This paper analyzes the relation between the ground leakage current and the power semiconductor parasitic components of a three-phase Current Source Inverter (CSI) topology named in literature H7 or CSI7. This topology is characterized by the presence of an additional switch which allows to reduce conduction power losses and decouple the DC side from the AC side during the application of the Zero current Vector to reduce ground leakage current. This work deals with the significant variation of this current depending on power converter parasitic capacitors. In this work the analysis is applied to grid-connected converters for a string photovoltaic applications, even if the behavior is the same also in case of electrical drives. The analysis is assessed by numerical simulations and experimental results with different parasitic capacitances that confirm also the validity of the proposed equivalent common-mode circuit.

Ecdysone signaling plays key roles in Drosophila oogenesis, as its activity is required at multiple steps during egg chamber maturation. Recently, its involvement has been reported on eggshell production by controlling chorion gene transcription and amplification. Here, we present evidence that ecdysone signaling also controls the expression of the eggshell gene VM32E, whose product is a component of vitelline membrane and endochorion layers. Specifically blocking the function of the different Ecdysone receptor (EcR) isoforms we demonstrate that EcR-B1 is responsible for ecdysone-mediated VM32E transcriptional regulation. Moreover, we show that the EcR partner Ultraspiracle (Usp) is also necessary for VM32E expression. By analyzing the activity of specific VM32E regulatory regions in usp2 clones we identify the promoter region mediating ecdysone-dependent VM32E expression. By in vitro binding assay and site-directed mutagenesis we demonstrate that this region contains a Usp binding site necessary for VM32E regulation.Our results further support the crucial role of ecdysone signaling in controlling transcription of eggshell structural genes and suggest that the heterodimeric complex EcR-B1/Usp mediates the ecdysone-dependent VM32E transcriptional activation in the main body follicle cells.

OBJETIVO: Estabelecer a prevalência de dislipidemias em uma amostra populacional brasileira ambulatorial de crianças e adolescentes de um centro hospitalar terciário público. MÉTODOS: Estudo retrospectivo de análise de resultados de lípides e lipoproteínas de 1.937 indivíduos de ambos os sexos, com idades entre dois e 19 anos, atendidos no ambulatório do Hospital de Clínicas (HC) da Universidade Estadual de Campinas, no Estado de São Paulo, entre 2000 e 2007. Os valores de referência adotados foram os recomendados por Kwiterovich. A análise estatística utilizou o programa SPSS. RESULTADOS: Valores alterados de colesterol, LDL-colesterol e triglicérides foram encontrados em 44, 36 e 56% das crianças de dois a nove anos e em 44, 36 e 50% dos adolescentes de dez a 19 anos, respectivamente. Foi observada redução de HDL-colesterol em 44% das crianças e 49% dos adolescentes. Hipercolesterolemia combinada com hipertrigliceridemia estava presente em 34% das crianças e dos adolescentes; hipercolesterolemia e hipoalfalipoproteinemia estavam presentes em 15% das crianças e 20% dos adolescentes; e hipertrigliceridemia e hipoalfalipoproteinemia ocorreram em 33% das crianças e 30% dos adolescentes. CONCLUSÕES: Este estudo demonstrou alta prevalência de dislipidemias em crianças e adolescentes atendidos em um ambulatório de hospital universitário público, o que aponta para a necessidade de estudos em outras comunidades brasileiras, tanto hospitalares quanto não hospitalares, para o diagnóstico precoce e tratamento das dislipidemias e para a adoção de medidas preventivas com relação às doenças cardiovasculares associadas.OBJECTIVE: To establish the prevalence of dyslipidemia among children and adolescents assisted at public university hospital outpatient clinic METHODS: This retrospective study analyzed the lipid profile of 1,937 outpatients of both genders, from two to 19 years old, assisted at the outpatient clinic of the Clinical Hospital of the State University of Campinas, Brazil, from 2000 to 2007. The cutoff values adopted were those recommended by Kwiterovich. Statistical analysis used the SPSS software. RESULTS: Altered values of cholesterol, LDL-cholesterol and triglycerides were found in 44, 36 and 56% of the children (2-9 years old) and in 44, 36 and 50% of the adolescents (10-19 years old), respectively. Reduced HDL-cholesterol concentrations were present in 44% of the children and 49% of the adolescents. A combination of hypercholesterolemia and hypertriglyceridemia was present in 34% of children and adolescents. Hypercholesterolemia combined with hypoalphalipoproteinemia was present in 15% of the children and 20% of the adolescents, and hypertriglyceridemia combined with hypoalphalipoproteinemia was present in 33% of the children and 30% of the adolescents. CONCLUSIONS: The frequency of dyslipidemias in children and adolescents of the out-patient clinic of a public university hospital was high. There is a need to further study hospital and non-hospital communities in order to establish early diagnosis and treatment of dyslipidemias and to avoid associated cardiovascular diseases.

