Abstract

A obra de Herberto Helder tem contribuído para pensar sobre a relação entre a poesia e as outras artes, como a fotografia e o cinema. Este artigo explora uma via possível dessa relação a partir do poema “Retratíssimo ou narração de um homem depois de maio”, estendendo a leitura do texto herbertiano a uma obra fotográfica, o autorretrato “Eu poeta”, de Fernando Lemos. Considerando que Helder apresenta um “Retratoblíquo” no qual inclui a expressão “Julgo ser eu”, preconizando a incerteza figurativa do poema, propõe-se uma ampliação da obliquidade do autorretrato poético para mostrar que a ambiguidade autorretratística pode sugerir o cruzamento de diferentes objetos artísticos aos olhos do leitor

Abstract

Common single-nucleotide polymorphisms (SNPs) are predicted to collectively explain 40-50% of phenotypic variation in human height, but identifying the specific variants and associated regions requires huge sample sizes1. Here, using data from a genome-wide association study of 5.4 million individuals of diverse ancestries, we show that 12,111 independent SNPs that are significantly associated with height account for nearly all of the common SNP-based heritability. These SNPs are clustered within 7,209 non-overlapping genomic segments with a mean size of around 90 kb, covering about 21% of the genome. The density of independent associations varies across the genome and the regions of increased density are enriched for biologically relevant genes. In out-of-sample estimation and prediction, the 12,111 SNPs (or all SNPs in the HapMap 3 panel2) account for 40% (45%) of phenotypic variance in populations of European ancestry but only around 10-20% (14-24%) in populations of other ancestries. Effect sizes, associated regions and gene prioritization are similar across ancestries, indicating that reduced prediction accuracy is likely to be explained by linkage disequilibrium and differences in allele frequency within associated regions. Finally, we show that the relevant biological pathways are detectable with smaller sample sizes than are needed to implicate causal genes and variants. Overall, this study provides a comprehensive map of specific genomic regions that contain the vast majority of common height-associated variants. Although this map is saturated for populations of European ancestry, further research is needed to achieve equivalent saturation in other ancestries.

11. Lowering the Voting Age to 16: Young People Making a Case for Political Education in Fostering Voting Competencies

Abstract

The intention of this article is to contribute to the debate about whether the voting age should be lowered to the age of 16, by examining quantitative and qualitative data collected in a local participation project with young people in Portugal: questionnaires (N = 961), interviews (N = 3), and focus group discussions (N = 15). Considering the coexistence of both willingness and reluctance to get engaged in formal politics--as youngsters often feel ill-equipped politically--it is argued that adequate political education needs to be provided by schools to enable young people to be confident and knowledgeable voters. We propose that governments recognise the importance of this area in the school curriculum, in order to enable the young people's acquisition of knowledge and skills that can sustain their growth as competent voters. This is crucial in legitimising democratic representative systems.

Abstract

Miscarriage is a common, complex trait affecting ~15% of clinically confirmed pregnancies. Here we present the results of large-scale genetic association analyses with 69,054 cases from five different ancestries for sporadic miscarriage, 750 cases of European ancestry for multiple (≥3) consecutive miscarriage, and up to 359,469 female controls. We identify one genome-wide significant association (rs146350366, minor allele frequency (MAF) 1.2%, P = 3.2 × 10-8, odds ratio (OR) = 1.4) for sporadic miscarriage in our European ancestry meta-analysis and three genome-wide significant associations for multiple consecutive miscarriage (rs7859844, MAF = 6.4%, P = 1.3 × 10-8, OR = 1.7; rs143445068, MAF = 0.8%, P = 5.2 × 10-9, OR = 3.4; rs183453668, MAF = 0.5%, P = 2.8 × 10-8, OR = 3.8). We further investigate the genetic architecture of miscarriage with biobank-scale Mendelian randomization, heritability, and genetic correlation analyses. Our results show that miscarriage etiopathogenesis is partly driven by genetic variation potentially related to placental biology, and illustrate the utility of large-scale biobank data for understanding this pregnancy complication.

Abstract

To investigate the genetic basis of type 2 diabetes (T2D) to high resolution, the GoT2D and T2D-GENES consortia catalogued variation from whole-genome sequencing of 2,657 European individuals and exome sequencing of 12,940 individuals of multiple ancestries. Over 27M SNPs, indels, and structural variants were identified, including 99% of low-frequency (minor allele frequency [MAF] 0.1-5%) non-coding variants in the whole-genome sequenced individuals and 99.7% of low-frequency coding variants in the whole-exome sequenced individuals. Each variant was tested for association with T2D in the sequenced individuals, and, to increase power, most were tested in larger numbers of individuals (>80% of low-frequency coding variants in ~82 K Europeans via the exome chip, and ~90% of low-frequency non-coding variants in ~44 K Europeans via genotype imputation). The variants, genotypes, and association statistics from these analyses provide the largest reference to date of human genetic information relevant to T2D, for use in activities such as T2D-focused genotype imputation, functional characterization of variants or genes, and other novel analyses to detect associations between sequence variation and T2D.

