The causative agent of Chagas disease is Trypanosoma cruzi , which is widely distributed throughout the South American continent and extends into North America. Its occurrence in bats is poorly described and may impact the disease's maintenance and epidemiology. The aim of this study was to detect the agent by PCR assays targeting kDNA and nuclear DNA in the organs of 203 urban bats and rural vampire bats from the Brazilian Atlantic Forest, São Paulo state, during the pandemic period from 2020 to 2022. In total, 6 of the 203 bats (2.97%) were positive for T. cruzi . Infection was detected in 2% (2/101) of Desmodus rotundus , 33% (1/3) of Nyctinomops laticaudatus , 25% (1/4) of Artibeus lituratus , 4% (1/24) of Eumops glaucinus and in 2% (1/41) of Molossus molossus . The gene sequences obtained were assessed for quality and deposited in a public repository. Fruit bats were statistically associated with positivity for T. cruzi . To our knowledge, this study detected T. cruzi for the first time in bats from São Paulo state and in N. laticaudatus and E. glaucinus species.

The aim of this study was to investigate the coinfection of feline retroviruses (feline immunodeficiency virus-FIV, and the feline leukemia virus-FeLV) with Leishmania infantum and Toxoplasma gondii and the factors associated with these pathogens in domestic cats from Mossoró, a city endemic for canine and human leishmaniasis situated in the semiarid region of Northeast Brazil. Blood samples from 120 cats were collected, and an epidemiological questionnaire was applied to investigate the risk factors associated with the infections. Retroviruses, L. infantum , and T. gondii infections were assessed using a point-of-care ELISA and quantitative PCR (qPCR), indirect fluorescent antibody test (IFAT) and qPCR, and IFAT, respectively. The overall seroprevalences observed were 35% (95% CI = 27.0-43.8%) for FIV, 0.8% (95% CI = 0.1-4.5%) for FeLV, 25.8% (95% CI = 18.8-34.3%) for T. gondii , and 4.2% (95% CI = 1.7-9.3%) for L. infantum . Coinfection with FIV and L. infantum was observed in 2.5% (3/120) of the assessed cats, while 12.5% (15/120) were coinfected with FIV and T. gondii . No significant association was found among the investigated agents ( p > 0.05). The factors associated with FIV infection in the multivariable analysis were male sex and age above 78 months. The findings of this study demonstrated a high rate of FIV infection in cats from the Brazilian semiarid region and the exposure of these animals to zoonotic and opportunistic agents. Due to the immunosuppressive potential of FIV, cats infected with this retrovirus should be screened for coinfections with L. infantum and T. gondii , and preventative measures should be adopted.

Background: Leptospirosis represents a One Health issue, affecting humans and animals. This study investigated pathogenic leptospires in small wild rodents in São Paulo, Brazil., Methods: Kidney samples from 164 rodents underwent qPCR testing, targeting pathogenic Leptospira spp., Results: Thirty-five animals (21.34%) tested positive, including five rodent species: Akodon montensis (2/21; 9.5%), Necromys lasiurus (1/4; 25%), Oligoryzomys nigripes (24/92; 26.1%), Oligoryzomys flavescens (5/26; 19.2%), and Sooretamys angouya (3/14; 21.4%). Botucatu municipality exhibited the highest prevalence, with 42.5% (20/47) of the animals testing positive., Conclusions: The presence of Leptospira spp. in wild rodents suggests they may be chronic carriers, contaminating the environment.

The performance of a commercial immunofluorescence assay (IFA commercial), an in-house immunofluorescence assay (IFA in-house ) and an indirect enzyme-linked immunosorbent assay (ELISA) were evaluated in the detection of antibodies anti- C. burnetii in the serum of Q fever patients and persons without the disease. For the study, seropositive and seronegative samples for Q fever ( n = 200) from a serum bank of the Instituto Adolfo Lutz in Brazil were used. Commercial IFA was considered in this study as the gold standard for diagnosing Q fever. The in-house IFA demonstrated good agreement with the commercial test, showing high sensitivity (91%) and specificity (97%) compared to the gold standard, with a Kappa coefficient of 0.8954. The indirect ELISA test showed lower agreement with the gold standard, showing low sensitivity (67%), although the specificity of the technique was high (97%) and the Kappa coefficient was moderate (0.6631). In-house IFA is an excellent alternative for diagnosing Q fever.

