Abstract The rate of sea level rise (SLR) along the Southeast Coast of the U.S. increased significantly after 2010. While anthropogenic radiative forcing causes an acceleration of global mean SLR, regional changes in the rate of SLR are strongly influenced by internal variability. Here we use observations and climate models to show that the rapid increase in the rate of SLR along the U.S. Southeast Coast after 2010 is due in part to multidecadal buoyancy-driven Atlantic meridional overturning circulation (AMOC) variations, along with heat transport convergence from wind-driven ocean circulation changes. We show that an initialized decadal prediction system can provide skillful regional SLR predictions induced by AMOC variations 5 years in advance, while wind-driven sea level variations are predictable 2 years in advance. Our results suggest that the rate of coastal SLR and its associated flooding risk along the U.S. southeastern seaboard are potentially predictable on multiyear timescales.

Abstract Arsenic (As) is a cancerogenic metalloid ubiquitously distributed in the environment, which can be easily accumulated in food crops like rice. Jasmonic acid (JA) and its derivatives play critical roles in plant growth and stress response. However, the role of endogenous JA in As accumulation and detoxification is still poorly understood. In this study, we found that JA biosynthesis enzymes Allene Oxide Synthases, OsAOS1 and OsAOS2, regulate As accumulation and As tolerance in rice. Evolutionary bioinformatic analysis indicated that AOS1 and AOS2 have evolved from streptophyte algae (e.g. the basal lineage Klebsormidium flaccidum) – sister clade of land plants. Compared to other two AOSs, OsAOS1 and OsAOS2 were highly expressed in all examined rice tissues and their transcripts were highly induced by As in root and shoot. Loss-of-function of OsAOS1 (osaos1–1) showed elevated As concentration in grains, which was likely attributed to the increased As translocation from root to shoot when the plants were subjected to arsenate [As(V)] but not arsenite [As (III)]. However, the mutation of OsAOS2 (osaos2–1) showed no such effect. Moreover, osaos1–1 and osaos2–1 increased the sensitivity of rice plants to both As(V) and As(III). Disrupted expression of genes involved in As accumulation and detoxification, such as OsPT4, OsNIP3;2, and OsOASTL-A1, was observed in both osaos1–1 and osaos2–1 mutant lines. In addition, a As(V)-induced significant decrease in Reactive Oxygen Species (ROS) production was observed in the root of osaos1–1 but not in osaos2–1. Taken together, our results indicate OsAOS1 modulates both As allocation and detoxification, which could be partially attributed to the altered gene expression profiling and ROS homeostasis in rice while OsAOS2 is important for As tolerance.

Abstract Long-term sea-level rise and multiyear to decadal sea level variations pose substantial risks for flooding and erosion in coastal communities. Here we use observations and climate model predictions to show that sea level variations along the U.S. East Coast are skillfully predictable 3 to 10 years in advance. The most predictable component of sea level is a basin scale upward trend, predictable a decade in advance and primarily a response to increasing greenhouse gases. Significant additional predictability comes from multidecadal variations of the Atlantic Meridional Overturning Circulation (AMOC). While perfect model simulations show AMOC-related sea level predictability of 5-7 years, model biases and initialization uncertainties reduce the realized predictive skill to 3-5 years, depending on location. Overall, greenhouse gas warming and predictable AMOC variations lead to multiyear to decadal prediction skill for sea level along the U.S. East Coast. Such skill could have significant societal benefit for planning and adaptation.

Salt stress poses a significant threat to global crop yield, prompting the need for understanding stress responses in crops like barley (Hordeum vulgare L.). This study investigated four barley genotypes - Giza129, Giza135, Line 1 and Line 2 - under 12 and 16 dS/m salt concentrations over a 30-day period. Results revealed a salinity-induced decrease in plant growth, particularly pronounced in Giza 129 and Giza 135. Ion analyses demonstrated distinct responses, with Giza genotypes accumulating more Na+ and less K+, leading to an increased Na+/K+ ratio, while both lines maintained K+, reducing the Na+/K+ ratio. Proline accumulation emerged as a crucial protective mechanism, evident in Line 1 and Line 2 exhibiting significantly higher proline content under salt stress compared to both Giza genotypes. Antioxidant enzyme activities, including catalase, ascorbate peroxidase and peroxidase, are markedly elevated in both lines, indicating superior salt tolerance mechanisms. This research contributes insights into barley salt tolerance, emphasizing the potential for genetic enhancement programs. Conclusively, the current study elucidated variations in salinity tolerance among new barley lines and their signaling mechanisms under salt stress. Given the global impact of salinity on crop yield, these findings hold promise for enhancing salt tolerance in barley and other crops through genetic advancements.

Warming of the subsurface Southern Ocean played only a minor role in shrinking Antarctic Sea ice extent in 2016, but was critical for the persistence of negative anomalies between 2016 and 2021, according to coupled climate model simulations partially constrained by observations.

In recent years, China has established many large water conservancy projects in the western mountainous areas. However, dam foundation excavation produces a huge amount of waste rock mixture. A soil-rock mixture with unique structural characteristics can easily cause geological disasters, such as collapse, landslide, and debris flow following an earthquake, rainfall, and engineering disturbance. Therefore, research on the strength and shear characteristics of the soil-rock mixture is beneficial to avoiding soil-rock mixture disasters. In this study, the soil-rock mixture in the slag dump site of Jinping Hydropower station is taken as the research object. The influence of different fillers on the gravel soil shear characteristics is investigated through a large-scale triaxial test of coarse-grained soil. The slope stability of the slag dump site is analyzed using Midas GTS. Then, the most unfavorable sliding surface of the slag dump site is discovered and the slope stability coefficient under the most unfavorable conditions is calculated. The results lay a scientific foundation for the design and construction of a slag dump slope.

