Abstract

Presents a study to clarify the temporal and causal relationships between mitogen-activated protein (MAP) kinase and maturation-promoting factor (MPF) during meiotic maturation, particularly during the resumption of meiosis. Use of pig oocytes matured synchronously in culture; Analysis of kinase activities; Demonstration that MAP kinase is activated simultaneously with MFP at germinal vesicle breakdown; Indication that its activation is not essential for the activation of MPF nor for the resumption of the first meiosis in pig oocytes.

Abstract

Ovine interferon tau (oIFN-τ) is an embryonic protein of critical importance in the establishment of pregnancy in the sheep. We have produced recombinant (r) oIFN-τ using a baculovirus expression system and demonstrated the biological activity of the protein produced. Bombyx mori larvae were infected with B. mori nuclear polyhedrosis virus (BmNPV), modified by inserting a cDNA coding for oIFN-τ downstream of the strong polyhedron promoter. Following infection, antiviral activity of the haemolymph rose to a maximum of 3.6 x 108 u/mL (equivalent to 3 mg roIFN-τ/mL) by day 5, when haemolymph was collected and stored frozen. Control haemolymph, collected from uninfected insects at an equivalent time, contained no antiviral activity. The roIFN-τ was partially purified by gel filtration column chromatography and the presence of roIFN-τ confirmed by western blotting. The biological activity of the partially purified roIFN-τ was tested in ewes. Treatment with roIFN-τ caused a significant delay in luteolysis confirming biological potency. 45. Sheep antiluteolytic interferon: cDNA sequence and analysis of mRNA levels

Abstract

A cloned cDNA has been isolated by probing a sheep blastocyst cDNA library using a synthetic oligonucleotide representing the N-terminal amino acid sequence of the antiluteolytic protein, ovine trophoblast protein-1. Sequence analysis of the cDNA confirms the 70% homology between the antiluteolysin and the interferon-α family of proteins; however, the sequence reported here differs at several points from previously reported amino acid and cDNA sequences for the antiluteolysin. In-vitro translation of day-16 poly(A)+RNA indicated that antiluteolysin mRNA is a major constituent of total mRNA at this stage of blastocyst development, and Northern blotting confirmed that antiluteolysin mRNA production occurred between days 13 and 22 after oestrus. This is consistent with the stage at which embryonic extracts are antiluteolytic on administration in vivo. These and other data confirm that the ovine trophoblast antiluteolysin is an interferon, and suggest that at least five isoforms of this protein may exist.

48. Ovarian steroid metabolism and oestrogens in the corpus luteum of the tammar wallaby

Abstract

Ovaries were obtained from tammar wallabies at various stages of the reproductive cycle to examine the occurrence of oestrogens in corpora lutea, and the synthesis and metabolism of steroids in the corpus luteum and ovarian cortical and interstitial tissues. Corpora lutea contained oestradiol-17β and oestrone during embryonic diapause and at all stages of pregnancy studied after blastocyst activation. Aryl sulphatase, 3β-hydroxysteroid dehydrogenase and 17β-oxidoreductase were shown to be present in luteal and other ovarian tissues by incubation in vitro with labelled substrates. Aromatase was undetectable in corpora lutea or in interstitial tissue, but was present in the ovarian tissues (including follicles) which remained after removal of corpora lutea. The probable source of the oestrogens detected in the corpus luteum is discussed in relation to their role in the inhibition of follicular development during embryonic diapause.
