Cell-specific aptamers offer a powerful tool to study membrane receptors at the single-molecule level. Most target receptors of aptamers are highly expressed on the cell surface, but difficult to analyze in situ because of dense distribution and fast velocity. Therefore, we herein propose a random sampling-based analysis strategy termed ligand dilution analysis (LDA) for easily implemented aptamer-based receptor study. Receptor density on the cell surface can be calculated based on a regression model. By using a synergistic ligand dilution design, colocalization and differentiation of aptamer and monoclonal antibody (mAb) binding on a single receptor can be realized. Once this is accomplished, precise binding site and detailed aptamer-receptor binding mode can be further determined using molecular docking and molecular dynamics simulation. The ligand dilution strategy also sets the stage for an aptamer-based dynamics analysis of two- and three-dimensional motion and fluctuation of highly expressed receptors on the live cell membrane.

As an early-stage tumor biomarker, microRNA (miRNA) has clinical application potential and its sensitive and accurate detection is significant for early tumor diagnosis. In this study, a photoelectrochemical (PEC) biosensing platform was fabricated for ultrasensitive miRNA-141 detection, which is based on a photocurrent polarity-switchable system using CdS quantum dots (QDs) in the presence of a 5,10,15,20-tetrakis (4-aminophenyl)-21H,23H-porphine (Tph-2H)-coated glassy carbon electrode (GCE). As an excellent photoactive material, Tph-2H has a narrow band gap that effectively gathers photoelectrons under visible light irradiation and improves the transfer ability of photogenerated electrons. Further, the detection sensitivity of miRNA-141 could be significantly improved by combining an enzyme-assisted recycle amplification reaction and a magnetic bead-based separation strategy. The proposed photocurrent polarity-switchable PEC biosensor could efficiently eliminate the false-positive or false-negative signals and achieve a wide linear response range from 1 fM to 1 nM with a low detection limit of 0.33 fM for miRNA-141, providing a potentially alternative solution for detecting other biomarkers in bioanalysis and clinical diagnosis.

A novel photoelectrochemical (PEC) biosensor based on an enzyme-free nucleic acid dual-amplification strategy combined with a mimic enzyme to catalyze the deposition of a quencher is reported for the ultrasensitive detection of miRNA-21. A limited amount of target miRNA-21 can trigger the formation of long DNA duplexes on the electrode, owing to the synergistic effect of the enzyme-free nucleic acid dual-amplification strategy of entropy-driven strand displacement reaction (ESDR) amplification and hybridization chain reaction (HCR) amplification. The embedded manganese porphyrin (MnPP) in the long DNA duplexes acts as a horseradish peroxidase (HRP)-mimicking enzyme to catalyze the transformation of benzo-4-chlorohexadienone on the electrode surface, resulting in a significant reduction in photocurrent intensity. As a photosensitive material, BiOCl-BiOI is used as a tag to provide strong initial PEC signals. Based on the cascade integration of the enzyme-free nucleic acid dual-amplification strategy and the mimic enzyme-catalyzed precipitation reaction, the current PEC biosensor exhibits outstanding performance for miRNA-21 detection with an ultralow detection limit (33 aM) and a wide quantification range (from 100 aM to 1 nM). This work provides a new avenue toward the ultrasensitive detection of miRNAs, and is expected to be used for clinical and biochemical samples. A unique PEC biosensor with the BiOCl-BiOI composite, as the photosensitive material, has been developed for ultrasensitive miRNA-21 determination based on the combination of an enzyme-free nucleic acid dual-amplification strategy and mimic enzyme catalytic precipitation reaction.