Abstract Background HAK/KUP/KT (High-affinity K+ transporters/K+ uptake permeases/K+ transporters) is the largest potassium transporter family in plants, and plays pivotal roles in K+ uptake and transport, as well as biotic and abiotic stress responses. However, our understanding of the gene family in barley (Hordeum vulgare L.) is quite limited. Results In the present study, we identified 27 barley HAK/KUP/KT genes (hereafter called HvHAKs) through a genome-wide analysis. These HvHAKs were unevenly distributed on seven chromosomes, and could be phylogenetically classified into four clusters. All HvHAK protein sequences possessed the conserved motifs and domains. However, the substantial difference existed among HAK members in cis-acting elements and tissue expression patterns. Wheat had the most orthologous genes to barley HAKs, followed by Brachypodium distachyon, rice and maize. In addition, six barley HAK genes were selected to investigate their expression profiling in response to three abiotic stresses by qRT-PCR, and their expression levels were all up-regulated under salt, hyperosmotic and potassium deficiency treatments. Conclusion Twenty seven HAK genes (HvHAKs) were identified in barley, and they differ in tissue expression patterns and responses to salt stress, drought stress and potassium deficiency.

To date, one of the challenges in Human-Computer Interaction (HCI) is fully immersive multisensory remote physical interaction technologies. The applications of haptic perception in HCI can enrich the interaction details and effectively improve the immersion and realism of interaction. In the human-in-the-loop haptic interaction system, the quality of experience (QoE) of the human operator plays an essential role. However, QoE in haptic interaction is still in its infancy. Based on the typical application scenarios of haptic operation, the paper constructs a haptic-visual interaction framework and analyzes the QoE influencing factors. Through subjective evaluation experiments, the paper establishes a haptic interaction database that can provide a research basis for further exploring the relationship between various influencing factors and interactive QoE.

Arsenic (As) contamination in agricultural soils has become a great threat to the sustainable development of agriculture and food safety. Although a lot of approaches have been proposed for dealing with soil As contamination, they are not practical in crop production due to high cost, time-taking, or operational complexity. The rapid development of nanotechnology appears to provide a novel solution to soil As contamination. This study investigated the roles of calcium oxide nanoparticles (CaO NPs) in alleviating As toxicity in two barley genotypes (LJZ and Pu-9) differing in As tolerance. The exposure of barley seedlings to As stress showed a significant reduction in plant growth, calcium and chlorophyll content (SPAD value), fluorescence efficiency (Fv/m), and a dramatic increase in the contents of reactive oxygen species (ROS), malondialdehyde (MDA) and As, with LJZ being more affected than Pu-9. The exogenous supply of CaO NPs notably alleviated the toxic effect caused by As in the two barley genotypes. Moreover, the expression of As transporter genes, that is, HvPHT1;1, HvPHT1;3, HvPHT1;4 and HvPHT1;6, was dramatically enhanced when barley seedlings were exposed to As stress and significantly reduced in the treatment of CaO NPs addition. It may be concluded that the roles of CaO NPs in alleviating As toxicity could be attributed to its enhancement of Ca uptake, ROS scavenging ability, and reduction of As uptake and transportation from roots to shoots.

In this study, with La0.8Sr0.2Cr0.5Fe0.5O3-δ (LSCrF) and Ce0.8Sm0.2O1.9 (SDC) taken as the electronic conducting phase and the oxide ion conductor, respectively, dual-phase composite ceramic membranes with or without 3 % (in mole) Y2O3 stable ZrO2 (3YSZ) support were prepared and tested as oxygen separation membranes. Then, the oxygen transmission performance was tested by the electrochemical method based on the limiting current. The results obtained with this method coincide with those acquired by means of gas chromatography. Furthermore, the dependences of oxygen permeation rate on oxygen partial pressure, temperature and ceramic membrane thickness were analyzed. When air was used as the sweeping gas on the porous 3YSZ supported SDC-LSCrF dual-phase membrane, the oxygen permeation rate can reach 0.73, 0.82, 0.90 and 0.97 mL cm−2 min−1 at 650, 700, 750 and 800 °C, respectively. Flowering is the key process for the sexual reproduction in seed plants. In gramineous crops, the process of flowering, which includes the actions of both glume opening and glume closing, is directly driven by the swelling and withering of lodicules due to the water flow into and out of lodicule cells. All these processes are considered to be controlled by aquaporins, which are the essential transmembrane proteins that facilitate the transport of water and other small molecules across the biological membranes. In the present study, the evolution of aquaporins and their contribution to flowering process in plants were investigated via an integration of genome-wide analysis and gene expression profiling. Across the barley genome, we found that HvTIP1;1, HvTIP1;2, HvTIP2;3, and HvPIP2;1 were the predominant aquaporin genes in lodicules and significantly upregulated in responding to glume opening and closing, suggesting the importance of them in the flowering process of barley. Likewise, the putative homologs of the above four aquaporin genes were also abundantly expressed in lodicules of the other monocots like rice and maize and in petals of eudicots like cotton, tobacco, and tomato. Furthermore, all of them were mostly upregulated in responding to the process of floret opening, indicating a conserved function of these aquaporin proteins in plant flowering. The phylogenetic analysis based on the OneKP database revealed that the homologs of TIP1;1, TIP1;2, TIP2;3, and PIP2;1 were highly conserved during the evolution, especially in the angiosperm species, in line with their conserved function in controlling the flowering process. Taken together, it could be concluded that the highly evolutionary conservation of TIP1;1, TIP1;2, TIP2;3 and PIP2;1 plays important roles in the flowering process for both monocots and eudicots.

