Abstract Background Immunoglobulin light-chain amyloidosis is a relatively rare condition with a worldwide incidence of 5.1–12.8 cases per million person-years (Baker, 2022). It is characterized by a clonal population of immunoglobulin-secreting cells that produce a monoclonal light chain of κ or λ type as either an intact molecule or a fragment. Case presentation A 69-year-old East Asian (Chinese) male patient who presented with progressive dysphagia visited multiple hospitals repeatedly for more than 2 years and was finally diagnosed with immunoglobulin light-chain amyloidosis. Conclusions Otolaryngologists should consider immunoglobulin light-chain amyloidosis when encountering suspicious clinical manifestations and intervene early to avoid misdiagnosis.

Abstract Background Bladder duplication is a rare congenital lower urinary tract anomaly disease characterized by the presence of two bladders, possibly with duplication of the urethra. This disease is rarely reported in cats. The clinical symptoms are commonly occult, with increased difficulty in making a definitive diagnosis, especially if there is no obvious urethral duplication. The diagnosis is typically based on radiographs and ultrasound, with computer tomography serving as a more advanced imaging diagnostic modality. Cases of duplicated bladders with accessory tubular tissues are even scarcer in both human and veterinary medicine. Case presentation A 6-year-old male neutered cat was brought to the hospital because of vomiting and constipation. Cystography revealed increased soft tissue density of a fusiform structure in the lower middle abdomen. The purulent-filled cavitary structure and the accessory tubular structure were removed via surgery, and histopathological examination confirmed a double bladder with attached accessory tubular tissue. After antibiotic treatment, the cat recovered uneventfully. Conclusion This is the first case of bladder duplication in China and the first case of feline bladder duplication with tubular structure attachment in the world. This information will provide a reference for the diagnosis and treatment of similar cases in the future.

Abstract Background Sarcoidosis is a multisystemic inflammatory disease, characterized by the presence of non-caseating, epithelioid granulomas. Glomerular disease in patients with sarcoidosis is rare and membranous nephropathy (MN) is cited as the most common. The association between the two diseases remained unclear. This article reported a case of co-occurrence of sarcoidosis and anti-PLA2R-associated MN, to provide a possible relationship between these two entities. Case presentation A 61-year-old Chinese Han woman with a history of sarcoidosis was admitted to our hospital for nephrotic syndrome. Her sarcoidosis was diagnosed according to the adenopathy observed on the computed tomography scan and the biopsy of lymph nodes. The MN presented with nephrotic syndrome with a PLA2R antibody titer of 357RU/ml, and the final diagnosis was based on a renal biopsy. The patient’s sarcoidosis was remitted after treatment with prednisone. One year later MN was diagnosed, and she was treated with prednisone combined with calcineurin inhibitors, based on a full dose of renin-angiotensin system (RAS) inhibitor. The patient’s sarcoidosis had been in remission while the MN was recurrent, and her renal function deteriorated to end-stage renal disease 6 years later due to discontinuation of immunosuppression. A genetic test led to the identification of the HLA-DRB1*0301 and HLA-DRB1*150 genes associated with both sarcoidosis and MN, which provides a new possible explanation of the co-occurrence of these two diseases. Conclusion This case suggested for the first time a potential genetic connection between idiopathic MN and sarcoidosis which needs further studies in the future.

Pen shell (Atrina pectinata) is a Bivalvia species with high commercial value in China. However, large scale production is difficult owing to the lack of seed. Pen shell uses external fertilization with sperm stored in the testis in a nonmotile state. Sperm motility is initiated when they are released from the reproductive tract into the aquatic environment, which enables fertilization. Various chemical signals including pH, ions, and cyclic nucleotides control sperm motility. However, there are interspecific differences in activation conditions between different Bivalvia species. Screening out an effective activating medium is the basis of artificial breeding technology.Swimming parameters, such as total motile sperm (TM), movement velocity, and beat-cross frequency (BCF) are common indicators to evaluate sperm quality. Sperm must have sufficient motility to reach the egg to complete fertilization. In addition, sperm needs a large amount of ATP to maintain its swimming movement. Intracellular ATP content controls the duration of the sperm movement phase in most marine species. Studying the movement characteristics and energy metabolism of sperm during activation will help develop and optimize artificial breeding technology. Studies of A. pectinata mainly focus on their oocytes. However, the activation conditions, moving characteristics, and energy metabolism of sperm during activation remains unknown. Thus, there is an urgent need to screen appropriate media and study the activation mechanisms of A. pectinata sperm.Adult A. pectinata were collected from Wuzhizhou Island, Hainan Province in November 2021. Artificial seawater with different levels of ammonia ions and pH were used to activate sperm. This study examined A. pectinata sperm activation in artificial seawater by varying the ammonia ion concentration and pH. The change in sperm motility, curvilinear velocity (VCL), straight-line velocity (VSL), average path velocity (VAP), and BCF were described. The ATP content, ATPase activities, and superoxide dismutase (SOD) activity were quantitatively recorded during the full activated stage. The motility was slightly improved by increasing the pH of seawater, but could not achieve the fully activated stage. Furthermore, the motility was significantly improved when activated by alkalized seawater containing ammonia ions, and the best results were observed in groups containing 3 mmol/L ammonia ion: The sperm motility index (MI) was in the fully activated stage (MI ≥ 4) until the end of the experiment, with TM ≥ 80%, VCL > 56 μm/s, VSL > 17 μm/s, VAP > 30 μm/s, and BCF > 6 Hz. Sperm ATP content decreased to 30.29% of their initial values [(128.80±66.92) μmol/g prot] during 5 min post activation and was maintained at this level during post-activation. ATPase activities were maintained at a constant level. Na+-K+-ATPase activity was lower [(0.62±0.03) U/mg prot] compared to Ca2+-Mg2+-ATPase activity [(6.08±0.04) U/mg prot]. The SOD activity of sperm steadily decreased to [(1.23±0.73) U/mg prot] during 15 min post activation and remained stable.In conclusion, pH was not the decisive factor in pen shell sperm motility. Abstract Background YouTube, a widely recognized global video platform, is inaccessible in China, whereas Bilibili and TikTok are popular platforms for long and short videos, respectively. There are many videos related to laryngeal carcinoma on these platforms. This study aims to identify upload sources, contents, and feature information of these videos on YouTube, Bilibili, and TikTok, and further evaluate the video quality. Methods On January 1, 2024, we searched the top 100 videos by default sort order (300 videos in total) with the terms “laryngeal carcinoma” and “throat cancer” on YouTube, “喉癌” on Bilibili and TikTok. Videos were screened for relevance and similarity. Video characteristics were documented, and quality was assessed by using the Patient Education Materials Assessment Tool (PEMAT), Video Information and Quality Index (VIQI), Global Quality Score (GQS), and modified DISCERN (mDISCERN). Results The analysis included 99 YouTube videos, 76 from Bilibili, and 73 from TikTok. Median video lengths were 193 s (YouTube), 136 s (Bilibili), and 42 s (TikTok). TikTok videos demonstrated higher audience interaction. Bilibili had the lowest ratio of original contents (69.7%). Treatment was the most popular topic on YouTube and Bilibili, while that was the prognosis on TikTok. Solo narration was the most common video style across all platforms. Video uploaders were predominantly non-profit organizations (YouTube), self-media (Bilibili), and doctors (TikTok), with TikTok authors having the highest certification rate (83.3%). Video quality, assessed using PEMAT, VIQI, GQS, and mDISCERN, varied across platforms, with YouTube generally showing the highest scores. Videos from professional authors performed better than videos from non-professionals based on the GQS and mDISCERN scores. Spearman correlation analysis showed no strong relationships between the video quality and the audience interaction. Conclusions Videos on social media platforms can help the public learn about the knowledge of laryngeal cancer to some extent. TikTok achieves the best flow, but videos on YouTube are of the best quality. However, the video quality across all platforms still needs enhancement. We need more professional uploaders to ameliorate the video quality related to laryngeal carcinoma. Content creators also should be aware of the certification, the originality, and the style of video shooting. As for the platforms, refining the algorithm will allow users to receive more high-quality videos.