A novel fluorescent Cd-MOF probe [Cd2(btec)(TPB)0.5(H2O)4]·2H2O (complex 1, TPB = 1,2,4,5-Tetra(4-pyridyl)benzene) has been rationally designed and synthesized, and its multipurpose sensing applications have been revealed. The thermo-sensitive property of complex 1 was investigated in a wide temperature range from 20 °C to 160 °C. A good linear relationship was observed between the fluorescence quenching efficiency of complex 1 and the circumstance temperature, which suggested that complex 1 could be a promising material for application in fluorescent thermometers. Stable suspensions could be obtained after dispersion of complex 1 in various solvents, especially in water. A remarkable red shift (about 20 nm) in the fluorescence emission spectrum of complex 1 was observed when H2O was used as the dispersion medium compared with that in D2O, which could be used for distinguishing D2O from H2O. Furthermore, based on the strong inner filtering effect (IFE), the fluorescence detection of acetone in water was carried out using complex 1-H2O suspensions. Real-time fluorescence quenching was observed after the addition of acetone into complex 1-H2O suspensions. Moreover, the fluorescence quenching efficiency vs. the concentration of acetone fitted well with a non-linear equation (R2 = 0.9929) with an experimental detection limit for acetone as 0.06% (vol%). In addition, the photoelectric responses study revealed that complex 1 might be modified and used as a photoelectric sensor.

In this research, a hydroxyl group functionalized metal-organic framework (MOF), UiO-66-(OH)2, was synthesized as a “on-off-on” fluorescent switching nanoprobe for highly sensitive and selective detection of Fe3+, ascorbic acid (AA) and acid phosphatase (ACP). UiO-66-(OH)2 emits yellow-green light under ultraviolet light, when Fe3+ was added, Fe3+ was chelated with hydroxyl group, the electrons in the excited state S1 of the MOF transferred to the half-filled 3d orbits of Fe3+, resulting in fluorescence quenching because of the nonradiative electron/hole recombination annihilation. AA could reduce Fe3+ to Fe2+, which can destroy the electron transfer between UiO-66-(OH)2 and Fe3+ after AA adding, resulted in nonoccurrence of the nonradiative electron transfer, leading to the recovery of UiO-66-(OH)2 fluorescence intensity. The probe can also be used to detect ACP based on the enzymolysis of 2-phospho- l -ascorbic acid (AAP) to produce AA. Benefitting from the hydroxyl group and the characteristics of UiO-66, including the high porosity and large surface area, the developed UiO-66-(OH)2 showed extensive advantages as a fluorescent probe for detection of multi-component, such as high sensitivity and selectivity, colorimetric detection, fast response kinetics and easy to operate, economical and secure. This is the first time to use active group functionalized MOFs as a multi-component sensor for these three substances detection.

Cancerous exosomes that carry multiple biomarkers are attractive targets for the early diagnosis and therapy of cancer. As one of the powerful molecular recognition tools, aptamers with excellent binding affinity and specificity toward biomarkers have been exploited to construct various aptamer-based biosensors (aptasensors) for exosome detection. Here, we review recent advances in aptasensors for the detection of cancerous exosomes. We first discuss the importance and potential of cancerous exosomes in cancer diagnosis and then summarize some conventional aptasensors from the perspective of biomarker recognition and signal collection strategies. Finally, we comment on the outlook for aptasensor research and new directions for cancerous exosome detection.

Photoelectrochemical (PEC) biosensors have shown great promise in bioanalysis and diagnostic applications in recent years. In this work, the CuO/Cu2O nanowire array (CuO/Cu2O Nanowire) supported on copper foam was prepared as a photocathode for detection of tyrosinase though quinone-chitosan conjugation chemistry method. The in-situ generated quinones that were the catalytic product of tyrosinase acted as electron acceptors, which were captured by the chitosan deposited on the surface of the electrode. Direct immobilization of electron acceptor on the electrode surface improved the photocurrent conversion efficiency and thus sensitivity. The as-prepared biosensor can realize a rapid response in a wide linear range of 0.05 U/mL to 10 U/mL with the detection limit as low as 0.016 U/mL of tyrosinase. The current work provides a new perspective to design and develop highly sensitive and selective PEC biosensor.