The Model-Analogs technique is used in the present study to assess the decadal sea surface temperature (SST) prediction skill over the Southern Ocean (SO). The Model-Analogs here is based on reanalysis products and model control simulations that have ∼1° ocean/ice (refined to 0.5° at high latitudes) components and 100 km atmosphere/land components. It is found that the model analog hindcasts show comparable skills with the initialized retrospective decadal hindcasts south of 50°S, with even higher skills over the Weddell Sea at longer lead years. The high SST skills primarily arise from the successful capture of SO deep convection states. This deep ocean memory and the associated decadal predictability are also clearly seen when we assess the Model-Analogs technique in a perfect model context. Within 30°S–50°S latitudinal band, the model analog hindcasts show low skills. When we include the externally forced signals estimated from the large ensemble simulations, the model analog hindcasts and initialized decadal hindcasts show identical skills. The Model-Analogs method therefore provides a great baseline for developing future decadal forecast systems. It is unclear whether such analog techniques would also be successful with models that explicitly resolve ocean mesoscale eddies or other small-scale processes. This area of research needs to be explored further.

Abstract Using GFDL's new coupled model SPEAR, we have developed a decadal coupled reanalysis/initialization system (DCIS) that does not use subsurface ocean observations. In DCIS, the winds and temperature in the atmosphere, along with sea surface temperature (SST), are restored to observations. Under this approach the ocean component of the coupled model experiences a sequence of surface heat and momentum fluxes that are similar to observations. DCIS offers two initialization approaches, called A1 and A2, which differ only in the atmospheric forcing from observations. In A1, the atmospheric winds/temperature are restored toward the JRA reanalysis; in A2, surface pressure observations are assimilated in the model. Two sets of coupled reanalyses have been completed during 1961–2019 using A1 and A2, and they show very similar multi‐decadal variations of the Atlantic Meridional Overturning Circulation (AMOC). Two sets of retrospective decadal forecasts were then conducted using initial conditions from the A1 and A2 reanalyses. In comparison with previous prediction system CM2.1, SPEAR‐A1/A2 shows comparable skill of predicting the North Atlantic subpolar gyre SST, which is highly correlated with initial values of AMOC at all lead years. SPEAR‐A1 significantly outperforms CM2.1 in predicting multi‐decadal SST trends in the Southern Ocean (SO). Both A1 and A2 have skillful prediction of Sahel precipitation and the associated ITCZ shift. The prediction skill of SST is generally lower in A2 than A1 especially over SO presumably due to the sparse surface pressure observations.

The recent advances in plant biology have significantly improved our understanding of reactive oxygen species (ROS) as signaling molecules in the redox regulation of complex cellular processes. In plants, free radicals and non-radicals are prevalent intra- and inter-cellular ROS, catalyzing complex metabolic processes such as photosynthesis. Photosynthesis homeostasis is maintained by thiol-based systems and antioxidative enzymes, which belong to some of the evolutionarily conserved protein families. The molecular and biological functions of redox regulation in photosynthesis are usually to balance the electron transport chain, photosystem II, photosystem I, mesophyll and bundle sheath signaling, and photo-protection regulating plant growth and productivity. Here, we review the recent progress of ROS signaling in photosynthesis. We present a comprehensive comparative bioinformatic analysis of redox regulation in evolutionary distinct photosynthetic cells. Gene expression, phylogenies, sequence alignments, and 3D protein structures in representative algal and plant species revealed conserved key features including functional domains catalyzing oxidation and reduction reactions. We then discuss the antioxidant-related ROS signaling and important pathways for achieving homeostasis of photosynthesis. Finally, we highlight the importance of plant responses to stress cues and genetic manipulation of disturbed redox status for balanced and enhanced photosynthetic efficiency and plant productivity.

β-amylase activity is related to the polymorphisms of Bmy1 intron III; however, no attention has been given to such relationships under environmental stresses such as drought. In this study, 73 cultivated barley genotypes and 52 Tibetan wild barley accessions were used to test the association between Bmy1 gene intron III polymorphisms and β-amylase activity under drought stress. Our results showed that three alleles, Bmy1.a, Bmy1.b, and Bmy1.c, existed in the examined barley genotypes. Tibetan wild barley had a higher proportion of Bmy1.b, whereas cultivated barley showed a higher proportion of Bmy1.a. Impressively, barley genotypes with Bmy1.b showed a significant increase in β-amylase activity under drought stress, compared with those with Bmy1.a or Bmy1.c, indicating that the Bmy1.b allele might provide more chances for developing barley cultivars with higher β-amylase activity under water stress than both the Bmy1.a and Bmy1.c alleles. Furthermore, the Tibetan wild barley XZ147, belonging to the Bmy1.b allele type, showed significantly higher β-amylase activity than Triumph under drought stress. This might be the result of the unique amino acid substitution M527 or the amino acid composition of R115, D165, A233, S347, and M527 of XZ147.

