Digital ulcers (DUs) are frequent manifestations of systemic scleroderma (SSc). This study assessed functional limitations due to DUs among patients enrolled in the Digital Ulcer Outcome (DUO) Registry, an international, multicentre, observational registry of SSc patients with DU disease.Patients completed at enrolment a DU-specific functional assessment questionnaire with a 1-month recall period, measuring impairment in work and daily activities, and hours of help needed from others. Physician-reported clinical parameters were used to describe the population. For patients who completed at least part of the questionnaire, descriptive analyses were performed for overall results, and stratified by number of DUs at enrolment.This study included 2327 patients who completed at least part of the questionnaire. For patients with 0, 1-2, and ≥3 DUs at enrolment, mean overall work impairment during the prior month among employed/self-employed patients was 28%, 42%, and 48%, respectively. Across all included patients, ability to perform daily activities was impaired on average by 35%, 54%, and 63%, respectively. Patients required a mean of 2.0, 8.7, and 8.8 hours of paid help and 17.0, 35.9, and 63.7 hours of unpaid help, respectively, due to DUs in the prior month. Patients with DUs had more complications and medication use than patients with no DUs.With increasing number of DUs, SSc patients reported more impairment in work and daily activities and required more support from others.

- Abstract
Escalating rates of suicide among U.S. military Veterans have prompted the Department of Veterans Affairs to prioritize Veteran suicide as a chief clinical concern. Veterans Affairs-funded research is consistently dedicated to suicide prevention initiatives, reflecting a commitment to addressing this urgent issue. Although general guidelines have been proposed for recognizing and responding to suicide risk among research participants, to date, no guidelines have been published that are Veteran specific. Veterans exhibit unique suicide risk factors compared to civilians, including higher rates of suicide, a tendency to utilize more lethal means when attempting suicide, and substantial stigma surrounding mental health and help seeking, underscoring the need for Veteran-specific suicide risk assessment and management protocols (SRAMs). This article offers a comprehensive SRAM to guide research with Veteran participants. The protocol provides guidance on (a) accurate assessment of suicide risk, (b) risk management strategies commensurate to presenting risk, and (c) tailoring SRAMs for diverse study designs and contexts. Low-pass genome sequencing is cost-effective and enables analysis of large cohorts. However, it introduces biases by reducing heterozygous genotypes and low-frequency alleles, impacting subsequent analyses such as model-based demographic history inference. Several approaches exist for inferring an unbiased allele frequency spectrum (AFS) from low-pass data, but they can introduce spurious noise into the AFS. Rather than correcting the AFS, here, we developed an approach that incorporates low-pass biases into the demographic modeling and directly analyzes the AFS from low-pass data. Our probabilistic model captures biases from the Genome Analysis Toolkit multisample calling pipeline, and we implemented it in the population genomic inference software dadi. We evaluated the model using simulated low-pass datasets and found that it alleviated low-pass biases in inferred demographic parameters. We further validated the model by downsampling 1000 Genomes Project data, demonstrating its effectiveness on real data. 6. Physiological adaptations of red blood cells during aestivation in the south American lungfish Lepidosiren paradoxa.

