The increasing use of chemical fungicides without effective control of phytopathogens has led to the development of resistance in microorganisms. As a promising alternative, products formulated with Trichoderma have emerged for their sustainable and effective potential in integrated disease management. However, the predominant formulations do not offer the necessary protection against abiotic factors. In this study, we investigated Trichoderma species encapsulated in sodium alginate through storage viability experiments and their antagonistic potential. The viability and storage conditions of the capsules were evaluated by plating at 5, 15, 30, 45, 60, and 150 days after production, kept in dry or refrigerated environments. The antagonistic potential was determined by the culture pairing method using the phytopathogen Fusarium sp. The results demonstrated that it was possible to maintain the viability of the conidia, with no differences between storage environments. Additionally, the capsule provided UV protection to the conidia encapsulated species possess antagonistic potential, inhibiting 52.54% of Fusarium sp. growth. Consequently, encapsulation is an alternative formulation method that ensures the viability of Trichoderma conidia and optimizes its use in biological control., Competing Interests: Declarations. Snake venoms have evolved in several families of Caenophidae, and their toxins have been assumed to be biochemical weapons with a role as a trophic adaptation. However, it remains unclear how venom contributes to the success of venomous species for adaptation to different environments. Here we compared the venoms from Bothrocophias hyoprora , Bothrops taeniatus , Bothrops bilineatus smaragdinus , Bothrops brazili , and Bothrops atrox collected in the Amazon Rainforest, aiming to understand the ecological and toxinological consequences of venom composition. Transcriptomic and proteomic analyses indicated that the venoms presented the same toxin groups characteristic from bothropoids, but with distinct isoforms with variable qualitative and quantitative abundances, contributing to distinct enzymatic and toxic effects. Despite the particularities of each venom, commercial Bothrops antivenom recognized the venom components and neutralized the lethality of all species. No clear features could be observed between venoms from arboreal and terrestrial habitats, nor in the dispersion of the species throughout the Amazon habitats, supporting the notion that venom composition may not shape the ecological or toxinological characteristics of these snake species and that other factors influence their foraging or dispersal in different ecological niches.

Weed coexistence with an agricultural crop can negatively affect its growth, development, and yield. From this perspective, this study aimed to evaluate weed management strategies and their effect on the agronomic parameters of cowpea cultivation in direct (SPD) and conventional (SPC) planting systems. The experiment was set up in a completely randomized block design with a split-plot arrangement with four replications. The plots received a source of variation referring to the planting systems (direct and conventional planting), and the subplots corresponded to ten weed management strategies (manual hoeing 18 days after planting (DAP); at 36 DAP; at 54 DAP; at 18 and 36 DAP; at 18 and 54 DAP; at 18 and 72 DAP; at 36 and 54 DAP; at 36-72 DAP; at 18, 36, and 54 DAP; and a control with no hoeing). Density and dry mass evaluations of the cowpea plants were performed at harvest (72 DAP) by determining the number of pods per plant, pod length, number of grains per pod, 1000-grain mass, and yield. A total of 28 species distributed in 12 botanical families were identified in the two cultivation systems. The family Poaceae showed the highest frequency, with 25% of the species identified. At the end of the assay, treatment 20 had the highest positive influence and provided significant quantitative gains to the complex of traits related to cowpea production (SPD and hoeing at 18, 36, and 54 DAP). UPGMA cluster analysis and canonical discriminant analysis were performed and allowed a better classification of the evaluated treatments. It was observed that the first two canonical variables explained 90.8% of the total variance contained in the original variables. The use of SPD with weeding at 18, 36, and 54 days after planting provides greater weed control and significant quantitative gains for the complex of characteristics related to cowpea production. The results underscore the importance of choosing the correct cropping system and implementing effective weeding practices to optimize weed control and improve crop performance.

