IntroductionThere are no reports in LATAM related to longitudinal humoral and cellular response to adenovirus based COVID-19 vaccines in people with Multiple Sclerosis (pwMS) under different disease modifying therapies (DMTs) and neutralization of the Omicron and Wuhan variants of SARS-COV-2.MethodsIgG anti- SARS-COV-2 spike titer were measured in a cohort of 101 pwMS under fingolimod, dimethyl fumarate, cladribine and antiCD20, as well as 28 healthy controls (HC) were measured 6 weeks after vaccination with 2nd dose (Sputnik V or AZD1222) and 3nd dose (homologous or heterologous schedule). Neutralizing capacity was against Omicron (BA.1) and Wuhan (D614G) variants and pseudotyped particles and Cellular response were analyzed.ResultsMultivariate regression analysis showed anti-cd20 (β= -,349, 95% CI: -3655.6 - -369.01, p=0.017) and fingolimod (β=-,399, 95% CI: -3363.8 - -250.9, p=0.023) treatments as an independent factor associated with low antibody response (r2 adjusted=0.157). Abstract In the evolving landscape of data science and computational biology, Causal Networks (CNs) have emerged as a robust framework for modelling causal relationships among elements of complex systems derived from experimental data. CNs can efficiently model causal relationships emerging in a single system while comparing multiple systems, allowing to understand rewiring in different cells, tissues, and physiological states with a deeper perspective. Despite the existence of network models, namely differential networks, that have been used to compare coexpression and correlation structures, causality needs to be introduced in differential analysis to robustly provide direction to the edges of such networks, in order to better understand the flows of information, and also to better intervene in their functioning, for example for agricultural or pharmacological purposes. Resolved to reach this ambitious goal, we introduce Differential Causal Networks (DCNs), a novel framework that represents differences between two existing CNs. A DCN is obtained from experimental data by comparing two CNs, and it is a power tool for highlighting differences in causal relations. After a careful definition and design of DCNs, we test our algorithm to model possible differential causal relationships between genes responsible for the onset of type 2 diabetes mellitus-related pathologies considering patients’ sex at the tissue level. DCNs allowed us to shed light on causal differences between sexes across nine tissues. We also compare differences among three possible definitions of DCNs to highlight similarities and differences of biological importance. Abstract This novel study applies targeted functional proteomics to examine tissues and cells obtained from a cohort of individuals with severe obesity who underwent bariatric surgery (BS), using a Reverse-Phase Protein Array (RPPA). In obese individuals, visceral adipose tissue (VAT), but not subcutaneous adipose tissue (SAT), shows activation of DNA damage response (DDR) markers including ATM, ATR, histone H2AX, KAP1, Chk1, and Chk2, alongside senescence markers p16 and p21. Additionally, stress-responsive metabolic markers, such as survivin, mTOR, and PFKFB3, are specifically elevated in VAT, suggesting both cellular stress and metabolic dysregulation. Conversely, peripheral blood mononuclear cells (PBMCs), while exhibiting elevated mTOR and JNK levels, did not present significant changes in DDR or senescence markers. Following BS, unexpected increases in phosphorylated ATM, ATR, and KAP1 levels, but not in Chk1 and Chk2 nor in senescence markers, were observed. This was accompanied by heightened levels of survivin and mTOR, along with improvement in markers of mitochondrial quality and health. This suggests that, following BS, pro-survival pathways involved in cellular adaptation to various stressors and metabolic alterations are activated in circulating PBMCs. Moreover, our findings demonstrate that the DDR has a dual nature. In the case of VAT from individuals with obesity, chronic DDR proves to be harmful, as it is associated with senescence and chronic inflammation. Conversely, after BS, the activation of DDR proteins in PBMCs is associated with a beneficial survival response. This response is characterized by metabolic redesign and improved mitochondrial biogenesis and functionality. This study reveals physiological changes associated with obesity and BS that may aid theragnostic approaches.

Abstract Depression is a prevalent and incapacitating condition with a significant impact on global morbidity and mortality. Although the immune system’s role in its pathogenesis is increasingly recognized, there is a lack of comprehensive understanding regarding the involvement of innate and adaptive immune cells. To address this gap, we conducted a multicenter case–control study involving 121 participants matched for sex and age. These participants had either an active (or current) major depressive episode (MDE) (39 cases) or a remitted MDE (40 cases), including individuals with major depressive disorder or bipolar disorder. We compared these 79 patients to 42 healthy controls (HC), analyzing their immunological profiles. In blood samples, we determined the complete cell count and the monocyte subtypes and lymphocyte T-cell populations using flow cytometry. Additionally, we measured a panel of cytokines, chemokines, and neurotrophic factors in the plasma. Compared with HC, people endorsing a current MDE showed monocytosis (p = 0.001), increased high-sensitivity C-reactive protein (p = 0.002), and erythrocyte sedimentation rate (p = 0.003), and an altered proportion of specific monocyte subsets. CD4 lymphocytes presented increased median percentages of activation markers CD69+ (p = 0.007) and exhaustion markers PD1+ (p = 0.013) and LAG3+ (p = 0.014), as well as a higher frequency of CD4+CD25+FOXP3+ regulatory T cells (p = 0.003). Additionally, patients showed increased plasma levels of sTREM2 (p = 0.0089). These changes are more likely state markers, indicating the presence of an ongoing inflammatory response during an active MDE. The Random Forest model achieved remarkable classification accuracies of 83.8% for MDE vs. HC and 70% for differentiating active and remitted MDE. Interestingly, the cluster analysis identified three distinct immunological profiles among MDE patients. Cluster 1 has the highest number of leukocytes, mainly given by the increment in lymphocyte count and the lowest proinflammatory cytokine levels. Cluster 3 displayed the most robust inflammatory pattern, with high levels of TNFα, CX3CL1, IL-12p70, IL-17A, IL-23, and IL-33, associated with the highest level of IL-10, as well as β-NGF and the lowest level for BDNF. This profile is also associated with the highest absolute number and percentage of circulating monocytes and the lowest absolute number and percentage of circulating lymphocytes, denoting an active inflammatory process. Cluster 2 has some cardinal signs of more acute inflammation, such as elevated levels of CCL2 and increased levels of proinflammatory cytokines such as IL-1β, IFNγ, and CXCL8. Similarly, the absolute number of monocytes is closer to a HC value, as well as the percentage of lymphocytes, suggesting a possible initiation of the inflammatory process. The study provides new insights into the immune system’s role in MDE, paving the ground for replication prospective studies targeting the development of diagnostic and prognostic tools and new therapeutic targets.

