Drought stress has an adverse effect on crop production and food quality. Milk thistle (Silybum marianum L.) is an oil and medicinal crop known as an alternative oil crop with high level of unsaturated fatty acids, which makes it a favourable edible oil for use in food production. Silymarin (a mixture of flavonolignans) is the main active medicinal component. Biochemical diversity, changes induced by water deficit stress in secondary metabolites, and their relationships with production traits in native germplasm are poorly understood in milk thistle. Twenty-six ecotypes mainly collected from different regions of Iran were evaluated for oil, fatty acid profile, triacylglycerol (TAG) composition, silymarin and agro-morphological traits under non-stress and water stress conditions for 2 years. Water stress increased oil and silymarin content while decreasing fruit yield and related traits. The most abundant fatty acid averaged over all ecotypes under both moisture conditions was linoleic acid (L, 39%), followed by oleic acid (O, 36%), palmitic acid (P, 9%) and stearic acid (E, 6%). Among the 24 detected TAGs, the five major compositions were OOL, OLL + OOLn (linolenic), POL, OOO, LLL + OLLn and EOL. Superior ecotypes rich in monounsaturated and polyunsaturated fatty acids were identified and can be introduced as candidates for food, medicinal and industrial purposes. Associations among different attributes are discussed.

Background: Better understanding of genetic structure of economic traits is crucial for identification and selection of superior genotypes in specific breeding programs. Best linear unbiased prediction (BLUP) is the most efficient method in this regards, which is poorly used in forage plant breeding. The present study aimed to assess genetic variation, estimate genetic parameters, and predict breeding values of five essential traits in full sib families (recognized by EST-SSR markers) of tall fescue using REML/BLUP procedure. Method: Forty-two full-sib families of tall fescue (included of 120 individual genotypes), recognized by EST-SSR markers’ along with twenty-one their corresponding parental genotypes were assessed for biomass production and agro-morphological traits at three harvests (spring, summer, and autumn) in the field during 4 years (2017-2020). Results: Considerable genotypic variability was observed for all traits. Low narrow-sense heritability (h2n) for dry forage yield (DFY) at three harvest indicates that non-additive gene actions may play an important role in the inheritance of this trait. Higher h2n of yield related traits and flowering time and also significant genetic correlation of these traits with forage yield, suggests that selection based on these traits via developing an index may lead to indirect genetic improvement of DFY. Conclusion: Our results showed the adequacy of REML/BLUP procedure for identification and selection of preferable parental genotypes and progenies with higher breeding values for future breeding programs such as variety development in tall fescue. Parental genotypes 21M, 1M, and 20L were identified as superior and stable genotypes and could also produce the best hybrid combinations when they were mostly used as maternal parent.

Background: The genetic basis and variability of persistence and summer dormancy and their association with forage production traits has been less investigated in tall fescue. Results: High genotypic variation was found for all the measured traits in both parental genotypes and half-sib families. Incomplete summer dormancy was found for most of the evaluated genotypes. Summer dormancy index were negatively correlated with forage production and yield related traits. Half-sib families had higher persistence than parental genotypes, which can be due to the heterosis expression in progenies. Moderate narrow-sense heritability for persistence indicates that both additive and dominant gene action may play a role in the expression of this trait. Yield-related traits and summer dormancy showed moderate to high estimates of narrow sense heritability (0.63-0.73) indicates additive gene action for these traits. This suggest phenotypic recurrent selection can be successful to achieve genetic progress. Conclusion: In conclusion, our results can enhance the knowledge of inheritance of summer dormancy and related traits which would be useful for future genetic studies. Based on the general combining ability and application of multivariate analysis, preferable genotypes for forage use (such as 21M and 1E) and turf application (such as 17M and 4E) were identified for future programs.

International audience; Half-sib (HS) mating is the most important breeding method of open-pollinated sexual forage grasses such as tall fescue. Progeny testing procedures of half-sib families can only measure general combining ability (GCA) due to the unknown pollen donor. In tall fescue, genetic studies using HS mating and application of molecular markers for paternity identification in polycross progenies are so far limited. In this study, 960 genotypes from sixteen half-sib families of tall fescue derived from polycrosses, along with their corresponding parental genotypes were evaluated for agro-morphological traits in the field during 2016 and 2017. Genetic diversity of the parental genotypes was assessed using twenty EST-SSR markers and then a minimal set of primers was used to identify the most likely male parent in the polycross progenies. Considerable genetic variation was observed among the parental genotypes based on both morphological and molecular assessments. Moderate to high estimates of narrow sense heritability (0.53-0.84) for morphological traits indicated that phenotypic selection can be successful to achieve genetic progress. The EST-SSR markers allowed to identify a unique paternal parent in 177 of the offspring which were divided into 66 full-sib families. The number of genotypes per full-sib family varied from 1 to 11. Marker data used to identify full-sib families, estimate specific combining ability (SCA), and select siblings based on the genetic value of both parents. No clear association was found between GCA of parents and the corresponding SCA for forage yield, indicating that both additive and dominant gene actions may play a role in the genetic control of this trait. The obtained results highlighted the importance of marker-based paternity identification to increase gain from selection in polycross breeding programs of tall fescue.