An increase in environmental pollution resulting from toxic heavy metals and metalloids [e.g., cadmium (Cd), arsenic (As), and lead (Pb)] causes serious health risks to humans and animals. Mitigation strategies need to be developed to reduce the accumulation of the toxic elements in plant-derived foods. Natural and genetically-engineered plants with hyper-tolerant and hyper-accumulating capacity of toxic minerals are valuable for phytoremediation. However, the molecular mechanisms of detoxification and accumulation in plants have only been demonstrated in very few plant species such as Arabidopsis and rice. Here, we review the physiological and molecular aspects of jasmonic acid and the jasmonate derivatives (JAs) in response to toxic heavy metals and metalloids. Jasmonates have been identified in, limiting the accumulation and enhancing the tolerance to the toxic elements, by coordinating the ion transport system, the activity of antioxidant enzymes, and the chelating capacity in plants. We also propose the potential involvement of Ca2+ signaling in the stress-induced production of jasmonates. Comparative transcriptomics analyses using the public datasets reveal the key gene families involved in the JA-responsive routes. Furthermore, we show that JAs may function as a fundamental phytohormone that protects plants from heavy metals and metalloids as demonstrated by the evolutionary conservation and diversity of these gene families in a large number of species of the major green plant lineages. Using ATP-Binding Cassette G (ABCG) transporter subfamily of six representative green plant species, we propose that JA transporters in Subgroup 4 of ABCGs may also have roles in heavy metal detoxification. Our paper may provide guidance toward the selection and development of suitable plant and crop species that are tolerant to toxic heavy metals and metalloids.

BackgroundAfter the elimination of leprosy in 1995, there were 10-30 newly detected leprosy cases every year in Zhejiang Province, and the epidemiological characteristics of the newly detected leprosy cases have changed. While most of the newly detected cases came from other provinces in China, not Zhejiang, it brought a new challenge for leprosy prevention and control in post- elimination era in Zhejiang, China. This study was aimed to understand the temporal-spatial distribution characteristics of newly detected leprosy cases, and provide the scientific rationales for the development of leprosy control strategy.MethodsData on the demographic of Zhejiang Province from 2011 to 2019 were obtained from the China Information System for Disease Control and Prevention, and the epidemiological data on leprosy cases newly detected in Zhejiang Province from 2011 to 2019 were obtained from the LEPROSY MANAGEMANT INFORMATION SYSTEM IN CHINA (LEPMIS), and temporal-spatial distributions were described. The geographic information system software-ArcGIS 10.4 was used to draw the statistical maps, and Geoda 1.14.0 was used for local spatial autocorrelation analysis (local Getis coefficient method). Ridley-Jopling classification was used to classify the clinical types into I, TT, BT, BB, BL or LL. Two-group classification system developed by the World Health Organization (WHO) was used and cases were classified into multibacillary (MB) type or paucibacillary (PB) type.ResultsA total of 167 leprosy cases were reported in Zhejiang Province during 2011-2019, including 107 cases in males and 60 in females. The mean age at diagnosis was 37.99±14.81 years, and 94.01% of the cases were detected through the examination at skin-clinics. The number of workers, MB cases, G2D cases were 81 (48.50%), 159 (94.01%), 24 (14.37%) respectively, and the rate of early detection increased from 45.16% in 2011 to 90.91% in 2019. Leprosy cases were reported in all the prefectures of Zhejiang except Zhoushan City. The cases in local population accounted for 23.35% (39 cases), and the cases in floating population (especially coming from high epidemic provinces in China) accounted for 76.65% (128 cases). The annual number of newly detected cases showed a decreasing trend, from 31 cases in 2011 to 11 in 2019. Time of the floating population living in Zhejiang Province ranged from several months to more than 10 years. The annual proportion of new cases with G2D declined from 22.58% in 2011 to 9.09% in 2019. The results of local indicators of autocorrelation (LISA) analysis showed that the high-high areas were mainly concentrated in the middle and northeast of Zhejiang Province, while the low-low areas were in the east and southwest.ConclusionA few scattered cases still can be seen in post-elimination era, and there was a spatial clustering of the newly detected leprosy cases in Zhejiang Province. Most of the cases in Zhejiang Province were from other high epidemic provinces in China, which brought a new challenge for leprosy control and prevention in post- elimination era in Zhejiang, and it is also necessary to strengthen the early detection and standard management of the leprosy cases in floating population in Zhejiang.

