Snakes of the Philodryadini tribe are included in the Dipsadidae family, which is a diverse group of rear-fanged snakes widespread in different ecological conditions, including habitats and diet. However, little is known about the composition and effects of their venoms despite their relevance for understanding the evolution of these snakes or even their impact on the occasional cases of human envenoming. In this study, we integrated venom gland transcriptomics, venom proteomics and functional assays to characterize the venoms from eight species of the Philodryadini tribe, which includes the genus Philodryas, Chlorosoma and Xenoxybelis. The most abundant components identified in the venoms were snake venom metalloproteinases (SVMPs), cysteine-rich secretory proteins (CRISPs), C-type lectins (CTLs), snake endogenous matrix metalloproteinases type 9 (seMMP-9) and snake venom serinoproteinases (SVSPs). These protein families showed a variable expression profile in each genus. SVMPs were the most abundant components in Philodryas, while seMMP-9 and CRISPs were the most expressed in Chlorosoma and Xenoxybelis, respectively. Lineage-specific differences in venom composition were also observed among Philodryas species, whereas P. olfersii presented the highest amount of SVSPs and P. agassizii was the only species to express significant amounts of 3FTx. The variability observed in venom composition was confirmed by the venom functional assays. Philodryas species presented the highest SVMP activity, whereas Chlorosoma species showed higher levels of gelatin activity, which may correlate to the seMMP-9 enzymes. The variability observed in the composition of these venoms may be related to the tribe phylogeny and influenced by their diets. In the presented study, we expanded the set of venomics studies of the Philodryadini tribe, which paves new roads for further studies on the evolution and ecology of Dipsadidae snakes.

Fifteen well-defined strains of Aeromonas of thirteen species were analyzed by SDS protein electrophoretic analysis (SDS-PAGE) and random amplified polymorphic DNA analysis (RAPD). The comparison between the patterns obtained by both methods allowed differentiating all the strains. Clusters formed by the unweighted pair group method with arithmetic averages applied to protein data correlates with the genetic and biochemical information about the species. The results show that protein fingerprinting has the potential to differentiate Aeromonas species, but the low qualitative variation indicates that this technique is not efficient for the characterization of strains within a species. Conversely, RAPD fingerprinting allows the identification of strains but the high variability limits its potential as an aiding method for species identification.Quinze linhagens de Aeromonas pertencentes a treze espécies foram avaliadas através de eletroforese de proteínas totais (SDS-PAGE) e segmentos de DNA amplificados ao acaso (RAPD). A comparação entre os padrões obtidos por ambos métodos permitiu diferenciar todas as linhagens. Agrupamentos formados com base nos dados protéicos mostraram relação com informações bioquímicas e genéticas a respeito das espécies. Os resultados mostraram que análises protéicas têm potencial para diferenciar espécies de Aeromonas, mas a baixa variação qualitativa indica que esta técnica não é eficiente para a caracterização entre linhagens dentro de espécies. Ao contrário, marcadores de RAPD permitem identificar linhagens, mas a alta variabilidade limita seu potencial como método auxiliar na identificação de espécies.

Southern Brazil is considered an area of low Hepatitis B endemicity, but some areas of higher endemicity have been described in the Southwest of Parana and Santa Catarina states. The aim of this study was to evaluate viral genotypes circulating throughout Parana state. PCR amplification and partial sequencing of the S gene was carried out in 228 samples from HBsAg positive candidate blood donors. Samples have been collected in seven different counties (Cascavel, Curitiba, Foz do Iguacu, Francisco Beltrao, Maringa, Londrina and Paranagua). The most common HBV genotype in Parana state was D (82.9%; 189/228), followed by A (14.1%; 32/228). Genotypes F (1.3%; 3/228), C (1.3%; 3/228) and H (0.4%; 1/228) were also found. Distribution of genotypes was different in the studied counties, but genotype D was the most frequent in all of them. In Francisco Beltrao, all studied samples belonged to genotype D. The high prevalence of HBV genotype D in South of Brazil is explained by the intense migration of settlers from Europeans countries. Subgenotypes A1 and A2 were identified circulating in all cities where HBV/A was found. As observed in other areas of Brazil, HBV/A1 is more frequent than the HBV/A2 in Parana state and its presence was significantly larger in black and mulatto individuals. Genotype C was found only in individuals with Asian ancestry from Londrina and Maringa. Most HBV/F sequences identified in this study were classified as subgenotype F2a that was previously described in Brazil. The sole case of subgenotype F4 was from Foz do Iguacu city, near to Northern Argentina, where F4 is highly prevalent. The single genotype H sample was from Curitiba. This is the first case of this genotype described in Brazil. Further studies should be carried out to determine if more genotype H samples can be found in other populations from Brazil.

