Abstract

Physical and selected physicochemical properties of distilled water treated with low-temperature low-frequency glow plasma (LPGP) for 5, 15, 30, 45, 60 and 90 min are given. Particular properties of PTW non-linearly vary with the time of exposure to plasma. PTW is characterized as declusterized solution of normal water consisting of clathrates of excited, probably singlet oxygen molecules, inside the clathrates. PWT is free of hydrogen peroxide and ozone. Raman spectroscopy is the best tool for diagnostic assessment of PTW.

36. Molecular Characterization of Wheat Amylopectins by Multiangle Laser Light Scattering Analysis

Abstract

Weight-average molecular weight (Mw) and chain length of eight amylopectins isolated from one Australian, two United States, and five Korean wheats were measured using multiangle laser light scattering (MALLS) and refractive index (RI) detectors operated in a microbatch mode, and in a high-performance size-exclusion chromatography (HPSEC) mode. The Mwof amylopectins measured in the microbatch mode ranged from 29 × 106to 349 × 106. Three amylopectins (Geuru, Tapdong and WW) showed significantly high Mwvalues over 200 × 106. The Mwmeasured by HPSEC mode with MALLS-RI detectors (42 × 106to 73 × 106) were significantly less than those obtained in the microbatch mode with exception of dark northern spring hard wheat (DNS) amylopectin, indicating the possible variation of Mwby the analysis mode. Root-mean square of the radius of gyration (Rg) also was greater when the microbatch mode was used (122–340 vs. 95–116 nm). Chain length distributions of debranched amylopectins of different cultivars, measured by the HPSEC-MALLS-RI system, were similar. Weight average degrees of polymerization (DPw) of A, B1, and larger B chains (B=2) had ranges of 13–22, 26–46, and 58–73, respectively, and mass ratios of A and B chains ranged from 0.7 to 1.1.

Abstract

Molecular weight (Mw) and radii of gyration (Rg) of starch polysaccharide chains, distribution of amylopectin structural units, crystallinity, thermal properties, pasting viscosity profiles, susceptibility to α-amylolysis, swelling power and solubility of spelt and wheat starches isolated from commercially available spelt and wheat flours were studied. Average molecular weight Mw of polysaccharide chains eluted under the whole complex peak from high-performance size exclusion chromatography (HPSEC) column was found to be 0.829 x 107 g·mol-1 for spelt starch and 2.307 x 107 g·mol-1 for wheat starch. Significant differences in amylopectin chain length distribution between spelt and wheat starch were found. Shortest A type chains with Mw = 3.418 x 10³ g·mol-1 and Rg = 65.4 nm constituted 88.2% of total mass of debranched spelt starch, whereas mass ratio of respective chains of wheat starch with Mw = 5.277 x 10³ g·mol-1 and Rg = 88.2 nm was found to be 32.7%. Wheat starch was more susceptible to α-amylolysis than spelt starch. Enzymatic hydrolysis, rate constant for the first stage of hydrolysis (k1 = 4.5 x 10-3 mg·ml-1min-1) and final hydrolysis extent (42.5%) for wheat starch were significantly higher than the respective values for spelt starch (k1 = 2.3 x 10-3 mg·ml-1min-1; 35%). Abstract

The alpha-amylolysis of potato, corn, waxy corn, and amaranthus starches with porcine pancreatic alpha-amylase upon illumination with visible polarized and nonpolarized light was studied. Samples were illuminated for 1 h either directly after blending the enzyme with starch or the enzyme solutions were illuminated for 1 h prior to the admixture of starch. Independently of the mode of illumination, no significant change in the hydrolysis kinetics was noted for corn, waxy corn, and amaranthus starches. The illumination of potato starch in the presence of alpha-amylase with polarized and nonpolarized light significantly accelerated the hydrolysis. In the first 5-h step the hydrolysis rate increased from 12.0 to 60.0 g x kg(-)(1) h(-)(1). Preillumination of enzyme in solution resulted in an increase in the rate of hydrolysis to 151.6 and 131.4 g x kg(-)(1) h(-)(1) after illumination with polarized and nonpolarized light, respectively. Circular dichroism spectra of alpha-amylase solutions stored in the dark and illuminated with visible polarized and nonpolarized light provided evaluation of the protein conformation, whereas exposure of enzyme solutions to the nonpolarized did not change the secondary structure of the protein. The illumination of the alpha-amylase solutions with polarized light significantly changed the amounts of alpha-helix and beta-form vs unilluminated samples: 42.3% and 25.5% vs 36.6% and 30.2%, respectively.
