Background: Cytoreductive radical prostatectomy (cRP) has emerged as a promising therapeutic approach for low-volume metastatic hormone-sensitive prostate cancer (mHSPC), but the best candidates for cRP are still unknown. This study aims to explore the potential value of 18F-PSMA-1007 PET/CT-derived quantitative volumetric tumor parameters in cRP treatment selection among patients with low-volume mHSPC. Methods: A total of 122 patients with primary low-volume mHSPC who underwent 18F-PSMA-1007 PET/CT followed by systemic therapy alone or plus cRP were included. The whole-body PSMA-derived tumor volume (PSMA-TV) was defined as the total volume of whole-body PSMA-avid tumor lesions, and prostate PSMA-TV was defined as the volume of prostate PSMA-avid tumor lesions. Spearman's correlation was used to analyze the relationships between whole-body PSMA-TV and clinicopathological characteristics. The primary endpoint was progression-free survival (PFS), and Cox regression analyses were performed to explore the independent predictors for PFS. Results: Among 122 patients, 37 (30.32%) underwent systemic therapy plus cRP. The median and optimal cutoff values of the whole-body PSMA-TV were 71.68 cm3 (41.28–157.41 cm3) and 78.57 cm3, respectively. Whole-body PSMA-TV was positively correlated with prostate-specific antigen (PSA), and patients with nonregional lymph node (NRLN) metastases had a greater whole-body PSMA-TV (P = 0.001). Cox regression analyses revealed that cRP, lower whole-body PSMA-TV and the absence of NRLN metastases were associated with better PFS (all P < 0.05). Subgroup analyses revealed that patients with a low whole-body PSMA or no NRLN metastases had a significant improvement in PFS for cRP versus no cRP (HR: 8.26; 95% CI: 2.72–25.06, P = 0.001; HR: 2.71; 95% CI: 1.25–5.93, P = 0.018). Moreover, among patients with higher prostate PSMA-TV and prostate PSMA-TV/whole-body PSMA-TV, cRP also significantly prolonged PFS compared with those without cRP (HR: 3.49; 95% CI: 1.49–8.18, P = 0.004; HR: 8.54; 95% CI: 2.47–29.50, P = 0.013). Conclusion: In management of primary low-volume mHSPC, whole-body and prostate PSMA-TV evaluations based on 18F-PSMA-1007 PET/CT could be helpful to identify the most suitable candidates for cRP. Background: Bernard–Soulier syndrome (BSS) is a rare inherited macrothrombocytopenia, usually autosomal recessive, which is characterized by prolonged bleeding, thrombocytopenia, and abnormally large platelets. Methods: For more than 6 years, we misdiagnosed a patient with BSS without an obvious bleeding tendency as having idiopathic thrombocytopenia purpura (ITP), prior to obtaining a genetic analysis. On admission, routine hematology showed a platelet count of 30 × 109/L and mean platelet volume (MPV) of 14.0 fL. Results: Whole-exome sequencing revealed two likely pathogenic heterozygous mutations (c.95_101del and c.1012del) in GP1BA. Flow cytometry analysis of platelet membrane glycoproteins indicated that the expression of GP1b was 0.28% of the normal level. Platelet aggregation tests indicated that platelet aggregation was inhibited by ristocetin- (1.7%), ADP- (14.5%), and arachidonic acid- (5.6%) induced platelet aggregation. A literature review identified reports on 53 mutations in the GP1BA gene in 253 patients, 29 mutations in the GP1BB gene in 90 patients, and 32 mutations in the GP9 gene in 114 patients. The Jinchuan Ni-Cu sulfide deposit in the Longshoushan terrane is among the world's largest magmatic sulfide deposits. This study uses magnetotelluric (MT) survey data imaging combined with previous geophysical data to investigate Segment III of the deposit. The image of MT data reveals a significant low-resistivity anomaly ~200 m beneath Segment III, aligning with known ore bodies, and an additional anomaly to the north, indicating further exploration potential. These findings highlight the deep-seated intrusions and their connection to surface mineralization, emphasizing the critical role of ultramafic rock encapsulation and fault-controlled morphology in ore body formation. The newly identified northern anomaly, resembling the Segment III ore bodies, suggests a promising target for future exploration. Integrating MT surveys with other geophysical methods enhances the understanding of subsurface structures and mineralization, providing a robust framework for discovering concealed mineral deposits and improving exploration efficiency. Objective To analyze the impact of mini plate and cannulated screw internal fixation on joint function recovery in patients with posterior malleolus fracture. Methods A total of 150 patients with posterior malleolus fractures, treated at our hospital from March 2021 to June 2023, were included in this study. They were divided into two groups using the odd-even number method. The control group consisted of 75 patients who underwent cannulated screw internal fixation, while the study group comprised 75 patients who received mini plate internal fixation. Clinical indicators, ankle range of motion, ankle function, and health status were compared and analyzed between the two groups. Additionally, levels of inflammatory factors, postoperative complications, and clinical efficacy were assessed. Results In terms of operation time and intraoperative blood loss, there was no statistically significant difference between the two groups (P > 0.05). However, the study group exhibited shorter ambulation days, fracture healing time, and hospitalization days compared to the control group (P < 0.05). Moreover, the study group demonstrated significantly improved ankle dorsiflexion, ankle plantar flexion, foot varus and foot valgus range of motion compared to the control group (P < 0.05). Additionally, higher AOFAS score and KPS score were observed in the study group as compared to the control group (P < 0.05). Furthermore, levels of IL-6, IL-8 and CRP were lower in the study group than in the control group (P < 0.05). The incidence of postoperative complica- tions was also lower in the study group than in the control group (P < 0.05). Conclusion Mini plate internal fixa- tion for posterior malleolus fracture yields ideal outcomes by promoting improvement in clinical indicators and ankle range of motion while effectively enhancing ankle function, reducing inflammatory reaction as well as minimizing postoperative complications. Objective To explore the role of Echinococcus granulosus aquaporins (EgAQPs) in the accumulation of hydatid fluid. Methods The secondary hydatids cultured in vitro and inoculated in BALB/c mice were collected respectively, and the relative transcription expression levels of 12 EgAQPs genes from the hydatid cysts cultured in vitro and in mice were detected by RT-qPCR. Six gene coding region fragments of EgAQP, EgAQP1, EgAQP3, EgAQP4-1, EgAQP4-4 and EgAQP9-1 were amplified using cDNA templates. The other 6 genes coding region fragments of EgAQPs (EgAQPAn.G, EgAQPFA-CHIP, EgAQP4-2, EgAQP4-3, EgAQP4-5 and EgAQP9-2) were amplified with chemical synthesis. The water permeability of these 12 EgAQPs genes were detected in Xenopus laevis (X. laevis), and the expression of EgAQPs on the oocytes membranes was detected by Western blotting. Results The volume of hydatid cultured in vitro and in vivo was increased gradually with the extension of culture time, and there was more and more transparent hydatid fluid in hydatid cyst. The results of RT-qPCR showed that the relative transcription levels of EgAQPs genes in the hydatid cysts were very low or even undetectable except for EgAQP3 and EgAQP9-1 at the beginning of culture in vitro. There were no significant differences in relative volume change rates and water permeability coefficients of X. laevis oocytes injected with EgAQPs genes when compared with the negative control, and these EgAQPs genes did not show water channel functions in the oocytes. Western blot assay displayed that there was no expression of EgAQPs genes in total, cytoplasmic and membrane protein on the oocytes. Conclusion The transcription levels of EgAQPs genes are quite low in the encystation process of protoscolex, and their heterologous expression do not show water channel functions in X. laevis oocytes.