In the present study, the effects of different levels of allicin supplementation (0.00 % as control, 0.30 %, 0.45 %, 0.60 %, 0.75 %, and 0.90 %) on the growth performance, biochemical composition and immune capacity of ivory shells (Babylonia areolata) were investigated. Allicin supplementation in the daily diet significantly enhanced growth performance, with the highest specific growth rates observed at concentrations ranging from 0.75 % to 0.90 % (2.18–2.20 %/d), and the lowest feed conversion ratio (1.39) recorded at 0.60 %. Correspondingly, mRNA expression levels of two appetite-related genes, Orexin and Neuropeptide Y (NPY), in the hepatopancreas significantly increased, while the expression of the appetite-suppressing gene Leptin significantly decreased with increasing allicin supplementation. The amounts of almost all detected fatty acids significantly increased when ivory shells were fed diets supplemented with allicin, while their profiles varied, showing increases in C20:1n9 and C22:6n3 (DHA), and decreases in C16:0, C18:2n6, and C20:4n6 (ARA). Lipase activities in both the intestine and hepatopancreas significantly increased in the allicin-supplemented groups, with peak values observed at 0.90 % and 0.75 % allicin supplementation, respectively (3.64 U/g prot in the intestine and 9.52 U/g prot in the hepatopancreas). Allicin supplementation improved the antioxidant and antimicrobial abilities of ivory shells, leading to increased activities of total antioxidant capacity (T-AOC), superoxide dismutase (SOD), catalase (CAT) and lysozyme in the hepatopancreas, and reduced malondialdehyde (MDA) contents. Similar trends were observed in mRNA levels of SOD, CAT and lysozyme genes. In conclusion, moderate allicin supplementation in artificial diets (approximately 0.75 %-0.90 %) can significantly enhance growth performance, increase muscle lipid content, and boost immune capabilities of ivory shells.

With the large-scale interconnection of wind power generation, the voltage problem of the power system becomes more and more prominent. Compared with adding external reactive power compensation devices, it is more economical and responsive for fans to adjust their control strategies to provide reactive power support. To make full use of reactive power supported by wind turbines, a mathematical model of doubly fed induction generator (DFIG) wind turbines is constructed to characterize the reactive power boundary of wind turbines. Then, active disturbance rejection control (ADRC) is used to generate a voltage control signal to effectively improve the unit’s reactive response speed; in addition, a variable gain coefficient is used to adjust the reactive power output of the unit, which effectively improves the reactive power response speed and its control adaptability and robustness under changing power grid conditions. Finally, a wind turbine generator (WTG) simulation model is built using MATLAB/Simulink simulation software, different fault locations are perturbed, and the effectiveness of reactive power support of the proposed ADRC-based strategy is simulated and verified. The proposed ADRC-based strategy could inject more reactive power to the grid to improve the voltage.

CSS-42L is a new generation carburized bearing steel for the critical service environment of aero-engine applications. Owing to the complex composition and heat treatment, this steel exhibits unique microstructural features at the subsurface, one of which is the formation of soft austenitic transition layers. So far, a plausible mechanism of this phenomenon is still absent. In this study, a systematic investigation into such austenitic transition layer formation process is conducted. By using various characterisation techniques along with thermodynamic analysis, it is found that the contents of Cr and C in the matrix are dominated by the formation of different types of carbides along depth. With increasing depth, the undissolved carbides vary from M7C3 and M23C6 to M6C and to nil. This is postulated to result in a drastic decrease of martensitic start (Ms) temperature in the M6C region, leading to the formation of austenitic transition layers. The simulations of both microstructure and Ms temperature evolution along depth during heat treatment are performed for different austenitic temperatures and compared against the experimental observation, with good agreement obtained. This study reveals the mechanism of austenitic transition layers for the first time and provides useful instructions for the further microstructural design of such high-performance bearing steels.

Abstract Background Bacterial drug resistance represents a significant global concern, with vancomycin-resistant Enterococcus faecalis posing a particularly grave threat to contemporary healthcare systems. This study aims to reveal the reasons for the prevalence of VRE in China. Methods This study collected data from the China Antimicrobial Resistance Detection System, China Statistical Yearbook, and China Meteorological Network. The collected data are meticulously organized and subjected to both single-factor and multi-factor analyses. An accurate multiple linear regression model was developed by utilizing this comprehensive dataset. Results Single-factor analysis revealed significant regional variations in the resistance rate of vancomycin-resistant Enterococcus faecalis (P = 0.003). Specifically, there were noteworthy disparities observed between regions experiencing temperate and monsoon climates (P = 0.029; P = 0.005). Furthermore, multi-factor regression analysis demonstrated a negative correlation between the drug resistance rate and both rainfall and rGDP, while a positive correlation was observed with nPI. Conclusion We successfully established a prediction model for the VRE and found that the resistance rate was low in areas with high rainfall and high per capita economic income, but high in areas with many specialized public health institutions. This is critical for public health strategies and helps policymakers and healthcare practitioners tailor antibiotic resistance approaches to local geography, meteorology, economic conditions.

Esophagus cancer (EC) is one of the most aggressive malignant digestive system tumors and has a high clinical incidence worldwide. Magnolol, a natural compound, has anticancer effects on many cancers, including esophageal carcinoma, but the underlying mechanism has not been fully elucidated. Here, we first find that magnolol inhibits the proliferation of esophageal carcinoma cells and enhances their autophagy activity in a dose- and time-dependent manner. This study demonstrates that magnolol increases the protein levels of LC3 II, accompanied by increased HACE1 protein levels in both esophageal carcinoma cells and xenograft tumors. HACE1-knockout (KO) cell lines are generated, and the ablation of HACE1 eliminates the anti-proliferative and autophagy-inducing effects of magnolol on esophageal carcinoma cells. Additionally, our results show that magnolol primarily promotes HACE1 expression at the transcriptional level. Therefore, this study shows that magnolol primarily exerts its antitumor effect by activating HACE1-OPTN axis-mediated autophagy. It can be considered a promising therapeutic drug for esophageal carcinoma.

Abstract This paper examines the application and evolution of philosophical practice in China, which employs philosophical principles and methods to facilitate insights and address life’s complexities. We aim to bridge the knowledge gap regarding the trajectory and contemporary landscape of China’s philosophical practice and its impact on individual and societal flourishing. Guided by the research question, “How has philosophical practice evolved in China, and what are its challenges and future prospects?” this study adopts a qualitative research methodology, including a literature review, historical analysis, and case studies from Chinese philosophical communities. Findings indicate a burgeoning growth of philosophical practice in China, with traditional Chinese philosophies being adapted to provide practical guidance for contemporary issues. The spread of philosophical practice communities across China reflects a growing embrace of these practices. This paper outlines the hurdles and prospects for philosophical practice, pinpointing opportunities for further research and cross-cultural engagement. The insights furnished by this study offer a framework for scholars, practitioners, and policymakers to enhance philosophical practice’s role in personal growth and societal well-being, both within China and internationally.