Q fever and brucellosis are zoonoses that cause fever and other systemic clinical signs in humans; their occurrences are neglected and the differential diagnosis for some diseases is disregarded. This study aimed to investigate the seropositivity for Coxiella burnetii and Brucella spp. antibodies in patients suspected of dengue from 38 municipalities in the state of São Paulo, Brazil. The samples (n = 604) were obtained by convenience from the Adolfo Lutz Institute serum bank. Sera were subjected to an indirect immunofluorescence assay (IFA) using in-house and commercial diagnostic protocols to evaluate C. burnetii positivity. For Brucella spp., sera were subjected to rapid plate serum agglutination with buffered acidified antigen (AAT), slow tube serum agglutination (SAL), and 2-mercaptoethanol (2-ME) techniques. Associations and statistical inferences of the results were performed by logistic regression according to the clinical and demographic variables collected from the patients. Statistical analyses were performed using Statistical Analysis Software (SAS) and associations were considered when p value was <0.05. In all, 129 patients showed positive results for Q fever, indicating a seropositivity of 21.4% (95% CI 18.15-24.85). Patients with 14-20 days of symptoms had 2.12 (95% CI 1.34-3.35) times more chances of being seropositive for Q fever than patients with 7-13 days, and patients with 21-27 days of fever had 2.62 (95% CI 1.27-5.41) times more chances of being seropositive for Q fever than patients with 7-13 days. For the other variables analyzed, there were no significant associations between the groups. No positivity for brucellosis was observed. This is the most comprehensive study of people seropositive for Q fever in São Paulo state and provides additional data for the medical community in Brazil. Background: The early symptoms of leptospirosis and dengue fever are difficult to distinguish and can cause diagnostic confusion. Due to the large dengue epidemics that has occurred in Brazil in recent years, it is possible that cases of leptospirosis were unreported. Therefore, we performed a retrospective study to detect leptospirosis in patients who were tested for dengue, but whose laboratory diagnoses were negative., Methods: Sera samples from 2,017 patients from 48 cities located in the central region of São Paulo state, Brazil, were studied. All samples were subjected to the microscopic agglutination test (MAT), 305 of which were taken from patients five days or less since the onset of symptoms, and were additionally subjected to real-time polymerase chain reaction (PCR)., Results: The overall prevalence of leptospirosis cases was 21 (1.04%), with 20 through MAT (18 for Icterohaemorrhagiae and two for the Cynopteri serogroup) and one through PCR (amplicon sequencing compatible with Leptospira interrogans ). According to previously established criteria, eight cases of leptospirosis were classified as "confirmed" and 13 as "probable". The Brazilian notification system for health surveillance had no records for 16 patients positive for leptospirosis and, thus, they were considered unreported cases. Statistical analyses revealed that the prevalence of leptospirosis was higher in men (1.56%) than in women (0.56%), and the mean age was higher in positive patients (43.7 years) than in negative ones (32.3 years)., Conclusion: The results indicated that patients suspected of dengue fever had evidence of leptospirosis or Leptospira infection, and most of these cases were unreported in the Brazilian notification system. The high burden of dengue may contribute to the misdiagnosis of leptospirosis, and health professionals should increase their awareness of leptospirosis as an important differential diagnosis of patients with suspicion of dengue., Competing Interests: Competing interests: The authors declare no conflicts of interest.

Antibodies to pathogens of public health importance were investigated in 33 free-ranging six-banded armadillos ( Euphractus sexcinctus ) from Brazil. The frequency of seropositive animals for Toxoplasma gondii , Leishmania spp., and Leptospira spp. were two, three, and two, respectively.