- Abstract
The South American lungfish Lepidosiren paradoxa inhabits areas with variable pluvial regimes. During aestivation (dormancy state observed in some species during dry seasons), the prolonged period of dryness imposes osmotic stress. We aimed to investigate the physiological and morphological adaptations of RBCs in Lepidosiren paradoxa during aestivation. Here, the lungfish were subjected to aestivation for 20 and 40 days and compared to a control group in an active period. The osmotic fragility, blood osmolality, and pH were measured. Blood smears were performed to assess morphological changes in the RBCs. Lepidosiren paradoxa presented lower hemolysis when compared with a teleost fish and tegu lizard. Hemolysis increased when the lungfish was subjected to 40 days of aestivation (from 6.04 % to 16.51 %; control vs 40-day aestivation). Hematocrit rose in both aestivation groups compared to the control (26.36 %, 41.36 % and 41 %, control, 20 and 40 days, respectively; p < 0.05) indicating changes in RBC volume and hydration status. Moreover, the results revealed increased osmolality in the 40-day aestivation group (244.4 vs 372.1 mmol/Kg; control vs 40-day aestivation). 40 days of aestivation led to a decrease in blood pH when compared to the control and 20-day aestivation groups. Both aestivation durations resulted in a reduction in the perimeter and cell diameters in at least one direction of the RBCs (24 % mean reduction in size from control to 40 days aestivation). The prevalence of panic disorder is two to four times higher in women compared to that in men, and hormonal changes during the menstrual cycle play a role in the occurrence of panic attacks. Here, we investigated the effect of the estrous cycle on the ventilatory and behavioral responses to CO
2 in mice. Female mice in proestrus, estrus, metestrus, or diestrus were exposed to 20% CO2 , and their escape behaviors, brain monoamines, and plasma levels of 17β-estradiol (E2 ) and progesterone (P4 ) were measured. Pulmonary ventilation (V̇E ), oxygen consumption (V̇O2 ), and body core temperature (TB ) were also measured during normocapnia followed by CO2 . Females exposed to 20% CO2 exhibited an escape behavior, but the estrous cycle did not affect this response. Females in all phases of the estrous cycle showed higher V̇E and lower TB during hypercapnia. In diestrus, there was an attenuation of CO2 -induced hyperventilation with no change in V̇O2 , whereas in estrus, this response was accompanied by a reduction in V̇O2 . Hypercapnia also increased the concentration of plasma P4 and central DOPAC, the main dopamine metabolite, in all females. There was an estrous cycle effect on brainstem serotonin, with females in estrus showing a higher concentration than females in the metestrus and diestrus phases. Therefore, our data suggest that hypercapnia induces panic-related behaviors and ventilatory changes that lead to an increase in P4 secretion in female mice, likely originating from the adrenals. While genetic variation in any species is potentially shaped by a range of processes, phylogeography and landscape genetics are largely concerned with inferring how environmental conditions and landscape features impact neutral intraspecific diversity. However, even as both disciplines have come to utilize SNP data over the last decades, analytical approaches have remained for the most part focused on either broad-scale inferences of historical processes (phylogeography) or on more localized inferences about environmental and/or landscape features (landscape genetics). Here we demonstrate that an artificial intelligence model-based analytical framework can consider both deeper historical factors and landscape-level processes in an integrated analysis. We implement this framework using data collected from two Brazilian anurans, the Brazilian sibilator frog (Leptodactylus troglodytes) and granular toad (Rhinella granulosa). Our results indicate that historical demographic processes shape most the genetic variation in the sibulator frog, while landscape processes primarily influence variation in the granular toad. CO
2 exposure has been used to investigate the panicogenic response in patients with panic disorder. These patients are more sensitive to CO2 , and more likely to experience the "false suffocation alarm" which triggers panic attacks. Imbalances in locus coeruleus noradrenergic (LC-NA) neurotransmission are responsible for psychiatric disorders, including panic disorder. These neurons are sensitive to changes in CO2 /pH. Therefore, we investigated if LC-NA neurons are differentially activated after severe hypercapnia in mice. Further, we evaluated the participation of LC-NA neurons in ventilatory and panic-like escape responses induced by 20% CO2 in male and female wild type mice and two mouse models of altered LC-NA synthesis. Hypercapnia activates the LC-NA neurons, with males presenting a heightened level of activation. Mutant males lacking or with reduced LC-NA synthesis showed hypoventilation, while animals lacking LC noradrenaline present an increased metabolic rate compared to wild type in normocapnia. When exposed to CO2 , males lacking LC noradrenaline showed a lower respiratory frequency compared to control animals. On the other hand, females lacking LC noradrenaline presented a higher tidal volume. Nevertheless, no change in ventilation was observed in either sex. CO2 evoked an active escape response. Mice lacking LC noradrenaline had a blunted jumping response and an increased freezing duration compared to the other groups. They also presented fewer racing episodes compared to wild type animals, but not different from mice with reduced LC noradrenaline. One key research goal of evolutionary biology is to understand the origin and maintenance of genetic variation. In the Cerrado, the South American savanna located primarily in the Central Brazilian Plateau, many hypotheses have been proposed to explain how landscape features (e.g., geographic distance, river barriers, topographic compartmentalization, and historical climatic fluctuations) have promoted genetic structure by mediating gene flow. Here, we asked whether these landscape features have influenced the genetic structure and differentiation in the lizard species Norops brasiliensis (Squamata: Dactyloidae). To achieve our goal, we used a genetic clustering analysis and estimate an effective migration surface to assess genetic structure in the focal species. Optimized isolation-by-resistance models and a simulation-based approach combined with machine learning (convolutional neural network; CNN) were then used to infer current and historical effects on population genetic structure through 12 unique landscape models. We recovered five geographically distributed populations that are separated by regions of lower-than-expected gene flow. The results of the CNN showed that geographic distance is the sole predictor of genetic variation in N. brasiliensis, and that slope, rivers, and historical climate had no discernible influence on gene flow. Our novel CNN approach was accurate (89.5%) in differentiating each landscape model. CNN and other machine learning approaches are still largely unexplored in landscape genetics studies, representing promising avenues for future research with increasingly accessible genomic datasets., (© 2024. 12. Gingival zenith level, position, and symmetry: A systematic review and meta-analysis.

Statement of Problem: The gingival zenith (GZ) influences the line angle position in the emergence profile and the axial inclination of the crown. Irregularities in GZ symmetry and contour have a negative impact on dental esthetics. GZ location is not consistently distal to the crown's long axis; instead, it tends to be more distal in anterior teeth. The GZ levelling of the lateral incisor is often described as either level with or slightly above the zenith line connecting the central incisor and canine teeth, but there are also reports of GZ being aligned apically to the zenith line. Controversial reports persist regarding GZ positioning, magnitude, and location, potentially leading to inadequate positioning and levelling of the GZ in esthetic restorative therapy., Purpose: The purpose of this systematic review and meta-analysis was to assess the scientific evidence related to GZ level, position, and symmetry in maxillary incisors and canines., Material and Methods: This study adhered to the population, variable, outcome (PVO) criteria and the preferred reporting items for systematic reviews and meta-analysis (PRISMA) checklist. Cross-sectional studies involving healthy adults with complete dentition of the maxillary anterior sextant were subjected to prevalence analysis and quantitative measurement to assess the gingival zenith level and position. Data were extracted, and methodological quality was appraised using the Joanna Briggs Institute critical appraisal checklist for analytical cross-sectional studies. Certainty of evidence was evaluated through the grading of recommendations, assessment, development, and evaluation (GRADE) system., Results: The GZs were distally positioned, coinciding with the tooth axis or mesially positioned in, respectively, 96%, 3%, and 1% on central incisors; 84%, 14%, and 1% on lateral incisors; and 43%, 44%, and 5% on canines. GZ distal positioning was greater in central incisors, followed by lateral incisors and canines. In lateral incisors, the GZ was levelled coronally in 82% of the population. Contralateral symmetry was observed for GZ levelling and positioning. The certainty of the evidence was very low for all comparisons., Conclusions: The frequency and magnitude of the distal position of the GZ increased the more anterior the tooth. Axially, the GZ was frequently levelled coronally to the zenith line. The present study aimed to evaluate concentrations of 16 priority PAHs in the mussel Perna perna and oyster Crassostrea rhizophorae of the Santos Estuary (Brazil) and the potential risk to human health associated with the consumption of these species. The levels of ΣPAHs in the tissues of bivalves ranged from 96.94 to 988.76 μg/kg for mussels and from 88.38 to 138.62 μg/kg for oysters. A general trend of higher concentrations of PAHs was observed according to the increase in log K
ow values in both species. The estimated amount of ΣPAHs via mussel and oyster intake ranged from 1.0 to 3.2 ng/kg and from 0.4 to 1.2 ng/kg of body weight per day, respectively. The local manufacture of advanced pharmaceutical products has been a long-standing objective of health and industry policy in many developing countries, including in Latin America. This strategy has been applied to fight epidemics such as HIV/AIDS, malaria, and the COVID-19 pandemic. However, we still know little about the politics and governance that enable such arrangements, especially when there is no consent from the originator company. This study focuses on the case of Brazil, a country that is well-known for its health-industry policy, which includes the local production of direct-acting antivirals (DAAs), a new treatment for hepatitis C. We seek to explain the factors that have contributed to Brazil's successful production of generic versions of DAAs, and, later, to the decision by the Ministry of Health (MoH) to procure drugs from multinational pharmaceutical companies rather than from local laboratories. A lack of support for domestic production by important stakeholders, the patent holder's attempt to block domestic production and the MoH's adoption of more modern treatment guidelines under a different procurement logic all created an unfavourable environment for local production and procurement of DAAs. Our study draws implications for middle-income countries that wish to produce drugs domestically without voluntary license agreements.

