1. How Crystalline is Low-Density Amorphous Ice?

Abstract

Low-density amorphous ice (LDA) is one of the most common solid materials in the Universe and a key material for understanding the many famous anomalies of liquid water. Yet, despite its significance and its discovery dating nearly 90 years, the structure of LDA is debated. It is unclear if LDA is a glassy state representing a liquid or a heavily disordered crystal; indeed, two forms (LDA-I and LDA-II) have been discussed as amorphous and partially crystalline in the literature, respectively. Here, with two widely used water models, we show that the experimental structure factor of LDA is best reproduced computationally by a partially crystalline structure. Models for both LDA-I and LDA-II are highly similar, with differences only due to subtle differences in crystallinity and/or experimental error. Further support for this structural model of LDA comes from experiment: if LDA is partially crystalline, then its route to formation should result in different nanocrystallite cubicities, and thus give rise to different cubicities upon recrystallisation. This memory effect of LDA's creation route is observed and it is incompatible with a fully amorphous material. The results present a unified computational and experimental view that LDA is not fully amorphous but instead a partially crystalline material. This impacts LDA's many roles in nature and potentially our understanding of liquid water. Furthermore, the "re-identification" of such an intensely studied material highlights that great care will be needed when classifying the nature of glassy materials going forward., Comment: 9 pages, 5 figures

4. The role of T‐type calcium channels in elderly human vascular function: A pilot randomized controlled trial

Abstract

Abstract Endothelial dysfunction develops with age and may precede cardiovascular disease. Animal data suggest that T‐type calcium channels play an important role in endothelial function, but data from humans are lacking. This study included 15 healthy, sedentary, elderly males for a double blinded, randomized controlled trial. For 8 weeks, they were given 40 mg/day of either efonidipine (L‐ and T‐type calcium channel blocker (CCB)) or nifedipine (L‐type CCB). Vascular function was evaluated by graded femoral arterial infusions of acetylcholine (ACh; endothelium‐dependent vasodilator) and sodium nitroprusside (endothelium‐independent vasodilator) both with and without co‐infusion of N‐acetylcysteine (NAC; antioxidant). We measured leg blood flow and mean arterial pressure and calculated leg vascular conductance to evaluate the leg vascular responses. Despite no significant change in blood pressure in either group, we observed higher leg blood flow responses (Δ 0.43 ± 0.45 l/min, P = 0.006) and leg vascular conductance (Δ 5.38 ± 5.67 ml/min/mmHg, P = 0.005) to intra‐arterial ACh after efonidipine, whereas there was no change in the nifedipine group, and no differences between groups. We found no upregulation of endothelial nitric oxide synthase in vastus lateralis muscle biopsies within or between groups. Smooth muscle cell responsiveness was unaltered by efonidipine or nifedipine. Intravenous co‐infusion of NAC did not affect endothelium‐dependent vasodilatation in either of the CCB groups. These results suggest that 8 weeks’ inhibition of T‐ and L‐type calcium channels augments endothelium‐dependent vasodilatory function in healthy elderly males. Further studies are required to elucidate if T‐type calcium channel inhibition can counteract endothelial dysfunction.

5. Thermal Desorption of H2O-Ice: From Nanoscale Films to the Bulk

Abstract

Desorption of H2O films ranging from 53 nanometres to 101 micrometre thicknesses have been investigated using a quartz-crystal microbalance (QCM) and temperature-programmed desorption. Three desorption stages are observed belonging to amorphous solid water (ASW), stacking disordered ice I (ice Isd), and hexagonal ice I (ice Ih). The desorption of ASW is only visible for the >10 micrometre films and is separated from the ice I desorption by 10-15 K and has an associated desorption energy of 64 kJ mol-1. The desorption energy of the 53 nanometre film was found to be near 50 kJ mol-1 as also noted in the literature, but with increasing film thickness the desorption energy of ice I rises until reaching a plateau around 65-70 kJ mol-1. The reason for the increased desorption energy is suggested to be due to molecules unable to desorb due to the thick covering layer of H2O and possibly re-adsorption. Before complete desorption of ice I which occurs around 220 K for the 100 micrometre film, a two-stage ice I desorption is observed with the QCM for the 10 and 20 micrometre films near 200 K. This event corresponds to the desorption of ice Isd as corroborated by X-ray diffraction patterns collected upon heating from 92-260 K at ambient pressure. Cubic ice is not observed as is commonly stated in the literature as resulting from the crystallisation of ASW. Therefore, ice Isd is the correct terminology for the initial crystallisation product of ASW.