Age-related macular degeneration (AMD) is a common retinal pathology characterized by degeneration of macula’s retinal pigment epithelium (RPE) and photoreceptors, visual impairment, or loss. Compared to wet AMD, dry AMD is more common, but lacks cures; therefore, identification of new potential therapeutic targets and treatments is urgent. Increased oxidative stress and declining antioxidant, detoxifying systems contribute to the pathophysiologic mechanisms underlying AMD. The present work shows that the Embryonic Lethal Abnormal Vision-Like 1/Human antigen R (ELAVL1/HuR) and the Vascular Endothelial Growth Factor (VEGF) protein levels are higher in the RPE of both dry and wet AMD patients compared to healthy subjects. Moreover, increased HuR protein levels are detected in the retina, and especially in the RPE layer, of a dry AMD model, the nuclear factor erythroid 2-related factor 2 (Nrf2) / peroxisome proliferator-activated receptor gamma coactivator-1α (PGC-1α) double knock-out mouse. The crosstalk among Nrf2, HuR and VEGF has been also studied in ARPE-19 cells in basal and stressful conditions related to the AMD context (i.e., oxidative stress, autophagy impairment, Nrf2 deficit), offering new evidence of the mutual influence between Nrf2 and HuR, of the dependence of VEGF expression and secretion by these two factors, and of the increased susceptibility of cells to stressful conditions in Nrf2- or HuR-impaired contexts. Overall, this study shows evidence of the interplay among Nrf2, HuR and VEGF, essential factors for RPE homeostasis, and represents an additional piece in the understanding of the complex pathophysiologic mechanisms underlying AMD.

Effective communication between humans and machines requires artificial tools to adopt a human-like social perspective. The Theory of Mind (ToM) enables understanding and predicting mental states and behaviours, crucial for social interactions from childhood through adulthood. Artificial agents with ToM skills can better coordinate actions, such as in warehouses or healthcare. Incorporating ToM in AI systems can revolutionise our interactions with intelligent machines. This proposal emphasises the current focus on first-order ToM models in the literature and investigates the potential of creating a computational model for higher-order ToM.

Summary: Mosquito behavioral assays are an important component in vector research and control tool development. Here, we present a protocol for rearing Anopheles mosquitoes, performing host-seeking behavioral bioassays, and collecting 3D flight tracks in a large wind tunnel. We describe steps for setting up host-seeking landing assays, both as a non-choice and as a dual-choice assay, and analyzing flight tracks. Background: Suicidal ideation and attempt (SI/SA) have been associated with dysregulation of the immune response and inflammation. However, few studies have explored how innate and acquired cellular immunity impact on the peripheral immune response. Our study addresses this gap by examining the composition of peripheral immune cells and humoral markers among individuals with current SI/SA, individuals with a history of SI/SA, and healthy controls (HC). Additionally, we aim to explore whether depressive symptoms settle the relationship between inflammation and SI/SA. Methods: This is a multicenter case-control study that included 105 participants. Clinical and demographic characterists together with hemogram parameters, soluble pro and anti-inflamatory factors, and specific innate and adaptive immune cell populations were compared among patients with current SI/SA (n = 21), a history of lifetime SI/SA (n = 42), and HC (n = 42). Results: Patients with both current and lifetime SI/SA had a significant increase in the absolute count of monocytes and in the monocyte/lymphocyte ratio (MLR). Additionally, patients with current and lifetime SI/SA showed a significant increase in high-sensitivity C- reactive protein (hs-CRP), and patients with lifetime SI/SA also showed higher levels of Erythrocyte Sedimentation Rate (ESR). The cellular inflammatory status of patients with SI/SA was characterized by altered proportions of monocytes with higher levels of nonclassical and intermediate monocytes. No differences were observed in the number of lymphocytes and the proportion of CD4 and CD8 between patients and HC, but we found differences in markers of exhaustion of CD4 lymphocytes, with increased levels of Programmed cell death protein 1 (PD1) in Current SI/SA and Lymphocyte activation gene 3 (LAG3) in Current SI/SA and Lifetime SI/SA compared to HC. The plasmainflammatory status was marked by higher levels of soluble Triggering receptor expressed on myeloid cells 2 (sTREM2) in patients with lifetime SI/SA compared to HC. Finally, the multinomial analysis indicates that inflammation and depressive symptoms are independently associated with SI/SA. Conclusion: This study highlights the association of immunological alterations with SI/SA. Furthremore, SI/SA is independently influenced by depressive symptoms and inflammation. This may have important therapeutic implications, as in these patients, it may be necessary to treat the inflammatory process beyond treating the depressive symptoms.

This study presents an updated interpretation of geological data collected between 1984 and 2022. The area under consideration holds significant regional importance as it is located between the Internal Nappe Zone (INZ) and the Regional Mylonitic Complex (RMC). Re-evaluation of the geological data has highlighted a more intricate structural framework than what is currently documented in the existing literature. This paper aims to illustrate, through structural analysis, that the Posada Valley Shear Zone (PVSZ) does not serve as the transitional boundary between the Inner Nappe Zone and the Regional Mylonitic Complex or High-Grade Metamorphic Complex (HGMC) as traditionally thought. Instead, the authors’ findings indicate that the transition boundary is confined to a shear band with a variable thickness ranging from 10 to 70 m at its widest points. The development of the Posada Valley Shear Zone is characterized by a series of transitions from mylonite I S-C to mylonite II S-C, extending over approximately 5 km. The formation of the Posada Valley Shear Zone is chronologically confined between the development of the East Variscan Shear Zone (EVSZ) and the emplacement of the Late Variscan granites. The differing orientations of Sm and S3 observed in the mylonitic events of the Posada Valley Shear Zone and the Regional Mylonitic Complex, respectively, are likely attributable to an anticlockwise rotation of the shortening directions during the upper Carboniferous period. Furthermore, this study proposes that the Condensed Isogrades Zone (CIZ), despite its unclear formation mechanism, should be recognized as the true transition zone between the Inner Nappe Zone and the Regional Mylonitic Complex or High-Grade Metamorphic Complex. This new interpretation challenges the previously accepted notion of increasing Variscan metamorphic zonation toward the northeast. This conclusion is supported by the identification of the same NE–SW orientation of the D2 tectonic event in both the Old Gneiss Complex (OGC in the Regional Mylonitic Complex) and the lithologies of the Inner Nappe Zone and the Condensed Isogrades Zone. The comprehensive analysis and new insights provided in this paper contribute to a refined understanding of the geological relationships and processes within this region, offering significant implications for future geological studies and interpretations.