Drought is a devastating environmental constraint affecting the agronomic production of barley. To facilitate the breeding process, abundant germplasm resources and reliable evaluation systems to identify the true drought-tolerant barley genotypes are needed. In this study, 237 cultivated and 190 wild barley genotypes, originating from 28 countries, were screened for drought tolerance under the conditions of both water deficit and polyethylene glycol (PEG)-simulated drought at seedling stage. Drought stress significantly reduced the plant growth of all barley genotypes, but no significant difference in drought-induced reduction in the performance of barley seedlings was observed under these two drought conditions. Both cultivated and wild barley subspecies displayed considerable genotypic variability in drought tolerance, which underpinned the identification of 18 genotypes contrasting in drought tolerance. A comparative analysis of drought effects on biomass, water relation, photosynthesis, and osmotic adjustment was undertaken using these contrasting barley genotypes, in order to verify the reliability of the screening and to obtain the credible traits as screening criteria of drought tolerance in barley. As expected, the selected drought-tolerant genotypes showed much less reduction in shoot biomass than drought-sensitive ones under water deficit, which was significantly positively correlated with the results of large-scale screening, confirming the reliability of the screening for drought tolerance under two drought conditions in this study. Likewise, the traits of water relation, photosynthetic activity, and osmotic adjustment differed greatly between the contrasting genotypes under water deficit stress, and they were highly correlated to the growth of barley seedlings, suggesting the potential of them to be the selection criteria for drought tolerance. The analysis of the variable importance of these traits in drought tolerance indicated that sap osmolality and relative water content in the youngest fully-expanded leaf are the suitable selection criteria of screening for drought tolerance in barley at seedling stage.

Abstract We document the development and simulation characteristics of the next generation modeling system for seasonal to decadal prediction and projection at the Geophysical Fluid Dynamics Laboratory (GFDL). SPEAR (Seamless System for Prediction and EArth System Research) is built from component models recently developed at GFDL—the AM4 atmosphere model, MOM6 ocean code, LM4 land model, and SIS2 sea ice model. The SPEAR models are specifically designed with attributes needed for a prediction model for seasonal to decadal time scales, including the ability to run large ensembles of simulations with available computational resources. For computational speed SPEAR uses a coarse ocean resolution of approximately 1.0° (with tropical refinement). SPEAR can use differing atmospheric horizontal resolutions ranging from 1° to 0.25°. The higher atmospheric resolution facilitates improved simulation of regional climate and extremes. SPEAR is built from the same components as the GFDL CM4 and ESM4 models but with design choices geared toward seasonal to multidecadal physical climate prediction and projection. We document simulation characteristics for the time mean climate, aspects of internal variability, and the response to both idealized and realistic radiative forcing change. We describe in greater detail one focus of the model development process that was motivated by the importance of the Southern Ocean to the global climate system. We present sensitivity tests that document the influence of the Antarctic surface heat budget on Southern Ocean ventilation and deep global ocean circulation. These findings were also useful in the development processes for the GFDL CM4 and ESM4 models.

In barley (Hordeum vulgare L.) breeding, heading date is one of the most important agricultural traits that is essential for the completion of its life cycle. Certain endogenous and environmental factors regulate floral transition, morphing the complex genetic mechanism of the heading phase, which could serve as a premise of orchestration for improved yields. To elaborate the network of genetic and environmental signals, a hydroponic experiment was carried out using two spring (i.e., DM65 and DM70) and two winter barley genotypes (i.e., DM269 and DM385). Our results confirmed that the vernalized environment produced a substantial reduction in plant height, biomass and photosynthetic activity compared with the control plants. A noticeable increase in oxidative stress was exhibited by DM65 and DM70 plants compared with their respective controls at 20 °C, while no significant difference was observed for any genotype grown in the greenhouse (25 °C). Simultaneously, increased antioxidant enzyme activity in winter barley genotypes showed a defensive mechanism under vernalized conditions (4 °C). Furthermore, the expression of key regulatory flowering genes revealed that the vernalization gene (HvVRN1) is the key regulator of floral induction after cold exposure, whereas Photoperiod Response Locus 1 (HvPpd-H1) had significantly higher expression under greenhouse conditions, along with Phytochrome C (HvPHY-C), validating their involvement as upstream heading time regulators. These findings contribute to enriching the study of environmental signals that substantially modulate the complex mechanism of barley heading date.

Rice (Oryza sativa) is a staple food that feeds more than half the world population. As rice is highly sensitive to soil salinity, current trends in soil salinization threaten global food security. To better understand the mechanistic basis of salinity tolerance in rice, three contrasting rice cultivars—Reiziq (tolerant), Doongara (moderately tolerant), and Koshihikari (sensitive)—were examined and the differences in operation of key ion transporters mediating ionic homeostasis in these genotypes were evaluated. Tolerant varieties had reduced Na+ translocation from roots to shoots. Electrophysiological and quantitative reverse transcription PCR experiments showed that tolerant genotypes possessed 2-fold higher net Na+ efflux capacity in the root elongation zone. Interestingly, this efflux was only partially mediated by the plasma membrane Na+/H+ antiporter (OsSOS1), suggesting involvement of some other exclusion mechanisms. No significant difference in Na+ exclusion from the mature root zones was found between cultivars, and the transcriptional changes in the salt overly sensitive signaling pathway genes in the elongation zone were not correlated with the genetic variability in salinity tolerance amongst genotypes. The most important hallmark of differential salinity tolerance was in the ability of the plant to retain K+ in both root zones. This trait was conferred by at least three complementary mechanisms: (1) its superior ability to activate H+-ATPase pump operation, both at transcriptional and functional levels; (2) reduced sensitivity of K+ efflux channels to reactive oxygen species; and (3) smaller upregulation in OsGORK and higher upregulation of OsAKT1 in tolerant cultivars in response to salt stress. These traits should be targeted in breeding programs aimed to improve salinity tolerance in commercial rice cultivars.