Abstract

Abstract Aldosterone has been suggested to be involved in the microvascular complications observed in type 2 diabetes. We aimed to investigate the effect of mineralocorticoid receptor (MR) blockade on endothelial function in individuals with type 2 diabetes compared to healthy controls. We included 12 participants with type 2 diabetes and 14 controls. We measured leg hemodynamics at baseline and during femoral arterial infusion of acetylcholine and sodium nitroprusside before and 8 weeks into treatment with MR blockade (eplerenone). Acetylcholine infusion was repeated with concomitant n‐acetylcysteine (antioxidant) infusion. No difference in leg blood flow or vascular conductance was detected before or after the treatment with MR blockade in both groups and there was no difference between groups. Infusion of n‐acetylcysteine increased baseline blood flow and vascular conductance, but did not change the vascular response to acetylcholine before or after treatment with MR blockade. Skeletal muscle eNOS content was unaltered by MR blockade and no difference between groups was detected. In conclusion, we found no effect of MR blockade endothelial function in individuals with and without type 2 diabetes. As the individuals with type 2 diabetes did not have vascular dysfunction, these results might not apply to individuals with vascular dysfunction.

Abstract

The recent discovery of a low-temperature endotherm upon heating hydrochloric-acid doped ice VI has sparked a vivid controversy. The two competing explanations aiming to explain its origin range from a new distinct crystalline phase of ice to deep-glassy states of the well-known ice VI. Problems with the slow kinetics of deuterated phases have been raised, which we circumvent here entirely by simultaneously measuring the inelastic neutron spectra and neutron diffraction data of H2O samples. These measurements clearly confirm the deep-glassy ice VI scenario and rule out alternative explanations. Additionally, we show that the crystallographic model of D2O ice XV, the ordered counterpart of ice VI, also applies to the corresponding H2O phase. The discovery of deep-glassy ice VI now provides a fascinating new example of ultra-stable glasses which are encountered across a wide range of other materials., Comment: 22 pages, 5 figures, 1 table (includes SI)

18. Ammonium Fluoride as a Hydrogen-disordering Agent for Ice

Abstract

The removal of residual hydrogen disorder from various phases of ice with acid or base dopants at low temperatures has been a focus of intense research for many decades. As an antipode to these efforts, we now show using neutron diffraction that ammonium fluoride (NH4F) is a hydrogen-disordering agent for the hydrogen-ordered ice VIII. Cooling its hydrogen-disordered counterpart ice VII doped with 2.5 mol% ND4F under pressure leads to a hydrogen-disordered ice VIII with ~31% residual hydrogen disorder illustrating the long-range hydrogen-disordering effect of ND4F. The doped ice VII could be supercooled by ~20 K with respect to the hydrogen-ordering temperature of pure ice VII after which the hydrogen-ordering took place slowly over a ~60 K temperature window. These findings demonstrate that ND4F-doping slows down the hydrogen-ordering kinetics quite substantially. The partial hydrogen order of the doped sample is consistent with the antiferroelectric ordering of pure ice VIII. Yet, we argue that local ferroelectric domains must exist between ionic point defects of opposite charge. In addition to the long-range effect of NH4F-doping on hydrogen-ordered water structures, the design principle of using topological charges should be applicable to a wide range of other 'ice-rule' systems including spin ices and related polar materials., Comment: 23 pages, 4 figures, 2 tables

21. Benchmarking acid and base dopants with respect to enabling the ice V to XIII and ice VI to XV hydrogen-ordering phase transitions

