Abstract

Gut microbiota dysbiosis has been implicated in rheumatoid arthritis (RA) and influences disease progression. Although molecular and culture‐independent studies revealed RA patients harbored a core microbiome and had characteristic bacterial species, the lack of cultured bacterial strains had limited investigations on their functions. This study aimed to establish an RA‐originated gut microbial biobank (RAGMB) that covers and further to correlates and validates core microbial species on clinically used and diagnostic inflammation and immune indices. We obtained 3200 bacterial isolates from fecal samples of 20 RA patients with seven improved and 11 traditional bacterial cultivation methods. These isolates were phylogenetically identified and selected for RAGMB. The RAGMB harbored 601 bacterial strains that represented 280 species (including 43 novel species) of seven bacterial phyla. The RAGMB covered 93.2% at species level of medium‐ and high‐abundant (relative abundances ≥0.2%) RA gut microbes, and included four rare species of the phylum Synergistota. The RA core gut microbiome was defined and composed of 20 bacterial species. Among these, Mediterraneibacter tenuis and Eubacterium rectale were two species that statistically and significantly correlated with clinically used diagnostic indices such as erythrocyte sedimentation rate (ESR) and IL‐10. Thus, M. tenuis and E. rectale were selected for experimental validation using DSS‐treated and not DSS‐treated mice model. Results demonstrated both M. tenuis and E. rectale exacerbated host inflammatory responses, including shortened colon length and increased spleen weight, decreased IL‐10 and increased IL‐17A levels in plasma. Overall, we established the RAGMB, defined the RA core microbiome, correlated and demonstrated core microbial species effected on host inflammatory and immune responses. This work provides diverse gut microbial resources for future studies on RA etiology and potential new targets for new biomedical practices. 20. Study on the Pb(II) adsorption mechanism on acid‐modified ceramsite made from sewage sludge and industrial solid wastes.

Abstract

Ceramsite was made from sludge, phosphogypsum, and red mud and then modified with different concentrations of hydrochloric, nitric, or citric acid. The resulting composite ceramsite was subsequently utilized for the adsorption of Pb(II). The maximum adsorption capacities of hydrochloric acid– and nitric acid–modified ceramsites were 90.38 and 88.57 mg/g, respectively, which were much higher than those of unmodified ceramsite (66.86 mg/g) and citric acid–modified ceramsite (57.85 mg/g). The variation of isoelectric point, acidic and basic groups, and Fourier transform infrared spectra of ceramsite were compared before and after adsorption. A decrease of isoelectric point implied weakening of electrostatic repulsion between Pb(II) and the modified ceramsite. Functional groups, such as O─H, C═O (or C═C), and Si─O─Si (or Al─O─Al), were involved in the adsorption. In addition, phenol hydroxyl, which exhibited a significant increase in modified ceramsite and a noticeable decrease after adsorption, was the most important group. Besides, the modified ceramsite had larger specific surface areas and pores (30.66 g/m2,.11256 cm3/g), more functional groups, and lower isoelectric points (8.57), which were favorable for adsorption. The adsorption processes described by pseudo‐second‐order kinetics were controlled by chemical adsorptions. Isothermal studies indicated that monolayer adsorption occurred on hydrochloric acid–modified ceramsite and multiple processes occurred on nitric acid– and citric acid–modified ceramsites. Abstract

It is well known that direct observation of interference and diffraction pattern in the intensity distribution requires a spatially coherent source. Optical waves emitted from portions beyond the coherence area possess statistically independent phases, and will degrade the interference pattern. In this paper we show an optical interference experiment, which seems contrary to our common knowledge, that the formation of the interference pattern is related to a spatially incoherent light source. Our experimental scheme is very similar to Gabor's original proposal of holography[1], just with an incoherent source replacing the coherent one. In the statistical ensemble of the incoherent source, each sample field produces a sample interference pattern between object wave and reference wave. These patterns completely differ from each other due to the fluctuation of the source field distribution. Surprisingly, the sum of a great number of sample patterns exhibits explicitly an interference pattern, which contains all the information of the object and is equivalent to a hologram in the coherent light case. In this sense our approach would be valuable in holography and other interference techniques for the case where coherent source is unavailable, such as x-ray and electron sources., Comment: 8 pages, 5 figures

44. Nitrogen Forms Regulate the Response of Microcystis aeruginosato Nanoplastics at Environmentally Relevant Nitrogen Concentrations

Abstract

As essential primary producers, cyanobacteria play a major role in global carbon and nitrogen cycles. Though the influence of nanoplastics on the carbon metabolism of cyanobacteria is well-studied, little is known about how nanoplastics affect their nitrogen metabolism, especially under environmentally relevant nitrogen concentrations. Here, we show that nitrogen forms regulated growth inhibition, nitrogen consumption, and the synthesis and release of microcystin (MC) in Microcystis aeruginosaexposed to 10 µg/mL amino-modified polystyrene nanoplastics (PS-NH2) with a particle size of 50 nm under environmentally relevant nitrogen concentrations of nitrate, ammonium, and urea. We demonstrate that PS-NH2inhibit M. aeruginosadifferently in nitrate, urea, and ammonium, with inhibition rates of 51.87, 39.70, and 36.69%, respectively. It is caused through the differences in impairing cell membrane integrity, disrupting redox homeostasis, and varying nitrogen transport pathways under different nitrogen forms. M. aeruginosarespond to exposure of PS-NH2by utilizing additional nitrogen to boost the production of amino acids, thereby enhancing the synthesis of MC, extracellular polymeric substances, and membrane phospholipids. Our results found that the threat of nanoplastics on primary producers can be regulated by the nitrogen forms in freshwater ecosystems, contributing to a better understanding of nanoplastic risks under environmentally relevant conditions.