Abstract Background This study aimed to explore potential indicators associated with the neoadjuvant efficacy of TCbHP regimen (taxane, carboplatin, trastuzumab, and pertuzumab) in HER2 + breast cancer (BrCa) patients. Methods A total of 120 plasma samples from 40 patients with HER2 + BrCa were prospectively collected at three treatment times of neoadjuvant therapy (NAT) with TCbHP regimen. Serum metabolites were analyzed based on LC-MS and GC-MS data. Random forest was used to establish predictive models based on pre-therapeutic differentially expressed metabolites. Time series analysis was used to obtain potential monitors for treatment response. Transcriptome analysis was performed in nine available pre‑therapeutic specimens of core needle biopsies. Integrated analyses of metabolomics and transcriptomics were also performed in these nine patients. qRT-PCR was used to detect altered genes in trastuzumab-sensitive and trastuzumab-resistant cell lines. Results Twenty-one patients achieved pCR, and 19 patients achieved non-pCR. There were significant differences in plasma metabolic profiles before and during treatment. A total of 100 differential metabolites were identified between pCR patients and non-pCR patients at baseline; these metabolites were markedly enriched in 40 metabolic pathways. The area under the curve (AUC) values for discriminating the pCR and non-PCR groups from the NAT of the single potential metabolite [sophorose, N-(2-acetamido) iminodiacetic acid, taurine and 6-hydroxy-2-aminohexanoic acid] or combined panel of these metabolites were greater than 0.910. Eighteen metabolites exhibited potential for monitoring efficacy. Several validated genes might be associated with trastuzumab resistance. Thirty-nine altered pathways were found to be abnormally expressed at both the transcriptional and metabolic levels. Conclusion Serum-metabolomics could be used as a powerful tool for exploring informative biomarkers for predicting or monitoring treatment efficacy. Metabolomics integrated with transcriptomics analysis could assist in obtaining new insights into biochemical pathophysiology and might facilitate the development of new treatment targets for insensitive patients.

Abstract Background Deleterious BRCA1/2 (BRCA) mutation raises the risk for BRCA mutation-related malignancies, including breast, ovarian, prostate, and pancreatic cancer. Germline variation of BRCA exhibits substantial ethnical diversity. However, there is limited research on the Chinese Han population, constraining the development of strategies for BRCA mutation screening in this large ethnic group. Methods We profile the BRCA mutational spectrum, including single nucleotide variation, insertion/deletion, and large genomic rearrangements in 2,080 apparently healthy Chinese Han individuals and 522 patients with BRCA mutation-related cancer, to determine the BRCA genetic background of the Chinese Han population, especially of the East Han. Incident cancer events were monitored in 1,005 participants from the healthy group, comprising 11 BRCA pathogenic/likely pathogenic (PLP) variant carriers and 994 PLP-free individuals, including 3 LGR carriers. Results Healthy Chinese Han individuals demonstrated a distinct BRCA mutational spectrum compared to cancer patients, with a 0.53% (1 in 189) prevalence of pathogenic/likely pathogenic (PLP) variant, alongside a 3 in 2,080 occurrence of LGR. BRCA1 c. 5470_5477del demonstrated high prevalence (0.44%) in the North Han Chinese and penetrance for breast cancer. None of the 3 LGR carriers developed cancer during the follow-up. We calculated a relative risk of 135.55 (95% CI 25.07 to 732.88) for the development of BRCA mutation-related cancers in the BRCA PLP variant carriers (mean age 42.91 years, median follow-up 10 months) compared to PLP-free individuals (mean age 48.47 years, median follow-up 16 months). Conclusion The unique BRCA mutational profile in the Chinese Han highlights the potential for standardized population-based BRCA variant screening to enhance BRCA mutation-related cancer prevention and treatment.

Abstract Background Breast cancer is the most prevalent cancer among women globally, and surgical procedures continue to be the primary treatment. However, over 50% of patients experience preoperative anxiety due to the unknown and fear associated with surgery. Although drug therapy is commonly used to address this anxiety, its side effects have led to a heated debate regarding its effectiveness. Consequently, non-pharmacological therapies, such as preoperative education, have emerged as an alternative approach to alleviate anxiety. WeChat, a widely popular social media platform, offers a public platform that can potentially be utilized for effective preoperative education. This study aims to evaluate the use of WeChat public platform as a tool for preoperative education in patients undergoing breast surgery. Methods This is a prospective, randomized, and controlled trial will involve 392 adult women scheduled for breast cancer resection. Participants will be randomly assigned to either the WeChat education group or the regular group. In addition to regular preoperative visits, the WeChat education group will also watch science videos through the WeChat public platform. The regular group will only receive education from ward nurses during preoperative visits. The primary outcome measure will be the incidence of preoperative anxiety, defined by scores of the State Anxiety Inventory (SAI) exceeding 40 points. Secondary outcome measures include the incidence of severe anxiety (SAI > 44) on the day before surgery, incidence of anxiety 72 h after surgery, incidence of severe anxiety 72 h after surgery, NRS scores for pain at rest and during activity 24, 48, and 72 h after surgery, incidence of nausea and vomiting within 24 h after surgery, subjective sleep score at 1 week postoperatively, quality of life QoR-15 scores at 1 and 3 months postoperatively, incidence of chronic pain at 3 months postoperatively, bowel function recovery, length of hospital stay, and hospitalization expenses. Discussion This is the first clinical trial to investigate the use of WeChat public platform for delivering preoperative education on perioperative anxiety in breast cancer patients. By utilizing the renowned WeChat public platform, our study aims to improve patient outcomes by providing video education that explains the disease, surgery, and anesthesia in a more accessible manner, thereby reducing the incidence of perioperative anxiety. If our hypothesis is confirmed, this non-pharmacological approach can be universally acknowledged as a cost-effective and practical method in clinical care. Its application can also be extended to other medical fields beyond breast cancer. Trial registration ClinicalTrials.gov, NCT05291494. Registered on 29 December 2021.

An integrated computer software system for macromolecular crystallography (MX) data collection at the BL02U1 and BL10U2 beamlines of the Shanghai Synchrotron Radiation Facility is described. The system, Finback, implements a set of features designed for the automated MX beamlines, and is marked with a user-friendly web-based graphical user interface (GUI) for interactive data collection. The Finback client GUI can run on modern browsers and has been developed using several modern web technologies including WebSocket, WebGL, WebWorker and WebAssembly. Finback supports multiple concurrent sessions, so on-site and remote users can access the beamline simultaneously. Finback also cooperates with the deployed experimental data and information management system, the relevant experimental parameters and results are automatically deposited to a database.

There has been no study examining the effectiveness of an accessible computing curriculum for students with autism spectrum disorder (ASD) on their learning of computational thinking concepts (CTCs), flow control, data representation, abstraction, user interactivity, synchronization, parallelism, and logic. This study aims to investigate the effects of an accessible computing curriculum for students with ASD on their learning of CTCs as measured by the scores of 312 computing artifacts developed by two groups of students with ASD. Conducted among 21 seventh-grade students with ASD (10 in the experimental group and 11 in the control), this study involved collecting data on the computing projects of these students over 24 instructional sessions. Group classification was considered the independent variable, and computing project scores were set as the dependent variables. The results showed that the original curriculum was statistically significantly more effective for students in learning logic than the accessible one when all seven CTCs were examined as a single construct. Both curriculums were statistically significantly effective in progressively improving students’ learning of data representation, abstraction, synchronization, parallelism, and all CTCs as a single construct when examining the gradual increase in their computing artifact scores over the 24 sessions. Both curriculums were statistically significantly effective in increasing the scores of synchronization and all CTCs as a single construct when the correlations between CTCs and sessions for individual groups were analyzed. The findings underscore that students with ASD can effectively learn computing skills through accessible or standard curriculums, provided that adjustments are made during delivery.