Abstract

Doping the hydrogen-disordered phases of ice V, VI and XII with hydrochloric acid (HCl) has led to the discovery of their hydrogen-ordered counterparts ices XIII, XV and XIV. Yet, the mechanistic details of the hydrogen-ordering phase transitions are still not fully understood. This includes in particular the role of the acid dopant and the defect dynamics that it creates within the ices. Here we investigate the effects of several acid and base dopants on the hydrogen ordering of ices V and VI with calorimetry and X-ray diffraction. HCl is found to be most effective for both phases which is attributed to a favourable combination of high solubility and strong acid properties which create mobile H3O+ defects that enable the hydrogen-ordering processes. Hydrofluoric acid (HF) is the second most effective dopant highlighting that the acid strengths of HCl and HF are much more similar in ice than they are in liquid water. Surprisingly, hydrobromic acid doping facilitates hydrogen ordering in ice VI whereas only a very small effect is observed for ice V. Conversely, lithium hydroxide (LiOH) doping achieves a performance comparable to HF-doping in ice V but it is ineffective in the case of ice VI. Sodium hydroxide, potassium hydroxide (as previously shown) and perchloric acid doping are ineffective for both phases. These findings highlight the need for future computational studies but also raise the question why LiOH-doping achieves hydrogen-ordering of ice V whereas potassium hydroxide doping is most effective for the 'ordinary' ice Ih., Comment: 18 pages, 7 figures, 1 table

Abstract

Cerebrospinal fluid contacting neurons (CSF-cNs) are a specific type of neurons located around the ventricles in the brain and the central canal in the spinal cord and have been demonstrated to be intrinsic sensory neurons in the central nervous system. One of the important channels responsible for the sensory function is the polycystic kidney disease 2-like 1 (PKD2L1) channel. Most of the studies concerning the distribution and function of the PKD2L1-expressing CSF-cNs in the spinal cord have previously been performed in non-mammalian vertebrates. In the present study immunohistochemistry was performed to determine the distribution of PKD2L1-immunoreactive (IR) CSF-cNs in the spinal cords of four mammalian species: mouse, rat, cat, and macaque monkey. Here, we found that PKD2L1-expressing CSF-cNs were present at all levels of the spinal cord in these animal species. Although the distribution pattern was similar across these species, differences existed. Mice and rats presented a clear PKD2L1-IR cell body labeling, whereas in cats and macaques the PKD2L1-IR cell bodies were more weakly labeled. Ectopic PKD2L1-IR neurons away from the ependymal layer were observed in all the animal species although the abundance and the detailed locations varied. The apical dendritic protrusions with ciliated fibers were clearly seen in the lumen of the central canal in all the animal species, but the sizes of protrusion bulbs were different among the species. PKD2L1-IR cell bodies/dendrites were co-expressed with doublecortin, MAP2 (microtubule-associated protein 2), and aromatic L-amino acid decarboxylase, but not with NeuN (neuronal nuclear protein), indicating their immature properties and ability to synthesize monoamine transmitters. In addition, in situ hybridization performed in rats revealed PKD2L1 mRNA expression in the cells around the central canal. Our results indicate that the intrinsic sensory neurons are conserved across non-mammalian and mammalian vertebrates. The similar morphology of the dendritic bulbs with ciliated fibers (probably representing stereocilia and kinocilia) protruding into the central canal across different animal species supports the notion that PKD2L1 is a chemo- and mechanical sensory channel that responds to mechanical stimulations and maintains homeostasis of the spinal cord. However, the differences of PKD2L1 distribution and expression between the species suggest that PKD2L1-expressing neurons may receive and process sensory signals differently in different animal species.

Abstract

Purpose: To investigate the experiences the target group had with physical activity as a physiotherapy intervention. Design, material and method: Six qualitative interviews were analyzed by Kirsti Malteruds´ principles of systematic text condensation (STC). This study was based on the biopsychosocial model of understanding (BPSS). Findings: Four categories described what the training group led to. 1) To change focus contributed to stop negative thoughts. The experience of being tired in a new way resulted in a calmer everyday life. 2) To have continuous routines created predictability, and the experience of being expected made attending manageable. 3) To be a part of a community gave a feeling of affiliation, even though exclusion was felt occasionally. 4) An experience of victory created a belief in facing, and managing other challenges in life. Conclusion: The findings gave an understanding of physical activity as an important impact in the patients’ lives (biologically, psychologically and socially). Physiotherapists can be an important part in the process of further developing the treatment for a group in need of a closer follow-up offer.