In the follicle cell (FC) epithelium that surrounds the Drosophila egg, a complex set of cell signals specifies two cell fates that pattern the eggshell: the anterior centripetal FC that produce the operculum and the posterior columnar FC that produce the main body eggshell structure. We have previously shown that the long-range morphogen DPP represses the expression of the bunched (bun) gene in the anterior-most centripetal FC. bun, which encodes a homolog of vertebrate TSC-22/GILZ, in turn represses anterior gene expression and antagonizes Notch signaling to restrict centripetal FC fates in posterior cells. From a screen for novel targets of bun repression we have identified the C/EBP homolog slow border cells (slbo). At stage 10A, slbo expression overlaps bun in anterior FC; by stage 10B they repress each other's expression to establish a sharp slbo/bun expression boundary. The precise position of the slbo/bun expression boundary is sensitive to Notch signaling, which is required for both slbo activation and bun repression. As centripetal migration proceeds from stages 10B–14, slbo represses its own expression and both slbo loss-of-function mutations and overexpression approaches reveal that slbo is required to coordinate centripetal migration with nurse cell dumping. We propose that in anterior FC exposed to a Dpp morphogen gradient, high and low levels of slbo and bun, respectively, are established by modulation of Notch signaling to direct threshold cell fates. Interactions among Notch, slbo and bun resemble a conserved signaling cassette that regulates mammalian adipocyte differentiation.

Proper assembly and maintenance of epithelia are critical for normal development and homeostasis. Here, using the Drosophila ovary as a model, we identify a role for the B1 isoform of the ecdysone receptor (EcR-B1) in this process. We performed a reverse genetic analysis of EcR-B1 function during oogenesis and demonstrate that silencing of this receptor isoform causes loss of integrity and multilayering of the follicular epithelium. We show that multilayered follicle cells lack proper cell polarity with altered distribution of apical and basolateral cell polarity markers including atypical-protein kinase C (aPKC), Discs-large (Dlg), and Scribble (Scrib) and aberrant accumulation of adherens junctions and F-actin cytoskeleton. We find that the EcR-B1 isoform is required for proper follicle cell polarity both during early stages of oogenesis, when follicle cells undergo the mitotic cell cycle, and at midoogenesis when these cells stop dividing and undergo several endocycles. In addition, we show that the EcR-B1 isoform is required during early oogenesis for follicle cell survival and that disruption of its function causes apoptotic cell death induced by caspase.

The Drosophila eggshell provides a model system for studying the assembly of extracellular matrix. Eggshell formation is a complex process that requires time-coordinated synthesis, cleavage, and transport of various proteins and finally cross-linking mediated by particular functional domains. It has been suggested that the eggshell can act as a storage site for spatial cues involved in embryonic pattern formation. Its structural components are synthesized in the somatic follicle cells in a precise temporally and spatially regulated manner. This review will summarize our knowledge of eggshell gene expression. We will discuss the amplification of the chorion gene clusters and the data acquired on the expression patterns and the regulatory elements controlling transcription of eggshell genes. We will then focus on the findings that correlate follicular epithelium patterning and eggshell gene expression, and discuss the interesting perspectives of an involvement in eggshell assembly of embryonic patterning cues. Drosophila vitelline membrane gene VM32E is expressed in the follicle cells of the stage 10 egg chamber and shows a peculiar temporal and spatial expression pattern compared to the other members of the same gene family. Previous work has led us to demonstrate that Decapentaplegic (Dpp) signaling represses the expression of the VM32E gene in the centripetal follicle cells. In this paper, we describe another level of complexity of the VM32E gene expression regulation. Through clonal analyses, we show that the expression of the VM32E gene in the main body follicle cells is modulated by the epidermal growth factor receptor (Egfr) activity. In follicle cell clones expressing a constitutively active form of the Egfr, the VM32E gene is downregulated, while the loss of the Egfr activity upregulates VM32E expression. In addition, we show that the ectopic expression of the Egfr-induced ETS transcription factor PointedP2 (PntP2) affects the expression of the VM32E gene. From these results and our previously published data, it appears that the proper patterning of follicle cells, defined by Dpp and Egfr signaling pathways, controls the VM32E gene expression pattern. This may suggest that a fine tuning of the expression of specific eggshell structural genes could be part of the complex process that leads to a proper eggshell assembly.

