Persister cells are a small subpopulation of non-growing bacteria within a population that can survive long exposures to antibiotic treatment. Following antibiotic removal, persister cells can regrow and populate, playing a key role in the chronic reoccurrence of bacterial infections. The development of new molecules and methods to kill bacterial persisters is critical. Essential oils and other natural products have long been studied for their antimicrobial effects. Here, we studied the effectiveness of tea tree essential oil (TTO), a common component in many commercial care products, against Escherichia coli and Staphylococcus epidermidis persister cells. Using biphasic kill curve assays, we found that concentrations of 0.5% and 1.0% TTO for E. coli and S. epidermidis , respectively, completely eradicated persister cells over a period of 24 h, with the component terpinen-4-ol responsible for most of the killing. Using a colorimetric assay, it was determined that the TTO exhibited its anti-persister effects through a membrane disruption mechanism.

Nitric oxide (NO) is a chemical weapon within the arsenal of immune cells, but is also generated endogenously by different bacteria. Pseudomonas aeruginosa are pathogens that contain an NO-generating nitrite (NO 2 - ) reductase (NirS), and NO has been shown to influence their virulence. Interestingly, P. aeruginosa also contain NO dioxygenase (Fhp) and nitrate (NO 3 - ) reductases, which together with NirS provide the potential for NO to be metabolically cycled (NO→NO 3 - →NO 2 - →NO). Deeper understanding of NO metabolism in P. aeruginosa will increase knowledge of its pathogenesis, and computational models have proven to be useful tools for the quantitative dissection of NO biochemical networks. Here we developed such a model for P. aeruginosa and confirmed its predictive accuracy with measurements of NO, O 2 , NO 2 - , and NO 3 - in mutant cultures devoid of Fhp or NorCB (NO reductase) activity. Using the model, we assessed whether NO was metabolically cycled in aerobic P. aeruginosa cultures. Calculated fluxes indicated a bottleneck at NO 3 - , which was relieved upon O 2 depletion. As cell growth depleted dissolved O 2 levels, NO 3 - was converted to NO 2 - at near-stoichiometric levels, whereas NO 2 - consumption did not coincide with NO or NO 3 - accumulation. Assimilatory NO 2 - reductase (NirBD) or NorCB activity could have prevented NO cycling, and experiments with ΔnirB, ΔnirS, and ΔnorC showed that NorCB was responsible for loss of flux from the cycle. Collectively, this work provides a computational tool to analyze NO metabolism in P. aeruginosa, and establishes that P. aeruginosa use NorCB to prevent metabolic cycling of NO., (Copyright © 2017 International Metabolic Engineering Society. Staphylococcus aureus is a notorious pathogen with a propensity to cause chronic, non-healing wounds. Bacterial persisters have been implicated in the recalcitrance of S. aureus infections, and this motivated us to examine the persistence of S. aureus to ciprofloxacin, a quinolone antibiotic. Upon treatment of exponential phase S. aureus with ciprofloxacin, we observed that survival was a non-monotonic function of ciprofloxacin concentration. Maximal killing occurred at 1 µg/mL ciprofloxacin, which corresponded to survival that was up to ~40-fold lower than that obtained with concentrations ≥ 5 µg/mL. Investigation of this phenomenon revealed that the non-monotonic response was associated with prophage induction, which facilitated killing of S. aureus persisters. Elimination of prophage induction with tetracycline was found to prevent cell lysis and persister killing. We anticipate that these findings may be useful for the design of quinolone treatments.

Nutrient demand is a fundamental characteristic of rapidly proliferating cells. Vitamin B12 is vital for cell proliferation; thus neoplastic cells have an increased demand for this essential nutrient. In this study we exploited the vitamin B12 uptake pathway to probe the nutritional demand of proliferating cells with a radiolabeled B12 derivative in various preclinical tumor models. We describe the synthesis and biological evaluations of copper-64-labeled B12 -ethylenediamine-benzyl-1,4,7-triazacyclononane-N,N',N''-triacetic acid (B12 -en-Bn-NOTA-(64) Cu), the first example of a B12 derivative for positron emission tomography (PET) imaging. Small-animal imaging and pharmacological evaluation show high tumor uptake ranging from 2.20 to 4.84% ID g(-1) at 6 h post-administration. Competition studies with excess native B12 resulted in a 95% decrease in tumor accumulation, indicating the specificity of this radiopharmaceutical for B12 endocytotic transport proteins. Bacterial persisters are phenotypic variants with extraordinary tolerances toward antibiotics. Persister survival has been attributed to inhibition of essential cell functions during antibiotic stress, followed by reversal of the process and resumption of growth upon removal of the antibiotic. Metabolism plays a critical role in this process, since it participates in the entry, maintenance, and exit from the persister phenotype. Here, we review the experimental evidence that demonstrates the importance of metabolism to persistence, highlight the successes and potential of targeting metabolism in the search for anti-persister therapies, and discuss the current methods and challenges to understand persister physiology.