Abstract

Aim: The SAMe-TT2R2 score helps identify patients with atrial fibrillation (AF) likely to have poor anticoagulation control during anticoagulation with vitamin K antagonists (VKA) and those with scores >2 might be better managed with a target-specific oral anticoagulant (NOAC). We hypothesized that in clinical practice, VKAs may be prescribed less frequently to patients with AF and SAMe-TT2R2 scores >2 than to patients with lower scores. Methods and results: We analyzed the Phase III dataset of the Global Registry on Long-Term Oral Antithrombotic Treatment in Patients with Atrial Fibrillation (GLORIA-AF), a large, global, prospective global registry of patients with newly diagnosed AF and ≥1 stroke risk factor. We compared baseline clinical characteristics and antithrombotic prescriptions to determine the probability of the VKA prescription among anticoagulated patients with the baseline SAMe-TT2R2 score >2 and ≤ 2. Among 17,465 anticoagulated patients with AF, 4,828 (27.6%) patients were prescribed VKA and 12,637 (72.4%) patients an NOAC: 11,884 (68.0%) patients had SAMe-TT2R2 scores 0-2 and 5,581 (32.0%) patients had scores >2. The proportion of patients prescribed VKA was 28.0% among patients with SAMe-TT2R2 scores >2 and 27.5% in those with scores ≤2. Conclusions: The lack of a clear association between the SAMe-TT2R2 score and anticoagulant selection may be attributed to the relative efficacy and safety profiles between NOACs and VKAs as well as to the absence of trial evidence that an SAMe-TT2R2-guided strategy for the selection of the type of anticoagulation in NVAF patients has an impact on clinical outcomes of efficacy and safety. The latter hypothesis is currently being tested in a randomized controlled trial. 39. Icy dust grains in the interstellar medium : their properties and impact

Abstract

The behaviour of atoms and molecules on interstellar dust grain analogue surfaces has been probed with surface science techniques such as reflection-absorption infrared spectroscopy (RAIRS) and temperature programmed desorption (TPD). Chemical systems under investigation include O atoms, O2, CO, H2O and N2O as deposited either on a silica (SiO2) surface representative of an interstellar dust grain or on H2O surfaces. As H2O is the dominant solid state chemical species in the interstellar medium (ISM) preliminary experiments have been conducted to investigate one of its formation pathways. Such experiments involved bombarding SiO2 or H2O surfaces with O atoms, producing O2 and O3 molecules as determined by RAIRS and TPD. During the initial stages of H2O growth on dust grains in the ISM, only small quantities will be found in the solid state. The de-wetting behaviour of such quantities was investigated by directly dosing H2O onto the SiO2 surface and observing the νOH vibrational band with time and temperature. Through such experiments, H2O has been observed to de-wet even at cryogenic temperatures of 17 K with an activation energy of about 2 kJ mol-1. The effect of this in the ISM is that bare dust grain surfaces will be left exposed for other molecules, such as CO, to interact with. Such interactions may be stronger than those of the molecules directly interacting with a water surface, meaning molecules will reside in the solid state for longer times. As icy mantles develop throughout the lifetime of a molecular cloud, adsorbed CO becomes a large part of the icy mantle. Multilayers of CO were investigated and shown to exhibit a bulk potential. This charge is due to a spontaneous electric (spontelectric) field arising as certain dipolar molecules align in the solid state. To investigate the spontelectric effect in CO, N2O was first examined and shows a correlation between IR features and the direct measurement of the spontelectric potential of N2O. Such a link was found through the temperature dependent shifts in the LO-TO splitting of the N2O. CO exhibits the same temperature dependence and spontelectric parameters have been extracted to show that a spontelectric potential of 6.7 mV per monolayer is created as multilayers adsorb on a surface. The effect of this in the ISM would be to reduce the gas-phase charge and ionisation fractions in molecular clouds of the ISM which in itself has the potential to have wide-reaching implications.