16. Pleistocene glaciations caused the latitudinal gradient of within-species genetic diversity.

- Abstract
Intraspecific genetic diversity is a key aspect of biodiversity. Quaternary climatic change and glaciation influenced intraspecific genetic diversity by promoting range shifts and population size change. However, the extent to which glaciation affected genetic diversity on a global scale is not well established. Here we quantify nucleotide diversity, a common metric of intraspecific genetic diversity, in more than 38,000 plant and animal species using georeferenced DNA sequences from millions of samples. Results demonstrate that tropical species contain significantly more intraspecific genetic diversity than nontropical species. To explore potential evolutionary processes that may have contributed to this pattern, we calculated summary statistics that measure population demographic change and detected significant correlations between these statistics and latitude. We find that nontropical species are more likely to deviate from neutral expectations, indicating that they have historically experienced dramatic fluctuations in population size likely associated with Pleistocene glacial cycles. By analyzing the most comprehensive data set to date, our results imply that Quaternary climate perturbations may be more important as a process driving the latitudinal gradient in species richness than previously appreciated., (© The Author(s) 2023. A screening for microcontaminants performed by gas chromatography detected several microcontaminants in 12 sediment samples from the eutrophic estuary Guanabara Bay (GB) in southeastern Brazil. Bisphenol A (BPA) ranged from 1.4 to 20.3 ng g
-1 , 4-octylphenol, from-1 , 4-nonylphenol, from -1 , gemfibrozil, from -1 , naproxen, from -1 m Ibuprofen, from -1 and diclofenac, from -1 . Among estrogens, estrone, estradiol, ethinylestradiol and estriol were detected, ranging, respectively from -1 , -1 , -1 and -1 . A strong and positive correlation between 4-nonylphenol and estrone and a moderate and positive correlation between bisphenol A and estradiol were noted. 18. Assessing model adequacy for Bayesian Skyline plots using posterior predictive simulation.

- Abstract
Bayesian skyline plots (BSPs) are a useful tool for making inferences about demographic history. For example, researchers typically apply BSPs to test hypotheses regarding how climate changes have influenced intraspecific genetic diversity over time. Like any method, BSP has assumptions that may be violated in some empirical systems (e.g., the absence of population genetic structure), and the naïve analysis of data collected from these systems may lead to spurious results. To address these issues, we introduce P2C2M.Skyline, an R package designed to assess model adequacy for BSPs using posterior predictive simulation. P2C2M.Skyline uses a phylogenetic tree and the log file output from Bayesian Skyline analyses to simulate posterior predictive datasets and then compares this null distribution to statistics calculated from the empirical data to check for model violations. P2C2M.Skyline was able to correctly identify model violations when simulated datasets were generated assuming genetic structure, which is a clear violation of BSP model assumptions. Conversely, P2C2M.Skyline showed low rates of false positives when models were simulated under the BSP model. We also evaluate the P2C2M.Skyline performance in empirical systems, where we detected model violations when DNA sequences from multiple populations were lumped together. Phylogeographic studies base inferences on large data sets and complex demographic models, but these models are applied in ways that could mislead researchers and compromise their inference. Researchers face three challenges associated with the use of models: (i) 'model selection', or the identification of an appropriate model for analysis; (ii) 'evaluation of analytical results', or the interpretation of the biological significance of the resulting parameter estimates, delimitations, and topologies; and (iii) 'model evaluation', or the use of statistical approaches to assess the fit of the model to the data. Harbours are located in major urban centres around the world and are of great economic importance to the cities in their surroundings. However, the intense traffic of boats and ships can generate environmental impacts that can directly affect the local biota as well as the population that lives in surrounding areas. Therefore, this work aimed to analyse the surface sediment of the Niterói Harbour using chemical, biological and micropalaeontological tools to investigate the environmental condition of this important harbour in Rio de Janeiro State. The pseudototal trace metal data analysed in the surface samples showed values far above those of the greater Guanabara Bay background. These data were corroborated by a high mortality rate of Artemia sp. and elevated presence of the bacterium Vibrio fischeri, indicating a high rate of local pollution. Dinoflagellate cysts also showed a direct response to high values of pseudototal trace metals. The data obtained in this study emphasize a need for greater monitoring of ports since the experience gained through this study in a Brazilian harbour can serve as an example for the management of other harbours located in large urban centres around the world., (Copyright © 2022 Elsevier Ltd. - Abstract
Melanoma is a type of skin cancer with low survival rates after it has metastasized. In order to find molecular differences that could represent targets of quercetin in anti-melanoma activity, we have chosen SKMEL-103 and SKMEL-28 melanoma cells and human melanocytes as models. Firstly, we observed that quercetin was able in reducing SKMEL-103 cell viability, but not in SKMEL-28. Besides that, quercetin treatment caused inhibition of AXL in both cell lines, but upregulation of PIM-1 in SKMEL-28 and downregulation in SKMEL-103. Moreover, HIF-1 alpha expression decreased in both cell lines. Interestingly, quercetin was more effective against SKMEL-103 than kinases inhibitors, such as Imatinib, Temsirolimus, U0126, and Erlotinib. Interestingly, we observed that while the levels of succinate dehydrogenase and voltage-dependent anion channel increased in SKMEL-103, both proteins were downregulated in SKMEL-28 after quercetin's treatment. Furthermore, AKT, AXL, PIM-1, ABL kinases were much more active and chaperones HSP90, HSP70 and GAPDH were highly expressed in SKMEL-103 cells in comparison with melanocytes. Our findings indicate, for the first time, that the efficacy of quercetin to kill melanoma cells depends on its ability in inhibiting tyrosine kinase and upregulating mitochondrial proteins, at least when SKMEL-103 and SKMEL-28 cells response were compared., (© 2021 Walter de Gruyter GmbH, Berlin/Boston.)