Abstract Peer pressure can influence risk-taking behavior and it is particularly felt during adolescence. With artificial intelligence (AI) increasingly present in a range of everyday human contexts, including virtual environments, it is important to examine whether AI can have an impact on human’s decision making processes and behavior. By using the balloon analogue risk task (BART) evaluating propensity to take risk, in this study 113 adolescents' risk-taking behavior was measured when playing alone and in the presence of either a robot avatar or human avatar. In the avatar conditions, participants performed the BART while the avatars either (1) verbally incited risk-taking or (2) discouraged risk-taking (experimental tasks). Risk-taking behavior in the BART was assessed in terms of total number of pumps, gain and explosions. Tendency to impulsivity was also evaluated, as well as the effects of age and gender on risky behavior. The main finding showed a significant effect of both avatars on risk-taking tendency, with riskier behavior during incitement than discouragement conditions, the latter being also substantially different from the playing-alone condition. The results of this study open up new questions in a very sensitive and timely topic and offer various insights into the effect of nudging on adolescents’ behavior in virtual contexts.

Summary: Malaria prevention relies on mosquito control interventions that use insecticides and exploit mosquito behavior. The rise of insecticide resistance and changing transmission dynamics urgently demand vector control innovation. To identify behavioral traits that could be incorporated into such tools, we investigated the flight and landing response of Anopheles coluzzii to human-like host cues. We show that landing rate is directly proportional to the surface area of thermal stimulus, whereas close-range orientation is modulated by both thermal and visual inputs. We modeled anopheline eye optics to theorize the distance at which visual targets can be detected under a range of conditions, and experimentally established mosquito preference for landing on larger targets, although landing density is greater on small targets. Target orientation does not affect landing rate; however, vertical targets can be resolved at greater distance than horizontal targets of the same size. Mosquito traps for vector control could be significantly enhanced by incorporating these features.

Abstract Bariatric surgery (BS) is an effective intervention for severe obesity and associated comorbidities. Although several studies have addressed the clinical and metabolic effects of BS, an integrative analysis of the complex body response to surgery is still lacking. We conducted a longitudinal data study with 36 patients with severe obesity who were tested before, 6 and 12 months after restrictive BS for more than one hundred blood biomarkers, including clinical, oxidative stress and metabolic markers, peptide mediators and red blood cell membrane lipids. By using a synthetic data-driven modeling based on principal component and correlation analyses, we provided evidence that, besides the early, well-known glucose metabolism- and weight loss-associated beneficial effects of BS, a tardive, weight-independent increase of the hepatic cholesterol metabolism occurs that is associated with potentially detrimental inflammatory and metabolic effects. Canonical correlation analysis indicated that oxidative stress is the most predictive feature of the BS-induced changes of both glucose and lipids metabolism. Our results show the power of multi-level correlation analysis to uncover the network of biological pathways affected by BS. This approach highlighted potential health risks of restrictive BS that are disregarded with the current practice to use weight loss as surrogate of BS success.

Objetivo: Conocer las características clínico-epidemiológicas de los pacientes ambulatorios del servicio de kinesiología de un hospital público de la Ciudad Autónoma de Buenos Aires y describir la prevalencia de sarcopenia en los mismos. Materiales y método: Estudio descriptivo, observacional, prospectivo realizado en el servicio de kinesiología de un hospital público de Ciudad Autónoma de Buenos Aires entre febrero y marzo de 2023. Se incluyeron individuos de 18 años o más, derivados al servicio. Se utilizó el algoritmo propuesto por el European Working Group on Sarcopenia in Older People 2 para el diagnóstico. Resultados: De 137 pacientes evaluados la media de edad fue de 51 (DE 15,6) años y el 63,5 % fueron mujeres. La mayoría de las derivaciones fueron por patologías musculoesqueléticas. La prevalencia de sarcopenia probable fue de 11,7 %, tuvieron una media de edad de 65,1 (DE 14,3) años y 81,3 % eran mujeres. El 75 % de los individuos no realizaban actividad física y tuvieron un índice de masa corporal (IMC) de 28,3 (DE 6,8). Conclusión: Las características de los pacientes ambulatorios del servicio de kinesiología de un hospital público de Ciudad Autónoma de Buenos Aires indicaron una media de edad de 51 (DE 15,6) años, predominando las mujeres. La mayoría no realizaban actividad física y tenían una mediana de IMC de 28,4 kg/m2. Si bien no se encontraron pacientes con sarcopenia confirmada, un 11,7 % tuvo sarcopenia probable.

Environmental issues are at the center of the political and cultural debate, representing one of the greatest challenges of our century. Sustainability and pro-environmental conducts are recognized as increasingly urgent to address the decay of ecosystems. To support the acquisition of attitudes that give greater consideration to environmental issues, experiencing a sense of connection with nature has been acknowledged in psychology as a particularly relevant individual component. Among the most commonly used scales in Anglo-Saxon context to analyses this feeling is the Connectedness to Nature Scale (CNS) assessing the emotional and experiential bond between humans and nature. To examine the reliability and validity of this scale in the Italian context, a study including 271 Italian adults (44,3% female; 55% males; Mean age = 34.70; SD = 13.584; age-range = 18-65 years) was conducted to establish evidence supporting the internal consistency of the CNS, as well as its ability to measure convergent, discriminant, and predictive validity. A Confirmatory Factor Analysis showed that CNS in Italian has a single-factor structure as reported in the original version by Mayer and Frantz. Furthermore, as expected, positive correlations were observed between the CNS and pro-environmental attitudes and negative correlations with civic moral disengagement. Finally, as assumed, the CNS positively correlated with mental well-being. A broad vision of this study concerns the idea that individuals who have a stronger connection with nature are likely to exhibit reduced tendencies to cause harm to it.

