Abstract

BackgroundAlthough native to North America, the invasion of the aphid-like grape phylloxera Daktulosphaira vitifoliae across the globe altered the course of grape cultivation. For the past 150 years, viticulture relied on grafting-resistant North American Vitis species as rootstocks, thereby limiting genetic stocks tolerant to other stressors such as pathogens and climate change. Limited understanding of the insect genetics resulted in successive outbreaks across the globe when rootstocks failed. Here we report the 294-Mb genome of D. vitifoliae as a basic tool to understand host plant manipulation, nutritional endosymbiosis, and enhance global viticulture.ResultsUsing a combination of genome, RNA, and population resequencing, we found grape phylloxera showed high duplication rates since its common ancestor with aphids, but similarity in most metabolic genes, despite lacking obligate nutritional symbioses and feeding from parenchyma. Similarly, no enrichment occurred in development genes in relation to viviparity. However, phylloxera evolved > 2700 unique genes that resemble putative effectors and are active during feeding. Population sequencing revealed the global invasion began from the upper Mississippi River in North America, spread to Europe and from there to the rest of the world.ConclusionsThe grape phylloxera genome reveals genetic architecture relative to the evolution of nutritional endosymbiosis, viviparity, and herbivory. The extraordinary expansion in effector genes also suggests novel adaptations to plant feeding and how insects induce complex plant phenotypes, for instance galls. Finally, our understanding of the origin of this invasive species and its genome provide genetics resources to alleviate rootstock bottlenecks restricting the advancement of viticulture.

Abstract

The present study investigated whether different grafting combinations (hypobiont-epibiont) of Vitis spp. influence root infection with phylloxera and thus vine biomass on potted 2-year-old plants. The study was conducted simultaneously at two locations in Slovenia (VEM) and Hungary (GF). The dormant canes of 'Johanniter' (JOH), 'Riesling' (RR) and Vitis berlandieri × Vitis riparia 'Teleki 5C' (5C) were hetero-grafted (each with each) and autografted (each with itself), so that nine plant combinations were used for the trial. The roots of the experimental plants at different ages (1–2 years) were infested with two phylloxera populations originating from two locations (VEM, GF). Plant growth was quantified 120 days after inoculation by measuring root and shoot biomass, while the extent of phylloxera infestation was assessed by the number of feeding sites (nodosities, tuberosities) and the number of larval stages of phylloxera. In most cases, the genotype of the hypobiont influenced the degree of phylloxera infestation on all roots of the two-year-old root system. At both locations, the highest number of nodosities and the highest increase in phylloxera population was observed on the autografted Teleki 5C (5C/5C). The phylloxera biotype derived from Vitis vinifera roots (GF) induced tuberosities, especially on roots of combinations where JOH and RR were used as hypobionts. No correlation was found between biomass and phylloxera infestation. The hypobiont genotype had no effect on cane biomass at the end of the growth cycle at either experimental site. Abstract

Grapevines are challenged by a range of diseases and pests, causing economic losses and requiring often costly approaches to mitigate damage. Public interest in reducing the use of chemicals is a related challenge, along with climate change. Yet, the Vitis gene pool provides vast resources for the development of genetic resistance in rootstock and scion cultivars. Traditional breeding approaches have made great strides in the development of adaptive traits, and recent access to ‘omic technologies has further facilitated the identification of useful loci along with rapid trait introgression from wild species. Moreover, marker technologies are now used to stack multiple genes for the same trait into a single genotype, a heretofore barely accessible technology. Genomic technologies are also impacting germplasm characterization, and thereby facilitating “Breeding by Design” approaches. Genetic transformation and gene-editing technologies are also applicable for both cultivar improvement as well as functional studies of genes. The landscape for acceptance of new resistant cultivars is complex and with wine grapes, subject to high degrees of regulation especially in the European Union. With rootstocks, as well as table/raisin grapes, gaining acceptance in the marketplace for new cultivars developed through either traditional or marker-assisted approaches is routine. Yet even in the highly regulated EU environment, the adoption of new wine cultivars of interspecific origins is beginning to take place in both traditional wine growing regions as well as non-traditional regions nearby.

34. Implications of Photosynthetic Performance on Vitis vinifera under Abiotic and Biotic Stress

Abstract

The effects of both temperature and infection by Daktulosphaira vitifoliae, on Vitis vinifera were investigated in two separate trials. Temperature is one of the principle environmental conditions that influences plants. From triggering many reproductive phenophases, to determining the ripeness of its fruit, to influencing how efficiently its photochemistry is, V. vinifera is extremely affected by temperature. In two different temperature chambers, of 25°C and 20°C, fluctuations in photosynthesis and gas exchange were observed. It was determined that this 5°C range was too insignificant to observe any real differences between the two treatments. Therefore, it is thought to be just along the plateau of the photosynthetic temperature optimum curve for V. vinifera. Inoculation of D. vitifoliae on V. vinifera was carried out to determine the subsequent short term phenotypic responses. It was hypothesized that the presence of D. vitifoliae, would reduce the photosynthetic operating efficiency of V. vinifera, before compensatory mechanisms, such as increased stomatal conductance, as well as adaxial stomata formation would equalize this reduction. It was found that there was no significant effect of the phylloxera on the functioning of the grapevine leaves. However, there was evidence that adaxial stomata did appear in response to the infection, something uncharacteristic for V. vinifera, as it is a hypostomatous plant (only possessing abaxial stomata). These two treatments were affected on V. vinifera to highlight its importance as one of the best candidates for climate adapted agricultural systems, given that it is a strongly plastic, and long lived perennial crop plant.

43. Pre-flowering leaf removal affects gamete viability and reduces cluster compactness in Vitis vinifera L. Grüner Veltliner

Abstract

Resumen del trabajo presentado en el International XI Symposium on Grapevine Physiology and Biotechnology, celebrado en Stellenbosch (Sudáfrica), del 31 de octubre al 5 de noviembre de 2021, Pre-flowering leaf removal (PFLR) is a management practice that involves the removal of basal leaves from selected nodes of the vine before the onset of flowering. Numerous works indicate the beneficial effect of this practice to mitigate yield loss derived from cluster rot diseases (such as gray and sour rots), particularly in grapevine cultivars with highly compact clusters. However, few of them indicate the effect of PFLR on the reproductive performance of the vine. Here, we tested the effect of two PFLR treatments applied at two different moments (one or two weeks before flowering time) on the cluster yield parameters of Grüner Veltliner, one of the most important cultivars for the Austrian wine industry. Our results indicate that PFLR treatments affect male gamete development and viability, probably due to the detrimental effect of pre-flowering carbohydrate source limitation on gametogenesis processes. This limitation affected fruit set processes, which in turn decreased the number of berries per cluster of defoliated vines. As it did not generate changes in rachis architecture or berry dimensions, the reduction in berry number had a loosening effect on Grüner Veltliner clusters. Interestingly, no effects on female gamete development were observed, as defoliated and no-defoliated plants showed a similar number of seeds per berry, suggesting some asynchrony between male and female gametes development and/or different degree of susceptibility to PFLR treatments. The lower number of berries per cluster observed in defoliated plants led to lower cluster weight compared to the undefoliated ones. However, this lower yield could be considered as an acceptable trade-off for the wine industry, as the significantly lower cluster compactness might contribute to a better sanitary status of Grüner Veltliner grapes for high-quality winemaking.