Adaptation to unfavorable abiotic stresses is one of the key processes in the evolution of plants. Calcium (Ca2+) signaling is characterized by the spatiotemporal pattern of Ca2+ distribution and the activities of multi-domain proteins in integrating environmental stimuli and cellular responses, which are crucial early events in abiotic stress responses in plants. However, a comprehensive summary and explanation for evolutionary and functional synergies in Ca2+ signaling remains elusive in green plants. We review mechanisms of Ca2+ membrane transporters and intracellular Ca2+ sensors with evolutionary imprinting and structural clues. These may provide molecular and bioinformatics insights for the functional analysis of some non-model species in the evolutionarily important green plant lineages. We summarize the chronological order, spatial location, and characteristics of Ca2+ functional proteins. Furthermore, we highlight the integral functions of calcium-signaling components in various nodes of the Ca2+ signaling pathway through conserved or variant evolutionary processes. These ultimately bridge the Ca2+ cascade reactions into regulatory networks, particularly in the hormonal signaling pathways. In summary, this review provides new perspectives towards a better understanding of the evolution, interaction and integration of Ca2+ signaling components in green plants, which is likely to benefit future research in agriculture, evolutionary biology, ecology and the environment.

Harmonia axyridis is an important natural predator used in the biological control of insect pests. Vitellogenin (Vg) supplementation to artificial diet can improve fecundity of H. axyridis, however, the effects of Vg on physiology of H. axyridis at the molecular level is unclear. This study investigated the effects of Vg on the physiology (digestive enzyme activities) and transcriptome patterns by feeding H. axyridis adults with treatment (artificial diet with Vg supplement) and control (artificial diet supplemented with bovine serum albumin (BSA). The transcriptome sequencing yielded 43.94 Gb of clean data, and 3,946 differentially expressed genes (DEGs) – including 93 upregulated and 3,853 downregulated genes between the treatment and control. Six DEGs related to development and digestive enzyme were chosen for quantitative real-time PCR (qRT-PCR) to validate the accuracy of the RNA-seq results and confirmed that the transcriptome analysis yielded reliable results. The Vg supplement has increased activities of digestive enzymes and related genes expression in H. axyridis. The transcript level of digestive enzyme genes (apolipoprotein D and phosphoenolpyruvate carboxykinase) were much higher in adults fed on diet supplemented with Vg compared with that of the control.

Cadmium (Cd) contamination in soils poses great risks to both agricultural production and human health. Calcium (Ca) is an essential element playing a significant role in protecting plants against Cd toxicity. However, how Ca affects Cd uptake and translocation in rice is still not fully elucidated. In this study, the regulatory role of Ca in Cd uptake and upward translocation was investigated in rice at different growth stages. Our results showed that the supplement of 5 mM Ca significantly reduced Cd uptake by rice roots, because of their competition for Ca-permeable channels as an absorption site and Ca-induced downregulation of OsNRAMP1 and OsNRAMP5. However, Ca application facilitated the upward translocation of Cd by both upregulating OsHMA2 to induce xylem loading of Cd and downregulating OsHMA3 to reduce vacuolar sequestration of Cd. Such contrary results suggested a double-edged role of Ca in regulating root Cd uptake and root-to-shoot Cd translocation in rice. Although it increased Cd content in the aboveground vegetative tissues during the whole growth period, the addition of 5 mM Ca eventually decreased Cd content in rice grains at the ripening stage. All these results suggest that Ca-based amendments possess great potential for the production of low-Cd rice grains.

Chromium (Cr) is recognized as a toxic metal that has detrimental effects on living organisms; notably, it is discharged into soil by various industries as a result of anthropogenic activities. Microbe-assisted phytoremediation is one of the most emergent and environmentally friendly methods used for the detoxification of pollutants. In this study, the alleviative role of Staphylococcus aureus strain K1 was evaluated in wheat (Triticum aestivum L.) under Cr stress. For this, various Cr concentrations (0, 25, 50 and 100 mg·kg−1) with and without peat-moss-based bacterial inoculum were applied in the soil. Results depicted that Cr stress reduced the plants’ growth by causing oxidative stress in the absence of S. aureus K1 inoculation. However, the application of S. aureus K1 regulated the plants’ growth and antioxidant enzymatic activities by reducing oxidative stress and Cr toxicity through conversion of Cr6+ to Cr3+. The Cr6+ uptake by wheat was significantly reduced in the S. aureus K1 inoculated plants. It can be concluded that the application of S. aureus K1 could be an effective approach to alleviate the Cr toxicity in wheat and probably in other cereals grown under Cr stress.

Waterlogging and salinity are two major abiotic stresses that hamper crop production world-wide resulting in multibillion losses. Plant abiotic stress tolerance is conferred by many interrelated mechanisms. Amongst these, the cell’s ability to maintain membrane potential (MP) is considered to be amongst the most crucial traits, a positive relationship between the ability of plants to maintain highly negative MP and its tolerance to both salinity and waterlogging stress. However, no attempts have been made to identify quantitative trait loci (QTL) conferring this trait. In this study, the microelectrode MIFE technique was used to measure the plasma membrane potential of epidermal root cells of 150 double haploid (DH) lines of barley (Hordeum vulgare L.) from a cross between a Chinese landrace TX9425 and Japanese malting cultivar Naso Nijo under hypoxic conditions. A major QTL for the MP in the epidermal root cells in hypoxia-exposed plants was identified. This QTL was located on 2H, at a similar position to the QTL for waterlogging and salinity tolerance reported in previous studies. Further analysis confirmed that MP showed a significant contribution to both waterlogging and salinity tolerance. The fact that the QTL for MP was controlled by a single major QTL illustrates the power of the single-cell phenotyping approach and opens prospects for fine mapping this QTL and thus being more effective in marker assisted selection.