In Europe, the mental health law legal framework has had several changes throughout the years to achieve and develop new reforms, better mental health care, and protect the human rights of patients. The UK national data shows rising detention rates and the disproportionate use of the legal framework among people from black and minority ethnic groups. At the national level, compulsory admissions are lower in Italy; it also shows that it has increased in the last few years in both countries. The lack of ethnic national data, especially in Italy, limited the ability to understand compulsory admission, discrimination, and stigma in mental health. The present study aims to compare the legal framework of mental health law and compulsory hospital admission in Italy and the UK. A review of each country’s latest amendments to mental health law and the number of compulsory hospital admissions was conducted to understand the impact of changes in mental health care.

Dimethyl fumarate (DMF) is an FDA-approved drug for the treatment of psoriasis and multiple sclerosis. DMF is known to stabilize the transcription factor Nrf2, which in turn induces the expression of antioxidant response element genes. It has also been shown that DMF influences autophagy and participates in the transcriptional control of inflammatory factors by inhibiting NF-κB and its downstream targets. DMF is receiving increasing attention for its potential to be repurposed for several diseases. This versatile molecule is indeed able to exert beneficial effects on different medical conditions through a pleiotropic mechanism, in virtue of its antioxidant, immunomodulatory, neuroprotective, anti-inflammatory, and anti-proliferative effects. A growing number of preclinical and clinical studies show that DMF may have important therapeutic implications for chronic diseases, such as cardiovascular and respiratory pathologies, cancer, eye disorders, neurodegenerative conditions, and systemic or organ specific inflammatory and immune-mediated diseases. This comprehensive review summarizes and highlights the plethora of DMF’s beneficial effects and underlines its repurposing opportunities in a variety of clinical conditions.

Sardinia (Italy) represents one of the most comprehensive cross-sections of the Variscan orogen. The metamorphic and structural complexity characterizing its axial zone still presents many unresolved issues in the current state of knowledge. The data presented from the structural study of the entire axial zone of this area have allowed the authors to propose a subdivision into two new structural complexes. In particular, a younger complex is identified as the New Gneiss Complex, containing remnants of an older and higher-grade metamorphic complex defined as the Old Gneiss Complex. The structural and geometric relationships between the two complexes suggest the redefinition of the axial zone of Sardinia as part of the intracontinental East Variscan Shear Zone/medium-temperature Regional Mylonitic Complex. Comparable relationships are also highlighted in many other areas of the Variscan chain (e.g., Morocco, Corsica, Maures Massif, and Argentera Massif). Extending this new structural interpretation to other axial zones of the South European Variscan orogen could provide new hints for reconstructing the collision boundaries between Gondwana and Laurussia in the late Carboniferous to the early Permian periods.

BackgroundSuicidal and non-suicidal self-injurious behaviors are among the leading causes of death and injury in adolescents and youth worldwide. Mobile app development could help people at risk and provide resources to deliver evidence-based interventions. There is no specific application for adolescents and young people available in Spanish. Our group developed CALMA, the first interactive mobile application with the user in Spanish, which provides tools based on Dialectical Behavioral Therapy to manage a crisis of suicidal or non-suicidal self-directed violence with the aim of preventing suicide in adolescents and youth.MethodsTo test the effectiveness, safety and level of engagement of the CALMA app in people aged 10 to 19 who are treated in mental health services of two public hospitals, we will conduct a parallel-group, two-arm randomized controlled trial. Participants will be assessed face-to-face and via video call at four timepoints: day-0 (baseline), day-30, day-60, and day-90. A total of 29 participants per group will be included. Change in the frequency of suicidal and non-suicidal self-injurious behaviors will be compared between groups, as well as the level of emotional dysregulation, level of app engagement and time of psychiatric admission during the follow-up period.DiscussionThis study is particularly relevant to young people given their widespread use of mobile technology, while there are currently no available smartphone app-based self-guided psychological strategies in Spanish that attempt to reduce suicidal behavior in adolescents who are assisted in the public health sector from low and middle-income countries in Latin America.Clinical trial registrationhttps://clinicaltrials.gov/, NCT05453370.

Las fracturas de platillos tibiales son lesiones frecuentes. Una de sus complicaciones postoperatorias es la pérdida de reducción con desviación de los ejes en los planos coronal, sagital y axial. La depresión ósea genera incongruencia articular con pérdida de tensión de estructuras ligamentarias indemnes, causando una pseudo laxitud con inestabilidad. Esto requiere de una corrección ósea para aumentar la tensión de dichas estructuras ligamentarias, y lograr así recuperar la congruencia y estabilidad articular en todo el rango de movilidad. El objetivo del presente trabajo es reportar un caso de inestabilidad medial en un paciente joven, quien, tras una fractura de platillo tibial medial mal consolidada, requirió una osteotomía selectiva biplanar e intraarticular para corregir dicha deformidad.

The present review aims to summarize the more recent scientific literature and updated state of the art on the research effort spent in adapting hardware–software tools to understand the true needs of honeybee colonies as a prerequisite for any sustainable management practice. A SWOT (Strengths, Weaknesses, Opportunities, and Threats) analysis was also performed with the aim of identifying the key factors that could support or impair the diffusion of precision beekeeping (PB) systems. Honeybee husbandry, or beekeeping, is starting to approach precision livestock farming (PLF), as has already happened in other animal husbandry sectors. A transition from the current paradigm of rational beekeeping to that of precision beekeeping (PB) is thus expected. However, due to the peculiarities of this species and the related farming practices, the PB technological systems (PB systems) are still undergoing a development process that, to some extent, limits their large-scale practical application. Several physical–chemical (weight, temperature, humidity, sound, gases) and behavioral traits (flight activity, swarming) of the hive are reviewed in light of the evolution of sensors, communication systems, and data management approaches. These advanced sensors are equipped with a microprocessor that records data and sends it to a remote server for processing. In this way, through a Wireless Sensor Network (WSN) system, the beekeeper, using specific applications on a personal computer, tablet, or smartphone, can have all the above-mentioned parameters under remote control. In general, weight, temperature, and humidity are the main hive traits monitored by commercial sensors. Surprisingly, flight activity sensors are rarely available as an option in modular PB systems marketed via the web. The SWOT analysis highlights that PB systems have promising strength points and represent great opportunities for the development of beekeeping; however, they have some weaknesses, represented especially by the high purchasing costs and the low preparedness of the addressed operators, and imply some possible threats for beekeeping in terms of unrealistic perception of the apiary status if they applied to some hives only and a possible adverse impact on the honeybees’ colony itself. Even if more research is expected to take place in the next few years, indubitably, the success of commercial PB systems will be measured in terms of return on investment, conditioned especially by the benefits (higher yields, better colonies’ health) that the beekeeper will appraise as a consequence of their use.

