Abstract The mechanisms underlying myopia remain not fully understood. We proposed to examine the function and underlying mechanisms of miR-204-5p in myopia development. The miR-204-5p expression level was assessed in the vitreous humor (VH) of a cohort consisting of 11 patients with high myopia (HM) and 16 control patients undergoing vitrectomy. Then the functional implications of miR-204-5p in ARPE-19 cells were assessed. Thioredoxin-interacting protein (TXNIP) was found as a possible target of miR-204-5p through mRNA sequencing, and its interaction with miR-204-5p was confirmed employing luciferase assay and western blotting. Furthermore, the miR-204-5p function in regulating oxidative stress was examined by measuring reactive oxygen species (ROS) accumulation. The results indicated a significant reduction of miR-204-5p in the VH of HM patients. Overexpression of miR-204-5p suppressed cell proliferation, migration, invasion, and apoptosis in ARPE-19 cells. The direct targeting of miR-204-5p on TXNIP has been confirmed, and its downregulation mediated the miR-204-5p impacts on ARPE-19 cells. Moreover, miR-204-5p overexpression significantly reduced ROS accumulation by targeting TXNIP. Our findings revealed the possible contribution of the miR-204-5p/TXNIP axis in myopia development by regulating oxidative stress, which may provide new targets to combat this prevalent and debilitating condition.

Abstract Background Despite being a prognostic predictor, cardiac autonomic dysfunction (AD) has not been well investigated in chronic obstructive pulmonary disease (COPD). We aimed to characterise computed tomography (CT), spirometry, and cardiopulmonary exercise test (CPET) features of COPD patients with cardiac AD and the association of AD with CT-derived vascular and CPET-derived ventilatory efficiency metrics. Methods This observational cohort study included stable, non-severe COPD patients. They underwent clinical evaluation, spirometry, CPET, and CT. Cardiac AD was determined based on abnormal heart rate responses to exercise, including chronotropic incompetence (CI) or delayed heart rate recovery (HRR) during CPET. Results We included 49 patients with FEV1 of 1.2–5.0 L (51.1–129.7%), 24 (49%) had CI, and 15 (31%) had delayed HRR. According to multivariate analyses, CI was independently related to reduced vascular volume (VV; VV ≤ median; OR [95% CI], 7.26 [1.56–33.91]) and low ventilatory efficiency (nadir VE/VCO2 ≥ median; OR [95% CI], 10.67 [2.23–51.05]). Similar results were observed for delayed HRR (VV ≤ median; OR [95% CI], 11.46 [2.03–64.89], nadir VE/VCO2 ≥ median; OR [95% CI], 6.36 [1.18–34.42]). Conclusions Cardiac AD is associated with impaired pulmonary vascular volume and ventilatory efficiency. This suggests that lung blood perfusion abnormalities may occur in these patients. Further confirmation is required in a large population-based cohort.

Porous carbonate reservoirs are characterized by strong heterogeneity and diverse pore types， with significant differences in waterflooding recovery and unclear main controlling factors. Based on the pore throat size distribution curves， the microscopic pore structure of the reservoir was qualitatively classified. Through digital core technology， a method for quantitative characterization of microscopic pore structure was established to clarify the pore throat characteristics of porous carbonate rocks in a region of the Middle Eastern， and indoor physical experiments on waterflooding were conducted to clarify the differences in the oil recovery of different types of cores and their main controlling factors. The results show that ① The microscopic pore structure of the reservoir in the study area can be classified into coarse unimodal type， fine unimodal type I， fine unimodal type II， coarse bimodal type， and multimodal type. There is no significant relationship between storage performance and core homogeneity； the single-phase flow capacity of non-unimodal type is better than that of unimodal type； unimodal type becomes worse with increasing core homogeneity. ②The average coordination number and pore-throat ratio of the non-unimodal type are higher than those of the unimodal type. The average coordination number and pore-throat ratio decreased with the increase of core homogeneity for the unimodal type. The upper limit of the pore-throat ratio tends to decrease with increasing throat radius for all types， and the upper limit of coordination number increases exponentially with increasing pore radius. ③Macroscopically， core homogeneity is the main controlling factor of oil displacement efficiency； microscopically， high connectivity at large pore-throat and large pore-throat ratio at small pore-throat are the fundamental reasons for non-unimodal type to have significantly lower oil displacement efficiency than unimodal type. Pore-throat size is the crucial factor of oil displacement efficiency of unimodal type.

ABSTRACT This study aims to explore the link between retinal vein occlusion (RVO), a blinding ocular condition, and alterations in gut microbiota composition, to offer insights into the pathogenesis of RVO. Fecal samples from 25 RVO patients and 11 non-RVO individuals were analyzed using 16S rRNA sequencing and liquid chromatography-mass spectrometry (LC-MS). Significant differences in the abundance of gut microbial species were noted between RVO and non-RVO groups. At the phylum level, the RVO group showed an elevation in the ratio of Firmicutes to Bacteroidetes. At the genus level, the RVO group showed higher abundance in Escherichia_Shigella (P < 0.05) and less abundance in Parabacteroides (P < 0.01) than the non-RVO group. Functional predictions indicated reduced folate synthesis, biotin metabolism, and oxidative phosphorylation, with an increase in butyric acid metabolism in the RVO group. LC-MS analysis showed significant differences in purine metabolism, ABC transporters, and naphthalene degradation pathways, especially purine metabolism. Pearson correlation analysis revealed significant associations between bacterial genera and fecal metabolites. Enrichment analysis highlighted connections between specific metabolites and bacterial genera. The findings showed that the dysregulation of gut microbiota was observed in RVO patients, suggesting the gut microbiota as a potential therapeutic target. Modulating the gut microbiota could be a novel strategy for managing RVO and improving patient outcomes. Furthermore, the study findings suggest the involvement of gut microbial dysbiosis in RVO development, underscoring the significance of understanding its pathogenesis for effective treatment development.IMPORTANCERetinal vein occlusion (RVO) is a blinding ocular condition, and understanding its pathogenesis is crucial for developing effective treatments. This study demonstrates significant differences in gut microbiota composition between RVO patients and non-RVO individuals, implicating the involvement of gut microbial dysbiosis in RVO development. Summary: N(6)-methyladenosine (m6A) critically regulates RNA dynamics in various biological processes. The m6A demethylase ALKBH5 promotes tumorigenesis of glioblastoma, while the intricate web that orchestrates its regulation remains enigmatic. Here, we discover that cell density affects ALKBH5 subcellular localization and m6A dynamics. Mechanistically, ALKBH5 is phosphorylated by the large tumor suppressor kinase 2 (LATS2), preventing its nuclear export and enhancing protein stability. Furthermore, phosphorylated ALKBH5 reciprocally erases m6A from LATS2 mRNA, thereby stabilizing this transcript. Unexpectedly, LATS2 depletion suppresses glioblastoma stem cell self-renewal independent of yes-associated protein activation. Additionally, deficiency in either LATS2 or ALKBH5 phosphorylation impedes tumor progression in mouse xenograft models. Moreover, high levels of LATS2 expression and ALKBH5 phosphorylation are associated with tumor malignancy in patients with gliomas. Collectively, our study unveils an oncogenic positive feedback loop between LATS2 and ALKBH5, revealing a non-canonical branch of the Hippo pathway for RNA processing and suggesting potential anti-cancer interventions.