The two-striped forest-pitviper ( Bothrops bilineatus ) is an arboreal snake that is currently represented by two subspecies ( B. b. bilineatus and B. b. smaragdinus ) that comprise a species complex, and its distribution is in the Amazon and the Atlantic Forest. The rarity of encounters with this snake is reflected in the low occurrence of cases of snakebites throughout its geographic distribution and the resulting low number of published clinical reports. However, in some areas, B. bilineatus proves to be more frequent and causes envenomations in a greater proportion. Herein, we review the main aspects of the species complex B. bilineatus , including its biology, ecology, taxonomy, morphology, genetic and molecular studies, geographic distribution, conservation status, venom, pathophysiology and clinical aspects, and epidemiology. Bites by Micrurus snakes in Brazilian Amazon represent about 0.4% of snakebite registered in that area. There is not information available about the M. annellatus snakebites In this report, we describe a case of envenoming caused by Micrurus annellatus bolivianus recorded in the municipality of Cruzeiro do Sul, state of Acre, western Brazilian Amazon. On admission, the patient was suffering from mild pain, mild edema and paraesthesia restricted to the site of the bite, darkened vision and nausea. Laboratorial evaluation revealed unclottable blood which improved only after 3 days of follow-up. Envenoming was classified as moderate, and 50 ml of antielapid serum was administered. The next day after the incident, vital signs were normal and the patient had mild thrombocytopenia (133,000/mm3). The patient did not present any bleeding during the hospitalization. On the fourth day after admission, the patient was discharged alleging no complaints. The patient showed coagulopathy, a rare condition in cases of Micrurus bites. The aim of this study was to evaluate the gene expression of proinflammatory (RANKL, TNF-a and IFN-g) and regulatory (TGF-b and IL-10) cytokines as reaction to experimental infection by mono or bi-association of Fusobacterium nucleatum (ATCC 10953) and Enterococcus faecalis (ATCC 19433). F. nucleatum and E. faecalis, either in mono- or bi-association were inoculated into the root canal system (RCS) of Balb/c mice. Animals were sacrificed at 10 and 20 days after infection and periapical tissues surrounding the root were collected. The mRNA expression of the cytokines RANKL, TNF-a, IFN- g, TGF-b and IL-10 was assessed using real-time PCR. The Kruskal-Wallis test was used for statistical analysis. F. nucleatum mono-infection induced high expression of RANKL and TNF-a, while its modulation was due to IL-10. High expression of IFN-g at day 20 was up-regulated by E. faecalis and RANKL; TNF-a was up-regulated by an independent mechanism via IL-10 and TGF-b. Bi-association (F. nucleatum and E. faecalis) stimulated high expression of RANKL, TNF-a and IFN-g, which seemed to be modulated by TGF-b 20 days later. The gene expression of proinflammatory cytokines was more prominent in the earlier periods of the experimental periapical infection, which concomitantly decreased in the later period. This expression may be regulated by IL-10 and TGF-b in an infection-specific condition.

Deep caries can induce severe inflammatory reactions. However, inflamed pulp can heal if the demineralization process is interrupted and inactivated sufficiently early. The aim of this case report is to describe the use of stepwise excavation to treat a mature permanent tooth that exhibited deep caries and apical periodontitis. A 12-year-old girl was experiencing lingering pain lasting for a few minutes in the mandibular left second molar when drinking cold water. Clinical and radiographic examinations suggested that a conservative therapeutic approach could be successful. The tooth was anesthetized and isolated, and the unsupported enamel was removed. The remaining affected dentin was left on the pulpal floor, which was protected by a thin layer of calcium hydroxide cement. The tooth was sealed with temporary cement. Three months later, pulpal sensitivity was reduced. The pulp was found to be healthy at a 9-month follow-up examination. The cavity was definitively restored with glass ionomer cement and composite resin applied with the sandwich technique. At the 4-year follow-up, the tooth remained functional, presenting standard color and satisfactory restoration. The periodontal tissues were healthy, and radiographic images indicated that the width of the periodontal ligament space was normal. This case demonstrates that clinical diagnosis should prevail over radiographic findings, even in cases where a radiographic widening of the periodontal ligament space suggests irreversible pulpal damage.

Sickle cell anemia (SCA) is the most prevalent genetic disease worldwide. Recurrent vaso-occlusive infarcts predispose SCA patients to infections, which are the primary causes of morbidly and mortality. This study aimed to evaluate the relationship between SCA and endodontic diseases. Personal information, medical data (hematological indices, virologic testing, blood transfusions, medications received, splenectomy) and information on the need for endodontic treatment were obtained from SCA patients who were registered and followed up by the Fundação Hemominas, Minas Gerais, Brazil.These data were compared with the need for root canal treatment in SCA patients. One hundred eight patients comprised the studied population, and the rate of the need for endodontic therapy was 10.2%. Among the medical data, a significant difference was observed for eosinophil (p = 0.045) counts and atypical lymphocyte counts (p = 0.036) when the groups (with and without the need for endodontic treatment) were compared. Statistical relevance was observed when comparing the patients with and without the need for root canal therapy concerned eosinophil counts and atypical lymphocyte counts. The differences in statistical medical data, observed between the groups suggest that both parameters are naturally connected to the stimulation of the immune system that can occur in the presence of root canal infections and that can be harmful to SCA individuals.