Toxoplasma gondii is one of the most important protozoa parasites worldwide. Although many seroprevalence studies have been performed in domestic and wild species, data on the cumulative incidence and the spatial distribution of T. gondii in animals are extremely scarce. In the present study, dogs from Botucatu municipality, São Paulo state, were followed for one year and their blood samples were collected on three moments: days 1, 180, and 360. The sera were submitted to the immunofluorescence antibody test (IFAT) to detect IgG antibodies to T. gondii. Age and sex were compared with IFAT results through statistical tests. Spatial analysis was used to detect clusters of seropositive dogs. Among the 350 dogs that were seronegative on day 1, 53 became seropositive in subsequent samplings; thus, cumulative incidence was 15.1% exposed dogs/year. Age and sex were not associated with serological results. The spatial analysis revealed that seropositive dogs were distributed in all the studied areas, with a significant cluster in a zone with poor sanitary conditions and low socioeconomic status. T. gondii is frequent and widely distributed in the urban area of Botucatu, and impoverished areas are possibly associated with high levels of environmental contamination by this parasite.

The aim of this study was to investigate the occurrence of Leishmania spp. antibodies, and its association with feline immunodeficiency virus (FIV) and feline leukemia virus (FeLV), in domestic cats from an area endemic for canine and human leishmaniasis in Rio Grande do Norte State, Brazil. Ninety-one cats were subjected to a complete clinical exam, and blood samples were collected. An epidemiological questionnaire was used to investigate the risk factors. IgG anti-Leishmania spp. antibodies were detected by immunofluorescence antibody test (IFAT), with a cut-off value of 1:40. Polymerase chain reaction (PCR) was performed to detect genetic material of Leishmania spp. in the blood samples. The presence of antibodies against FIV and antigens of FeLV was evaluated using an immunochromatographic test. Seropositivity for Leishmania spp., FIV, and FeLV was observed in 14/91 (15.38%), 26/91 (28.57%), and 3/91 (3.29%) cats, respectively. All samples gave negative results on PCR analysis. Based on these data, no significant statistical association was observed between seropositivity for Leishmania spp., and sex, age, presence of clinical signs, evaluated risk factors, and positivity for retroviruses. These findings demonstrated for the first time that cats from Mossoró, Rio Grande do Norte, are being exposed to this zoonosis and might be part of the epidemiological chain of transmission of visceral leishmaniasis.

Antibodies against smooth Brucella were investigated in serum samples of 526 freeranging mammals (21 species) in Brazil. All animals were seronegative, which indicated lowlevel exposure to these bacteria.

Hemoparasites are vector-borne parasites that infect wild carnivores worldwide. Since data on hemoparasite infections in Nasua nasua from Brazil are lacking, the aim of this study was to investigate the occurrence of hemoparasites and ectoparasites in N. nasua from different areas of Brazil. Blood samples and ectoparasites from 83 N. nasua were collected in Botucatu, Palmital, and São Paulo municipalities. Samples were screened via microscopy and molecular methods to detect hemoparasites. Tissues from two N. nasua were obtained for histopathological and molecular analyses. All 83 samples were negative for piroplasms on morphological and molecular examination. Thin blood smears of nine animals were positive for Hepatozoon gamonts. The gamonts shared morphological characteristics of Hepatozoon procyonis. Meronts were detected in the liver and spleen tissue of one animal. Twenty-one blood samples and four tissue samples were PCR positive for Hepatozoon sp. The sequences obtained were 97% identical to those of Hepatozoon felis, Hepatozoon ursi, and Hepatozoon sp. Based on searches for similarity and morphology, we identified the sequences as belonging to H. procyonis. This study provides epidemiological data on hemoparasite infections and redescribe H. procyonis based on morphological, morphometrical, and molecular analyses.