The rise in non-heme iron (NHI) concentration observed in skeletal muscle of aging rodents is thought to contribute to the development of sarcopenia. The source of the NHI has not been identified, nor have the physiological ramifications of elevated iron status in aged muscle been directly examined. Therefore, we assessed plantaris NHI and heme iron (HI) levels in addition to expression of proteins involved in iron uptake (transferrin receptor-1; TfR1), storage (ferritin), export (ferroportin; FPN), and regulation (iron regulatory protein-1 (IRP1) and -2 (IRP2)) of male F344xBN F1 rats (n=10/group) of various ages (8, 18, 28, 32, and 36 months) to further understand iron regulation in aging muscle. In a separate experiment, iron chelator (pyridoxal isonicotinoyl hydrazone; PIH) or vehicle was administered to male F344xBN F1 rats (n=8/group) beginning at 30 months of age to assess the impact on plantaris muscle mass and function at ~36 months of age. Principle findings revealed the increased NHI concentration in old age was consistent with concentrating effects of muscle atrophy and reduction in HI levels, with no change in the total iron content of the muscle. The greatest increase in muscle iron content occurred during the period of animal growth and was associated with downregulation of TfR1 and IRP2 expression. Ferritin upregulation did not occur until senescence and the protein remained undetectable during the period of muscle iron content elevation. Lastly, administration of PIH did not significantly (p>0.05) impact NHI or measures of muscle atrophy or contractile function. In summary, this study confirms that the elevated NHI concentration in old age is largely due to the loss in muscle mass. The increased muscle iron content during aging does not appear to associate with cytosolic ferritin storage, but the functional consequences of elevated iron status in old age remains to be determined., (© 2013.)

Human PYY(3-36) (hPYY3-36) is a 34 amino acid hormone that has received a great deal of attention due to its effects on appetite regulation. hPYY(3-36) was modified at the N-terminus with an octahistidine tag and factor Xa protease sequence along with the small ubiquitin-like modifier (SUMO) tag and expressed in Escherichia coli. The protein was purified from clarified E. coli lysate by immobilized metal affinity chromatography (IMAC) with a yield of 30±7 mg/L of induced culture returned as an average over seven runs, and its identity was confirmed by Western blot and hPYY antibody recognition. The SUMO-tagged hPYY(3-36) was digested with two different proteases to return either His-tagged hPYY(3-36) or unmodified hPYY(3-36): (1) digestion with SUMO protease proceeded at about 50% efficiency yielding His-tagged hPYY(3-36); (2) digestion with factor Xa protease proceeded at greater than 90% efficiency yielding final hPYY(3-36). Products were purified from the digestion mixtures by reverse-phase high-performance liquid chromatography (C(18)) or IMAC, respectively, the identities were confirmed by mass spectrometry and hPYY antibody recognition, and the folded state of His-tagged hPYY(3-36) was investigated by circular dichroism spectroscopy., (Copyright © 2012 Elsevier Inc. hPYY(3-36) injections have shown positive effects on appetite regulations, sparking increased interest in hPYY(3-36) research. Of great interest is oral delivery of hPYY(3-36) that can achieve clinically relevant weight-loss outcomes in what would be a highly patient compliant route. Successful oral delivery of other peptides utilizing the vitamin B12 pathway has been shown but below clinically relevant levels. Herein, we present clinically relevant in vivo oral delivery of B12-hPYY(3-36) conjugates.

Isostructural, "clamshell"-like, neutral dimeric pyrophosphato complexes of general formula {[M(bipy)](2)(μ-P(2)O(7))} [M = Pd(II) (1) or Pt(II) (2)] were synthesized and studied through single-crystal X-ray diffraction, IR, (31)P NMR spectroscopy, and MALDI-TOF mass spectrometry. Compound 1 was synthesized through the reaction of palladium(II) acetate, 2,2'-bipyridine (bipy), and sodium pyrophosphate (Na(4)P(2)O(7)) in water. Compound 2 was prepared through two different routes. The first involved the reaction of the Pt(IV) precursor Na(2)PtCl(6), bipy, and Na(4)P(2)O(7) in water, followed by reduction in DMF. The second involved the reaction of the Pt(II) precursor K(2)PtCl(4), bipy, and Na(4)P(2)O(7) in water. Both complexes crystallize in the monoclinic chiral space group Cc as hexahydrates, 1·6H(2)O (1a, yellow crystals) and 2·6H(2)O (2a, orange crystals), and exhibit a zigzag chain-like supramolecular packing arrangement with short and long intra/intermolecular metal-metal distances [3.0366(3)/4.5401(3) Å in 1a; 3.0522(3)/4.5609(3) Å in 2a]. A second crystalline phase of the Pt species was also isolated, with formula 2·3.5H(2)O (2b, deep green crystals), characterized by a dimer-of-dimers (pseudo-tetramer) structural submotif. Green crystals of 2b could be irreversibly converted to the orange form 2a by exposure to air or water, without retention of crystallinity, while a partial, reversible crystal-to-crystal transformation occurred when 2a was dried in vacuo. (31)P NMR spectra recorded for both 1 and 2 at various pHs revealed the occurrence of a fluxional protonated/deprotonated system in solution, which was interpreted as being composed, in the protonated form, of [HO=PO(3)](+) (P(α)) and O=PO(3) (P(β)) pyrophosphate subunits. Compounds 1 and 2 exhibited two successive one-electron oxidations, mostly irreversible in nature; however, a dependence upon pH was observed for 1, with oxidation only occurring in strongly basic conditions. Density functional theory and atoms in molecules analyses showed that a d(8)-d(8) interaction was present in 1 and 2.
