Exposure to air can cause amine solutions in CANada Deuterium Uranium (CANDU) reactor secondary coolant circuit feed tanks to absorb carbon dioxide (CO2). Likewise, carbon dioxide can be absorbed directly into the amine-containing secondary coolant by air ingress during shutdown, lay-up, and startup. Sampling operations, including transferring the sample to the laboratory and subsequent analyses, can also provide opportunities for CO2 contamination. This paper reports the results of laboratory and chemical modeling studies to examine the effects of CO2 contamination on aqueous morpholine solutions. The chemistry of CO2 uptake by feed tanks containing up to 50 wt% (11.5 mol·kg-1) morpholine at 25°C was modeled using the OLI Studio 9.5.2 chemical equilibrium model, and the speciation was confirmed by 13C nuclear magnetic resonance spectroscopy (NMR) measurements. The effects of CO2 contamination on the pH of the secondary coolant containing 60 ppm (0.006 wt%, 7.00 × 10-4 mol·kg-1) morpholine and the resulting effects on the solubility of magnetite and nickel oxide from 25°C and 250°C at steam saturation were modeled as a function of CO2 loading using the Electrical Power Research Institute chemical modeling software MULTEQ v.8. The chemical modeling calculations show that concentrated alkaline morpholine solutions at room temperature and pressure would be expected to have a strong tendency to absorb CO2 and have additional uptake abilities due to the formation of morpholine carbamates. For dilute morpholine solutions at room temperature and pressure, the solutions are still sufficiently alkaline to absorb enough CO2 to cause a measurable change in the pH of the secondary coolant. This effect was shown to be negligible under reactor operating conditions. The absorption of CO2 would potentially have the most effect on either unprotected feed tanks or during lay-up conditions in the steam generators, as it could depress the pH of the lay-up solution and adversely affect the rate of corrosion in the internal components of the steam generators (e.g., carbon steel materials). The 13C NMR measurements on samples of 50 wt% aqueous morpholine solutions from feed tanks at the Ontario Power Generation's Pickering Nuclear Generating Station found that CO2 was below the 0.02 wt% detection limit, and suggest that the procedures used to avoid CO2 contamination in feed tanks are effective. The 13C NMR was shown to be an effective tool for monitoring CO2 uptake by morpholine solution in the feed tanks under conditions in which they may have undergone abnormal exposure to air. The nanocrystal (NC) technology has become one of the most commonly used strategies for the formulation of poorly soluble actives. Given their large specific surface, NCs are mainly used to enhance the oral absorption of poorly soluble actives. Differently from conventional nanoparticles, which require the use of carrier materials and have limited drug loadings, NCs' drug loading approaches 100% since they are formed of the pure drug and surrounded by a thin layer of a stabilizer. In this work, we report the covalent decoration of curcumin NCs with folic acid (FA) using EDC/NHS chemistry and explore the novel systems as highly loaded "Trojan horses" to target cancer cells. The decorated NCs demonstrated a remarkable improvement in curcumin uptake, exhibiting enhanced growth inhibition in cancer cells (HeLa and MCF7) while sparing healthy cells (J774A.1). Cellular uptake studies revealed significantly heightened entry of FA-decorated NCs into cancer cells compared to unmodified NCs while also showing reduced uptake by macrophages, indicating a potential for prolonged circulation in vivo . These findings underline the potential of NC highly loaded nanovectors for drug delivery and, in particular, for cancer therapies, effectively targeting folate receptor-overexpressing cells while evading interception by macrophages, thus preserving their viability and offering a promising avenue for precise and effective treatments.

Adeleorinid parasites commonly infect turtles and tortoises in nature. Currently, our knowledge about such parasites is extremely poor. Their characterization is based on morphological and molecular approaches using the 18S rDNA molecular marker. However, there is a limitation with the 18S rDNA due to its slow rate of evolution. For that reason, the goals of this study were to 1) design primers for new molecular mitochondrial markers to improve the phylogenetic reconstructions of adeleorinid parasites and 2) to determine the morphological and genetic diversity of Haemogregarina infecting turtles and tortoises in Colombia. Turtles from 16 species representing six families were examined for the presence of haemoparasites. We analyzed 457 samples using PCR, and 203 of them were also analyzed by microscopy. Using a mitochondrial genome of Haemogregarina sequenced in this study, we designed primers to amplify fragments of the cytochrome oxidase I (coxI), cytochrome oxidase III (coxIII), and cytochrome b (cytb) mitochondrial markers in adeleorinid parasites. Lineages obtained from nuclear and mitochondrial molecular markers clustered according to the turtle lineages from which they were isolated. It is noteworthy that we found different evolutionary lineages within the same morphotype, which may indicate heteroplasmy and/or cryptic diversity in Haemogregarina. Due to this situation, we could not make a species delimitation, even when integrating the different lines of evidence we had in this study. However, the primers presented here are useful for diagnosis and, moreover, according to the available information, all three genes retain phylogenetic signals; thereby fragments amplified can be used in reconstructing evolutionary relationships. The irritable bowel syndrome (IBS) is a complex disorder in which psychosocial, cultural and biological factors, interact. Recent knowledge in the pathophysiology of IBS, seem to combine issues such as a low grade inflammation or immune activation and dysbiosis that can trigger or exacerbate IBS. On the other hand, stress mediated through the hypothalamic-pituitary-adrenal axis can produce motility abnormalities that can modify the microbiota as well, with the subsequent immune activation in the mucosa and stimulation of nerve terminals, generating symptoms of IBS. Also, we speculate that, stress, dysbiosis or an underlying genetic predisposition, may increase the epithelial permeability leading to a contact between pathogens-associated molecular patterns and toll-like receptors in the deeper layers of the gut, developing a host immunity response and IBS generation. We believe that the role of toll-like receptors in IBS and elucidating the communication processes between the immune and the nervous system, warrant future research.

Background: Development of animal models has been one of the most remarkable achievements for Irritable Bowel Syndrome (IBS) research. These models need to exhibit face validity, construct and predictive validity, as well as internal (replicability) and external validity (generalizability). Among these models, chronic stress induced by water avoidance exposure (WA) has been validated in rats through increasing visceral hypersensitivity, motility impairment, anxiety and, colonic immune activity, all related to IBS., Objective: To assess the external validity of WA indirectly on colonic motility in Wistar rats., Method: Ten male-Wistar rats were exposed to WA and compared with ten Wistar rats not exposed (controls). At the end of each exposure, the quantity of fecal pellets were determined and considered as a sign of autonomic regulation of colonic motility. Data was analyzed with a general linear model for repeated measures., Results: Rats exposed to WA had higher number or pellets than controls: 7.46 ± 0.45 (95%CI: 6.51, 8.41) vs. 2.88 ± 0.45 (1.93, 3.83), p < 0.001. The higher number of pellets was related to WA exposure as there were no other significant interactions. In both groups, the number of pellets was higher during the first day and then decreased progressively., Conclusions: Chronic stress induced through WA in Wistar rats, exhibits external validity as an experimental model for IBS research and our findings of increased number of fecal pellets coincide with the appearance hypermotility related to IBS. The model is optimum for research studies on this disorder.