Enteric glial cells (EGCs), the major components of the enteric nervous system (ENS), are implicated in the maintenance of gut homeostasis, thereby leading to severe pathological conditions when impaired. However, due to technical difficulties associated with EGCs isolation and cell culture maintenance that results in a lack of valuable in vitro models, their roles in physiological and pathological contexts have been poorly investigated so far. To this aim, we developed for the first time, a human immortalized EGC line (referred as ClK clone) through a validated lentiviral transgene protocol. As a result, ClK phenotypic glial features were confirmed by morphological and molecular evaluations, also providing the consensus karyotype and finely mapping the chromosomal rearrangements as well as HLA-related genotypes. Lastly, we investigated the ATP- and acetylcholine, serotonin and glutamate neurotransmitters mediated intracellular Ca2+ signaling activation and the response of EGCs markers (GFAP, SOX10, S100β, PLP1, and CCL2) upon inflammatory stimuli, further confirming the glial nature of the analyzed cells. Overall, this contribution provided a novel potential in vitro tool to finely characterize the EGCs behavior under physiological and pathological conditions in humans.

Attributing mental states to others, such as feelings, beliefs, goals, desires, and attitudes, is an important interpersonal ability, necessary for adaptive relationships, which underlies the ability to mentalize. To evaluate the attribution of mental and sensory states, a new 23-item measure, the Attribution of Mental States Questionnaire (AMS-Q), has been developed. The present study aimed to investigate the dimensionality of the AMS-Q and its psychometric proprieties in two studies. Study 1 focused on the development of the questionnaire and its factorial structure in a sample of Italian adults (N = 378). Study 2 aimed to confirm the findings in a new sample (N = 271). Besides the AMS-Q, Study 2 included assessments of Theory of Mind (ToM), mentalization, and alexithymia. A Principal Components Analysis (PCA) and a Parallel Analysis (PA) of the data from Study 1 yielded three factors assessing mental states with positive or neutral valence (AMS-NP), mental states with negative valence (AMS-N), and sensory states (AMS-S). These showed satisfactory reliability indexes. AMS-Q’s whole-scale internal consistency was excellent. Multigroup Confirmatory Factor Analysis (CFA) further confirmed the three-factor structure. The AMS-Q subscales also showed a consistent pattern of correlation with associated constructs in the theoretically predicted ways, relating positively to ToM and mentalization and negatively to alexithymia. Thus, the questionnaire is considered suitable to be easily administered and sensitive for assessing the attribution of mental and sensory states to humans. The AMS-Q can also be administered with stimuli of nonhuman agents (e.g., animals, inanimate things, and even God); this allows the level of mental anthropomorphization of other agents to be assessed using the human as a term of comparison, providing important hints in the perception of nonhuman entities as more or less mentalistic compared to human beings, and identifying what factors are required for the attribution of human mental traits to nonhuman agents, further helping to delineate the perception of others’ minds.

The superconformal central charge is an important quantity for theories emerging from geometrical engineering of Quantum Field Theory since it is linked, for example, to the scaling dimension of fields. Butti and Zaffaroni construction of the central charge for toric Calabi-Yau threefold geometries is a powerful tool but its implementation could be quite tricky. Here we present an equivalent new construction based on a 2-simplexes decomposition of the toric diagram.

In recent decades, per- and polyfluoroalkyl substances (PFASs) have garnered widespread public attention due to their persistence in the environment and detrimental effects on the health of living organisms, spurring the generation of several transcriptome-centered investigations to understand the biological basis of their mechanism. In this study, we collected 2144 publicly available samples from seven distinct animal species to examine the molecular responses to PFAS exposure and to determine if there are conserved responses. Our comparative transcriptional analysis revealed that exposure to PFAS is conserved across different tissues, molecules and species. We identified and reported several genes exhibiting consistent and evolutionarily conserved transcriptional response to PFASs, such as ESR1, HADHA and ID1, as well as several pathways including lipid metabolism, immune response and hormone pathways. This study provides the first evidence that distinct PFAS molecules induce comparable transcriptional changes and affect the same metabolic processes across inter-species borders. Our findings have significant implications for understanding the impact of PFAS exposure on living organisms and the environment. We believe that this study offers a novel perspective on the molecular responses to PFAS exposure and provides a foundation for future research into developing strategies for mitigating the detrimental effects of these substances in the ecosystem.

Virtual environments have been widely adopted for design and training tasks in the industrial domain. Low-cost automation (LCA) is a technology that automatizes some activities using mostly standard automation mechanisms available off the shelf. However, LCA systems should adapt to existing standard production lines and workstations. Thus, workers must customize standard LCA templates and perform adaptation and customization steps. This activity can be very time consuming with physical LCA systems, and in case of errors, it may be necessary to rebuild many parts from scratch. Thus, LCA systems would greatly benefit from a design and prototyping step experienced in a virtual simulation environment. An immersive virtual reality (IVR) application for rapid and easy prototyping of LCA solutions has been investigated in previous work; the assessment of the system usability proved that the users highly appreciated the proposed solutions. This research explores further improvements to exploit the existing IVR application as a training tool for LCA prototyping trainees. The proposed application now provides users with two different interaction paradigms based on the VIVE controllers and the Manus Prime II data gloves. The application’s interface has been revised to allow a proper comparison of the two interaction models. The two interfaces have been compared, involving 12 participants in an LCA building task. The System Usability Scale (SUS) and the NASA Task Load Index (TLX) questionnaires have been used to assess the usability and workload of the two solutions.

