Metabolic dysfunction-associated fatty liver disease (MAFLD) is a newly proposed definition of fatty liver disease (FLD) independent of excessive alcohol consumption (EAC) and hepatitis viral infection. Evidence on the mortality risk in different types of FLD [nonalcoholic FLD (NAFLD), alcoholic FLD (AFLD), and MAFLD] is sparse, hindering the identification of high-risk populations for preferential clinical surveillance.A total of 11,000 participants in the Third National Health and Nutrition Examination Survey were enrolled. Participants were categorized into three groups [FLD( - ), MAFLD( - ), and MAFLD( +)] according to FLD and MAFLD criteria, and further categorized into six groups by EAC. Multivariate Cox proportional hazard model was used to estimate the risk of all-cause, cardiovascular-related, and cancer-related mortality.During a median follow-up of 23.2 years, a total of 3240 deaths were identified. Compared with FLD( - )/EAC( - ) participants, MAFLD( +) individuals had higher all-cause mortality risk [hazard ratio (HR) = 1.28, 95% confidence interval (CI) = 1.18-1.39] regardless of EAC status [MAFLD( +)/NAFLD: HR = 1.22, 95%CI = 1.11-1.34; MAFLD( +)/AFLD: HR = 1.83, 95%CI = 1.46-2.28], while not for MAFLD( - ) individuals. Furthermore, diabetes-driven-MAFLD had higher mortality risk (HR = 2.00, 95%CI = 1.77-2.27) followed by metabolic dysregulation-driven-MAFLD (HR = 1.30, 95%CI = 1.06-1.60) and overweight/obesity-driven-MAFLD (HR = 1.11, 95%CI = 1.00-1.22). Additionally, MAFLD( - ) participants with elevated fibrosis score were also associated with statistically significantly higher mortality risk (HR = 3.23, 95%CI = 1.63-6.40).Utilizing a representative sample of the US population, we proved the validity of MAFLD subtype and fibrosis score, rather than the traditional definition (NAFLD and AFLD), in the risk stratification of FLD patients. These findings may be applied to guide the determination of surveillance options for FLD patients.

Acute Myeloid Leukemia (AML) is a highly lethal blood cancer arising due to aberrant differentiation of haematopoietic stem cells. Here we obtained 3D genome organization maps by Hi-C in the CD34+ haematopoietic stem cells from three healthy individuals and eight individuals with AML, and found that AML have increased loops to oncogenes compared with normal CD34+ cells. The MEIS1 oncogenic transcription factor is regulated by a Frequently Interacting Region (FIRE). This FIRE is only present in normal bone marrow samples, and four of eight AML sample. FIRE presence is associated with MEIS1 expression. CRISPR excision of a FIRE boundary led to loss of MEIS1 and reduced cell growth. Moreover, MEIS1 can bind to the promoter of HOXA9, and HOXA9 shows gain of Acute Myeloid Leukemia-specific super-enhancers that loop to the HOXA9 promoter.SignificanceWe found that Acute Myeloid Leukemias have more chromatin loops to oncogenes compared with normal blood stem cells. We identified heterogeneity in chromatin interactions at oncogenes, and heterogeneity in super-enhancers that loop to oncogenes, as two key epigenetic mechanisms that underlie MEIS1 and HOXA9 oncogene expression respectively.

To examine the effect of subcutaneous injection of insulin-like growth factor-1 (IGF-1) on the expression of the amyloid protein (Aβ1–40), α-secretase (ADAM10), β-secretase (BACE1), and γ-secretase (PS1) in APP/PS1 double transgenic mice. APP/PS1 double transgenic mice and wild-type mice were divided into wild-type group, wild-type therapy group, transgenome group, and transgenic therapy group. Subcutaneous injection of IGF-1 (50 μg/kg day) was administered once daily to the wild-type therapy group and transgenic therapy group for 8 weeks, respectively. The expression of the Aβ1–40 in the cortex and hippocampus was detected by immunohistochemistry 8 weeks after administration. Periodontitis is a highly prevalent infectious disease associated genetically with coronary heart disease (CHD). The effects of proprotein convertase subtilisin/kexin type 9 (PCSK9), a critical regulator of CHD, on periodontitis have not been studied to date. Here, we found that PCSK9 expression was increased in periodontitis patients and Porphyromonas gingivalis (Pg)-infected mice. Loss of PCSK9 attenuated Pg-induced periodontal bone loss in mice. First, PCSK9 deficiency reduced the release of inflammation-associated cytokines, such as tumor necrosis factor alpha (TNF-α) and interleukin 1β, in vitro and in vivo. Second, its deficiency enhanced Pg and endotoxin clearance during Pg invasion in part by upregulating CD36 and low-density lipoprotein receptor (LDLR), respectively. However, after berberine treatment, periodontal bone regeneration in the PCSK9 knockout group was significantly lower than that in wild-type. This was because PCSK9 overexpression promoted osteogenic differentiation of periodontal ligament stem cells (PDLCs) prechallenged by TNF-α. Furthermore, PCSK9 could rescue PDLC osteogenesis by repressing the NF-κB signaling pathway by interacting with TRAF2. These results suggest that PCSK9 may be a potent drug target for treating periodontitis.

