Abstract

Long-range, terrestrial quantum networks will require high brightness single-photon sources emitting in the telecom C-band for maximum transmission rate. Many applications additionally demand triggered operation with high indistinguishability and narrow spectral linewidth. This would enable the efficient implementation of photonic gate operations and photon storage in quantum memories, as for instance required for a quantum repeater. Especially, semiconductor quantum dots (QDs) have shown these properties in the near-infrared regime. However, the simultaneous demonstration of all these properties in the telecom C-band has been elusive. Here, we present a coherently (incoherently) optically-pumped narrow-band (0.8 GHz) triggered single-photon source in the telecom C-band. The source shows simultaneously high single-photon purity with $g^{(2)}(0) = 0.026$ ($g^{(2)}(0) = 0.014$), high two-photon interference visibility of 0.508 (0.664) and high application-ready rates of 0.75 MHz (1.45 MHz) of polarized photons. The source is based on a QD coupled to a circular Bragg grating cavity combined with spectral filtering. Coherent (incoherent) operation is performed via the novel SUPER scheme (phonon-assisted excitation).

Abstract

CRF mediates numerous stress-related endocrine, autonomic, metabolic, and behavioral responses. We present the synthesis and chemical and biological properties of astressin B analogues {cyclo(30-33)[D-Phe(12),Nle(21,38),C(α)MeLeu(27,40),Glu(30),Lys(33)]-acetyl-h/r-CRF(9-41)}. Out of 37 novel peptides, 17 (2, 4, 6-8, 10, 11, 16, 17, 27, 29, 30, 32-36) and 16 (3, 5, 9, 12-15, 18, 19, 22-26, 28, 31) had k(i) to CRF receptors in the high picomolar and low nanomole ranges, respectively. Peptides 1, 2, and 11 inhibited h/rCRF and urocortin 1-induced cAMP release from AtT20 and A7r5 cells. When Astressin C 2 was administered to adrenalectomized rats at 1.0 mg subcutaneously, it inhibited ACTH release for >7 d. Additional rat data based on the inhibitory effect of (2) on h/rCRF-induced stimulation of colonic secretory motor activity and urocortin 2-induced delayed gastric emptying also indicate a safe and long-lasting antagonistic effect. The overall properties of selected analogues may fulfill the criteria expected from clinical candidates.

Abstract

Rural regions in Austria have been under increasing pressure for change in the last decades. On a regional level, varying development patterns of shrinkage and growth arise alongside economic, demographic and social parameters. In this paper, regional ambassador concepts will be explicated as a new instrument of regional development. Additionally, potential positive impacts of these approaches on the problems faced by rural, structurally weak regions will be highlighted. Notable advantages of these approaches are network and feedback effects between stakeholders. These, in turn, are capable of improving the economic and social situation in those regions. However, these measures require a high degree of control capacities which structurally weak areas often lack.

Abstract

The bacterial human pathogen Helicobacter pylori produces a type IV secretion system (cagT4SS) to inject the oncoprotein CagA into gastric cells. The cagT4SS external pilus mediates attachment of the apparatus to the target cell and the delivery of CagA. While the composition of the pilus is unclear, CagI is present at the surface of the bacterium and required for pilus formation. Here, we have investigated the properties of CagI by an integrative structural biology approach. Using Alpha Fold 2 and Small Angle X-ray scattering, it was found that CagI forms elongated dimers mediated by rod-shape N-terminal domains (CagIN) prolonged by globular C-terminal domains (CagIC). Three Designed Ankyrin Repeat Proteins (DARPins) K2, K5 and K8 selected against CagI interacted with CagIC with subnanomolar affinities. The crystal structures of the CagI:K2 and CagI:K5 complexes were solved and identified the interfaces between the molecules, thereby providing a structural explanation for the difference in affinity between the two binders. Purified CagI and CagIC were found to interact with adenocarcinoma gastric (AGS) cells, induced cell spreading and the interaction was inhibited by K2. The same DARPin inhibited CagA translocation by up to 65% in AGS cells while inhibition levels were 40% and 30% with K8 and K5, respectively. Our study suggests that CagIC plays a key role in cagT4SS-mediated CagA translocation and that DARPins targeting CagI represent potent inhibitors of the cagT4SS, a crucial risk factor for gastric cancer development.