Contaminant substances consist of chemical elements that present the potential to adversely impact the ecology of the environment, thus representing a threat to local fauna and flora. In this context, heavy metals are critical agents that, depending on the nature and level, are potentially toxic to living organisms. In order to evaluate the bioaccumulation of heavy metals in the Santos estuary and to determine the potential influence of salinity gradient on the heavy metal bioconcentration, the present study measured the concentrations of As, Cd, Pb, Cu, Cr, Fe, Ni, and Zn in two bivalve species (Crassostrea rhizophorae and Perna Perna) sampled at different sites of Santos Bay, in the southeastern region of Brazil. Throughout the study, the "sentinel species" used were effective in bioaccumulating contaminants. In oysters, based on the Brazilian legislation, critical limits were exceeded for As, Zn, Cu, and Cr. In the case of mussels, on the other hand, only for As, Zn, and Cr, the critical limits were overcome. In the present study, obtained data suggested salinity as a determinant parameter in As incorporation processes of bivalve mollusks. PRACTITIONER POINTS: The present study presents important results for the development of environmental management policies in estuarine environments. The present study points out differences between different organisms as biomonitors, providing subsidies for the decision of an effective biomonitoring program. The estuarine channel of Vitória Bay was evaluated regarding bioaccumulation of metals (Al, Ba, Mn, Fe, Zn, Cu, Cr, Pb, Ni, Cd, Hg) and As in mollusks. Mussels from an aquaculture farm and transplanted into the estuary, whereas oysters were collected in situ in the same area. Concentrations of Al, Mn, Fe, Cr and As were higher in P. perna, whereas C. rhizophorae bioaccumulated more Ba, Zn and Cu. Arsenic concentrations in P. perna exceeded the limit of the Brazilian legislation in the outer estuary. Salinity seemed to influence metal uptake differently for each bivalve, with P. perna absorbing more metal at higher salinities and C. rhizophorae in areas of lower salinity. Hazard index (HI) >1 revealed risk for both bivalves for high level consumers. Bottom sediment quality from the Niterói harbor at Guanabara Bay (Rio de Janeiro, Brazil) was evaluated based on concentrations of organic phosphorus (OP) and inorganic phosphorus (IP) and bioavailability of trace metals through BCR fractionation analysis (Zn, Cu, Pb, Cr, Ni, Cd). The study area revealed elevated concentrations of fine sediments and organic matter (TOC: 2.26-7.31%). OP presented extremely elevated concentrations between 0.57 and 47.04 μmol/g, whereas IP reached a maximum concentration of 4.99 μmol/g. The anoxic bottom at the study area was confirmed by high TOC/OP values in most stations. Phosphorus enrichment index (PEI) varied between 0.07 and 2.57, pointing to ecological risk at some stations. Trace metals were mostly bonded to the bioavailable fractions (exchangeable, reducible, oxidizable), and decreasing order of mobility was Zn > Cu > Pb > Cr > Ni > Cd. The Risk Assessment Code (RAC) suggested a high risk of bioavailability for Zinc and a medium risk for the other metals. 26. Phylogeographic model selection using convolutional neural networks.