Many oncogenes, such as MEIS1 and HOXA9, are overexpressed in some but not all cancers. We identified two key epigenetic mechanisms underlying this heterogeneity in oncogene expression in Acute Myeloid Leukemia. Background: Coffee is a popular beverage in Korea recent years. The purpose of this study was to investigate the relationship between coffee consumption and the risk of metabolic syndrome in Korean adults based on the 2010 Korean National Health and Nutrition Examination Survey (KNHANES V-1). Methods: Dietary intake status and the factors of metabolic syndrome were assessed. Three groups (no coffee consumption, moderate intake, and high intake) were divided into tertile according to black coffee cream (include brewed coffee) consumption per day. Results: Our results showed that the Tertile 3 group consumed more calories from fat, and niacin was higher than in the Tertile 1 and Tertile 2 group. INQ for protein and vitamin B1 was significantly higher in no coffee consumption group than the other groups and in Tertile 3 exhibited significantly higher niacin intake. The subjects in Tertile 3 showed significantly higher consumption in grain and oil intake, and Tertile 1 group showed higher consumption in milk and dairy products. In the logistic regression analysis, adjusting for sex, age, energy intake, smoking, and drinking, being in the high coffee consumption group (Tertile 3) was significantly and inversely associated with abdominal obesity (OR = 0.76, CI = 0.71–0.82), hypertension (OR = 0.70, CI = 0.54–0.87), high glucose(OR = 0.71, CI = 0.61–0.86). However, no significant association was found between coffee consumption and metabolic syndrome. Conclusion: Coffee consumption has not a considerably relationship with nutrient intake. Appropriate consumption of coffee may have potentially helpful effects on certain metabolic risk factors, such as abdominal obesity, hypertension, and high glucose. The purpose of this research was to investigate the relationship between white rice consumption and dietary intake, the risk of metabolic syndrome in Korean based on (1998), (2007~2009), (2013) KNHANES. This study included 25,799 subjects who were age 10 or over. Three groups(low, medium, high) were divided according to white rice consumption (per day). In the low group, the meat and oil intake were significantly higher than the high group. In the survey year, the BMI, waist circumstances, hypertriglyceridemia and fasting blood glucose in the high group were higher than other groups, however the and survey year showed no significant results. In the survey year the risk of waist circumstances was higher in the high white rice consumption group and also had high risk with hypertriglyceridemia. Since the lower white rice consumption in than survey year we found no significant results. But in the 6th survey year with the lowest white rice consumption the risk of high diastolic pressure was reduced in high group. Because continuously reduced white rice consumption meantime showed more intake of meat and oil, it can be concluded that defensive effects with the Korean health.

Background: Several studies have previously focused on associations between the (GT)n repeat polymorphism of the heme oxygenase-1 (HO-1) gene promoter region and risk of cancers, but results are complex. We conducted the present meta-analysis to integrate relevant findings and evaluate the association between HO-1 (GT)n repeat polymorphism and cancer susceptibility. Materials and Methods: Published literature was retrieved from the PubMed/MEDLINE, EMBASE and ISI Web of Science databases before November 2013. For all alleles and genotypes, odds ratios were pooled to assess the strength of the associations using either fixed-effects or random-effects models according to heterogeneity. Subgroup analysis was conducted according to ethnicity and histopathology. Results: A total of 10 studies involving 2,367 cases and 2,870 controls were identified. The results showed there was no association between HO-1 (GT)n repeat polymorphism and the cancer risk both at the allelic and genotypic level. However, in the stratified analysis, we observed an increased risk of squamous cell carcinoma in persons carrying the LL genotype and the LL+LS genotype as compared with those carrying the SS genotype. When the LS and SS genotypes were combined, the odds ratio for squamous cell carcinoma in LL-genotype carriers, were also significantly increased. No publication bias was observed. Conclusions: The LL genotype and L-allele carrying genotypes (LL+LS) of HO-1 (GT)n repeat polymorphism are potential genetic factors for developing squamous cell carcinoma. More large and well-designed studies are required for further validations.