Invasive cervical resorption is an external resorption that begins below the epithelial attachment. It is caused primarily by dental trauma, orthodontic treatment, or dental bleaching. This case report involved an invasive Class III cervical resorption resulting from trauma to the superior right central incisor. Root canal treatment was followed by surgical intervention. The resorptive defect was debrided, and part of the tooth was restored with resin-modified glass ionomer cement. Postoperative follow-up revealed complete healing and healthy gingival attachment.

Introduction: Root canal treatment typically involves cleaning and shaping procedures followed by treatment with antibacterial endodontic dressing between appointments and, ultimately, 3-dimensional,hermetic filling. Chlorhexidine (CHX) is effective as an irrigation solution and is used as an endodontic dressing. The aim of this study was to examine the influence of CHX on periapical cytokine expression., Methods: Expression levels of the cytokines interferon γ, tumor necrosis factor α, interleukin (IL)-1β, IL-17A, IL-10, and the chemokine monocyte chemoattractant protein-1 (CCL2/MCP-1) were assayed by real-time polymerase chain reaction immediately after root canal cleaning and 15 days later., Results: Messenger RNA expression of IL-1β, interferon γ, IL-10, and CCL2/MCP-1 was increased on day 15 in teeth without endodontic dressing. No statistical change was observed in the messenger RNA expression of cytokines when comparing sampling times for teeth that received endodontic dressing., Conclusions: The results show that CHX application between appointments prevented the increase of both proinflammatory and immunoregulatory cytokines 15 days after the dental procedure., (Copyright © 2013 American Association of Endodontists. Introduction: The use of calcium hydroxide is an effective step in killing bacteria that remain after cleaning and shaping procedures. It also induces hard-tissue formation and is effective for stopping inflammatory exudates., Methods: The aim of this study was to assay and to compare the influence of calcium hydroxide on periapical interstitial fluid from human root canals. The mRNA expression levels of the cytokines interferon (IFN)-γ, tumor necrosis factor-α, interleukin (IL)-1β, IL-17A, and IL-10 as well as the chemokine MCP-1 were assayed by real-time polymerase chain reaction immediately after root canal cleaning and 15 days later., Results: Levels of IL-1β, IFN-γ, IL-10, and the chemokine CCL2/MCP-1 were increased in teeth without endodontic dressings. With calcium hydroxide interappointment dressings, no statistically significant changes were observed in cytokine mRNA expression. However, when comparing teeth that received the medication with those that did not, expression levels of IL-1β, IFN-γ, and IL-10 were statistically lower in those teeth that received calcium hydroxide., Conclusions: Analyses of cytokines and the chemokine CCL-2/MCP-1 demonstrated the benefits of calcium hydroxide as a root canal dressing because it impedes the increase of all mediators during the experimental time., (Copyright © 2012 American Association of Endodontists. Introduction: Failure in endodontic treatment is often caused by the persistence of microorganisms in the root canal after therapy. When treatment fails, an immune response develops that is characterized by an extensive network of immunologic mechanisms that lead to the production of cytokines and chemokines., Methods: The objective of this study was to determine the relative messenger RNA (mRNA) expression of IFN-γ, TNF-α, IL-1β, IL-17A, IL-10, and MCP-1 in periapical dental lesions refractory to treatment. Clinical samples were taken from teeth presenting periapical lesions refractory to endodontic treatment (the experimental group) or from healthy teeth with pulp vitality (the control group). Three paper points passing through the root apex (2 mm) were used to collect the samples. The total RNA was extracted from each sample, complementary DNA was synthesized, and quantitative polymerase chain reaction analysis was performed. The Mann-Whitney U test was used to determine the statistical significance of our findings (P < .05)., Results: Significant differences in the levels of IFN-γ, TNF-α, IL-17A, and MCP-1 mRNA expression were observed in cases refractory to endodontic treatment as compared with the control group. The expression of IL-1β mRNA was not significantly different between the groups. The expression of IL-10 mRNA was insignificant in both the experimental and control groups., Conclusions: A significantly increased expression of TNF-α, IFN-γ, IL-17A, and MCP-1 mRNA was observed in the periapical immune response in cases of endodontic failure.