Grain weight and protein content will be reduced and increased, respectively, when barley is subjected to water stress after anthesis, consequently deteriorating the malt quality. However, such adverse impact of water stress differs greatly among barley genotypes. In this study, two Tibetan wild barley accessions and two cultivated varieties differing in water stress tolerance were used to investigate the genotypic difference in metabolic profiles during grain-filling stage under drought condition. Totally, 71 differently accumulated metabolites were identified, including organic acids, amino acids/amines, and sugars/sugar alcohols. Their relative contents were significantly affected by water stress for all genotypes and differed distinctly between the wild and cultivated barleys. The principal component analysis of metabolites indicated that the Tibetan wild barley XZ147 possessed a unique response to water stress. When subjected to water stress, the wild barley XZ147 showed the most increase of β-amylase activity among the four genotypes, as a result of its higher lysine content, less indole-3-acetic acid (IAA) biosynthesis, more stable H2O2 homeostasis, and more up-regulation of BMY1 gene. On the other hand, XZ147 had the most reduction of β-glucan content under water stress than the other genotypes, which could be explained by the faster grain filling process and the less expression of β-glucan synthase gene GSL7. All these results indicated a great potential for XZ147 in barley breeding for improving water stress tolerance.

The green lacewing, Chrysopa pallens Rambur, is one of the most important natural predators because of its extensive spectrum of prey and wide distribution. However, what we know about the nutritional and reproductive physiology of this species is very scarce.By cDNA amplification and Illumina short-read sequencing, we analyzed transcriptomes of C. pallens female adult under starved and fed conditions. In total, 71236 unigenes were obtained with an average length of 833 bp. Four vitellogenins, three insulin-like peptides and two insulin receptors were annotated. Comparison of gene expression profiles suggested that totally 1501 genes were differentially expressed between the two nutritional statuses. KEGG orthology classification showed that these differentially expression genes (DEGs) were mapped to 241 pathways. In turn, the top 4 are ribosome, protein processing in endoplasmic reticulum, biosynthesis of amino acids and carbon metabolism, indicating a distinct difference in nutritional and reproductive signaling between the two feeding conditions.Our study yielded large-scale molecular information relevant to C. pallens nutritional and reproductive signaling, which will contribute to mass rearing and commercial use of this predaceous insect species.

In this work, the effect of drought on K+ uptake in root and its translocation from root to shoot was investigated using six barley genotypes contrasting in drought tolerance. Results showed that drought conditions caused significant changes in K+ uptake and translocation in a time- and genotype-specific manner, which consequently resulted in a significant difference in tissue K+ contents and drought tolerance levels between the contrasting barley genotypes. The role of K+ transporters and channels and plasma membrane (PM) H+-ATPase in barley’s adaptive response to drought stress was further investigated at the transcript level. The expression of genes conferring K+ uptake (HvHAK1, HvHAK5, HvKUP1, HvKUP2 and HvAKT1) and xylem loading (HvSKOR) in roots were all affected by drought stress in a time- and genotype-specific manner, indicating that the regulation of these K+ transporters and channels is critical for root K+ uptake and root to shoot K+ translocation in barley under drought stress. Furthermore, the barley genotypes showed a strong correlation between H+ efflux and K+ influx under drought stress, which was further confirmed by the significant up-regulation of HvHA1 and HvHA2. These results suggested an important role of plasma membrane H+-ATPase activity and/or expression in regulating the activity of K+ transporters and channels under drought stress. Taken together, it may be concluded that the genotypic difference in drought stress tolerance in barley is conferred by the difference in the ability to regulate K+ transporters and channels in root epidermis and stele.

Waterlogging is a serious environmental problem that limits agricultural production in low-lying rainfed areas around the world. The major constraint that plants face in a waterlogging situation is the reduced oxygen availability. Accordingly, all previous efforts of plant breeders focused on traits providing adequate supply of oxygen to roots under waterlogging conditions, such as enhanced aerenchyma formation or reduced radial oxygen loss. However, reduced oxygen concentration in waterlogged soils also leads to oxygen deficiency in plant tissues, resulting in an excessive accumulation of reactive oxygen species (ROS) in plants. To the best of our knowledge, this trait has never been targeted in breeding programs and thus represents an untapped resource for improving plant performance in waterlogged soils. To identify the quantitative trait loci (QTL) for ROS tolerance in barley, 187 double haploid (DH) lines from a cross between TX9425 and Naso Nijo were screened for superoxide anion (O2•−) and hydrogen peroxide (H2O2)—two major ROS species accumulated under hypoxia stress. We show that quantifying ROS content after 48 h hypoxia could be a fast and reliable approach for the selection of waterlogging tolerant barley genotypes. The same QTL on chromosome 2H was identified for both O2•− (QSO.TxNn.2H) and H2O2 (QHP.TxNn.2H) contents. This QTL was located at the same position as the QTL for the overall waterlogging and salt tolerance reported in previous studies, explaining 23% and 24% of the phenotypic variation for O2•− and H2O2 contents, respectively. The analysis showed a causal association between ROS production and both waterlogging and salt stress tolerance. Waterlogging and salinity are two major abiotic factors affecting crop production around the globe and frequently occur together. The markers associated with this QTL could potentially be used in future breeding programs to improve waterlogging and salinity tolerance.