Abstract Background The N-terminal regulatory element (NRE) of Receptor-like kinases (RLKs), consisting of the juxtamembrane segment in receptor kinases (RKs) and the N-terminal extension segment in RLCKs, is a crucial component that regulates the activities of these proteins. However, the features and functions of the NRE have remained largely unexplored. Herein, we comprehensively analyze 510,233 NRE sequences in RLKs from 528 plant species, using information theory and data mining techniques to unravel their common characteristics and diversity. We also use recombinant RKs to investigate the function of the NRE in vitro. Results Our findings indicate that the majority of NRE segments are around 40–80 amino acids in length and feature a serine-rich region and a 14-amino-acid consensus sequence, ‘FSYEELEKAT[D/N]NF[S/D]’, which contains a characteristic α-helix and ST motif that connects to the core kinase domain. This conserved signature sequence is capable of suppressing FERONIA’s kinase activity. A motif discovery algorithm identifies 29 motifs with highly conserved phosphorylation sites in RK and RLCK classes, especially the motif ‘VGPWKpTGLpSGQLQKAFVTGVP’ in LRR-VI-2 class. Phosphorylation of an NRE motif in an LRR-VI-2 member, MDIS1, modulates the auto-phosphorylation of its co-receptor, MIK1, indicating the potential role of NRE as a ‘kinase switch’ in RLK activation. Furthermore, the characterization of phosphorylatable NRE motifs improves the accuracy of predicting phosphorylatable sites. Conclusions Our study provides a comprehensive dataset to investigate NRE segments from individual RLKs and enhances our understanding of the underlying mechanisms of RLK signal transduction and kinase activation processes in plant adaptation.

The use of antibacterial dressings is crucial in the prevention and treatment of wound infection in emergency situations. However, the efficacy of dressings is compromised by long-term storage or exposure to harsh conditions. Here, an ultrastable in-situ silver nanoparticle dressing (AgSNP@CD) was prepared for effective prevention and treatment of wound infection in emergency. The fabrication process of AgSNP@CD is simple, suitable for large-scale production. Due to the strong interaction between the in-situ synthesized AgNPs and the cotton fabric, AgSNP@CD owned remarkable stability, thus retaining its antimicrobial efficacy even after long-term storage (up to 2 years) and under extreme conditions (damp heat, low temperatures, low-oxygen, water immersion, acid-alkali erosion). Both in vitro and in vivo results demonstrated the extraordinary antibacterial efficacy and stability of AgSNP@CD, facilitating infection prevention and wound healing in extreme conditions. In particular, AgSNP@CD exhibited a superior treatment effect on severe bacteria-infected trauma and can prevent the occurrence of sepsis effectively. The exceptional stability and antibacterial efficacy of AgSNP@CD under complex and extreme conditions make it a well-suited dressing strategy for the prevention and treatment of wound infection in emergency.

Cracking is an important factor affecting the durability of concrete. Rheological properties are important performance indicators of the workability of fresh concrete. Ion chelator (CA) is one kind of crystalline admixture, which can enhance crack-healing of concrete by promoting the formation of calcium carbonate in wet environment. Calcium alginate hydrogel (CaAlg) can maintain the humidity of cementitious matrix and promote the crystallization of inorganic minerals. Therefore, in this study, CaAlg bead was prepared to further improve the crack-healing of cementitious materials containing CA. Self-healing performance was evaluated by visual crack closure and water permeability test. Rheological property was measured by rotor rheometer. Healing mechanism of cementitious materials with CA and CaAlg was analyzed. Results showed that with the increase of CaAlg dosage, yield stress and plastic viscosity of paste decreased gradually. Meanwhile, fly ash and slag reduced viscosity of pastes, while silica fume increased the viscosity of pastes. CA could evidently improve the permeability and mechanical properties of mortar, especially for slag mortar. Self-healing performance test and characterization showed that CaAlg could further enhance crack-healing process of mortar with CA, especially for pure cement and slag mortar. Microscopic testing indicates that CaAlg could induce the formation of vaterite and calcite on its surface to further improve crack-healing efficiency.

Staphylococcal Enterotoxin B (SEB), produced by Staphylococcus aureus (S. aureus), is a powerful superantigen that induces severe immune disruption and toxic shock syndrome (TSS) upon binding to MHC-II and TCR. Despite its significant impact on the pathogenesis of S. aureus, there are currently no specific therapeutic interventions available to counteract the mechanism of action exerted by this toxin. In this study, we have identified a human monoclonal antibody, named Hm0487, that specifically targets SEB by single-cell sequencing using PBMCs isolated from volunteers enrolled in a phase I clinical trial of the five-antigen S. aureus vaccine. X-ray crystallography studies revealed that Hm0487 exhibits high affinity for a linear B cell epitope in SEB (SEB138–147), which is located distantly from the site involved in the formation of the MHC-SEB-TCR ternary complex. Furthermore, in vitro studies demonstrated that Hm0487 significantly impacts the interaction of SEB with both receptors and the binding to immune cells, probably due to an allosteric effect on SEB rather than competing with receptors for binding sites. Moreover, both in vitro and in vivo studies validated that Hm0487 displayed efficient neutralizing efficacy in models of lethal shock and sepsis induced by either SEB or bacterial challenge. Our findings unveil an alternative mechanism for neutralizing the pathogenesis of SEB by Hm0487, and this antibody provides a novel strategy for mitigating both SEB-induced toxicity and S. aureus infection.

Objective To determine the efficacy and safety of Astragalus combined with renin-angiotensin-aldosterone system (RAAS) blockers in treating stage III diabetic nephropathy (DN) by meta-analysis.Methods PubMed, Embase, Cochrane Library, Wiley, and Web of Science databases were searched for articles published between August 2007 and August 2022. Clinical studies on Astragalus combined with RAAS blockers for the treatment of stage III DN were included. Meta-analysis was performed by RevMan 5.1 and Stata 14.3 software.Results A total of 32 papers were included in this meta-analysis, containing 2462 patients from randomized controlled trials, with 1244 receiving the combination treatment and 1218 solely receiving RAAS blockers. Astragalus combined with RAAS blockers yielded a significantly higher total effective rate (TER) (mean difference [MD] 3.63, 95% confidence interval [CI] 2.59–5.09) and significantly reduced urinary protein excretion rate (UPER), serum creatinine (Scr), blood urine nitrogen (BUN) and glycosylated hemoglobin (HbAlc) levels. In subgroup analysis, combining astragalus and angiotensin receptor blocker significantly lowered fasting plasma glucose (FPG) and 24 h urinary protein (24hUTP) levels, compared with the combined astragalus and angiotensin-converting enzyme inhibitor treatment. Meanwhile, the latter significantly decreased the urinary microprotein (β2-MG). Importantly, the sensitivity analysis confirmed the study’s stability, and publication bias was not detected for UPER, BUN, HbAlc, FPG, or β2-MG. However, the TER, SCr, and 24hUTP results suggested possible publication bias.Conclusions The astragalus-RAAS blocker combination treatment is safe and improves outcomes; however, rigorous randomized, large-scale, multi-center, double-blind trials are needed to evaluate its efficacy and safety in stage III DN.

Given the global dominance of American movies, they play a significant role in presenting U.S. ideology and culture to the world, thus raising concern of undermining domestic culture. This paper investigates whether trade shapes culture by studying the impact of imported American movies on Chinese cultural values measured by Hofstede Index. The results show that demand for American movies has a positive and statistically significant effect on individualism, power distance, long-term orientation and indulgence. While many studies show evidence that cultural similarity stimulates trade, there is almost no empirical evidence on how international trade affects culture. This study contributes to the literatures related to the relationship between culture and trade by filling this void. The findings of this study also have strong policy implications by bringing empirical evidence to the debate on free trade of cultural products.