Introduction: This study aimed to detect anti-Leptospira spp antibodies and Leptospira DNA in domestic dogs., Methods: Blood and urine from 106 dogs were evaluated by microscopic agglutination test (MAT) and polymerase chain reaction (PCR), respectively., Results: Six (5.7%) and one (1%) animals were positive by MAT and PCR, respectively., Conclusions: These results show a low prevalence of infection by Leptospira spp. The absence of positive results for the Icterohaemorrhagiae serogroup indicates the small relevance of these dogs as sources of human leptospirosis.

Leptospirosis is a disease of great importance in tropical regions. Infection occurs mainly through contact with water contaminated with the urine of infected animals, especially that of rodents. Despite the diversity and abundance of wild fauna in Brazil, little is known about the role of other wild species in the epidemiology of leptospirosis. This study aimed to investigate new reservoirs of Leptospira among wildlife in Brazil, using serological and molecular diagnoses in a large-sized sample. Biological samples were collected from 309 free-ranging mammals, belonging to 16 species. The majority of the animals included were opossums (Didelphis albiventris) and coatis (Nasua nasua). Blood and urine samples were subjected to the microscopic agglutination test (MAT) and real-time PCR, respectively. Genetic characterization of genomospecies was performed using PCR amplicons. Statistical analysis was applied to test associations between positive diagnoses and age, sex, season and type of environment. The prevalence of infection found via MAT and PCR was 11% and 5.5%, respectively. If these tests are taken to be complementary, the overall prevalence was 16%. The most common serogroups were Djasiman and Australis, while L. santarosai was the prevalent genomospecies. Significant differences in prevalence between animal species were observed. Greater risk of infection was detected among adult opossums than among young ones. The influence of each serogroup and genomospecies was tested for the same variables, and this revealed higher risk of infection by L. santarosai among male opossums than among females. The present study highlights the exposure and carrier status of several wild species in Brazil and it indicates that coatis and other carnivores are priorities for further investigations., (Copyright © 2017 Elsevier B.V. Hepatozoon species are vector-borne pathogens that infect domestic and wild animals. Marsupials of the species Didelphis albiventris are adapted to urban and peri-urban areas and act as reservoir hosts for several parasites. The present study evaluated the occurrence of infection by Hepatozoon species in synantropic D. albiventris from Botucatu, São Paulo, Brazil. Blood samples and ectoparasites from 19 D. albiventris were collected from urban and peri-urban areas. Hepatozoon spp. detection was performed by microscopy and molecular analysis. One opossum was positive for Hepatozoon spp. in microscopy analysis and PCR, while another animal was positive only in PCR. The obtained sequences were 100% identical to Hepatozoon canis. Six species of ticks and two species of fleas were detected on D. albiventris. This is the first report of H. canis in synantropic D. albiventris. In Brazil, H. canis transmission among dog populations is not well established, which highlights the importance of investigating the role that opossums might play in the epidemiology of this protozoan., (Copyright © 2017 Elsevier GmbH. The present study reports the first case of the presence of the intestinal nematode Strongyloides sp. in fecal examinations of a male Lutreolina crassicaudata, considered a synanthropic marsupial species with zoonotic potential. The Willis technique was used for the diagnosis. Presence of typical eggs of Strongyloides species in feces was detected. A fecal culture was performed to obtain larval stages, free-living adults and infective third stage larvae (L3) of this nematode after seven days, which was morphologically identified as Strongyloididae. This is the first report of infection by Strongyloides sp. in a tick-tailed opossum from Brazil.

