Abstract

The motor thalamus (MTh) plays a crucial role in the basal ganglia (BG)-cortical loop in motor information codification. Despite this, there is limited evidence of MTh functionality in normal and Parkinsonian conditions. To shed light on the functional properties of the MTh, we examined the effects of acute and chronic dopamine (DA) depletion on the neuronal firing of MTh neurons, cortical/MTh interplay and MTh extracellular concentrations of glutamate (GLU) and gamma-aminobutyric acid (GABA) in two states of DA depletion: acute depletion induced by the tetrodotoxin (TTX) and chronic denervation obtained by 6-hydroxydopamine (6-OHDA), both infused into the medial forebrain bundle (MFB) in anesthetized rats. The acute TTX DA depletion caused a clear-cut reduction in MTh neuronal activity without changes in burst content, whereas the chronic 6-OHDA depletion did not modify the firing rate but increased the burst firing. The phase correlation analysis underscored that the 6-OHDA chronic DA depletion affected the MTh-cortical activity coupling compared to the acute TTX-induced DA depletion state. The TTX acute DA depletion caused a clear-cut increase of the MTh GABA concentration and no change of GLU levels. On the other hand, the 6-OHDA-induced chronic DA depletion led to a significant reduction of local GABA and an increase of GLU levels in the MTh. These data show that MTh is affected by DA depletion and support the hypothesis that a rebalancing of MTh in the chronic condition counterbalances the profound alteration arising after acute DA depletion state.

Abstract

Background: Non-ST-elevation myocardial infarction (NSTEMI) and unstable angina (UA) are caused often by destabilization of non-flow limiting inflamed coronary artery plaques. 18F-fluorodeoxyglucose (FDG) uptake with positron emission tomography/computed tomography (PET/CT) reveals plaque inflammation, while intracoronary optical coherence tomography (OCT) reliably identifies morphological features of coronary instability, such as plaque rupture or erosion. We aimed to prospectively compare these two innovative biotechnologies in the characterization of coronary artery inflammation, which has never been attempted before. Methods: OCT and FDG PET/CT were performed in 18 patients with single vessel coronary artery disease, treated by percutaneous coronary intervention (PCI) with stent implantation, divided into 2 groups: NSTEMI/UA (n = 10) and stable angina (n = 8) patients. Results: Plaque rupture/erosion recurred more frequently [100% vs 25%, p = 0.001] and FDG uptake was greater [TBR median 1.50 vs 0.87, p = 0.004] in NSTEMI/UA than stable angina patients. FDG uptake resulted greater in patients with than without plaque rupture/erosion [1.2 (0.86–1.96) vs 0.87 (0.66–1.07), p = 0.013]. Among NSTEMI/UA patients, no significant difference in FDG uptake was found between ruptured and eroded plaques. The highest FDG uptake values were found in ruptured plaques, belonging to patients with NSTEMI/UA. OCT and PET/CT agreed in 72% of patients [p = 0.018]: 100% of patients with plaque rupture/erosion and increased FDG uptake had NSTEMI/UA. Conclusion: For the first time, we demonstrated that the correspondence between increased FDG uptake with PET/CT and morphology of coronary plaque instability at OCT is high.

Abstract

In this article, the Moderate Resolution Imaging Spectroradiometer (MODIS) Bidirectional Reflectance Distribution Function (BRDF)/Albedo product (MCD43) is evaluated over a heterogeneous agricultural area in the framework of the Earth Observation: Optical Data Calibration and Information Extraction (EODIX) project campaign, which was developed in Barrax (Spain) in June 2011. In this method, two models, the RossThick-LiSparse-Reciprocal (RTLSR) (which corresponds to the MODIS BRDF algorithm) and the RossThick-Maignan-LiSparse-Reciprocal (RTLSR-HS), were tested over airborne data by processing high-resolution images acquired with the Airborne Hyperspectral Scanner (AHS) sensor. During the campaign, airborne images were retrieved with different view zenith angles along the principal and orthogonal planes. Comparing the results of applying the models to the airborne data with ground measurements, we obtained a root mean square error (RMSE) of 0.018 with both RTLSR and RTLSR-HS models. The evaluation of the MODIS BRDF/Albedo product (MCD43) was performed by comparing satellite images with AHS estimations. The results reported an RMSE of 0.04 with both models. Additionally, taking advantage of a homogeneous barley pixel, we compared in situ albedo data to satellite albedo data. In this case, the MODIS albedo estimation was (0.210 +/- 0.003), while the in situ measurement was (0.204 +/- 0.003). This result shows good agreement in regard to a homogeneous pixel.