BackgroundRadioactive iodine (RAI) therapy is a widely used treatment for Graves’ Hyperthyroidism (GH). However, various factors can impact the non-remission rate of GH after single RAI therapy. This study aimed to develop an online dynamic nomogram to assist physicians in providing personalized therapy for GH.MethodsData from 454 GH patients who received RAI therapy were retrospectively reviewed and included in the present study. The univariate and multivariate analysis were conducted to investigate and identify independent influencing factors. The nomogram was developed based on the training cohort to explore non-remission rates. Finally, the reliability and accuracy of the constructed nomogram model were verified in the validation cohort via the calibration, receiver operating characteristic (ROC) curve, and decision curve analysis (DCA).Results24-hours radioactive iodine uptake (RAIU24h), effective half-life (Teff), total iodine dose (TID) and iodine dose per gram of thyroid tissue (IDPG) were independent predictors. The nomogram had a high C-index 0.922 (95% CI: 0.892–0.953), for predicting non-remission. The calibration curves demonstrated excellent consistency between the predicted and the actual probability of non-remission. ROC analysis showed that the AUC of the nomogram model and the four independent factors in the training cohort were 0.922, 0.673, 0.760, 0.761, and 0.786, respectively. The optimal cutoff value for the total nomogram scores was determined to be 155. A total score of ≥155 indicates a higher likelihood of non-remission after a single RAI therapy for GH, whereas a score below 155 suggests a greater likelihood of remission. Additionally, the DCA curve indicated that this nomogram had good clinical utility in predicting non-remission.ConclusionAn online nomogram was constructed with good predictive performance, which can be used as a practical approach to predict and assist physicians in making personalized therapy decisions for GH patients.

BackgroundAlzheimer's disease (AD) is a progressive and irreversible neurodegenerative disorder that has become one of the major health concerns for the elderly. Computer-aided AD diagnosis can assist doctors in quickly and accurately determining patients' severity and affected regions.MethodsIn this paper, we propose a method called MADNet for computer-aided AD diagnosis using multimodal datasets. The method selects ResNet-10 as the backbone network, with dual-branch parallel extraction of discriminative features for AD classification. It incorporates long-range dependencies modeling using attention scores in the decision-making layer and fuses the features based on their importance across modalities. To validate the effectiveness of our proposed multimodal classification method, we construct a multimodal dataset based on the publicly available ADNI dataset and a collected XWNI dataset, which includes examples of AD, Mild Cognitive Impairment (MCI), and Cognitively Normal (CN).ResultsOn this dataset, we conduct binary classification experiments of AD vs. CN and MCI vs. CN, and demonstrate that our proposed method outperforms other traditional single-modal deep learning models. Furthermore, this conclusion also confirms the necessity of using multimodal sMRI and DTI data for computer-aided AD diagnosis, as these two modalities complement and convey information to each other. We visualize the feature maps extracted by MADNet using Grad-CAM, generating heatmaps that guide doctors' attention to important regions in patients' sMRI, which play a crucial role in the development of AD, establishing trust between human experts and machine learning models.ConclusionWe propose a simple yet effective multimodal deep convolutional neural network model MADNet that outperforms traditional deep learning methods that use a single-modality dataset for AD diagnosis.

BackgroundThe incidence of severe infections (SIs) in patients with autoimmune nephropathy after rituximab (RTX) treatment varies significantly. Our study aims to identify high-risk populations, specifically by comparing the differences in the risk of SIs between patients with primary nephropathy and those with nephropathy in the context of systemic autoimmune diseases (referred to as secondary nephropathy).MethodsThis retrospective cohort study investigated the occurrence of SIs in adult patients with immune-related kidney disease who received RTX treatment at our institution from 2017 to 2022. Multivariable COX regression models were used to analyze the association between the type of nephropathy (primary or secondary) and SIs. Propensity score analyses, subgroup analyses, and E-value calculations were performed to ensure the reliability of the results.ResultsOut of 123 patients, 32 (26%) developed 39 cases of SIs during a mean follow-up period of 19.7 ± 14.6 months post-RTX treatment, resulting in an incidence rate of 18.9/100 patient-years. The multivariable COX regression analysis indicated that patients with secondary nephropathy had a significantly higher risk of SIs compared to those with primary nephropathy (HR = 5.86, 95% CI: 1.05–32.63, P = 0.044), even after accounting for confounding variables including gender, age, BMI, history of prior SIs, baseline eGFR, lymphocyte counts, IgG levels, and the utilization of other immunosuppressive therapies. Various sensitivity analyses consistently supported these findings, with an E-value of 5.99. Furthermore, advanced age (HR: 1.03; 95% CI: 1.01–1.06; P = 0.023), low baseline IgG levels (HR: 0.75; 95% CI: 0.64–0.89; P < 0.001), and recent history of SIs (HR: 5.68; 95% CI: 2.2–14.66; P < 0.001) were identified as independent risk factors.ConclusionThe incidence of SIs following RTX administration in patients with autoimmune nephropathy is significant. It is crucial to note that there are distinct differences between the subgroups of primary and secondary nephropathy. Patients with secondary nephropathy, particularly those who are elderly, have low baseline IgG levels, and have a recent history of SI, are more susceptible to SIs.

Abstract Corticotropin-releasing hormone-binding protein (CRHBP) is involved in many physiological processes. However, it is still unclear what role CRHBP has in tumor immunity and prognosis prediction. Using databases such as the Cancer Genome Atlas (TCGA), Gene Expression Omnibus (GEO), Tumor Protein Database, Timer Database, and Gene Expression Profiling Interactive Analysis (GEPIA), we evaluated the potential role of CRHBP in diverse cancers. Further research looked into the relationships between CRHBP and tumor survival prognosis, immune infiltration, immune checkpoint (ICP) indicators, tumor mutation burden (TMB), microsatellite instability (MSI), mismatch repair (MMR), DNA methylation, tumor microenvironment (TME), and drug responsiveness. The anticancer effect of CRHBP in liver hepatocellular carcinoma (LIHC) was shown by Western blotting, EdU staining, JC-1 staining, transwell test, and wound healing assays. CRHBP expression is significantly low in the majority of tumor types and is associated with survival prognosis, ICP markers, TMB, and microsatellite instability (MSI). The expression of CRHBP was found to be substantially related to the quantity of six immune cell types, as well as the interstitial and immunological scores, showing that CRHBP has a substantial impact in the TME. We also noticed a link between the IC50 of a number of anticancer medicines and the degree of CRHBP expression. CRHBP-related signaling pathways were discovered using functional enrichment. Cox regression analysis showed that CRHBP expression was an independent prognostic factor for LIHC. CRHBP has a tumor suppressor function in LIHC, according to cell and molecular biology trials. CRHBP has a significant impact on tumor immunity, treatment, and prognosis, and has the potential as a cancer treatment target and prognostic indicator.

Abstract Purpose The purpose of this study was to develop a functional clinical nomogram for predicting 8-year overall survival (OS) of patients with prostate cancer (PCa) primary based on peripheral lymphocyte. Patients and methods Using data from a single-institutional registry of 94 patients with PCa in China, this study identified and integrated significant prognostic factors for survival to build a nomogram. The discriminative ability was measured by concordance index (C-index) and ROC curves (Receiver Operating Characteristic Curves). And the predictive accuracy was measured by the calibration curves. Decision curve analyses (DCA) was used to measure the clinical usefulness. Results A total of 94 patients were included for analysis. Five independent prognostic factors were identified by LASSO-Cox regression and incorporated into the nomogram: age, the T stage, the absolute counts of peripheral CD3(+)CD4(+) T lymphocytes, CD3(-)CD16(+)CD56(+) NK cells and CD4(+)/CD8(+) ratio. The area under the curve (AUC) values of the predictive model for 5-, 8-, and 10-year overall survival were 0.81, 0.76, and 0.73, respectively. The calibration curves for probability of 5-,8- and 10-year OS showed optimal agreement between nomogram prediction and actual observation. The stratification into different risk groups allowed significant distinction. DCA indicated the good clinical application value of the model. Conclusion We developed a novel nomogram that enables personalized prediction of OS for patients diagnosed with PCa. This finding revealed a relative in age and survival rate in PCa, and a more favorable prognosis in patients exhibiting higher levels of CD4 + T, CD4+/CD8 + ratio and CD3(-)CD16(+)CD56(+) NK cells specifically. This clinically applicable prognostic model exhibits promising predictive capabilities, offering valuable support to clinicians in informed decision-making process.