As coleções biológicas constituem uma das mais importantes ferramentas para obtenção de informações sobre a biodiversidade. Nas coleções zoológicas de vertebrados, um dos meios clássicos de preservação de espécimes é através de fixação em solução aquosa de formaldeído a 10% (ou formalina), seguida de preservação em etanol 70%. A formalina pode ser considerada como um bom fixador de tecidos biológicos, ainda que não chegue a níveis de preservação morfológica suficientemente refinada para a utilização da microscopia eletrônica, o que necessariamente requer o uso de fixadores mais precisos contendo glutaraldeído. Por outro lado, com a aplicação da biologia molecular, tornaram-se cada vez mais frequentes os estudos que necessitam da preservação do DNA de amostras, principalmente retiradas do músculo ou do fígado, visando o acesso à sistemática molecular. Essas amostras, porém, devem necessariamente ser fixadas em etanol, não podendo ter contato com fixadores aldeídicos. Para tanto, muitos pesquisadores zoólogos passaram a fixar os espécimes coletados em campo diretamente no etanol 70%, capaz de preservar o DNA das amostras. Essa prática se tornou especialmente comum durante expedições de campo para coleta de anfíbios, talvez devido à pouca sobrevida dos espécimes coletados e à grande permeabilidade e fragilidade cutâneas, que permitem que esses animais sejam rapida e facilmente fixados em etanol 70%. No entanto, essa prática fornece material inadequadamente fixado às coleções zoológicas, muitas vezes inviabilizando o seu aproveitamento para possíveis estudos morfológicos. Entre os anfíbios, a pele é diferenciada em cada espécie considerada, o que a torna um bom material para a avaliação das condições de preservação morfológica. Dessa forma, visando à conscientização dos curadores de coleções e dos pesquisadores responsáveis pela coleta de animais no campo, a proposta desse estudo é a avaliação comparativa da preservação morfológica em uma espécie de perereca da Mata Atlântica, Trachycephalus mesophaeus, submetida à preservação pelo uso de diferentes agentes fixadores. Em paralelo, utilizamos as peles preservadas em fixador contendo glutaraldeído para realizarmos um estudo morfológico detalhado da pele dessa perereca, principalmente no que se refere às glândulas de veneno e às glândulas mucosas. Biological collections are important tools for obtaining information about biodiversity. In vertebrate zoological collections, one of most classic methods of specimen preservation is through fixation using 10% aqueous formaldehyde solution (or formalin), followed by 70% ethanol preservation. Formalin can be considered a good fixative of biological tissues, although it does not reach sufficiently refined morphological preservation levels for electron microscopy, which necessarily requires the use of more accurate glutaraldehyde-containing fixatives. On the other hand, application of molecular biology studies requiring DNA preservation especially of muscle or liver samples aiming to access the molecular systematics, have become increasingly frequent. DNA samples, however, cannot have contact with aldehyde fixatives and necessarily must be preserved in ethanol. For this reason, aiming at the preservation of DNA, many zoologists are fixing field specimens directly in 70% ethanol. This practice has become especially common in field expeditions for amphibian collection, perhaps due to the poor survival of the collected specimens and the high skin permeability and fragility that allow these animals to be quickly and easily fixed in 70% ethanol. This practice, however, provides material badly fixed to zoological collections, often turning its use impossible for morphological purposes. Among amphibians, the skin is differentiated in each species considered, which makes it a good material for the evaluation of morphological preservation conditions. Thus, aiming at the awareness of the zoological collection curators and of field researchers, the main purpose of this study is the comparative evaluation of the morphological preservation in a common tree frog species of the Atlantic Rainforest, Trachycephalus mesophaeus, submitted to preservation using different fixative agents. In parallel, we used the skin preserved with glutaraldehyde-containing fixative to perform a detailed morphological study of the skin of this tree frog, especially regarding the poison and mucous glands.