The discipline of phylogeography has evolved rapidly in terms of the analytical toolkit used to analyse large genomic data sets. Despite substantial advances, analytical tools that could potentially address the challenges posed by increased model complexity have not been fully explored. For example, deep learning techniques are underutilized for phylogeographic model selection. In non-model organisms, the lack of information about their ecology and evolution can lead to uncertainty about which demographic models are appropriate. Here, we assess the utility of convolutional neural networks (CNNs) for assessing demographic models in South American lizards in the genus Norops. Three demographic scenarios (constant, expansion, and bottleneck) were considered for each of four inferred population-level lineages, and we found that the overall model accuracy was higher than 98% for all lineages. We then evaluated a set of 26 models that accounted for evolutionary relationships, gene flow, and changes in effective population size among the four lineages, identifying a single model with an estimated overall accuracy of 87% when using CNNs. The inferred demography of the lizard system suggests that gene flow between non-sister populations and changes in effective population sizes through time, probably in response to Pleistocene climatic oscillations, have shaped genetic diversity in this system. Approximate Bayesian computation (ABC) was applied to provide a comparison to the performance of CNNs. ABC was unable to identify a single model among the larger set of 26 models in the subsequent analysis. Air-breathing vertebrates undergo respiratory adjustments when faced with disturbances in the gas composition of the environment. In mammals, the medullary raphe nuclei are involved in the neuronal pathway that mediates the ventilatory responses to hypoxia and hypercarbia. We investigate whether the serotoninergic neurons of the medullary raphe nuclei of toads (Rhinella diptycha) play a functional role in respiratory control during resting conditions (room air), hypercarbia (5% CO
2 ), and hypoxia (5% O2 ). The raphe nuclei were located and identified based on the location of the serotoninergic neurons in the brainstem. We then lesioned the medullary raphe (raphe pallidus, obscurus and magnus) with anti-SERT-SAP and measured ventilation in both control and lesioned groups and we observed that serotonin (5-HT) specific chemical lesions of the medullary raphe caused reduced respiratory responses to both hypercarbia and hypoxia. In summary, we report that the serotoninergic neurons of the medullary raphe of the cururu toad Rhinella diptycha participate in the chemoreflex responses during hypercarbia and hypoxia, but not during resting conditions. 28. Epidermal Growth Factor Is Associated with Loss of Mucosae Sealing and Peri-Implant Mucositis: A Pilot Study.

This study aimed to evaluate the correlation between epidermal growth factor (EGF) and receptor (EGFR) levels in different clinical stages of dental implant rehabilitation and trace mucositis development's biological profile. Thirty-six participants from the Specialization in Implant Dentistry, Universidade Federal Fluminense, Brazil, were included in the study and underwent sample collection: inside the alveolar socket, immediately before implant placement (Group 1, n = 10); at the peri-implant crevicular fluid (PICF) during reopening (Group 2, n = 10); PICF from healthy peri-implant in function (Group 3, n = 8); and PICF from mucositis sites (Group 4, n = 18). Quantitative polymerase chain reaction (PCR) evaluated EGF/EGFR gene expression using the SYBR Green Master Mix detection system. The results showed that EGF expression in the peri-implant crevicular fluid was statistically different. There was a higher EGF expression for group C (peri-implant health) ( p = 0.04) than for the other groups. Regarding EGFR, there was no statistical difference among the groups ( p = 0.56). It was concluded that low levels of EGF gene expression in the peri-implant crevicular fluid are related to the development of peri-implant mucositis and the absence of mucosae sealing. There was no correlation between EGFR gene expression with health or mucositis.

The present study aimed to evaluate the influence of manual torque (10 Ncm) versus clinical torque (30 Ncm), which is recommended by the manufacturer, on the total length of morse cone implant abutments. Twenty specimens were prepared and distributed into two groups: group 1 with ten analogs for morse cone type implant, and group 2 with ten morse type implants, size 4.3 × 15 cm. In each group, the distance between the implant platform to the top of the prosthetic abutment (abutment height) was measured and subjected to a torque of 10 Ncm. Then, the 30 Ncm torque was applied to the same abutment, and abutment height was measured. The distance between the top of the abutment and the implant/analog base was measured. In order to verify the clinical reproducibility of the experiment, comparisons between the abutment height of the analog at 10 Ncm and the implant at 30 Ncm were performed, showing a greater discrepancy in torque for the 10 Ncm analog ( p < 0.05). In order to verify if the change in the laboratory protocol from 10 to 30 Ncm could minimize the differences in the height of the prosthetic abutments, the abutment height in groups 1 and 2 was compared with 30 Ncm, and no significant difference was observed ( p > 0.05). The data indicated that the manual torque and the torque recommended by the manufacturer influence the total length of the prosthetic abutments of morse cone implants.

