Increasing local people and indigenous communities' participation on restoration programs is widely recommended as a way to increase restoration long‐term benefits and local people's commitment with restored lands. Here, we demonstrate how traditional knowledge can provide crucial information on usefulness of plant species that increases direct value of forest and landscape restoration programs. Our study analyzed secondary data from 73 studies that assessed indigenous and local knowledge regarding plant's utility across the densely populated dry forest of the Brazilian Caatinga. Our results suggest that highly valued plants represent a relatively small number of useful species (52 out of 632 useful species) that account for more than half of the plant services (i.e. functional space represented by the uses people give to plants). Interestingly, these plants belonged to closely related lineages, suggesting that people must select plants based on phylogenetically conserved traits that provide utility. We propose that accounting for indigenous and local knowledge in early phases of restoration process such as species selection can both maximize classical restoration targets (e.g. species diversity) and increase delivery of ecosystem services to forest‐dependent people. Such an interdisciplinary integration moves us a step closer to ambitious targets set for global ecosystem restoration based on fair and inclusive participation of indigenous people and local communities. Excitable temperament disrupts physiological events required for reproductive development in cattle, but no research has investigated the impacts of temperament on growth and puberty attainment in Bos indicus females. Hence, this experiment evaluated the effects of temperament on growth, plasma cortisol concentrations and puberty attainment in B. indicus heifers. A total of 170 Nelore heifers, weaned 4 months before the beginning of this experiment (days 0 to 91), were managed in two groups of 82 and 88 heifers each (mean ± SE; initial BW=238±2 kg, initial age=369±1 days across groups). Heifer temperament was evaluated via exit velocity on day 0. Individual exit score was calculated within each group by dividing exit velocity into quintiles and assigning heifers with a score from 1 to 5 (1=slowest; 5=fastest heifer). Heifers were classified according to exit score as adequate (ADQ, n =96; exit score⩽3) or excitable temperament (EXC, n =74; exit score>3). Heifer BW, body condition score (BCS) and blood samples were obtained on days 0, 31, 60 and 91. Heifer exit velocity and score were recorded again on days 31, 60 and 91. Ovarian transrectal ultrasonography was performed on days 0 and 10, 31 and 41, 60 and 70, 81 and 91 for puberty evaluation. Heifer was declared pubertal at the first 10-day interval in which a corpus luteum was detected. Exit velocity and exit score obtained on day 0 were correlated (r ⩾0.64, P <0.01) with evaluations on days 31, 60 and 91. During the experiment, ADQ had greater (P <0.01) mean BCS and BW gain, and less (P <0.01) mean plasma cortisol concentration compared with EXC heifers. Temperament × time interactions were detected (P <0.01) for exit velocity and exit score, which were always greater (P <0.01) in EXC v. ADQ heifers. A temperament × time interaction was also detected (P =0.03) for puberty attainment, which was delayed in EXC v. ADQ heifers. At the end of the experiment, a greater (P <0.01) proportion of ADQ were pubertal compared with EXC heifers. In summary, B. indicus heifers classified as EXC had reduced growth, increased plasma cortisol concentrations and hindered puberty attainment compared to ADQ heifers. The present study evaluated the growth and puberty attainment of Bos indicus heifers administered recombinant bovine somatotropin (bST) or saline injections during preweaning and/or postweaning. On day 0, 177 suckling Nellore heifers were stratified by initial age and body weight (BW) (80 ± 10 d; 97 ± 16 kg), and randomly assigned, in a 2 × 2 factorial design (n = 44 to 45 heifers/treatment), to receive s.c. injections of saline (5 mL 0.9% NaCl) or sometribove zinc (Posilac; Elanco, Greenfield, IN; 6.14 mg/kg of BW0.75) on days 0 and 10 (PRE) and/or days 167 and 177 (POS). All heifers were managed as a single group in Brachiaria decumbens pastures from day 0 until 24 d postweaning (day 191), and then provided a corn silage–based TMR from days 191 to 380 to achieve 65% to 70% of mature BW at the end of the study (day 380). Heifer full BW was collected on days 0, 10, 167, 177, and monthly from days 191 to 380. Transrectal ultrasonography of ovaries was performed on days 1 and 10 of each month from days 229 to 380 to assess the percentage of pubertal heifers. Liver biopsies and blood samples from jugular vein were collected on days 0, 10, 167, 177, and 380. Additional blood samples were collected monthly from days 259 to 380 (n = 10 to 15 heifers/treatment). No interactions among day of the study, PRE, and POS injections of saline or bST were detected (P ≥ 0.11). Preweaning bST injections increased heifer average daily gain (ADG) from days 0 to 10 and plasma IGF-1 on day 10 (P ≤ 0.03), did not affect ADG from days 0 to 177, plasma IGF-1 from days 259 to 380, and any liver gene mRNA expression (P ≥ 0.19), but tended to decrease ADG from days 191 to 380 (P = 0.07) and percentage of pubertal heifers on days 349 (P = 0.07), 359 (P = 0.002), and 380 (P = 0.0001) compared with saline injections. Postweaning bST injections increased plasma IGF-1 on day 177 and overall liver mRNA expression of GHR-1A (P ≤ 0.05), decreased plasma IGF-1 from days 259 to 380 (P = 0.03), tended to decrease liver mRNA expression of GHR-1B on day 177 (P = 0.08), but did not affect ADG from days 167 to 177 and 191 to 380, and puberty attainment from days 229 to 380 (P ≥ 0.12) compared with saline injections. Thus, preweaning and postweaning injections of bST successfully increased heifer plasma IGF-1 concentrations 10 d after first injection. Postweaning injections of bST had no impact on puberty attainment, whereas preweaning bST injections of bST impaired puberty attainment of Nellore beef heifers. [ABSTRACT FROM AUTHOR]