Abstract Cost‐effective non‐noble metal‐based catalysts for selective hydrogenation with excellent activity, selectivity, and durability are still the holy grail. Herein, an oxygen‐doped carbon (OC) chainmail encapsulated dilute Cu–Ni alloy is developed by simple pyrolysis of Cu/Ni‐metal–organic framework. The CuNi0.05@OC catalyst displays superior performance for atmospheric pressure transfer hydrogenation of p‐chloronitrobenzene and p‐nitrophenol, and for hydrogenation of furfural, all in water and with exceptional durability. Comprehensive characterizations confirm the close interactions between the diluted Ni sites, the base Cu, and optimized three‐layered graphene chainmail. Theoretical calculations demonstrate that the properly tuned lattice strain and Schottky junction can adjust electron density to facilitate specific adsorption on the active centers, thus enhancing the catalytic activity and selectivity, while the OC shell also offers robust protection. This work provides a simple and environmentally friendly strategy for developing practical heterogeneous catalysts that bring the synergistic effect into play between dilute alloy and functional carbon wrapping.

Abstract Multiple linear stapler firings is a risk factor for anastomotic leakage (AL) in laparoscopic low anterior resection (LAR) using double stapling technique (DST) anastomosis. In this study, our objective was to establish the risk factors for ≥ 3 linear stapler firings, and to create and validate a predictive model for ≥ 3 linear stapler firings in laparoscopic LAR using DST anastomosis. We retrospectively enrolled 328 mid–low rectal cancer patients undergoing laparoscopic LAR using DST anastomosis. With a split ratio of 4:1, patients were randomly divided into 2 sets: the training set (n = 260) and the testing set (n = 68). A clinical predictive model of ≥ 3 linear stapler firings was constructed by binary logistic regression. Based on three-dimensional convolutional networks, we built an image model using only magnetic resonance (MR) images segmented by Mask region-based convolutional neural network, and an integrated model based on both MR images and clinical variables. Area under the curve (AUC), sensitivity, specificity, accuracy, positive predictive value (PPV), and Youden index were calculated for each model. And the three models were validated by an independent cohort of 128 patients. There were 17.7% (58/328) patients received ≥ 3 linear stapler firings. Tumor size ≥ 5 cm (odds ratio (OR) = 2.54, 95% confidence interval (CI) = 1.15–5.60, p = 0.021) and preoperative carcinoma embryonic antigen (CEA) level > 5 ng/mL [OR = 2.20, 95% CI = 1.20–4.04, p = 0.011] were independent risk factors associated with ≥ 3 linear stapler firings. The integrated model (AUC = 0.88, accuracy = 94.1%) performed better on predicting ≥ 3 linear stapler firings than the clinical model (AUC = 0.72, accuracy = 86.7%) and the image model (AUC = 0.81, accuracy = 91.2%). Similarly, in the validation set, the integrated model (AUC = 0.84, accuracy = 93.8%) performed better than the clinical model (AUC = 0.65, accuracy = 65.6%) and the image model (AUC = 0.75, accuracy = 92.1%). Our deep-learning model based on pelvic MR can help predict the high-risk population with ≥ 3 linear stapler firings in laparoscopic LAR using DST anastomosis. This model might assist in determining preoperatively the anastomotic technique for mid–low rectal cancer patients.

The status monitoring system of train bogie is key to the overall health monitoring of a train. Typically dozens of sensors, like axle temperature monitoring, instability detection, vibration impact monitoring, etc., are deployed on each bogie, making the overall bogie wiring complex and maintenance challenging. In this paper, the ZigBee wireless communication technology and a cluster-based networking approach are employed to form a wireless sensor network for each bogie system. This significantly reduces the deployment time and cost and makes operation and maintenance more convenient. By utilizing network simulation tool NS2, a train bogie wireless sensor network model based on the IEEE 802.15.4 standard and the Ad Hoc on-demand distance vector (AODV) routing protocol was built. To mitigate packet loss rate and decrease end-to-end communication delay, modifications to the AODV protocol were undertaken based on the route timeout mechanism, the lifecycle of route request (RREQ), and the direction of RREQ propagation. Simulation results illustrate that, following the improvements, the AODV protocol reduces by 32% the time to reestablish communication after interruption. Simultaneously, packet loss rate and end-to-end communication delay are both reduced by over 50%. These results demonstrate that the wireless sensor network design proposed in this paper can satisfy the data transmission requirements of train bogie sensors.

Fibrotic diseases, such as idiopathic pulmonary fibrosis (IPF) and systemic scleroderma (SSc), are commonly associated with high morbidity and mortality, thereby representing a significant unmet medical need. Interleukin 11 (IL11)-mediated cell activation has been identified as a central mechanism for promoting fibrosis downstream of TGFβ. IL11 signaling has recently been reported to promote fibroblast-to-myofibroblast transition, thus leading to various pro-fibrotic phenotypic changes. We confirmed increased mRNA expression of IL11 and IL11Rα in fibrotic diseases by OMICs approaches and in situ hybridization. However, the vital role of IL11 as a driver for fibrosis was not recapitulated. While induction of IL11 secretion was observed downstream of TGFβ signaling in human lung fibroblasts and epithelial cells, the cellular responses induced by IL11 was quantitatively and qualitatively inferior to that of TGFβ at the transcriptional and translational levels. IL11 blocking antibodies inhibited IL11Rα-proximal STAT3 activation but failed to block TGFβ-induced profibrotic signals. In summary, our results challenge the concept of IL11 blockade as a strategy for providing transformative treatment for fibrosis.

Under accelerated domestic economic cycle, it is significant to predict the embodied carbon transfer network (ECTNs) to identify key emission regions to improve emission reduction efficiency. Based on the existing China Multi-regional Input-Output Table (CMRIOs) for 2002, 2007, 2010, 2012, 2015 and 2017, the CMRIOs for 2002-2017 were updated, then the ECTNs were predicted and constructed from 2018 to 2025 through Particle Swarm Optimization- Support Vector Model. Finally, the spatial and temporal evolution trends of the ECTNs’ features were explored through complex networks analysis. The results showed that carbon leakage between provinces has been becoming increasingly serious. The small-world features of the ECTNs were becoming increasingly obvious. The distribution of provinces with great influence on carbon transfer was transferred from north to south, and then to the central region. Hebei, Jiangsu, Henan, Zhejiang, Inner Mongolia, and other resource-intensive and manufacturing provinces played an important "bridge" role in the trade between economic developed and developing provinces. Trade ties between non neighboring provinces have become increasingly close, which means the development of China’s integration has strengthened. This study provides a theoretical reference for the formulation of China’s overall carbon emission reduction policy.

Alternaria mycotoxins are the secondary metabolites of Alternaria species, which have obvious toxicity （mutagenicity, teratogenicity and carcinogenicity, etc） to humans and animals. Alternariol （AOH）, alternariol monomethyl ether （AME）, alternuene （ALT）, tenuazonic acid （TeA） and tenoxin （TEN） are the most studied at present. Alternaria mycotoxins was reported in apples, orange, tomatoes, corns and other agricultural products. Eating these foods contaminated by Alternaria mycotoxins will cause serious harm to human health. In this paper, the pollution status and the detection technology of Alternaria mycotoxins were summarized, and the future research direction of Alternaria mycotoxins was prospected. The purpose was to provide references for the risk assessment, detection and prevention of Alternaria mycotoxins in foods.