The tribe Philodryadini is constituted by a rich group of neotropical snakes that are highly diverse ecologically and morphologically. Currently, 24 species compose the tribe, and are recognized as common components of the ophidian diversity in several regions of South America. The species of Philodryadini exhibit two great geographical distribution patterns, with most species occurring in the lowlands of the cis-Andean region of the American continent, while another not so diverse group is distributed in the trans-Andean region of the central and southern Andes, in Ecuador, Peru and Chile. The richness of the tribe and its evolutionary relationships has varied greatly in recent years, mainly due to the recent formulation of diverse phylogenetic hypotheses based on molecular evidence. In the same way, in recent years many taxonomic complexes have been studied and the taxonomic status of several species has been clarified. However, many questions about the status of some complexes and phylogenetic relationships within the tribe are still unknown. To understand the evolutionary relationships between Philodryadini and the other Xenodontinae tribes we performed a phylogenetic analysis including molecular evidence of a representative sample of all tribes of the subfamily. Simultaneously, we evaluated the relationships within Philodryadini using DNA sequences from the vast majority of the species of the tribe described so far. Likewise, we performed a taxonomic revision of the tribe species, using a combination of morphological and molecular evidence. Our phylogenetic analyzes revealed that the tribe Philodryadini is a non-monophyletic group, and is currently composed of two different lineages of unrelated xenodontine snakes. To provide a phylogenetic structure that reflected the relations of the tribes in the interior of the subfamily, we erected a new tribe and a new genus to accommodate the group of species that constituted a completely different radiation of xenodontine snakes from the Andes. Within Philodryadini (sensu stricto), we recognize a particular pattern of diversification, with a first clade, composed of two groups , closely related to the clade that contains the type species of Philodryadini. To best represent the pattern of evolutionary diversification within the tribe, we restructured its generic composition by resurrecting the genera Chlorosoma and Xenoxybelis. Additionally, with our taxonomic revision we resolve the taxonomic status of three species complexes and recognize four taxa previously located in the synonymy of Philodryas. With our study, the relationships within Philodryadini are now better understood and their diversity is currently consisted of three genera and 24 species. A tribo Philodryadini é composta por um rico grupo de serpentes neotropicais altamente diversas ecológica e morfologicamente. Na atualidade, 24 espécies fazem parte da tribo, sendo amplamente reconhecidas como um dos componentes comuns da diversidade de ofídio-fauna de América do Sul. As espécies que fazem parte de Philodryadini apresentam dois grandes padrões de distribuição geográfica, sendo que a grande maioria das espécies ocorrem nas terras baixas da região cis-Andina do continente americano, enquanto que um outro grupo não tão diverso distribui-se na região trans-Andina dos Andes centrais e do Sul, no Equador, Peru e Chile. O conhecimento da diversidade da tribo e das suas relações evolutivas tem variado muito nos últimos anos, principalmente pela recente formulação de diversas hipóteses filogenéticas baseadas em evidência de biologia molecular. Do mesmo jeito, muitos complexos taxonômicos têm sido abordados recentemente e o status taxonômico de várias espécies esclarecido. No entanto, ainda se desconhecem muitas questões sobre o status de alguns complexos e as relações filogenéticas do interior da tribo. Para entender as relações evolutivas entre Philodryadini e as demais tribos de Xenodontinae realizamos uma análise filogenética incluindo evidência molecular de uma amostra representativa de todas as tribos da subfamília. Simultaneamente, avaliamos as relações ao interior de Philodryadini empregando sequências de ADN da grande maioria das espécies da tribo descritas até o momento. De igual forma, realizamos uma revisão taxonômica das espécies da tribo, empregando uma combinação de variáveis morfológicas e moleculares. As nossas análises filogenéticas mostraram que a tribo Philodryadini é um grupo não monofilético, estando na atualidade composto por duas linhagens diferentes de serpentes xenodontineas não relacionadas. Para fornecer uma estrutura filogenética que refletisse as relações das tribos no interior da subfamília, erigimos uma nova tribo e um gênero novo para acomodar o grupo de espécies que constituem uma radiação completamente diferente de serpentes xenodontineas dos Andes. Já no interior de Philodryadini (sensu stricto), reconhecemos um padrão de diversificação particular, com um primeiro clado, composto por dois grupos (as cobras cipó e as cobras de focinho afiado da Amazônia), estreitamente relacionado com o clado que contem a espécie tipo de Philodryadini. Pra melhor representar o padrão de diversificação evolutivo no interior da tribo, reestruturamos a sua composição genérica ao ressuscitar os gêneros Chlorosoma e Xenoxybelis. Adicionalmente, com a nossa revisão taxonômica reconhecemos o status taxonômico de três complexos de espécies e reconhecemos a validade de quatro táxons previamente localizadas na sinonímia de Philodryas. Com o nosso estudo, as relações no interior da tribo Philodryadini ficaram melhor resolvidas e a sua diversidade ficou constituída por três gêneros e 24 espécies.