Concerns about the interface between wildlife, domestic animals, and humans in the transmission of visceral leishmaniasis (VL) have been growing due to natural or anthropogenic environmental changes. In this context, investigations of the infection in wild mammals are important to assess their exposure to the vector and the parasite. A study of anti-Leishmania (Leishmania) infantum antibodies was carried out using the direct agglutination test (DAT) on 528 free-ranging wild mammals of 38 species from the region of Botucatu, state of São Paulo, Brazil, a municipality that has no records of the vector or of human or canine autochthony. Antibodies were detected, with a cutoff of 1:320, in 9/528 (1.7%; 95% confidence interval [CI] 0.6-2.8%) mammals of the species Callithrix jacchus, Lepus europaeus, Sphiggurus villosus, Nasua nasua, Eira barbara, and Galictis cuja, with high titers (≥1280) for the last three. These three are little-studied species, and previous records of the detection of anti-Leishmania spp. antibodies in Brazil exist only for coatis (N. nasua), whereas worldwide, infection by L. (L.) infantum has been confirmed only in hares (Le. europaeus). On the other hand, opossums and canids, the species most commonly reported to be naturally infected by L. (L.) infantum, were not seropositive. Fifty-eight (58/528; 10.9%) mammals were found to have antibody titers ranging from 20 to 160 and were not included among the seropositive animals due to the adopted cutoff. However, the possibility of infection in these animals should not be discarded, because there is no standard cutoff point for the different wild species. Our findings indicate the need for investigations into the exact role of the seropositive species in the epidemiology of VL and for effective epidemiological surveillance to prevent its expansion, because even in regions where there are no records of canine or human autochthonous cases, there may be parasite circulation among wild mammals.

Toxoplasma gondii is one of the most significant parasite, due its importance in veterinary medicine and in public health, considered a food-borne pathogens, there is no available drug treatments to eliminate it from animal tissue, this reinforce the search for a vaccine against this parasite. This study was aimed to evaluate the dynamic of the distribution of T. gondii in tissues of female Wistar rats and their milk, after the immunization by oral rote with irradiated tachyzoites. One week after pregnancy confirmation, rats was challenged by gavage with T. gondii bradyzoites, oocysts or tachyzoites of T. gondii. Forty-eight pregnant rats were grouped as follows: immunized and challenged with bradyzoites (BZ*); non-immunized and challenged with bradyzoites (BZ); immunized and challenged with oocysts (OC*); non-immunized and challenged with oocysts (OC); immunized and challenged with tachyzoites (TZ*); non-immunized and challenged with tachyzoites (TZ); only immunized (I); control group (C). After parturition, milk samples were collected for 3 weeks and then rats were sacrificed and the tissues and milk samples were researched for T. gondii parasite load determined by the quantitative PCR (qPCR). It was verified that the immunization with irradiated tachyzoites of T. gondii induced the reduction of parasitic load in muscle samples in rats challenged by bradyzoites and oocysts, although not enabled the development of sterile immunity. The detection of parasite DNA in milk was found throughout the lactation period, from immunized and non-immunized rats, however no differences were found in the parasite load caused by immunization., (Copyright © 2015. Pterothominx pulchra (Freitas, 1934) are little known gastric nematodes of Nyctinomops laticaudatus (Chiroptera: Molossidae). Information about the occurrence and host range of these parasites in Neotropical region is still scanty, and the only two morphological descriptions available in the literature are divergent about the presence or absence of a spiny spicular sheath in males, which may lead to incorrect taxonomical positioning, since this feature represents the main difference between the genera Pterothominx and Aonchotheca. Based on the absence of this morphological feature in specimens of this nematode obtained from N. laticaudatus and Nyctinomops macrotis bats captured in two municipalities in the state of São Paulo, Brazil, the present study reclassifies the aforementioned species in the genus Aonchotheca and allocates it to the subgenus Aonchotheca. Additional morphometric data and new host and locality records are also provided.

Toxoplasmosis and neosporosis are parasitic diseases of global importance. The present study had the objective to determine the influence of age, sex and breed in the prevalence of antibodies against both diseases in dogs from Brotas city, São Paulo State, Brazil. Blood samples of 342 dogs were collected, and the age, sex and breed of each animal were recorded. The serological diagnosis for toxoplasmosis and neosporosis were performed using the immunofluorescent antibody test (IFAT). The Fischer's test was used to calculate the association probability of the variables, with α = 5%. For toxoplasmosis the prevalence of antibodies was 26.9% (CI 95% 22.4-31.8%), and for neosporosis 4.97% (CI 95% 3.1-7.8%). The statistical analysis revealed a higher risk of infection for T. gondii in dogs with more than five years.