The massive amount of human biological, imaging, and clinical data produced by multiple and diverse sources necessitates integrative modeling approaches able to summarize all this information into answers to specific clinical questions. In this paper, we present a hypermodeling scheme able to combine models of diverse cancer aspects regardless of their underlying method or scale. Describing tissue-scale cancer cell proliferation, biomechanical tumor growth, nutrient transport, genomic-scale aberrant cancer cell metabolism, and cell-signaling pathways that regulate the cellular response to therapy, the hypermodel integrates mutation, miRNA expression, imaging, and clinical data. The constituting hypomodels, as well as their orchestration and links, are described. Two specific cancer types, Wilms tumor (nephroblastoma) and non-small cell lung cancer, are addressed as proof-of-concept study cases. Personalized simulations of the actual anatomy of a patient have been conducted. The hypermodel has also been applied to predict tumor control after radiotherapy and the relationship between tumor proliferative activity and response to neoadjuvant chemotherapy. Our innovative hypermodel holds promise as a digital twin-based clinical decision support system and as the core of future in silico trial platforms, although additional retrospective adaptation and validation are necessary.

Abstract Chinese formulation‐based medicinal food has been widely used in clinical trials, but its safety is not well studied. In this research, the edible safety assessment of Balanced Health Care Dan—a formulation containing traditional edible ingredients that were initially formulated to reduce side effects for lung cancer patients—was studied in mice based on biochemical and gut microbial analyses. The experimental mice were subcutaneously loaded with lung tumor A549 cells and then administrated with Balanced Health Care Dan (200 mg/kg or 400 mg/kg b.w. in gavage feeding) for 4 weeks. The body weight, blood parameters, and pathogenic phenotype in tissues were examined. No toxicological symptom was found in experimental mice compared with the normal control. Comprehensive analyses were also conducted to evaluate intestinal microbiota that are associated with many diseases. Balanced Health Care Dan modified the gut microbiota structure in a positive way. In conclusion, the Chinese formulation‐based medicinal food has shown no toxicological effect in mice within 4 weeks of feeding experiment and has the potential to be used in clinical trials.

Abstract Background Individual organisms may exhibit phenotypic plasticity when they acclimate to different conditions. Such plastic responses may facilitate or constrain the adaptation of their descendant populations to new environments, complicating their evolutionary trajectories beyond the genetic blueprint. Intriguingly, phenotypic plasticity itself can evolve in terms of its direction and magnitude during adaptation. However, we know little about what determines the evolution of phenotypic plasticity, including gene expression plasticity. Recent laboratory-based studies suggest dominance of reversing gene expression plasticity—plastic responses that move the levels of gene expression away from the new optima. Nevertheless, evidence from natural populations is still limited. Results Here, we studied gene expression plasticity and its evolution in the montane and lowland populations of an elevationally widespread songbird—the Rufous-capped Babbler (Cyanoderma ruficeps)—with reciprocal transplant experiments and transcriptomic analyses; we set common gardens at altitudes close to these populations’ native ranges. We confirmed the prevalence of reversing plasticity in genes associated with altitudinal adaptation. Interestingly, we found a positive relationship between magnitude and degree of evolution in gene expression plasticity, which was pertinent to not only adaptation-associated genes but also the whole transcriptomes from multiple tissues. Furthermore, we revealed that genes with weaker expressional interactions with other genes tended to exhibit stronger plasticity and higher degree of plasticity evolution, which explains the positive magnitude-evolution relationship. Conclusions Our experimental evidence demonstrates that species may initiate their adaptation to new habitats with genes exhibiting strong expression plasticity. We also highlight the role of expression interdependence among genes in regulating the magnitude and evolution of expression plasticity. This study illuminates how the evolution of phenotypic plasticity in gene expression facilitates the adaptation of species to challenging environments in nature.

Abstract As the largest source of carbon emissions in China, the thermal power industry is the only emission-controlled industry in the first national carbon market compliance cycle. Its conversion to clean-energy generation technologies is also an important means of reducing CO2 emissions and achieving the carbon peak and carbon neutral commitments. This study used fractional Brownian motion to describe the energy-switching cost and constructed a stochastic optimization model on carbon allowance (CA) trading volume and emission-reduction strategy during compliance period with the Hurst exponent and volatility coefficient in the model estimated. We defined the optimal compliance cost of thermal power enterprises as the form of the unique solution of the Hamilton–Jacobi–Bellman equation by combining the dynamic optimization principle and the fractional Itô’s formula. In this manner, we obtained the models for optimal emission reduction and equilibrium CA price. Our numerical analysis revealed that, within a compliance period of 2021–2030, the optimal reductions and desired equilibrium prices of CAs changed concurrently, with an increasing trend annually in different peak-year scenarios. Furthermore, sensitivity analysis revealed that the energy price indirectly affected the equilibrium CA price by influencing the Hurst exponent, the depreciation rate positively impacted the CA price, and increasing the initial CA reduced the optimal reduction and the CA price. Our findings can be used to develop optimal emission-reduction strategies for thermal power enterprises and carbon pricing in the carbon market.

Diffuse large B-cell lymphoma (DLBCL) is the most common pathological type of non-Hodgkin lymphoma, and is closely associated with hepatitis B virus (HBV) infection status and hepatitis B X (HBx) gene integration. This project investigated the cellular biological effects and molecular mechanisms responsible for lymphomagenesis and the progression of HBx integration in DLBCL. The data showed that clinical DLBCL cells demonstrated HBx integration, and the sequencing analysis of integrated sites validated HBx integration in the constructed HBx-transfected cells. Compared with control cells, HBx-transfected cells had a significantly reduced proportion of mitochondrial membrane potential, signals of chromosomal DNA breaks, and proportion of apoptotic cells. Further studies found that this decreased apoptosis level was associated with a significant reduction of cleaved Caspase-3 and downstream poly ADP-ribose polymerase (PARP) proteins, revealing the molecular mechanisms of HBx-associated apoptosis in DLBCL. Animal experiments also demonstrated that the protein expression of cleaved Caspase-3 and PARP was prominently reduced in HBx-transfected cells from subcutaneous tumors in mice. Furthermore, the HBx-integrated cells in clinical tissues had significantly lower cleaved PARP levels than the HBx-negative samples. Therefore, HBx integration inhibits cell apoptosis through the Caspase-3-PARP pathway in DLBCL indicating a potential biomarker and therapeutic target in HBV related DLBCL.

RNA 5-methylcytosine (m5C) is a widespread post-transcriptional modification involved in diverse biological processes through controlling RNA metabolism. However, its roles in uveal melanoma (UM) remain unknown. Here, we describe the biological roles and regulatory mechanisms of RNA m5C in UM. Initially, we identified significantly elevated global RNA m5C levels in both UM cells and tissue specimens using ELISA assay and dot blot analysis. Meanwhile, NOP2/Sun RNA methyltransferase family member 2 (NSUN2) was upregulated in both types of these samples, whereas NSUN2 knockdown significantly decreased RNA m5C level. Such declines inhibited UM cell migration and suppressed cell proliferation through cell cycle G1 arrest. Furthermore, bioinformatic analyses, m5C-RIP-qPCR, and luciferase assay identified β-Catenin (CTNNB1) as a direct target of NSUN2-mediated m5C modification in UM cells. Additionally, overexpression of miR-124a in UM cells diminished NSUN2 expression levels indicating that it is an upstream regulator of this response. Our study suggests that NSUN2-mediated RNA m5C methylation provides a potential novel target to improve the therapeutic management of UM pathogenesis.