Abstract

To characterize type 2 diabetes (T2D)-associated variation across the allele frequency spectrum, we conducted a meta-analysis of genome-wide association data from 26,676 T2D case and 132,532 control subjects of European ancestry after imputation using the 1000 Genomes multiethnic reference panel. Promising association signals were followed up in additional data sets (of 14,545 or 7,397 T2D case and 38,994 or 71,604 control subjects). We identified 13 novel T2D-associated loci (P < 5 × 10-8), including variants near the GLP2R, GIP, and HLA-DQA1 genes. Our analysis brought the total number of independent T2D associations to 128 distinct signals at 113 loci. Despite substantially increased sample size and more complete coverage of low-frequency variation, all novel associations were driven by common single nucleotide variants. Credible sets of potentially causal variants were generally larger than those based on imputation with earlier reference panels, consistent with resolution of causal signals to common risk haplotypes. Stratification of T2D-associated loci based on T2D-related quantitative trait associations revealed tissue-specific enrichment of regulatory annotations in pancreatic islet enhancers for loci influencing insulin secretion and in adipocytes, monocytes, and hepatocytes for insulin action-associated loci. These findings highlight the predominant role played by common variants of modest effect and the diversity of biological mechanisms influencing T2D pathophysiology.

Abstract

To dissect the genetic architecture of blood pressure and assess effects on target organ damage, we analyzed 128,272 SNPs from targeted and genome-wide arrays in 201,529 individuals of European ancestry, and genotypes from an additional 140,886 individuals were used for validation. We identified 66 blood pressure-associated loci, of which 17 were new; 15 harbored multiple distinct association signals. The 66 index SNPs were enriched for cis-regulatory elements, particularly in vascular endothelial cells, consistent with a primary role in blood pressure control through modulation of vascular tone across multiple tissues. The 66 index SNPs combined in a risk score showed comparable effects in 64,421 individuals of non-European descent. The 66-SNP blood pressure risk score was significantly associated with target organ damage in multiple tissues but with minor effects in the kidney. Our findings expand current knowledge of blood pressure-related pathways and highlight tissues beyond the classical renal system in blood pressure regulation.

Abstract

Carbon dioxide released permanently from soils in diffuse degassing areas may constitute a permanent hazard for the population. Several villages in the Azores archipelago (Portugal) are placed in areas with anomalous soil CO2 degassing and lethal indoor CO2 concentration (>10 vol%) has been already recorded in some buildings. The 2021-2022 dislodgements of population at Vulcano (Italy) and La Palma (Spain) volcanic islands due to high soil CO2 degassing highlight the importance of defining criteria to produce human CO2 exposure risk maps, which are useful to mitigate the risk and should constitute valuable tools for land-use planners. Risk is assessed in the current study by combining susceptibility, exposure, and vulnerability maps. The defined criteria were applied to two villages in Furnas Volcano (Sao Miguel Island, Azores), showing that 58% and 98% of the buildings, respectively, at Furnas and Ribeira Quente villages are at high risk of CO2 exposure. Abstract

Strategies for the adaptive reuse of vernacular architecture are of utmost importance in the current context of social, economic, and environmental vulnerability. This article examines the design strategies of adaptive reuse in three cases of renowned architects of the so-called School of Porto developed across the second half of the 20th century, specifically between 1956 and 1991. The paper aims to introduce a new and deeper knowledge of the selected practices by critically documenting the whole process of the intervention (before, during, after) and not only the final result, as is common practice in specialized publications. The research methodology combines the bibliographical and archival research and interpretation of diverse graphic, photographic, and textual documentation with the production of analytical drawings. The demolitions/additions color code (black/yellow/red) is applied to plans, sections, and elevations as an essential tool for understanding and communicating the transformations undertaken. The selected case studies are Além House (1956–1967) by Fernando Távora, Alcino Cardoso House (1971–1973; 1988–1991) by Álvaro Siza, and the House in Gerês (1980–1982) by Eduardo Souto de Moura. These projects show different strategies of intervention in built heritage, providing lessons on the reactivation of obsolete or abandoned rural constructions with new functions that are compatible with the preservation of their values (historical, landscape, constructive, social, and aesthetic) and guidelines for sustainable reuse.