Background: Intelligent garments, a burgeoning class of wearable devices, have extensive applications in domains such as sports training and medical rehabilitation. Nonetheless, existing research in the smart wearables domain predominantly emphasizes sensor functionality and quantity, often skipping crucial aspects related to user experience and interaction. Methods: To address this gap, this study introduces a novel real-time 3D interactive system based on intelligent garments. The system utilizes lightweight sensor modules to collect human motion data and introduces a dual-stream fusion network based on pulsed neural units to classify and recognize human movements, thereby achieving real-time interaction between users and sensors. Additionally, the system in- corporates 3D human visualization functionality, which visualizes sensor data and recognizes human actions as 3D models in realtime, providing accurate and comprehensive visual feedback to help users better understand and analyze the details and features of human motion. This system has significant potential for applications in motion detection, medical monitoring, virtual reality, and other fields. The accurate classification of human actions con- tributes to the development of personalized training plans and injury prevention strategies. Conclusions: This study has substantial implications in the domains of intelligent garments, human motion monitoring, and digital twin visualization. The advancement of this system is expected to propel the progress of wearable technology and foster a deeper comprehension of human motion.

Electric-drive-reconstructed on-board chargers (EDROCs) have emerged as a viable solution to alleviate mileage anxiety in electric vehicles (EVs) over the past decade. Nevertheless, the inclusion of additional components and separate control strategies in the previous EDROCs undeniably results in an escalation of system cost and control complexity. To address such an issue, an enhanced dual-vector model predictive current integrated control (MPCIC) for EDROCs is proposed, which is designed for the solar-powered electric vehicle (SPEV) that is equipped with a symmetrical six-phase machine. Firstly, the proposed topology implements the integration of charging and driving operations by re-leveraging the inherent traction system, significantly minimizing the incremental components. Secondly, the integrated control strategy for all operation modes is devised by incorporating the enhanced dual-vector model predictive control (MPC), mitigating the complexities and enhancing reliability. In this enhanced MPC, 12 virtual vectors, which consist of two active vectors, along with the optimized cost function are introduced, effectively suppressing harmonic currents. To this end, the effectiveness of the proposed EDROC is verified through a 2-kW experimental prototype under different operation modes.

Aluminum sulfate was used as a demulsifier to demulsify the lad-made emulsion of wire-drawing oil with different concentration. After demulsification，the COD，turbidity and pH value were measured，and the removal rate were calculated. Then，the demulsification effect of the aluminum sulfate alone and the composite demulsifier of aluminum sulfate-poly aluminum chloride were examined. The results show that the aluminum sulfate possesses the more stronger demulsification in several demulsifiers，the COD and turbidity in its treated water layer reached 70% and 60% respectively. However，the use of aluminum sulfate alone for the waste emulsion of wire-drawing oil of medium and high concentration from actual production showed poor performance，due to the emulsifying waste oil and water could not be layered. The composite demulsifier of aluminum sulfate and poly aluminum chloride，exhibited more effective demulsification on this waste. After stirring for 10 min，the removal rate of COD and turbidity of the medium concentration of waste emulsion reached 98% and 91% respectively，while 99% and 97% respectively for the waste of high concentration waste.

Abstract Correction of leg length discrepancy (LLD) in skeletally mature patients with osteosarcoma was rarely reported and quite challenging. This study aimed to propose a treatment strategy of staged lengthening and reconstruction with a standard static prosthesis to address LLD and restore limb function. It also evaluated the effectiveness of the strategy in terms of leg lengthening, functional outcomes, and complications. The strategy for lengthening included three stages. In stage 1, the previous prosthesis was removed and an external fixator with a temporary rod-cement spacer was placed. In this stage, the external fixator was used to lengthen the limb to the appropriate length. In stage 2, the external fixator was removed and the old rod-cement spacer was replaced with a new one. In stage 3, the rod-cement spacer was removed and the standard static prosthesis was planted. Nine skeletally mature distal femoral osteosarcoma patients with unacceptable LLD were treated in our institution from 2019 to 2021. We performed a chart review on nine patients for the clinical and radiographic assessment of functional outcomes, LLD, and complications. The mean (range) leg lengthening was 7.3 cm (3.6–15.6). The mean (range) LLD of the lower limbs decreased from 7.6 cm (4.1–14.2) before the lengthening to 0.3 cm (− 0.3 to 2.1) at the final follow-up with statistical significance (P = 0.000). The mean (range) Musculoskeletal Tumor Society score improved from 30.3% (16.7%–53.3%) before the lengthening to 96.3% (86.7%–100%) at the final follow-up with statistical significance (P = 0.000). Three patients (33.3%) had a minor complication; none needed additional surgical intervention. In the short term, the current staged lengthening and reconstruction with standard static prosthesis provided satisfactory functional outcomes and LLD correction with few complications. The long-term effects of this method need further exploration.

Abstract Point cloud registration is a multifaceted problem that involves a series of procedures. Many deep learning methods employ complex structured networks to achieve robust registration performance. However, these intricate structures can amplify the challenges of network learning and impede gradient propagation. To address this concern, the soft-hard correspondence (SHC) framework is introduced in the present paper to streamline the registration problem. The framework encompasses two modes: the hard correspondence mode, which transforms the registration problem into a correspondence pair search problem, and the soft correspondence mode, which addresses this new problem. The simplification of the problem provides two advantages. First, it eliminates the need for intermediate operations that lead to error fusion and counteraction, thereby improving gradient propagation. Second, a perfect solution is not necessary to solve the new problem, since accurate registration results can be achieved even in the presence of errors in the found pairs. The experimental results demonstrate that SHC successfully simplifies the registration problem. It achieves performance comparable to complex networks using a simple network and can achieve zero error on datasets with perfect correspondence pairs.

Accurate power load forecasting is conducive to ensuring the safe and economic operation of the power system. Aiming at the problems of low prediction accuracy and long time consuming of the current prediction algorithms, an improved deep learning (DL) short-term load forecasting model based on random forest (RF) algorithm and rough set theory (RST), namely RF-DL-RST, was proposed. Firstly, based on historical data, the model used RF algorithm to extract the key features that affected the load forecasting. Then, the key features and historical load data were trained as the input and output items of deep neural network (DNN), and the prediction results were corrected by RST. After that, the rough set method was used to revise the prediction results. Finally, the simulation was verified by an example. The results show that the prediction accuracy of the model is higher than that of a single DNN model and a model without RST revised.

Abstract Patients who receive heart valve surgery need anticoagulation prophylaxis to reduce the risk of thrombosis. Warfarin often is a choice but its dosage varies due to gene and clinical factors. We aim to study, among them, if there is an interaction between acute kidney injury and two gene polymorphisms from this study. We extracted data of heart valve surgery recipients from the electronic health record (EHR) system of a medical center. The primary outcome is about the average daily dose of warfarin, measured as an additive interaction effect (INTadd) between acute kidney injury (AKI) and warfarin-related gene polymorphisms. The confounders, including age, sex, body surface area (BSA), comorbidities (i.e., atrial fibrillation [AF], hypertension [HTN], congestive heart failure [CHF]), serum albumin level, warfarin-relevant gene polymorphism (i.e., CYP2C9, VKORC1), prosthetic valve type (i.e., metal, bio), and warfarin history were controlled via a multivariate-linear regression model. The study included 200 patients, among whom 108 (54.00%) are female. Further, the mean age is 54.45 years, 31 (15.50%) have CHF, and 40 (20.00%) patients were prescribed concomitant amiodarone, which potentially overlays with the warfarin prophylaxis period. During the follow-up, AKI occurred in 30 (15.00%) patients. VKORC1 mutation (1639G>A) occurred in 25 (12.50%) patients and CYPC29 *2 or *3 mutations presented in 20 patients (10.00%). We found a significant additive interaction effect between AKI and VKORC1 (− 1.17, 95% CI − 1.82 to − 0.53, p = 0.0004). This result suggests it is probable that there is an interaction between acute kidney injury and the VKORC1 polymorphism for the warfarin dose during the initial period of anticoagulation prophylaxis.