The objectives of the study were to evaluate the growth, physiological parameters, and liver trace mineral status of beef heifers provided low-quality warm-season forage and different forms (meal vs. block) of trace mineral-fortified supplementation. One hundred yearling Nellore heifers were blocked by initial body weight (BW) (184 ± 2.5 kg) and randomly assigned into 1 of 20 drylot pens (5 heifers/pen). Treatments were randomly assigned to pens (5 pens/treatment) and consisted of heifers receiving: 1) a loose meal trace mineral supplement (TM; De Heus Animal Nutrition Industry); 2) free choice access to a low-moisture, cooked sugarcane molasses-based protein block (LMB); 3) isocaloric and isonitrogenous, loose meal protein supplement pair-fed to LMB supplement dry matter (DM) intake (PSPF); and (4) loose meal protein supplement offered at 0.2% of BW (PS). Supplements were formulated to achieve same daily intake of supplemental trace mineral among treatments. Hence, TM supplement was offered at 66.6% of the supplement DMI of LMB heifers. Heifers were offered free choice access to water and ground brachiaria (Brachiaria brizantha) hay from day 0 to 45. Overall average daily gain from day 0 to 45 was the least for TM heifers (P ≤ 0.05) and did not differ among LMB, PSPF, and PS heifers (P ≥ 0.60). Daily hay DMI did not differ among treatments (P ≥ 0.63). Total intake of DM and TDN were least for TM heifers (P ≤ 0.03) and did not differ (P ≥ 0.66) among LMB, PSPF, and PS heifers. Total supplemental intake of crude protein (CP) and rumen degradable protein (RDP) and total intake of CP and RDP (supplement + hay) were least for TM and greatest for PS heifers (P ≤ 0.05), and intermediate for LMB and PSPF heifers (P ≥ 0.70). Effects of treatment × day and treatment were not detected (P ≥ 0.61) for plasma concentrations of insulin-like growth factor 1 (IGF-1), and non-esterified fatty acids (NEFA). Effects of treatment were detected for plasma concentrations of PUN (P = 0.005) and tended to be detected for plasma concentrations of glucose (P = 0.08), which were least for TM heifers (P ≤ 0.03) and did not differ (P ≥ 0.17) among LMB, PSPF, and PS heifers. Trace mineral intake and liver concentrations of all trace minerals did not differ (P ≥ 0.13) among treatments. Hence, the use of LMB supplementation resulted in positive effects on growth without impacting trace mineral status compared to a loose meal trace mineral salt, and similar growth performance and trace mineral status compared to a conventional protein supplementation offered at 0.2% of body weight. Agents able to bind tightly and selectively to disease markers can greatly benefit disease diagnosis and therapy. Aptamers are functional molecules, usually DNA or RNA oligonucleotides, with the appropriate sequence and structure to form a complex with a target molecule. MUC1 is a well-known tumour marker present in a variety of malignant tumours and it has been a target of interest for many years. In this work we report the selection of DNA aptamers that bind with high affinity and selectivity to the MUC1 peptides. Combinatorial chemistry techniques based on the SELEX methodology were used for the identification of the specific aptamers. These were selected from an initial library containing a 25-base-long variable region, resulting in 425 random sequences of single-stranded DNA molecules, for their ability to bind to synthetic forms of MUC1. Ten rounds of in vitro selection were performed enriching for MUC1 binding. By round ten more than 90% of the pool of sequences consisted of MUC1-binding molecules. Selected aptamer families were cloned, sequenced and found to be unique, sharing no sequence consensus. The binding properties of these aptamers were quantitated by enzyme-linked immunosorbent assay and surface plasmon resonance, whereas their specificity for MUC1-expressing cancer cells has been validated using fluorescent microscopy. Aptamers offer significant advantages over existing antibody-based recognition procedures in that they offer higher binding affinity (higher retention/reduced dissociation) and specificity to the target (ability to determine variations on the protein target down to single amino acid changes), higher selectivity against mutated protein epitopes and potentially reduced immunogenicity and increased tumour penetration associated with their size. Unlabelled: Low-grade inflammation adversely influences metabolism and cardiovascular prognosis, nevertheless increased intake of fruits and vegetables has rarely been studied in this context., Objective: In a prospective controlled study, the effect on C-reactive protein (CRP) levels was assessed., Methodology: Sixty consecutive women undergoing cosmetic abdominal surgery were instructed to consume six servings each of fruits and vegetables during the first postoperative month. Detailed 24 h interviewer-administered dietary recall was conducted at baseline and at the end of the study, with weekly returns to monitor unscheduled dietary changes and compliance with the protocol. Variance (ANOVA) and covariance (ANCOVA) were evaluated to confirm significance and minimize confounding variables., Results: No differences concerning age (42.2±5.3 vs 41.1±6.0 years) or BMI (25.5±3.1 vs 25.0±3.0 kg/m²) occurred. Ingestion of fruits increased to approximately 5.2 vs 3.9 and of vegetables 5.9 vs 3.4 servings/ day, respectively. CRP decreased more conspicuously in the treated group (P=0.028), and correlation between vitamin C input and CRP in supplemented participants was demonstrated (P=0.014)., Conclusions: Higher intake of antioxidant foods was feasible, and an antiinflammatory effect occurred. Further studies with longer administration and follow-up period are recommended.
