Extension professionals help important agricultural sectors across the country resolve challenges using science-based practices that enhance environmental and social wellbeing while supporting businesses. Nursery and greenhouse growers comprise one of the largest sectors of U.S. agriculture, and this group is challenged to conserve water without compromising their economic viability. While Extension professionals educate and support nursery and greenhouse growers, there is a deficiency of research on adoption processes within this sector. To better understand this important Extension audience, this research examined the influence of critical thinking and problem-solving style on perceived characteristics of water conservation technologies and in turn the perceived characteristics relationship with their implementation. A route to adoption was established to inform effective Extension activities that promote water conservation. Problem-solving style predicts trialability while critical thinking style predicts none of the five characteristics of innovations. Of the five characteristics of water conservation innovations, relative advantage, trialability, and observability play a role in nursery and greenhouse growers' implementation, and implementation does influence adoption. When designing water conservation programs for nursery and greenhouse growers, Extension professionals should consider participants' problem-solving style and incorporate strategies to increase trialability, relative advantage, and observability.

Fragile X Associated Tremor/Ataxia Syndrome (FXTAS) is a neurodegenerative disorder affecting carriers of premutation alleles (PM) of the X-linked FMR1 gene, which contain CGG repeat expansions of 55-200 range in a non-coding region. This late-onset disorder is characterised by the presence of tremor/ataxia and cognitive decline, associated with the white matter lesions throughout the brain, especially involving the middle cerebellar peduncles. Nearly half of older male and ~ 20% of female PM carriers develop FXTAS. While there is evidence for mitochondrial dysfunction in neural and some peripheral tissues from FXTAS patients (though less obvious in the non-FXTAS PM carriers), the results from peripheral blood mononuclear cells (PBMC) are still controversial. Motor, cognitive, and neuropsychiatric impairments were correlated with measures of mitochondrial and non-mitochondrial respiratory activity, AMPK, and TORC1 cellular stress-sensing protein kinases, and CGG repeat size, in a sample of adult FXTAS male and female carriers. Moreover, the levels of these cellular measures, all derived from Epstein- Barr virus (EBV)- transformed and easily accessible blood lymphoblasts, were compared between the FXTAS (N = 23) and non-FXTAS (n = 30) subgroups, and with baseline data from 33 healthy non-carriers. A significant hyperactivity of cellular bioenergetics components as compared with the baseline data, more marked in the non-FXTAS PMs, was negatively correlated with repeat numbers at the lower end of the CGG-PM distribution. Significant associations of these components with motor impairment measures, including tremor-ataxia and parkinsonism, and neuropsychiatric changes, were prevalent in the FXTAS subgroup. Moreover, a striking elevation of AMPK activity, and a decrease in TORC1 levels, especially in the non-FXTAS carriers, were related to the size of CGG expansion. The bioenergetics changes in blood lymphoblasts are biomarkers of the clinical status of FMR1 carriers. The relationship between these changes and neurological involvement in the affected carriers suggests that brain bioenergetic alterations are reflected in this peripheral tissue. A possible neuroprotective role of stress sensing kinase, AMPK, in PM carriers, should be addressed in future longitudinal studies. A decreased level of TORC1-the mechanistic target of the rapamycin complex, suggests a possible future approach to therapy in FXTAS.