The Portofino Conglomerate (PC) cropping out in the Eastern Liguria is an approximately 500 m thick, very gently folded succession mainly composed of poorly bedded and mostly matrix-supported conglomerates. It stratigraphically rests on the Helminthoid Flysch (UA3) thrusted onto the Antola Unit. We vertically distinguished three mostly ruditic litho/petrofacies: (i) Paraggi (fP) with carbonate clasts from an Helminthoid Flysch succession; (ii) Monte Pallone (fMP) with prevailing carbonate and meta-carbonate clasts and minor quartz-rich (meta)siliciclastic and high-pressure–low-pressure (HP-LP) metabasite clasts; and (iii) Monte Bocche (fMB) with dominant quartz-rich (meta)siliciclastic, meta-carbonate clasts, and minor granitoid elements and medium-temperature–high-temperature (MT-HT) regional metamorphic rocks. The middle-upper Eocen age of Paraggi litho/petrofacies is constrained by well-preserved microforaminifers (e.g., Globigerinatheka) recovered in the matrix. During its sedimentation, the directions of the paleocurrents would indicate that the PC underwent a counterclockwise rotation coeval with the first Cenozoic rotational phase of the Sardinia–Corsica system (50–30 Ma) and then stopped before the sedimentation of the Monte Pallone and Monte Bocche litho/petrofacies. The vertical compositional variation in the sedimentary inputs suggested that the PC is the result of a progressive deepening of the erosional level of a tectonic pile that can be located in the Ligurian Alps Chain. We considered the PC as the likely apical part of a submarine fan deposited in a piggy-back/thrust-top basin within the Alpine nappe stack. This sedimentary body was later tectonically transported eastward with its UA3 Helminthoid Flysch substrate (similarly to Epiligurian Units of the Northern Apennines) onto the Apenninic orogenic system (i.e., the Antola Unit).

Since July 1st., 2020, Law 27.551 entered into effectiveness in our country, changing the Civil and Commercial Code mainly regarding the property lease contract and in particular regarding the property with residence purposes lease. The legislation is divided into four headings, each of them corresponding to the modifications of the Code, the passing of complementary normative, the creation of the National Program of social ‘rent’, and the promotion of alternative means of conflict resolutions, respectively, reaffirming a protective nature in sights of the protection of the lessee. This paper will focus on the first two headings, gathering different jurists’ opinions in that regard. Additionally, it will analyze the connection between Law 27.551 and the emergency legislation passed as a result of de Covid-19 pandemic regarding leases, on the grounds of the so called “sources dialogue”.

The COVID-19 pandemic is showing troubling othering demographic discourses. For older adults in particular, there are concerning thematics that should be shined light on. In this editorial, we provide perspectives from three countries: Norway, Italy and the United States. We provide four topics of discussion that can be utilized to further understand othering discoures of the COVID-19 pandemic, as well as potential future disasters.

Abstract Objectives To assess the trueness of a solid index (SI) in the full-arch (FA) implant impression, and to compare it with that of two intraoral scanners (IOSs). A type-IV gypsum model of a completely edentulous patient with 8 implant scanbodies (SBs) was scanned with a desktop scanner (7Series®) to obtain a reference virtual model (RVM), and with two IOSs (CS 3700® and Emerald S®). Five scans were taken with each IOS. Based on the RVM, an SI (custom tray consisting of hollow cylinders connected by a bar) was fabricated and used to capture a physical impression of the model; from this, a second gypsum model was derived and scanned with a desktop scanner (D15®). The SI-derived and the IOSs-derived models were superimposed onto the RVM, to evaluate trueness. Results The overall mean trueness was 29 μm (± 26) for the SI-derived model, versus 42.4 μm (± 14.7) for CS 3700® and 52.2 μm (± 4.6) for Emerald S®. Despite its limitations (in vitro design, a limited number of models evaluated, RVM captured with a desktop scanner) this study supports the use of SI for FA implant impressions. Further studies are needed to confirm this evidence.

Abstract Background and objective Radiation pneumonitis (RP) could be a lethal complication of lung cancer treatment. No reliable predictors of RP severity have been recognized. This prospective pilot study was performed to identify early predictors of high grade lung toxicity and to evaluate clinical, biological or dosimetric features associated with different grades of toxicity. Method Sixteen patients with non-small cell lung cancer with indication of concurrent chemoradiotherapy using 60 Gy/2 Gy/fraction starting at cycle one of platinum based chemotherapy were included. Bronchoalveolar lavage (BAL), pulmonary function testing (PFT), and 18F-2-fluoro-2-deoxy-D-glucose positron-emission tomography was performed before radiotherapy (RT), after three weeks of treatment, and two months post-RT. For analysis, patients were grouped by grade (low [G1-G2] vs. high [G3-G5]). The two groups were compared to identify predictors of RP. Protein expression BAL and lung tissue metabolism was evaluated in two patients (RP-G1 vs. RP-G3). Categorical variables such as comorbidities, stages and locations were summarized as percentages. Radiation doses, pulmonary function values and time to RP were summarized by medians with ranges or as means with standard deviation. Longitudinal analysis PFT was performed by a T-test. Results All 16 patients developed RP, as follows: G1 (5 pts; 31.3%); G2 (5 pts; 31.3%); G3 (5 pts; 31.3%); and G5 (1 pts; 6.1%). Patients with high grade RP presented significant decrease (p = 0.02) in diffusing lung capacity for carbon monoxide (DLCO) after three weeks of RT. No correlation between dosimetric values and RP grades was observed. BAL analysis of the selected patients showed that CXCL-1, CD154, IL-1ra, IL-23, MIF, PAI-1 and IFN-γ were overexpressed in the lungs of the RP-G3 patient, even before treatment. The pre-RT SUVmax value in the RP-G3 patient was non-significantly higher than in the patient with RP-G1. Conclusions RT induces some degree of RP. Our data suggest that decrease in DLCO% is the most sensitive parameter for the early detection of RP. Moreover, we detect biological differences between the two grades of pneumonitis, highlighting the potential value of some cytokines as a prognostic marker for developing high grade lung toxicity. Further multicenter studies with larger sample size are essential to validate these findings.