Research on water stress strategies (WSS) and crop response to water stress is important to improve irrigation water productivity (WPI) and crop production especially for regions where water is a limited resource. This study was conducted to assess (i) the response of silage maize to water stress at different growth stages for contrasting water stress strategies in the semi-arid environment under pot and field experiments irrigated by a drip-tape system, (ii) to determine the relationship between applied water and total biomass (TB) in different WSS in both experiments. The hybrid 704-single-cross of maize was assessed at pot experiment during 2009 and 2010 and field experiment was conducted in 2017 under two WSS including WS1 (same irrigation event with different applied water depth) and WS2 (same applied water depth in different irrigation event) at three growth stages. Each WSS consisted of four different irrigation levels, including severe, moderate, and mild drought stress and a full irrigation. In both pot and field experiments, severe drought stress under WS1 and WS2 strategies led to a depression in LA and TB at tassel (S2) and silage harvest (S3) stages. At each growth stage, WPI values decreased with increasing water-deficit levels in both WSS’s and experiments. We found a linear relationship between TB and applied water for WS1 and WS2 in both experiments. However, the yield performance per unit of water used was higher for WS1 than for WS2. The yield response factor to stress (Ky) values for LA and TB were higher under WS2 than WS1 in both experiments, indicating that WS1 can be employed as an effective deficit irrigation management under the drip-tape irrigation system for maize in an arid area with shortage of water.

Identification and selection of plant genotypes with survival, recovery and sustainable performance during drought periods is one of the main objectives of plant improvement for arid and semi-arid regions. This study aimed to evaluate root traits and recovery of tall fescue genotypes after a period of drought stress. A total of twenty four tall fescue genotypes were selected from a wide polycross population and assessed for field drought tolerance based on stress tolerance index (STI) and recovery during 2010–2013. The genotypes were classified as tolerant, moderately tolerant and susceptible based on STI. Then, in 2014, genotypes were assessed in a pot experiment for root characteristics under two levels of moisture environments (control and intense drought stress). In both moisture environments, root length (RL), root area (RA), root volume (RV) and root weight (RW) were negatively correlated with days to recovery (DR). Genotypes that recovered from drought had greater RL, RA, RV and RW than the genotypes unable to recover. Principle component analysis (PCA) was performed to identify genotypes with superior root characteristics, stress tolerance and recovery potential that can be used for future breeding programmes. Results indicated that selection based on combining STI, recovery and extensive root system might improve drought tolerance of tall fescue.

Analysis of genetic diversity in germplasm collections is an important component of crop improvement programs. This study was conducted to analyze genetic variation and to classify tall fescue genotypes based on phenotypic evaluation and EST-SSR molecular markers. Twenty-five genotypes were assessed based on phenotypic and 42 EST-SSR molecular markers according to a completely randomized block design with three replications during eight years (2007–2014). Results indicated that the effect of year, genotype and their interaction were significant for all of the measured traits. Both morphological and molecular assessments showed considerable genetic variation among genotypes. The estimates of broad-sense heritability (h 2 b ) were moderate to high (h 2 b = 42.1–78.4) for the traits studied. Based on EST-SRR analysis, a total number of 229 alleles were detected with an average of 4.58 alleles per marker. Average PIC value was 0.49 with a range of 0.014 for NFA140 to 0.95 for NFA047 . Phenotypic evaluations and EST-SSR molecular marker classified genotypes into 3 and 7 clusters, respectively which mainly supported geographical origins. The general correspondence was observed between morphological and molecular classification. Therefore, combining the molecular markers with morphological responses could be more beneficial to describe genetic variation and distinguish superior genotypes for future breeding programs.