30. Plastic microfibre pollution: how important is clothes' laundering?

- Abstract
Plastic microfibre pollution produced by domestic and commercial laundering of synthetic textiles has recently been incriminated in the press and the scientific literature as the main source (up to 90%) of primary microplastics in the oceans. Polyethylene terephthalate (PET) is the most common microfibre encountered. This review aims to provide updated information on worldwide plastic microfibre pollution caused by textile laundering and some possibilities for its control. Release of microfibres during domestic washing and tumble drying, their fate in wastewater treatment plants (WWTPs) and the oceans, and their environmental effects on the aquatic biota are discussed, as well as potential control methods at the levels of textile modification and laundry procedures. Environmental effects on aquatic biota are important; as a result of their small size and length-to-diameter ratio, microfibers are more effectively incorporated by organisms than other plastic particle groups. Simulation laundering studies may be useful in the development of a Standard Test Method and modification of WWTPs may reduce microfibre release into aquatic systems. However, improvements will be necessary in textile design and appliance design, and recommendations should be made to consumers about reducing their personal impact on the environment through their laundering choices, which can include appliances, fabric care products and washing conditions. Vaccines against SARS-CoV-2 continue to be developed at an astonishingly quick speed and the early ones, like Pfizer and Moderna, have been shown to be more effective than many public health scientists had dared to hope. As COVID-19 vaccine research continues to progress, the world's eyes are turning toward medicine regulators. COVID-19 vaccines need to be authorized for use in each country in which the pharmaceutical industry intends to commercialize its product. This results in a patchwork of regulations that can influence the speed at which products are launched and the standards that govern them. In this research forum article, we discuss several key questions about COVID-19 vaccine regulations that should shape research on the next stage of the pandemic response. The ubiquitous presence of contaminants in the marine environment is considered a global threat to marine organisms. Heavy metals and microplastics are two distinct classes of pollutants but there are interactions between these two stressors that are still poorly understood. We examined the potential relationship between heavy metals (Al, Cr, Mn, Fe, Ni, Cu, Zn, As, Cd, Ba, Hg, Pb) and microplastic particles in oysters sampled along the Paranaguá Estuarine System. The results suggested high levels of As and Zn in the bivalves, which are destined for human consumption. Microplastic particles were found in oysters from all sampled locations, demonstrating the spread of this pollutant in the marine environment and its ability to bioaccumulate in oysters. However, our data did not demonstrate a direct relationship between microplastics and heavy metals, suggesting that these particles are not the main route for heavy metal contamination of oysters in the Paranaguá Estuarine System., (Copyright © 2021 Elsevier Ltd. - Abstract
In pre-metamorphic tadpoles, the neural network generating lung ventilation is present but actively inhibited; the mechanisms leading to the onset of air breathing are not well understood. Orexin (ORX) is a hypothalamic neuropeptide that regulates several homeostatic functions, including breathing. While ORX has limited effects on breathing at rest, it potentiates reflexive responses to respiratory stimuli mainly via ORX receptor 1 (OX1R). Here, we tested the hypothesis that OX1Rs facilitate the expression of the motor command associated with air breathing in pre-metamorphic bullfrog tadpoles (Lithobates catesbeianus). To do so, we used an isolated diencephalic brainstem preparation to determine the contributions of OX1Rs to respiratory motor output during baseline breathing, hypercapnia and hypoxia. A selective OX1R antagonist (SB-334867; 5-25 µmol l-1) or agonist (ORX-A; 200 nmol l-1 to 1 µmol l-1) was added to the superfusion media. Experiments were performed under basal conditions (media equilibrated with 98.2% O2 and 1.8% CO2), hypercapnia (5% CO2) or hypoxia (5-7% O2). Under resting conditions gill, but not lung, motor output was enhanced by the OX1R antagonist and ORX-A. Hypercapnia alone did not stimulate respiratory motor output, but its combination with SB-334867 increased lung burst frequency and amplitude, lung burst episodes, and the number of bursts per episode. Hypoxia alone increased lung burst frequency and its combination with SB-334867 enhanced this effect. Inactivation of OX1Rs during hypoxia also increased gill burst amplitude, but not frequency. A 23-year-old woman was diagnosed with Graves' disease 5 months ago with decompensated thyroid function, for which she is taking thiamazole and propranolol. She developed progressive respiratory dyspnoea [New York Heart Association (NYHA) class III] and frequent palpitations. On emergency admission, the patient was tachypnoeic, hypotensive (77/54 mm Hg) and tachycardic (120 beats per minute), with an oxygen saturation of 94%. She also presented with cold, swollen and shaky extremities, with extended capillary filling time, and a significant reduction in heart sounds. Echocardiogram showed massive pericardial effusion compatible with cardiac tamponade. Pericardiocentesis was performed, with a drainage of 1420 mL serosanguinolent fluid, with prompt haemodynamic recovery. Analysis of the pericardial fluid showed exudates. A diagnosis of pericardial effusion secondary to Graves' disease was determined and corticotherapy, lithium carbonate, cholestyramine and phenobarbital were prescribed. An oral iodine-131 was performed and the patient showed reasonable control of the clinical manifestations of hyperthyroidism. The fragmentation of plastic materials into nanoparticles of less than 1000 nm (secondary nanoplastics) and their possible accumulation in the environment is a recent matter of concern. There are still no suitable standard methods for determining the concentrations and chemical makeup of these particles in aquatic systems and the fate and effect of nanoplastics in the aquatic environment has been little explored, although there has been research using engineered nanoparticles as models. In this review, we give a summary of the (mainly laboratory-based) studies on the influences of nanoplastics. We aim to provide an updated overview of this emerging topic, reviewing the literature mainly from 2018 onwards and considering the effects of nanoplastics on ecosystems, their uptake and transport of polluting molecules, and the challenges that are faced by workers in this area. The mussel Perna perna is one of the most used bioindicators of coastal areas and the most economically exploited species in Brazil through mariculture. In the present study, P. perna was used to investigate metal pollution in the estuarine area of Vitória Bay. Four sampling sites were located along an estuarine branch of Vitória Bay and stations were sampled during three campaigns. Trace metals in the tissues of P. perna were evaluated as well as dissolved trace metals and other ancillary variables in the water column. Dissolved Cd, Pb, Cu, Ni, and Fe concentrations surpassed the tolerance limits stablished by legislation in all the sampling campaigns. P. perna exhibited concentrations in disagreement with the Brazilian legislation for Cr and As. A general trend of higher concentrations in outer stations was observed for most metals, what suggested the occurrence of flocculation process in the lower estuary, reducing the concentrations of dissolved elements and increasing their bioavailability for the biota through the particulate form. Cd was highlighted with elevated concentrations in dissolved fraction but not detected in P. perna, probably due to chlor-complex formation under influence of more saline waters. Al, Ba, Mn, Fe, Cu, Zn, Ni, Cr, Pb, and As were considered bioavailable, once they were accumulated in the mussels' tissues. Hazard index (HI) and target cancer risk (TCR) showed that the consumption of mussels from the study area offers health risk issues, being iron and arsenic the main contributors for the high indexes.