Disassembly is the basis of static analysis of binary code and is used in malicious code detection, vulnerability mining, software optimization, etc. Disassembly of arbitrary suspicious code blocks (e.g., for suspicious traffic packets intercepted by the network) is a difficult task. Traditional disassembly methods require manual specification of the starting address and cannot automate the disassembly of arbitrary code blocks. In this paper, we propose a disassembly method based on code extension selection network by combining traditional linear sweep and recursive traversal methods. First, each byte of a code block is used as the disassembly start address, and all disassembly results (control flow graphs) are combined into a single flow graph. Then a graph attention network is trained to pick the correct subgraph (control flow graph) as the final result. In the experiment, the compiler-generated executable file, as well as the executable file generated by hand-written assembly code, the data file and the byte sequence intercepted by the code segment were tested, and the disassembly accuracy was 93%, which can effectively distinguish the code from the data.

The heavy metal ion Cu2+ was used as the research object in this study. Cu2+ was detected using a cysteine- and glutathione-modified cadmium telluride quantum dot solution. Compounds of Cu2+ and modified quantum dots on the surface caused the fluorescence quenching of quantum dots. A linear relationship existed between the concentration and the intensity of the change in fluorescence quenching. Using this linear relationship, the quantitative detection of the Cu2+ content in the solution by quantum dots was achieved; the detection band was 476-800 nm. Under optimized conditions, the detection limit of the method was 3.58×10-5 mol/L (S/N=3, n=11), and the linear range was 3×10-7 to 8×10-4 mol/L. In a certain experimental environment, standard normal variate preprocessing of the characteristic band and partial least squares modeling and analysis yielded good self-prediction and actual prediction abilities. The correction coefficient was 0.9006. The RMSEC was 8.83084 µmol/L, the determination coefficient of the validation set was 0.8686, and the RMSEP was 5.4320 µmol/L. The results showed that the modeling effect was good in the experiment, the operation was simple and convenient, and the experiment was fast, reliable, and nonpolluting, indicating that the method of detecting Cu2+ using a quantum dot fluorescent probe was feasible.

In the paper, N1212 working face was taken as the research object, the law and mechanism of surface cracks in shallow coal seam mining were studied by means of field investigation, dynamic monitoring and numerical simulation. The results show that the development degree of surface cracks in shallow coal seam mining is high, mainly “step” cracks, and the in-plane cracks are distributed at parallel intervals with an interval distance of 8-15 m. The cracks have two kinds of activity characteristics: “opening-closing” and “only opening and disconnecting”; the stress evolution and failure in the overlying strata extend forward in the form of “arch”, and the overlying rock fissures and surface cracks are connected with each other; the advanced generation and activity characteristics of surface cracks are closely related to surface movement and deformation, surface “tension-compression” deformation transformation and slope slip. The research results can provide a theoretical basis for the prevention and control of ground collapse in coal mining areas.

Currently, China will promote county towns’ urbanization, and few studies have analyzed spatial effects of urban land-use intensity on CO2 emissions and predicted CO2 emissions at the county level accurately. Here, taking data from 2010 to 2015 at the county level of Zhejiang Province as an example, we analyzed the spatial effect of urban land-use intensity from three aspects of input, density and output on CO2 emissions by the spatial Durbin model (SDM). And then a machine learning method of Back Propagation Neural Network (BPNN) was proposed to predict CO2 emissions for 2035 nonlinearly under the different promotions of urban land-use intensity. The main result and conclusion showed that: (1) The spillover effects of urban land-use density were negatively related to CO2 emissions, and urban land-use output intensity showed positive spillover effects on CO2 emissions; (2) The prediction of BPNN showed that the improvement of urban land-use intensity would not be effective in CO2 emissions reductions for southwestern counties with both low levels of urbanization and urban land-use intensity; (3) It was effective in CO2 emissions reduction by slowing down the growth rate of urban land-use capital input intensity, especially for the northeast region which was developed by the port economy. Our study encouraged a regional differentiated urban land-use intensity improvement strategy for Zhejiang to achieve low carbon development.

Myocardial infarction (MI) is caused by the formation of plaques in the arterial walls, leading to a decrease of blood flow to the heart and myocardium injury as a result of hypoxia. Ferroptosis is a crucial event in myocardial injury, and icariin (ICA) exerts protective effects against myocardial injury. Here, we investigated the protective mechanism of ICA in hypoxia/reoxygenation (H/R)‐induced ferroptosis of cardiomyocytes. H9C2 cells were subjected to H/R induction. The content of lactate dehydrogenase and the levels of oxidative stress and intracellular ferrous ion Fe2+ were measured. The levels of ferroptosis markers (ACSL4 and GPX4) were detected. H/R‐induced H9C2 cells were cultured with ICA in the presence or absence of ferroptosis inducer (erastin). Znpp (an HO‐1 inhibitor) was added to ICA‐treated H/R cells to verify the role of the Nrf2/HO‐1 pathway. H/R‐induced H9C2 cells showed reduced viability, enhanced oxidative stress and lactate dehydrogenase content, increased levels of Fe2+ and ACSL4, and decreased levels of GPX4. ICA inhibited H/R‐induced ferroptosis and oxidative stress in cardiomyocytes. Erastin treatment reversed the inhibitory effect of ICA on ferroptosis in H/R cells. The expression of Nrf2 and HO‐1 in H/R‐induced H9C2 cells was reduced, whereas ICA treatment reversed this trend. Inhibition of the Nrf2/HO‐1 pathway reversed the protective effect of ICA on H/R‐induced ferroptosis. Collectively, our results suggest that ICA attenuates H/R‐induced ferroptosis of cardiomyocytes by activating the Nrf2/HO‐1 signaling pathway.

BackgroundLaparoscopic colectomy is widely practiced for colon cancer, but many variations exist for anastomosis after laparoscopic colon cancer radical resection.MethodWe retrospectively analyzed 226 patients who underwent laparoscopic-assisted radical resection for left colon cancer with knotless hand-sewn end-to-end anastomosis (KHEA) technique with barbed V-loc™ suture material and compared perioperative outcomes, safety, and efficacy to those undergoing stapled anastomosis from 2010 to 2021.ResultsAfter the 1:2 propensity score matching, 123 participants with similar preoperative characteristics (age, body mass index, TNM stage, and tumor location) were enrolled in the study: 41 in the KHEA and 82 in the stapler group. Statistically significant differences were found in time to accomplish the anastomosis (mean 7.9 vs. 11.9 min, p 0.05) in the KHEA and stapler group, respectively.ConclusionKHEA is feasible and safe for anastomosis after laparoscopic left hemicolectomy. The KHEA technique could reduce operation time and hospital costs with complication rates comparable to stapling.