Hylodidae is composed of 46 species distributed in three genera: Crossodactylus (14 spp), Hylodes (25 spp), and Megaelosia (7 spp). These torrent-frogs are diurnal and associated with riverine habitats throughout the Atlantic Rain Forest. The high degree of habitat specificity observed in this group seems to be associated with an extremely conservative external morphology; however, other sources of evidence have been proved useful to distinguish species. My study was designed to review the status of the current hylodid systematics, performing a total evidence analysis that represents as many species (and populations) as possible; confirm the monophyly of currently valid nominal taxa; investigate the evolutionary history of some morphological characters; and reconstruct biogeographical changes in the distribution of hylodids. My total evidence analysis included morphological (293 characters) and molecular data (four mitochondrial and five nuclear genes) for up to 371 hylodid terminals plus 45 outgroups, and resulted in 713 most parsimonious trees. I recovered Hylodidae and its compounding genera as monophyletic, and recognized 58 lineages within this family. Three synapomorphies were identified for Hylodidae based on vocal sac morphology (external double vocal sac, internal double vocal sac, and opening of the m. interhyoideus through which the diverticulum passes). Analysis of the distribution of hylodids revealed the origin of this frog family in the East Atlantic river basin, with subsequent allopatric speciation in adjacent basins Hylodidae é composta por 46 espécies distribuídas em três gêneros: Crossodactylus (14 spp), Hylodes (25 spp) e Megaelosia (7 spp). Essas rãs-de-corredeiras são diurnas e associadas à ambientes encachoeiradas ao longo da Mata Atlântica. O alto nível de especificacidade à esses habitats observado nesse grupo parece estar relacionado com a morfologia externa extremamente conservada; entretanto, outras fontes de evidências tem provado ser úteis para distinção de espécies. Meu estudo foi designado para revisar o status atual da sistemática dos hilodídeos através de uma análise de evidência total que representa o máximo de espécies (e populações) possíveis; confirmar o monofiletismo dos taxa válidos atualmente; avaliar a história evolutiva de alguns caracteres; e reconstruir mudanças biogeográficas na distribuição de Hylodidae. Minha análise de evidência total incluiu dados morfológicos (293 caracteres) e moleculares (quatro genes mitocondriais e cinco genes nucleares) para 34371 terminais de hilodídeos mais 45 terminais de grupo externo, resultando em 713 árvores mais parcimoniosas com 32.405 passos. Eu recuperei Hylodidae e todos os seus gêneros como monofiléticos e reconheci 59 linhagens dentro da família. Três sinapomorfias putativas foram identificadas para Hylodidae baseadas na morfologia do saco vocal (saco vocal externo duplo, saco vocal interno duplo e abertura no m. interhyoideus por onde o divertículo passa). Análises da distribuição dos hilodídeos revelaram a origem de Hylodidae na bacia Atlântico Leste com subsequentes especiações alopátricas em bacias subjacentes

A Amazônia abriga uma grande parcela da biodiversidade mundial, no entanto, existem grandes incertezas sobre os processos de geração e manutenção dessa enorme diversidade, com várias hipóteses propostas até o momento. Diversas características como tamanho pequeno, baixa capacidade de dispersão e fidelidade ao ambiente estritamente florestal fazem de lagartos do gênero Loxopholis (Squamata: Gymnophthalmidae) candidatos ideais para rastrear a história de modificações da paisagem no norte da América do Sul durante o Neogeno. Nesta tese, revisamos a variação morfológica, taxonomia e a distribuição geográfica das espécies do gênero Loxopholis, assim como estimamos as relações filogenéticas e o tempo de divergência entre as espécies com base em sequências de DNA provenientes do genoma mitocondrial e nuclear. Com essa base de dados, construímos um modelo de como ocorreu a história evolutiva de Loxopholis, discutindo os resultados a partir de reconstruções geológicas provenientes da literatura, e fornecendo uma datação independente para eventos geológicos que potencialmente influenciaram a história evolutiva do gênero. Neste estudo, também revelamos que a atual diversidade descrita para Loxopholis está severamente subestimada. Encontramos que a espécie L. osvaldoi representa um complexo de espécies crípticas, contendo de 8 a 14 potenciais espécies novas, com profundas divergências genéticas tanto nos genomas mitocondrial e nuclear, praticamente dobrando a diversidade conhecida no gênero. No entanto, a morfologia em Loxopholis é extremamente conservada, dificultando a diagnose das espécies novas. Finalmente, descrevemos a primeira população bissexual da espécie