Halophytes species can be used as a highly convenient model system to reveal key ionic and molecular mechanisms that confer salinity tolerance in plants. Earlier, we reported that quinoa (Chenopodium quinoa Willd.), a facultative C3 halophyte species, can efficiently control the activity of slow (SV) and fast (FV) tonoplast channels to match specific growth conditions by ensuring that most of accumulated Na+ is safely locked in the vacuole (Bonales-Alatorre et al. (2013) Plant Physiology). This work extends these finding by comparing the properties of tonoplast FV and SV channels in two quinoa genotypes contrasting in their salinity tolerance. The work is complemented by studies of the kinetics of net ion fluxes across the plasma membrane of quinoa leaf mesophyll tissue. Our results suggest that multiple mechanisms contribute towards genotypic differences in salinity tolerance in quinoa. These include: (i) a higher rate of Na+ exclusion from leaf mesophyll; (ii) maintenance of low cytosolic Na+ levels; (iii) better K+ retention in the leaf mesophyll; (iv) a high rate of H+ pumping, which increases the ability of mesophyll cells to restore their membrane potential; and (v) the ability to reduce the activity of SV and FV channels under saline conditions. These mechanisms appear to be highly orchestrated, thus enabling the remarkable overall salinity tolerance of quinoa species.

Since the upper ocean takes up much of the heat added to the earth system by anthropogenic global warming, one would expect that global warming would lead to an increase in stratification and a decrease in the ventilation of the ocean interior. However, multiple simulations in global coupled climate models using an ideal age tracer which is set to zero in the mixed layer and ages at 1 yr/yr outside this layer show that the intermediate depths in the low latitudes, Northwest Atlantic, and parts of the Arctic Ocean become younger under global warming. This paper reconciles these apparently contradictory trends, showing that the decreases result from changes in the relative contributions of old deep waters and younger surface waters. Implications for the tropical oxygen minimum zones, which play a critical role in global biogeochemical cycling are considered in detail.

The gap gene hunchback (hb) is a key regulator in the anteroposterior patterning of insects. Loss-of-function of hb resulted in segmentation defects in the next generation. In this paper, hb expression level was investigated at different developmental stages of the pea aphid, Acyrthosiphon pisum (Ap). Aphb mRNA was most early detected at the first instar stage and showed an incontinuous increase in the whole life cycle. Ingested RNA interference was performed at the second instar stage to knockdown the Aphb expression. Continuous feeding of Aphb double-stranded RNA mixed in artificial diet led to reduction of Aphb transcripts and rise of insect lethality. These results indicated that hunchback was a good RNAi target in the management of insect pests.

Context • Gonorrhea, a highly communicable, sexually transmitted infection, remains a major public-health concern globally. In recent years, Zhejiang province, an eastern province, has had the highest incidence of gonorrhea in China. Objective • The study intended to identify the geographic distribution patterns and spaciotemporal clustering characteristics of the disease’s incidence in Zhejiang between 2016 and 2020, to understand the spatial epidemiology of gonorrhea and to pinpoint the locations with relatively high risks of gonorrhea, the hotspots, which could be the key areas for disease prevention and control. Design • The research team performed a retrospective, spaciotemporal-clustering analysis of data about newly reported gonorrhea cases from January 2016 to December 2020 in Zhejiang province, using the China Information System for Disease Control and Prevention. Setting • The study took place at the Zhejiang Provincial Institute of Dermatology in Huzhou, China. Outcome Measures • The research team: (1) used the Geographic Information System software-ArcGIS 10.8 software to draw statistical maps; (2) conducted a spatialpattern clustering analysis at the district or county level; (3) performed an autocorrelation analysis using Getis-Ord (Gi*) statistics to detect spatial patterns and the hotspots of gonorrhea incidence; and (4) used SaTScan9.7 to analyze the space-time clusters. Results • Zhejiang province reported 85 904 gonorrhea cases from 2016 to 2020, with a male to female ratio of 3.81:1. The average annual incidence rate of gonorrhea was 30.50 per 100 000 individuals in the population, ranging from 22.73 cases to 39.65 cases, and the annual incidence showed a significant downward trend over the five years (χ²=16.142, P<.001). The northern and central areas had a higher incidence than the southern area did. Autocorrelation analysis showed that the gonorrhea incidence had a significantly clustered distribution (Moran’s I from 0.197 to 0.295, Z score from 4.749 to 6.909, P<.001). The high-high cluster areas were mainly in the urban districts of Hangzhou and some counties and districts of Jiaxing. The Gi* statistics further indicated that the hotspots of gonorrhea were mainly in Hangzhou, Jiaxing, and Huzhou. The Kuldorff ’s scan identified two clusters, mainly composed of 36 counties or districts in northern Zhejiang, such as Hangzhou and Jiaxing, and central Zhejiang, such as Jinhua and Shaoxing. Conclusions • The gonorrhea incidence rates in northern and central Zhejiang from 2016 to 2020 were higher than those in southern Zhejiang. An area of relatively higher risk for gonorrhea existed mainly in the urban districts of Hangzhou and some counties and districts of Jiaxing, Jinhua, and Shaoxing. In the future, the research team plans to focus on strengthening the prevention and control measures against gonorrhea in those areas. [ABSTRACT FROM AUTHOR]