In recent years, crop yield prediction has become a research hotspot in the field of agricultural science, playing a decisive role in the economic development of every country. Therefore, accurate and timely prediction of crop yields is of great significance for the national formulation of relevant economic policies and provides a reasonable basis for agricultural decision-making. The results obtained through prediction can selectively observe the impact of factors such as crop growth cycles, soil changes, and rainfall distribution on crop yields, which is crucial for predicting crop yields. Although traditional machine learning methods can obtain an estimated crop yield value and to some extent reflect the current growth status of crops, their prediction accuracy is relatively low, with significant deviations from actual yields, and they fail to achieve satisfactory results. To address these issues, after in-depth research on the development and current status of crop yield prediction, and a comparative analysis of the advantages and problems of domestic and foreign yield prediction algorithms, this paper summarizes the methods of crop yield prediction based on deep learning. This includes analyzing and summarizing existing major prediction models, analyzing prediction methods for different crops, and finally providing relevant views and suggestions on the future development direction of applying deep learning to crop yield prediction research.

The escalating impact of human activities has led to the accumulation of microplastics (MPs) and heavy metals in marine environments, posing serious threats to marine ecosystems. As essential components of oceanic ecosystems, large seaweeds such as Caulerpa lentillifera play a crucial role in maintaining ecological balance. This study investigated the effects of MPs and cadmium (Cd) on the growth, physiology, biochemistry, and Cd accumulation in C. lentillifera while elucidating the underlying molecular regulatory mechanisms. The results demonstrated that exposure to MPs alone significantly promoted the growth. In contrast, exposure to Cd either alone or in combination with MPs significantly suppressed growth by reducing stem and stolon length, bud count, weight gain, and specific growth rates. Combined exposure to MPs and Cd exhibited the most pronounced inhibitory effect on growth. MPs had negligible impact while Cd exposure either alone or combined with MPs impaired antioxidant defenses and exacerbated oxidative damage; with combined exposure being the most detrimental. Analysis of Cd content revealed that MPs significantly increased Cd accumulation in algae intensifying its toxic effects. Gene expression analysis revealed that Cd exposure down-regulated key genes involved in photosynthesis, impairing both photosynthetic efficiency and energy conversion. The combined exposure of MPs and Cd further exacerbated these effects. In contrast, MPs alone activated the ribosome pathway, supporting ribosomal stability and protein synthesis. Additionally, both Cd exposure alone or in combination with MPs significantly reduced chlorophyll B and soluble sugar content, negatively impacting photosynthesis and nutrient accumulation. In summary, low concentrations of MPs promoted C. lentillifera growth, but the presence of Cd hindered it by disrupting photosynthesis and antioxidant mechanisms. Furthermore, the coexistence of MPs intensified the toxic effects of Cd. These findings enhance our understanding of how both MPs and Cd impact large seaweed ecosystems and provide crucial insights for assessing their ecological risks.

Canine malignant melanoma (CMM) is highly aggressive and mostly located in the oral cavity. CMM is the predominant type of canine oral malignancy and shows striking homologies with human mucosal melanoma. In comparative oncology, canine oral melanomas (COMs), as spontaneous tumor models, have the potential to acquire a unique value as a translational model of rare human melanoma subtypes. This review aims to provide a comprehensive summary of targeted therapies for canine malignant melanoma and to enrich the field of comparative oncology. Following the PRISMA guidelines, a comprehensive literature search was conducted across databases for studies from 1976 to April 2024. Studies were selected based on their relevance to targeted treatments. A total of 30 studies met the inclusion criteria. Based on the treatment approaches, the studies were further categorized into immunotherapies, small molecule signaling inhibitors, indirect kinase inhibitors, and other alternative strategies. Some treatments have been shown to result in stable disease or partial response, accounting for 29% (monoclonal antibody) and 76.5% (micro-RNA therapies) in clinical trials. Moreover, in vitro experiments of small molecule inhibitors, including cell signaling inhibitors and indirect kinase inhibitors, have shown the potential to be an effective treatment option for the development of therapeutic strategies in canine malignant melanoma. The observed response in in vitro experiments of CMM (particularly the oral and certain cutaneous subtypes) to drugs used in the treatment of human melanoma underlines the resemblance to human melanoma, therefore supporting the notion that CMM may be a valuable model for understanding rare human melanoma subtypes and exploring potential therapeutic avenues in preclinical trials. Finally, this literature review serves as a valuable resource for the development of therapeutic strategies for CMM and highlights the potential for translating these findings to human cancer treatment.

Secure instant communication is an important topic of information security. A group chat is a highly convenient mode of instant communication. Increasingly, companies are adopting group chats as a daily office communication tool. However, a large volume of messages in group chat communication can lead to message overload, causing group members to miss important information. Additionally, the communication operator’s server may engage in the unreliable behavior of stealing information from the group chat. To address these issues, this paper proposes an attribute-based end-to-end policy-controlled signcryption scheme, aimed at establishing a secure and user-friendly group chat communication mode. By using the linear secret sharing scheme (LSSS) with strong expressive power to construct the access structure in the signcryption technology, the sender can precisely control the recipients of the group chat information to avoid message overload. To minimize computational cost, a signcryption step with constant computational overhead is designed. Additionally, a message-sending mechanism combining “signcryption + encryption” is employed to prevent the operator server from maliciously stealing group chat information. Rigorous analysis shows that PCE-EtoE can resist adaptive chosen-ciphertext attacks under the standard model. Simulation results demonstrate that our theoretical derivation is correct, and that the PCE-EtoE scheme outperforms existing schemes in terms of computational cost, making it suitable for group chat communication.

Seed germination is a pivotal stage in the plant life cycle, orchestrated by a myriad of internal and external factors, notably plant hormones. The underlying molecular mechanisms governing rice seed germination remain largely unelucidated. Herein, we uncover OsMBF1a as a crucial regulatory factor that employs a dual strategy to promote seed germination: positively activating genes involved in gibberellin (GA) biosynthesis pathways, while negatively regulating key genes responsible for abscisic acid (ABA) synthesis. Furthermore, OsMBF1a modulates the endogenous levels of ABA and GA in rice seeds, reinforcing its central role in the germination process. The expression of ZmMBF1a and ZmMBF1b, the homologous genes in maize, in rice seeds similarly affects germination, indicating the conserved functionality of MBF1 family genes in regulating seed germination. This study provides novel insights into the molecular mechanisms underlying rice seed germination and underscores the significance of MBF1 family genes in plant growth and development.

Traditional Chinese music had a long history, with rich and extensive aesthetic concepts. The integration of traditional music and aesthetic ideas has a significant impact on the formation of China’s traditional music system. This article explored the interactive influence of traditional music and aesthetic ideas, and elaborated on the musical ideas in traditional Chinese aesthetics. The content of musical aesthetic ideas of Confucianism, Taoism and Mohism was discussed, and the position of aesthetic ideas in traditional Chinese music was analyzed. By combing and analyzing the relevant literature, ancient books and historical materials, it is possible to understand the historical evolution and mutual influence of traditional music and aesthetic thought. Traditional aesthetic concepts not only played an important role in the development of Chinese music, but also played an undeniable role in promoting the development of Chinese music over thousands of years of history. This article aimed to explore the interaction between traditional music and aesthetic ideas, as well as the impact of traditional aesthetics on music, in order to promote the integration and development of traditional music and aesthetic ideas. In the evaluation of the interaction between traditional music and aesthetic thought, there may be a situation that the concept is too simplified. The relationship between traditional music and aesthetic thought is very complicated, involving many schools, historical periods and regions. It may not be able to fully cover all the influencing factors if it is only summarized by Confucianism, Taoism and Buddhism. It is suggested that the education and inheritance of traditional music and aesthetic ideas should be strengthened in music education. More traditional music artists and aesthetic theorists should be cultivated, so that they can pass on traditional music and aesthetic ideas to future generations and continue to develop and innovate in contemporary society.