partenogenética L. percarinatum, discutindo as implicações deste achado para o entendimento relativo a aparição da partenogênese em Loxopholis The Amazon rainforest houses a significant portion of the world biodiversity, however, there is still a lot of uncertainty about the processes involved in its generation and maintenance, with several hypotheses proposed so far. Loxopholis lizards (Squamata: Gymnophthalmidae) are small-sized; show low vagility, and high fidelity to the humid forest environment, making them ideal to test hypotheses on the history of landscape modifications in Northern South America, during the Neogene. In this thesis, we revise the morphological variation, the taxonomy, and the geographic distribution of Loxopholis species, as well as estimated a phylogenetic hypothesis and divergence times for these species based on sequences from mitochondrial and nuclear DNA. With this dataset, we build a model for the evolutionary history of Loxopholis and compared the results with geologic reconstructions of the Amazon available in the literature, which provided independent time estimates for geological events that potentially affected the evolutionary history of genus. In this study, we also showed that the actual diversity described for Loxopholis was severely underestimated. We found that L. osvaldoi represents a complex of cryptic species with 8-14 potential new species, and strong divergences in both the mitochondrial and the nuclear genomes, virtually doubling the actual diversity in the genus. Finally, we describe the first bisexual population for the parthenogenetic species L. percarinatum, discussing its implications to the origin of parthenogenesis in the genus

Visando contribuir para a compreensão dos processos que originaram e mantiveram a biodiversidade na Mata Atlântica (MA), o presente estudo teve como objetivo inferir a história evolutiva - bem como os processos biogeográficos envolvidos - de duas espécies de aves da MA, P. atra (120 indivíduols) e P. leucoptera (434 indivíduos). Realizamos uma análise filogeográfica envolvendo marcadores com diferentes tipos de herança: materna (gene mitocondrial da subunidade II da NADH desidrogenase), biparental (três marcadores anônimos VIDY, GK439 e 55J7) e ligada ao cromossomo Z (íntron 18 da helicase com cromo-domínio de ligação ao DNA, íntron 15 da helicase Brahma dependente de ATP e íntron 1 da fosfolipase A2). Duas questões principais foram abordadas: (i) Qual hipótese de diversificação é mais congruente com os resultados do presente estudo? (ii) Os resultados baseados em marcadores com diferentes tipos de herança são congruentes? Para responder a primeira questão, algumas das previsões de cada hipótese foram testadas. Foi encontrada evidência de que as mudanças climáticas no final do Pleistoceno parecem ter sido importantes na estrutura filogeográfica de Pyriglena atra e Pyriglena leucoptera na MA. Os resultados encontrados foram congruentes com a Hipótese dos Isolados de Montanha, mas aparentemente refutam outras hipóteses, tais como: a Hipótese dos Refúgios do Pleistoceno no contexto do modelo paleoclimático proposto por Carnaval & Moritz (2008), Hipótese de estruturas fisiográficas (rios ou montanhas) como barreiras, e a Hipótese de Gradientes Ecológicos. Além disso, os rios Doce e Contas parecem ser barreiras secundárias interrompendo parcialmente o gênico. Finalmente, os marcadores revelaram diferentes níveis de estrutura geográfica e sinais nem sempre totalmente congruentes. Assim, embora os marcadores mitocondriais sejam muito informativos em reconstruções filogeográficas, é importante usar vários tipos de marcadores para recuperar de forma mais completa a história dos processos evolutivos dos organismos e os possíveis fatores que geraram essa biodiversidade To contribute to the understanding of the processes that originated and maintained biodiversity in the Atlantic Forest (AF), the present study aimed to infer the evolutionary history - as well as the biogeographic processes involved - of two AF bird species, P. atra (120 individuals) and P. leucoptera (434 individuals). We performed a phylogeographic analysis based on markers with different types of inheritance: maternal (mitochondrial gene of the NADH dehydrogenase subunit II), biparental (three annonymous markers: VIDY, GK439, 55J7) and Z-linked (intron 18 of the chromo-DNA-binding domain helicase, intron 15 of the ATP-dependent Brahma helicase and intron 1 of the Phospholipase A2). Two major questions were addressed: (i) Which diversification hypothesis best fits the results of the present study? (ii) Are the results based on markers with different types of inheritance congruent? In order to answer the first question, some of the predictions of each hypothesis were tested. We found evidence that climate changes in the