Mercury detection remains an important task because of its high toxicity. Herein a new dual-signal probe based on a boric acid (BA)-functionalized lanthanide metal-organic framework (BA-Eu-MOF) was developed for the detection of Hg2+ and CH3Hg+ ions for the first time. The BA-Eu-MOF was synthesized by coordination of Eu3+ with 5-boronobezene-1, 3-dicarboxylic acid (5-bop) through a one-pot method. The 5-bop ligand not only acted as the "antenna" to sensitize the luminescence of Eu3+ but also provided reaction sites for Hg2+ and CH3Hg+. Owing to the electron-withdrawing effect of the BA group, the "antenna" effect of the ligand was passivating and the BA-Eu-MOF showed weak red emission in water. Upon addition of Hg2+ or CH3Hg+ into the system, a transmetalation reaction took place, i.e., BA groups were replaced by Hg2+ or CH3Hg+; therefore, the "antenna" effect of the ligand was triggered, leading to the enhancement of red emission. As Hg2+ or CH3Hg+ concentration increased, the red emission was gradually enhanced, and the color change was also observed with the naked eye under 365 nm ultraviolet light. Owing to the porous characteristics and the surface effect of the MOF, as well as the unique transmetalation reaction between the BA group and Hg2+ or CH3Hg+, the developed nanoprobe showed excellent characteristics for simultaneous detection of Hg2+ and CH3Hg+, such as simple preparation, convenient operation, "turn-on" signal output, high sensitivity, and selectivity. The unique features of the BA-Eu-MOF make it an attractive probe for monitoring Hg2+ and CH3Hg+.

Herein, a dual-emission metal-organic framework based ratiometric fluorescence nanoprobe was reported for detecting copper(II) ions. In particular, carbon dots (CDs) and gold nanoclusters (AuNCs) were embedded into ZIF-8 (one of the classical metal-organic frameworks) to form CDs/AuNCs@ZIF-8 nanocomposites, which exhibited dual-emission peaks at UV excitation. In the presence of Cu2+, the fluorescence attributed to AuNCs can be rapidly quenched, while the fluorescence of CDs serves as reference with undetectable changes. Therefore, the CDs/AuNCs@ZIF-8 nanocomposites were utilized as a ratiometric fluorescence nanoprobe for sensitive and selective detection of Cu2+. A good linear relationship between the ratiometric fluorescence signal of CDs/AuNCs@ZIF-8 and Cu2+ concentration was obtained in the range of 10−3–103 μM, and the detection limit was as low as 0.3324 nM. The current ratiometric fluorescence nanoprobe showed promising prospects in cost-effective and rapid determination of Cu2+ ions with good sensitivity and selectivity. Furthermore, this nanoprobe has been successfully applied for the quantitative detection of Cu2+ in serum samples, indicating its value of practical application.

A sensitive photoelectrochemical (PEC) aptasensor was constructed for prostate-specific antigen (PSA) detection using an enhanced photocurrent response strategy. The p-n heterostructure CdS-Cu2O nanorod arrays were prepared on Ti mesh (CdS-Cu2O NAs/TM) by a simple hydrothermal method and successive ionic-layer adsorption reactions. Compared with the original CdS/TM, the synergistic effect of p-n type CdS-Cu2O NAs/TM and the internal electric field realizes the effective separation of photoinduced electron–hole pairs and improves the PEC performance. In order to construct the aptasensor, an amino-modified aptamer was immobilized on CdS-Cu2O NAs/TM to serve as a recognition unit for PSA. After the introduction of PSA, PSA was specifically captured by the aptamer on the PEC aptasensor, which can be oxidized by photogenerated holes to prevent electron–hole recombination and increase photocurrent. Under optimal conditions, the constructed PEC aptasensor has a linear range of 0.1–100 ng·mL−1 and a detection limit as low as 0.026 ng·mL−1. The results of aptasensor detection of human serum indicate that it has broad application prospects in biosensors and photoelectrochemical analysis.