Background. Studies on the effect of intensive insulin therapy (IIT) in septic patients with hyperglycemia have given inconsistent results. The primary purpose of this meta-analysis was to evaluate whether it is effective in reducing mortality.Methods. We searched PubMed, Embase, the Cochrane Library, clinicaltrials.gov, and relevant reference lists up to September 2013 and including randomized controlled trials that compared IIT with conventional glucose management in septic patients. Study quality was assessed using the Cochrane Risk of Bias Tool. And our primary outcome measure was pooled in the random effects model.Results. We identified twelve randomized controlled trials involving 4100 patients. Meta-analysis showed that IIT did not reduce any of the outcomes: overall mortality (risk ratio [RR] = 0.98, 95% CI [0.85, 1.15],P=0.84), 28-day mortality (RR = 0.66, 95% CI [0.40, 1.10],P=0.11), 90-day mortality (RR = 1.10, 95% CI [0.97, 1.26],P=0.13), ICU mortality (RR = 0.94, 95% CI [0.77, 1.14],P=0.52), hospital mortality (RR = 0.98, 95% CI [0.86, 1.11],P=0.71), severity of illness, and length of ICU stay. Conversely, the incidence of hypoglycemia was markedly higher in the IIT (RR = 2.93, 95% CI [1.69, 5.06],P=0.0001).Conclusions. For patients with sepsis, IIT and conservative glucose management show similar efficacy, but ITT is associated with a higher incidence of hypoglycemia.

This paper carried out temperature analysis in the working state of the electron gun structure, the thermal deformation under high temperature and thermal stress analysis. Firstly, from the steady state temperature cloud chart of the electron gun we can see the cathode temperature is 985 °C, shadow grid temperature is same as cathode temperature, and the controlled-grid temperature is as high as 540 °C. Secondly, on the basis of thermal analysis, the coupled thermo-mechanical simulation technology is broken through; further thermal stress and thermal deformation at high temperature are analyzed. Thermal stress analysis results show that the maximum stress point is at contact area of cathode and its supporting tube. And the thermal deformation result shows that at high temperature under the action of thermal deformation, the axial distance of cathode-grid distance reduced 0.078 mm. Based on thermal deformation, the development personnel of microwave tube can use simulator to analysis the influence of thermal deformation to the electron gun design parameters, which can provide theoretical basis for tube design.

The pre-stressed modal and random vibration of electron gun are calculated by finite element software ANSYS Workbench. The results show that electron gun under the excitation of radial load (X direction) can be in danger of disable structure. Some measures can be taken to avoid electron gun failure due to resonance, such as replacing the materials, changing the structure of weak links and changing TWT installation to make external incentives along the axial direction (Y direction). The analysis results can provide effective reference and guidance for TWT development.

A nonlinear transient three-dimensional finite element model for the laser welding process is developed to investigate the temperature and residual stress distribution. All the major physical phenomena associated to the laser welding process, including temperature dependent thermo-physical and mechanical properties are taken into account in the model. The heat input to the model is assumed to be a Gaussian heat source. Results show that the temperature distribution of laser welding is consistent with the actual, and along-welding line longitudinal stress and transverse stress are retained in the weld zone after laser welding and cooling to the room temperature.

Based on analysis of five different conditions’ full scale model tests of the fastener tubular steel scaffolding, the paper introduces that notional lateral loads can imitate the effect which generalized imperfections of the falsework(initial imperfections, node semi-rigid and so on) give the bracket’s stability capacity. According to the finite element software ANSYS, the falseworks’ stability capacities of different conditions have been imitated and analysed, Analysis results compare with test results. The final result indicates that it is reasonable that notional lateral loads in certain scope can imitate generalized initial imperfection. Research results can be used for on-site t technical personnels’ design and safety control.