late Pleistocene seem to have been important for shaping the phylogeographic structure of Pyriglena atra and Pyriglena leucoptera in the AF. The results are congruent with the Montane Isolation Hypothesis but apparently refute other hypotheses, such as: The Pleistocene Refuge Hypothesis in the context of the paleoclimatic model proposed by Carnaval & Moritz (2008), Hypothesis of physiographic structures (i.e. rivers or mountains) as barriers, and the Ecological Gradients Hypothesis. Additionaly, the Doce and Contas rivers seem to act as secondary barriers partially disrupting gene flow. Finally, the markers revealed varying levels of geographical structure that were not totally congruent. Thus, although mitochondrial markers are very informative for phylogeographic reconstructions, it is important to use several types of markers in order to recover the history of the evolutionary processes of organisms and the possible factors that generate this biodiversity

A delimitação de espécies é essencial para a caracterização e conservação da biodiversidade. No entanto, representa um desafio para grupos onde há pouca variação em morfologia, como no caso dos lagartos Iphisini. São conhecidas oito espécies pertencentes a seis gêneros na tribo, com base em análises moleculares e de anatomia hemipeniana foram detectadas quatro espécies candidatas para Iphisa. A tribo filogeneticamente aparentada Gymnophthalmini apresenta maior riqueza de espécies e diversidade morfológica, principalmente em espécies com adaptações para a vida fossorial. No entanto, ainda se conhece pouco sobre os mecanismos históricos e ecológicos que causam distintos padrões de especiação, apesar de historicamente o papel de isolamento geográfico ter sido enfatizado para a biota Neotropical. Em tempos recentes, foram desenvolvidos métodos quantitativos para abordar questões evolutivas como probabilidades de especiação, variações em taxas de diversificação e reconstrução de demografia histórica de populações e migração. Implementei neste estudo uma combinação de métodos quantitativos com uso de dados moleculares, morfológicos e ambientais para testar as hipóteses que: há diversidade não reconhecida em Iphisini; as diferenças em riqueza e disparidade morfológica entre Iphisini e outras tribos de Gymnophthalminae se deve a diferenças em tempo e modo de diversificação, e que a diversificação em Acratosaura ocorreu por isolamento geográfico causado por flutuações paleoclimáticas. Com base em análises moleculares, foram delimitadas quatro espécies candidatas para Iphisini, aumentando em 33% a diversidade conhecida para a tribo. Não houve variação entre as espécies candidatas de Acratosaura em morfometria. A filogenia estimada para Gymnophthalminae apresentou alto suporte para a relação entre Iphisini e Heterodactylini, e demonstra um tempo de diversificação e riqueza neste clado similar a Gymnophthalmini. As tribos apresentaram padrões de diversificação semelhantes, mas taxas distintas. Os padrões de evolução morfológica foram congruentes com a diversificação em Gymnophthalmini, porém distintas em Heterodactylini sensu lato, indicando que disparidade independe de diversificação. As análises filogeográficas indicam que as populações de Acratosaura apresentaram estabilidade demográfica e espacial ao longo do tempo, com evidências de fluxo gênico entre linhagens diferenciadas. Desta forma, os resultados sugerem que Acratosaura diversificou sem influência de variações históricas no clima, e provavelmente sem isolamento reprodutivo completo Species delimitation is essential for characterization and conservation of biodiversity. Nevertheless, it represents a challenge for groups in which morphological variation is subtle, such as the microteiid lizards of the tribe Iphisini. Eight species from six genera are currently recognized in the tribe but recently, based on molecular analysis and hemipenial anatomy, four candidate species were inferred for Iphisa. The phylogenetically related tribe Gymnophthalmini presents higher species richness and morphological diversity, specially in forms with adaptations to fossoriality. Nevertheless, the historical and ecological mechanisms involved in the distinct speciation patterns are poorly known, although geographical isolation have been historically emphasized for neotropical biota. In recent times, quantitative methods were developed to address evolutionary questions such as speciation probabilities, variation in diversification rates and reconstruction of historical demography of populations and migration. In this study I used a combination of quantitative methods based on molecular, morphological and environmental data for testing the hypothesis that: there is unrecognized diversity within Iphisini; differences in species richness and disparity among Iphisini and related