Orientador: Eliana Cotta de Faria Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Ciencias Medicas Resumo: As análises laboratoriais estão sujeitas a diversos interferentes e, mesmo em indivíduos saudáveis, sofrem variações decorrentes de fatores biológicos e/ou laboratoriais, que podem gerar equívocos na interpretação dos resultados de exames. Com o aperfeiçoamento dos equipamentos e da implantação de rígidos sistemas de controle de qualidade, ocorreu uma sensível diminuição da variabilidade analítica e pós-analítica nos exames dos laboratórios clínicos. Conseqüentemente, o enfoque atual está na busca da origem das variações préanalíticas. Um componente biológico muito estudado é o intra-individual, que é inerente ao próprio indivíduo e modifica-se ao longo do tempo. o objetivo deste trabalho foi o de avaliar a ocorrência de ritmos biológicos em analitos bioquímicos séricos (correlacionando-os com a frequência de doenças) e analisar a prevalência de dislipidemias, assim como a regulação sérica da lipoproteína(a), numa população brasileira. Para tal foram realizados quatro estudos retrospectivos e um prospectivo de avaliação dos resultados de diversos exames laboratoriais de pacientes atendidos no Hospital das Clínicas da Unicamp, no período de 1996 a 2003. Os parâmetros analisados foram: ácido úrico, cálcio, colesterol, creatinina, fósforo, HDL-colesterol, LDL-colesterol, lipoproteína(a), triglicérides e uréia. Análises estatísticas foram realizadas utilizando-se o programa SAS e SPSS para análises descritivas e comparativas. Para a análise temporal foi utilizado o método Cosinor. Os valores encontrados para colesterol, HDL-colesterol, LDL-colesterol, triglicérides e Lp(a) estão fora dos recomendados pelo NCEP e adotados pela IIIDBSD em uma grande parcela da população estudada. Foram detectados ritmos sazonais nas determinações séricas de colesterol, HDL-colesterol, LDL-col~sterol, cálcio e creatinina. Além disso, os ritmos encontrados para colesterol e LDL-colesterol se associaram a uma maior freqüência de manifestações clínicas da aterosclerose na região estudada. Conclui-se que os valores observados para colesterol, HDL-colesterol, LDL-colesterol, triglicérides e Lp(a), tanto em adultos como crianças e adolescentes, indicam a altaprevalência de dislipidemias nesta região. Os bioritmos de colesterol, HDL-colesterol, LDL-colesterol, cálcio e creatinina devem ser valorizados como uma importante causa de variação pré-analítica no laboratório clínico e pelo fato dos ritmos dos lípides e lipoproteínas se associarem ao ritmo de prevalência de aterosclerose na região. Estes resultados resgatam a importância dos "relógios biológicos" na prática médica Abstract: Laboratory exams are subject to several interfering aspects and even healthy individuaIs may present variations, stemming from biological and/or laboratory factors, and generating problems in the outcome interpretation of exams. As a consequence of laboratory technological improvement, information technology and strict quality control systems, a marked decrease in analytical and post-analytical variability has taken place in clinical laboratories today. Accordingly, studies have preferably focused on the origin of pre-analytical errors. Among the biological components current1y being investigated there is the intra-individual, which is inherent to the individual and changes over time. The objective of this work was to evaluate the occurrence of biological rhythms in several biochemical serum parameters (correlating with atheroscleroses) and to measure the prevalence of dyslipidemia, as well as the modulation of serum lipoprotein(a), in a Brazilian population. For such four retrospective studies and a prospective one the results of severaI biochemical serum parameters of individuaIs registered at a referral University Hospital in Campinas - Unicamp,duringthe yearsof 1996to 2003 were evaluated. The analyzed parameters were: uric acid, calcium, cholesterol, creatinine, phosphorus, HDL-cholesterol, LDL-cholesterol, lipoprotein(a), triglycerides and urea. Statistical analyses