica sono direttamente implicate nei meccanismi di proliferazione della malattia. Si è ritenuto pertanto e risulta pertanto particolarmente interessante valutare le modificazioni delle varie componenti lipidiche in relazione alla progressione della malattia tumorale ed alla proliferazione cellulare. Dopo estrazione della frazione lipidica totale, (Folch et.al.), le principali componenti sono state separate con l’utilizzo di colonnine SPE Discovery DSC-Si che hanno permesso di ottenere le seguenti frazioni: a) esteri del colesterolo, squalene (CHOLE), b) Trigliceridi (TG), c) colesterolo, dolicoli (CHOL), d) fosfolipidi (PH). L’analisi delle frazioni è stata effettuata mediante HPLC con una colona C18 a fase inversa ASCENTIS (Supelco). Differenze tra tessuto sano e tessuto tumorale si sono evidenziate nella frazione CHOL: in particolare per quanto riguarda i dolicoli essi sono presenti nel tessuto sano e assenti in quello tumorale. La frazione del CHOLE mostra come gli esteri del colesterolo siano molto più concentrati nel tumore rispetto al tessuto sano. La variazione di queste due componenti è da mettere in relazione al ruolo che esse hanno nei confronti della fluidità di membrana (dolicoli) e nella angiogenesi nei tessuti tumorali (esteri del colesterolo). Variazioni sostanziali non sono state evidenziate per ubichinone e colesterolo libero.

open 5 no The mitochondrial permeability transition pore (PTP), which drives regulated cell death when Ca2+ concentration suddenly increases in mitochondria, was related to changes in the Ca2+-activated F1FO-ATPase. The effects of the gadolinium cation (Gd3+), widely used for diagnosis and therapy, and reported as PTP blocker, were evaluated on the F1FO-ATPase activated by Mg2+ or Ca2+ and on the PTP. Gd3+ more effectively inhibits the Ca2+-activated F1FO-ATPase than the Mg2+-activated F1FO-ATPase by a mixed-type inhibition on the former and by uncompetitive mechanism on the latter. Most likely Gd3+ binding to F1, is favoured by Ca2+ insertion. The maximal inactivation rates (Kinact) of pseudo-first order inactivation are similar either when the F1FO-ATPase is activated by Ca2+ or by Mg2+. The half-maximal inactivator concentrations (KI) are 2.35 ± 0.35 mM and 0.72 ± 0.11 mM, respectively. The potency of a mechanism-based inhibitor (Kinact/KI) also highlights a higher inhibition efficiency of Gd3+ on the Ca2+-activated F1FO-ATPase (0.59 ± 0.09 mM-1∙s-1) than on the Mg2+-activated F1FO-ATPase (0.13 ± 0.02 mM-1∙s-1). Consistently, the PTP is desensitized in presence of Gd3+. The properties of the mitochondrial F1 FO -ATPase catalytic site, which can bind Mg2+ , Mn2+ , or Ca2+ and hydrolyze ATP, were explored by inhibition kinetic analyses to cast light on the Ca2+ -activated F1 FO -ATPase connection with the permeability transition pore (PTP) that initiates cascade events leading to cell death. While the natural cofactor Mg2+ activates the F1 FO -ATPase in competition with Mn2+ , Ca2+ is a noncompetitive inhibitor in the presence of Mg2+ . Selective F1 inhibitors (Is-F1 ), namely NBD-Cl, piceatannol, resveratrol, and quercetin, exerted different mechanisms (mixed and uncompetitive inhibition) on either Ca2+ - or Mg2+ -activated F1 FO -ATPase, consistent with the conclusion that the catalytic mechanism changes when Mg2+ is replaced by Ca2+ . In a partially purified F1 domain preparation, Ca2+ -activated F1 -ATPase maintained Is-F1 sensitivity, and enzyme inhibition was accompanied by the maintenance of the mitochondrial calcium retention capacity and membrane potential. The data strengthen the structural relationship between Ca2+ -activated F1 FO -ATPase and the PTP, and, in turn, on consequences, such as physiopathological cellular changes.

In mammalian cells enzymatic and non-enzymatic pathways produce H2S, a gaseous transmitter which recently emerged as promising therapeutic agent and modulator of mitochondrial bioenergetics. To explore this topic, the H2S donor NaHS, at micromolar concentrations, was tested on swine heart mitochondria. NaHS did not affect the F1FO-ATPase activated by the natural cofactor Mg2, but, when Mg2+ was replaced by Ca2+, a slight 15% enzyme inhibition at 100 µM NaHS was shown. Conversely, both the NADH-O2 and succinate-O2 oxidoreductase activities were totally inhibited by 200 μM NaHS with IC50 values of 61.6 ± 4.1 and 16.5 ± 4.6 μM NaHS, respectively. Since the mitochondrial respiration was equally inhibited by NaHS at both first or second respiratory substrates sites, the H2S generation may prevent the electron transfer from complexes I and II to downhill respiratory chain complexes, probably because H2S competes with O2 in complex IV, thus reducing membrane potential as a consequence of the cytochrome c oxidase activity inhibition. The Complex IV blockage by H2S was consistent with the linear concentration-dependent NADH-O2 oxidoreductase inhibition and exponential succinate-O2 oxidoreductase inhibition by NaHS, whereas the coupling between substrate oxidation and phosphorylation was unaffected by NaHS. Even if H2S is known to cause sulfhydration of cysteine residues, thiol oxidizing (GSSG) or reducing (DTE) agents, did not affect the F1FO-ATPase activities and mitochondrial respiration, thus ruling out any involvement of post-translational modifications of thiols. The permeability transition pore, the lethal channel which forms when the F1FO-ATPase is stimulated by Ca2+, did not open in the presence of NaHS, which showed a similar effect to ruthenium red, thus suggesting a putative Ca2+ transport cycle inhibition.

Through a multiple approach, the present study on the mitochondrial membranes from mussel gills and swine heart combines some biochemical information on fatty acid composition, sterol pattern, and temperature dependence of the F1FO-ATPase activity (EC 3.6.3.14.) with fluorescence data on mitochondrial membranes and on liposomes obtained from lipid extracts of mitochondria. The physical state of mussel gills and swine heart was investigated by Laurdan steady state fluorescence. Quite surprisingly, the similar temperature dependence of the F1FO complex, illustrated as Arrhenius plot which in both mitochondria exhibits the same discontinuity at approximately 21°C and overlapping activation energies above and below the discontinuity, is apparently compatible with a different composition and physical state of mitochondrial membranes. Accordingly, mussel membranes contain highly unsaturated fatty acids, abundant sterols, including phytosterols, while mammalian membranes only contain cholesterol and in prevalence shorter and less unsaturated fatty acids, leading to a lower membrane unsaturation with respect to mussel mitochondria. As suggested by fluorescence data, the likely formation of peculiar microdomains interacting with the membrane-bound enzyme complex in mussel mitochondria could produce an environment which somehow approaches the physical state of mammalian mitochondrial membranes. Thus, as an adaptive strategy, the interaction between sterols, highly unsaturated phospholipids and proteins in mussel gill mitochondria could allow the F1FO-ATPase activity to maintain the same activation energy as the mammalian enzyme.