Vibration capability and reliability is necessarily to be considered for electronic components design. In this paper, the vibration characteristics of electronic components were studied using FEM, for example, gridded electron gun. The dynamical mode of girded electron gun was established. Modal frequency and model obtained by simulation shows tendency as tested experiment results, verifying the correctness of the simulation model and method used. The Parameters showed an inherent property of structure, which provided a theoretical basis for production design, the optimization of parameters and the vibration-resistance reliability improvement of microwave tube. In the aspects of electronic components reliability analysis, the variations of production capability parameters under various stress environments can be gained by FEM simulation technology to evaluate environment adaptability and reliability.

BACKGROUND A model of simulated Alzheimer's disease (AD) induced by aggregated amyloid protein (Abeta(1-40)) was built in Wistar rats to observe the behavioral and pathological changes of Abeta(1-40) and the effect of hypodermic insulin injected on the function of study and memory and the expression of Abeta(1-40) from the CA1 area of the hippocampus. METHODS Experimental groups were as follows: contrast, simulated AD model, contrast of Nacl, and insulin treated. The simulated AD model was built by microinjection of aggregated Abeta(1-40) at the CA1 area of the hippocampus, and was hypodermically injected with 0.9% NaCl (1 ml/kg) and insulin (0.1 U/kg) separately the next day. Two weeks after the modeling, the four groups were tested with water maze about the study and memory function of rats. Three weeks after the injection, the expression of Abeta(1-40) at the CA1 area of the hippocampus was examined by pathological tests (HE, Congo red) and immunohistochemical methods. RESULTS The study and memory abilities of rats were ameliorated significantly by the place navigation test and the spatial probe test after the application of insulin. Insulin could decrease the expression of Abeta(1-40) at the CA1 area of the hippocampus to reduce the pathological damage of Abeta(1-40) to the hippocampal area of rats. CONCLUSIONS The injection of aggregated Abeta(1-40) to the hippocampal area could simulate the behavioral and pathological features of AD such as the difficulty of study and memory and the damage to neurons. Insulin is effective to improve the function of study and memory and amend the pathological damage of simulated AD model rats. The results give a experimental proof of insulin in the clinical treatment of AD.

Along with the infrared technical rapid development, infrared radiation (IR) thermal imaging technology is widely applied in device and circuit reliability research. For device and circuit reliability study, IR thermal imaging technology is best for getting the peak temperature and temperature distribution. With the peak temperature and temperature distribution, the thermal information of the devices were evaluated and optimized to meet the design requirement. Secondly, IR thermal imaging technology is used to position hotspot for failures analysis, example for especially increases in elevated leakage currents. However, IR thermal imaging technology can provide evidence of excessive temperature rise due to power dissipation. It can identify root-cause of failures at temperatures below expectations.

Epithelial ovarian carcinoma is the most lethal gynecological cancer due to its silent onset and recurrence with resistance to chemotherapy. Overexpression of oncogene c-Myc is one of the most frequently encountered events present in ovarian carcinoma. Disrupting the function of c-Myc and its downstream target genes is a promising strategy for cancer therapy. Our objective was to evaluate the potential effects of small-molecule c-Myc inhibitor, 10058-F4, on ovarian carcinoma cells and the underlying mechanisms by which 10058-F4 exerts its actions. Using MTT assay, colony formation, flow cytometry and Annexin V FITC assays, we found that 10058-F4 significantly inhibited cell proliferation of both SKOV3 and Hey ovarian cancer cells in a dose dependent manner through induction of apoptosis and cell cycle G1 arrest. Treatment with 10058-F4 reduced cellular ATP production and ROS levels in SKOV3 and Hey cells. Consistently, primary cultures of ovarian cancer treated with 10058-F4 showed induction of caspase-3 activity and inhibition of cell proliferation in 15 of 18 cases. The response to 10058-F4 was independent the level of c-Myc protein over-expression in primary cultures of ovarian carcinoma. These novel findings suggest that the growth of ovarian cancer cells is dependent upon c-MYC activity and that targeting c-Myc-Max heterodimerization could be a potential therapeutic strategy for ovarian cancer.

Microwave tubes for communication must have high reliability, high power, and a long life of 10 to 15 years. Cathode as the electric source module in microwave vacuum tubes, its reliability and life directly affect the tube's reliability and life. In this paper, electronic gun in short tubes are accelerated to evaluate the lifetime of one kind of microwave vacuum (such as traveling wave tube). In order to shorten the testing process, accelerated life-test tests method is established at higher operating temperature and currents of cathode. The highest acceleration is obtained at the cathode temperature of 1100 °Cb with the cathode current density of 1.5A/cm2. One of the most common equations used to analyze cathode lifetimes is the Arrhenius model. Lifetimes were estimated based the time at which 50% cumulative failures had occurred using OriginPro75 software.