BackgroundAlthough neoadjvuant chemoradiotherapy (CRT) improves the local control rate of locally advanced rectal cancer (LARC), it fails to significantly improve disease-free survival (DFS) and overall survival (OS). We explored the efficacy of prolonged neoadjuvant chemotherapy (pNCT) without radiation and compared this schema with total neoadjuvant therapy (TNT).Material and methodsPatients diagnosed with LARC and received TNT (4 cycles of induction CapeOX/FOLFOX followed with CRT) or pNCT (6~8 cycles of CapeOX/FOLFOX) between June 2016 and October 2021 were retrospective analyzed. All patients underwent total mesorectal excision (TME). A 1:1 propensity score match was performed to adjust baseline potential confounders. The tumor response, toxicity, recurrence-free survival (RFS) and OS were observed.ResultsA total of 184 patients with 92 patients in each group were finally enrolled. The median follow-up time was 35 months. TNT showed better pathological complete response (pCR) rate (25.0% vs 16.3%) and objective regression rate (73.9% vs 59.8%) than pNCT. TNT and pNCT produce similar 3-year RFS and OS rates in patients with mid-to-upper rectal cancer. TNT was associated with improved tumor responsiveness in all patients and improved 3-year RFS rates in those with low rectal cancer.ConclusionpNCT is an option for patients with mid-to-upper rectal cancer, but radiation is still necessary for low rectal cancer. To determine optimal schema for neoadjuvant therapy and patient selection, additional randomized controlled studies are needed.

Background Glioblastoma (GBM) is the most common malignant brain tumor with poor clinical outcomes. Immunotherapy has recently been an attractive and promising treatment of extracranial malignancies, however, most of clinical trials for GBM immunotherapy failed due to predominant accumulation of tumor-associated microglia/macrophages (TAMs).Results High level of LRIG2/soluble LRIG2 (sLRIG2) expression activates immune-related signaling pathways, which are associated with poor prognosis in GBM patients. LRIG2/sLRIGs promotes CD47 expression and facilitates TAM recruitment. Blockade of CD47–SIRPα interactions and inhibition of sLRIG2 secretion synergistically suppress GBM progression in an orthotropic murine GBM model.Conclusions GBM cells with high level LRIG2 escape the phagocytosis by TAM via the CD47-SIRPα axis, highlighting a necessity for an early stage of clinical trial targeting LRIG2 and CD47-SIRPα as a novel treatment for patients with GBM.

BackgroundCXCL12/CXCR4 signaling is essential in cardiac development and repair, however, its contribution to aortic valve stenosis (AVS) remains unclear. In this study, we tested the role of endothelial CXCR4 on the development of AVS.Materials and methodsWe generated CXCR4 endothelial cell-specific knockout mice (EC CXCR4 KO) by crossing CXCR4fl/fl mice with Tie2-Cre mice to study the role of endothelial cell CXCR4 in AVS. CXCR4fl/fl mice were used as controls. Echocardiography was used to assess the aortic valve and cardiac function. Heart samples containing the aortic valve were stained using Alizarin Red for detection of calcification. Masson’s trichrome staining was used for the detection of fibrosis. The apex of the heart samples was stained with wheat germ agglutinin (WGA) to visualize ventricular hypertrophy.ResultsCompared with the control group, the deletion of CXCR4 in endothelial cells led to significantly increased aortic valve peak velocity and aortic valve peak pressure gradient, with decreased aortic valve area and ejection fraction. EC CXCR4 KO mice also developed cardiac hypertrophy as evidenced by increased diastolic and systolic left ventricle posterior wall thickness (LVPW), cardiac myocyte size, and heart weight (HW) to body weight (BW) ratio. Our data also confirmed increased microcalcifications, interstitial fibrosis, and thickened valvular leaflets of the EC CXCR4 KO mice.ConclusionThe data collected throughout this study suggest the deletion of CXCR4 in endothelial cells is linked to the development of aortic valve stenosis and left ventricular hypertrophy. The statistically significant parameters measured indicate that endothelial cell CXCR4 plays an important role in aortic valve development and function. We have compiled compelling evidence that EC CXCR4 KO mice can be used as a novel model for AVS.

China is now facing the challenges of high leverage. Our study has examined the role of firms’ rising debt in explaining the decrease in labor share in recent years. We developed a model to show that firms may strategically use debt to increase their bargaining power, resulting in lower labor share. Empirical evidence shows the labor share is robustly negatively related to debt per worker, and the negative correlation varies across ownership, labor union and maturity of debt. The findings imply that the de-leveraging policy may help alleviate the worsening income inequality.

Abstract Background People with type 2 diabetes mellitus (T2DM) generally spend a large amount of time sitting. This increases their risk of cardiovascular disease, premature mortality, diabetes-related complications and mental health problems. There is a paucity of research that has evaluated interventions aimed at reducing and breaking up sitting in people with T2DM. The primary aim of this study is to assess the feasibility of delivering and evaluating a tailored intervention to reduce and break up sitting in ambulatory adults with T2DM. Methods This is a mixed-methods randomised controlled feasibility trial. Participants (n=70) with T2DM aged 18-85 years who sit ≥7 h/day and are able to ambulate independently will be randomly allocated to receive the REgulate your SItting Time (RESIT) intervention or usual care (control group) for 24 weeks. RESIT is a person-focused intervention that delivers a standardised set of behaviour change techniques to the participants, but the mode through which they are delivered can vary depending on the tools selected by each participant. The intervention includes an online education programme, health coach support, and a range of self-selected tools (smartphone apps, computer-prompt software, and wearable devices) that deliver behaviour change techniques such as self-monitoring of sitting and providing prompts to break up sitting. Measures will be taken at baseline, 12 and 24 weeks. Eligibility, recruitment, retention and data completion rates will be used to assess trial feasibility. Sitting, standing and stepping will be measured using a thigh-worn activity monitor. Cardiometabolic health, physical function, psychological well-being, sleep and musculoskeletal symptoms will also be assessed. A process evaluation will be conducted including evaluation of intervention acceptability and fidelity. Discussion This study will identify the feasibility of delivering a tailored intervention to reduce and break up sitting in ambulatory adults with T2DM and evaluating it through a randomised controlled trial (RCT) design. The findings will inform a fully powered RCT to evaluate the effectiveness of the intervention. Trial registration ISRCTN, ISRCTN14832389 ; Registered 6 August 2020.

Fatty acid amides (FAAs) are endogenous lipid molecules that exhibit various physiological activities. FAAs are usually present at nanomolar levels in biological samples. In this study, a method for the qualitative and quantitative determination of six FAAs (linoleamide, linoleoyl ethanolamide, oleoyl ethanolamide, palmitic amide, oleamide, and octadecanamide) in edible vegetable oils was established. All six FAAs were detected in sesame, peanut, soybean (decolorized and non-decolorized), and blended oils; five in sunflower oil; four in rice oil; three in linseed and olive oils; and two in corn and canola oils. The total contents of FAAs were highest in sesame oil (104.88 ± 3.01 μg/mL), followed by peanut oil (34.96 ± 3.87 μg/mL), soybean oil (16.75 ± 1.27 μg/mL), and blended oil (13.33 ± 0.77 μg/mL), and the contents in the other edible vegetable oils were all

Despite significant advancements in object detection technology, most existing detection networks fail to investigate global aspects while extracting features from the inputs and cannot automatically adjust based on the characteristics of the inputs. The present study addresses this problem by proposing a detection network consisting of three stages: preattention, attention, and prediction. In the preattention stage, the network framework is automatically selected based on the features of the images’ objects. In the attention stage, the transformer structure is introduced. Taking into account the global features of the target, this study combines a self-attention module in the transformer model and convolution operation to integrate image features from global to local and for detection, thus improving the ship target accuracy. This model uses mathematical methods to obtain results of predictive testing in the prediction stage. The above improvements are based on the You Only Look Once version 4 (YOLOv4) framework, named “Auto-T-YOLO”. The model achieves the highest accuracy of 96.3% on the SAR Ship Detection dataset (SSDD) compared to the other state-of-the-art (SOTA) model. It achieves 98.33% and 91.78% accuracy in the offshore and inshore scenes, respectively. The experimental results verify the practicality, validity, and robustness of the proposed model.