Statement of Problem: Unsatisfactory adaptation of restorations with subgingival margins can cause problems such as accumulation of biofilm, secondary caries, and inflammation of the periodontal tissue. Therefore, special attention should be given to gingival displacement and impression procedures to optimize marginal fit., Purpose: The purpose of this systematic review and meta-analysis was to compare gingival displacement with conventional cords and cordless techniques and determine the reliability of the measurement methodologies., Material and Methods: This study followed the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) statement and identified studies through September 2018. The studies were submitted to the Cochrane risk-of-bias assessment. The gingival displacement was evaluated by using the Review Manager Software., Results: Nine studies were selected, and the most common risks of bias were random sequence generation, blinding of outcome assessment, and absence of sample size calculation. Most of the studies reported obtaining a width greater than 0.2 mm., Conclusions: The cord technique resulted in increased displacement when compared with the cordless technique. - Abstract
Paint particles are part of the increasingly important microplastics (MPs) pollution of our oceans. They contain polyurethanes, polyesters, polyacrylates, polystyrenes, alkyls and epoxies. In spite of their prevalence, paint fragments are often excluded from MP audits. This review, citing 127 references, discusses detection, characteristics, sources and ecological effects of paint fragments in our oceans, as well as the abundance of paint fragments in MP samples around the world and their colonization by marine microorganisms, which differs from that of non-paint MPs. Paint MPs arise from shipping and boating activities, road markings and external surfaces of buildings. Many paint fragments come from antifouling paints used on commercial vessels and leisure boats; these may be regarded as particular pollutants, not only containing but also leaching heavy metals and biocides. 39. Medical software applications for in-hospital insulin therapy: A systematic review.

Background: In-hospital hyperglycemia (HH) is frequent and related to higher morbidity and mortality. Despite the benefits of HH treatment, glycemic control is often poor and neglected. The use of health applications to support diagnosis and therapy is now incorporated into medical practice. Medical applications for inpatient glycemic management have potential to standardize this handling by the nonspecialist physician. However, related studies are scarce. We aim to evaluate the efficacy in inpatient glycemic control parameters of medical software applications in non-critical care settings., Methods: This systematic review on in-hospital insulin applications was performed according to PRISMA guidelines. Data were extracted in triplicate and methodological quality was verified. Specific outcomes of interest were glycemic control efficacy, hypoglycemia risk, length of in-hospital stay, integration with the electronic medical record and healthcare staff acceptance., Results: Among the 573 articles initially identified and subsequent revision of the references of each one, seven studies involving six applications were eligible for the review. A better glycemic control was reported with the use of most in-hospital insulin applications in the studies evaluated, but there was no mention of the time to reach the glycemic goal. The risk of hypoglycemia was low. Different reasons influenced the varied acceptance of the use of applications among health professionals., Conclusion: The six applications of inpatient insulin therapy in a non-critical care environment proved to be useful and safe compared to the usual management. Light/dark cycle affects the physiology of vertebrates and hypothalamic orexin neurons (ORX) are involved in this function. The breathing pattern of the green iguana changes from continuous to episodic across the light/dark phases. Since the stimulatory actions of ORX on breathing are most important during arousal, we hypothesized that ORX regulates changes of breathing pattern in iguanas. Thus, we: (1) Localized ORX neurons with immunohistochemistry; (2) Quantified cyclic changes in plasma orexin-A levels by ELISA; (3) Compared breathing pattern at rest and during hypoxia and hypercarbia; (4) Evaluated the participation of the ORX receptors in ventilation with intracerebroventricular microinjections of ORX antagonists during light and dark phases. We show that the ORX neurons of I. iguana are located in the periventricular hypothalamic nucleus. Orexin-A peaks during the light/active phase and breathing parallels these cyclic changes: ventilation is higher during the light phase than during the dark phase. However, inactivation of ORX-receptors does not affect the breathing pattern. Iguanas increase ventilation during hypoxia only during the light phase. Conversely, CO
2 promotes post-hypercarbic hyperpnea during both phases. We conclude that ORXs potentiate the post-hypercarbic (but not the hypoxic)-drive to breathe and are not involved in light/dark changes in the breathing pattern.- Published

COVID-19 has created a ramifying public health, economic, and political crisis throughout many countries in the world. While globally the pandemic is at different stages and far from under control in some countries, now is the time for public health researchers and political scientists to start understanding how and why governments responded the way they have, explore how effective these responses appear to be, and what lessons we can draw about effective public health policymaking in preparation of the next wave of COVID-19 or the next infectious disease pandemic. We argue that there will be no way to understand the different responses to COVID-19 and their effects without understanding policy and politics. We propose four key focuses to understand the reasons for COVID-19 responses: social policies to crisis management as well as recovery, regime type (democracy or autocracy), formal political institutions (federalism, presidentialism), and state capacity (control over health care systems and public administration). A research agenda to address the COVID-19 pandemic that takes politics as a serious focus can enable the development of more realistic, sustainable interventions in policies and shape our broader understanding of the politics of public health.

In this study, we analyzed the bioconcentration of Cd, Cr, Cu, Pb, Ni, and Zn in the soft tissue of transplanted oysters in two sites in the Potengi estuary for six months. Native oysters collected before and after the transplantation experiment provided the background for statistical analyses. Cd, Cr, and Ni showed a strong inverse correlation with oyster weight in both sites. Transplantation upstream of the estuary presented increasing concentrations of Zn, Cu, and Pb and condition index (CI) and decreasing trends for Cd and Ni, whereas Cr oscillated significantly. In the downstream transplantation, Cu, Pb, and Zn and the CI tended to decrease, whereas for Ni, Cd, and Cr, the concentrations increased. Spatiotemporal principal component analysis correlated these results mainly with proximity to the polluting source, seasonality, and previous exposure to heavy metals. These results helped interpret the responses provided by these biomonitors to environmental changes, whether they are natural or anthropogenic.