Flatworms are among the best studied animal models for regeneration; however, they also represent an emerging opportunity to investigate other biological processes as well. For instance, flatworms are nocturnal and sleep during the day, a state that is regulated by sleep/wake history and the action of the sleep‐promoting neurotransmitter gamma‐aminobutyric acid (or GABA). Sleep is widespread across the animal kingdom, where it serves many nonexclusive functions. Notably, sleep saves energy by reducing metabolic rate and by not doing something more energetically taxing. Whether the conservation of energy is apparent in sleeping flatworms is unclear. We measured the oxygen consumption rate (OCR) of flatworms dosed with either (1) GABA (n = 29) which makes flatworms inactive or (2) dopamine (n = 20) which stimulates flatworms to move, or (3) day and night neurotransmitter‐free controls (n = 28 and 27, respectively). While OCR did not differ between the day and night, flatworms treated with GABA used less oxygen than those treated with dopamine, and less than the day‐time control. Thus, GABA affected flatworm physiology, ostensibly by enforcing energy‐conserving sleep. Evidence that dopamine increased metabolism was less strong. This work broadens our understanding of flatworm physiology and expands the phylogenetic applicability of energy conservation as a function of sleep. The fragile X premutation (PM) allele contains a CGG expansion of 55-200 repeats in the FMR1 gene's promoter. Male PM carriers have an elevated risk of developing neurological and psychiatric changes, including an approximately 50% risk of the fragile X-associated tremor/ataxia syndrome (FXTAS). The aim of this study was to assess the relationships of regional white matter hyperintensities (wmhs) semi-quantitative scores, clinical status, motor (UPDRS, ICARS, Tremor) scales, and cognitive impairments, with FMR1-specific genetic changes, in a sample of 32 unselected male PM carriers aged 39-81 years. Half of these individuals were affected with FXTAS, while the non-FXTAS group comprised subcategories of non-affected individuals and individuals affected with non-syndromic changes. The dynamics of pathological processes at the cellular level relevant to the clinical status of PM carriers was investigated using the enzyme AMP-activated protein kinase (AMPK), which is a highly sensitive cellular stress-sensing alarm protein. This enzyme, as well as genetic markers - CGG repeat number and the levels of the FMR1 mRNA - were assessed in blood lymphoblasts. The results showed that the repeat distribution for FXTAS individuals peaked at 85-90 CGGs; non-FXTAS carriers were distributed within the lowest end of the PM repeat range, and non-syndromic carriers assumed an intermediate position. The size of the CGG expansion was significantly correlated, across all three categories, with infratentorial and total wmhs and with all motor scores, and the FMR1 mRNA levels with all the wmh scores, whilst AMPK activity showed considerable elevation in the non-FXTAS combined group, decreasing in the FXTAS group, proportionally to increasing severity of the wmhs and tremor/ataxia. We conclude that the size of the CGG expansion relates to the risk for FXTAS, to severity of infratentorial wmhs lesions, and to all three motor scale scores. FMR1 mRNA shows a strong association with the extent of wmhs, which is the most sensitive marker of the pathological process. However, the AMPK activity findings - suggestive of a role of this enzyme in the risk of FXTAS - need to be verified and expanded in future studies using larger samples and longitudinal assessment.

While governments and individuals strive to maintain the availability of high-quality water resources, many factors can "change the landscape" of water availability and quality, including drought, climate change, saltwater intrusion, aquifer depletion, population increases, and policy changes. Specialty crop producers, including nursery and greenhouse container operations, rely heavily on available high-quality water from surface and groundwater sources for crop production. Ideally, these growers should focus on increasing water application efficiency through proper construction and maintenance of irrigation systems, and timing of irrigation to minimize water and sediment runoff, which serve as the transport mechanism for agrichemical inputs and pathogens. Rainfall and irrigation runoff from specialty crop operations can contribute to impairment of groundwater and surface water resources both on-farm and into the surrounding environment. This review focuses on multiple facets of water use, reuse, and runoff in nursery and greenhouse production including current and future regulations, typical water contaminants in production runoff and available remediation technologies, and minimizing water loss and runoff (both on-site and off-site). Water filtration and treatment for the removal of sediment, pathogens, and agrichemicals are discussed, highlighting not only existing understanding but also knowledge gaps. Container-grown crop producers can either adopt research-based best management practices proactively to minimize the economic and environmental risk of limited access to high-quality water, be required to change by external factors such as regulations and fines, or adapt production practices over time as a result of changing climate conditions.

