Abstract

Introduction and Objectives Nutritional assessment in individuals infected with HIV-1 holds significance as it allows for indirect monitoring of physiological and morphological changes, identification of nutritional risks, and effective intervention to maintain or restore nutritional status. This, in turn, contributes to enhancing the quality of life for this population. This study aims to establish a connection between self-image perceptions and body composition, biochemical profiles, and the use of lipid-lowering medications among adults with HIV who are undergoing outpatient follow-up in São Paulo, Brazil. Methods A retrospective cross-sectional study was conducted involving 231 adults, both on and off antiretroviral drugs. The study subjectively assessed two aspects: (1) alterations in the redistribution of body fat (referred to as self-reported lipodystrophy) and (2) self-perceptions of body image. These variables were subsequently correlated with the individuals' self-perceived body image. Results The study included 67% (n = 154) men and 33% (n = 77) women, with 28.5% (n = 66) having been exposed to some form of protease inhibitor. Among the participants, 40% (n = 98) perceived alterations in the redistribution of body fat (lipodystrophy). Notable changes included fat loss in the face, arms, thighs, and buttocks (p = 0.004, 0.006, 0.001, and 0.001, respectively), along with increased waistlines (p = 0.001), prominent veins (p = 0.001), and the presence or increase of lipomas (p = 0.046). Conclusion A positive correlation was observed between the use of lipid-lowering medications and self-perceptions of body image. In this study, anthropometric and biochemical parameters proved more effective in assessing changes in the distribution of body fat among people living with HIV/AIDS compared to the subjective self-reported method. Abstract

Lung cancer is one of the leading causes of death in the world. Some tumor events are attributed to an important group of molecules (cadherins and integrins). We evaluated the interactions of cell adhesion molecules in cell lines from lung cancer. Two lung cancer cell lines were nonmetastatic (H358 and H441) and two were metastatic (H1299 and H292). All cell lines were treated with epidermal growth factor (EGF), and Western blot analysis was performed to assess the interactions between these proteins. The bronchoalveolar cells H358 showed the three analyzed proteins: E-cadherin, β-catenin, and p120 catenin. The adenocarcinoma cells H441 did not present p120 catenin, and carcinoma cells did not show E-cadherin (H1299) or p120 catenin (H292). FAK (pTyr925) was dephosphorylated in adenocarcinoma cells H441, absent in carcinoma cells H1299, and upregulated in the other carcinoma cells H292. p130Cas showed no difference when the cell lines were treated with EGF for 30 min; it was absent in the metastatic carcinoma cells H1299. Paxillin was dephosphorylated in adenocarcinoma cells H441 and also absent in other metastatic carcinoma cells H292. Vinculin showed the same results, and talin was downregulated in adenocarcinoma cells H441 when the cells were treated with EGF. Rap1 was downregulated and PYK2 was upregulated in the same cell line. Our data help to comprehend the mechanism involved in cell migration to the blood and metastasis generation. In conclusion, the expression patterns of cell-cell adhesion were not affected by EGF treatment but it affected cell-extracellular matrix adhesion. No. of bitstreams: 0 Previous issue date: 2012-03-07 Painful bladder syndrome associated with interstitial cystitis (PBS/IC) is a clinical condition characterized pelvic pain, urinary urgency, and urinary frequency. In this study, 22 patients were assigned to make two visits over a three weeks period. The patients were randomly, double-blinded assigned in two groups. The first group received Cystex® capsules. The second group received placebo capsules. Two capsules were taken three times a day away from meals. The change from baseline in the O'Leary-Sant IC symptom and problem index was the primary outcome parameter. Changes in functional bladder capacity and intensity of pain and urgency have been chosen as secondary outcome parameters. Mood as well as physical and sexual activity were rated by 10 questions on a scale 0 to 6. The ratings were analyzed and the average for each patient in both groups Cystex® and placebo was determined as the quality of life index. For the primary outcome there was a statistically significant difference between the groups. Mean symptom score-sum decreased from 28.4 to 20.5 in the Cystex® group compared with 29.5 to 26.8 in the placebo group (p0.05). In our study, Cystex® enhanced quality of life over the placebo showing a statistically significant. This trial have shown that the efficacy and safety of therapy with Cystex® in the treatment of interstitial cystitis and is an alternative for patients suffering from this pathology. Therefore, it can be concluded that the composition of Cystex®, increased the quality of life in treated patients. 45. Peripheral Blood c-erbB-2Expression by Reverse Transcriptase—Polymerase Chain Reaction in Breast Cancer Patients Receiving Chemotherapy

Abstract

Minimal residual disease (MRD) evaluation in breast cancer patients is a promising tool to improve current staging procedures. In a previous work employing a CK-19—based reverse transcriptase—polymerase chain reaction (RT-PCR) technique for MRD detection, we identified a group of women who exhibited persistent negativity for this assay and for whom this technique was considered noninformative. In order to improve the yield of MRD detection in these patients, we evaluated the usefulness of RT-PCR detection of c-erbB-2 expression. We were able to detect up to 1 MCF-7 cell (positive for c-erbB-2expression) in a mixture of 1,000,000 CCRF-CEM cells (negative for c-erbB-2expression). We evaluated the specificity of this technique in the peripheral-blood mononuclear cells (PBMCs) of 20 healthy women and found that 2 of these women were positive for c-erbB-2expression. In the PBMCs of a group of 16 women with breast cancer, 25% of the samples were positive for c-erbB-2expression before chemotherapy. Except for race (P= 0.017), no other significant correlations were found, including c-erbB-2expression in the primary tumor by immunoperoxidase. Interestingly, in the subgroup of 6 patients for whom this technique was informative, we found that 80% of the samples obtained while on chemotherapy were negative compared to only 10% obtained off treatment (P= 0.017). Additionally, 2 patients for whom CK-19expression was noninformative had at least 1 c-erbB-2—positive sample. We conclude that this technique might be useful for MRD detection in breast cancer patients, but further studies are necessary to confirm our findings.