43. Corticosteroid injection for coccydynia: a review of 241 patients

Abstract

Aims: We aimed to establish the short- and long-term efficacy of corticosteroid injection for coccydynia, and to determine if betamethasone or triamcinolone has the best effect. Methods: During 2009 to 2016, we treated 277 patients with chronic coccydynia with either one 6 mg betamethasone or one 20 mg triamcinolone cortisone injection. A susequent injection was given to 62 (26%) of the patients. All were reviewed three to four months after injection, and 241 replied to a questionnaire a mean of 36 months (12 to 88) after the last injection. No pain at the early review was considered early success. When the patient had not been subsequently operated on, and indicated on the questionnaire that they were either well or much better, it was considered a long-term success. Results: At the three- to four-month review, 22 (9%) reported that they had no pain. The long-term success of one injection was 15% and rose to 29% after a second injection. Logistic regression tests showed that both early success (odds ratio (OR) 5.5, 95% confidence interval (CI) 2.1 to 14.4; p = 0.001) and late success (OR 3.7, 95% CI 1.7 to 8.3; p = 0.001) was greater with triamcinolone than with betamethasone. Late success was greater for patients with symptoms for less than 12 months (OR 3.0, 95% CI 1.4 to 6.7; p = 0.006). We saw no complications of the injections. Conclusion: We conclude that the effect of corticosteroid injection for coccygodynia is moderate, possibly because we used modest doses of the drugs. Even so, they seem worthwhile as they are easily and quickly performed, and complications are rare. If the choice is between injections of betamethasone or triamcinolone, the latter should be selected.

Abstract

Abstract Preclinical and clinical proof-of-concept studies have suggested the effectiveness of pharmacological modulation of inflammatory cytokines in ischemic stroke. Experimental evidence shows that targeting tumor necrosis factor (TNF) and interleukin (IL)-1 holds promise, and these cytokines are considered prime targets in the development of new stroke therapies. So far, however, information on the cellular expression of TNF and IL-1 in the human ischemic brain is sparse. We studied 14 cases of human post-mortem ischemic stroke, representing 21 specimens of infarcts aged 1 to > 8 days. We characterized glial and leukocyte reactions in the infarct/peri-infarct (I/PI) and normal-appearing tissue (NAT) and the cellular location of TNF, TNF receptor (TNFR)1 and TNFR2, IL-1α, IL-1β, and IL-1 receptor antagonist (IL-1Ra). The immunohistochemically stained tissue sections received a score reflecting the number of immunoreactive cells and the intensity of the immunoreactivity (IR) in individual cells where 0 = no immunoreactive cells, 1 = many intermediately to strongly immunoreactive cells, and 2 = numerous and intensively immunoreactive cells. Additionally, we measured blood TNF, TNFR, and IL-1 levels in surviving ischemic stroke patients within the first 8 h and again at 72 h after symptom onset and compared levels to healthy controls. We observed IL-1α and IL-1β IR in neurons, glia, and macrophages in all specimens. IL-1Ra IR was found in glia, in addition to macrophages. TNF IR was initially found in neurons located in I/PI and NAT but increased in glia in older infarcts. TNF IR increased in macrophages in all specimens. TNFR1 IR was found in neurons and glia and macrophages, while TNFR2 was expressed only by glia in I/PI and NAT, and by macrophages in I/PI. Our results suggest that TNF and IL-1 are expressed by subsets of cells and that TNFR2 is expressed in areas with increased astrocytic reactivity. In ischemic stroke patients, we demonstrate that plasma TNFR1 and TNFR2 levels increased in the acute phase after symptom onset compared to healthy controls, whereas TNF, IL-1α, IL-1β, and IL-1Ra did not change. Our findings of increased brain cytokines and plasma TNFR1 and TNFR2 support the hypothesis that targeting post-stroke inflammation could be a promising add-on therapy in ischemic stroke patients.

Abstract

Increasing evidence suggests that Alzheimer's disease pathogenesis is not restricted to the neuronal compartment, but includes strong interactions with immunological mechanisms in the brain. Misfolded and aggregated proteins bind to pattern recognition receptors on microglia and astroglia, and trigger an innate immune response characterised by release of inflammatory mediators, which contribute to disease progression and severity. Genome-wide analysis suggests that several genes that increase the risk for sporadic Alzheimer's disease encode factors that regulate glial clearance of misfolded proteins and the inflammatory reaction. External factors, including systemic inflammation and obesity, are likely to interfere with immunological processes of the brain and further promote disease progression. Modulation of risk factors and targeting of these immune mechanisms could lead to future therapeutic or preventive strategies for Alzheimer's disease.

Abstract Individuals with type 2 diabetes have an increased risk of cardiovascular disease. A correlation between plasma aldosterone and hyperinsulinemia has been demonstrated in vivo, and hyperinsulinemia and insulin resistance are independently associated with the development of cardiovascular complications.