Lime reaction rate in a container substrate is influenced by temperature and moisture, which are factors that vary between batches of substrate during manufacturing, storage, and crop production. The effects of temperature and moisture on the duration required to achieve a stable substrate-pH are useful information for substrate companies and growers, as well as a key component when modeling lime reaction over time. The pH of a 70 peat : 30 perlite (by volume) substrate was quantified over time under a range of different storage temperature (1.9 to 33.3°C) and substrate volumetric water content (VWC, 0.168 to 0.568 L of H2O/L of substrate). The lime source was a horticultural dolomitic carbonate limestone screened to the fraction that passed through a 100 US mesh but was retained on a 200 US mesh (0.075–0.15 mm) incorporated at 2.67 g·L−1 of substrate. Experiments provided two data sets for calibration and validation. Lime reaction rate increased with increasing substrate temperature and substrate moisture level. The duration required to reach a target substrate-pH value of 6.0 was used to indicate 90% of maximum pH effect from lime. Duration varied from 4 days with the combined high temperature (20.6 and 33.3 °C) and high VWC (0.468 and 0.568 L H2O/L of substrate) to 53 days with low temperature (1.9 °C) and low VWC (0.168 L H2O/L of substrate) for the calibration data set. At an example low VWC of 0.168 L of H2O/L of substrate, the duration required to reach substrate-pH 6.0 at 1.9, 7.8, 10, 20.6, and 33.3°C was 53, 38, 34, 23, and 17 days, respectively. Similarly, if the temperature was held constant at 33.3°C, reducing VWC from 0.568 L H2O/L to 0.128 L H2O/L would decrease lime reaction rate from 100% to 21%, requiring five times the duration to reach an equilibrium pH. Results can be used to compare the relative effects of moisture and temperature on lime reaction rate for substrate manufacturers and growers. [ABSTRACT FROM AUTHOR]

The main purpose of this study was to improve the accuracy and reliability of nutrient analysis on soilless substrates by comparing two different saturated medium extract (SME) methods, using either water (SMEW) or DTPA (diethylenetriamine-pentaacetic acid, SMED) as extractants. A series of analyses were performed to compare ion levels quantified from either SMEW or SMED. Substrate pH, electrical conductivity (EC) and nutrient data were analyzed from a survey of substrate components (coconut coir, peat, bark, perlite, vermiculite), and blended greenhouse propagation substrates using the SMEW and SMED methods. In addition, a series of SMEW and SMED extracted samples were sent to three commercial soil testing laboratories for nutrient analysis. The objectives of these analyses were to (1) validate that published relationships between SMEW and SMED methods were consistent with correlation curves for pH, EC and nutrient levels across a wide range of soilless substrates, (2) compare statistical variability of results from SMEW and SMED methods and (3) compare variability in results between different laboratories. The results validated that previously published standards for interpreting SMEW extractions can be used for SMED method for macronutrients and EC due to an approximately 1:1 relationship for test results between extraction methods. Up to 2 orders of magnitude higher levels of micronutrients were extracted for iron (Fe), manganese (Mn), zinc (Zn), and copper (Cu) using SMED compared with SMEW, with low correlation between extractants. Concentrations of ions from SMED extractions were less variable than SMEW for micronutrient analysis but were comparable for macronutrient analysis. Nutrient solution pH and water quality are challenges faced by small-scale hydroponic growers. The objective was to evaluate the use of the pH buffer 2-(N-morpholino) ethanesulfonic acid (MES) as an initial dose to simplify pH management in small-scale hydroponic systems, and its effectiveness within the range of alkalinity that commonly occurs in irrigation water. Two experiments tested pH buffering capacity in response to acid-base titrations, and when growing compact tomatoes (Lycopersicon esculentum) over a nine-week cycle. MES was applied at 0 to 10 mM in water and nutrient solutions at five water qualities ranging from 4 to 295 mg.L−1 CaCO3 alkalinity. Solution buffering increased with addition of fertilizer, increasing doses of MES, and increasing water alkalinity. The MES decreased pH variability over time in all water qualities, without affecting plant growth or causing nutritional disorders. The pH of solutions without MES ranged from 4.5 to 8.0 over the growing cycle, whereas in 10 mM MES solutions it ranged from 5.7 to 7.0 for alkalinities 0 to 146 mg.L−1 CaCO3 and reached 7.6 for 295 mg.L−1 CaCO3. Titration of nutrient solutions after tomato growth indicates that MES acid-buffering capacity was partially consumed along the trial, which was probably the result of MES decomposition or interaction with acids and bases in solution. Results indicate that a single 10 mM MES application could be used by small-scale hydroponic growers to provide acceptable pH buffering for low to moderate alkalinity levels, but initial pH adjustment may be necessary and solution pH may still increase with high alkalinity.