Genetic analysis of root and physiological traits and selection of genotypes with higher drought tolerance through these traits is generally limited in tall fescue. In this study, some parental genotypes of tall fescue first were assessed for field drought tolerance in 2014–2015 and then the polycross seeds were harvested to provide half-sib families. Sixteen half-sib families along with their corresponding parental genotypes were assessed in a pot experiment for root and physiological characteristics under three irrigation levels (control, mild and intense) in 2016. The results showed that drought stress decreased dry forage yield (DFY), relative water content and total chlorophyll and increased carotenoid and proline in both parental genotypes and half-sib families. Intense drought stress decreased most of the root traits at 0-30 cm soil depth while at 30–60 cm depth length, area, volume and dry weight of roots were increased. A broad range of general combining ability (GCA) was observed for DFY (21M and 9E), root (21M, 12L and 20L) and physiological characteristics (12L and 9E) at three irrigation levels. Moderate to high estimates of narrow sense heritability (0.40–0.72) as well as genetic variation for root and physiological traits, indicated that phenotypic selection can be successful to attain genetic progress. Indirect selection to improve DFY was more effective through selection for root and some physiological traits. Significant associations of root and some physiological traits with drought tolerance demonstrated that these traits could be used as appropriate selection criteria to elevate forage yield and identify superior genotypes for arid and semi-arid regions.

Research on crop response to deficit irrigation is important to reduce agricultural water use in areas where water is a limited resource. The objective of this study was to evaluate changes in physiological and root traits under mild and intense drought stress in tall fescue. It also sought to find associations between these changes and field drought tolerance. A total of 24 tall fescue genotypes were selected from a wide polycross population and assessed for field drought tolerance during 2011–2012 in the field. The genotypes were classified as tolerant, moderately tolerant, and susceptible based on drought stress tolerance index (STI), tolerance index (TOL), and yield reduction (YR). In 2013, 24 genotypes were clonally propagated and planted in polyvinylchloride (PVC) tubes under, three levels of moisture regimes. Root characteristics were investigated at 0–30 and 30–60 cm depths of soil. Also 11 physiological traits, dry forage yield, STI, TOL, and YR were recorded. At the 30–60 cm depths of soil, the root length increased by 5.95 and 7.30 % under mild and intense stress, respectively. Under mild stress, root area and root volume were positively correlated with STI. Consequences of drought stress, manifested as declined relative water content and chlorophyll, could be associated with a decrease in the activity of antioxidant enzymes. Some tall fescue genotypes had extensive root systems, high photosynthetic capacity, and less YR in the field. These genotypes may adapt to drought through drought avoidance and drought tolerance mechanisms. The application of principle component analysis for screening suitable genotypes was also discussed.

International audience; Climate change models predict more frequent and severe droughts in the world. For the future adaptation of perennial forage grasses, identification and selection of genotypes with enhanced recovery and persistence under repeated drought is crucial. The aim of the present study was to investigate traits related to persistence, recovery, and root adaptation in three sets of tall fescue genotypes with different flowering date, after successive drought stress. A total of 72 genotypes of tall fescue (24 medium, 24 late, 24 early flowering) were assessed for agro-morphological traits and persistence during 2010-2014 under two levels of soil moisture. In 2015, irrigation was withheld and all genotypes were evaluated for drought recovery in the field. Then 18 genotypes were chosen based on field results to evaluate their root traits at two irrigation levels (control and intense drought stress) in pots. In the field, plant persistence decreased from year 2 to 5 under both control and intense drought stress conditions in all three sets of genotypes, although persistence of late flowering genotypes was higher. The results also indicated that most of the genotypes with greatest persistence had higher recovery after the final severe drought. Persistence and recovery were positively correlated with root length (RL), root area (RA), root volume (RV), root weight (RW) and root to shoot ratio (R/S) under both soil moisture. Moreover, genotypes with higher recovery after drought had more RL, RA, RV and R/S than others. Application of principle component analysis to identify genotypes of interest for future breeding programs is discussed.

The genetic basis of post-drought recovery, summer dormancy and persistence under drought stress is little investigated in orchardgrass (Dactylis glomerata L.). In this study, polycross populations (half-sib genotypes) were evaluated under normal and regulated deficit irrigation for 3 years (2012–14) in the field. Irrigation was then withheld in 2015, and the genotypes were evaluated for post-drought recovery and persistence. The results showed that regulated deficit irrigation decreased forage yield, yield components, persistence and recovery. A broad range of general combining ability was observed for most of the measured traits. Moderate to high estimation of narrow-sense heritability for yield components, persistence, recovery and summer dormancy index indicated that phenotypic selection may be successful to attain genetic progress. Under deficit irrigation, flowering time was positively correlated with persistence and recovery, indicating that selection based on lateness would lead to improvement in these traits. Stable and superior families across environments were identified based on regression coefficients and principle component analysis that would be desirable in future breeding programs.