Many clinical trials have attempted to use stem cells to treat ischemic heart diseases (IHD), but the benefits have been modest. Though coronary collaterals can be a “natural bypass” for IHD patients, the regulation of coronary collateral growth (CCG) and the role of endogenous stem cells in CCG are not fully understood. In this study, we used a bone marrow transplantation scheme to study the role of bone marrow stem cells (BMSCs) in a rat model of CCG. Transgenic GFP rats were used to trace BMSCs after transplantation; GFP bone marrow was harvested or sorted for bone marrow transplantation. After recovering from transplantation, the recipient rats underwent 10 days of repetitive ischemia (RI), with echocardiography before and after RI, to measure cardiac function and myocardial blood flow. At the end of RI, the rats were sacrificed for the collection of bone marrow for flow cytometry or heart tissue for imaging analysis. Our study shows that upon RI stimulation, BMSCs homed to the recipient rat hearts’ collateral-dependent zone (CZ), proliferated, differentiated into endothelial cells, and engrafted in the vascular wall for collateral growth. These RI-induced collaterals improved coronary blood flow and cardiac function in the recipients’ hearts during ischemia. Depletion of donor CD34+ BMSCs led to impaired CCG in the recipient rats, indicating that this cell population is essential to the process. Overall, these results show that BMSCs contribute to CCG and suggest that regulation of the function of BMSCs to promote CCG might be a potential therapeutic approach for IHD.

ALDH1A3 and Linc00284 involve in colorectal cancer (CRC) development; however, the regulatory mechanism is still unclear. In this study, we collected clinicopathological characteristics and tissue samples from 73 CRC patients to analyze the expression of ALDH1A3, Linc00284, TGFβ signaling and miR-361-5p using qPCR, Western blotting, and ELISA. Multiple CRC cell lines were evaluated in this study, and the highest level of ALDH1A3 was observed in SW480 cells. To investigate the regulatory mechanism, RIP and luciferase assays were used to validate the interaction between Linc00284, miR-361-5p, and TGFβ. Proliferation, viability, migration, and invasion assays were performed to profile the effects of the ALDH1A3–Linc00284 axis in CRC cell functions, which was upregulated in CRC tissues. Knockdown ALDH1A3 or Linc00284 significantly reduced TGFβ expression and suppressed the EMT process, while overexpression had opposite effects. miR-361-5p targeted TGFβ directly, which negatively correlated with ALDH1A3–Linc00284 expression and CRC progression. Mechanistically, upregulation of ALDH1A3–Linc00284 promotes colorectal cancer invasion and migration by regulating miR-361-5p/TGFβ signaling pathway. Dysregulation of the ALDH1A3–Linc00284-miR-361-5p-TGFβ axis causes CRC invasion, which might provide a new insight into the treatment of CRC.

Cognitive diversity is an important concept stemming from western management research in the 1990s. With the rapid development of science and technology, there is a growing interest in the composition of an academic research team, such as team diversity. However, there is no tool available for measuring team cognitive diversity (TCD) for academic research teams. Based on Van der Vegt’s theoretical model of TCD, an Academic Research Team Cognitive Diversity Scale (ATCDS) is developed and validated for an academic research team in our research with two studies (N=737). In Study One, in-depth interviews and panel discussions were conducted to generate a preliminary questionnaire. In Study Two, the questionnaire was administered among academic research teams. Exploratory factor analysis revealed four factors regarding cognitive diversity: (1) the way of thinking, (2) knowledge and skills, (3) the view of the world, and (4) beliefs about what is right and wrong. The factor structure was further validated by confirmatory factor analysis. Objective To investigate the transcriptomic changes in atherosclerosis after cetp knockout of zebrafish. Methods The gene editing technology CRISPR/Cas9 was used to knock out cetp of zebrafish to construct a homozygous mutant model. Total RNA of livers from wild type and cetp-/- model was extracted by the Trizol method, and gene expression profiles of cetp knockout were observed by transcriptomics. KEGG enrichment analysis was performed on significantly different expressed genes to explore the signal pathways it may affect. Results Homozygous mutant zebrafish that has a frameshift mutation of adding 5 base pairs was created through CRISPR/Cas9 technology. Transcriptomics analysis showed that the gene expression profile of zebrafish liver changed significantly after cetp knockout. A total of 3 808 different expressed genes were screened (P＜0.05), of which 1 918 genes were up-regulated and 1 890 genes were down-regulated. Genes related to lipid metabolism such as abca1 and abcg2a were significantly up-regulated and lipg were significantly down-regulated. Genes related to autophagy signaling pathways such as atg13 were significantly up-regulated, and down-regulated genes were enriched in related pathways such as DNA and RNA synthesis and the tricarboxylic acid cycle. Conclusions A zebrafish model with cetp knockout is established to study the mechanism of cholesteryl ester transfer protein (CETP) in atherosclerotic diseases. Although increased high-density lipoprotein cholesterol due to the inhibition of CETP prevents atherosclero-sis, inflammation is activated after cetp knockout in zebrafish and causes atherosclerosis. It suggests that CETP has a dual regulation mechanism in the development of atherosclerosis.

AIM: To evaluate the therapeutic effect of combined phacovitrectomy with membrane peeling and intraocular lens (IOL) implantation in patients with severe idiopathic epiretinal membrane (iERM) and concurrent cataract. METHODS: A total of 34 eyes from 34 patients who underwent phacovitrectomy and epiretinal membrane (ERM) peeling at the First Affiliated Hospital of Zhejiang University between 2015 and 2017. Circular Synthetic Aperture Radar (CSAR) is a novel imaging mode, which has the advantages of alldirectional observation, high spatial resolution, and three-dimensional imaging. With the development of airborne CSAR imaging techniques, it has become one of the effective methods for key point area observation. This paper introduces works on airborne CSAR imaging techniques performed by our research team in recent years, including airborne CSAR imaging mode, spatial resolution evaluation, two-dimensional CSAR imaging, three-dimensional target image reconstruction based on a single CSAR, and three-dimensional holographic SAR imaging. In this paper, experimental results based on raw data acquired using airborne CSAR systems with P and X bands are presented. The obtained research results prove the effectivity and practicability of the airborne CSAR imaging mode. The content of this paper is based on a keynote speech presented by the author at the Fifth Young Scientists Forum of Journal of Radars on August 15, 2019.