A sensitive enzyme immunoassay including microtiter plates coated with porcine thyroglobulin is used to measure the amount of active phytohemagglutinin (PHA) in dietary supplements containing proteins extracts from kidney bean (Phaseolus vulgaris). It was found that phytohemagglutinin induces serious consequences for metabolism and health when consumed raw or only lightly cooked.

Abstract

Cyclic adenosine monophosphate (cAMP) regulates long-term potentiation (LTP) and ameliorates memory in healthy and diseased brain. Increasing evidence shows that, under physiological conditions, low concentrations of amyloid β (Aβ) are necessary for LTP expression and memory formation. Here, we report that cAMP controls amyloid precursor protein (APP) translation and Aβ levels, and that the modulatory effects of cAMP on LTP occur through the stimulation of APP synthesis and Aβ production.

Abstract

Pathological oscillations characterize the firing discharge of different basal ganglia (BG) stations in rat models of Parkinson's disease. Most recent literature focused on the prominence of the beta frequency band in awake rats. Yet, in 6-hydroxydopamine-lesioned animals, the firing discharge of the globus pallidus (GP) and the substantia nigra reticulata are in phase with urethane-induced slow wave cortical activity. The neuronal basis of this pathological synergy at low frequency is widely debated. In order to understand the role of substantia nigra pars compacta (SNc) signalling in the development of pathological synchronization, we performed a pharmacological inactivation of the medial forebrain bundle (MFB) through tetrodotoxin (TTX), which led to a dramatic, but reversible, reduction of the dopamine content in the striatum. This procedure caused a significant contralateral akinesia, detectable as soon as anaesthesia vanished, and lasting about 3-4 h. We sought to determine the electrophysiological counterpart of this transient Parkinsonian-like hypokinetic syndrome. Hence, we obtained the electrocorticogram (ECoG) and single unit recordings from GP and subthalamic nucleus (STN) in normal rats before and after the TTX injection in MFB. Intriguingly, the TTX-mediated inactivation of MFB induced a fast developing coherence between cortex and GP and a significant increase of the cortex/STN synchronization. The intra-GP iontophoretic delivery of haloperidol or the GABA(A) receptor antagonist bicuculline induced a short term cortex/GP synchronization. Strikingly, STN inactivation by local muscimol reversed both haloperidol- and TTX-mediated coherence between cortex and GP. Our data show that an abnormal cortical/BG synchronization, at low frequency, can be reproduced also without SNc neuronal loss and striatal cytoarchitectonic alterations. In addition, our results, which represent an acute and reversible Parkinsonism based upon impaired cable properties, seem compatible with the interpretation of acute changes of the functional interplay between cortex and the STN-GP pathway as a key factor mechanism underlying the fast deep brain stimulation-induced acute Off-On transitions.

Abstract

Nutritional science is increasingly seen in sports practice as an instrument capable of influencing the performance, recovery and gene expression of the athlete. Vice versa, the performance itself is able to modify human metabolism and the use of substrates for energy purposes. Since even small changes are important for the athlete, nutritional science must be considered a precision medicine in which standardized protocols apply to the needs of the individual athlete (based on the type of sport, the frequency of training and its intensity, the goals in terms of weight and muscle mass) in order to minimize measurement errors. The study of the baseline composition of the athlete's body is essential to build up a nutritional plan that follows the athlete before, during and after competition, with the aim of optimizing performance and preventing the onset of fatigue. The purpose of this review is to sum up the most recent guidelines, underlining the key points of the current state of art on the best strategies to achieve specific goals in terms of changes or maintenance of body weight, preparing adequately the athlete for competition and encouraging recovery, with a brief mention to the psycho-behavioural dimension that nutrition acquires in sports practice. Abstract

It is universally known the link between physical inactivity and coronary heart disease. It is estimated that over 30% of coronary heart disease worldwide is due to physical inactivity. This evidence draws attention to the importance of exercise in primary prevention of heart disease and in secondary prevention measures, such as cardiac rehabilitation. International guidelines started to include exercise training beside pharmacological therapies, developing a specific type of training modality tailored for each subpopulation of cardiovascular patients and their risk factors. Many different types of exercise protocols, arranged for intensity, method, type, application, control and setting, exist and cardiopulmonary exercise test represents the gold standard for the assessment of functional evaluation in exercise prescription. In clinical practice, it is not always feasible to group patients into a specific category, due to the variety of their co-morbidities and risk factors: in secondary prevention, exercise intensity must yet be safe but also tailored to achieve the outcome goal of rehabilitation. The purpose of this report is to introduce the innovation of recent notions published in the scientific literature, trying to insert them into clinical practice giving a simple, but yet effective, instrument to the health care professionals.