- Abstract
The marine environment is constantly being impacted by anthropogenic activities. Nowadays, microplastics (MPs) representing one of the most deleterious material among of all substances and material from anthropogenic origin. The Microplastics (MPs) are particles smaller than 5 mm. This study presents information on abundance, distribution, type and colour of microplastics in the bottom sediments of the continental Shelf of Rio de Janeiro State. This area is strongly impacted due to its location in front of one of the most polluted coastal bays in the Brazilian Coastline. It receives untreated sewage from an Ipanema Beach submarine emissary and also a great amount of sediments dredged from Rio de Janeiro Harbour, which strongly influences the distribution of MPs in the area. The analyses detected the presence of MP in 100% of the samples, composed mainly by secondary microplastics, and almost 50% were fibers, followed by plastic films, plastic fragments and pellets. Based on the nature of the sources of the MP, a great variety of colours was shown, dominated by four main colours: blue, white, transparent, and black, this pattern could potentially increase their bioavailability due to resemblance to prey items, especially to visual raptorial species., (Copyright © 2019 Elsevier Ltd. The means and standard deviations of roughness were evaluated for the meta-analysis using Review Manager software., Results: The 17 studies that met all inclusion criteria presented a medium risk of bias. All the treatment methods tested were able to create a roughness on the yttria-stabilized tetragonal zirconia polycrystal surface. The I2 test values presented a high heterogeneity among the studies., Conclusions: The presintered specimens submitted to airborne-particle abrasion had higher surface roughness compared with abrasion after the sintering process. Irradiation with the neodymium-doped YAG and CO2 lasers was destructive to the zirconia surfaces. - Abstract
Aim: To evaluate the efficiency of fluoride-releasing adhesives for the prevention of secondary caries on in-vitro tests., Materials and Methods: This study was performed according to the PRISMA checklist. The bibliographic research was performed to identify in-vitro studies between January 2000 and September 2017. The selected studies were submitted to bias risk assessment and data extraction. In the meta-analysis the data were evaluated using Review Manager Software., Results: The 11 studies that met all inclusion criteria showed that the most common risks of bias were: absence of the sample size calculation description, absence of the single operator protocol and blind operation of the test machine. The meta-analysis was performed with six studies that used polarised light microscopy in order to quantify the mineral content of dentine and presented the values as means and standard deviations., Conclusions: Most of the authors presented that the fluoride-containing adhesive systems had a significant effect to reduce in-vitro secondary caries. The meta-analysis did not present statistical difference between fluoride-free and fluoride-containing adhesive systems. The artificial caries challenge methodology may have affected the results.

Purpose: The purpose of this meta-analysis was to evaluate how the thickness variations in ceramic restorations of lithium disilicate and the use of different photopolymerizers influence the degree of conversion of the resin cements., Methods: This study was performed according to the PRISMA checklist. The bibliographic research was performed to identify in vitro studies until December 2017 in the databases MEDLINE via PubMed, Web of Science, and Scopus. The selected studies were submitted to bias risk assessment. In the meta-analysis the data were evaluated using Review Manager Software v.5.3, with random-effect model at a 5% significance level., Results: The meta-analysis in this study showed that despite the translucency of lithium disilicate, significant reduction of light was observed when activated through the ceramic. The degree of conversion was higher in the control groups in which the resin cement was photopolymerized without the interposition of the ceramic material., Conclusions: The thinner the ceramic material, the higher the degree of conversion. A thickness greater than 1.0 mm drastically reduces the degree of conversion of dual-curing or light-curing resins. The curing unit with irradiance of 3200 to 3505 mW/cm

Objective: The beneficial mechanical properties of zirconia, together with the adhesive techniques, provide a wide range of clinical applications, including the construction of thin structures and minimally invasive adhesive restorations., Clinical Considerations: A periodontal disease-related missing maxillary central incisor, in a pregnant patient with chronic periodontal disease in the first trimester of pregnancy, and with type II diabetes treated with resin-bonded fixed partial denture using the missing tooth cemented to a zirconia framework and then bonded to the abutment teeth., Conclusions: According to the systemic conditions presented in this case report, using a low-cost technology and the missing tooth it was possible to obtain an easier and satisfactory esthetic and functional result., Clinical Significance: The mechanical properties of zirconia made it possible to treat a pregnant patient with chronic periodontal disease in the first trimester of pregnancy, and with type II diabetes with a thin zirconia framework and minimally invasive adhesive restorations., (© 2018 Wiley Periodicals, Inc.)

- Abstract
The Pleistocenic Arc Hypothesis (PAH) posits that South American Seasonally Dry Tropical Forests (SDTF) were interconnected during Pleistocene glacial periods, enabling the expansion of species ranges that were subsequently fragmented in interglacial periods, promoting speciation. The lizard genus Lygodactylus occurs in Africa, Madagascar, and South America. Compared to the high diversity of African Lygodactylus, only two species are known to occur in South America, L. klugei and L. wetzeli, distributed in SDTFs and the Chaco, respectively. We use a phylogenetic approach based on mitochondrial (ND2) and nuclear (RAG-1) markers covering the known range of South American Lygodactylus to investigate (i) if they are monophyletic relative to their African congeners, (ii) if their divergence is congruent with the fragmentation of the PAH, and (iii) if cryptic diversity exists within currently recognized species. Maximum likelihood and Bayesian phylogenetic analyses recovered a well-supported monophyletic South American Lygodactylus, presumably resulting from a single trans-Atlantic dispersal event 29 Mya. Species delimitation analyses supported the existence of five putative species, three of them undescribed. Divergence times among L. klugei and the three putative undescribed species, all endemic to the SDTFs, are not congruent with the fragmentation of the PAH. However, fragmentation of the once broader and continuous SDTFs likely influenced the divergence of L. wetzeli in the Chaco and Lygodactylus sp. 3 (in a SDTF enclave in the Cerrado)., (Copyright © 2018 Elsevier Inc.