We report the facile synthesis of gold–silver (Au–Ag) nanostructures in a niche hybrid form of ultrathin wire–dots, where the ultrathin nanowires are embedded with quasi-spherical dots along the longitudinal direction. The key to the success of this work relies on the use of seeded growth and the introduction of excessive Ag ions in the growth stage to facilitate the formation of such a unique architecture. Several reaction parameters, including the amount of silver and reductant, seed quantity and size/morphology, are systematically investigated to reveal their role in determining the morphology of the final products. We also recorded their UV-vis extinction spectra and compared them with pure ultrathin wires to investigate the effect of dot regions on the optical properties.

After being functionalized with His, the resultant His@ZIF-8 had abundant carboxyl and amino groups, which like tongs help His@ZIF-8 “grasp” Tb3+ firmly to form a stable lanthanide-doped nanoparticle (His@ZIF-8/Tb3+). Owing to the unsaturated coordination of Tb3+ with the amino acid group, the resultant His@ZIF-8/Tb3+ showed reserved response sites of Tb3+ to DPA because of its unique molecular structure. After the His@ZIF-8/Tb3+ coordination with DPA, the intrinsic fluorescence emission of the Tb3+ ions was triggered through energy transfer, leading to bright yellow green luminescence owing to the antenna role of DPA. Benefitting from the His functionalization and the characteristics of ZIF-8, especially the high porosity and large surface area, the developed His@ZIF-8/Tb3+ sensing platform exhibited attractive features as a fluorescent sensor for monitoring DPA such as fast response kinetics (10 s), high sensitivity and selectivity, and being portable, easy to operate, economical and secure. This sensor platform showed a satisfactory linear relationship (R2 = 0.999) ranging from 0.08 to 10 μmol L−1 and an ultralow limit of detection (LOD) of 0.02 μmol L−1. This strategy for the design of functionalized MOFs to construct sensing probes and the resultant His@ZIF-8/Tb3+ would provide a potential strategy for the exploitation of other functionalized materials used in other research fields and promising fluorescence platforms for the detection of other targets.

Although recent studies on the oxygen evolution reaction (OER) have achieved great progress, it remains a great challenge to achieve a smaller overpotential at high current density. In order to further improve the performance of the OER, we adopted a novel method to load Fe nanoparticles on a Ni3S2 nanoplate array grown on nickel foam (Fe NPs-Ni3S2 NA/NF) which formed an amorphous structure. In particular, this catalyst exhibited an overpotential of 206 mV at 150 mA cm−2, outmatching most non-noble OER catalysts reported so far; it also operates smoothly in various stability tests.

A novel "turn-on" fluorescent trypsin detection platform dependant on carbon dot-MnO

Surface modification of inorganic nanomaterials with biomolecules has enabled the development of composites integrated with extensive properties. Lanthanide ion-doped upconversion nanoparticles (UCNPs) are one class of inorganic nanomaterials showing optical properties that convert photons of lower energy into higher energy. Additionally, DNA oligonucleotides have exhibited powerful capabilities for organizing various nanomaterials with versatile topological configurations. Through rational design and nanotechnology, DNA-based UCNPs offer predesigned functionality and potential. To fully harness the capabilities of UCNPs integrated with DNA, various DNA-UCNP composites have been developed for diagnosis and therapeutics. In this review, beginning with the introduction of the UCNPs and the conjugation of DNA strands on the surface of UCNPs, we present an overview of the recent progress of DNA-UCNP composites while focusing on their applications for bioanalysis and therapeutics.

Herein, an efficient electrochemical sensing platform is proposed for selective and sensitive detection of nitrite on the basis of Cu@C@Zeolitic imidazolate framework-8 (Cu@C@ZIF-8) heterostructure. core–shell Cu@C@ZIF-8 composite was synthesized by pyrolysis of Cu-metal-organic framework@ZIF-8 (Cu-MOF@ZIF-8) in Ar atmosphere on account of the difference of thermal stability between Cu-MOF and ZIF-8. For the sensing system of Cu@C@ZIF-8, ZIF-8 with proper pore size allows nitrite diffuse through the shell, while big molecules cannot, which ensures high selectivity of the sensor. On the other hand, Cu@C as electrocatalyst promotes the oxidation of nitrite, thereby resulting high sensitivity of the sensor. Accordingly, the Cu@C@ZIF-8 based sensor presents excellent performance for nitrite detection, which achieves a wide linear response range of 0.1–300.0 μM, and a low limit of detection of 0.033 μM. In addition, the Cu@C@ZIF-8 sensor possesses excellent stability and reproducibility, and was employed to quantify nitrite in sausage samples with recoveries of 95.45%–104.80%.

