CoSAXS is a state-of-the-art SAXS/WAXS beamline exploiting the high brilliance of the MAX IV 3 GeV synchrotron. By coupling advances in sample environment control with fast X-ray detectors, millisecond time-resolved scattering methods can follow structural dynamics of proteins in solution. In the present work, four sample environments are discussed. A sample environment for combined SAXS with UV–vis and fluorescence spectroscopy (SUrF) enables a comprehensive understanding of the time evolution of conformation in a model protein upon acid-driven denaturation. The use of microfluidic chips with SAXS allows the mapping of concentration with very small sample volumes. For highly reproducible sequences of mixing of components, it is possible using stopped-flow and SAXS to access the initial effects of mixing at 2 millisecond timescales with good signal to noise to allow structural interpretation. The intermediate structures in a protein are explored under light and temperature perturbations by using lasers to “pump” the protein and SAXS as the “probe”. The methods described demonstrate that features at low q, corresponding to cooperative motions of the atoms in a protein, could be extracted at millisecond timescales, which results from CoSAXS being a highly-stable, low background, dedicated SAXS beamline.

Native mucus is heterogeneous, displays high inter-individual variation and is prone to changes during harvesting and storage. To overcome the lack of reproducibility and availability of native mucus, commercially available purified mucins, porcine gastric mucin (PGM) and mucin from bovine submaxillary gland (BSM), have been widely used. However, the question is to which extent the choice of mucin matters in studies of their interaction with polymers as their composition, structure and hence physicochemical properties differ. Accordingly, the interactions between PGM or BSM with two widely used polymers in drug delivery, polyethylene oxide and chitosan, was studied with orthogonal methods: turbidity, dynamic light scattering, and quartz crystal microbalance with dissipation monitoring. Polymer binding and adsorption to the two commercially available and purified mucins, PGM and BSM, is different depending on the mucin type. PEO, known to interact weakly with mucin, only displayed limited interaction with both mucins as confirmed by all employed methods. In contrast, chitosan was able to bind to both PGM and BSM. Interestingly, the results suggest that chitosan interacts with BSM to a greater extent than with PGM indicating that the choice of mucin, PGM or BSM, can affect the outcome of studies of mucin interactions with polymers.

Plant protein-based nanofibers generated by eco-friendly waterborne electrospinning are emerging as sustainable and innovative materials with vast applications in different biomedical areas. In this study, we fabricated electrospun nanofibers based on potato, pea, and soy protein isolates, achieving remarkably high protein content without the use of organic solvents, strong bases, or surfactants. The different protein nanofibers were characterized by means of quantitative fluorescence imaging, optical spectroscopy, and dynamic mechanical analysis. Results indicated that the intrinsic nature of the proteins modulated the properties of the nanofibers in terms of morphology, fluorescence fingerprints, mechanical strength, and stability in aqueous environments. Pea and soy protein isolates, both rich in beta-structure, led to the formation of robust and dense nanofibers, which slowly disintegrated in water. On the contrary, less dense and highly soluble nanofibers were generated from the structurally more flexible potato protein isolate, and these nanofibers demonstrated lower resistance to breakage. Our findings indicate the importance of protein structural elements when designing protein-based electrospun nanofibers with specific features. Deciphering the intricate relationship between protein structure at the molecular level and properties of nanofiber holds promise for the development of biomaterials with enhanced efficacy in diverse biomedical applications.

Amyloid aggregation is associated with many diseases and may also occur in therapeutic protein formulations. Addition of co-solutes is a key strategy to modulate the stability of proteins in pharmaceutical formulations and select inhibitors for drug design in the context of diseases. However, the heterogeneous nature of this multicomponent system in terms of structures and mechanisms poses a number of challenges for the analysis of the chemical reaction. Using insulin as protein system and polysorbate 80 as co-solute, we combine a spatially resolved fluorescence approach with single molecule microscopy and machine learning methods to kinetically disentangle the different contributions from multiple species within a single aggregation experiment. We link the presence of interfaces to the degree of heterogeneity of the aggregation kinetics and retrieve the rate constants and underlying mechanisms for single aggregation events. Importantly, we report that the mechanism of inhibition of the self-assembly process depends on the details of the growth pathways of otherwise macroscopically identical species. This information can only be accessed by the analysis of single aggregate events, suggesting our method as a general tool for a comprehensive physicochemical characterization of self-assembly reactions.

Low-frequency Raman (LFR) spectroscopy is presented as a viable tool for studying the hydration characteristics of lyotropic liquid crystal systems herein. Monoolein was used as a model compound, and its structural changes were probed both in situ and ex situ which enabled a comparison between different hydration states. A custom-built instrumental configuration allowed the advantages of LFR spectroscopy to be utilized for dynamic hydration analysis. On the other hand, static measurements of equilibrated systems (i.e., with varied aqueous content) showcased the structural sensitivity of LFR spectroscopy. The subtle differences not intuitively observed between similar self-assembled architectures were distinguished by chemometric analysis that directly correlated with the results from small-angle X-ray scattering (SAXS), which is the current “gold standard” method for determining the structure of such materials.