were carried out by the SAS or SPSS programs for descriptive and comparative analyses. Temporal analyses were carried out using the Cosinor method. The values found for cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides and Lp(a) are out of the recommended ranges by NCEP and adopted by IIIDBSD in a great portion of the studied population. Seasonal rhythms was observed in serum cholesterol, HDL-cholesterol, LDL-cholesterol, calcium ~nd creatinine. The correlation between the rhythms of cholesterol and LDLcholesterol and the prevalence of manifestations of atherosclerosis strongly suggest their impact of seasonality on coronary artery disease. In conc1usion, the serum concentrations of cholesterol, HDL-cholesterol, LDL-cholesterol, triglycerides and Lp(a), so much in aduIts as children and adolescents, indicated the high prevalence of dyslipidemia in this area. The sazonality observed for cholesterol, HDLcholesterol, LDL-cholesterol, calcium and creatinine should be considered a significant cause of pre-analytical variation in these laboratory tests and also the rhythms of lipids and lipoproteins are associated with atherosc1erosis rhythmicity in the studied area. We have analysed the chromatin structure of the ade6 gene of Schizosaccharomyces pombe and its flanking regions both in the chromosome and in plasmids. The chromatin structure is independent of the chromosomal or extrachromosomal location. The ade6 gene contains eight precisely positioned nucleosomes on the 5' half, 'not positioned' nucleosomes around the 3' end and a nuclease-sensitive promoter region. Precisely positioned nucleosomes, but no nuclease-sensitive region were also detected on the ura4 gene in the chromosome and on a plasmid. The results show that S. pombe chromosomal and extrachromosomal genes have chromatin structures similar to those of S. cerevisiae and higher eukaryotes.

Orientador: Eliana Cotta de Faria Tese (doutorado) - Universidade Estadual de Campinas, Faculdade de Ciencias Medicas Resumo: Esta tese envolveu estudos com 287 mulheres, 69 não menopausadas e 218 pós-menopausadas; destas 84 em e 124 sem terapia de reposição hormonal (TRH), todas atendidas em hospitais públicos da Universidade Estadual de Campinas, Estado de São Paulo. As voluntárias foram recrutadas junto aos ambulatórios de dislipidemias (n=193), do Hospital das Clínicas Unicamp, e do ambulatório de menopausa (n=94), do CAISM/Unicamp. Seu intervalo de idades foi de 20 a 82 anos. As pós-menopausadas apresentavam idade acima de 40 anos e amenorréia por período superior a um ano. O grupo em terapia de reposição hormonal foi subdividido de acordo com o tipo de TRH em 2 subgrupos: pacientes em uso de estrógenos isoladamente (0.625mg/dia, n=48) ou pacientes em reposição hormonal combinada com acetato de medroxiprogesterona, (2,5mg/dia, 10% e 5mg/dia, 90%, n=36), por no mínimo um ano. Caracterizaram a metodologia a definição da menopausa, através do preenchimento de questionário sobre o tempo de amenorréia natural, a determinação do uso e tipo de terapia de reposição hormonal por meio de entrevistas, seguidas de exame médico clínico. Foi objetivo a determinação dos efeitos do uso da terapia de reposição hormonal oral no período pós-menopausal, estrogênica ou estrogênica associada à progestágenos sobre diversos marcadores séricos de oxidação no plasma. Como evento ponto-final da aterosclerose precoce determinou-se os efeitos da menopausa e da menopausa tratada com reposição hormonal sobre a aterosclerose precoce carotidiana e sua regulação metabólica. A abordagem de efeitos metabólicos da TRH foi realizada com a determinação após uso da terapia de reposição hormonal oral das atividades de proteínas reguladoras do metabolismo das lipoproteínas plasmáticas: a lípase hepática, a lipoproteína lipase, a proteína de transferência de colesteril-éster e a proteína de transferência de fosfolípides. Foram analisados também os seguintes parâmetros: colesterol, não HDL colesterol (NHDLcol), colesterol de lipoproteínas de alta densidade (HDLcol), colesterol de lipoproteínas de baixa densidade (LDLcol), triglicérides (TG), apolipoproteínas (AI e B 100), lipoproteína (a) Lp(a), autoanticorpos anti-LDL oxidada, anticorpos anti-epítopos proteicos da apolipoproteína B oxidada (anti-D, anti-D2 e anti-A); atividades das proteínas de transferência de colesteril-éster (CETP) e de fosfolípides (PLTP), da lipase hepática (LH), da lipoproteína lipase (LPL), a atividade séricas da catalase, determinação do nitrato, substâncias reativas ao ácido tiobarbitúrico (TBARS) e PCR séricos. O parâmetro radiológico medido foi o espessamento íntimo-médio da camada carotídiana (EIM) das artérias carótidas comuns direita e esquerda (ultra-sonografia Doppler). A análise estatística dos dados foi realizada através do programa SAS. Procedeu-se à correções para idade e IMC, quando indicado. A analise de regressão linear múltipla foi utilizada para acessar a influência dos diversos parâmetros bioquímicos e antropométricos sobre a EIM carotídea. Foram observados vários efeitos bioquímicos e antropométricos pró-aterogênicos da menopausa: aumento do EIM, do IMC, da medida da cintura e títulos de autoanticorpos anti-LDL oxidada e anti-D. A terapia de reposição hormonal apresentou efeitos modificadores benéficos reduzindo a lipase hepática (maior magnitude com a terapia conjugada), aumento de HDLcol, redução de autoanticorpos anti-D2 e aumento da concentração da catalase, (maior magnitude na terapia combinada). Outros marcadores de estresse oxidativo os nitratos, as substancias reativas ao ácido tiobarbitúrico e os lipoperóxidos não se modificaram com a TRH. Nas análises multivariadas a TRH conjugada e estrogênica modulou a EIM através de três fatores: via triglicérides, CETP (negativo) e lipoperóxidos (ao contrário do grupo sem TRH, com sete fatores de regulação); a terapia estrogênica atuou apenas via TG. Houve influência positiva do tratamento sobre a regulação positiva pela PCR a qual desapareceu. Este estudo reitera o risco aumentado para a doença cardiovascular (DCV) pelo aumento de um conjunto de fatores de risco na mulher em pós-menopausa, fato já demonstrado em estudos prévios. A TRH foi benéfica do ponto de vista de melhora do perfil de lípides. Modificou favoravelmente a lípase hepática aumentando o colesterol da HDL, lipoproteína anti-aterogênica. A redução de autoanticorpos contra a oxidação apoproteica B100 e o aumento da atividade sérica da catalase demonstram capacidade antioxidante maior e dredução do estersse oxidativo plasmático. Não menos importante e apesar do efeito ter sido insuficiente amostra populacional para alterar a EIM, a TRH modificou a modulação da aterosclerose precoce no sentido de maior ateroproteção Abstract: This thesis was composed of studies conducted on 287 women: pre menopausal (69) and post (218); the last with (n=84, WHRT) and without (n=134, WTRT) hormone replacement therapy (HRT), attended at the UNICAMP university hospitals, São Paulo state. The volunteers were recruited from Hospital de Clínicas (n=193) and CAISM hospital (n=94). They aged from 20 to 82 years (y). Postmenopausal women were 40y old and above and presented amenorrhea for at least 1 year. WHRT women were subdivided in 2 groups: one using conjugated estrogens (0.625mg/day, n=48) or estrogen associated with medroxyprogesterone acetate (2.5, 10% of all or 5mg/day 90% of all, n=36). The methodology was characterized by the menopause definition and through questionnaires and a clinical exam. The objective of this study was to verify if HRT, estrogenic or combined, modified plasmatic oxidative markers. The end-point for atherosclerosis was the measurement of common carotid intima-media thickness (IMT), as well as its metabolic regulation. The study also dtermined the activities of several proteins of lipid metabolism: lipoprotein lipase, hepatic lipase, cholesteryl ester and phospholipid transfer protein under HRT estrogenic or combined. ELISA, nephelometric, enzymatic and radiometric methods were used to determine several parameters: cholesterol, non HDL cholesterol (NHDLchol), HDL-cholesterol, LDL-cholesterol triglycerides (TG), apolipoproteins (AI e B 100), lipoprotein (a), Lp(a), autoantibodies against oxidized LDL, epitopes of oxidized apolipoprotein B100 (anti-D, anti-D2 e anti-A); activities of CETP, PLTP, HL LH and LPL, catalase, nitrates, TBARS, lipid peroxides, CRP. The radiologic common carotid intima-media thickness was done by Doppler ultrasound. The data were analyzed by the SAS statistical package. Multiple linear regression analyses were used to assess the influence of diverse biochemical markers on carotid IMT. In this study several postmenopausal anthropometric and biochemical effects were pro-atherogenic: increases in IMT, BMI (body mass index), WC (waist circumference), antibodies against oxidized LDL and anti-apoD antibodies titers. HRT showed beneficial actions, decreasing HL activity, reducing anti-D2 antibody titers, increasing HDLchol and catalase activity. The nitrate concentration, TBARS and hydroperoxides showed no changes with HRT. HRT improved the women¿s lipid profiles but not ApoAI and B100. decreased hepatic lipase and increased HDLchol, an anti-atherogenic lipoprotein, reduced anti-D2 and increased catalase activity. Although HRT was insufficient to modify IMT, the multivariate analysis demonstrated that conjugated and estrogen HRT modulated IMT through triglycerides concentration, CETP (negative) and lipid peroxides a situation differently from non-treated women that presented 7 modulators; under estrogenic treatment only TG regulated IMT. As well the hormone treatment influenced favorably excluding the effects positive of CRP. This study reinforces the higher risk of CAD in post-menopausal women and the beneficial action of HRT by improving lipid profiles. It changed favorably HL, HDL-cholesterol, decreased antibodies against oxidized apoB100 and increased catalase activity indicating reduced oxidative stress; not less important are the results showing that HRT although not changing carotid IMT, modified beneficially the relationship of precocious atherosclerosis and its modulators suggesting an atheroprotective action Doutorado Patologia Clínica Doutor em Ciências Médicas

Orientador: Eliana Cotta de Faria Dissertação (mestrado) - Universidade Estadual de Campinas, Faculdade de Ciencias Medicas Resumo: Está bem estabelecida na literatura especializada a associação inversa entre as concentrações plasmáticas de colesterol das lipoproteínas de alta densidade (HDL-C) e a incidência de doença arterial coronariana (DAC). Além de propriedades anti-oxidante, anti-inflamatória e anti-trombótica, a HDL participa do transporte reverso de colesterol, via pela qual o colesterol é captado das lipoproteínas e das membranas células periféricas e transportado ao fígado para sua excreção na forma livre ou de ácidos biliares. A lipase hepática (LH) possui função crucial no transporte reverso do colesterol, por sua atividade lipolítica e pela função de ligante à lipoproteínas facilitando sua captação tissular. A proteína de transferência de ésteres de colesterol (CETP), e mesma importância metabólica, promove a troca de ésteres de colesterol por triglicérides entre a HDL e as lipoproteínas ricas em triglicérides. Mutações nos genes que codificam estas proteínas têm sido muito estudadas para se compreender a função destas no metabolismo lipídico. O modelo experimental da hiperalfalipoproteinemia tem sido utilizado no decorrer dos últimos anos com o intuito de elucidar os mecanismos de ação da HDL e das proteínas reguladoras do seu metabolismo. A hiperalfalipoproteinemia é caracterizada pelo aumento das concentrações de HDL-C e é causada principalmente por deficiências genética de CETP e/ou LH. Os objetivos desta dissertação foram o de se estabelecer à modulação da hiperalfalipoproteinemia sobre os parâmetros antropométricos, bioquímicos, moleculares (¿514C/T do gene da LH e I405V do gene da CETP) e radiológicos (espessura da camada íntima média de carótidas) em uma amostra populacional brasileira. O estudo foi conduzido em 291 voluntários de ambos os sexos, classificados como hiperalfalipoproteinemicos (Hiper-A), HDL-C =68mg/dL, ou controles, HDL-C