A marine vessel re-identification system has to determine whether or not different images represent the same vessel. Accurate vessel re-identification improves onshore closed-circuit television monitoring in a vessel traffic services system as well as onboard surveillance of surrounding vessels. However, because ships are rigid bodies and the marine environment is harsh, the accurate re-identification of vessels at sea can be very difficult. We describe a marine vessel-re-identification framework, Global-and-Local Fusion-based Multi-view Feature Learning (GLF-MVFL), which is based on a combination of global and fine-grained local features. GLF-MVFL combines cross-entropy loss with our newly-developed orientation-guided quintuplet loss. We exploit intrinsic features of marine vessels to optimize multi-view representation learning for re-identification. GLF-MVFL uses ResNet-50 as the backbone network to extract features for simultaneous quintuple input. It detects and discriminates between features and estimates viewpoints to form a comprehensive re-identification framework. We created an annotated large-scale vessel retrieval dataset, VesselID-539, which contains images from viewpoints similar to those of an autonomous surface vessel, to use in evaluating the performance of the model. Extensive experiments and analysis of the results obtained from using VesselID-539 demonstrate that our approach significantly increases the accuracy of vessel re-identification and is more effective and robust for images from different viewpoints than other approaches.

In recent years, many studies have been conducted in the field of firearm identification with the objective of providing an objective method of evaluating the comparison of cartridge cases. However, less attention has been paid to the objective evaluation of bullet comparisons. In this study, 1 000 registered Chinese Norinco QSZ-92 pistols were used, and a database of 2 996 bullets was constructed. Both the receiver operating characteristic (ROC) curve and the score-based likelihood ratio method were used to objectively evaluate the similarity scores derived by the Evofinder® system. The results indicate that this system has excellent ability to discriminate between the selected pistols. This paper proposes an objective evaluation method, which serves as a response to the ongoing debates regarding the foundation of the discipline.

Distant metastasis is the principal cause of mortality for breast cancer patients. Targeting specific mutations that have been acquired during the evolution process of advanced breast cancer is a potential means of enhancing the clinical efficacy of treatment strategies. In metastatic breast cancer, ARID1A is the most prevalent mutation of the SWI/SNF complex, which regulates DNA repair, recombination, and gene transcription. The low expression of ARID1A is associated with poor disease-free survival and overall survival of patients with luminal A or HER2-rich breast cancer. In addition, ARID1A plays a prominent role in maintaining luminal characteristics and has an advantage for identifying responses to treatment, including endocrine therapies, HDAC inhibitors and CDK4/6 inhibitors. The therapeutic vulnerabilities initiated by ARID1A alterations encourage us to explore new approaches to cope with ARID1A mutant-related drug resistance or metastasis. In this review, we describe the mutation profiles of ARID1A in metastatic breast cancer and the structure and function of ARID1A and the SWI/SNF complex as well as discuss the potential mechanisms of ARID1A-mediated endocrine resistance and therapeutic potential.

Economic development is accompanied by the production of pollutants. Coordinating the relationship between economic development and haze pollution is of great significance to future development in China. Based on the original decoupling index (Tapio decoupling index), this study developed a new decoupling index that overcomes some of the original defects and highlights the trends to a better extent. It also analyzes the transformation relationship among the new decoupling index, the original decoupling index and the EKC curve. Firstly, we compared the changing trends of the two decoupling indexes and then derived the factors that influence the new and original decoupling indexes based on C-D-Kaya equation. Finally, we compared the factors influencing the two decoupling indexes from the perspective of decomposition analysis and measurement. The conclusions are as follows: (1) Based on the limit theory, the boundary point for relative decoupling and absolute decoupling derived from the original decoupling index can be used to determine the inflection point of the EKC curve. The new decoupling index can also reflect this property. At the inflection point of the EKC curve, the ratio of the new decoupling index of two adjacent periods satisfies equation (12), which is only related to GDP. (2) In 2003, the new decoupling index showed a downward trend, and the fluctuation was smaller than that of the original decoupling index. The original decoupling index is more sensitive to short-term policies than the new decoupling index, but the new decoupling index is more conducive to the comprehensive analysis of China’s decoupling trend. (3) After decomposing the new decoupling index and the original decoupling index, per capita GDP and the emission coefficient are the largest positive and negative driving forces, respectively. However, the trend of the contribution made by each factor was more obvious when the new decoupling index was used. The emission coefficient for Shandong made the largest negative contribution, and the per capita GDP for Shanxi made the greatest positive contribution. (4) The geographically and temporally weighted regression (GTWR) analysis showed that the fitting degree of the new decoupling index is much higher than that of the original decoupling index. The analysis of the influencing factors affecting the new decoupling index showed that there was temporal and spatial heterogeneity among the factors in east, middle, and west China, and there were significant differences in the impacts due to energy structure, energy intensity, technological progress, and urban population factors, respectively. These findings will significantly improve the formulation of efficient and pertinent policies for energy conservation and haze reduction.

Lung cancer (LC) is a malignant tumor with the highest incidence and mortality rates worldwide. Linc00284, a long non-coding RNA, is a newly discovered regulator of LC. This study aimed to explore the role of Linc00284 in LC progression. Gene expression levels were detected by RT-qPCR and/or western blot analysis. Cell migratory and invasive capabilities were measured by wound healing and transwell assays. Subcutaneous xenograft models were constructed to examine tumor growth of LC cells. Data showed that Linc00284 was significantly upregulated in LC tissues compared to adjacent normal lung tissues and predicted poor prognosis in patients with LC. In vitro, Linc00284 was highly expressed in LC cells and was mainly localized in the cytoplasm. Mechanistically, Linc00284 directly bound to miR-205-3p, leading to the upregulation of c-Met expression. A significant negative correlation was observed between Linc00284 and miR-205-3p expression levels, and the Linc00284 level was positively correlated with the c-Met expression. Linc00284/miR-205-3p/c-Met regulatory axis promotes LC cell proliferation, migration, and invasion. Furthermore, the in vivo results indicated that Linc00284 knockdown markedly suppressed tumor growth. Taken together, these data suggest that Linc00284 facilitates LC progression by targeting the miR-205-3p/c-Met axis, which may be a potential target for LC treatment.

Electrical impedance tomography (EIT) is a non-invasive detection technology that uses the electrical response value at the boundary of an observation field to image the conductivity changes in an area. When EIT is applied to the thoracic cavity of the human body, the conductivity change caused by the heartbeat will be concentrated in a sub-region of the thoracic cavity, that is, the heart region. In order to improve the spatial resolution of the target region, two sensor optimization methods based on conformal mapping theory were proposed in this study. The effectiveness of the proposed method was verified by simulation and phantom experiment. The qualitative analysis and quantitative index evaluation of the reconstructed image showed that the optimized model could achieve higher imaging accuracy of the heart region compared with the standard sensor. The reconstruction results could effectively reflect the periodic diastolic and systolic movements of the heart and had a better ability to recognize the position of the heart in the thoracic cavity.

Abstract Since its inception in 2017, npj Digital Medicine has attracted a disproportionate number of manuscripts reporting on uses of artificial intelligence. This field has matured rapidly in the past several years. There was initial fascination with the algorithms themselves (machine learning, deep learning, convoluted neural networks) and the use of these algorithms to make predictions that often surpassed prevailing benchmarks. As the discipline has matured, individuals have called attention to aberrancies in the output of these algorithms. In particular, criticisms have been widely circulated that algorithmically developed models may have limited generalizability due to overfitting to the training data and may systematically perpetuate various forms of biases inherent in the training data, including race, gender, age, and health state or fitness level (Challen et al. BMJ Qual. Saf. 28:231–237, 2019; O’neil. Weapons of Math Destruction: How Big Data Increases Inequality and Threatens Democracy, Broadway Book, 2016). Given our interest in publishing the highest quality papers and the growing volume of submissions using AI algorithms, we offer a list of criteria that authors should consider before submitting papers to npj Digital Medicine.