tribes are congruent with differences in time and mode of diversification; and that diversification in Acratosaura occurred with geographical isolation caused by paleoclimatic fluctuations. Based on the results of molecular analyses, four candidate species were delimited for Iphisini, rising in 33% the tribe diversity. There was no significant variation in morphometry between candidate species of Acratosaura. The phylogeny of Gymnophthalminae presented high support for the relationship between Iphisini and Heterodactylini, and showed diversification timming and species richness comparable between this clade and Gymnophthalmini. The tribes presented similar diversification patterns but distinct rates. The patterns of morphological evolution were congruent with diversification patterns in Gymnophthalmini but distinct in Heterodactylini sensu lato, indicating that disparity is independent from diversification in the group. The phylogeographic analyses indicate that populations of Acratosaura presented demographic and spatial stability through time, with evidences of gene flow among lineages after differentiation. Thus, the results suggest that diversification of Acratosaura was not influenced by variations in historical climate, and probably occurred without complete reproductive isolation

The genus Pristimantis has 500 species of anurans with distribution in northwest South America, specially in the Andes. Due to the large number of species morphologically diverse, the genus was divided in 11 species groups, with 312 species not allocated in any of them. The great number of species not allocated in species groups is due to the lack of morphological characters to better define and diagnose these groups, and because most of the anteriorly recognized groups in the literature of 70, 80 and 90s decades of the last century were not recovered monophyletic in the most recent phylogenetic analyses. The present study aims to perform a phylogenetic analysis of Pristimantis, with phenotypic and genotypic characters, through total evidence with dynamic homology and parsimony. As a result, Pristimantis is not monophyletic and some of its species were reallocated in the genus Tachiramantis. The species groups within Pristimantis were reformulated with proposed diagnosis. O grande número de espécies não alocadas em grupos se deve especialmente pela falta de dados morfológicos para definir e diagnosticar esses grupos, e pelo fato dos grupos anteriormente reconhecidos nas décadas de 70, 80 e 90, serem em sua grande maioria não monofiléticos. O presente trabalho tem como objetivo uma análise filogenética do gênero Pristimantis, utilizando caracteres fenotípicos e genotípicos através de uma análise de evidência total utilizando homologia dinâmina e parcimônia. Pristimantis não resultou monofilético e algumas de suas espécies foram realocadas no gênero Tachiramantis. Os grupos de espécies em Pristimantis foram reformulados com diagnoses propostas. No total 25 grupos de espécies são reconhecidos

Bothrops bilineatus é uma das seis espécies presentes no grupo taeniatus, é reconhecida atualmente por duas sub-espécies: B. b. smaragdinus que se distribui pelo oeste Amazônico e B. b. bilineatus com distribuição disjunta pela Amazônia e Mata Atlântica. Este trabalho tem como objetivo testar a validade das duas sub-espécies com base em ferramentas moleculares, morfométricas, merísticas, hemipenianas e osteológicas, reconhecendo e delimitando as linhagens encontradas. Pretende-se ainda investigar as relações filogeográficas entre as populações ao longo da sua distribuição, analisando o tempo de divergência e os possíveis eventos associados a sua diversificação. Além disso, pretende-se comparar a variação molecular e hemipeniana de B. bilineatus com B. taeniatus (espécie filogeneticamente próxima) e as existentes nos outros grupos de espécies do gênero. Os resultados mostram que a variação genética e hemipeniana encontram se em diferentes níveis ao longo dos grupos de espécies em Bothrops, fornecendo informação relevante a nível intra e inter-específico. Foram recuperados seis clados estruturados para Bothrops bilineatus, representando quatro linhagens independentes e diagnosticáveis com base conjunta nos caracteres morfológicos, hemipenianos, osteológicos e moleculares: linhagem 1 - Amazônia central, basal as outras, linhagem 2 - Mata Atlântica+Escudo das Guianas externa às linhagens 3 e 4 Amazônia oeste parte sul e norte, respectivamente. Já para B. taeniatus quatro clados representando três linhagens foram recuperadas com bases nos caracteres moleculares e hemipenianos, porém se faz necessário buscar por mais caracteres morfológicos e osteológicos para melhor delimitar e diagnosticar as linhagens. As mudanças paleoclimáticas e geomorfológicas ocorridas na transição Terciário/Quaternário