Along with the infrared technical rapid development, infrared radiation (IR) infrared thermal image technology is widely applied in nondestructive testing. In this paper, the theoretical basis of IR thermal imaging technology and the method of emissivity and background correction are introduced. The thermal characteristic of one kind of packaging device was studied using infrared thermal imaging technology and FEM (Finite element method), for example, microwave module. Through the comparison and analysis of the infrared images of one type of microwave module at working condition, the junction temperature of electronic chips is obtained.

Influence of weld technology on reliability and life to cathode in vacuum tube was studied by requests of microwave tube. The quality of weld technology quite affects the emission capability of cathode. The mode of weld quite affects thermal efficiency of cathode in vacuum tube. Thermal efficiency of cathode can increase availably by changing the weld technology on the heat transfer path. Thermal analysis and optimization design of cathode module for vacuum tube was carried out with the FEM in order to obtain required cathode temperature with lower heat power. The weld mode quite affects the cathode heat power, for example, the cathode heat power can reduce 28.6% with 8+8 laser spot welding. We have also built the life test system of TWT's electron gun structure of 8+8 laser spot welding, using this life test system, up to 30 years life of cathode is forecasted.

Thermal performance and reliability are necessary to be considered for high-power electronic packaging device design. In this paper, the thermal characteristic of one kind of packaging device was studied using FEM and experimentation, for phase shifter. The model for thermal condition of phase shifter was established. The top junction temperature obtained by simution shows tendency as tested experiment results, verifying the correctness of the simulation model and method used. The relation of relative temperature rising to different substrate thickness and material is studied by simution. The quantitative relations of these parameters and temperature provided a theoretical basis for the thermal design and reliability estimate, the optimization of parameters and the reliability improvement of phase shifter.

This study was designed to investigate whether EGB protect against ethanol-induced oxidative injury in rat testes.The rats were pretreated with EGB (4.8,9.6 mg/100g bw per day) before 30% ethanol administration (2.37g/kg bw by gastric incubation). Ninety days later, the rats were killed in order to measure the activities of Superoxide Dismutase (SOD), Glutathione Reductase (GST), Glutathione Peroxidase (GSH-Px), Catalase (CAT) and the contents of Glutathione (GSH), Reactive Oxygen Species (ROS), Malondialdehyde (MDA)in rat testes. The microsomal fractions were obtained by the method of differential centrifugation, and HO-1 (Heme Oxygenase-1) activity in testicular microsomal was measured. To examine the expression of HO-1 mRNA by reverse transcription polymerase chain reaction (RT-PCR).Compared with ethanol group, ROS, and MDA contents significantly decreased in EGB group testes, respectively (P0.05). GST, GSH-Px, CAT, SOD activities, and GSH content in EGB group testes elevated markedly (P0.05). EGB could enhance HO-1 mRNA expression remarkably.It is suggested that EGB as a preventive antioxidant can protect against ethanol-induced testicular injury, which may be associated with HO-1 activity enhancement, free radicals elimination, lipid per-oxidation reaction inhibition.

To investigate the protective effects of flavonoids of ginkgo biloba on anti-oxidizing system damaged by acute alcohol administration.Adult male Kunming mice were employed and divided into randomly flavonoid intervention group, normal control and ethanol control group according to body weight. After pretreated with flavonoids of ginkgo biloba (96mg/kg bw), the mice in flavonoid intervention group ingested alcohol (ethanol 4.8g/kg bw) via i.g. and were decapitated after 0.5, 1, 2, 4, 6, 9, and 15 h of ethanol administration. The same treatment was carried out on ethanol control group except that physiological saline was applied instead of flavonoid of ginkgo biloba. Meanwhile, the normal control group was established.The concentration of glutathione (GSH), malondialdehyde (MDA) and the activity of glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) in the serum and liver were determined. The experiment displays that the content of GSH and the activities of GSH-Px and SOD decreased rapidly after 1 h of treatment with alcohol and dropped to the lowest level at 4h of treatment. After 6h of treatment, these indexes came to the normal level rapidly. The level of MDA of serum and liver increased rapidly after 1 h of treatment and reached the climax at 4h and 6h respectively.