Facile and sensitive determination of formaldehyde (FA) in indoor environments still remains challenging. Herein, a fluorescent probe, termed PHN@MOF, was synthesized by embedding the fluorescent molecule of N-propyl-4-hydrazine-naphthalimide (PHN) into a metal-organic framework (MOF) for sensitive and visual monitoring of FA. The hydrazine group of PHN acts as the specific reaction group with FA based on the condensation reaction. The host of MOF (UiO-66-NH2) offers the surrounding confinement space required for the reaction. Owing to the enrichment effect and molecular sieve selection of UiO-66-NH2 to FA, PHN@MOF, compared with free PHN, exhibits very high sensitivity and selectivity based on space confinement-induced sensitivity enhancement (SCISE). Moreover, the fluorescence of UiO-66-NH2 offers a reference signal for FA detection. Using this ratiometric fluorescent PHN@MOF probe, a colorimetric gel plate and test paper were developed and used to visually monitor FA in air.

β-Glucuronidase (β-GCU) is closely related to the occurrence of multiple diseases, and it has been applied as a biomarker and therapeutic target in clinical diagnosis. However, reliable methods with high selectivity and sensitivity for monitoring β-GCU are still lacked. Herein, we designed a novel fluorescent nanoprobe, rhodamine B encapsulated MOF-5 (RhB@MOF-5) for the first time, to detect β-GCU through the synergistic effect of inner filter effect (IFE) and static quenching effect (SQE) by employing 4-nitrophenyl-β-D-glucuronide (PNPG) as the substrate. After encapsulating into MOF-5, the fluorescence emission of RhB at 550 nm excited by 320 nm was greatly enhanced. The major overlap between the fluorescence excitation spectrum of RhB@MOF-5 (about 320 nm) and the ultraviolet absorption spectrum of PNPG (about 310 nm) lead to PNPG being a good IFE absorber in this sensing system. Under the optimized conditions, the excitation spectrum of the RhB@MOF-5 could be absorbed by PNPG, resulting in the dramatically decrease of fluorescence emission. After adding β-GCU into the system, the substrate of PNPG would be enzymatic hydrolyzed to p-nitrophenol (PNP) and glucose, then the IFE disappearance and the fluorescence recovered. The current sensing platform was interference-free and exhibited a broad linearity relationship for β-GCU range of 0.1–10 U L−1 (R2 = 0.9957) with a limit of detection as low as 0.03 U L−1, which was reduced by more than one orders compared with the reported methods. Moreover, the encapsulation of dyes using porous nanoparticles to achieve some tailor-made characteristics will enrich experimental design inspiration.

Ferric ion (Fe3+) plays a vital role in cellular homeostasis. However, the detection of Fe3+ with rhodamine B (RhB) has potential problems, such as poor selectivity and low photostability. To address these problems, we rationally designed an RhB@MOF nanocomposite-based "on-off-on" fluorescent switching nanoprobe for highly sensitive and selective detection of Fe3+ and ascorbic acid. This RhB@MOF nanoprobe was prepared through a facile one-pot synthesis. Here MOF served as a selectivity regulator for the detection of Fe3+. By embedding RhB into the porous crystalline MOF, enhanced photostability and fluorescence lifetime of RhB to Fe3+ were achieved. The as-prepared RhB@MOF was demonstrated to be an ultrasensitive and selective nanoprobe for the detection of Fe3+ in human serum and ascorbic acid in rat brain microdialysate. Furthermore, inner filter effect (IFE) and photoinduced electron transfer (PET) were proposed and discussed to explain the selectivity and sensitivity of RhB to Fe3+ against other interfering substances. Our novel "on-off-on" nanoprobe provides insight into the rational design of MOF-based biosensors for selective and sensitive detection of analytes.