The diurnal and nocturnal valve gaping behavior of the pearl oyster Pteria penguin was characterized through video analysis aided by open-source software. Juvenile pearl oysters were video-recorded for 48 hours under laboratory conditions, using commonly used surveillance hardware. The direct measuring of the distance changes between the two valve edges effectively revealed the pearl oysters’ tendency to open their valves wider at night-time, than during day-time. Moreover, the differences observed were also attained at the individual level. The description for the first time of this behavior has broader implications for the research in pearl oyster aquaculture. Moreover, it introduces a non-invasive tool for comprehensive assessment of adult oyster behavior.

IntroductionActivation of complement through the alternative pathway (AP) has a key role in the pathogenesis of IgA nephropathy (IgAN). We previously showed, by intraperitoneal injection of Lactobacillus casei cell wall extract (LCWE), C57BL/6 mice develop mild kidney damage in association with glomerular IgA deposition. To further address complement activity in causing glomerular histological alterations as suggested in the pathogenesis of IgAN, here we used mice with factor H mutation (FHW/R) to render AP overactivation in conjunction with LCWE injection to stimulate intestinal production of IgA.MethodsDose response to LCWE were examined between two groups of FHW/R mice. Wild type (FHW/W) mice stimulated with LCWE were used as model control.ResultsThe FHW/R mice primed with high dose LCWE showed elevated IgA and IgA-IgG complex levels in serum. In addition to 100% positive rate of IgA and C3, they display elevated biomarkers of kidney dysfunction, coincided with severe pathological lesions, resembling those of IgAN. As compared to wild type controls stimulated by the same high dose LCWE, these FHW/R mice exhibited stronger complement activation in the kidney and in circulation.DiscussionThe new mouse model shares many disease features with IgAN. The severity of glomerular lesions and the decline of kidney functions are further aggravated through complement overactivation. The model may be a useful tool for preclinical evaluation of treatment response to complement-inhibitors.

Urban construction generates significant amounts of construction residue soil. This paper introduces a novel soil stabilizer based on industrial waste to improve its utilization. This stabilizer is primarily composed of blast furnace slag (BFS), steel slag (SS), phosphogypsum (PG), and other additives, which enhance soil strength through physical and chemical processes. This study investigated the mechanical properties of construction residue soil cured with this stabilizer, focusing on the effects of organic matter content (Oo), stabilizer dosage (Oc), and curing age (T) on unconfined compressive strength (UCS). Additionally, water stability and wet–dry cycle tests of the stabilized soil were conducted to assess long-term performance. According to the findings, the UCS increased with the higher stabilizer dosage and longer curing periods but reduced with the higher organic matter content. A stabilizer content of 15–20% is recommended for optimal stabilization efficacy and cost-efficiency in engineering applications. The samples lost their strength when immersed in water. However, adding more stabilizers to the soil can effectively enhance its water stability. Under wet–dry cycle conditions, the UCS initially increased and then decreased, remaining lower than that of samples cured under standard conditions. The findings can provide valuable data for the practical application in construction residual soil stabilization.

A number of studies have reported that drug addiction is associated with microRNAs (miRNAs). However, the roles of plasma miRNAs in methamphetamine (METH) addicts have not been clearly explained. This study aimed to profile a panel of miRNAs as non-invasive predictive biomarkers and therapeutic targets for METH addiction. Differentially expressed miRNAs were derived from next-generation sequencing technology (NGS) and were validated by quantitative real-time PCR (RT-qPCR). The diagnostic value of specific altered miRNAs was evaluated by receiver operating characteristic (ROC) analysis and area under the curve (AUC). NGS results revealed that 63 miRNAs were significantly altered in the METH-exposed paradigm. The levels of hsa-miR-592, hsa-miR-9-3p, hsa-miR-206 and hsa-let-7b-3p were significantly elevated in the plasma of METH addicts. Hsa-miR-9-3p was a useful biomarker discriminating METH addicts from normal (AUC was 0.756). Importantly, combining detection of hsa-miR-592 and hsa-miR-9-3p achieved the highest AUC of 0.87, with a sensitivity and specificity of 82.7% and 78.9%, respectively. Target gene BDNF decreased significantly in METH addicts. Although METH addicts showed significant depressive symptoms, there was no correlation between the expression level of miR-592 and miR-9-3p and the degree of depression. Our findings suggested that hsa-miR-592, hsa-miR-9-3p, hsa-miR-206, and hsa-let-7b-3p may play a potential role in the pathology of METH addiction, and a combination of hsa-miR-592 and hsa-miR-9-3p could serve as potential peripheral biomarker and therapeutic target for METH addiction.

To address the challenges posed by the complex shapes of hollow parts, this study examined the ultrasonic-assisted granular medium hydroforming (UGMF) process for tubular components. The dynamics of the deformation behavior and deformation control during 6063-T5 aluminum alloy tube free forming by UGMF were studied via simulations and experiments. Based on the ABAQUS software platform, a coupled method based on finite element (FE) simulation analysis and discrete element (DE) analysis for the UGMF free forming process was used. The results showed that ultrasonic vibration (UV) could reduce the forming force required for expansion and promote the flow of material at the end to the forming area as well as inhibit the decrease in the wall thickness. The accuracy of the FE-DE coupled simulation model and a parabolic geometric model was verified by testing. The results found that UV enhances material flow, decreases the forming force needed, and minimizes damage to the granular surface.

Incorporating renewable energy sources into the grid poses challenges due to their volatility and uncertainty in optimizing dispatch strategies. In response, this article proposes a cloud–edge collaborative scheduling strategy for distributed multi-energy systems, operating across various time scales. The strategy integrates day-ahead dispatch, intra-day optimization, and real-time adjustments to minimize operational costs, reduce the wastage of renewable energy, and enhance overall system reliability. Furthermore, the cloud–edge collaborative framework helps mitigate scalability challenges. Crucially, the strategy considers the multi-timescale characteristics of two types of energy storage systems (ESSs) and three types of demand response (DR), aimed at optimizing resource allocation efficiently. Comparative simulation results evaluate the strategy, providing insights into the significant impacts of different ESS and DR types on system performance. By offering a comprehensive approach, this strategy aims to address operational complexities. It aims to contribute to the seamless integration of renewable energy into distributed systems, potentially enhancing sustainability and resilience in energy management.

Transcriptome analysis based on high-throughput sequencing of a cDNA library has been widely applied to functional genomic studies. However, the cDNA dependence of most RNA sequencing techniques constrains their ability to detect base modifications on RNA, which is an important element for the post-transcriptional regulation of gene expression. To comprehensively profile the N6-methyladenosine (m6A) and N5-methylcytosine (m5C) modifications on RNA, direct RNA sequencing (DRS) using the latest Oxford Nanopore Technology was applied to analyze the transcriptome of six tissues in rice. Approximately 94 million reads were generated, with an average length ranging from 619 nt to 1013 nt, and a total of 45,707 transcripts across 34,763 genes were detected. Expression profiles of transcripts at the isoform level were quantified among tissues. Transcriptome-wide mapping of m6A and m5C demonstrated that both modifications exhibited tissue-specific characteristics. The transcripts with m6A modifications tended to be modified by m5C, and the transcripts with modifications presented higher expression levels along with shorter poly(A) tails than transcripts without modifications, suggesting the complexity of gene expression regulation. Gene Ontology analysis demonstrated that m6A- and m5C-modified transcripts were involved in central metabolic pathways related to the life cycle, with modifications on the target genes selected in a tissue-specific manner. Furthermore, most modified sites were located within quantitative trait loci that control important agronomic traits, highlighting the value of cloning functional loci. The results provide new insights into the expression regulation complexity and data resource of the transcriptome and epitranscriptome, improving our understanding of the rice genome.
