Abstract

IntroductionChildren with high-grade gliomas (pHGGs) represent a clinical population in substantial need of new therapeutic options given the inefficacy and toxicity of current standard-of-care modalities. Although immunotherapy has emerged as a promising modality, it has yet to elicit a significant survival benefit for pHGG patients. While preclinical studies address a variety of underlying challenges, translational clinical trial design and management also need to reflect the most updated progress and lessons from the field.Areas coveredThe authors will focus our discussion on the design of clinical trials, the management of potential toxicities, immune monitoring, and novel biomarkers. Clinical trial design should integrate appropriate patient populations, novel, and preclinically optimized trial design, and logical treatment combinations, particularly those which synergize with standard of care modalities. However, there are caveats due to the nature of immunotherapy trials, such as patient selection bias, evidenced by the frequent exclusion of patients on high-dose corticosteroids. Robust immune-modulating effects of modern immunotherapy can have toxicities. As such, it is important to understand and manage these, especially in pHGG patients.Expert opinionAdequate integration of these considerations should allow us to effectively gain insights on biological activity, safety, and biomarkers associated with benefits for patients.

17. Methodological Challenges in Conducting Cross-Cultural/Language Research with Spanish Speakers: The Role of the Researcher/Translator.

Abstract

With the increase in global research, it is common for researchers to investigate topics in intercultural settings, both in their own home countries and abroad. Although findings from this research are prolific, rarely are detailed examples given or practical suggestions offered, particularly in relation to the role of the translator/researcher. The significant and often undervalued role of the translator/researcher in cross cultural/language qualitative research warrants methodological considerations at the onset and throughout the research. Nonetheless, few qualitative studies transparently report the process of how the translation findings were developed. This paper addresses this gap by examining a Latinx postgraduate student's role in the qualitative research on teacher induction in Chile. I used reflexivity to assess my positionality of insiderness and outsiderness and its influence in the process of recruitment, conducting interviews, transcription, and translation. Interviews were conducted, transcribed and analysed in Spanish. Examples of data translation that can help to identify the main issues associated with reporting the findings in English are provided. These applied examples are used to illustrate the gaps and misinterpretations possible in intercultural research. 18. Beginning Teachers' Induction in Chile: The Perspective of Principals

Abstract

This case study investigated the role of school principals in the induction of beginning teachers in Copiapó, Chile. Building upon group and individual interviews, and review of extant literature, the following findings were established: (a) principals in this study expect beginning teachers to be fully formed as classroom teachers; (b) principals were unlikely to talk about induction practices that might help beginning teachers to learn pedagogical strategies for classrooms; and (c) principals' induction practices focused on the symbolic role that principals play as the highest authority within the school which has little practical influence on helping beginning teachers to develop their pedagogy. Implications for policy and practice are discussed.

19. Characterization of a stem cell population derived from human peripheral blood and its therapeutic potential in brain tumors

Abstract

Adherent CD34+ cells are a stem cell enriched population with a high frequency of primitive stem cells that are genetically primed to differentiate into tissue-specific lineages. These cells make up the adherent CD34+ cells in peripheral blood which are the focus of this study. The first aim of this study was to transcriptionally characterize adherent CD34+ cells relative to nonadherent CD34+ cells using a whole human genome Affymetrix U133 plus 2 array. The subsequent analysis demonstrated transcriptional differences in genes which are involved in quiescence, cell cycle, homing and adhesion, which are likely to be relevant to the suitability of adherent CD34+ cells for clinical application. To verify the microarray analysis, a selection of representative transcripts was chosen and their relative expression was compared using real-time qPCR. To analyze further the stem cell characteristics of adherent CD34+ cells, the DNA replication timing kinetics of pluripotency-associated genes in bone marrow-derived adherent CD34+ cells, non-adherent CD34+ cells, and foetal mesenchymal stem cells were conducted and compared to those of embryonic stem (ES) cells. This showed that pluripotency-associated genes Oct4 and Nanog replicate early in the cell cycle similar to ES cells. The second aim of this study was to examine the potential use of adherent CD34+ cells in neural injury. As adherent CD34+ cells are primitive stem cells, it was reasonable to hypothesize that these cells would differentiate into neural cells in vitro, but these experiments revealed that these cells are not neurogenic and, in our hands, do not differentiate into neural cells. The experiments revealed that adherent CD34+ cells differentiate efficiently and reproducibly into microglia-like cells (CD34MG cells). CD34MG cells upregulated microglia-associated antigens and demonstrated functional characteristics of microglia cells including TNF and IL-6 release in response to lipopolysaccharide. CD34MG also stimulate responders in a mixed lymphocyte reaction, suggesting antigen presenting capabilities. To determine if these cells might be exploited for use in brain tumour therapy, it was shown that CD34MG migrate and invade in response to brain tumour glioma cells. Results also suggest that these cells are capable of presenting glioma specific antigen to immune cells in an autologous lymphocyte reaction in vitro. The combined results suggest that CD34MG cells might be exploited for therapeutic purposes in brain tumours. Stem cells are currently being explored for their therapeutic potential. This study demonstrates that adherent CD34+ cells and CD34MG cells may be useful for clinical use.

Abstract

This research aimed to determine the effect of innovative leadership on teachers’ job satisfaction as mediated by the role of a supportive environment. The study was guided by the following hypotheses: (1)The Supportive Environment does not significantly mediate the relationship between the extent of Innovative Leadership and the level of Teachers’ Job Satisfaction with Operating Procedures, and (2) The Supportive Environment does not significantly mediate the relationship between the extent of Innovative Leadership and the level of Teachers’ Job Satisfaction on Performance-Based Rewards. In this study, the descriptive-correlational research design was used and quantitative research was conducted. A total of 165 teacher-respondents in 11 elementary and 3 secondary schools in the District of Alaminos, Laguna. The researcher adapted and modified survey questionnaires from related studies. Research findings revealed that there is a significant relationship between the extent of innovative leadership on teachers’ job satisfaction. The correlation between innovative leadership and teachers’ job satisfaction with operating procedures and performance-based rewards was found to have manifested a strong significant relationship. Moreover, findings also suggest that a supportive environment partially mediates the relationship between the extent of innovative leadership and the level of teachers’ job satisfaction. This study provides useful findings which have implications for school leaders and future researchers.