To ensure a high signal-to-noise ratio and high image volume, a geostationary orbiting ocean remote-sensing system needs to maintain high platform stability over a long integration time because it is affected by satellite attitude changes. When the observation target is the ocean, it is difficult to extract image features because of the lack of characteristic objects in the target area. In this paper, we attempt to avoid using image data for satellite attitude and image pixel offset estimation. We obtain the satellite attitude by using equipment such as gyroscopes and performing time registration between the satellite attitude and the image data to achieve pixel offset matching between images. According to the law of satellite attitude change, we designed a Kalman-like filter fitting (KLFF) algorithm based on the satellite attitude change model and the Nelder–Mead search principle. The discrete attitude data were time-marked by a synchronization system, and high-precision estimation of the satellite attitude was achieved after fitting with the KLFF algorithm. When the measurement accuracy of the equipment was 1.0 × 10−3°, the average prediction error of the algorithm was 1.09 × 10−3°, 21.58% better than the traditional interpolation prediction result of 1.39 × 10−3°. The peak value of the fitting angle error reached 2.5 × 10−3°. Compared with the interpolation prediction result of 6.2 × 10−3°, the estimated stability of the satellite attitude improved by about 59.68%. After using the linear interpolation method to compensate for the estimated pixel offset, its discrete range was 0.697 pixels. Compared with the 1.476 pixels of the interpolation algorithm, it was 52.8% lower, which improved the noise immunity of the algorithm. Finally, a KLFF algorithm was designed based on the satellite attitude change model by using external measurement data and the synchronous clock as a benchmark. The instantaneous attitude of the satellite was accurately estimated in real time, and the offset matching between the images was realized, which lays the foundation for in-orbit satellite data processing.

BackgroundSerum thyroglobulin (Tg) serves as a sensitive and easily available tumor marker for patients with metastatic differentiated thyroid carcinoma (m-DTC). The aim of the present study was to evaluate the predictive value of suppressed Tg changes (Δsup-Tg) and/or stimulated Tg changes (Δsti-Tg) to evaluate the efficacy of radioiodine therapy (RT).MethodsWe studied 117 patients with m-DTC who received RT. Δsup-Tg and Δsti-Tg were compared after the first RT in different therapeutic response groups and a receiver-operating characteristic (ROC) curve was used to determine the cut-off values to predict non-remission. Univariate and multivariate analyses were used to investigate the effects of 17 observed factors on the efficacy of RT.ResultsA total of 218 RT events in 117 patients with m-DTC were analyzed. After the last RT, the remission rate was 70.94% (83/117), and the proportion of remission events accounted for 74.77% (163/218). ROC curve analysis showed that the cut-off values for Δsup-Tg and Δsti-Tg after the first RT to predict the non-remission of RT were 21.54% and 27.63%, respectively. Age, the size of the metastasis, the maximum count of target metastatic lesions and the average count of contralateral non-target tissue on tomographic imaging (Tmax/NTmean) of the first RT, and Δsup-Tg after the first RT were identified as independent factors associated with RT efficacy.ConclusionsTg response was valuable to predict RT efficacy for patients with m-DTC. Age, the size of the metastasis, Tmax/NTmean, and Δsup-Tg after the first RT were verified as independent predictive factors of RT efficacy.

Abstract Background and aims Thrombolytic therapy is widely used to treat acute ischemic stroke (AIS) patients. As intracerebral hemorrhage is a life‐threatening complication of this therapy, monitoring the fibrinolytic and coagulation systems is imperative. However, existing studies on plasmin inhibitor complex (PIC) and thrombin‐antithrombin III complex (TAT) mostly apply the enzyme‐linked immunosorbent assay (ELISA) method. The aim of this study is to establish the baseline of thrombolytic treatment for AIS patients; to monitor the fibrinolytic and coagulation system following alteplase administration; to ascertain the proper time point to predict intracerebral hemorrhage. Methods The method used to assess a patient's intravascular situation, namely chemiluminescence, was used to quantitatively assess the PIC, TAT, and thrombomodulin (TM). Immuno‐turbidimetric was used to assess the concentration of D‐dimer, fibrin/fibrinogen degradation products (FDP), and the Von Willebrand factor (vWF). The Clauss clotting method was used to assay the activated partial thromboplastin time (APTT), prothrombin time (PT) and FIB. Results PIC increased to its peak concentration at 3 hours post intravenous (IV) alteplase infusion and decreased by nearly 50% every 3 hours thereafter. After 24 hours, PIC returned to its normal range, while D‐dimer and FDP decreased 3 hours later compared to PIC. PT and APTT exhibited no obvious change during the 24‐hour period. TM also exhibited no changes during the treatment. Conclusion PIC decreased 3 hours earlier than D‐dimer and FDP. The combined test of PIC, D‐dimer, and fibrinogen can be used to monitor the fibrinolytic system after the IV alteplase infusion. The use of IV alteplase had no impact on the endothelium. Creating a patient's individual data curve could assist in the prediction of hemorrhagic transformation (HT) and a stroke occurring.

The annotation of the tumour from medical scans is a crucial step in nephroblastoma treatment. Therefore, an accurate and reliable segmentation method is needed to facilitate the evaluation and the treatments of the tumour. The proposed method serves this purpose by performing the segmentation of nephroblastoma in MRI scans. The segmentation is performed by adapting and a 2D free hand drawing tool to select a region of interest in the scan slices. Results from 24 patients show a mean root-mean-square error of 0.0481 ± 0.0309, an average Dice coefficient of 0.9060 ± 0.0549 and an average accuracy of 99.59% ± 0.0039. Thus the proposed method demonstrated an effective agreement with manual annotations.

Abstract Background Elevation of exogenous free fatty acid (FFA) level leads to insulin resistance (IR) in liver, IR is manifested by elevated hepatic glucose production. We aim to study whether inhibition of endogenous fatty acid synthesis could decrease hepatic glucose production. Methods Low-passage HepG2 cells derived from human liver tissue were cultured in medium supplemented with FFA to induce IR, the influences of sterol regulatory element binding protein-1c (SREBP-1c) silencing on glucose production of HepG2 cells were investigated, and genes responsible for fatty acid and glucose metabolism were detected by real-time PCR. This paper presents an approach to implement multi-parameter (i.e., pressure, temperature, vibration, current, and liquid level) signals for fault diagnosis of the reciprocating compressor (RC). Due to the complexity of structure and motion of such compressor, the acquired signals involve transient impacts and noises. This causes the useful information to be corrupted and makes it difficult to diagnose the fault patterns accurately. A component estimating empirical mode decomposition (CEEMD) method is proposed to remove the random noise and improve data quality. Furthermore, a new template matching algorithm called de-dimension template with double-loop correction (DDT-DLC) is applied to diagnose the fault pattern contained in the time series signals. The DDT employs a judging criterion for key characterization parameters extraction and a multicellular parameter fusion method to reduce the dimension of the matching template, and then, the DLC supplies a double-loop correction algorithm to build a parameter state array computing model of the time series data by adjusting the dynamic factors. The proposed approach is validated with three fault patterns and the healthy pattern in a two-stage reciprocating air compressor. To confirm the superiority of the proposed method, its performance is compared with that of the traditional methods. The results have indicated that the proposed approach is of highly diagnostic accuracy and shortly computing time in the fault diagnosis.