Abstract Background Veno-arterial extracorporeal life support (VA-ECLS) results in cardiopulmonary shunting with reduced native cardiac output (NCO). Low NCO occurrence is common and associated with risk of thromboembolic and pulmonary complications. Practical tools for monitoring NCO during VA-ECLS would therefore be valuable. Pulse pressure (PP) and end-tidal carbon dioxide (EtCO2) are known to be related to cardiac output. We have designed a study to test whether PP and EtCO2 were efficient for the monitoring of NCO during VA-ECLS. Methods In this prospective single-center observational study, patients who underwent a VA-ECLS for cardiogenic shock from January 2016 to October 2017 were included, provided low NCO was suspected by a PP

Group B Streptococcus (GBS) is a Gram-positive bacterium able to switch from a harmless commensal of healthy adults to a pathogen responsible for invasive infections in neonates. The signals and regulatory mechanisms governing this transition are still largely unknown. CodY is a highly conserved global transcriptional regulator that links nutrient availability to the regulation of major metabolic and virulence pathways in low-G+C Gram-positive bacteria. In this work, we investigated the role of CodY in BM110, a GBS strain representative of a hypervirulent lineage associated with the majority of neonatal meningitis. Deletion of codY resulted in a reduced ability of the mutant strain to cause infections in neonatal and adult animal models. The observed decreased in vivo lethality was associated with an impaired ability of the mutant to persist in the blood, spread to distant organs, and cross the blood-brain barrier. Notably, the codY null mutant showed reduced adhesion to monolayers of human epithelial cells in vitro and an increased ability to form biofilms, a phenotype associated with strains able to asymptomatically colonize the host. RNA-seq analysis showed that CodY controls about 13% of the genome of GBS, acting mainly as a repressor of genes involved in amino acid transport and metabolism and encoding surface anchored proteins, including the virulence factor Srr2. CodY activity was shown to be dependent on the availability of branched-chain amino acids, which are the universal cofactors of this regulator. These results highlight a key role for CodY in the control of GBS virulence.

As the literature clearly shows, supporting the development of reflective awareness skills is undoubtedly an important element in learning processes. In psychology, multiple theoretical approaches and research work have delved into the study of what is termed implicit knowledge. In particular, the relationship between human activity (in terms of actions, thoughts, beliefs, motivation, and reasoning) and different levels of awareness is a relevant subject of analysis that we consider as the core of this paper. In order to deepen our understanding of the concepts of awareness and reflexive activity, we refer to Pierre Vermersch’s psycho-phenomenological approach and Piaget’s theory of cognitive awareness. In this paper, we aim to show the use of the reflective approach centered on the elicitation of specific lived experiences. The objective is to promote in students a process of awareness of their activity and their role within the university context. Two students case studies from the University of Salerno took part in the research. The method used was based on narrative interviews that makes use of some techniques and principles of elicitation interviews, a conversational approach that supports the participant in focusing and describing a specific experience. The interviewer guides subjects, without induction, through the transition from the implicit of lived experience (particularly action) to the explicit of reflected awareness of that action. The data collected show how reflective activity by means of guided evocations of lived experiences helped participants become aware of how some distortions and irrational thoughts (related to the self and context) negatively affected them during the activities. The reflective work fostered by the elicitation of experiences often allows for enhanced self-awareness; the subject takes ownership of the action, analyzes it, and understands the difficulties.

Berberine (BBR) is known for its antitumor activity and photosensitizer properties in anti-cancer photodynamic therapy (PDT), and it has previously been favorably assayed against glioblastoma multiforme (GBM)-derived cells. In this work, two BBR hydrophobic salts, dodecyl sulfate (S) and laurate (L), have been encapsulated in PLGA-based nanoparticles (NPs), chitosan-coated by the addition of chitosan oleate in the preparation. NPs were also further functionalized with folic acid. All the BBR-loaded NPs were efficiently internalized into T98G GBM established cells, and internalization increased in the presence of folic acid. However, the highest mitochondrial co-localization percentages were obtained with BBR-S NPs without folic acid content. In the T98G cells, BBR-S NPs appeared to be the most efficient in inducing cytotoxicity events and were therefore selected to assess the effect of photodynamic stimulation (PDT). As a result, PDT potentiated the viability reduction for the BBR-S NPs at all the studied concentrations, and a roughly 50% reduction of viability was obtained. No significant cytotoxic effect on normal rat primary astrocytes was observed. In GBM cells, a significant increase in early and late apoptotic events was scored by BBR NPs, with a further increase following the PDT scheme. Furthermore, a significantly increased depolarization of mitochondria was highlighted following BBR-S NPs’ internalization and mostly after PDT stimulation, compared to untreated and PDT-only treated cells. In conclusion, these results highlighted the efficacy of the BBR-NPs-based strategy coupled with photoactivation approaches to induce favorable cytotoxic effects in GBM cells.

The Echus-Kasei region on Mars has been exposed to different episodic volcanic, fluvial, and glacial events in Amazonian time. The goal of the present work is to demonstrate the usefulness of radar instruments to find preserved late Amazonian subsurface structures that may have been encapsulated underneath recent lava flows on Mars. We have analysed 27 radar observations of the SHAllow RADar (SHARAD) instrument on board the Mars Reconnaissance Orbiter (MRO), over the region of Echus Chasma. We discovered the presence of subsurface reflectors in five consecutive SHARAD radargrams at a depth from 35 to 79 m beneath the structure of a lava fan that formed about 59 ± 4 Ma ago. Some vents are preserved above the surface of this lava flow, which stands at a height of 80 m above the surrounding surface. A few kilometres to the north, we find other subsurface reflectors at a depth of about 30 m and a long pit chain formed by the collapse of a lava tube. These kinds of subsurface late Amazonian structures are of interest for astrobiology because they date from the last period when the planet still experienced intense volcanic activity over regions that were previously extensively covered by water.

The pandemic from COVID-19 causes a health threat for many countries and requires an internationally coordinated response due to the high spread of the infection. The current local and international situation gives rise to logistical and ethical considerations regarding the imbalance between needs for assistance and availability of health resources in the continuation of the emergency. A shortage condition will require healthcare professionals to choose between patients who will have access to respiratory support and those who will have to continue without. The sharing of criteria for the introduction of patients to the different therapeutic paths is fundamental to prevent the onset of ethical issues. The present paper analyzes the critical issues related to the scarcity of healthcare resources and the limitation of access to intensive care with the aim of proposing ethically sustainable principles for the management of the current pandemic situation.

Dimethyl fumarate (DMF) is a small molecule currently approved and used in the treatment of psoriasis and multiple sclerosis due to its immuno-modulatory, anti-inflammatory, and antioxidant properties. As an Nrf2 activator through Keap1 protein inhibition, DMF unveils a potential therapeutical use that is much broader than expected so far. In this comprehensive review we discuss the state-of-art and future perspectives regarding the potential repositioning of this molecule in the panorama of eye pathologies, including Age-related Macular Degeneration (AMD). The DMF’s mechanism of action, an extensive analysis of the in vitro and in vivo evidence of its beneficial effects, together with a search of the current clinical trials, are here reported. Altogether, this evidence gives an overview of the new potential applications of this molecule in the context of ophthalmological diseases characterized by inflammation and oxidative stress, with a special focus on AMD, for which our gene–disease (KEAP1-AMD) database search, followed by a protein–protein interaction analysis, further supports the rationale of DMF use. The necessity to find a topical route of DMF administration to the eye is also discussed. In conclusion, the challenge of DMF repurposing in eye pathologies is feasible and worth scientific attention and well-focused research efforts.