The composite of metal-organic frameworks (MOFs) and graphene aerogels (GAs) is getting growing attention in the field of electrochemical sensor owing to its interconnected meso-/macro-porous structure, high conductivity, and large surface area. How to effectively compound the two components in a simple way and maximize the performance advantages of materials is still a challenge. In this paper, a simple in-situ growth method was proposed by using polypyrrole (PPy) as a crosslinking agent for the synthesis of hierarchically porous PPy@ZIF-8/GAs. Wherein, PPy was initially decorated on GAs and then ZIF-8 nanocrystals were orderly grown on the surface of PPy/GAs through the coordination interaction between Zn2+ and amino ( NH ) groups in PPy chains. This hybrid 3D architecture provided open channels for electrolyte transportation and improved electron transfer between the electrode and the catalysts, which was exploited as electrochemical sensing platform for the detection of highly toxic 2, 2-methylenebis (4-chlorophenol) (dichlorophenol, Dcp). Benefiting from the synergistic effects between PPy, GAs and ZIF-8, the PPy@ZIF-8/GAs electrode showed high sensitivity for Dcp detection with a low detection limit down to 0.1 nM. In addition, the current sensor presented several merits, such as good reproducibility, high selectivity and long stability. More importantly, the sensor was successfully applied for the detection of Dcp in lake water samples.

Industrial production of NH3 from N2 and H2 significantly relies on Haber–Bosch process, which suffers from high energy consume and CO2 emission. As a sustainable and environmentally-benign alternative process, electrochemical artificial N2 fixation at ambient conditions, however, is highly required efficient electrocatalysts. In this study, we demonstrate that hexagonal boron nitride nanosheet (h-BNNS) is able to electrochemically catalyze N2 to NH3. In acidic solution, h-BNNS catalyst attains a high NH3 formation rate of 22.4 μg·h–1·mg–1cat. and a high Faradic efficiency of 4.7% at–0.75 V vs. reversible hydrogen electrode, with excellent stability and durability. Density functional theory calculations reveal that unsaturated boron at the edge site can activate inert N2 molecule and significantly reduce the energy barrier for NH3 formation.

We present a fluorescence turn-on and colorimetric dual-readout sensing system for the sensitive detection of alkaline phosphatase (ALP) activity via self-assembled gold nanoclusters (AuNCs) based on fluorescence resonance energy transfer (FRET). The positively charged polyallylamine hydrochloride (PAH)-crosslinked AuNCs (PAH-AuNCs) with aggregation-induced enhancement (AIE) characteristics can electrostatically adsorb the negatively charged 2, 6-dichlorophenolindophenol (DCIP). Thus, the fluorescence of PAH-AuNCs can be significantly quenched by the occurrence of FRET from PAH-AuNCs to DCIP. However, the reduction reaction of DCIP from blue to colourless by L -ascorbic acid (AA) which is generated by the ALP catalyse hydrolysis of 2-Phospho- L -ascorbic acid (AAP) disturbs the FRET between PAH-AuNCs to DCIP. The quenched PAH-AuNCs fluorescence can be recovered efficiently. The strategy of first creating AIE-enhanced PAH-AuNCs, followed by the effective FRET manipulation, is an important contribution to the sensitive detection of ALP. More importantly, the distinct colorimetric signal change can be used to visually distinguish the presence of ALP. Good linear relationships of fluorescence and colorimetric sensing towards ALP were obtained in the range from 0.5 to 100 U/L, and the detection limits were 0.2 U/L and 0.5 U/L, respectively. In addition, the proposed FRET sensing system was applied to the detection of ALP in human serum samples with satisfactory results. The simple and efficient sensing approach proposed here has the potential to promote the development of chemo/biodetection methods using fluorescence and colorimetric dual-readout.