Leptospirosis is an infectious disease of worldwide importance. The development of diagnostic techniques allows sick animals to be identified, reservoirs to be eliminated and the disease prevented and controlled. The present study aimed to compare different techniques for diagnosing leptospirosis in sheep. Samples of kidney, liver and blood were collected from 465 animals that originated from a slaughterhouse. The sera were analyzed by the Microscopic Agglutination Test (MAT), and kidney and liver samples of seropositive animals were analyzed using four techniques: bacteriological culture, the Warthin Starry (WS) technique, conventional PCR (cPCR), and quantitative PCR (qPCR). With the MAT, 21 animals were positive (4.5%) to serovars Hardjo (n=12), Hebdomadis (n=5), Sentot (n=2), Wolfii (n=1) and Shermani (n=1). Titers were 100 (n=10), 200 (n=2), 400 (n=6) and 1600 (n=3). No animal was positive by bacteriological culture; four animals were positive by the WS technique in kidney samples; six animals were positive by cPCR in kidney samples; and 11 animals were positive by qPCR, eight of which in kidney samples and three in liver. The bacterial quantification revealed a median of 4.3 bacteria/μL in liver samples and 36.6 bacteria/μL in kidney samples. qPCR presented the highest sensitivity among the techniques, followed by cPCR, the WS technique and bacteriological culture. These results indicate that sheep can carry leptospires of the Sejroe serogroup, and demonstrate the efficiency of quantitative PCR to detect Leptospira spp. in tissue samples., (Published by Elsevier B.V.)

Considering that Toxoplasma gondii is a parasite of global importance which affects several animal species including humans, the current study aimed to investigate the prevalence of antibodies against T. gondii among 72 white-eared opossums (Didelphis albiventris) from Botucatu Municipality (22°53'S 48°26'W), São Paulo State, Brazil. The investigation was carried out from January 2008 to December 2009, when the animals had their blood samples collected and subjected to the modified agglutination test (MAT); 12 specimens had brain samples bioassayed in mice. Seroprevalence was 5.5% (n=4) and bioassays were negative. Older animals had higher prevalence of antibodies against T. gondii. Canine distemper virus (CDV) has been reported in several wild animal species, but there have been no reports of CDV in hoary fox (Lycalopex vetulus). This paper characterizes the first case of natural CDV infection in hoary fox, including the clinical and pathologic aspects of the disease as well as the viral strain phylogeny.

Domestic pigs are considered to be important sources of Toxoplasma gondii infection for humans. Due to the increased consumption of wild boar meat in Brazil, this species may also be an important source of the parasite. The objective of the present study was to detect T. gondii infection in 306 blood samples collected from wild boars bred in the state of São Paulo, Brazil. Samples were analyzed using the modified agglutination test (MAT), and 14 (4.5%) of them yielded positive results. Modern breeding techniques may have contributed to the low frequency of infection observed. Results indicated that wild boars were exposed to T. gondii and that the consumption of this kind of meat may represent a source of infection for humans.

Toxoplasmosis is a worldwide zoonosis caused by Toxoplasma gondii and its definitive host is the domestic and wild felids infecting human beings and other warmblooded animals. Dogs are considered a potential risk on the transmission due they can mechanically transmit oocysts to man. In this study, a retrospective analysis of toxoplasmic infection in dog serum samples sent to Serviço de Diagnóstico de Zoonoses/FMVZ-UNESP/Botucatu, SP, in the period of 1998 to 2007 was performed. During this period 1097 serum samples were analyzed by the indirect fluorescent antibody test (IFAT), with 299 (27.25%) positive. The most frequent titer was 16 (42.80%), followed by 64 (37.79%). The results indicate that T.gondii is distributed in the environment showing the role of the dog as sentinel animal to toxoplasmosis to monitor public health actions to the control of this zoonosis.