parecem ser os principais eventos a influenciar na diversificação das linhagens encontradas em Bothrops bilineatus e B. taeniatus Bothrops bilineatus is one of six species in the taeniatus group, is now recognized by two subspecies: B. b. smaragdinus that distributes through western Amazon and B. b. bilineatus with disjunct distribution through Amazon and Atlantic Forest. This work aims to test the validity of these two sub-species, based on molecular, morphometric tools, meristic, osteological and hemipenial data, recognizing and delimiting lineages found. We also intend to investigate the phylogeographic relationships among populations throughout its distribution, analyzing the divergence time and possible events associated with it diversification. In addition, we intend to compare the molecular and hemipenial variation presented in B. bilineatus with B. taeniatus (phylogenetically close species) and those in the other groups of the genus. The genetic and hemipenial results show different levels of variation along the groups of Bothrops species, providing significant intra-and inter-specific information. Six clades structured to Bothrops bilineatus were recovered, representing four independent lineages diagnosable with joint based on morphology, hemipenial, osteological and molecular characters: lineage 1 - central Amazon, basal the others, lineage 2 - Atlantic Forest + Guayana Shield region recovered outside the sisters lineages 3 and 4 - western Amazon, southern part and western Amazon, northern part, respectively. As for B. taeniatus four clades representing three lineages were recovered with bases in molecular and hemipenial characters, however it is necessary to search for more morphological and osteological characters to better delimit and diagnose the lineages. Paleoclimatic and geomorphic changes in the transition Tertiary/Quaternary, probably are the major events influencing the diversification of lineages found in Bothrops bilineatus and B. taeniatus

Estudos morfológicos constataram que a espécie Tropidurus torquatus apresenta dois morfotipos geograficamente distintos ao longo da área ocupada pela espécie. Um com preferência por ambientes saxícolas, situado mais para o interior do continente e, outro, mais costeiro, ocupando predominantemente os solos arenosos de dunas e restingas. Estes morfotipos possuem características morfológicas externas distintas associadas a estes ambientes. Porém, estas diferenças morfológicas observadas não foram determinantes para um estudo mais conclusivo dos padrões de diferenciação e da história zoogeográfica da espécie em questão. Dentro deste contexto teórico, este trabalho abordou a história das populações de T. torquatus com o uso de dados moleculares mostrando como a variabilidade genética da espécie esta distribuída geograficamente. Esta espécie se mostrou muito mais diversa do que se imaginava inicialmente. Quatro diferentes linhagens foram recuperadas na maioria das análises e apresentam relação com a geografia. Os resultados mostraram que esta espécie está se expandido e que possivelmente pode ocorrer uma diferenciação intraespecífica. Também mostrou que pode estar ocorrendo fluxo gênico não só dentro desta espécie, mas como entre as outras espécies do gênero, porém somente uma filogenia mais robusta poderá elucidar a relação entre elas Morphological studies have showed that Tropidurus torquatus presents two geographically distinct morphotypes along the area occupied by the species. The first morphotype is saxicolous and exhibits a preference for rocky environments located farther inland, and the other morphotype is located in more coastal areas, occupying predominantly sandy soils of dunes and sandbanks. These morphotypes have distinct external morphological traits associated with the environments they occupy. However, these differences were not observed morphological determinants for a more conclusive study of the differentiation patterns of differentiation and zoogeographical history of the species. Within this theoretical context, this work addressed the history of T. torquatus populations using molecular data, in order to understand the geographical distribution of genetic variability. Results showed that this species is far more diverse than initially hypothesized. Four different lineages were recovered in most analyzes, each one related to its intrinsic distributional pattern. Besides, this study shows that this species is expanding its distribution area and for this reason a differentiation among populations is likely to occur, potentially with formation of new species. Also results show that gene flow may be occurring not only within the species, but also between other Tropidurus species. Nonetheless, only a more robust phylogeny may elucidate the relationship between these species