Fentanyl is a synthetic L-opioid receptor agonist, approximately 100 times more potent than morphine, that is experiencing an upward trend in the field of abuse. Fentanyl patches’ abusive consumption can occur either by transdermal absorption or through other atypical and ingenious routes. In the present case, a 29-year-old man with a history of illicit drug use was found dead in a suburban neighborhood of an Italian city. At autopsy, lungs appeared increased in weight and showed minute subpleural hemorrhages. Airways contained abundant reddish foamy material; in addition, a fentanyl patch protective film was found inside the left main bronchus. Toxicological analysis revealed the presence of morphine, fentanyl, BEG and ethyl alcohol in peripheric blood; 6-MAM was also revealed in urine. Findings collected during post-mortem investigations allowed us to identify fentanyl consumption as the cause of death. Fentanyl consumption presumably took place by chewing of a transdermal patch, with subsequent aspiration of the protective film. The pathophysiology of death can be identified as combined respiratory failure—both central suppression and a fentanyl-induced increase in muscular stiffness; a further minor contribution may be identified in the mechanical airflow obstruction caused by the presence of the protective film at the bronchial level.

Pupil detection plays a key role in eye and gaze video-based tracking algorithms. Various algorithms have been proposed through the years in order to improve the performances or the robustness in real-world scenarios. However, the development of an algorithm which excels in both execution time and pupil detection precision is still an open challenge. This paper presents a novel, feature-based eye-tracking algorithm for pupil detection. Morphological operators are used to remove corneal reflections and to reduce noise in the pupil area prior to the pupil detection step: this solution allows to significantly reduce the computational overhead without lowering the tracking precision. Moreover, a shape validation step is performed after the elliptical fitting and, if the elliptical shape is not detected properly, a set of additional steps is performed to improve the pupil estimation. The proposed solution, Pupil Detection after Isolation and Fitting (PDIF), has been compared with other state-of-the-art tracking algorithms that use morphological operations such as ElSe (Ellipse Selection) and ExCuSe (Exclusive Curve Selector) to evaluate both speed and robustness; the proposed algorithm has been tested over numerous datasets offering different pupil detection challenges. Obtained results show how PDIF provides comparable tracking precision at a significantly lower computational cost compared to ElSe and ExCuSe.

Background: Depression is a highly prevalent disorder that is one of the leading causes of disability worldwide. Despite an unknown aetiology, evidence suggests that the innate and adaptive immune systems play a significant role in the development and maintenance of major depressive disorder (MDD). The non-competitive glutamatergic N-methyl-D-aspartate receptor (NMDAR) antagonist, (R,S)-ketamine (ketamine), has demonstrated rapid and robust efficacy as an antidepressant when administered at sub-anaesthetic doses. Methods: Our goal was to characterize the pro-inflammatory profile of patients with MDD by measuring pro-inflammatory cytokines in plasma and circulating monocyte subsets and to understand how ketamine induces an anti-inflammatory program in monocyte and macrophages in vitro and vivo. Finding: Our results show that patients with MDD without other comorbidities (N = 33) exhibited significantly higher levels of pro-inflammatory IL-12 and IL-6 in plasma and that these cytokines were associated with increased numbers of non-classical (CD11b+CD16brightCD14neg) monocytes and increased activation state (CD40+CD86+) of classical monocytes in circulation. Remarkably, we have demonstrated that sub-anaesthetic doses of ketamine programs human monocytes into M2c-like macrophages by inducing high levels of CD163 and MERTK with intermediate levels of CD64 and stimulating mTOR-associated gene expression in vitro. The NMDAR antagonist MK-801, but not the α-amino-3‑hydroxy-5-methyl-4-isoxazolepropionic acid receptor (AMPAR) antagonist, NBQX, also polarizes macrophages to an M2c-like phenotype, but this phenotype disappears upon mTOR pathway inhibition. Sub-anaesthetic doses (10 mg/kg) of ketamine administration in mice both promote reduction of circulating classical pro-inflammatory monocytes and increase of alternative M2 macrophage subtypes in the spleen and CNS. Interpretation: Our results suggest an anti-inflammatory property of ketamine that can skew macrophages to an M2-like phenotype, highlighting potential therapeutic implications not only for patients with MDD but also other inflammatory-based diseases. This article proposes to work with the Industry Life Cycle (ILC) approach for a better understanding of technological innovations role in industry of manufacturers of mobile cellular telephony devices (TMC devices), as well as in evolution of this sector. Three qualitative strategies of information processing and analysis are used for construction of meaning. This information was organized, systematized and confronted with assumptions of the industry life cycle model (ILC) proposed by Utterback and Abernathy (1975) for the period between 1997 and 2017. The analysis suggests that the behavior of the TMC device industry conforms to the assumptions of the ILC model. It is also worth noting Apple’s iPhone as the dominant design since its appearance (2007) until today (2019). It also shows that, as of the date of this investigation and according to the ILC assumptions, this industry is in transition between the “transitional” and “specific” phases, as well as the importance of technological convergence as a source of innovation in this industry. Some trajectories or possible future directions in the evolution and technological innovations for this industry are also proposed, always from the perspective of the ILC model

Abstract Background Intensive care unit (ICU)-acquired weakness (ICU-AW) and ICU-acquired diaphragm dysfunction (ICU-DD) occur frequently in mechanically ventilated (MV) patients. It is unknown whether they have different risk factors and different impacts on outcome. This study was designed to (1) describe the respective risk factors associated with ICU-AW and severe ICU-DD and (2) evaluate the respective impact of ICU-AW and severe ICU-DD on outcome. Methods Post hoc analysis of two prospective cohort studies conducted in two ICUs. In patients mechanically ventilated for at least 24 h undergoing a first spontaneous breathing trial, severe ICU-DD was defined as diaphragm twitch pressure
